Development of a Functional Food Product Using Guavas

Guava Cheese (GC) is a semi-solid... The global market of functional foods and demand for a healthy lifestyle among consumers in recent years is growing. Guava is a highly perishable fruit and a rich source of vitamin C. Guava Cheese (GC) is a semi-solid concentrated fruit product that could be consumed as a snack. The objectives were to develop a guava cheese (GC) and determine the antioxidant potential of the product. Formulation consisted of guava puree with added sucrose or agave, chia seeds and almonds. Pectin was added at select concentrations (0.5%, 1%, 1.5%) along with citric acid to enhance the consistency of GC. Physiochemical and sensory parameters were analyzed for extended shelf life (3 months) studies. Moisture content, pH, water activity, color (L*, a*, b*) and texture profile analysis (hardness, springiness, cohesiveness, gumminess, chewiness) did not change over the 90-day period. Ash, protein and fat contents of 0% pectin were 2% lower than GC with pectin. Sensory parameters (firmness, mouthfeel, flavor and overall acceptability) of GC 0% pectin and 1% GC were similar. Total content in 0%, 0.5%, 1%, and 1.5% pectin added GC were 150.49 ± 32.76, 340.17 ± 54.65, 346.39 ± 53.04, and 355.72 ± 14.24 mg gallic acid equivalent (GAE)/100g. Flavonoid content in 0%, 0.5%, 1%, and 1.5% GC were 159.73 ± 13.31, 332.77 ± 13.31, 341.65 ± 15.37, and 350.52 ± 16.60 mg catechin equivalent (CE)/100g. Similarly, antioxidant of potential measure by DPPH radical scavenging was similar in all samples (IC50 at 0.8 mg/ml). Guava cheese may be utilized as a healthy fruit snack because of added alternative sweeteners and functional ingredients to obtain health benefits. Adding pectin to guava cheese improved functionality by increasing antioxidant potential as well as physical properties. 展开更多

Objective:As an age-related neurodegenerative disease,the prevalence of mild cognitive impairment(MCI)increases with age.Within the framework of traditional Chinese medicine,spleen-kidney deficiency syndrome(SKDS)is recognized as the most frequent MCI subtype.Due to the covert and gradual onset of MCI,in community settings it poses a significant challenge for patients and their families to discern between typical aging and pathological changes.There exists an urgent need to devise a preliminary diagnostic tool designed for community-residing older adults with MCI attributed to SKDS(MCI-SKDS).Methods:This investigation enrolled 312 elderly individuals diagnosed with MCI,who were randomly distributed into training and test datasets at a 3:1 ratio.Five machine learning methods,including logistic regression(LR),decision tree(DT),naive Bayes(NB),support vector machine(SVM),and gradient boosting(GB),were used to build a diagnostic prediction model for MCI-SKDS.Accuracy,sensitivity,specificity,precision,F1 score,and area under the curve were used to evaluate model performance.Furthermore,the clinical applicability of the model was evaluated through decision curve analysis(DCA).Results:The accuracy,precision,specificity and F1 score of the DT model performed best in the training set(test set),with scores of 0.904(0.845),0.875(0.795),0.973(0.875)and 0.973(0.875).The sensitivity of the training set(test set)of the SVM model performed best among the five models with a score of 0.865(0.821).The area under the curve of all five models was greater than 0.9 for the training dataset and greater than 0.8 for the test dataset.The DCA of all models showed good clinical application value.The study identified ten indicators that were significant predictors of MCI-SKDS.Conclusion:The risk prediction index derived from machine learning for the MCI-SKDS prediction model is simple and practical;the model demonstrates good predictive value and clinical applicability,and the DT model had the best performance. 展开更多

