OsPATA1 deficiency enhances ABA-dependent salt tolerance by sequestering OsEULD1b in the cytosol of rice

Abiotic stresses,particularly salinity,pose a major threat to rice productivity,highlighting the need to identify novel genetic resources to improve stress tolerance.Gamma irradiation remains one of the most widely used tools breeding stress-tolerant plant varieties.In this study,we identified a salt-tolerant rice mutant,salt-insensitive TILLING line 4(sitl4),generated via gamma irradiation and linked its enhanced tolerance to a loss-of-function mutation in Oryza sativa protein acyltransferase for ABA response 1(OsPATA1),which encodes a DHHC-type palmitoyl acyltransferase.Functional analyses using both sitl4 and a CRISPR/Cas9-mediated OsPATA1-knockout line(ospata1)revealed that disruption of OsPATA1 leads to increased abscisic acid(ABA)accumulation and upregulation of ABA-responsive genes under salt stress conditions.We identified OsEULD1b,a previously uncharacterized Euonymus lectin(EUL)domaincontaining protein,as an interactor of OsPATA1.In sitl4 and ospata1,OsEULD1b displayed cytosolic retention,suggesting that its subcellular redistribution enhances its role in ABA-mediated stress signaling.Taken together,our findings demonstrate that OsPATA1 and OsEULD1b form a regulatory module that modulates the ABA-dependent salt stress responses in rice.These results provide new insights into the molecular mechanisms underlying abiotic stress tolerance and will help to identify potential genetic targets for developing stress-tolerant rice cultivars through molecular breeding or genome editing. 展开更多

Since the beginning of the 21st century,modern medical technology has advanced rapidly,and the cryomedicine has also seen significant progress.Notable developments include the application of cryomedicine in assisted reproduction and the cryopreservation of sperm,eggs and embryos,as well as the preservation of skin,fingers,and other isolated tissues.However,cryopreservation of large and complex tissues or organs remains highly challenging.In addition to the damage caused by the freezing and rewarming processes and the inherent complexity of tissues and organs,there is an urgent need to address issues related to damage detection and the investigation of injury mechanisms.It provides a retrospective analysis of existing methods for assessing tissue and organ viability.Although current techniques can detect damage to some extent,they tend to be relatively simple,time-consuming,and limited in their ability to provide timely and comprehensive assessments of viability.By summarizing and evaluating these approaches,our study aims to contribute to the improvement of viability detection methods and to promote further development in this critical area. 展开更多

BACKGROUND Extracellular vesicles derived from mesenchymal stromal cells(MSC-EVs)can be used for anti-aging therapy and treating various aging-related diseases.However,the clinical application of MSC-EVs is still limited,mainly due to insufficient in-formation on the preparation process,quality,and mechanism of action of MSC-EVs.To study the biological effects of MSC-EVs in regulating cellular senescence.METHODS In this study,we developed a clinical-grade production process for MSC-EVs and defined the release criteria for products suitable for human use.To support the clinical use of our product as a therapeutic agent,we performed efficacy assays to evaluate the anti-aging capacity of MSC-EVs in vitro and in vivo.RESULTS The functional analysis results revealed that MSC-EVs significantly reduced the levels of senescence-associatedβ-galactosidase,matrix metallopeptidase 1,P21,and interleukin-1βand increased the level of collagen I in a naturally aged cell model of human dermal fibroblasts.Similarly,treatment with MSC-EVs effectively improved D-gal-induced subacute aging in mice,aging-related histopathological changes,oxidative stress,and aging-related gene expression.CONCLUSION These findings indicate that MSC-EVs can partially alleviate D-gal-induced senescence by reducing oxidative stress and regulating metabolism.Overall,these findings strongly suggest that MSC-EVs hold promise for aging therapy. 展开更多

