BACKGROUND Proximal gastrectomy(PG)for gastric cancer requires a delicate balance between oncological radicality and postoperative quality of life to mitigate reflux complications.Although the Kamikawa anastomosis has...BACKGROUND Proximal gastrectomy(PG)for gastric cancer requires a delicate balance between oncological radicality and postoperative quality of life to mitigate reflux complications.Although the Kamikawa anastomosis has gained attention for its theoretical anti-reflux advantages,robust clinical evidence remains limited.AIM To comprehensively evaluate the efficacy,safety,and nutritional outcomes of the Kamikawa anastomosis in PG,thereby addressing a critical gap in surgical decision-making.METHODS Following PRISMA guidelines,we systematically searched PubMed,Embase,and the Cochrane Library for studies on Kamikawa anastomosis after PG.Data were pooled using fixed-or random-effects models based on heterogeneity levels(I2 statistics).The risk of bias was evaluated using Risk of Bias in Non-randomized Studies of Interventions.The protocol was prospectively registered in PROSPERO.RESULTS Of 106 screened studies,20 involving 2291 patients were included.Most studies(85%)originated from Japan,with a mean patient age of 54-73 years and a male predominance(68.1%).Overall pooled incidence of 30-day postoperative complications was 9.9%[95%confidence interval(95%CI):6.8-12.9],with major complications(Clavien-Dindo grade≥III)occurring in 6.1%(95%CI:4.6-7.7).Anastomosis-related complications were observed in 7.2%of cases,comprising leakage 1.8%(95%CI:1.1-2.4),stenosis in 7.2%(95%CI:5.8-8.5),and bleeding in 0.7%(95%CI:0.1-1.2).Pooled incidence of reflux esophagitis was 4%(95%CI:2.7-5.3)for all LA grades at 12-month follow-up,with 28.3%(95%CI:14.7-41.9)of patients requiring regular proton pump inhibitor use.Operative outcomes demonstrated a pooled mean operative time of 349.5 minutes(95%CI:331-380),estimated blood loss of 351.5 mL(95%CI:264-495),and postoperative hospital stay of 12.3 days(95%CI:11.5-13.1).Nutritional outcomes revealed 11.4%(95%CI:10.6-12.2)body weight loss at one year.Comparative analyses showed equivalent safety profiles between the Kamikawa anastomosis and double-tract reconstruction but longer operative times(P<0.001)compared with total gastrectomy.CONCLUSION These findings underscore the clinical viability of the Kamikawa anastomosis following PG,demonstrating favorable anti-reflux efficacy,acceptable incidence of anastomotic strictures,and preservation of nutritional status.展开更多
The aim of this work is to find an alternative lubricating grease formulation that can be produced from renewable and biodegradable sources with minimal risks to human health and the environment.We used a castor oil a...The aim of this work is to find an alternative lubricating grease formulation that can be produced from renewable and biodegradable sources with minimal risks to human health and the environment.We used a castor oil and electrospun cellulose acetate propionate(CAp)as raw materials.We hypothesized that the acetyl and propionyl groups could provide an adequate chemical compatibility with the castor oil and that the electrospun nanostructures could enable improved physical stability by creating a variety of morphologies allowing the tailoring of the rheological and tribological properties of the resulting greases.The experimental results show that the use of electrospun CAp nanostructures can indeed yield physically stable formulations,even when used at low concentrations(3 wt%).The resulting dispersions went through structural transitions due to changes in the thickener morphologies and/or concentration,as shown by oscillatory rheology,oil holding capacity,tackiness,and lubrication performance in metal–metal contact.We found that the formulations,containing smooth or porous CAp nanofibers,at 5 wt%as a thickener,possess suitable rheological and tribological properties with a performance comparable to that of traditional lithium lubricating greases.展开更多