BACKGROUND Dental follicle stem cell(DFSC)sheets demonstrate strong extracellular secretion capabilities and efficacy in periodontal regeneration.However,existing methods for producing DFSC sheets lack a comprehensive discussion on the most efficient and cost-effective approaches at the good manufacturing practice(GMP)level.AIM To investigate the culture condition of GMP-compliant DFSC sheets and to compare the properties of DFSC sheets and cell suspensions.METHODS This study explored the optimal conditions for culturing GMP-compliant DFSC sheets,focusing on four key factors:Cell passage,cell concentration,L-ascorbic acid content,and culture duration.We evaluated the characteristics of the cell sheets under varying culture conditions,including cell viability,cell count,appearance,osteogenesis,chondrogenesis,odontogenesis,aging,relative telomere length,and extracellular matrix secretion.A comparison was also made between the periodontal regeneration,osteogenesis,and paracrine capacity of cell sheets cultured under optimal conditions and those of the cell suspensions.RESULTS The GMP-compliant DFSC sheets cultured from passage 4 cells exhibited the highest viability(≥99%,P<0.05)and optimal osteogenic differentiation capacity(optical density≥0.126,P<0.05).When cultured for 10 days,DFSC sheets demonstrated maximal expression of osteogenic,chondrogenic and periostin genes[alkaline phosphatase,Runt-related transcription factor 2,collagen type I,osteopontin,cartilage associated protein,and PERIOSTN(P<0.001);osteocalcin(P<0.01)].Concurrently,they showed the lowest senescent cell count(P<0.01)with no progression to late-stage senescence.At a seeding density of 2500 cells/cm^(2),GMP-compliant DFSC sheets achieved better osteogenic differentiation(P<0.01)and maximal osteogenic,chondrogenic and periostin gene expression(P<0.001),coupled with the highest hydroxyproline secretion(P<0.001)and moderate sulfated glycosaminoglycan production.No statistically significant difference in senescent cell count was observed compared to DFSC sheets at a seeding density of 5000 cells/cm^(2).Supplementation with 25μg/mL L-ascorbic acid significantly enhanced osteogenic gene expression(P<0.001)and elevated hydroxyproline(P<0.01)and sulfated glycosaminoglycan secretion to high ranges.Compared with the cell suspension,the cell sheet demonstrated improved osteogenic,paracrine,and periodontal regenerative capacities in Sprague-Dawley rats.The optimized DFSC sheets demonstrated significantly higher levels of vascular endothelial growth factor and angiopoietin-1(P<0.001)compared to DFSC suspensions,along with enhanced osteogenic induction outcomes(optical density=0.1333±0.01270 vs 0.1007±0.0005774 in suspensions,P<0.05).Following implantation into the rat periodontal defect model,micro-computed tomography analysis revealed superior bone regeneration metrics in the cell sheet group compared to both the cell suspension group and control group(percent bone volume,trabecular thickness,trabecular number),while trabecular spacing exhibited an inverse pattern.CONCLUSION Optimized DFSC sheets cultured under the identified conditions outperform DFSC suspensions.This study contributes to the industrial-scale production of DFSC sheets and establishes a foundation for cell therapy applications. 展开更多

Current methods for nasal spray formulations have been elementary evaluating the dripping characteristics of a formulation and have not assessed the behavior of the nasal formulation in the presence of varying types of mucus depending on the indication or diseased state. This research investigated the effects of nasal mucus on the dripping behavior of nasal formulations and focused on developing an improved in vitro analytical test method that is more physiologically relevant in characterizing nasal formulation dripping behavior. Method development was performed using simulated nasal mucus preparations for both healthy and diseased states as coatings for the dripping experiment representing a wide range of viscosity. Factors evaluated during development of this in vitro test method included amount of mucus, application of mucus, drying times, and compatibility of the mucus on a C18 Thin Layer Chromatography(TLC) substrate. The dripping behavior of nasal formulations containing a range of 1%Avicel to 3.5% Avicel was assessed by actuating the nasal spray on a perpendicular TLC plate coated with either healthy or diseased simulated nasal mucus. After actuation of the nasal spray, the dripping of the formulation on the coated TLC plate was measured after the plate was repositioned vertically. The method that was developed generated reproducible results on the dripping behavior of nasal formulations and provided critical information about the compatibility of the formulation with the nasal mucus for different diseased states, aiding in nasal spray formulation development and physical characterization of the nasal spray. 展开更多

