AIM To evaluate the sex-specific effects of a hydroalcoholic extract from Eugenia punicifolia(HEEP)leaves on gastric ulcer healing.METHODS In this rat study involving males,intact(cycling)females,and ovariectomized females,gastric ulcers were induced using acetic acid.A vehicle,lansoprazole,or HEEP was administered for 14 d after ulcer induction.Body weight was monitored throughout the treatment period.At the end of treatment,the rats were euthanized and the following in vivo and in vitro investigations were performed:macroscopic examination of the lesion area and organ weights,biochemical analysis,zymography,and evaluation of protein expression levels.Additionally,the concentration-dependent effect of HEEP was evaluated in terms of subacute toxicity and cytotoxicity.RESULTS Compared to the vehicle,HEEP demonstrated a great healing capacity by substantially reducing the ulcerative lesion area in males(52.44%),intact females(85.22%),and ovariectomized females(65.47%),confirming that HEEP accelerates the healing of acetic acidinduced gastric lesions and suggesting that this effect is modulated by female sex hormones.The antiulcer effect of HEEP was mediated by prostaglandin E2 only in male rats.Overall,the beneficial effect of HEEP was the highest in intact females.Notably,HEEP promoted the expression of vascular endothelial growth factor(intact vs ovariectomized females)and decreased the expression of Caspase-8 and Bcl-2(intact female vs male or ovariectomized female).Additionally,HEEP enhanced fibroblast proliferation and migration into a wounded area in vitro,confirming its healing effect.Finally,no sign of subacute toxicity or cytotoxicity of HEEP was observed.CONCLUSION In gastric ulcers,HEEP-induced healing(modulated by female sex hormones;in males,mediated by prostaglandin)involves extracellular matrix remodeling,with gastric mucosa cell proliferation and migration. 展开更多

Radical Scavenging and Antioxidant Potential of Nuts and Leaves Extracts of <i>Semecarpus anacardium</i>(L.)

In the present investigation, antioxidant activity of the crude extracts from nuts and leaves of S. anacardium was carried out. The antioxidant potential of various extracts (ethanol, acetone and aqueous) of S. anacardium was determined by using DPPH assay, ABTS assay and metal chelating activity assay. Among the extracts the ethanol extract of nut showed significant scavenging activity (DPPH-88.73 ± 2.26;ABTS assay-81.65 ± 1.57 and metal chelating activity-72.37 ± 2.26) compared with antioxidant controls, ascorbic acid and BHA respectively. The findings of the present investigation suggest that the S. anacardium extracts can prove to be a potent source of biologically active compounds that can be further subjected to isolation of therapeutic antioxidant agents. 展开更多

Osmoporation is a novel encapsulation approach for bioactive compounds based on the osmoresistance mechanisms of microbial cells.To the best of our knowledge,this is the first study investigating the production of fisetin-enriched yoghurt using Lactobacillus acidophilus-based bio-capsules via osmoporation as the starter culture.Results showed that the milk acidification with fisetin-loaded L.acidophilus progressed at a slower pace due to complex mechanisms induced by osmopo-ration and internalized fisetin.Milk fermentation using fisetin bio-capsules reached a maximum acidification rate of 0.18 pH units/h after 23 h and pH 4.6 was achieved after 32 h.Besides,the antioxidant activity of yoghurts produced with fisetin bio-capsules did not change during cold storage,while the antioxidant activity of yoghurt produced with non-encapsulated fisetin was reduced by 2.5-fold after 28 days.Overall,this study shows that fisetin osmoporation using L.acidophilus is a versatile encapsulation bioprocess that enables the delivery of preserved phytoactives into fermented foods like yoghurt.This strategy has the potential to be extended to other applications in the dairy industry using lactic acid bacteria as both the encapsulation matrix and fermentation agent. 展开更多
