Melatonin alleviates neuroinflammation in ischemic stroke by regulating cyclic GMP-AMP synthase-mediated microglial pyroptosis signaling

Inflammation plays a key role in driving the secondary brain injury that follows ischemic stroke.Melatonin is an endogenous neuroendocrine hormone that regulates mitochondrial homeostasis.However,the role and mechanisms by which melatonin regulates microglial pyroptosis and the inflammatory cascade through double-stranded DNA(dsDNA)-sensing cyclic GMP-AMP synthase(cGAS)signaling warrant further study.Using middle cerebral artery occlusion mice,we investigated the effects of melatonin on cGAS-mediated pyroptosis and neuroinflammation.Middle cerebral artery occlusion model mice exhibited significantly increased DNA damage and cytoplasmic dsDNA release,as reflected byγH2AX staining,as well as heightened activation of the cytosolic dsDNA-sensing cGAS-STING pathway,both of which were notably suppressed by melatonin treatment.Melatonin also mitigated NOD-like receptor family pyrin domain-containing protein 3(NLRP3)inflammasome activation and nuclear factor(NF)-κB/gasdermin D-mediated pyroptosis in microglia following ischemic stroke,while exhibiting the capacity to attenuate the immune response to ischemia in mice.This led to reduced infiltration of peripheral neutrophils and monocytes/macrophages in the ischemic brain.Specifically,melatonin administration resulted in reductions in the numbers of ionized calcium-binding adapter molecule 1-positive cells and production of interleukin-6 and tumor necrosis factor-αby microglia.Regarding neurological outcomes,melatonin significantly reduced cerebral infarct volume and ameliorated neurological deficits in mice.Notably,the neuroprotective effect of melatonin was correlated with the inhibition of cGAS activity.We also developed and tested melatonin co-loaded macrophage membrane-biomimetic reactive oxygen species-responsive nanoparticles(Mф-MLT@FNGs),which exhibited therapeutic properties in middle cerebral artery occlusion mice.Our findings suggest that melatonin acts on microglial pyroptosis to inhibit neuroinflammation and reshape the immune microenvironment through regulation of the cGAS-STING-NF-κB signaling pathway.By doing so,melatonin rescues damaged brain tissue and protects neurological function,highlighting its potential as a neuroprotective treatment for ischemic stroke. 展开更多

The luminance in the road tunnel threshold zone attracts broad attention due to its enormous energy consumption and direct influence on tunnel transportation security.Current lighting design methods in threshold zones mostly adopt the reduction coefficient method.However,the determination of reduction coefficient k simply considers tunnel design speed and flow rate,while excluding outside tunnel luminance and threshold zone color temperature and luminance,which have a major impact on driver visual adaptation.Existing problems in the determination of k value are analyzed;a visual performance experiment is utilized;and the reaction time of drivers in changeable outside tunnel luminance and threshold zone color temperature and luminance conditions is obtained;thus,the equations concerning reduction coefficient variation law are derived.In the end,a comparative analysis is made of the k values of the reduction coefficient stipulated by various norms under different color temperature conditions. 展开更多

Designing heterogeneous grain structure(HGS)has been proven to be an effective strategy for overcoming the strength-plasticity dilemma in copper and copper alloys.However,the construction of HGS in dispersionstrengthened copper(DSC)for enhancing strength-plasticity synergy remains challenging.Here,we proposed a novel method,multistep ball milling and reduction process followed by spark plasma sintering,to prepare DSC with an HGS to ameliorate the strength-plasticity dilemma in DSC.Micron-and nano-CuO and nano-Y_(2)O_(3)powders were chosen as raw materials in this new method.The Cu-7vol%Y_(2)O_(3)composite,exhibiting a compressive yield strength of 438 MPa and a failure strain of 46.3%,exhibits a superior strength-plasticity tradeoff in comparison with other DSC materials.Systematic experiments indicate that the back-stress at the heterointerfaces between coarse grains and fine grains maybe not only raise the yield strength of Cu-Y_(2)O_(3)composite,but also significantly enhance the strain hardening to increase the plasticity of the material.The new HGS designing route in this study offers a feasible pathway to develop DSC with a remarkable enhancement in strength and plasticity. 展开更多