Aim: To identify the serum biomarkers of prostate cancer (PCa) by protein chip and bioinformatics. Methods: Serum samples from 83 PCa patients and 95 healthy men were taken from a mass screening in Changchun, China. Protein profiling was carried out using surface-enhanced laser desorption/ionization time-of-flight mass spectrometry (SELDI-TOF MS). The data of spectra were analyzed using two bioinformatics tools. Results: Eighteen serum differential proteins were identified in the PCa group compared with the control group (P 〈 0.01). There were four proteins at the higher serum level and 14 proteins at the lower serum level in the PCa group. A decision tree classification algorithm that used an eight-protein mass pattern was developed to correctly classify the samples. A sensitivity of 92.0 % and a specificity of 96.7 % for the study group were obtained by comparing the PCa and control groups. Conclusion: We identified new serum biomarkers of PCa. SELDI-TOF MS coupled with a decision tree classification algorithm will provide a highly accurate and innovative approach for the early diagnosis of PCa. (Asian J Androl 2006 Jan; 8: 45-51) 展开更多

Western countries are seeing a constant decline in the incidence of Helicobacter pylori-associated gastritis, coupled with a rising epidemiological and clinical impact of autoimmune gastritis. This latter gastropathy is due to autoimmune aggression targeting parietal cells through a complex interaction of auto-antibodies against the parietal cell proton pump and intrinsic factor, and sensitized T cells. Given the specific target of this aggression, autoimmune gastritis is typically restricted to the gastric corpus-fundus mucosa. In advanced cases, the oxyntic epithelia are replaced by atrophic(and metaplastic) mucosa, creating the phenotypic background in which both gastric neuroendocrine tumors and(intestinal-type) adenocarcinomas may develop. Despite improvements in our understanding of the phenotypic changes or cascades occurring in this autoimmune setting, no reliable biomarkers are available for identifying patients at higher risk of developing a gastric neoplasm. The standardization of autoimmune gastritis histology reports and classifications in diagnostic practice is a prerequisite for implementing definitive secondary prevention strategies based on multidisciplinary diagnostic approaches integratingendoscopy, serology, histology and molecular profiling. 展开更多

Several studies have shown that fibroblast growth factor-2 (FGF2) can directly affect axon regeneration after peripheral nerve damage. In this study, we performed sensory tests and histological analyses to study the effect of recombinant human FGF-2 (rhFGF2) treatment on damaged mental nerves. The mental nerves of 6-week-old male Sprague-Dawley rats were crush-injured for 1 minute and then treated with 10 or 50 μg/mL rhFGF2 or PBS in crush injury area with a mini Osmotic pump. Sensory test using von Frey filaments at 1 week revealed the presence of sensory degeneration based on decreased gap score and increased difference score. However, at 2 weeks, the gap score and difference score were significantly rebounded in the mental nerve crush group treated with 10 μg/mL rhFGF2. Interestingly, treatment with 10 μg/mL rhFGF had a more obviously positive effect on the gap score than treatment with 50 μg/mL rhFGF2. In addition, retrograde neuronal tracing with Dil revealed a significant increase in nerve regeneration in the trigeminal ganglion at 2 and 4 weeks in the rhFGF2 groups (10 μg/mL and 50 μg/mL) than in the PBS group. The 10 μg/mL rhFGF2 group also showed an obviously robust regeneration in axon density in the mental nerve at 4 weeks. Our results demonstrate that 10 μg/mL rhFGF induces mental nerve regeneration and sensory recovery after mental nerve crush injury. 展开更多

