Total P (TP), total participate P (PP), total dissolved P (TDP), molybdatereactive P (MRP) and dissolved organic P (DOP) were determined in waters from pipe-drains (at 65-cmdepth) from the Broadbalk Experiment at Rothamsted Research, UK. Average TPand PP exceeded 1 mgL^(-1) in about half of the 12 plots receiving superphosphate for the 5 measurements taken betweenDecember 2000 and April 2001. Ranging between 33.8% and 87.3% of TP, PP was the largest P fractionin drainage waters, with DOP, ranging from 0.5% to 26.2% of TP, being the smallest fraction Meanproportions of PP, MRP and DOP in TP in drainage waters were 63.4%, 32.5% and 4.1%, respectively.These findings support previous findings that P losses from soil to drainage waters were muchlargerthan previously thought, and could therefore make a significant contribution to eutrophication. 展开更多

Remobilisation of nitrate in plants, especially in vacuole of plant, is mostly related to the qua- lity of agricultural products and the high nitrogen use efficiency in plants. Ion-selective microelectrodes offer a non-destructive and non-interruptive method to measure NO 3 gradients and electric potential differences across both the plasma membrane and tonoplast. Thus, a double-barrelled microelectrode backfilled with a membrane sensor for NO 3 embedded in poly vinyl chloride (PVC) can record the NO 3 activity in cytoplasm and vacuole of a cell. This paper presented how to make this kind of microelectrode and how to do the intracellular measurements on intact plants. Our result showed that nitrate activity was about 2.7 mmol L 1 in cytoplasm while 70 mmol L 1 in vacuole, which implicated that vacuole was a pool of nitrate in plants. 展开更多

嗅觉在疟蚊寄主发现行为过程中起主要作用.基于生物信息学技术,对按蚊嗅觉结合蛋白基因进行了全基因组分析,结果总共鉴定了32条嗅觉结合蛋白(OBP)候选基因序列.进一步采用半定量反转录聚合酶链反应(semi-quantitativeRT-PCR),以疟蚊激... 嗅觉在疟蚊寄主发现行为过程中起主要作用.基于生物信息学技术,对按蚊嗅觉结合蛋白基因进行了全基因组分析,结果总共鉴定了32条嗅觉结合蛋白(OBP)候选基因序列.进一步采用半定量反转录聚合酶链反应(semi-quantitativeRT-PCR),以疟蚊激动蛋白基因为内部表达参照标准,研究了冈比亚按蚊所有嗅觉结合蛋白候选基因的组织特异性表达谱.结果显示:其中20个OBP候选基因在嗅觉组织(雌蚊触角)中有强的表达,这说明OBP在疟蚊嗅觉行为中起重要作用.还研究了冈比亚按蚊复合体(A.gambiaecomplex)中两种最重要的疟蚊,即冈比亚按蚊(A.gambiae)和阿拉伯按蚊(A.arabiensis)的蚊种嗅觉特异性表达型,结果发现其中12个OBP候选基因显示种间差异表达,同时发现阿拉伯按蚊触角中OBP的累积相对表达强度高于冈比亚按蚊,其比例为1441.45︰1314.12.这可能是由二者的不同寄主偏好行为所致,因为冈比亚按蚊高度嗜吸人血,而阿拉伯按蚊则介于嗜吸人血和嗜畜血之间,因此后者需要表达更多的OBPs以支持其寄主选择行为模式.疟蚊OBP基因的鉴定及其组织和蚊种特异性表达型的确定,是分析疟蚊嗅觉探测的分子机制的第一步,如嗅觉结合蛋白重组表达及其配体鉴定. 展开更多

Chemosensory proteins (CSPs) are identifiable by four spatially conserved Cys-teine residues in their primary structure or by two disulfide bridges in their tertiary structure according to the previously identified olfactory specific-D related proteins. A genomics- and bioinformatics-based approach is taken in the present study to identify the putative CSPs in the malaria-carrying mosquito, Anopheles gambiae. The results show that five out of the nine annotated candidates are the most possible Anopheles CSPs of A. gambiae. This study lays the foundation for further functional identification of Anopheles CSPs, though all of these candidates need additional experimental verification. 展开更多