A bibliometric analysis of core articles ofinternational nursing research frontiersbased on the Web of Science database

Objective:To review and analyze the basic information and distribution of core articles in the global research frontier of nursing,so as to ascertain the current trend in the field.Methods:A total of 37 highly cited e... Objective:To review and analyze the basic information and distribution of core articles in the global research frontier of nursing,so as to ascertain the current trend in the field.Methods:A total of 37 highly cited essential science indicator(ESI)papers in the nursing discipline were retrieved,which were compared with the core article collection of the ESI research frontier in January 2019 to understand the current nursing research frontiers.Subsequently,a statistical analysis of the core articles that constituted the nursing research frontiers was performed using a bibliometric method.The analysis was conducted with multiple aspects including the number of core articles,total number of citations,average publication year,issuing country,participating institution,citing paper,journal distribution,and core researcher.Results:At present,the two international research frontiers of nursing are patient-centered care(Research Frontier 1)and missed care(Research Frontier 2).These two frontiers include a total of 12 core articles,6 of which are highly cited Web of Scienceindexed nursing papers.Research Frontier 1 includes three core articles that were cited 389 times.Research Frontier 2 includes nine core articles that were cited 841 times.Articles in the two frontiers were cited by 454 and 841 papers,respectively,most of which are in the United States.In addition,the 12 core articles were published across eight journals,the impact factors of which are all relatively high.Lastly,Research Frontier 1 involves 11 authors,whereas Research Frontier 2 involves 49 authors,of which seven authors have published more than two articles.Conclusions:The core articles in the frontier of international nursing research demonstrate distinctive features in their issuing country/region,journal distribution,and participating institution.Review and analysis of the core articles of international nursing research frontiers can help nursing staff understand the current research hotspots and consequently perform corresponding scientific research to promote the development of the nursing discipline. 展开更多

Analysis on the Driving Factors and Realization Path of the Value of Tutor-Student Relationship of Postgraduates-with Medical and Pharmaceutical Universities as an Example

A harmonious relationship between teachers and postgraduates presents the comprehensive value of psychological education,which guarantees training quality and acts as an effective carrier of mental health education of the postgraduates.Based on Bandura’s theory of reciprocal determinism and Austin’s input-output theory,this paper constructs a model concerning the value of psychological education in the tutor-student relationship.The path of realization is also explored in a structural equation model through a questionnaire survey of 1112 graduate supervisors and administrators in medical colleges.The experimental result indicates that(1)The interaction between teachers and postgraduates promotes the value of psychological education in the highest measure,among which the quality and frequency of tutor-student communication,mutual trust,and respect act as the key influencing indicators.(2)Tutor’s professional quality is considered as the fundamental driving force in the realization of psychological education value in the tutor-student relationship,the maximum mediating effect is observed in the developing path of tutor’s professional quality-interaction behavior between teachers and postgraduates-psychological growth of teachers and postgraduates.(3)The objective results obtained by teachers and postgraduates,such as the improvement of quality and quantity,positively promote the overall quality of life satisfaction of both sides,which improves the major psychological growth indicators.(4)In the path that affects the psychological growth of teachers and postgraduates,female groups mainly focusing on natural sciences pay more attention to the tutor-student emotional experience,while male and lower-grade groups pay more attention to behavioral interaction;In the path that affects the interaction between teachers and postgraduates,the tutor does not value the difference in the quality of graduate students,lower-grade and natural science professional groups pay more attention to institutional environment. 展开更多

