Urban sprawl affects the sustainable development of the world’s economy and society.Confirming urban sprawl trends and proposing countermeasures have received significant attention.Cities are self-organizing systems ...Urban sprawl affects the sustainable development of the world’s economy and society.Confirming urban sprawl trends and proposing countermeasures have received significant attention.Cities are self-organizing systems with dissipative attributes,and city development is accompanied by urban entropy changes.Urban entropy change reveals the essence of urban sprawl;thus,it can be used tomeasure urban sprawl and better understand its phenomena.However,the literature on entropy change in urban sprawl research is limited to qualitative descriptions,and no convenient or effective quantitativemetrics exist.This study bridges entropy changes and urban formmetrics,analyzing the driving forces in Beijing as a demonstration case.Themain contributions and results are as follows:(1)Anewmethod was constructed to confirm compact or sprawling urban form development.(2)This research shows that the essence of urban sprawl is urban form distortions in an urban self-organizing system resulting from dominant driving forces.(3)This research also shows that urban development with general sprawl characteristics is accompanied by temporally fine-grained compact urban development,and the empirical results show that the urban development state paradigm was neither sprawl nor compact.In future urban management work,it is necessary to regularly monitor changes in various driving factors to prevent urban entropy increases and formulate or adjust urban development plans based on effective assessments.This study contributes by developing a new universal analysis tool and exploring the urban sprawl mechanism,with potential application scenarios that can be extended to any city or region worldwide.展开更多

Objective: To explore the relationship of postoperative cognitive dysfunction (POCD) in one-lung ventilation (OLV) patients and regional cerebral oxygen saturation (rSO2). Methods: Twenty-nine male and twenty-...Objective: To explore the relationship of postoperative cognitive dysfunction (POCD) in one-lung ventilation (OLV) patients and regional cerebral oxygen saturation (rSO2). Methods: Twenty-nine male and twenty-one female cases of OLV received thoracic surgery, with American Standards Association (ASA) physical status being at Grades Ⅰ-Ⅲ. Neuropsychological tests were performed on the day before operation and 7 d after operation, and there was an intraoperative continuous monitoring of rSO2. The values of rSO2 before anesthesia induction (t1), at the beginning of OLV (t2), and at the time of OLV 30 min (t3), OLV 60 min (t4), the end of OLV (t5), and the end of surgery (t6) were recorded. The intraoperative average of rSO2 ( rSO2 ), the intraoperative minimum value of rSO2 (rSO2, rn=n), and the reduced maximum percentage of rSO2 (rSO2, %max) when compared with the baseline value were calculated. The volume of blood loss, urine output, and the amount of fluid infusion was recorded. Results: A total of 14 patients (28%) in the 50 cases suffered from POCD. The values of mini-mental state examination (MMSE), the digit span and the digit symbol on the 7th day after the operation for POCD in OLV patients were found to be significantly lower than those before the operation (P〈0.05). The values of MMSE and vocabulary fluency scores were significantly lower than those in the non-POCD group (P〈0.05). The values of rSO2 in the POCD group of OLV patients at t2 and t3 and the values of rSO2 in the non-POCD group at t2 were found to be significantly higher than those at tl (P〈0.05). The values of rSO2, %max in the POCD group were significantly higher than those in the non-POCD group (P〈0.05). When the value of rSO2, %max is more than 10.1%, it may act as an early warning index for cognitive function changes, Conclusions: POCD after OLV may be associated with a decline in rSO2.展开更多

Animal model of anti-tuberculosis drug-induced liver injury was established by intragastric administration isoniazid + rifampicin. PDTC group received intraperitoneal injection of PDTC,and AG490 group received intraperitoneal injection of AG490. Twenty-eight days after intervention,the rats were executed,and the liver injury indexes,inflammation indexes and oxidative stress indexes in serum as well as JAK2/STAT3 expression,liver injury indexes,inflammation indexes and oxidative stress indexes in liver tissue were determined. Results:p-JAK2,p-STAT3,TNF-α,IL-1β,IL-6,ROS,8-OHdG and MDA expression in liver tissue as well as TBIL,ALT,AST,γ-GT,TNF-α,IL-1β,IL-6,ROS,8-OHdG and MDA levels in serum of model group were significantly higher than those of normal group while p-JAK2,p-STAT3,TNF-α,IL-1β,IL-6,ROS,8-OHdG and MDA expression in liver tissu as well as TBIL,ALT,AST,γ-GT,TNF-α,IL-1β,IL-6,ROS,8-OHdG and MDA levels in serum of PDTC group and AG490 group were significantly lower than those of model group. Conclusions:PDTC can inhibit the inflammation and oxidative stress mediated by JAK2/STAT3 signaling pathway to alleviate the anti-tuberculosis drug-induced liver injury.展开更多