METHODS: H pylo...AIM: To evaluate whether celecoxib, a selective cyclooxygenase 2 (COX-2) inhibitor, could reduce the severity of gastric precancerous lesions following Hel/cobacter pylori (H pylorl) eradication. METHODS: H pylori-eradicated patients with gastric precancerous lesions randomly received either celecoxib (n = 30) or placebo (n = 30) for up to 3 mo. COX-2 expression and activity was determined by immunostaining and prostaglandin E2 (PGE2) assay, cell proliferation by Ki-67 immunostaining, apoptosis by TUNEL staining and angiogenesis by microvascular density (MVD) assay using CD31 staining.RESULTS: COX-2 protein expression was significantly increased in gastric precancerous lesions (atrophy, intestinal metaplasia and dysplasia, respectively) compared with chronic gastritis, and was concomitant with an increase in cell proliferation and angiogenesis. A significant improvement in precancerous lesions was observed in patients who received celecoxib compared with those who received placebo (P 〈 0.001). Of these three changes, 84.6% of sites with dysplasia regressed in patients treated with celecoxib (P = 0.002) compared with 60% in the placebo group, suggesting that celecoxib was effective on the regression of dysplasia. COX-2 protein expression (P 〈 0.001) and COX-2 activity (P 〈 0.001) in the gastric tissues were consistently lower in celecoxib-treated patients compared with the placebo-treated subjects. Moreover, it was also shown that celecoxib suppressed cell proliferation (P 〈 0.01), induced cell apoptosis (P 〈 0.01) and inhibited angiogenesis with decreased MVD (P 〈 0.001). However, all of these effects were not seen in placebo-treated subjects. Furthermore, COX-2 inhibition resulted in the up-regulation of PPARy expression, a protective molecule with anti-neoplastic effects. CONCLUSION: H pylori eradication therapy followed by celecoxib treatment improves gastric precancerous lesions by inhibiting COX-2 activity, inducing apoptosis, and suppressing cell proliferation and angiogenesis.展开更多
Chlorine has been widely used in different advanced oxidation processes(AOPs)for micropollutants removal.In this study,different chlorine-based AOPs,namely medium pressure(MP)UV/chlorine,low pressure(LP)UV/chlorine,an...Chlorine has been widely used in different advanced oxidation processes(AOPs)for micropollutants removal.In this study,different chlorine-based AOPs,namely medium pressure(MP)UV/chlorine,low pressure(LP)UV/chlorine,and in-situ chlorination,were compared for carbamazepine(CBZ)removal efficiency,energy consumption,and disinfection by-products(DBPs)formation.All three processes could achieve nearly 100%CBZ removal,while the reaction time needed by in-situ chlorination was double the time required by UV/chlorine processes.The energy consumed per magnitude of CBZ removed(EE/O)of MP UV/chlorine was 13 times higher than that of LP UV/chlorine,and relative to that of in-situ chlorination process.Accordingly,MP and LP UV/chlorine processes generated one to two orders of magnitude more hydroxyl radicals(^(·)OH)and reactive chlorine species(RCS)than in-situ chlorination.Besides,RCS were the dominant reactive species,contributing to 78.3%,75.6%,and 71.6% of CBZ removal in MP,LP UV/chlorine,and in-situ chlorination,respectively.According to the Gibbs free energy barriers between CBZ and RCS/^(·)OH calculated based on density functional theory(DFT),RCS had more reaction routes with CBZ and showed lower energy barrier in the main CBZ degradation pathways like epoxidation and formation of iminostilbene.When applied to secondary wastewater effluent,UV/chlorine and in-situ chlorination produced overall DBPs ranging from 104.77 to 135.41μg/L.However,the production of chlorate during UV/chlorine processes was 15 times higher than that during in-situ chlorination.展开更多

The degenerative brain disease, chronic traumatic encephalopathy (CTE), presents clinically with memory loss, aggression, difficulty in rational thinking and other cognitive problems. This spectrum, which mimics Alzheimer’s disease, is diagnosed post-mortem through a brain biopsy in many professional athletes. However, little is known about the process of development and how to identify vulnerable individuals who may be on course for developing CTE. Boxing is a sport that has a severe toll on athletes’ health, primarily on their brain health and function. This review addresses the concerns of brain injury, describes the pathologies that manifest in multiple scales, e.g., molecular and cognitive, and also proposes possible diagnostic and prognostic markers to characterize the early onset of CTE along with the aim to identify a starting point for future precautions and interventions.展开更多

