It is shown that, the model can reasonably simulate the annual cycle and interannual variability of sea surface temperature (SST) in the tropical Pacif- ic, as well as the seasonal phase-locking of the ENSO. The model also captures two prerequisites for the E1 Nino onset, i.e., a westerly anomaly and a warm SST anomaly in the equatorial western Pacific. Owing to too strong forcing from an extratropical meridional wind, however, the westerly anomaly in this region is largely overestimated. Moreover, the simulated thermocline is much shallower with a weaker slope. As a result, the warm SST anomaly from the western Pacific propagates eastward more quickly, leading to a faster develop- ment of an E1 Nino. During the decay stage, owing to a stronger E1Nino in the model, the secondary Gill-type response of the tropical atmosphere to the eastern Pacific warming is much stronger, thereby resulting in a persistent easterly anomaly in the western Pacific. Meanwhile, a cold anomaly in the warm pool appears as a result of a lifted thermocline via Ekman pumping. Finally, an E1 Nino decays into a La Nina through their interactions. In addition, the shorter period and larger amplitude of the ENSO in the model can be attribut- ed to a shallower thermocline in the equatorial Pacific, which speeds up the zonal redistribution of a heat content in the upper ocean.展开更多

For the so-called “Atheists” originally proposed by Karl Max and Friedrich Engels, “there is a Beginning, there must be an End”, meaning our Earth will finally no longer exist in the entire Universe by colliding with the other planet. According to Holly Bible, however, Jesus, will send out his angels to separate the wicked from the righteous and throw the former into the fiery furnace. For such a special time-period, many useful ideas or outcomes can be acquired by the Internet Institutes.展开更多

文摘Downregulation of the inwardly rectifying potassium channel Kir4.1 is a key step for inducing retinal Müller cell activation and interaction with other glial cells, which is involved in retinal ganglion cell apoptosis in glaucoma.Modulation of Kir4.1 expression in Müller cells may therefore be a potential strategy for attenuating retinal ganglion cell damage in glaucoma. In this study,we identified seven predicted phosphorylation sites in Kir4.1 and constructed lentiviral expression systems expressing Kir4.1 mutated at each site to prevent phosphorylation.Following this,we treated Müller glial cells in vitro and in vivo with the m Glu R I agonist DHPG to induce Kir4.1 or Kir4.1 Tyr^(9)Asp overexpression. We found that both Kir4.1 and Kir4.1 Tyr^(9)Asp overexpression inhibited activation of Müller glial cells.Subsequently,we established a rat model of chronic ocular hypertension by injecting microbeads into the anterior chamber and overexpressed Kir4.1 or Kir4.1 Tyr^(9)Asp in the eye,and observed similar results in Müller cells in vivo as those seen in vitro. Both Kir4.1 and Kir4.1 Tyr^(9)Asp overexpression inhibited Müller cell activation, regulated the balance of Bax/Bcl-2,and reduced the m RNA and protein levels of pro-inflammatory factors, including interleukin-1βand tumor necrosis factor-α.Furthermore,we investigated the regulatory effects of Kir4.1 and Kir4.1 Tyr^(9)Asp overexpression on the release of pro-inflammatory factors in a co-culture system of Müller glial cells and microglia. In this co-culture system,we observed elevated adenosine triphosphate concentrations in activated Müller cells, increased levels of translocator protein(a marker of microglial activation),and elevated interleukin-1βm RNA and protein levels in microglia induced by activated Müller cells. These changes could be reversed by Kir4.1 and Kir4.1 Tyr^(9)Asp overexpression in Müller cells.Kir4.1 overexpression, but not Kir4.1 Tyr^(9)Asp overexpression, reduced the number of proliferative and migratory microglia induced by activated Müller cells.Collectively, these results suggest that the tyrosine residue at position nine in Kir4.1 may serve as a functional modulation site in the retina in an experimental model of glaucoma.Kir4.1 and Kir4.1 Tyr^(9)Asp overexpression attenuated Müller cell activation, reduced ATP/P2X receptor–mediated interactions between glial cells, inhibited microglial activation, and decreased the synthesis and release of pro-inflammatory factors, consequently ameliorating retinal ganglion cell apoptosis in glaucoma.