The branched-chain amino acids (BCAAs) valine, leucine and isoleucine are essential amino acids that are critical for animal growth and development. Animals need to obtain BCAAs from their diet because they cannot synthesize them. Plants are the ultimate source of these amino acids. Acetolactate synthase (ALS) is the first common enzyme in the biosynthesis of BCAAs. The metabolic control of BCAA biosynthesis involves allosteric regulation of ALS by the end-products of the pathway, i.e., valine, leucine and isoleucine. ALS holoenzyme seems to consist of two large catalytic subunits and two small regulatory subunits. In a previous study, using homologous recombination dependent gene targeting we created rice plants in which W548Land S627I mutations were induced into the endogenous gene encoding the ALS catalytic subunit. These two amino acid substitutions conferred hypertolerance to the ALS-inhibiting herbicide bispyripac-sodium. In this study, we revealed that feedback regulation by valine and leucine was reduced by these two amino acid substitutions. Furthermore, in leaves and seeds of ALS mutants with W548Land/or S627I substitution, a 2- to 3-fold increase in BCAAs was detected. Our results suggest that the ALS catalytic subunit is also involved in feedback regulation of ALS, and that judicious modification of the regulatory and catalytic subunits of ALS-coding genes by gene targeting can lead to the efficient accumulation of BCAA in plants. 展开更多

AIM: To investigate the(-)-epigallocatechin-3-gallate(EGCG) binding to transforming growth factor-β(TGF-β) type Ⅱ receptor(TGFRⅡ).METHODS: The expression of α-smooth muscle actin(α-SMA) was used as a marker for fibrotic change inhuman lung fibroblast MRC-5 cells. The α-SMA expression level was determined by western blotting and immunohistological analysis. We examined whether the anti-fibrotic effects of EGCG on MRC-5 cells was dependent on antioxidant mechanism by using edaravone and N-acetylcysteine(NAC). The suppression effects of EGCG on Smad2/3 activation were studied by confocal fluorescence microscopy. The binding of EGCG to recombinant TGFRⅡ protein was analyzed by immunoprecipitation and affinity chromatography.RESULTS: When MRC-5 cells were treated with TGF-β, EGCG decreased the expression of α-SMA in a dose dependent manner, whereas catechin did not influence the α-SMA expression in the cells. Except for EGCG, antioxidant compounds(e.g., edaravone and NAC) had no effects on the TGF-β-induced α-SMA expression. Nuclear localization of phosphorylated Smad2/3 was observed after TGF-β treatment; however, EGCG treatment attenuated the nuclear transportation of Smad2/3 in the presence or absence of TGF-β. After a TGFRⅡ expression vector was introduced into COS-7 cells, cell lysates were untreated or treated with EGCG or catechin. The immunoprecipitation experiments using the lysates showed that EGCG dose-dependently bound to TGFRⅡ and that catechin did not at all. Affinity chromatography study indicated that EGCG would bind to TGFRⅡ.CONCLUSION: Our results demonstrate that EGCG interacts with TGFRⅡ and inhibits the expression of α-SMA via the TGF-β-Smad2/3 pathway in human lung fibroblast MRC-5 cells. 展开更多

Objective:To identify the prevalence of soil-transmitted helminthiasis(STH)among school-age children in the Cagayan Valley,the Philippines,assess their level of awareness on the disease,and determine predisposing factors of the disease.Methods:A total of 478 GradesⅢ-Ⅴschool-age children in Pamplona and Sanchez-Mira School Districts in the Cagayan Valley answered the questionnaire assessing their knowledge,attitude,and practices on STH,subjected to anthropometric measurements,and provided faecal samples for parasitologic assessment(direct smear,Kato-Katz,and formol-ether concentration techniques).Results:The participants of the study,with 55.86%females,were 8 to 14 years old.Their nutritional status was assessed‘normal’(84.31%),‘severely wasted’(6.49%),‘wasted’(5.23%),‘overweight’(2.72%),and‘obese’(1.26%).The prevalence of infection with at least 1 STH species was 25.99%in Pamplona and 19.40%in SanchezMira.Overall,the prevalence of heavy intensity was 7.11%for Ascaris lumbricoides and 1.67%for Trichuris trichiura.All hookworm infections had light intensities.The majority of the school-age children had a low score in the KAP test.In knowledge of STH,‘stunted growth as a symptom of infection’was associated with a lower risk of Ascaris lumbricoides infection(OR 0.448;95%CI 0.212,0.945;P=0.035)while‘playing with soil as a mode of transmission’was associated with an increased risk of Ascaris lumbricoides infection(OR 2.067;95%CI 1.014,4.212;P=0.046).In attitude towards STH,‘I think I have intestinal worm now’was associated with a higher risk of Ascaris lumbricoides infection(OR 1.681;95%CI 1.061,2.662;P=0.027).Conclusions:The prevalence rate of Ascaris lumbricoides among the school-age children in the Cagayan Valley shows the need to further intensify intervention in the area to meet the threshold set by the World Health Organization.The identified predictors of infection,which concerns the school-age children’s knowledge and attitude toward STH,can be used in augmenting intervention programs in the future. 展开更多

