Alt...Mitochondrial dysfunction has emerged as a critical factor in the etiology of various neurodevelopmental disorders, including autism spectrum disorders, attention-deficit/hyperactivity disorder, and Rett syndrome. Although these conditions differ in clinical presentation, they share fundamental pathological features that may stem from abnormal mitochondrial dynamics and impaired autophagic clearance, which contribute to redox imbalance and oxidative stress in neurons. This review aimed to elucidate the relationship between mitochondrial dynamics dysfunction and neurodevelopmental disorders. Mitochondria are highly dynamic organelles that undergo continuous fusion and fission to meet the substantial energy demands of neural cells. Dysregulation of these processes, as observed in certain neurodevelopmental disorders, causes accumulation of damaged mitochondria, exacerbating oxidative damage and impairing neuronal function. The phosphatase and tensin homolog-induced putative kinase 1/E3 ubiquitin-protein ligase pathway is crucial for mitophagy, the process of selectively removing malfunctioning mitochondria. Mutations in genes encoding mitochondrial fusion proteins have been identified in autism spectrum disorders, linking disruptions in the fusion-fission equilibrium to neurodevelopmental impairments. Additionally, animal models of Rett syndrome have shown pronounced defects in mitophagy, reinforcing the notion that mitochondrial quality control is indispensable for neuronal health. Clinical studies have highlighted the importance of mitochondrial disturbances in neurodevelopmental disorders. In autism spectrum disorders, elevated oxidative stress markers and mitochondrial DNA deletions indicate compromised mitochondrial function. Attention-deficit/hyperactivity disorder has also been associated with cognitive deficits linked to mitochondrial dysfunction and oxidative stress. Moreover, induced pluripotent stem cell models derived from patients with Rett syndrome have shown impaired mitochondrial dynamics and heightened vulnerability to oxidative injury, suggesting the role of defective mitochondrial homeostasis in these disorders. From a translational standpoint, multiple therapeutic approaches targeting mitochondrial pathways show promise. Interventions aimed at preserving normal fusion-fission cycles or enhancing mitophagy can reduce oxidative damage by limiting the accumulation of defective mitochondria. Pharmacological modulation of mitochondrial permeability and upregulation of peroxisome proliferator-activated receptor gamma coactivator 1-alpha, an essential regulator of mitochondrial biogenesis, may also ameliorate cellular energy deficits. Identifying early biomarkers of mitochondrial impairment is crucial for precision medicine, since it can help clinicians tailor interventions to individual patient profiles and improve prognoses. Furthermore, integrating mitochondria-focused strategies with established therapies, such as antioxidants or behavioral interventions, may enhance treatment efficacy and yield better clinical outcomes. Leveraging these pathways could open avenues for regenerative strategies, given the influence of mitochondria on neuronal repair and plasticity. In conclusion, this review indicates mitochondrial homeostasis as a unifying therapeutic axis within neurodevelopmental pathophysiology. Disruptions in mitochondrial dynamics and autophagic clearance converge on oxidative stress, and researchers should prioritize validating these interventions in clinical settings to advance precision medicine and enhance outcomes for individuals affected by neurodevelopmental disorders.展开更多

METHODS We retrospectively investigated the patients with colorectal polyps who were diagnosed by colonoscopy and pathology in our hospital from 1990 to 2004. The analysis involved the incidence, average patient age, polyp location and pathological types. We recorded the changes of the polyp clinicopathologic features by comparing the clinicopathologic types of colorectal polyps over fi ve-year periods. RESULTS Of the 21,853 patients who received a colonoscopy, 2,889(13.2%) were diagnosed with colorectal polyps. Their average age was 46.6±16.5 years, with a male to female ratio of 1.8:1. The males were older than females (47.1±17.5 vs. 45.5±14.5, P<0.05). Location of the polyps: 41% in the rectum, 27.7% in the sigmoid colon, 35.8% in the left side verus 23.1% in the right side (P<0.05). Patients with polyps located in the transverse and ascending colon were older than those with polyps in the rectum and sigmoid colon (P<0.05). Adenomatous polyps comprised the most common type (67%) and the rectum was the most common site for each type, especially juvenile and retention polyps. Juvenile polyps were found in the youngest patients(12±4.8, P<0.05) and the adenomatous in the oldest (52±14, P<0.05). The ratio of patients with polyposis comprised 1.2%, and patients with polyps accompanied with colorectal cancer comprised 6.1%. Examination of the changes in the incidence, the average patient age, and adenomatous type showed that they had all increased, but the frequency of infl ammatory and retention polyps decreased. CONCLUSION Colorectal polyps are a common problem. The frequency is greater in males compared to females and the rectum and sigmoid colon had the highest incidence. Most juvenile and retention polyps were found in young patients, but most adenomatous occurred in adults. In recent years, the incidence of colorectal polyps, the average patient age, and the adenomatous type have increased in the Nanchang area, but inflammatory and retention types decreased.展开更多