Methods: Fifty patients undergoing elective laparoscopic pancreaticoduodenectomy received five groups of neuropsychological tests 1 d pre-operatively and 7 d post-operatively, with continuous monitoring of rSO2 intra-operatively. Before anesthesia induction(t0), at the beginning of laparoscopy(t1), and at the time of pneumoperitoneum 120 min(t2), pneumoperitoneum 240 min(t3), pneumoperitoneum 480 min(t4), the end of pneumoperitoneum(t5), and 24 h after surgery, jugular venous blood was drawn respectively for the measurement of Aβ by enzyme-linked immunosorbent assay(ELISA). Results: Twenty-one cases of the fifty patients suffered from POCD after operation. We found that the maximum percentage drop in rSO2(rSO2, %max) was significantly higher in the POCD group than in the non-POCD group. The rSO2, %max value of over 10.2% might be a potential predictor of neurocognitive injury for those patients. In the POCD group, the plasma Aβ levels after 24 h were significantly higher than those of pre-operative values(P〈0.01). After 24 h, levels of plasma Aβ in the POCD group were significantly higher than those in the non-POCD group(P〈0.01). Conclusions: The development of POCD in patients undergoing laparoscopic pancreaticoduodenectomy is associated with alterations of rSO2 and Aβ. Monitoring of rSO2 might be useful in the prediction of POCD, and Aβ might be used as a sensitive biochemical marker to predict the occurrence of POCD.展开更多

The westem Europe has a longer history of agricultural land development than...The northeastern China, the United States, and the western Europe are important agricultural regions both on the global and regional scales. The westem Europe has a longer history of agricultural land development than the eastem United States. These two regions have changed from the deforestation and reclamation phase in the past to the current land abandonment and reforestation phase. Compared with the two regions, large-scale land exploitation has only been practiced in the northeastern China during the last century. After a short high-intensity deforestation and reclamation period, agricultural and forest lands are basically in a dynamic steady state. By comparing domestic and international agro-forestry development and considering the ecological environment and socio-economic bene- fits that can be derived from agro-forestry, this paper suggests that large area of reforestation would be inevitable in future though per- sistent and large agricultural demand in coming decades even more. And local reforestation at slope farmland with ecological vulner- ability should be imperative at present to avoid severer damage. At the same time, from the perspective of Land Change Science, the results demonstrate that the research on land use change in the agro-forestry ecotone is typical and critical, particularly those dealing with the analysis of spatial and temporal characteristics and the simulation of climate, hydrology, and other environmental effects.展开更多

The treatment was performed once a day, 7 times was 1 course, and the curative effect was evaluated after 1 course of treatment.Results: Among 20 patients, 12 were cured, improvement was observed in 7 patients, and 1 case had no response to treatment. The effective rate was up to 95.0%;improvement was observed in 14 patients after treatment for 3 times. According to follow-up visit 1 month after treatment, 1 patient relapsed due to excessive mouth opening, but the symptom was mild and relieved after rubbing.Conclusion: The clinical efficacy of treatment for temporomandibular disorders with warming-needle moxibustion on ginger at ST 7 was ideal.展开更多

This study investigated land use pattern and spatial he...Ecotones have received great attention due to its critical function in energy flux, species harbor, global carbon sequestration, and land-atmosphere interaction. This study investigated land use pattern and spatial heterogeneity of the ecotones among agricultural land, forest land, and grassland of the southeastern Da Hinggan Mountains in the northeastern China. The change of these delineated ecotones under different slopes and aridity conditions was analyzed by two landscape indices, edge density(ED) and core area percentage of landscape(CPL), to explore the inter-linkage between spatial structure of ecotones and socioeconomic development and land management. Specifically, the ecotones such as agriculture-forest(AF) ecotone, forest-grassland(FG) ecotone, and agriculture-forestgrassland(AFG) ecotone moved from the arid southeast to the humid northwest. The flat area with small slope is more edge-fragmented than the steep area since the ED decreases as the slope increases. The AF ecotone mostly found in the humid region is moving to more humid areas while the agriculture-grassland(AG) ecotone mostly found in the dry region is moving towards the drier region.展开更多