Objective Indirubin,isolated from Indigo Naturals,has been reported to have the inhibitory activity of MCF-7 human breast cancer cells in vitro.However,studies on its anti-breast cancer activity in vivo and the underlying mechanism are insufficient.We explored whether indirubin could trigger ferroptosis of breast cancer cells to exert anti-tumor activity.Methods Bioinformatical analysis was performed to detect the expression of prostaglandin-endoperoxide synthase 2(Ptgs2)in breast cancer tissues and Ptgs2-related prognosis for patients with breast cancer.The inhibitory effects of indirubin on 4T1 cells were assessed using MTT assay and LDH activity detection.The levels of 4-HNE,GPX4,PTGS2 and GSK-3βproteins were detected by Western blotting,and the mRNA of Ptgs2 was tested by qPCR.The contents of GSH and MDA were determined by commercial kits.Molecular docking was employed to study the interaction between indirubin and GSK-3β.A 4T1 murine breast cancer was adopted to evaluate the in vivo antitumor activity of indirubin.Results Indirubin promoted ferroptosis of 4T1 breast cancer cells with depleting of GSH,increased MDA and 4-HNE levels,as well as decreased GPX4 expression.Indirubin suppressed 4T1 breast tumor in vivo.The mechanism study showed indirubin up-regulated Ptgs2 expression by promoting phosphorylation(Ser 9)of GSK-3β.Conclusion Indirubin suppresses 4T1 murine breast cancer in vitro and in vivo by induction of ferroptosis. 展开更多

In order to assess the responses of different skin types stimulated by tape stripping,after which two kinds of brand cosmetics of cream and hydrogel,were separately applied to evaluate the individual soothing effect of the product on different skin types.A total of 30 volunteers aged 18~65 years were recruited,which were divided into four groups(oily,dry,normal and combined)according to the self-oiling ability of facial and T zones.Four 3 cm×3 cm of skins on inner forearm of each volunteer were chosen and randomly marked as blank,negative control,soothing cream and soothing gel.The blank area was kept untreated,and the remaining areas were tape-stripped by 10 times(Transparent universal 1527C-1,3M China Co.,Ltd.,)to disrupt the skin corneum and induce erythema.Images,erythema index(EI)and transcutaneous water loss rate(TEWL)of the marked skin areas before and 5 min after tape-stripping were taken to assess the distinct responses of different skin type to the taping treatment;Ten minutes after tape-stripping,The soothing products were applied to the test area,the skin images,EI and TEWL at 20 min and 50 min(corresponding to 30 min and 60 min after tape-stripping,respectively)were measured after cosmetics application to compare and evaluate the soothing effects of cream and hydrogel on different skin types.Results showed that different skin displayed diverse reactions to the tape-stripping,and the oily skin was more sensitive with longer duration of erythema than other skin types.After the application of soothing products,both cream and hydrogel could relieve skin erythema,while the soothing effects are dependent both on skin types and the cosmetic base materials:Hydrogel has a better efficacy in alleviating erythema and TEWL of dry skin,while the cream was more effective on normal,oily and combined skin types.The results suggest that the modelling method for soothing evaluation should consider the skin types of the volunteers,and the cosmetics of different base materials might have the target skin types. 展开更多