The designed grafts are characterized by the physical properties as well as mechanical properties according to ISO 7198 ∶ 1998 Standard.Moreover,the process of the grafts fabrication will be explained considering the stages affected the critical properties of the elaborated textile implants. The increase of PLAGA melt-electrospun tubular structures mechanical properties was observed due to the thermal stabilization use. The ethylene oxide( EO) and steam sterilization had a destructive effect on the designed PLAGA meltelectrospun tubular, fibrous structures, whereas the irradiation yielded in the changes in mechanical and physical properties on the acceptable level,taking into account the future clinical applications.展开更多

Then, the recombinant plasmid pET32a-htra was transformed into E. coli BL21 competent cells. The over-expression of His-HtrA protein was induced by the addition of IPTG at a final concentration of 1 mmol/L, and verified by SDA-PAGE and Western blot. In addition, the protein was purified with Ni-NTA agarose, and used to immunize mice,and the polyclonal antibody specifically bound to E. coli HtrA protein. The results will provide a theoretical basis for studying the roles of HtrA protein and developing vaccines for fur-bearing animals against E. coli.展开更多

METHODS: The antitumor efficacy of SG300 in hepatocellular carcinoma was assessed in vitro and in vivo. In vitro cell viability by MTT assay was used to assess the tumor-selective oncolysis and safety features of SG300, and in vivo antitumor activity of SG300 was assessed in established hepatocellular carcinoma models in nude mice. RESULTS: SG300 could lyse hepatocellular carcinoma cells at a low multiplicity of infection (MOI), but could not affect growth of normal cells even at a high MOI. Both in Hep3B and SMMC-7721 xenograft models of hepatocellular carcinoma, SG300 had an obvious antitumor effect, resulting in a decrease in tumor volume. Its selective oncolysis to tumor cells and safety to normal cells was also superior to that of ONYX-015. Pathological examination of tumor specimens showed that SG300 replicated selectively in cancer cells and resulted in apoptosis and necrosis of cancer cells. CONCLUSION: hTERT promoter-regulated replicativeadenovirus SG300 has a better cancer-selective replication-competent ability, and can specifically kill a wide range of cancer cells with positive telomerase activity, and thus has better potential for targeting therapy of hepatocellular carcinoma.展开更多
Glial cells play an important role in signal transduction,energy metabolism,extracellular ion homeostasis and neuroprotection of the central nervous system.However,few studies have explained the potential effects of e...Glial cells play an important role in signal transduction,energy metabolism,extracellular ion homeostasis and neuroprotection of the central nervous system.However,few studies have explained the potential effects of exosomes from glial cells on central nervous system health and disease.In this study,the genes expressed in exosomes from astrocytes and microglia were identified by deep RNA sequencing.Kyoto Encyclopedia of Genes and Genomes analysis indicated that several pathways in these exosomes are responsible for promoting neurodegenerative diseases,including Alzheimer's disease,Parkinson's disease and Huntington's disease.Gene ontology analysis showed that extracellular exosome,mitochondrion and growth factor activity were enriched in exosomes from the unique astrocyte group,while extracellular exosome and mitochondrion were enriched in exosomes from the unique microglia group.Next,combined with the screening of hub genes,the protein-protein interaction network analysis showed that exosomes from astrocytes influence neurodegenerative diseases through metabolic balance and ubiquitin-dependent protein balance,whereas exosomes from microglia influence neurodegenerative diseases through immune inflammation and oxidative stress.Although there were differences in RNA expression between exosomes from astrocytes and microglia,the groups were related by the hub genes,ubiquitin B and heat shock protein family A(Hsp70) member 8.Ubiquitin B appeared to be involved in pleiotropic regulatory functions,including immune regulation,inflammation inhibition,protein catabolism,intracellular protein transport,exosomes and oxidative stress.The results revealed the clinical significance of exosomes from glia in neurodegenerative diseases.This study was approved by the Animal Ethics Committee of Nantong University,China(approval No.S20180102-152) on January 2,2018.展开更多