文摘Diabetic retinopathy is a prominent cause of blindness in adults, with early retinal ganglion cell loss contributing to visual dysfunction or blindness. In the brain, defects inγ-aminobutyric acid synaptic transmission are associated with pathophysiological and neurodegenerative disorders, whereas glucagon-like peptide-1 has demonstrated neuroprotective effects. However,it is not yet clear whether diabetes causes alterations in inhibitory input to retinal ganglion cells and whether and how glucagon-like peptide-1 protects against neurodegeneration in the diabetic retina through regulating inhibitory synaptic transmission to retinal ganglion cells. In the present study,we used the patch-clamp technique to recordγ-aminobutyric acid subtype A receptor-mediated miniature inhibitory postsynaptic currents in retinal ganglion cells from streptozotocin-induced diabetes model rats.We found that early diabetes(4 weeks of hyperglycemia)decreased the frequency of GABAergic miniature inhibitory postsynaptic currents in retinal ganglion cells without altering their amplitude, suggesting a reduction in the spontaneous release ofγ-aminobutyric acid to retinal ganglion cells.Topical administration of glucagon-like peptide-1 eyedrops over a period of 2 weeks effectively countered the hyperglycemia-induced downregulation of GABAergic mIPSC frequency, subsequently enhancing the survival of retinal ganglion cells.Concurrently, the protective effects of glucagon-like peptide-1 on retinal ganglion cells in diabetic rats were eliminated by topical administration of exendin-9-39,a specific glucagon-like peptide-1 receptor antagonist,or SR95531,a specific antagonist of theγ-aminobutyric acid subtype A receptor.Furthermore, extracellular perfusion of glucagon-like peptide-1 was found to elevate the frequencies of GABAergic miniature inhibitory postsynaptic currents in both ON-and OFF-type retinal ganglion cells. This elevation was shown to be mediated by activation of the phosphatidylinositol-phospholipase C/inositol 1,4,5-trisphosphate receptor/Ca2+/protein kinase C signaling pathway downstream of glucagon-like peptide-1 receptor activation. Moreover, multielectrode array recordings revealed that glucagon-like peptide-1 functionally augmented the photoresponses of ON-type retinal ganglion cells.Optomotor response tests demonstrated that diabetic rats exhibited reductions in visual acuity and contrast sensitivity that were significantly ameliorated by topical administration of glucagon-like peptide-1.These results suggest that glucagon-like peptide-1 facilitates the release ofγ-aminobutyric acid onto retinal ganglion cells through the activation of glucagon-like peptide-1 receptor, leading to the de-excitation of retinal ganglion cell circuits and the inhibition of excitotoxic processes associated with diabetic retinopathy.Collectively, our findings indicate that theγ-aminobutyric acid system has potential as a therapeutic target for mitigating early-stage diabetic retinopathy.Furthermore, the topical administration of glucagon-like peptide-1 eyedrops represents a non-invasive and effective treatment approach for managing early-stage diabetic retinopathy.

文摘The impact of spinal cord injury(SCI)on the immune system is increasingly recognized in a field traditionally focused on motor impairments.SCI can seriously affect the immune system by progressively disrupting the regulatory mechanisms that control immune responses. This dysregulation varies widely among patients and can evolve over time, ranging from systemic inflammatory responses to immunosuppression, greatly contributing to the morbidity and mortality of individuals with SCI(Bao et al.,2011;Brennan et al.,2024).
文摘Experimental therapies targeting immune and stromal cells, such as mast cells,cancer-associated fibroblasts, dendritic cells, and tumor endothelial cells,in the treatment of gastrointestinal solid tumors pose new and complex surgical and medico-legal challenges. These innovative treatments require that informed consent not be limited to simple acceptance of the medical procedure, but instead reflect a true relational and cognitive process grounded in understanding, free choice, and the ability to revoke consent at any time.In particular,it is essential that the patient understands the experimental nature of the therapy, its development stage, potential benefits and risks,as well as the implications for their health and personal dignity. In the case of stromal cell-based treatments, which may exert complex immunomodulatory effects or activate angiogenic pathways that are not yet fully understood, patients must be made fully aware that they are participating in a non-standardized therapy whose outcomes, whether beneficial or harmful, cannot yet be predicted with certainty. This requires particularly careful medical communication, using simple yet scientifically accurate explanations delivered in appropriate language, along with a final verification of the patient’s actual understanding.

文摘Based on reanalysis data from 1979 to 2021,this study explores the spatial distribution of the Southern Indian Ocean Dipole(SIOD)and its individual and synergistic effects with the El Niño-Southern Oscillation(ENSO)on summer precipitation in China. The inverse phase spatial distribution of sea surface temperature anomalies(SSTAs)in the southwest and northeast of the southern Indian Ocean is defined as the SIOD.Positive SIOD events(positive SSTAs in the southwest, negative SSTAs in the northeast)are associated with La Niña events(Central Pacific(CP)type),while negative SIOD events(negative SSTAs in the southwest, positive SSTAs in the northeast)are associated with El Niño events(Eastern Pacific(EP)type). Both SIOD and ENSO have certain impacts on summer precipitation in China.Precipitation in the Yangtze River basin decreases, while precipitation in southern China increases during pure positive SIOD(P_PSIOD)events.During pure negative SIOD(P_NSIOD)events,the changes in precipitation are exactly the opposite of those during P_PSIOD events, which may be due to differences in the cross-equatorial flow in the southern Indian Ocean, particularly in low-level Australian cross-equatorial flow. When positive SIOD and CP-type La Niña events occur simultaneously(PSIOD+La_Niña),precipitation increases in the Yangtze-Huaihe River basin, while it decreases in northern China. When negative SIOD and EP-type El Niño events occur simultaneously(NSIOD+El_Niño),precipitation in the Yangtze-Huaihe River basin is significantly lower than during P_NSIOD events. This is caused by differences in water vapor originating from the Pacific Ocean during different events.