Red alga species in the phylum Rhodophyta are ecologically and economically important,which are widely used as food and medicinal products because they are rich in bioactive compounds.Unfortunately,researchers often meet challenges in identifying red algal species and understanding the evolutionary relationship among them.Shooting away these limitations necessitates detailed genomic studies,including those of cellular organelles.In this study,we sequenced circular organelle genomes of three red macroalgal species,Grateloupia asiatica,Pachymeniopsis lanceolata and Polyopes affinis,yielding two complete chloroplast genomes of P.lanceolata and P.affinis and one mitochondrial genome of P.affinis.The average chloroplast and mitochondrial genome sizes were 192,724 bp and 29,699 bp,respectively,which encoded 202 and 25 pro-teins,respectively,on average.The short-and long-repeat sequences,gene rearrangements,the nucleotide diversity,and phylogenetic relationship among these three species were analyzed.Memo:revise conjunctions and prepositions to form a clearer list(A,B,C,and D).The 194 chloroplast and 23 mitochondrial protein-coding genes shared by the species belonging to Florideophyceae and Bangiophyceae were used to reconstruct a maximum-likelihood phylogenetic tree.In addition,we developed 15 species-specific PCR markers,five for each species,using single-nucleotide polymorphism information.Our results should aid in identifying these species and deciphering the evolutionary relationship among species in the phylum Rhodophyta. 展开更多

Objective To isolate and identify the chemical constituents from mycelia and spores of the fungus Cordyceps cicadae,respectively.Methods The chemical constituents were isolated and purified by repeated silica gel,Sephadex LH-20,and reversed phase HPLC.The structures of these compounds were elucidated on the basis of extensive spectroscopic analysis,including 1 D and 2D NMR.Results Nine known sterols such as ergosterol(1),ergosterol peroxide(2),9,11-dehydroergosterol peroxide(3),3β,5α,9α-trihydroxy-(22E,24R)-ergosta-7,22-dien-6-one(4),3β,5α,9α,14α-tetrahydroxy-(22E,24R)-ergosta-7,22-dien-6-one(5),5 α,6α-e poxy-(22 E,,24 R)-e rgosta-8(14),22-diene-3α,7α-diol (6), 3β,5α,6β-(22E,24R)-ergosta-7,22-dien-3,5,6-triol (7), 3β,5α,6α-6- methoxyergosta-(22E,24R)-7,22-diene-3,5-diol (8), 4-hydroxy-17R-methylincisterol (9), together with a resorcinol derivative, 5-n-nonadecylresorcinol (10), a cydo- desipeptide, beauvericin (11), and a nucleoside, N^6-(2-hydroxyethyl)adenosine (12) were successively isolated from the cultivated C. cicadaemycelia and spores. Conclusion Compounds 3-10 are reported for the first time from the title sample, beauvericin exhibits significant cytotoxicity against human leukemia cell line and human lung cancer cell line. 展开更多

The β-1,3-glucan recognition protein gene from Spodoptera exigua (SeβGRP) was cloned and characterized. The cDNA of this gene is 1?644 nucleotides in length and the predicted polypeptide is 491 amino acids (aa) in length, with a calculated molecular mass of 54.8 kDa. The first 22 aa encode a predicted secretion signal peptide. A BLAST search, multiple sequence alignment, and phylogenetic analysis of the aa sequence of SeβGRP revealed that this protein is most similar to the β-1,3-glucan recognition protein (βGRP) family of pattern recognition proteins. Using reverse-transcription polymerase chain reaction, we detected the presence of SeβGRP transcripts in the egg, larval, pupal, and adult stages of S. exigua. In addition, the SeβGRP transcript was expressed in all the tissues examined including the brain, hemocytes, fat body, intestine, and cuticle. There were no changes in SeβGRP mRNA levels in larvae infected with ultraviolet (UV)-killed Escherichia coli DH5α compared with the control larvae inoculated with the water; however, SeβGRP mRNA levels were markedly elevated 4–8 h after infection and slightly induced 12–24 h after infection in larvae injected with UV-killed Fusarium oxysporum. This may be because β-1,3-glucan is the main component of the cell wall of F. oxysporum, but not E. coli DH5α. 展开更多