Recent studies on the pathogenesis and progression of multiple sclerosis have highlighted the multiple roles of mononuclear macrophages in the neuroinflammatory process. Monocytes play a significant role in neuroinflammation, and managing neuroinflammation by manipulating peripheral monocytes stands out as an effective strategy for the treatment of multiple sclerosis, leading to improved patient outcomes. This review outlines the steps involved in the entry of myeloid monocytes into the central nervous system that are targets for effective intervention: the activation of bone marrow hematopoiesis, migration of monocytes in the blood, and penetration of the blood–brain barrier by monocytes. Finally, we summarize the different monocyte subpopulations and their effects on the central nervous system based on phenotypic differences. As activated microglia resemble monocyte-derived macrophages, it is important to accurately identify the role of monocyte-derived macrophages in disease. Depending on the roles played by monocyte-derived macrophages at different stages of the disease, several of these processes can be interrupted to limit neuroinflammation and improve patient prognosis. Here, we discuss possible strategies to target monocytes in neurological diseases, focusing on three key aspects of monocyte infiltration into the central nervous system, to provide new ideas for the treatment of neurodegenerative diseases.展开更多

文摘Ischemia–reperfusion injury is a common pathophysiological mechanism in retinal degeneration.PANoptosis is a newly defined integral form of regulated cell death that combines the key features of pyroptosis, apoptosis, and necroptosis.Oligomerization of mitochondrial voltage-dependent anion channel 1 is an important pathological event in regulating cell death in retinal ischemia–reperfusion injury. However, its role in PANoptosis remains largely unknown. In this study,we demonstrated that voltage-dependent anion channel 1 oligomerization-mediated mitochondrial dysfunction was associated with PANoptosis in retinal ischemia–reperfusion injury.Inhibition of voltage-dependent anion channel 1 oligomerization suppressed mitochondrial dysfunction and PANoptosis in retinal cells subjected to ischemia–reperfusion injury.Mechanistically,mitochondria-derived reactive oxygen species played a central role in the voltagedependent anion channel 1-mediated regulation of PANoptosis by promoting PANoptosome assembly. Moreover, inhibiting voltage-dependent anion channel 1 oligomerization protected against PANoptosis in the retinas of rats subjected to ischemia–reperfusion injury.Overall, our findings reveal the critical role of voltage-dependent anion channel 1 oligomerization in regulating PANoptosis in retinal ischemia–reperfusion injury, highlighting voltage-dependent anion channel 1 as a promising therapeutic target.