Cholangiocarcinoma(CCA), the adenocarcinoma of the biliary duct, is commonly reported in Asia with the highest incidence in northeastern Thailand. Chemotherapy of this type of cancer is limited due to the lack of effective chemotherapeutic drugs. A series of previous studies support further research and development of Atractylodes lancea(Thunb) DC.(AL) as a potential candidate for the treatment of CCA as a crude ethanolic extract. In the present study, we aimed to develop an oral pharmaceutical formulation(capsule) of the standardized AL crude ethanolic extract for further clinical development in patients with CCA. Major steps included macroscopic and microscopic authentication of the AL rhizomes, preparation of standardized AL extract, preparation of oral pharmaceutical formulation(capsule) of the standardized AL extract, quantitative and qualitative analysis of the marker compound(atractylodin) in the formulated AL extract, evaluation of contaminations of heavy metals, pesticides residues, and microorganisms in the ground AL rhizomes and the formulated(capsule) powder of AL, physicochemical and pharmaceutical properties of the formulated AL extract/capsule, and cytotoxicity evaluation of the formulated AL extract. One of the most interesting ways to improve mechanical properties is to reduce secondary dendrite arm spacing (SDAS).SDAS also plays an important role in controlling and providing the well distributed and fine microstructure resulting in better tensile strength and elongation.To reduce SDAS,it is commonly known by increasing cooling rate and increasing interface instability by limited-soluble alloy addition.It is,however, unclear that how both cooling rate and limited-soluble alloy,e.g.Sb,relate to each other.This may be the reason that the limited-soluble alloy may not effectively reduce SDAS.To better understand this phenomenon, influences of Sb on solid/liquid interface instability using columnar to equiaxed transition (CET) were studied in the directionally solidification experiment.From macrographs and micrographs,it was observed that at 0.06-0.18 wt pct Sb the CET min,CET max,CET zone and %CET area gradually increased.The increases of CET max and CET zone in samples with 0.06 to 0.18 wt pct Sb addition results from recalesced zone.On the other hand,the variations of CET max and CET zone in samples with 0.24 to 0.30 wt pct Sb results from equiaxed grain formations that prohibit the growth of columnar grain and latent heat from intermetallic phase solidified. 展开更多

Latent dimensions between water use and socio-economic development: A global exploratory statistical analysis

Water use and socio-economic development are interconnected in complex ways.Causalities are not easy to identify but it is evident that a nexus between water use and socio-economic development does exist.Considering the diversity of national situations relating to these interrelated phenomena,its study should be considered from a global perspective.This article critically reviews the literature and information from official sources on the relevance of water use and circular economy in order to create a global picture,linking water with socio-economic development.Data from 195 countries were analyzed statistically.A factor analysis defined five essential latent dimensions on the nexus between water use and socio-economic development:development and basic services,population and resources,economic volume,health and well-being,and population density.Based on the identified factors,countries were classified into six groups:Global South in difficulty,global semi-periphery,advanced economy,Middle East and other Global South developing economy,global weight,and small highly developed economy.The clustering results clarify connections between water use conditions and socioeconomic development.Understanding the variety of national profiles is helpful to reveal the magnitude and urgency of dealing with the nexus between water use and socio-economic development for many countries. 展开更多

Extraction and Powder Product of Fructo-oligosaccharide from Jerusalem Artichoke

It can not be digested by enzymes in the body. It also helps to improve a digestion in the intestine, an excretion system and prevent the cancer. Jerusalem artichoke is scanned and analyzed by High Performance Liquid Chromatography (HPLC) for quantity of FOS. They are selected for extraction and drying process. Studies on the extraction for a powder making, the conditions are optimum for maximum yield. Experiments manage using a factorial real 2 × 4 × 6 in 2 blocks of Randomized Complete Block Design (RCBD) models. The first factor studies on two types of solvent. The second factor is the four levels of temperature in the extraction with water at 25 ℃, 35 ℃, 50℃ and 60℃ for 30 min. The third factor is the ratio of sample per solvent as 5, 6, 10, 15, 20 and 30 times. The data and comparison of average is analyzed by Duncan's New Mutiple Range Test at the significant level 0.05. The concentrated extracts are processed to powder by freeze drying, a hot air and vacuum drying. Optimal conditions control the temperature and time by heating in both vacuum and conventional oven. The results of temperature on the extraction are significantly different at a = 0.05. In drying process with high temperature, the color of product is more yellow (b) and less white (L). The difference of color value is statistically significant at level a = 0.05. The sensory evaluation of food products are added the extracted powder and attribute by panelist. High level acceptance of product is aspectual about texture and flavor. FOS is the trend of functional foods for health. It is focus on low energy and on obesity including increasing the absorption of calcium. 展开更多