In multicellular organisms, controlling the timing and extent of asymmetric cell divisions (ACDs) is crucial for correct patterning. During post-embryonic root development in Arabidopsis thaliana, ground tissue (GT) maturation involves an additional ACD of the endodermis, which generates two different tissues: the endo- dermis (inner) and the middle cortex (outer). It has been reported that the abscisic acid (ABA) and gibberellin (GA) pathways are involved in middle cortex (MC) formation. However, the molecular mechanisms under- lying the interaction between ABA and GA during GT maturation remain largely unknown. Through transcriptome analyses, we identified a previously uncharacterized C2H2-type zinc finger gene, whose expression is regulated by GA and ABA, thus named GAZ (GA- AND ABA-RESPONSIVE ZINC FINGER). Seedlings ectopically overexpressing GAZ (GAZ-OX) were sensitive to ABA and GA during MC formation, whereas GAZ-SRDX and RNAi seedlings displayed opposite phenotypes. In addition, our results indicated that GAZ was involved in the transcriptional regulation of ABA and GA homeostasis. In agreement with pre- vious studies that ABA and GA coordinate to control the timing of MC formation, we also confirmed the unique interplay between ABA and GA and identified factors and regulatory networks bridging the two hor- mone pathways during GT maturation of the Arabidopsis root. 展开更多

13-Lipoxygenases(LOXs)initiate the synthesis of jasmonic acid(JA),the best-understood oxylipin hormone in herbivory defense.However,the roles of 9-LOX-derived oxylipins in insect resistance remain unclear.Here,we repo... 13-Lipoxygenases(LOXs)initiate the synthesis of jasmonic acid(JA),the best-understood oxylipin hormone in herbivory defense.However,the roles of 9-LOX-derived oxylipins in insect resistance remain unclear.Here,we report a novel anti-herbivory mechanism mediated by a tonoplast-localized 9-LOX,ZmLOX5,and its linolenic acid-derived product,9-hydroxy-10-oxo-12(Z),15(Z)-octadecadienoic acid(9,10-KODA).Transposon-insertional disruption of ZmLOX5 resulted in the loss of resistance to insect herbivory.lox5 knockout mutants displayed greatly reduced wound-induced accumulation of multiple oxylipins and defense metabolites,including benzoxazinoids,abscisic acid(ABA),and JA-isoleucine(JA-Ile).However,exogenous JA-Ile failed to rescue insect defense in lox5 mutants,while applications of 1 mM 9,10-KODA or the JA precursor,12-oxo-phytodienoic acid(12-OPDA),restored wild-type resistance levels.Metabolite profiling revealed that exogenous 9,10-KODA primed the plants for increased production of ABA and 12-OPDA,but not JA-Ile.While none of the 9-oxylipins were able to rescue JA-Ile induction,the lox5 mutant accumulated lower wound-induced levels of Ca^(2+),suggesting this as a potential explanation for lower wound-induced JA.Seedlings pretreated with 9,10-KODA exhibited rapid or more robust woundinduced defense gene expression.In addition,an artificial diet supplemented with 9,10-KODA arrested fall armyworm larvae growth.Finally,analysis of single and double lox5 and lox10 mutants showed that ZmLOX5 also contributed to insect defense by modulating ZmLOX10-mediated green leaf volatile signaling.Collectively,our study uncovered a previously unknown anti-herbivore defense and hormonelike signaling activity for a major 9-oxylipin α-ketol. 展开更多