The novel severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)is the cause of a rapidly spreading illness,coronavirus disease 2019(COVID-19),affecting more than seventeen million people around the world.Diagnosis and treatment guidelines for clinicians caring for patients are needed.In the early stage,we have issued"A rapid advice guideline for the diagnosis and treatment of 2019 novel coronavirus(2019-nCoV)infected pneumonia(standard version)";now there are many direct evidences emerged and may change some of previous recommendations and it is ripe for develop an evidence-based guideline.We formed a working group of clinical experts and methodologists.The steering group members proposed 29 questions that are relevant to the management of COVID-19 covering the following areas:chemoprophylaxis,diagnosis,treatments,and discharge management.We searched the literature for direct evidence on the management of COVID-19,and assessed its certainty generated recommendations using the Grading of Recommendations,Assessment,Development and Evaluation(GRADE)approach.Recommendations were either strong or weak,or in the form of ungraded consensus-based statement.Finally,we issued 34 statements.Among them,6 were strong recommendations for,14 were weak recommendations for,3 were weak recommendations against and 11 were ungraded consensus-based statement.They covered topics of chemoprophylaxis(including agents and Traditional Chinese Medicine(TCM)agents),diagnosis(including clinical manifestations,reverse transcription-polymerase chain reaction(RT-PCR),respiratory tract specimens,IgM and IgG antibody tests,chest computed tomography,chest X-ray,and CT features of asymptomatic infections),treatments(including lopinavirritonavir,umifenovir,favipiravir,interferon,remdesivir,combination of antiviral drugs,hydroxychloroquine/chloroquine,interleukin-6 inhibitors,interleukin-1 inhibitors,glucocorticoid,qingfei paidu decoction,lianhua qingwen granules/capsules,convalescent plasma,lung transplantation,invasive or noninvasive ventilation,and extracorporeal membrane oxygenation(ECMO)),and discharge management(including discharge criteria and management plan in patients whose RT-PCR retesting shows SARS-CoV-2 positive after discharge).We also created two figures of these recommendations for the implementation purpose.We hope these recommendations can help support healthcare workers caring for COVID-19 patients. 展开更多

Alzheimer’s disease(AD)is the most common cause of dementia.Neuropathological changes in AD patients occur up to 10–20 years before the emergence of clinical symptoms.Specific diagnosis and appropriate intervention strategies are crucial during the phase of mild cognitive impairment(MCI)and AD.The detection of biomarkers has emerged as a promising tool for tracking the efficacy of potential therapies,making an early disease diagnosis,and prejudging treatment prognosis.Specifically,multiple neuroimaging modalities,including magnetic resonance imaging(MRI),positron emission tomography,optical imaging,and single photon emissioncomputed tomography,have provided a few potential biomarkers for clinical application.The MRI modalities described in this review include structural MRI,functional MRI,diffusion tensor imaging,magnetic resonance spectroscopy,and arterial spin labelling.These techniques allow the detection of presymptomatic diagnostic biomarkers in the brains of cognitively normal elderly people and might also be used to monitor AD disease progression after the onset of clinical symptoms.This review highlights potential biomarkers,merits,and demerits of different neuroimaging modalities and their clinical value in MCI and AD patients.Further studies are necessary to explore more biomarkers and overcome the limitations of multiple neuroimaging modalities for inclusion in diagnostic criteria for AD. 展开更多

In this special issue of Genes&Diseases,Primary Immune Deficiencies(PIDs)are the main focus.This is an exciting and a rapidly emerging field of basic and clinical immunology wherein upwards of 400 clinical conditions,with approximately 350 defined mutations,are now recognized.Contrary to common perception,PIDs as a group are not uncommon.Epidemiologic studies show that population prevalence of PIDs is approximately 1:2000.Statistical extrapolations of this figure would suggest that in a large country like China or India,approximately 1 million individuals would be expected to have a PID.However,at present because of lack of awareness of these conditions amongst both the laity as well as medical professionals,majority of these patients remain undiagnosed and untreated in many developing countries.This is clearly unfortunate. 展开更多