During high-speed rotation,the surface of aeronautic spiral bevel gears will generate significant pressure and viscous forces,which will cause a certain amount of windage power loss and reduce the efficiency of the transmission system.Based on the computational fluid dynamics,this paper analyzes the windage power loss of a single spiral bevel gear and a spiral bevel gear pair under oil injection lubrication.In addition,the shroud is used to suppress gear windage loss,and the clearance size and opening angle of the designed shroud are optimized.Finally,by comparing and analyzing the experimental results,the following conclusions were obtained:(1)For a single gear,the speed is the most important factor affecting windage loss,followed by the hand of spiral,and rotation direction;(2)For gear pairs,under oil injection lubrication,the input speed has the greatest impact on windage power loss,followed by the influence of oil injection port speed,temperature and oil injection port pressure;(3)Installing a shroud is an effective method to reduce windage power loss;(4)In the pure air phase,the smaller the clearance between the shroud and the gear surface,and the smaller the radial direction between the shroud and the shaft,the better the effect of reducing windage;(5)In the two-phase flow of oil and gas,it is necessary to design oil drainage holes on the shroud to ensure the smooth discharge of lubricating oil and improve the drag reduction effect. 展开更多

Thin-film lithium niobate(TFLN)possesses great potential because it enables high-speed modulation by voltage,which allows higher resolution and lower power consumption for laser beam scanning than direct laser modulation.To achieve these functions,a red,green,and blue(RGB)multiplexer using TFLN is required as an important building block for photonic integrated circuits.We fabricated an RGB multiplexer using TFLN and experimentally confirmed its operation.Three different laser lights of red(λ=638 nm),green(λ=520 nm),and blue(λ=473 nm)were successfully coupled as a single laser beam by an RGB multiplexer composed of multimode interferometers.Furthermore,the TFLN was fabricated by sputter deposition,whereas conventionally,it is fabricated via bulk-lithium niobate adhesion to the substrate.The sputterdeposited TFLN is advantageous for large-volume mass production. 展开更多

Aqueous Zinc-metal batteries(AZBs)hold great promise for energy storage applications,yet their practical deployment is hindered by challenges such as dendrite formation and parasitic side reactions at the Zn anode.Herein,we developed a three-dimensional Cu-coated flexible host via an electroless plating strategy on cotton cloth(Cu@CT).This design effectively homogenizes the local current density,spatially regulates Zn-ion flux,and accommodates substantial volume changes during cycling.Additionally,the zincophilic Cu coating facilitates Zn nucleation and deposition by forming Cu-Zn alloys,which reduce the Zn nucleation overpotential and promote uniform Zn plating.As a result,the Cu@CT based anode exhibits highly reversible Zn plating/stripping behavior with an average Coulombic efficiency of 99.58%over 800 cycles,accompanied by low polarization and dendrite-free behavior.Moreover,the Zn-I_(2) full cell demonstrates excellent rate capability,delivering a discharge capacity of 114 mA h g^(-1) at 10 A g^(-1),along with stable long-term cycling performance over 950 cycles.The electroless plating strategy may represent a promising pathway for advancing high-performance AZBs. 展开更多

Organohydrogel-based strain sensors are gaining attention for real-time health services and human-machine interactions due to their flexibility,stretchability,and skin-like compliance.However,these sensors often have limited sensitivity and poor stability due to their bulk structure and strain concentration during stretching.In this study,we designed and fabricated diamond-,grid-,and peanut-shaped organohydrogel based on positive,near-zero,and negative Poisson’s ratios using digital light processing(DLP)-based 3D printing technology.Through structural design and optimization,the grid-shaped organohydrogel exhibited record sensitivity with gauge factors of 4.5(0–200%strain,ionic mode)and 13.5/1.5×10^(6)(0-2%/2%-100%strain,electronic mode),alongside full resistance recovery for enhanced stability.The 3D-printed grid structure enabled direct wearability and breathability,overcoming traditional sensor limitations.Integrated with a robotic hand system,this sensor demonstrated clinical potential through precise monitoring of paralyzed patients’grasping movements(with a minimum monitoring angle of 5°).This structural design paradigm advanced flexible electronics by synergizing high sensitivity,stability,wearability,and breathability for healthcare,and human-machine interfaces. 展开更多

Layer-by-layer tear film measurement in patients with dry eye and meibomian gland dysfunction