The inflammatory microenvironment and neurotoxicity can hinder neuronal regeneration and functional recovery after spinal cord injury.Ruxolitinib,a JAK-STAT inhibitor,exhibits effectiveness in autoimmune diseases,arth...The inflammatory microenvironment and neurotoxicity can hinder neuronal regeneration and functional recovery after spinal cord injury.Ruxolitinib,a JAK-STAT inhibitor,exhibits effectiveness in autoimmune diseases,arthritis,and managing inflammatory cytokine storms.Although studies have shown the neuroprotective potential of ruxolitinib in neurological trauma,the exact mechanism by which it enhances functional recovery after spinal cord injury,particularly its effect on astrocytes,remains unclear.To address this gap,we established a mouse model of T10 spinal cord contusion and found that ruxolitinib effectively improved hindlimb motor function and reduced the area of spinal cord injury.Transcriptome sequencing analysis showed that ruxolitinib alleviated inflammation and immune response after spinal cord injury,restored EAAT2 expression,reduced glutamate levels,and alleviated excitatory toxicity.Furthermore,ruxolitinib inhibited the phosphorylation of JAK2 and STAT3 in the injured spinal cord and decreased the phosphorylation level of nuclear factor kappa-B and the expression of inflammatory factors interleukin-1β,interleukin-6,and tumor necrosis factor-α.Additionally,in glutamate-induced excitotoxicity astrocytes,ruxolitinib restored EAAT2 expression and increased glutamate uptake by inhibiting the activation of STAT3,thereby reducing glutamate-induced neurotoxicity,calcium influx,oxidative stress,and cell apoptosis,and increasing the complexity of dendritic branching.Collectively,these results indicate that ruxolitinib restores glutamate homeostasis by rescuing the expression of EAAT2 in astrocytes,reduces neurotoxicity,and effectively alleviates inflammatory and immune responses after spinal cord injury,thereby promoting functional recovery after spinal cord injury.展开更多
Targeted genotyping is an extremely powerful approach for the detection of known genetic variations that are biologically or clinically important.However,for non-model organisms,large-scale target geno-typing in a cos...Targeted genotyping is an extremely powerful approach for the detection of known genetic variations that are biologically or clinically important.However,for non-model organisms,large-scale target geno-typing in a cost-effective manner remains a major challenge.To address this issue,we present an ultrahigh-multiplex,in-solution probe array-based high-throughput diverse marker genotyping(HD-Marker)approach that is capable of targeted genotyping of up to 86000 loci,with coverage of the whole gene repertoire,in what is a 27-fold and six-fold multiplex increase in comparison with the conventional Illumina GoldenGate and original HD-Marker assays,respectively.We perform extensive analyses of var-ious ultrahigh-multiplex levels of HD-Marker(30 k-plex,56 k-plex,and 86 k-plex)and show the power and excellent performance of the proposed method with an extremely high capture rate(about 96%)and genotyping accuracy(about 96%).With great advantages in terms of cost(as low as 0.0006 USD per geno-type)and high technical flexibility,HD-Marker is a highly efficient and powerful tool with broad appli-cation potential for genetic,ecological,and evolutionary studies of non-model organisms.展开更多

