Gross signs...Studies were conducted to determine the cause of the acute mortality of half-smooth tongue sole Cynoglossus semilaevis Günther juveniles in a fish farm in Jimo, Shandong Province, China, in June 2006. Gross signs of the diseased tongue sole included several petechiae and ecchymoses on the body and fin necrosis and hemorrhagic lesion at the base of the fin. Bacteria were isolated from kidney, liver and hemorrhagic lesions of the diseased tongue sole. Among 14 strains, SJ060621 was proved to be highly virulent to juvenile tongue sole with LD50 value of 〈1.0×10^5 colony forming units (CFU)mL^-1, while the remaining 13 were avirulent. Among the 16 antibiotics tested, SJ060621 was sensitive to gentamicin and nitrofurantoin. It was identified as Listonella anguillantm with conventional plate and tube tests in combination with API 20E analysis. 16S rRNA gene and partial HSP60 gene sequenceing analysis revealed that the strain was highly homologous with L. anguillarum. Examination of the infected musculature by electron microscopy indicated numerous bacteria and lots of macrophages containing phagocytosed bacteria. Histopathological investigations revealed severe necrotic degenerative changes in the infected organs. Indirect immunofluorescence assay (IFA) was employed to detect the location of occurrence of bacteria, and bacteria were found in aggregations in the inflammatory areas in musculature.展开更多
An effort was made to couple FVCOM (a three-dimensional (3D),unstructured grid,Finite Volume Coastal Ocean Model) and FVCOM-SWAVE (an unstructured grid,finite-volume surface wave model) for the study of nearshore ocea...An effort was made to couple FVCOM (a three-dimensional (3D),unstructured grid,Finite Volume Coastal Ocean Model) and FVCOM-SWAVE (an unstructured grid,finite-volume surface wave model) for the study of nearshore ocean processes such as tides,circulation,storm surge,waves,sediment transport,and morphological evolution.The coupling between FVCOM and FVCOM-SWAVE was achieved through incorporating 3D radiation stress,wave-current-sediment-related bottom boundary layer,sea surface stress parameterizations,and morphology process.FVCOM also includes a 3D sediment transport module.With accurate fitting of irregular coastlines,the model provides a unique tool to study sediment dynamics in coastal ocean,estuaries,and wetlands where local geometries are characterized by inlets,islands,and intertidal marsh zones.The model was validated by two standard benchmark tests: 1) spectral waves approaching a mild sloping beach and 2) morphological changes of seabed in an idealized tidal inlet.In Test 1,model results were compared with both analytical solutions and laboratory experiments.A further comparison was also made with the structured grid Regional Ocean Model System (ROMS),which provides an insight into the performance of the two models with the same open boundary forcing.展开更多

The visible fossils are preserved in thinlaminated siltstone or muddy siltstone intercalated between 8-15 ram-thick carbonate deposits, probably representing sedimentary settings of a constrained local depression in the shallow water carbonate platform during the Early Cambrian Meishucunian Stage. However, controversial results have been reported regarding the exact role of 20-HETE on blood pressure. In the present study, we used recombinant adenoassociated viral vector (rAAV) to deliver CYP 4A1 cDNA and antisense 4A1 cDNA into Sprague-Dawley (SD) rats and spontaneously hypertensive rats (SHR), respectively, to investigate the effects of long-term modifications of blood pressure and the potential for gene therapy of hyperténsion. The mean systolic pressure increased by 14.2±2.5 mm Hg in rAAV.4A 1-treated SD rats and decreased by 13.7±2.2 mm Hg in rAAV.anti4A l-treated SHR rats 5 weeks after the injection compared with controls and these changes in blood pressure were maintained until the experiments ended at 24 weeks. In 4A1 treated animals CYP4A was overexpressed in various tissues, but preferentially in the kidney at both mRNA and protein levels. In anti-4Al-treated SHR, CYP4A mRNA in various tissues was probed, especially in kidneys, but 4A l protein expression was almost completely inhibited. These results suggest that arachidonic acid CYP hydroxylases contribute not only to the maintenance of normal blood pressure but also to the development of hypertension. rAAV-mediated anti4A administration strategy has the potential to be used as targeted gene therapy in human hypertension by blocking expression of CYP 4A in kidneys.展开更多

(JCL) seeds were extracted and transesterified in-situ using supercritical methanol extraction in the absence of catalyst at different temperatures (200-280℃) and pressures (8-12 MPa), and at...Jatropha curcas L. (JCL) seeds were extracted and transesterified in-situ using supercritical methanol extraction in the absence of catalyst at different temperatures (200-280℃) and pressures (8-12 MPa), and at a fixed reaction time of 30 min with seeds-to-methanol ratio of 1:40 w/v. Design of experiment approach using five-level-two-factors design of Response Surface Methodology (RSM) was used to observe the effect of two independent variables i.e. temperature and pressure and the percent of biodiesel yield which required 13 runs. For optimization of the variables, Central Composite Rotatable Design (CCRD) was used for regression analysis and analysis of variance (ANOVA). The optimize conditions suggested by RSM were at T = 280℃ and P = 12.04 MPa. The predicted and experimental biodicsel yields were found to be 56.8% and 59.9%, respectively, with relatively small deviation errors of 1.59%.展开更多
