Background Although 15 mA transcranial alternating current stimulation(tACS)has a therapeutic effect on depression,the activations of brain structures in humans accounting for this tACS configuration remain largely un...Background Although 15 mA transcranial alternating current stimulation(tACS)has a therapeutic effect on depression,the activations of brain structures in humans accounting for this tACS configuration remain largely unknown.Aims To investigate which intracranial brain structures are engaged in the tACS at 77.5 Hz and 15 mA,delivered via the forehead and the mastoid electrodes in the human brain.Methods Actual human head models were built using the magnetic resonance imagings of eight outpatient volunteers with drug-naïve,first-episode major depressive disorder and then used to perform the electric field distributions with SimNIBS software.Results The electric field distributions of the sagittal,coronal and axial planes showed that the bilateral frontal lobes,bilateral temporal lobes,hippocampus,cingulate,hypothalamus,thalamus,amygdala,cerebellum and brainstem were visibly stimulated by the 15 mA tACS procedure.Conclusions Brain-wide activation,including the cortex,subcortical structures,cerebellum and brainstem,is involved in the 15 mA tACS intervention for first-episode major depressive disorder.Our results indicate that the simultaneous involvement of multiple brain regions is a possible mechanism for its effectiveness in reducing depressive symptoms.展开更多
Objective Long noncoding RNAs(lncRNAs)and microRNAs(miRNAs)are widely expressed in the brain and are associated with the development of neurological and neurodegenerative diseases.However,their roles and molecular mec...Objective Long noncoding RNAs(lncRNAs)and microRNAs(miRNAs)are widely expressed in the brain and are associated with the development of neurological and neurodegenerative diseases.However,their roles and molecular mechanisms in major depressive disorder(MDD)remain largely unknown.This study aimed to identify lncRNAs and miRNAs involved in the development of MDD and elucidate their molecular mechanisms.Methods Transcriptome and bioinformatic analyses were performed to identify miRNAs and lncRNAs related to MDD.C57 mice were subjected to chronic unpredictable mild stress(CUMS)to establish a depression model.Lentiviruses containing either lncRNA NPTN-IT1-201 or miR-142-5p were microinjected into the hippocampal region of these mice.Behavioral tests including the sucrose preference test(SPT),tail suspension test(TST),and forced swim test(FST)were conducted to evaluate depressive-like behaviors.Results The results revealed that overexpression of lncRNA NPTN-IT1-201 or inhibition of miR-142-5p significantly ameliorated depressive-like behaviors in CUMS-treated mice.Dual-luciferase reporter assays confirmed interactions between miR-142-5p with both brain-derived neurotrophic factor(BDNF)and NPTN-IT1-201.ELISA analysis revealed significant alterations in relevant biomarkers in the blood samples of MDD patients compared to healthy controls.Histological analyses,including HE and Nissl staining,showed marked structural changes in brain tissues following CUMS treatment,which were partially reversed by lncRNA NPTN-IT1-201 overexpression or miR-142-5p inhibition.Immunofluorescence imaging demonstrated significant differences in the levels of BAX,Bcl2,p65,Iba1 among different treatment groups.TUNEL assays confirmed reduced apoptosis in brain tissues following these interventions.Western blotting showed the significant differences in BDNF,BAX,and Bcl2 protein levels among different treatment groups.Conclusion NPTN-IT1-201 regulates inflammation and apoptosis in MDD by targeting BDNF via miR-142-5p,making it a potential therapeutic target for MDD.展开更多
Background With the continuous improvement of people's material living standards,the consumption of alcoholic beverages is increasing.Alcohol dependence caused by alcohol addiction has become a widespread concern ...Background With the continuous improvement of people's material living standards,the consumption of alcoholic beverages is increasing.Alcohol dependence caused by alcohol addiction has become a widespread concern in society.As the brand-new medical and health model created by the modern biomedical technology revolution and the information technology revolution,mobile health has been used more and more widely in the field of medical health with the characteristics of convenience,rapidity and generalisability.With the general use of the social software WeChat,it is feasible to implement mobile health on the WeChat platform.Aim We aimed to explore the clinical effect of WeChat platform used in the cognitive behavioural therapy(CBT)on the maintenance period of alcohol dependence and addiction.Methods Seventy alcohol-dependent patients in the withdrawal maintenance stage admitted into our hospital were randomly divided into the CBT on WeChat platform group(the experiment group,n=35)and the general clinical treatment using sertraline group(the control group,n=35)with block randomisation grouping.They were evaluated with the Severity of Alcohol Dependence Questionnaire(SADQ-C)before the treatment,at the end of the fourth week,at the end of the eighth week and at the end of the twelfth week of treatment.In addition,Zung's Self-Rating Anxiety Scale(SAS)and Self-Rating Depression Scale(SDS)were employed to evaluate the degree of anxiety and depression of the two groups.Results The SADQ-C,SAS and SDS scores of the two groups after treatment decreased markedly compared with those before treatments,and the differences were statistically significant.Between-group comparison:the SADQ-C scores of the experiment group at the end of the fourth,eighth and twelfth weeks were lower than those of the control group,and the difference was statistically significant,which indicated that the score reduction in the experiment group was larger than that in the control group.The SAS and SDS scores of the experiment group at these time points were not significantly different from those of the control group,which indicated that these two treatment methods were comparable in treating patients1 anxiety and depression.Conclusion The CBT in tervention using WeChat may improve the subjectively reported severity of alcohol dependence of patients who had completed detoxification.It is conducive to prevention of relapse,and is convenient for patients.It is worthy of clinical application and further study.展开更多
Objective:The purpose of the research was to systematically evaluate the clinical efficacy and safety of Shenkang injection combined with Western medicine in treating chronic glomerulonephritis.Methods:Randomized cont...Objective:The purpose of the research was to systematically evaluate the clinical efficacy and safety of Shenkang injection combined with Western medicine in treating chronic glomerulonephritis.Methods:Randomized controlled trials of Shenkang Injection combined with Western medicine in treating CGN were obtained from 8 Science databases including China National Knowledge Infrastructure(CNKI),China Biomedical Database(SinMed),Weipu Database(VIP),Wanfang Database(Wanfang),PubMed,The Cochrane Library,Embase,Web of Science from establishment to June 2022.Two researchers screened literature and extracted useful data according to inclusion and exclusion criteria,and used RevMan 5.3 and STATA SE 14.0 software to perform meta-analysis.Results:1,214 patients were included in 15 studies,with 607 in treatment group and 607 in control group.The results of meta-analysis showed that the treatment group could improve the clinical effective rate[RR=1.33,95%CI(1.25,1.40),P<0.00001],reduce serum creatinine[MD=-31.28,95%CI(-42.90,-19.66),P<0.00001],blood urea nitrogen[MD=-2.26,95%CI(-3.08,-1.44),P<0.00001],24-hour urine protein quantification[MD=-0.37,95%CI(-0.49,-0.25),P<0.00001],tumor necrosis factor-α[MD=-7.93,95%CI(-10.56,-5.30),P<0.00001],systolic blood pressure[MD=-10.81,95%CI(-13.66,-7.96),P<0.00001]and diastolic blood pressure[MD=-7.36,95%CI(-9.34,-5.38),P<0.00001].Conclusion:Shenkang injection combined with Western medicine can enhance the clinical effective rate of CGN and reduce the levels of serum creatinine,blood urea nitrogen,24-hour urine protein quantification,tumor necrosis factor-α,systolic blood and diastolic blood pressure.However,due to the low quality of literature,the conclusion still needs to be supported by more multi-center,large-sample studies with rigorous design and standardized implementation.展开更多