Effective health care teams include members who have defined, yet intersecting roles, where mutual respect characterises professional interaction and the patient’s well-being is central. The aim was to explore the perception of psychologists, psychiatry registrars and psychiatrists with respect to GPs’ role in managing difficult-to-treat-depression (DTTD). Methods: A previously developed semi-structured interview schedule comprising six questions was used. Thirty-two health professionals participated. Data were analysed using the Framework method. Findings: Four main themes emerged: 1) The team approach was important, particularly to ensure information accuracy and/or when responding to patient needs and pressures;2) Referrals, usually generated by GPs can be a vehicle for other health professionals to provide advice to the GP;3) Availability and accessibility often depended on health professionals work location and knowing how to navigate the system;4) Limited availability of government funding impacts on patients’ accessibility to health professionals. Discussion: Interprofessional relationships were described as paramount. Appropriate and timely referrals are integral to patient management, regardless of challenges. Ongoing challenges include program funding, workforce numbers and costs to patients. Improvement to mental health care access was noted, even for patients among relatively disadvantaged groups and those receiving Medicare Benefits Schedule-subsidised services. Conclusion: Despite adequate GP/specialist communication, the delivery of optimal team based care to patients with difficult-to-treat depression is compromised by lack of access to specialised services and inadequate funding.展开更多

文摘AIM: To evaluate whether celecoxib, a selective cyclooxygenase 2 (COX-2) inhibitor, could reduce the severity of gastric precancerous lesions following Hel/cobacter pylori (H pylorl) eradication. METHODS: H pylori-eradicated patients with gastric precancerous lesions randomly received either celecoxib (n = 30) or placebo (n = 30) for up to 3 mo. COX-2 expression and activity was determined by immunostaining and prostaglandin E2 (PGE2) assay, cell proliferation by Ki-67 immunostaining, apoptosis by TUNEL staining and angiogenesis by microvascular density (MVD) assay using CD31 staining.RESULTS: COX-2 protein expression was significantly increased in gastric precancerous lesions (atrophy, intestinal metaplasia and dysplasia, respectively) compared with chronic gastritis, and was concomitant with an increase in cell proliferation and angiogenesis. A significant improvement in precancerous lesions was observed in patients who received celecoxib compared with those who received placebo (P 〈 0.001). Of these three changes, 84.6% of sites with dysplasia regressed in patients treated with celecoxib (P = 0.002) compared with 60% in the placebo group, suggesting that celecoxib was effective on the regression of dysplasia. COX-2 protein expression (P 〈 0.001) and COX-2 activity (P 〈 0.001) in the gastric tissues were consistently lower in celecoxib-treated patients compared with the placebo-treated subjects. Moreover, it was also shown that celecoxib suppressed cell proliferation (P 〈 0.01), induced cell apoptosis (P 〈 0.01) and inhibited angiogenesis with decreased MVD (P 〈 0.001). However, all of these effects were not seen in placebo-treated subjects. Furthermore, COX-2 inhibition resulted in the up-regulation of PPARy expression, a protective molecule with anti-neoplastic effects. CONCLUSION: H pylori eradication therapy followed by celecoxib treatment improves gastric precancerous lesions by inhibiting COX-2 activity, inducing apoptosis, and suppressing cell proliferation and angiogenesis.