文摘To explore the formation mechanism of anisotropy in Ti-6Al-4V alloy fabricated by selective laser melting(SLM),the compressive mechanical properties, microhardness, microstructure, and crystallographic orientation of the alloy across different planes were investigated. The anisotropy of SLM-fabricated Ti-6Al-4V alloys was analyzed, and the electron backscatter diffraction technique was used to investigate the influence of different grain types and orientations on the stress-strain distribution at various scales. Results reveal that in room-temperature compression tests at a strain rate of 10^(-3) s^(-1),both the compressive yield strength and microhardness vary along the deposition direction, indicating a certain degree of mechanical property anisotropy. The alloy exhibits a columnar microstructure;along the deposition direction, the grains appear equiaxed, and they have internal hexagonal close-packed(hcp)α/α'martensitic structure.α'phase has a preferential orientation approximately along the<0001>direction.Anisotropy arises from the high aspect ratio of columnar grains, along with the weak texture of the microstructure and low symmetry of the hcp crystal structure.
文摘Mitochondrial dysfunction has emerged as a critical factor in the etiology of various neurodevelopmental disorders, including autism spectrum disorders, attention-deficit/hyperactivity disorder, and Rett syndrome. Disruptions in mitochondrial dynamics and autophagic clearance converge on oxidative stress, and researchers should prioritize validating these interventions in clinical settings to advance precision medicine and enhance outcomes for individuals affected by neurodevelopmental disorders.
文摘Dissimilar AZ31B magnesium alloy and DC56D steel were welded via AA1060 aluminum alloy by magnetic pulse welding. The effects of primary and secondary welding processes on the welded interface were comparatively investigated.Macroscopic morphology, microstructure, and interfacial structure of the joints were analyzed using scanning electron microscope, energy dispersive spectrometer, and X-ray diffractometer(XRD).The results show that magnetic pulse welding of dissimilar Mg/Fe metals is achieved using an Al interlayer, which acts as a bridge for deformation and diffusion.Specifically, the AZ31B/AA1060 interface exhibits a typical wavy morphology, and a transition zone exists at the joint interface, which may result in an extremely complex microstructure. The microstructure of this transition zone differs from that of AZ31B magnesium and 1060 Al alloys, and it is identified as brittle intermetallic compounds(IMCs)Al_(3)Mg_(2) and Al_(12)Mg_(17).The transition zone is mainly distributed on the Al side, with the maximum thickness of Al-side transition layer reaching approximately 13.53μm.Incomplete melting layers with varying thicknesses are observed at the primary weld interface, while micron-sized hole defects appear in the transition zone of the secondary weld interface. The AA1060/DC56D interface is mainly straight, with only a small number of discontinuous transition zones distributed intermittently along the interface. These transition zones are characterized by the presence of the brittle IMC FeAl_(3),with a maximum thickness of about 4μm.

文摘Parkinson’s disease is characterized by synucleinopathy-associated neurodegeneration.Previous studies have shown that glucagon-like peptide-1(GLP-1)has beneficial effects in a mouse model of Parkinson’s disease induced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine.However,the effect of GLP-1 on intrinsic synuclein malfunction remains unclear. In this study,we investigated the effect of Lactococcus lactis MG1363-pMG36e-GLP-1 on parkinsonism in SncaA53T transgenic mice and explored the underlying mechanisms. Our data showed that Lactococcus lactis MG1363-pMG36e-GLP-1 inhibited dopaminergic neuronal death, reduced pathological aggregation ofα-synuclein,and decreased movement disorders in SncaA53T transgenic mice.Furthermore,Lactococcus lactis MG1363-pMG36e-GLP-1 downregulated lipopolysaccharide-related inflammation, reduced cerebral activation of microglia and astrocytes, and promoted cell survival via the GLP-1 receptor/PI3K/Akt pathway in the substantia nigra.Additionally,Lactococcus lactis MG1363-pMG36e-GLP-1 decreased serum levels of pro-inflammatory molecules including lipopolysaccharide, lipopolysaccharide binding protein,interleukin-1β,and interleukin-6.Gut histopathology and western blotting further revealed that Lactococcus lactis MG1363-pMG36e-GLP-1 increased the expression of gut integrity-related proteins and reduced lipopolysaccharide-related inflammation by reversing gut dysbiosis in SncaA53T transgenic mice. Our findings showed that the beneficial effect of Lactococcus lactis MG1363-pMG36e-GLP-1 on parkinsonism traits in SncaA53T transgenic mice is mediated by microglial polarization and the reversal of dysbiosis.Collectively, our findings suggest that Lactococcus lactis MG1363-pMG36e-GLP-1 is a promising therapeutic agent for the treatment of Parkinson’s disease.