The fusion gene was cloned into pENTR/D-TOPO transfer vector after the base pairs of DNA (CACC) sequence was added to the 5′ end. Adenoviral expression vector containing CRT-HBsAg fusion gene was constructed by homologous recombinan-tion. The human embryo kidney (HEK) 293A cells were transfected with linearized DNA plasmid of the recombi-nant adenoviral vector to package and amplify recombi-nant adenovirus. The recombinant adenovirus titer was characterized using the end-dilution assay. The expres-sion of the CRT/HBsAg fusion protein in Ad-CRT/HBsAg infected 293A cells was detected by Western blotting.RESULTS: The CRT-HBsAg fusion gene was char-acterized by PCR and sequencing and its length and sequence were confirmed to be accurate. The CRT-HB-sAg fusion gene recombinant pENTR/D-TOPO transfer vector was constructed. The recombinant adenoviral vector, Ad-CRT/HBsAg, was generated successfully. The titer of Ad-CRT/HBsAg was characterized as 3.9 × 1011 pfu/mL. The CRT-HBsAg fusion protein was ex-pressed by HEK 293A cells correctly. CONCLUSION: CRT/HBsAg fusion gene recombinant replication-defective adenovirus expression vector is constructed successfully and this study has provided an experimental basis for further studies of Hepatitis B vi-rus gene therapy.展开更多

Methods: The xenograft model was e...Objective: The aim of this study was to investigate the effect of grape proanthocyanidins(GPC) on the growth and angiogenesis of hepatocellular carcinoma H22 cells xenograft in mice. Methods: The xenograft model was established using injected subcutaneously H22 cells into the right axilla of the mice. Each group was treated with different doses of GPC and Endostar. All these treatments were maintained for 10 days, and mice were sacrificed. The xenograft tumors in mice were measured. The proliferation activity level of H22 cells was determined by MTT assay, and the levels of vascular endothelial growth factor(VEGF) protein were examined by immunohistochemistry. Results: When treated with 50, 100 and 200 mg/kg of GPC and Endostar, the tumor inhibition rates were 13.17%, 23.37%, 36.15% and 14.71%, respectively. The tumor weight of xenograft was significantly lighter in high GPC group than the control group(P < 0.05). The ODs in GPC groups were 0.835, 0.666 and 0.519, respectively. The absorbances in middle and high GPC groups were statistically significant, compared with control group(P < 0.01). Immunohistochemical technique showed the expression of VEGF of the GPC groups was downregulated significantly compared with the control group(P < 0.01). Conclusion: GPC can inhibit the growth of hepatocellular carcinoma H22 cell xenograft in mice. The inhibition of angiogenesis by the down-regulation of VEGF expression may play a key role in the anti-neoplastic effect of GPC.展开更多

Tumor necrosis factor (TNF), as one of importa...BACKGROUND: Lots of evidences have demonstrated that acute inflammatory reaction plays an important role in cerebral ischemia and cerebral ischemia/reperfusion injury. Tumor necrosis factor (TNF), as one of important inflammatory cytokines, also participates in the injury. OBJECTIVE: To observe the changes in TNF-α expression and myeloperoxidase (MPO) activity of mouse models of local cerebral infarction induced by photochemical method, and analyze the correlation of TNF-α expression and MPO activity. DESIGN: Randomized controlled experiment. SETTING: Laboratory of Cerebral Microcirculation, Taishan Medical College. MATERIALS: Sixty involved male adult Kunming mice were provided by the Experimental Animal Center of Shandong University. TNF-α primary antibody, kits for enzyme-linked immunosorbent assay(ELISA) and streptavidin-biotin complex immunohistochemical dyeing kit were purchased from Boster Company(Wuhan). MPO kit was purchased from Jiancheng Bioengineering Institute (Nanjing). Cold light source was developed by Hengfa Co.,Ltd.( LG-150, Xuzhou). METHODS: This experiment was carried out in the Laboratory of Cerebral Microcirculation of Taishan Medical College between July 2004 and July 2005. The involved 60 Kumning mice were randomized into 3 groups: normal control group (n =6), sham-operation group (n =6) and model group (n =48). Mice in the model group were observed at 30 minutes, 1, 3, 6, 12, 24, 48 and 72 hours after illumination, separately, 6 mice at each time point. In the model group, mice models of local cerebral infarction were developed as follows: The mice were anesthetized to expose left skulls. Taking 2 mm left to sagittal suture and 2 mm posterior to coronal suture as center, a field with diameter of 3 mm for illumination was set. The optical fiber detecting head of cold light source was vertically close to exposed skull. The mice were injected with rose Bengal for 5 minutes, and then cold light source was open for 10 minutes, Illumination was omitted in the sham-operation group. Mice in the control group were not modeled. At postoperative 6 hours, TNF-α expression in infracted-side cortex was detected with immunohistochemical method and ELISA, and MPO activity in infracted-side cortex with chromatometry. MPO activity could reflect the infiltration degree of neutrophils in tissue. Stronger activity indicated severer infiltration. Single-factor analysis of variance was used for comparison among groups, q test for pairwise comparison and correlative analysis for detecting the inter-parameter correlation. MAIN OUTCOME MEASURES: Changes in TNF-α expression and MPO activity of left cortex of mice in each group. RESULTS: Sixty mice were involved in the final analysis. After cerebral infarction, TNF-α positive cells were neurons and glial cells mainly, distributing in and around the infarct region. TNF-α expression in cortex of mice of sham-operation group was (615.7 ±16.1 ) ng/L, and that of model group increased to (792.2± 17.8) ng/L at 3 hours after illumination, and reached peak [ (921.9±23.9) ng/L] at 6 hours after illumination, and decreased to (848.0±30.6) ng/L at 12 hours after illumination and recovered to the normal level [ (625.3 ± 14.3) ng/L] at 72 hours after illumination. MPO activity of sham-operation group was (7.151±0.433) nkat/g, and that of model group increased to (10.469±0.600) nkat/g at 3 hours after illumination, reached the peak [ ( 15.486 ± 0.650) nkat/g] at 12 hours after illumination, decreased to (11.052 ± 0,617) nkat/g at 24 hours after illumination and recovered to the normal level [ (7.418 ± 0.617) nkat/g] at 72 hours after illumination. Change of MPO activity lagged behind that of TNF-α, and correlative analysis showed that the both were positively correlated (r =0.953, P 〈 0.01 ) . CONCLUSION: In the acute stage of cerebral infarction of mice induced by photochemical method, TNF-α expression in infarcted-side cortex is closely related with infiltration of neutrophils. TNF-α induces inflammatory cells to intrude into ischemic brain tissue, and participates in the inflammatory reaction process at the early stage of cerebral ischemia.展开更多