文摘Chlorine has been widely used in different advanced oxidation processes(AOPs)for micropollutants removal.In this study,different chlorine-based AOPs,namely medium pressure(MP)UV/chlorine,low pressure(LP)UV/chlorine,and in-situ chlorination,were compared for carbamazepine(CBZ)removal efficiency,energy consumption,and disinfection by-products(DBPs)formation.All three processes could achieve nearly 100%CBZ removal,while the reaction time needed by in-situ chlorination was double the time required by UV/chlorine processes.The energy consumed per magnitude of CBZ removed(EE/O)of MP UV/chlorine was 13 times higher than that of LP UV/chlorine,and relative to that of in-situ chlorination process.Accordingly,MP and LP UV/chlorine processes generated one to two orders of magnitude more hydroxyl radicals(^(·)OH)and reactive chlorine species(RCS)than in-situ chlorination.Besides,RCS were the dominant reactive species,contributing to 78.3%,75.6%,and 71.6% of CBZ removal in MP,LP UV/chlorine,and in-situ chlorination,respectively.According to the Gibbs free energy barriers between CBZ and RCS/^(·)OH calculated based on density functional theory(DFT),RCS had more reaction routes with CBZ and showed lower energy barrier in the main CBZ degradation pathways like epoxidation and formation of iminostilbene.When applied to secondary wastewater effluent,UV/chlorine and in-situ chlorination produced overall DBPs ranging from 104.77 to 135.41μg/L.However,the production of chlorate during UV/chlorine processes was 15 times higher than that during in-situ chlorination.
文摘The observed images of the asteroid and the asteroid reference images are used to obtain the probe-to-asteroid direction and the location of the limb features of the asteroid in the inertial coordinate. These informa-tion in combination with the shape model of the asteroid and attitude information of the probe are utilized to ob-tain the position of the probe. The position information is then input to the UKF which determines the real-timeorbit of the probe. Finally, the autonomous orbit determination algorithm is validated using digital simulation.The determination of orbit using UKF is compared with that using extended Kalman filter (EKF), and the resultshows that UKF is superior to EKF.
文摘Brain damage sustained from repeated blows in boxing, wrestling, and other combat sports has serious physical and mental health consequences. The degenerative brain disease, chronic traumatic encephalopathy (CTE), presents clinically with memory loss, aggression, difficulty in rational thinking and other cognitive problems. This spectrum, which mimics Alzheimer’s disease, is diagnosed post-mortem through a brain biopsy in many professional athletes. However, little is known about the process of development and how to identify vulnerable individuals who may be on course for developing CTE. Boxing is a sport that has a severe toll on athletes’ health, primarily on their brain health and function. This review addresses the concerns of brain injury, describes the pathologies that manifest in multiple scales, e.g., molecular and cognitive, and also proposes possible diagnostic and prognostic markers to characterize the early onset of CTE along with the aim to identify a starting point for future precautions and interventions.

文摘The general object of research is to design new idea of the semi-biodegradable,multilayered vascular prosthesis used for the reconstruction of significantly small diameter vascular vessels( microreconstruction). The paper studies the selection of appropriate sterilization technique for resorbable tubular structures made of poly( L-lactide-co-glicolide)( PLAGA) and nonbiodegradable tubular structures made of polypropylene( PP). The designed grafts are characterized by the physical properties as well as mechanical properties according to ISO 7198 ∶ 1998 Standard.Moreover,the process of the grafts fabrication will be explained considering the stages affected the critical properties of the elaborated textile implants. The increase of PLAGA melt-electrospun tubular structures mechanical properties was observed due to the thermal stabilization use. The ethylene oxide( EO) and steam sterilization had a destructive effect on the designed PLAGA meltelectrospun tubular, fibrous structures, whereas the irradiation yielded in the changes in mechanical and physical properties on the acceptable level,taking into account the future clinical applications.

文摘To investigate the roles of HtrA protein in bacterial adaption to stresses, htra gene was cloned from an Escherichia coli strain isolated from silver fox, double digested, and ligated into the prokaryotic expression vector pET32a. Then, the recombinant plasmid pET32a-htra was transformed into E. coli BL21 competent cells. The over-expression of His-HtrA protein was induced by the addition of IPTG at a final concentration of 1 mmol/L, and verified by SDA-PAGE and Western blot. In addition, the protein was purified with Ni-NTA agarose, and used to immunize mice,and the polyclonal antibody specifically bound to E. coli HtrA protein. The results will provide a theoretical basis for studying the roles of HtrA protein and developing vaccines for fur-bearing animals against E. coli.

文摘AIM: To construct a tumor-selective replication-competent adenovirus (RCAd), SG300, using a modified promoter of human telomerase reverse transcriptase (hTERT). METHODS: The antitumor efficacy of SG300 in hepatocellular carcinoma was assessed in vitro and in vivo. In vitro cell viability by MTT assay was used to assess the tumor-selective oncolysis and safety features of SG300, and in vivo antitumor activity of SG300 was assessed in established hepatocellular carcinoma models in nude mice. RESULTS: SG300 could lyse hepatocellular carcinoma cells at a low multiplicity of infection (MOI), but could not affect growth of normal cells even at a high MOI. Both in Hep3B and SMMC-7721 xenograft models of hepatocellular carcinoma, SG300 had an obvious antitumor effect, resulting in a decrease in tumor volume. Its selective oncolysis to tumor cells and safety to normal cells was also superior to that of ONYX-015. Pathological examination of tumor specimens showed that SG300 replicated selectively in cancer cells and resulted in apoptosis and necrosis of cancer cells. CONCLUSION: hTERT promoter-regulated replicativeadenovirus SG300 has a better cancer-selective replication-competent ability, and can specifically kill a wide range of cancer cells with positive telomerase activity, and thus has better potential for targeting therapy of hepatocellular carcinoma.