文摘On the basis of more than 200-year control run, the performance of the climate system model of Chinese Academy of Sciences (CAS-ESM-C) in simulating the E1 Nifio-Southern Oscillation (ENSO) cycle is evalu- ated, including the onset, development and decay of the ENSO. In addition, the shorter period and larger amplitude of the ENSO in the model can be attribut- ed to a shallower thermocline in the equatorial Pacific, which speeds up the zonal redistribution of a heat content in the upper ocean.

文摘Supported by the spatial analysis feature of geographic information science and assessment model of regional debris flows, hazards degrees of the debris flows in the Upper Yangtze River Watershed (UYRW) are divided into five grades based on grid cell. The area of no danger, light danger, medium danger, severe danger and extreme severe danger regions respectively are 278 000, 288 000, 217 000, 127 000, 15 000 km^2. Furthermore, the counties in the UYRW are classified into four classes based on the hazards degrees in each county. The number of severe danger, medium danger, light danger and no danger counties respectively are 49, 82, 77 and 105. The assessment results will be provided for the hazards forecasting and mitigation in the UYRW and ongoing regionalization of Main Function Regions in China as data and technique framework.
文摘Resilience is the psychological capability to recover from difficulties quickly.Healthcare professionals are especially vulnerable to job-related stress and burnout.Unitary Caring Science is the framework for Watson’s Human Caring Theory, providing a philosophy of practice in healthcare. With the high rates of clinician burnout and psychological issues,it will be significant to unify the human caring theory with research-informed psychological and neuroscience evidence to develop clinicians’resilience-building strategies. The purpose of this article is to introduce a Unitary Caring Science Resilience Model and explain the science behind the core strategies based on Unitary Caring Science philosophy and the psychological and neuroscience research. This model includes six strategies:Embracing loving-kindness for self and others;Nurturing interpersonal and intersubjective connections/relations;Deepening a creative use of self and sense of belonging;Balancing self-learning,self-awareness,and an evolved selfconsciousness;Valuing forgiveness and releasing negativity;Inspiring and maintaining faith-hope.The caring-theory guided resilience-building strategies are proven to alleviate the depletion of clinicians’energy and emotions.Healthcare practices are challenging but rewarding.Clinicians can be emotionally, psychologically, and physically exhausted if they always consider themselves‘giving’and‘doing’institutional tasks without a sense of purpose or fulfillment. The practice can be rewarding if it becomes more aligned with clinicians’value to serve humanity.Through the unitary caring science resilience strategies, clinicians can build resilience as an antidote to clinician burnout and depletion.
文摘It is extremely fearful for the pestilences covering our Earth. Does that mean the “World End” is around the corner? For the so-called “Atheists” originally proposed by Karl Max and Friedrich Engels, “there is a Beginning, there must be an End”, meaning our Earth will finally no longer exist in the entire Universe by colliding with the other planet. According to Holly Bible, however, Jesus, will send out his angels to separate the wicked from the righteous and throw the former into the fiery furnace. For such a special time-period, many useful ideas or outcomes can be acquired by the Internet Institutes.
文摘The damage of coronavirus to our Earth is unprecedented that is going to kill the entire human beings. Facing this kind of situation, there are two completely different kinds of viewpoints. According to the philosophy of “Atheists” Karl Max and Friedrich Engels, “there is a Bigging, there must be an End”, meaning our Earth will eventually collide with the other planet and be destroyed completely. According to Holly Bible, however, Jesus, will come down again to bring Godly people to the Heaven and leave the remaining in the coronavirus-hell. During this waiting period, the “Internet Institutes” can provide the most useful knowledge for the other planets.
文摘The most unethical behaviors in science are of falsifying data and stealing ideas from previous investigators. But for publishing papers with high similarity and editing papers with coercion, it is necessary to carry out a concrete analysis case by case.

文摘Lithium-oxygen(Li-O2)batteries are perceived as a promising breakthrough in sustainable electrochemical energy storage, utilizing ambient air as an energy source, eliminating the need for costly cathode materials, and offering the highest theoretical energy density(~3.5 k Wh kg^(-1))among discussed candidates.Contributing to the poor cycle life of currently reported Li-O_(2)cells is singlet oxygen(1O_(2))formation,inducing parasitic reactions, degrading key components, and severely deteriorating cell performance. Here,we harness the chirality-induced spin selectivity effect of chiral cobalt oxide nanosheets(Co_(3)O_(4)NSs)as cathode materials to suppress 1O_(2)in Li-O_(2)batteries for the first time.Operando photoluminescence spectroscopy reveals a 3.7-fold and 3.23-fold reduction in 1O_(2)during discharge and charge, respectively, compared to conventional carbon paperbased cells, consistent with differential electrochemical mass spectrometry results, which indicate a near-theoretical charge-to-O_(2)ratio(2.04 e-/O_(2)).Density functional theory calculations demonstrate that chirality induces a peak shift near the Fermi level, enhancing Co 3d-O 2p hybridization, stabilizing reaction intermediates, and lowering activation barriers for Li_(2)O_(2)formation and decomposition. These findings establish a new strategy for improving the stability and energy efficiency of sustainable Li-O_(2)batteries,abridging the current gap to commercialization.