The levels of neurotransmitters, nerve cytokines and stress indexes in serum were measured before treatment as well as 14 days and 28 days after treatment.Results: Compared with those before treatment, serum Glu,γ-GABA, 5-HT, 5-HIAA, BDNF and NGF levels of both groups significantly increased whereas NE, TNF-α, IL-6, ATCH, Cor, CRP, SOD and GSH levels significantly decreased after treatment, and serum Glu,γ-GABA, 5-HT, 5-HIAA, BDNF and NGF levels of dexzopiclone group after treatment were higher than those of estazolam group whereas NE, TNF-α, IL-6, ATCH, Cor, CRP, SOD and GSH levels were lower than those of estazolam group.Conclusion:Dexzopiclone is more effective than estazolam to regulate the secretion of neurotransmitters and nerve cytokines and reduce the stress state in patients with insomnia after stroke.展开更多

The differences in the levels of hk10 and DJ-1 in preoperative serum as well as the expression of ovarian cancer-related proliferation genes and invasion genes in intraoperative lesion tissue were compared between the two groups. Correlation analysis was used to assess the inner link of serum hk10 and DJ-1 levels with tumor malignancy in patients with ovarian cancer.Results: Serum hk10 and DJ-1 levels of ovarian cancer group were higher than those of ovarian cyst group;proliferation genes Clusterin, FUNDC1, AKT, TNFAIP8 and HBXIP mRNA expression in lesion tissues were higher than those of ovarian cyst group;invasion genes Beclin1, BRMS1 and SIRT2 mRNA expression in lesion tissues were lower than those of ovarian cyst group whereas EFEMP1 mRNA expression was higher than that of ovarian cyst group. The Pearson test showed that serum hk10 and DJ-1 levels in ovarian cancer were directly correlated with the expression of proliferation genes and invasion genes. Conclusion: hk10 and DJ-1 are highly expressed in serum of patients with ovarian cancer, and their levels are directly correlated with the proliferation and invasion activity of cancer cells.展开更多

文摘Urban sprawl affects the sustainable development of the world’s economy and society.Confirming urban sprawl trends and proposing countermeasures have received significant attention.Cities are self-organizing systems with dissipative attributes,and city development is accompanied by urban entropy changes.Urban entropy change reveals the essence of urban sprawl;thus,it can be used tomeasure urban sprawl and better understand its phenomena.However,the literature on entropy change in urban sprawl research is limited to qualitative descriptions,and no convenient or effective quantitativemetrics exist.This study bridges entropy changes and urban formmetrics,analyzing the driving forces in Beijing as a demonstration case.Themain contributions and results are as follows:(1)Anewmethod was constructed to confirm compact or sprawling urban form development.(2)This research shows that the essence of urban sprawl is urban form distortions in an urban self-organizing system resulting from dominant driving forces.(3)This research also shows that urban development with general sprawl characteristics is accompanied by temporally fine-grained compact urban development,and the empirical results show that the urban development state paradigm was neither sprawl nor compact.In future urban management work,it is necessary to regularly monitor changes in various driving factors to prevent urban entropy increases and formulate or adjust urban development plans based on effective assessments.This study contributes by developing a new universal analysis tool and exploring the urban sprawl mechanism,with potential application scenarios that can be extended to any city or region worldwide.