文摘Fenton-like technology based on peroxymonosulfate activation has shown great potential in refractory organics degradation. In this work, single Fe atom catalysts were synthesized through facile ball milling and exhibited very high performance in peroxymonosulfate activation. The Fe single-atom filled an N vacancy on the triazine ring edge of C_(3)N_(4),as confirmed through X-ray absorption fine structure, density functional calculation and elec-tron paramagnetic resonance. The SAFe_(0.4)–C_(3)N_(4)/PMS system could completely remove phenol(20 mg/L)within 10 min and its first-order kinetic constant was 12.3 times that of the Fe_(3)O_(4)/PMS system.Under different ini-tial pH levels and in various anionic environments,SAFe_(0.4)–C_(3)N_(4) still demonstrated excellent catalytic activity, achieving a removal rate of over 90%for phenol within 12 min.In addition,SAFe_(0.4)–C_(3)N_(4) exhibited outstanding selectivity in reaction systems with different pollutants, showing excellent degradation effects on electron-rich pollutants only.Hydroxyl radicals(•OH),singlet oxygen(1O_(2))and high-valent iron oxide(Fe(Ⅳ)=O)were de-tected in the SAFe_(0.4)–C_(3)N_(4)/PMS system through free radical capture experiments.Further experiments on the quenching of active species and a methyl phenyl sulfoxide probe confirmed that 1O_(2) and Fe(Ⅳ)=O played dom-inant roles.Additionally, the change in the current response after adding PMS and phenol in succession proved that a direct electron transfer path between organic matter and the catalyst surface was unlikely to exist in the SAFe_(0.4)–C_(3)N_(4)/PMS/Phenol degradation system. This study provides a new demonstration of the catalytic mech-anism of single-atom catalysts.

文摘With the gradual advancement of research methods and technologies, various biological processes have been identified as playing roles in the pathogenesis of neurodegenerative diseases. However, current descriptions of these biological processes do not fully explain the onset, progression, and development of these conditions. Therefore, exploration of the pathogenesis of neurodegenerative diseases remains a valuable area of research. This review summarizes the potential common pathogeneses of Alzheimer’s disease,Parkinson’s disease, amyotrophic lateral sclerosis,Huntington’s disease, frontotemporal lobar dementia, and Lewy body disease.Research findings have indicated that several common biological processes, including aging, genetic factors, progressive neuronal dysfunction, neuronal death and apoptosis, protein misfolding and aggregation, neuroinflammation, mitochondrial dysfunction, axonal transport defects, and gut microbiota dysbiosis, are involved in the pathogenesis of these six neurodegenerative diseases.Based on current information derived from diverse areas of research, these biological processes may form complex pathogenic networks that lead to distinctive types of neuronal death in neurodegenerative diseases.Furthermore, promoting the regeneration of damaged neurons may be achievable through the repair of affected neural cells if the underlying pathogenesis can be prevented or reversed.Hence, these potential common biological processes may represent only very small, limited elements within numerous intricate pathogenic networks associated with neurodegenerative diseases. In clinical treatment, interfering with any single biological process has proven insufficient to completely halt the progression of neurodegenerative diseases. Therefore, future research on the pathogenesis of neurodegenerative diseases should focus on uncovering the complex pathogenic networks, rather than isolating individual biological processes.Based on this, therapies that aim to block or reverse various targets involved in the potential pathogenic mechanisms of neurodegenerative diseases may be promising directions,as current treatment methods that focus on halting a single pathogenic factor have not achieved satisfactory efficacy.

文摘MnO_(x)-CeO_(2)catalysts for the low-temperature selective catalytic reduction(SCR)of NO remain vulnerable to water and sulfur poisoning, limting their practical applications.Herein,we report a hydrophobic-modified MnO_(x)-CeO_(2)catalyst that achieves enhanced NO conversion rate and stability under harsh conditions. The catalyst was synthesized by decorating MnOx crystals with amorphous CeO_(2),followed by loading hydrophobic silica on the external surfaces. The hydrophobic silica allowed the adsorption of NH_(3)and NO and diffusion of H,suppressed the adsorption of H_(2)O,and prevented SO_(2)interaction with the Mn active sites, achieving selective molecular discrimination at the catalyst surface. At 120℃,under H_(2)O and SO_(2)exposure,the optimal hydrophobic catalyst maintains 82%NO conversion rate compared with 69%for the unmodified catalyst. The average adsorption energies of NH_(3),H_(2)O,and SO_(2)decreased by 0.05,0.43,and 0.52 eV,respectively. The NO reduction pathway follows the Eley-Rideal mechanism,NH_(3)^(*)+*→NH_(2)^(*)+H^(*)followed by NH_(2)^(*)+NO^(*)→N_(2)^(*)+H_(2)O^(*),with NH_(3)dehydrogenation being the rate determining step.Hydrophobic modification increased the activation energy for H atom transfer, leading to a minor decrease in the NO conversion rate at 120℃.This work demonstrates a viable strategy for developing robust NH_(3)-S CR catalysts capable of efficient operation in water-and sulfur-rich environments.