文摘Glial cells play an important role in signal transduction,energy metabolism,extracellular ion homeostasis and neuroprotection of the central nervous system.However,few studies have explained the potential effects of exosomes from glial cells on central nervous system health and disease.In this study,the genes expressed in exosomes from astrocytes and microglia were identified by deep RNA sequencing.Kyoto Encyclopedia of Genes and Genomes analysis indicated that several pathways in these exosomes are responsible for promoting neurodegenerative diseases,including Alzheimer's disease,Parkinson's disease and Huntington's disease.Gene ontology analysis showed that extracellular exosome,mitochondrion and growth factor activity were enriched in exosomes from the unique astrocyte group,while extracellular exosome and mitochondrion were enriched in exosomes from the unique microglia group.Next,combined with the screening of hub genes,the protein-protein interaction network analysis showed that exosomes from astrocytes influence neurodegenerative diseases through metabolic balance and ubiquitin-dependent protein balance,whereas exosomes from microglia influence neurodegenerative diseases through immune inflammation and oxidative stress.Although there were differences in RNA expression between exosomes from astrocytes and microglia,the groups were related by the hub genes,ubiquitin B and heat shock protein family A(Hsp70) member 8.Ubiquitin B appeared to be involved in pleiotropic regulatory functions,including immune regulation,inflammation inhibition,protein catabolism,intracellular protein transport,exosomes and oxidative stress.The results revealed the clinical significance of exosomes from glia in neurodegenerative diseases.This study was approved by the Animal Ethics Committee of Nantong University,China(approval No.S20180102-152) on January 2,2018.

文摘The inflammatory microenvironment and neurotoxicity can hinder neuronal regeneration and functional recovery after spinal cord injury.Ruxolitinib,a JAK-STAT inhibitor,exhibits effectiveness in autoimmune diseases,arthritis,and managing inflammatory cytokine storms.Although studies have shown the neuroprotective potential of ruxolitinib in neurological trauma,the exact mechanism by which it enhances functional recovery after spinal cord injury,particularly its effect on astrocytes,remains unclear.To address this gap,we established a mouse model of T10 spinal cord contusion and found that ruxolitinib effectively improved hindlimb motor function and reduced the area of spinal cord injury.Transcriptome sequencing analysis showed that ruxolitinib alleviated inflammation and immune response after spinal cord injury,restored EAAT2 expression,reduced glutamate levels,and alleviated excitatory toxicity.Furthermore,ruxolitinib inhibited the phosphorylation of JAK2 and STAT3 in the injured spinal cord and decreased the phosphorylation level of nuclear factor kappa-B and the expression of inflammatory factors interleukin-1β,interleukin-6,and tumor necrosis factor-α.Additionally,in glutamate-induced excitotoxicity astrocytes,ruxolitinib restored EAAT2 expression and increased glutamate uptake by inhibiting the activation of STAT3,thereby reducing glutamate-induced neurotoxicity,calcium influx,oxidative stress,and cell apoptosis,and increasing the complexity of dendritic branching.Collectively,these results indicate that ruxolitinib restores glutamate homeostasis by rescuing the expression of EAAT2 in astrocytes,reduces neurotoxicity,and effectively alleviates inflammatory and immune responses after spinal cord injury,thereby promoting functional recovery after spinal cord injury.