文摘Objective: To explore the relationship of postoperative cognitive dysfunction (POCD) in one-lung ventilation (OLV) patients and regional cerebral oxygen saturation (rSO2). Methods: Twenty-nine male and twenty-one female cases of OLV received thoracic surgery, with American Standards Association (ASA) physical status being at Grades Ⅰ-Ⅲ. Neuropsychological tests were performed on the day before operation and 7 d after operation, and there was an intraoperative continuous monitoring of rSO2. The values of rSO2 before anesthesia induction (t1), at the beginning of OLV (t2), and at the time of OLV 30 min (t3), OLV 60 min (t4), the end of OLV (t5), and the end of surgery (t6) were recorded. The intraoperative average of rSO2 ( rSO2 ), the intraoperative minimum value of rSO2 (rSO2, rn=n), and the reduced maximum percentage of rSO2 (rSO2, %max) when compared with the baseline value were calculated. The volume of blood loss, urine output, and the amount of fluid infusion was recorded. Results: A total of 14 patients (28%) in the 50 cases suffered from POCD. The values of mini-mental state examination (MMSE), the digit span and the digit symbol on the 7th day after the operation for POCD in OLV patients were found to be significantly lower than those before the operation (P〈0.05). The values of MMSE and vocabulary fluency scores were significantly lower than those in the non-POCD group (P〈0.05). The values of rSO2 in the POCD group of OLV patients at t2 and t3 and the values of rSO2 in the non-POCD group at t2 were found to be significantly higher than those at tl (P〈0.05). The values of rSO2, %max in the POCD group were significantly higher than those in the non-POCD group (P〈0.05). When the value of rSO2, %max is more than 10.1%, it may act as an early warning index for cognitive function changes, Conclusions: POCD after OLV may be associated with a decline in rSO2.

文摘Objective:To study the effect of pyrrolidine dithiocarbamate(PDTC) on the anti-tuberculosis drug-induced liver injury and the molecular mechanism. Methods:Clean male SD rats were selected as experimental animals and randomly divided into normal group,model group,PDTC group and AG490 group. Animal model of anti-tuberculosis drug-induced liver injury was established by intragastric administration isoniazid + rifampicin. PDTC group received intraperitoneal injection of PDTC,and AG490 group received intraperitoneal injection of AG490. Twenty-eight days after intervention,the rats were executed,and the liver injury indexes,inflammation indexes and oxidative stress indexes in serum as well as JAK2/STAT3 expression,liver injury indexes,inflammation indexes and oxidative stress indexes in liver tissue were determined. Results:p-JAK2,p-STAT3,TNF-α,IL-1β,IL-6,ROS,8-OHdG and MDA expression in liver tissue as well as TBIL,ALT,AST,γ-GT,TNF-α,IL-1β,IL-6,ROS,8-OHdG and MDA levels in serum of model group were significantly higher than those of normal group while p-JAK2,p-STAT3,TNF-α,IL-1β,IL-6,ROS,8-OHdG and MDA expression in liver tissu as well as TBIL,ALT,AST,γ-GT,TNF-α,IL-1β,IL-6,ROS,8-OHdG and MDA levels in serum of PDTC group and AG490 group were significantly lower than those of model group. Conclusions:PDTC can inhibit the inflammation and oxidative stress mediated by JAK2/STAT3 signaling pathway to alleviate the anti-tuberculosis drug-induced liver injury.