AIM:To evaluate parameters measured using the tear film imager(TFI)prototype,a new technology that enables to quantify the tear film thickness of lipid and mucoaqueous layers.METHODS:In this cross-sectional study,patients with dry eye,meibomian gland dysfunction(MGD),and non-dry eye/MGD from February 2020 to January 2021 were analyzed.Quantified TFI outputs included lipid layer thickness(LLT),mucoaqueous layer thickness(MALT),MALT rate of change(MALTR),and lipid breakup time.Two other interferometry devices,LipiView2 and DR-1α,were used for comparison.TFI outputs and other clinical parameters were analyzed using correlation coefficients.Each patient underwent one or several study visits.Baseline values of three device outputs,other clinical parameters,and their changes were examined.RESULTS:This study involved 28 patients(8 patients with dry eyes,13 with MGD,and 7 with non-dry eye/MGD).Baseline TFI,LipiView2,and DR-1αvalues were associated with various clinical parameters.The LLT values estimated using TFI had a correlation with the plugging score in the upper eyelid(r=−0.42).Several TFI values have correlated better than LipiView2 and DR-1α,particularly with questionnaire scores.MALTR by TFI revealed a correlation between standardized patient evaluation on eye dryness(SPEED)and dry eye-related quality of life score(DEQS)scores(r=0.59,0.43),respectively.CONCLUSION:TFI enabled to quantify the LLT and MALT separately over time and shows the moderate correlations between TFI measurements and clinical parameters,which yields the potential for TFI to serve as a complementary tool for assessing dry eye and MGD. 展开更多

Micro-alloying is an effective approach for improving the corrosion resistance of cast AZ91.However,the effect of micro-alloyed elements on corrosion resistance can be varied depending on the solidification rate influencing the diffusion and precipitation behavior of micro-alloying elements.This study investigated the effects of the cooling rate on the microstructure and corrosion behavior of micro-Ca and-Y alloyed cast AZ91 alloy(i.e.,AZXW9100).To achieve various cooling rates,the alloys were prepared using three methods:steel mold casting(SMC),copper step mold casting(CSMC),and high-pressure die casting(HPDC).The corrosion behavior was analyzed through weight loss measurements,electrochemical impedance spectroscopy,and corrosion morphology observations.The results showed that the key microstructural factors influencing corrosion resistance differed between short-and long-term corrosion.As the cooling rate increased,the short-term corrosion rate was lowered from 0.91 mm/y(SMC)to 0.38 mm/y(HPDC),which was attributed to the decrease in the total area fractions of the eutecticαandβphases acting as galvanic corrosion sources.The long-term corrosion rate was reduced from 17.20 mm/y(SMC)to 0.71 mm/y(HPDC),which was revealed to be due to the enhanced connectivity of theβphase acting as corrosion barriers.Meanwhile,the increase in the cooling rate led to a modification of the Zn molar ratio in theβphase,reducing the Volta potential of theβphase from 101.8 m V to 66.9 m V.This reduction in the Volta potential of the main galvanic source also contributed to improved corrosion resistance.The HPDC AZXW9100 alloy produced in this study exhibited the lowest corrosion rate compared to other alloys.These findings suggest that controlling the cooling rate is a promising strategy for enhancing the corrosion resistance of AZXW9100 alloys. 展开更多

Quantitative scale validation of the Dimensional Anhedonia Rating Scale in the treatment of Chinese patients with major depressive disorder

Background The patient-reported Dimensional Anhedonia Rating Scale(DARS)has been adapted into Chinese,so there is a need to evaluate its measurement properties in a Chinese population.Aims To evaluate the reliability and validity of the DARS among Chinese individuals with major depressive disorder(MDD)and its treatment sensitivity in a prospective clinical study.Methods Data were from a multicentre,prospective clinical study(NCT03294525),which recruited both patients with MDD,who were followed for 8 weeks,and healthy controls(HCs),assessed at baseline only.The analysis included confirmatory factor analysis,validity and sensitivity to change.Results Patients’mean(standard deviation(SD))age was 34.8(11.0)years,with 68.7%being female.75.2%of patients with MDD had melancholic features,followed by 63.8%with anxious distress.Patients had experienced MDD for a mean(SD)of 9.2(18)months.DARS scores covered the full range of severity with no major floor or ceiling effects.Confirmatory factor analysis showed adequate fit statistics(comparative fit index 0.976,goodness-of-fit index 0.935 and root mean square error of approximation 0.055).Convergent validity with anhedonia-related measures was confirmed.While the correlation between the DARS and the Hamilton Depression Rating Scale was not strong(r=0.31,baseline),the DARS was found to differentiate between levels of depression.Greater improvements in DARS scores were seen with the Hamilton Rating Scale for Depression responder group(effect size 1.16)compared with the non-responder group(effect size 0.46).Conclusions This study comprehensively evaluated the measurement properties of the DARS using a Chinese population with MDD.Overall,the Chinese version of DARS demonstrates good psychometric properties and has been found to be responsive to change during antidepressant treatment.The DARS is a suitable scale for assessing patient-reported anhedonia in future clinical trials. 展开更多