文摘AIM:To compare spontaneous brain regional activities between diabetic vitreous hemorrhage patients(DVHs)and healthy controls(HCs).METHODS:Thirty-two DVHs and 32 HCs were enrolled in this study.Baseline demographic and vision data were compared between groups using an independent sample t-test.Resting-state functional magnetic resonance imaging(rs-fMRI)was used in all participants.fMRI data was obtained and analyzed using MRIcro and SPM8 software.Fractional amplitude of low-frequency fluctuation(fALFF)technology was used to measure regional spontaneous brain activity, and sensitivity was tested using receiver operating characteristic curves(ROCs).The fALFF values were analyzed using REST software and two-sample t-tests were used to compare values between groups.Hospital anxiety and depression scale(HADS)score was assessed in DVHs and Pearson’s correlation was used to test relationships between mean fALFF value and both HADS score and duration of DVH.RESULTS:Except for the best-corrected visual acuity(BCVA)in both eyes, which showed a statistically significant difference(P<0.05),there were no statistically significant differences in the other indicators(P>0.05)between the HCs and DVHs group.Compared with controls,fALFF value was higher in DVH in cerebellum posterior lobe(CPL)and lower in right anterior cingulate cortex(ACC)and right medial orbitofrontal cortex(OFC).In DVH patients, mean fALFF value of CPL was positively correlated with HADS score and duration of diabetes. However,no such correlation was found, for right ACC or right medial OFC.DVH may lead to abnormal activities in certain brain regions related to visual control and mood.CONCLUSION:Visual impairment caused by DVH may lead to adjustment in regional visual brain activities and may be related to depression or reward system processing in some brain regions.

文摘THE mechanical response and deformation mechanisms of pure nickel under nanoindentation were systematically investigated using molecular dynamics(MD)simulations, with a particular focus on the novel interplay between crystallographic orientation, grain boundary(GB)proximity, and pore characteristics(size/location).This study compares single-crystal nickel models along[100],[110],and[111]orientations with equiaxed polycrystalline models containing 0,1,and 2.5 nm pores in surface and subsurface configurations. Our results reveal that crystallographic anisotropy manifests as a 24.4%higher elastic modulus and 22.2%greater hardness in[111]-oriented single crystals compared to[100].Pore-GB synergistic effects are found to dominate the deformation behavior:2.5 nm subsurface pores reduce hardness by 25.2%through stress concentration and dislocation annihilation at GBs,whereas surface pores enable mechanical recovery via accelerated dislocation generation post-collapse.Additionally,size-dependent deformation regimes were identified, with 1 nm pores inducing negligible perturbation due to rapid atomic rearrangement,in contrast with persistent softening in 2.5 nm pores. These findings establish atomic-scale design principles for defect engineering in nickel-based aerospace components, demonstrating how crystallographic orientation, pore configuration, and GB interactions collectively govern nanoindentation behavior.