文摘Targeted genotyping is an extremely powerful approach for the detection of known genetic variations that are biologically or clinically important.However,for non-model organisms,large-scale target geno-typing in a cost-effective manner remains a major challenge.To address this issue,we present an ultrahigh-multiplex,in-solution probe array-based high-throughput diverse marker genotyping(HD-Marker)approach that is capable of targeted genotyping of up to 86000 loci,with coverage of the whole gene repertoire,in what is a 27-fold and six-fold multiplex increase in comparison with the conventional Illumina GoldenGate and original HD-Marker assays,respectively.We perform extensive analyses of var-ious ultrahigh-multiplex levels of HD-Marker(30 k-plex,56 k-plex,and 86 k-plex)and show the power and excellent performance of the proposed method with an extremely high capture rate(about 96%)and genotyping accuracy(about 96%).With great advantages in terms of cost(as low as 0.0006 USD per geno-type)and high technical flexibility,HD-Marker is a highly efficient and powerful tool with broad appli-cation potential for genetic,ecological,and evolutionary studies of non-model organisms.
文摘Background: Team based care is an essential ingredient of chronic disease management including chronic mental illness. Effective health care teams include members who have defined, yet intersecting roles, where mutual respect characterises professional interaction and the patient’s well-being is central. The aim was to explore the perception of psychologists, psychiatry registrars and psychiatrists with respect to GPs’ role in managing difficult-to-treat-depression (DTTD). Methods: A previously developed semi-structured interview schedule comprising six questions was used. Thirty-two health professionals participated. Data were analysed using the Framework method. Findings: Four main themes emerged: 1) The team approach was important, particularly to ensure information accuracy and/or when responding to patient needs and pressures;2) Referrals, usually generated by GPs can be a vehicle for other health professionals to provide advice to the GP;3) Availability and accessibility often depended on health professionals work location and knowing how to navigate the system;4) Limited availability of government funding impacts on patients’ accessibility to health professionals. Discussion: Interprofessional relationships were described as paramount. Appropriate and timely referrals are integral to patient management, regardless of challenges. Ongoing challenges include program funding, workforce numbers and costs to patients. Improvement to mental health care access was noted, even for patients among relatively disadvantaged groups and those receiving Medicare Benefits Schedule-subsidised services. Conclusion: Despite adequate GP/specialist communication, the delivery of optimal team based care to patients with difficult-to-treat depression is compromised by lack of access to specialised services and inadequate funding.

文摘In this study,a green and pollution-free multifunctional superhydrophobic paper-based material was prepared using a simple and efficient dipping method.The superhydrophobic paper with a water contact angle(WCA)of 160°was prepared by attaching micro-and nanocomposite particles,made of stearic acid-modified chitosan and two kinds of titanium dioxide(TiO_(2))nanoparticles of different sizes,to a paper substrate.The surface morphology,elemental composition,and wetting properties of the coatings were examined using scanning electron microscopy(SEM),X-ray photoelectron spectroscopy(XPS),Fourier transform infrared spectroscopy(FT-IR),and contact angle measurements.Additionally,superhydrophobic coatings exhibited good self-cleaning properties,liquid repellency,ease of repair,and antifouling properties in organic solutions.

文摘Myeloproliferative neoplasms(MPNs)occur due to the abnormal proliferation of one or more terminal myeloid cell lines in peripheral blood.Subjects suffering from MPNs display a high burden of cardiovascular risk factors,and thrombotic events are often the cause of death in this population of patients.Herein,we provide a brief overview of dyslipidemia and metabolic syndrome and their epidemiology in MPNs and examine the common molecular mechanisms between dyslipidemia,metabolic syndrome,and MPNs,with a special focus on cardio-vascular risk,atherosclerosis,and thrombotic events.Furthermore,we investigate the impact of dyslipidemia and metabolic syndrome on the occurrence and survival of thrombosis in MPN patients,as well as the management of dyslipidemia in MPNs,and the impact of MPN treatment on serum lipid concentrations,particularly as side/adverse effects reported in the context of clinical trials.
