文摘Stroke, particularly ischemic stroke,is the leading cause of long-term disability and mortality worldwide. It occurs due to the occlusion of the cerebral arteries, which significantly reduces the delivery of blood, oxygen, and essential nutrients to brain tissues. This deprivation triggers a cascade of cellular events that ultimately leads to neuronal death.Recent studies have clarified the multifactorial pathogenesis of ischemic stroke, highlighting the roles of energy failure, excitotoxicity, oxidative stress, neuroinflammation, and apoptosis. This review aimed to provide a comprehensive insight into the fundamental mechanisms driving neuronal death triggered by ischemia and to examine the progress of neuroprotective therapeutic approaches designed to mitigate neuronal loss and promote neurological recovery after a stroke.Additionally,we explored widely accepted findings regarding the potential pathways implicated in neuronal death during ischemic stroke, including the interplay of apoptosis, autophagy, pyroptosis, ferroptosis, and necrosis, which collectively influence neuronal fate.We also discussed advancements in neuroprotective therapeutics, encompassing a range of interventions from pharmacological modulation to stem cell-based therapies, aimed at reducing neuronal injury and enhancing functional recovery following ischemic stroke.Despite these advancements, challenges remain in translating mechanistic insights into effective clinical therapies.Although neuroprotective strategies have shown promise in preclinical models, their efficacy in human trials has been inconsistent, often due to the complex pathology of ischemic stroke and the timing of interventions. In conclusion, this review synthesizes mechanistic insights into the intricate interplay of molecular and cellular pathways driving neuronal death post-ischemia.It sheds light on cutting-edge advancements in potential neuroprotective therapeutics, underscores the promise of regenerative medicine, and offers a forward-looking perspective on potential clinical breakthroughs. The ongoing evolution of precision-targeted interventions is expected to significantly enhance preventative strategies and improve clinical outcomes.

文摘Global brain ischemia and neurological deficit are consequences of cardiac arrest that lead to high mortality.Despite advancements in resuscitation science, our limited understanding of the cellular and molecular mechanisms underlying post-cardiac arrest brain injury have hindered the development of effective neuroprotective strategies.Previous studies primarily focused on neuronal death, potentially overlooking the contributions of non-neuronal cells and intercellular communication to the pathophysiology of cardiac arrest-induced brain injury. To address these gaps,we hypothesized that single-cell transcriptomic analysis could uncover previously unidentified cellular subpopulations, altered cell communication networks, and novel molecular mechanisms involved in post-cardiac arrest brain injury. In this study,we performed a single-cell transcriptomic analysis of the hippocampus from pigs with ventricular fibrillation-induced cardiac arrest at 6 and 24 hours following the return of spontaneous circulation, and from sham control pigs.Sequencing results revealed changes in the proportions of different cell types, suggesting post-arrest disruption in the blood-brain barrier and infiltration of neutrophils. These results were validated through western blotting, quantitative reverse transcription-polymerase chain reaction, and immunofluorescence staining. We also identified and validated a unique subcluster of activated microglia with high expression of S100A8,which increased over time following cardiac arrest. This subcluster simultaneously exhibited significant M1/M2 polarization and expressed key functional genes related to chemokines and interleukins.Additionally,we revealed the post-cardiac arrest dysfunction of oligodendrocytes and the differentiation of oligodendrocyte precursor cells into oligodendrocytes.Cell communication analysis identified enhanced post-cardiac arrest communication between neutrophils and microglia that was mediated by neutrophil-derived resistin, driving pro-inflammatory microglial polarization. Our findings provide a comprehensive single-cell map of the post-cardiac arrest hippocampus, offering potential novel targets for neuroprotection and repair following cardiac arrest.

文摘After spinal cord injury, impairment of the sensorimotor circuit can lead to dysfunction in the motor, sensory, proprioceptive, and autonomic nervous systems.Functional recovery is often hindered by constraints on the timing of interventions, combined with the limitations of current methods. To address these challenges, various techniques have been developed to aid in the repair and reconstruction of neural circuits at different stages of injury.Notably, neuromodulation has garnered considerable attention for its potential to enhance nerve regeneration, provide neuroprotection, restore neurons, and regulate the neural reorganization of circuits within the cerebral cortex and corticospinal tract. To improve the effectiveness of these interventions, the implementation of multitarget early interventional neuromodulation strategies, such as electrical and magnetic stimulation,is recommended to enhance functional recovery across different phases of nerve injury. This review concisely outlines the challenges encountered following spinal cord injury, synthesizes existing neurostimulation techniques while emphasizing neuroprotection, repair, and regeneration of impaired connections, and advocates for multi-targeted,task-oriented,and timely interventions.