文摘In response to the challenges in highway pavement distress detection, such as multiple defect categories, difficulties in feature extraction for different damage types, and slow identification speeds, this paper proposes an enhanced pavement crack detection model named Star-YOLO11.This improved algorithm modifies the YOLO11 architecture by substituting the original C3k2 backbone network with a Star-s50 feature extraction network. The enhanced structure adjusts the number of stacked layers in the StarBlock module to optimize detection accuracy and improve model efficiency. To enhance the accuracy of pavement crack detection and improve model efficiency, three key modifications to the YOLO11 architecture are proposed.Firstly, the original C3k2 backbone is replaced with a StarBlock-based structure, forming the Star-s50 feature extraction backbone network. This lightweight redesign reduces computational complexity while maintaining detection precision.Secondly,to address the inefficiency of the original Partial Self-attention(PSA)mechanism in capturing localized crack features, the convolutional prior-aware Channel Prior Convolutional Attention(CPCA)mechanism is integrated into the channel dimension, creating a hybrid CPC-C2PSA attention structure.Thirdly, the original neck structure is upgraded to a Star Multi-Branch Auxiliary Feature Pyramid Network(SMAFPN)based on the Multi-Branch Auxiliary Feature Pyramid Network architecture, which adaptively fuses high-level semantic and low-level spatial information through Star-s50 connections and C3k2 extraction blocks.Additionally,a composite dataset augmentation strategy combining traditional and advanced augmentation techniques is developed. This strategy is validated on a specialized pavement dataset containing five distinct crack categories for comprehensive training and evaluation.Experimental results indicate that the proposed Star-YOLO11 achieves an accuracy of 89.9%(3.5%higher than the baseline),a mean average precision(mAP)of 90.3%(+2.6%),and an F1-score of 85.8%(+0.5%),while reducing the model size by 18.8%and reaching a frame rate of 225.73 frames per second(FPS)for real-time detection. It shows potential for lightweight deployment in pavement crack detection tasks.

文摘The precise tuning of magnetic nanoparticle size and magnetic domains, thereby shaping magnetic properties. However, the dynamic evolution mechanisms of magnetic domain configurations in relation to electromagnetic(EM)attenuation behavior remain poorly understood. To address this gap,a thermodynamically controlled periodic coordination strategy is proposed to achieve precise modulation of magnetic nanoparticle spacing. This approach unveils the evolution of magnetic domain configurations, progressing from individual to coupled and ultimately to crosslinked domain configurations.A unique magnetic coupling phenomenon surpasses the Snoek limit in low-frequency range, which is observed through micromagnetic simulation. The crosslinked magnetic configuration achieves effective low-frequency EM wave absorption at 3.68 GHz,encompassing nearly the entire C-band.This exceptional magnetic interaction significantly enhances radar camouflage and thermal insulation properties.Additionally,a robust gradient metamaterial design extends coverage across the full band(2–40 GHz),effectively mitigating the impact of EM pollution on human health and environment. This comprehensive study elucidates the evolution mechanisms of magnetic domain configurations, addresses gaps in dynamic magnetic modulation, and provides novel insights for the development of high-performance,low-frequency EM wave absorption materials.

文摘OBJECTIVE To study the clinicopathologic characteristics, changes in the nature and incidence of colorectal polyps in the Nanchang area, Jiangxi, Province. In recent years, the incidence of colorectal polyps, the average patient age, and the adenomatous type have increased in the Nanchang area, but inflammatory and retention types decreased.

文摘Mononuclear macrophage infiltration in the central nervous system is a prominent feature of neuroinflammation. 文摘BACKGROUND Patients with hepatitis B virus(HBV)infection require chronic and personalized care to improve outcomes.Large language models(LLMs)can potentially provide medical information for patients.AIM To examine the performance of three LLMs,ChatGPT-3.5,ChatGPT-4.0,and Google Gemini,in answering HBV-related questions.METHODS LLMs’responses to HBV-related questions were independently graded by two medical professionals using a four-point accuracy scale, and disagreements were resolved by a third reviewer. Each question was run three times using three LLMs.Readability was assessed via the Gunning Fog index and Flesch-Kincaid grade level.RESULTS Overall, all three LLM chatbots achieved high average accuracy scores for subjective questions(ChatGPT-3.5:3.50;ChatGPT-4.0:3.69;Google Gemini:3.53,out of a maximum score of 4).With respect to objective questions,ChatGPT-4.0 achieved an 80.8%accuracy rate, compared with 62.9%for ChatGPT-3.5 and 73.1%for Google Gemini.Across the six domains,ChatGPT-4.0 performed better in terms of diagnosis, whereas Google Gemini demonstrated excellent clinical manifestations.Notably,in the readability analysis, the mean Gunning Fog index and Flesch-Kincaid grade level scores of the three LLM chatbots were significantly higher than the standard level eight, far exceeding the reading level of the normal population.CONCLUSION Our results highlight the potential of LLMs,especially ChatGPT-4.0,for delivering responses to HBV-related questions.LLMs may be an adjunctive informational tool for patients and physicians to improve outcomes.Nevertheless, current LLMs should not replace personalized treatment recommendations from physicians in the management of HBV infection.