文摘Objective: To investigate the relationship between post-operative cognitive dysfunction(POCD) and regional cerebral oxygen saturation(rSO2) and β-amyloid protein(Aβ) in patients undergoing laparoscopic pancreaticoduodenectomy. Methods: Fifty patients undergoing elective laparoscopic pancreaticoduodenectomy received five groups of neuropsychological tests 1 d pre-operatively and 7 d post-operatively, with continuous monitoring of rSO2 intra-operatively. Before anesthesia induction(t0), at the beginning of laparoscopy(t1), and at the time of pneumoperitoneum 120 min(t2), pneumoperitoneum 240 min(t3), pneumoperitoneum 480 min(t4), the end of pneumoperitoneum(t5), and 24 h after surgery, jugular venous blood was drawn respectively for the measurement of Aβ by enzyme-linked immunosorbent assay(ELISA). Results: Twenty-one cases of the fifty patients suffered from POCD after operation. We found that the maximum percentage drop in rSO2(rSO2, %max) was significantly higher in the POCD group than in the non-POCD group. The rSO2, %max value of over 10.2% might be a potential predictor of neurocognitive injury for those patients. In the POCD group, the plasma Aβ levels after 24 h were significantly higher than those of pre-operative values(P〈0.01). After 24 h, levels of plasma Aβ in the POCD group were significantly higher than those in the non-POCD group(P〈0.01). Conclusions: The development of POCD in patients undergoing laparoscopic pancreaticoduodenectomy is associated with alterations of rSO2 and Aβ. Monitoring of rSO2 might be useful in the prediction of POCD, and Aβ might be used as a sensitive biochemical marker to predict the occurrence of POCD.

文摘The northeastern China, the United States, and the western Europe are important agricultural regions both on the global and regional scales. The westem Europe has a longer history of agricultural land development than the eastem United States. These two regions have changed from the deforestation and reclamation phase in the past to the current land abandonment and reforestation phase. Compared with the two regions, large-scale land exploitation has only been practiced in the northeastern China during the last century. After a short high-intensity deforestation and reclamation period, agricultural and forest lands are basically in a dynamic steady state. By comparing domestic and international agro-forestry development and considering the ecological environment and socio-economic bene- fits that can be derived from agro-forestry, this paper suggests that large area of reforestation would be inevitable in future though per- sistent and large agricultural demand in coming decades even more. And local reforestation at slope farmland with ecological vulner- ability should be imperative at present to avoid severer damage. At the same time, from the perspective of Land Change Science, the results demonstrate that the research on land use change in the agro-forestry ecotone is typical and critical, particularly those dealing with the analysis of spatial and temporal characteristics and the simulation of climate, hydrology, and other environmental effects.
文摘Objective: To observe the clinical efficacy of treatment for temporomandibular disorders with warmingneedle moxibustion on ginger at Xiàguan(下关ST 7).Methods: Twenty patients with temporomandibular disorders were given acupuncture at ST 7 of the affected side, and then the self-prepared round cake-shaped ginger with moxa was set on the needle closing to skin surface, Ignited moxa, burn out as one cone, three cones were given for once. The treatment was performed once a day, 7 times was 1 course, and the curative effect was evaluated after 1 course of treatment.Results: Among 20 patients, 12 were cured, improvement was observed in 7 patients, and 1 case had no response to treatment. The effective rate was up to 95.0%;improvement was observed in 14 patients after treatment for 3 times. According to follow-up visit 1 month after treatment, 1 patient relapsed due to excessive mouth opening, but the symptom was mild and relieved after rubbing.Conclusion: The clinical efficacy of treatment for temporomandibular disorders with warming-needle moxibustion on ginger at ST 7 was ideal.

文摘Ecotones have received great attention due to its critical function in energy flux, species harbor, global carbon sequestration, and land-atmosphere interaction. This study investigated land use pattern and spatial heterogeneity of the ecotones among agricultural land, forest land, and grassland of the southeastern Da Hinggan Mountains in the northeastern China. The change of these delineated ecotones under different slopes and aridity conditions was analyzed by two landscape indices, edge density(ED) and core area percentage of landscape(CPL), to explore the inter-linkage between spatial structure of ecotones and socioeconomic development and land management. Specifically, the ecotones such as agriculture-forest(AF) ecotone, forest-grassland(FG) ecotone, and agriculture-forestgrassland(AFG) ecotone moved from the arid southeast to the humid northwest. The flat area with small slope is more edge-fragmented than the steep area since the ED decreases as the slope increases. The AF ecotone mostly found in the humid region is moving to more humid areas while the agriculture-grassland(AG) ecotone mostly found in the dry region is moving towards the drier region.