文摘Ischemia–reperfusion injury is a common pathophysiological mechanism in retinal degeneration.PANoptosis is a newly defined integral form of regulated cell death that combines the key features of pyroptosis, apoptosis, and necroptosis.Oligomerization of mitochondrial voltage-dependent anion channel 1 is an important pathological event in regulating cell death in retinal ischemia–reperfusion injury. However, its role in PANoptosis remains largely unknown. In this study,we demonstrated that voltage-dependent anion channel 1 oligomerization-mediated mitochondrial dysfunction was associated with PANoptosis in retinal ischemia–reperfusion injury.Inhibition of voltage-dependent anion channel 1 oligomerization suppressed mitochondrial dysfunction and PANoptosis in retinal cells subjected to ischemia–reperfusion injury.Mechanistically,mitochondria-derived reactive oxygen species played a central role in the voltagedependent anion channel 1-mediated regulation of PANoptosis by promoting PANoptosome assembly. Moreover, inhibiting voltage-dependent anion channel 1 oligomerization protected against PANoptosis in the retinas of rats subjected to ischemia–reperfusion injury.Overall, our findings reveal the critical role of voltage-dependent anion channel 1 oligomerization in regulating PANoptosis in retinal ischemia–reperfusion injury, highlighting voltage-dependent anion channel 1 as a promising therapeutic target.

文摘Transformer-based models have significantly advanced binary code similarity detection(BCSD)by leveraging their semantic encoding capabilities for efficient function matching across diverse compilation settings.Although adversarial examples can strategically undermine the accuracy of BCSD models and protect critical code, existing techniques predominantly depend on inserting artificial instructions, which incur high computational costs and offer limited diversity of perturbations. To address these limitations,we propose AIMA,a novel gradient-guided assembly instruction relocation method. Our method decouples the detection model into tokenization, embedding, and encoding layers to enable efficient gradient computation.Since token IDs of instructions are discrete and nondifferentiable,we compute gradients in the continuous embedding space to evaluate the influence of each token. The most critical tokens are identified by calculating the L2 norm of their embedding gradients. We then establish a mapping between instructions and their corresponding tokens to aggregate token-level importance into instructionlevel significance. To maximize adversarial impact,a sliding window algorithm selects the most influential contiguous segments for relocation, ensuring optimal perturbation with minimal length. This approach efficiently locates critical code regions without expensive search operations. The selected segments are relocated outside their original function boundaries via a jump mechanism, which preserves runtime control flow and functionality while introducing“deletion”effects in the static instruction sequence.Extensive experiments show that AIMA reduces similarity scores by up to 35.8%in state-of-the-art BCSD models. When incorporated into training data,it also enhances model robustness, achieving a 5.9%improvement in AUROC.

文摘Formamidinium lead iodide(FAPbI_(3))perovskite exhibits an impressive X-ray absorption coefficient and a large carrier mobility-lifetime product(μτ),making it as a highly promising candidate for X-ray detection application. However, the presence of larger FA^(+)cation induces to an expansion of the Pb-I octahedral framework, which unfortunately affects both the stability and charge carrier mobility of the corresponding devices. To address this challenge,we develop a novel low-dimensional(HtrzT)PbI_(3) perovskite featuring a conjugated organic cation(1H-1,2,4-Triazole-3-thiol,HtrzT^(+))which matches well with theα-FAPbI_(3) lattices in two-dimensional plane.Benefiting from the matched lattice between(HtrzT)PbI_(3) andα-FAPbI_(3),the anchored lattice enhances the Pb-I bond strength and effectively mitigates the inherent tensile strain of theα-FAPbI_(3) crystal lattice. The X-ray detector based on(HtrzT)PbI_(3)(1.0)/FAPbI_(3) device achieves a remarkable sensitivity up to 1.83×10^(5)μC Gy_(air)^(−1) cm^(−2),along with a low detection limit of 27.6 nGy_(air) s^(−1),attributed to the release of residual stress, and the enhancement in carrier mobility-lifetime product.Furthermore, the detector exhibits outstanding stability under X-ray irradiation with tolerating doses equivalent to nearly 1.17×10^(6) chest imaging doses.

文摘In the age of big data, ensuring data privacy while enabling efficient encrypted data retrieval has become a critical challenge.Traditional searchable encryption schemes face difficulties in handling complex semantic queries.Additionally, they typically rely on honest but curious cloud servers, which introduces the risk of repudiation.Furthermore, the combined operations of search and verification increase system load, thereby reducing performance.Traditional verification mechanisms, which rely on complex hash constructions, suffer from low verification efficiency. To address these challenges, this paper proposes a blockchain-based contextual semantic-aware ciphertext retrieval scheme with efficient verification.Building on existing single and multi-keyword search methods, the scheme uses vector models to semantically train the dataset, enabling it to retain semantic information and achieve context-aware encrypted retrieval, significantly improving search accuracy.Additionally,a blockchain-based updatable master-slave chain storage model is designed, where the master chain stores encrypted keyword indexes and the slave chain stores verification information generated by zero-knowledge proofs, thus balancing system load while improving search and verification efficiency. Finally,an improved non-interactive zero-knowledge proof mechanism is introduced, reducing the computational complexity of verification and ensuring efficient validation of search results.Experimental results demonstrate that the proposed scheme offers stronger security, balanced overhead, and higher search verification efficiency.