文摘AIM: To generate recombinant adenoviral vector con-taining calreticulin (CRT)-hepatitis B surface antigen (HBsAg) fusion gene for developing a safe, effective and HBsAg-specific therapeutic vaccine.METHODS: CRT and HBsAg gene were fused using polymerase chain reaction (PCR), endonuclease diges-tion and ligation methods. The fusion gene was cloned into pENTR/D-TOPO transfer vector after the base pairs of DNA (CACC) sequence was added to the 5′ end. Adenoviral expression vector containing CRT-HBsAg fusion gene was constructed by homologous recombinan-tion. The human embryo kidney (HEK) 293A cells were transfected with linearized DNA plasmid of the recombi-nant adenoviral vector to package and amplify recombi-nant adenovirus. The recombinant adenovirus titer was characterized using the end-dilution assay. The expres-sion of the CRT/HBsAg fusion protein in Ad-CRT/HBsAg infected 293A cells was detected by Western blotting.RESULTS: The CRT-HBsAg fusion gene was char-acterized by PCR and sequencing and its length and sequence were confirmed to be accurate. The CRT-HB-sAg fusion gene recombinant pENTR/D-TOPO transfer vector was constructed. The recombinant adenoviral vector, Ad-CRT/HBsAg, was generated successfully. The titer of Ad-CRT/HBsAg was characterized as 3.9 × 1011 pfu/mL. The CRT-HBsAg fusion protein was ex-pressed by HEK 293A cells correctly. CONCLUSION: CRT/HBsAg fusion gene recombinant replication-defective adenovirus expression vector is constructed successfully and this study has provided an experimental basis for further studies of Hepatitis B vi-rus gene therapy.

文摘Objective: The aim of this study was to investigate the effect of grape proanthocyanidins(GPC) on the growth and angiogenesis of hepatocellular carcinoma H22 cells xenograft in mice. Methods: The xenograft model was established using injected subcutaneously H22 cells into the right axilla of the mice. Each group was treated with different doses of GPC and Endostar. All these treatments were maintained for 10 days, and mice were sacrificed. The xenograft tumors in mice were measured. The proliferation activity level of H22 cells was determined by MTT assay, and the levels of vascular endothelial growth factor(VEGF) protein were examined by immunohistochemistry. Results: When treated with 50, 100 and 200 mg/kg of GPC and Endostar, the tumor inhibition rates were 13.17%, 23.37%, 36.15% and 14.71%, respectively. The tumor weight of xenograft was significantly lighter in high GPC group than the control group(P < 0.05). The ODs in GPC groups were 0.835, 0.666 and 0.519, respectively. The absorbances in middle and high GPC groups were statistically significant, compared with control group(P < 0.01). Immunohistochemical technique showed the expression of VEGF of the GPC groups was downregulated significantly compared with the control group(P < 0.01). Conclusion: GPC can inhibit the growth of hepatocellular carcinoma H22 cell xenograft in mice. The inhibition of angiogenesis by the down-regulation of VEGF expression may play a key role in the anti-neoplastic effect of GPC.

文摘BACKGROUND: Lots of evidences have demonstrated that acute inflammatory reaction plays an important role in cerebral ischemia and cerebral ischemia/reperfusion injury. Tumor necrosis factor (TNF), as one of important inflammatory cytokines, also participates in the injury. OBJECTIVE: To observe the changes in TNF-α expression and myeloperoxidase (MPO) activity of mouse models of local cerebral infarction induced by photochemical method, and analyze the correlation of TNF-α expression and MPO activity. DESIGN: Randomized controlled experiment. SETTING: Laboratory of Cerebral Microcirculation, Taishan Medical College. MATERIALS: Sixty involved male adult Kunming mice were provided by the Experimental Animal Center of Shandong University. TNF-α primary antibody, kits for enzyme-linked immunosorbent assay(ELISA) and streptavidin-biotin complex immunohistochemical dyeing kit were purchased from Boster Company(Wuhan). MPO kit was purchased from Jiancheng Bioengineering Institute (Nanjing). Cold light source was developed by Hengfa Co.,Ltd.( LG-150, Xuzhou). METHODS: This experiment was carried out in the Laboratory of Cerebral Microcirculation of Taishan Medical College between July 2004 and July 2005. The involved 60 Kumning mice were randomized into 3 groups: normal control group (n =6), sham-operation group (n =6) and model group (n =48). Mice in the model group were observed at 30 minutes, 1, 3, 6, 12, 24, 48 and 72 hours after illumination, separately, 6 mice at each time point. In the model group, mice models of local cerebral infarction were developed as follows: The mice were anesthetized to expose left skulls. Taking 2 mm left to sagittal suture and 2 mm posterior to coronal suture as center, a field with diameter of 3 mm for illumination was set. The optical fiber detecting head of cold light source was vertically close to exposed skull. The mice were injected with rose Bengal for 5 minutes, and then cold light source was open for 10 minutes, Illumination was omitted in the sham-operation group. Mice in the control group were not modeled. At postoperative 6 hours, TNF-α expression in infracted-side cortex was detected with immunohistochemical method and ELISA, and MPO activity in infracted-side cortex with chromatometry. MPO activity could reflect the infiltration degree of neutrophils in tissue. Stronger activity indicated severer infiltration. Single-factor analysis of variance was used for comparison among groups, q test for pairwise comparison and correlative analysis for detecting the inter-parameter correlation. MAIN OUTCOME MEASURES: Changes in TNF-α expression and MPO activity of left cortex of mice in each group. RESULTS: Sixty mice were involved in the final analysis. After cerebral infarction, TNF-α positive cells were neurons and glial cells mainly, distributing in and around the infarct region. TNF-α expression in cortex of mice of sham-operation group was (615.7 ±16.1 ) ng/L, and that of model group increased to (792.2± 17.8) ng/L at 3 hours after illumination, and reached peak [ (921.9±23.9) ng/L] at 6 hours after illumination, and decreased to (848.0±30.6) ng/L at 12 hours after illumination and recovered to the normal level [ (625.3 ± 14.3) ng/L] at 72 hours after illumination. MPO activity of sham-operation group was (7.151±0.433) nkat/g, and that of model group increased to (10.469±0.600) nkat/g at 3 hours after illumination, reached the peak [ ( 15.486 ± 0.650) nkat/g] at 12 hours after illumination, decreased to (11.052 ± 0,617) nkat/g at 24 hours after illumination and recovered to the normal level [ (7.418 ± 0.617) nkat/g] at 72 hours after illumination. Change of MPO activity lagged behind that of TNF-α, and correlative analysis showed that the both were positively correlated (r =0.953, P 〈 0.01 ) . CONCLUSION: In the acute stage of cerebral infarction of mice induced by photochemical method, TNF-α expression in infarcted-side cortex is closely related with infiltration of neutrophils. TNF-α induces inflammatory cells to intrude into ischemic brain tissue, and participates in the inflammatory reaction process at the early stage of cerebral ischemia.

文摘Objective:To study the effects of dexzopiclone and estazolam on neurotransmitters, nerve cytokines and stress states in patients with insomnia after stroke.Methods: The patients with insomnia after stroke who were treated in Linyi Rongjun Hospital between March 2015 and October 2017 were chosen and randomly divided into dexzopiclone group and estazolam group. The levels of neurotransmitters, nerve cytokines and stress indexes in serum were measured before treatment as well as 14 days and 28 days after treatment.Results: Compared with those before treatment, serum Glu,γ-GABA, 5-HT, 5-HIAA, BDNF and NGF levels of both groups significantly increased whereas NE, TNF-α, IL-6, ATCH, Cor, CRP, SOD and GSH levels significantly decreased after treatment, and serum Glu,γ-GABA, 5-HT, 5-HIAA, BDNF and NGF levels of dexzopiclone group after treatment were higher than those of estazolam group whereas NE, TNF-α, IL-6, ATCH, Cor, CRP, SOD and GSH levels were lower than those of estazolam group.Conclusion:Dexzopiclone is more effective than estazolam to regulate the secretion of neurotransmitters and nerve cytokines and reduce the stress state in patients with insomnia after stroke.
文摘Objective:To discuss the value of serum hk10 and DJ-1 levels in patients with ovarian cancer for evaluating the malignant biology of tumor.Methods:A total of80 patients with ovarian cancer who underwent surgical treatment in our hospital between August 2015 and February 2018 were selected as ovarian cancer group, and 67 patients with ovarian cyst were selected as ovarian cyst group. The differences in the levels of hk10 and DJ-1 in preoperative serum as well as the expression of ovarian cancer-related proliferation genes and invasion genes in intraoperative lesion tissue were compared between the two groups. Correlation analysis was used to assess the inner link of serum hk10 and DJ-1 levels with tumor malignancy in patients with ovarian cancer.Results: Serum hk10 and DJ-1 levels of ovarian cancer group were higher than those of ovarian cyst group;proliferation genes Clusterin, FUNDC1, AKT, TNFAIP8 and HBXIP mRNA expression in lesion tissues were higher than those of ovarian cyst group;invasion genes Beclin1, BRMS1 and SIRT2 mRNA expression in lesion tissues were lower than those of ovarian cyst group whereas EFEMP1 mRNA expression was higher than that of ovarian cyst group. The Pearson test showed that serum hk10 and DJ-1 levels in ovarian cancer were directly correlated with the expression of proliferation genes and invasion genes. Conclusion: hk10 and DJ-1 are highly expressed in serum of patients with ovarian cancer, and their levels are directly correlated with the proliferation and invasion activity of cancer cells.