文摘Driven by advancements in mobile internet technology, images have become a crucial data medium.Ensuring the security of image information during transmission has thus emerged as an urgent challenge. This study proposes a novel image encryption algorithm specifically designed for grayscale image security. This research introduces a new Cantor diagonal matrix permutation method. The proposed permutation method uses row and column index sequences to control the Cantor diagonal matrix, where the row and column index sequences are generated by a spatiotemporal chaotic system named coupled map lattice(CML).The high initial value sensitivity of the CML system makes the permutation method highly sensitive and secure.Additionally, leveraging fractal theory, this study introduces a chaotic fractal matrix and applies this matrix in the diffusion process. This chaotic fractal matrix exhibits selfsimilarity and irregularity.Using the Cantor diagonal matrix and chaotic fractal matrix, this paper introduces a fast image encryption algorithm involving two diffusion steps and one permutation step. Moreover, the algorithm achieves robust security with only a single encryption round, ensuring high operational efficiency.Experimental results show that the proposed algorithm features an expansive key space, robust security, high sensitivity, high efficiency, and superior statistical properties for the ciphered images. Thus, the proposed algorithm not only provides a practical solution for secure image transmission but also bridges fractal theory with image encryption techniques, thereby opening new research avenues in chaotic cryptography and advancing the development of information security technology.

文摘Adult hippocampal neurogenesis is linked to memory formation in the adult brain, with new neurons in the hippocampus exhibiting greater plasticity during their immature stages compared to mature neurons.Abnormal adult hippocampal neurogenesis is closely associated with cognitive impairment in central nervous system diseases.Targeting and regulating adult hippocampal neurogenesis have been shown to improve cognitive deficits. This review aims to expand the current understanding and prospects of targeting neurogenesis in the treatment of cognitive impairment.Recent research indicates the presence of abnormalities in AHN in several diseases associated with cognitive impairment, including cerebrovascular diseases,Alzheimer's disease,aging-related conditions, and issues related to anesthesia and surgery. The role of these abnormalities in the cognitive deficits caused by these diseases has been widely recognized, and targeting AHN is considered a promising approach for treating cognitive impairment. However, the underlying mechanisms of this role are not yet fully understood, and the effectiveness of targeting abnormal adult hippocampal neurogenesis for treatment remains limited, with a need for further development of treatment methods and detection techniques.By reviewing recent studies,we classify the potential mechanisms of adult hippocampal neurogenesis abnormalities into four categories:immunity, energy metabolism, aging, and pathological states. In immunity-related mechanisms, abnormalities in meningeal, brain, and peripheral immunity can disrupt normal adult hippocampal neurogenesis.Lipid metabolism and mitochondrial function disorders are significant energy metabolism factors that lead to abnormal adult hippocampal neurogenesis.During aging, the inflammatory state of the neurogenic niche and the expression of aging-related microRNAs contribute to reduced adult hippocampal neurogenesis and cognitive impairment in older adult patients.Pathological states of the body and emotional disorders may also result in abnormal adult hippocampal neurogenesis.Among the current strategies used to enhance this form of neurogenesis, physical therapies such as exercise, transcutaneous electrical nerve stimulation, and enriched environments have proven effective.Dietary interventions, including energy intake restriction and nutrient optimization, have shown efficacy in both basic research and clinical trials. However, drug treatments, such as antidepressants and stem cell therapy, are primarily reported in basic research, with limited clinical application. The relationship between abnormal adult hippocampal neurogenesis and cognitive impairment has garnered widespread attention, and targeting the former may be an important strategy for treating the latter. However, the mechanisms underlying abnormal adult hippocampal neurogenesis remain unclear, and treatments are lacking. This highlights the need for greater focus on translating research findings into clinical practice.

文摘Climate models are essential for understanding past, present, and future changes in atmospheric circulation, with circulation modes providing key sources of seasonal predictability and prediction uncertainties for both global and regional climates. This study assesses the performance of models participating in phase 6 of the Coupled Model Intercomparison Project in simulating interannual variability modes of Northern Hemisphere 500-hPa geopotential height during winter and summer, distinguishing predictable(potentially predictable on seasonal or longer timescales)and unpredictable(intraseasonal and essentially unpredictable at long range)components, using reanalysis data and a variance decomposition method.Although most models effectively capture unpredictable modes in reanalysis, their ability to reproduce dominant predictable modes-specifically the Pacific-North American pattern,Arctic Oscillation, and Western Pacific Oscillation in winter, and the East Atlantic and North Atlantic Oscillations in summer-varies notably. An optimal ensemble is identified to distinguish(a)predictable-external modes, dominated by external forcing,and(b)predictable-internal modes, associated with slow internal variability, during the historical period(1950-2014)and the SSP5-8.5 scenario(2036-2100).Under increased radiative forcing, the leading winter/summer predictable-external mode exhibits a more uniform spatial distribution, remarkably larger trend and annual variance, and enhanced height-sea surface temperature(SST)covariance under SSP5-8.5 compared to historical conditions. The dominant winter/summer predictable-internal modes also exhibit increased variance and height-SST covariance under SSP5-8.5,along with localized changes in spatial configuration.Minimal changes are observed in spatial distribution or variance for dominant winter/summer unpredictable modes under SSP5-8.5.This study, from a predictive perspective, deepens our understanding of model uncertainties and projected changes in circulations.
