Preparation of diacylglycerols from Acer truncatum seed oil via enzymatic glycerolysis and evaluation of their neuroprotective effects

Acer truncatum seed oil(ATO),rich in nervonic acid,was enzymatically converted to diacylglycerols(DAG)via optimized glycerolysis(1:1 ATO:glycerol ratio,2%Novozym 435,60℃,24 h),achieving a>50%DAG yield.The process demonstrated scalability(50-fold)and enzyme reusability(6 cycles),yielding 50%DAG content in scaled batches.Purification through molecular distillation and silica gel chromatography produced high-purity DAG(98.7%)while preserving fatty acid composition.In H_(2)O_(2)-induced PC12 cells,ATO-derived DAG(40μg mL^(-1))enhanced cell viability by 32%(vs.H_(2)O_(2)controls),reduced reactive oxygen species(ROS)by 49%,lowered malondialdehyde(MDA)levels to 20.8μM mg^(-1)protein,and restored the activities of antioxidant enzymes,including superoxide dismutase(SOD)and glutathione peroxidase(GPx).These results establish enzymatic glycerolysis as a scalable method for neuroprotective DAG production and position ATO-derived DAG as a promising therapeutic against oxidative neurodegeneration. 展开更多

Jujube is one of the most produced dried fruits in China,and it is also a traditional Chinese medicine that enhances immunity and has anti-cancer activity.Jujube fruit is rich in phenolic compounds,but few reports are available on its biological activities.To evaluate the biological activities of the phenolic compounds in dried jujube fruit,the composition and content of metabolites in dried jujube fruit were determined by metabolomics,and the antibacterial and anticancer activities of the phenolic compounds were analyzed in dried jujube fruit.The results showed that 463 compounds were identified in dried‘Junzao’fruit,including 102 phenolic compounds.The in vitro activity test showed that the jujube phenolic metabolites had extensive antibacterial effects and caused disruption and nuclear sclerosis of hepatocellular carcinoma(HepG2)cells.Canonical correlation analysis showed that total phenolic content,quercetin-3-rutinose,and procyanidin B1 were the main active antibacterial and anticancer components.The study provides data supporting the application of dried jujube fruit in the development of functional foods,pharmaceuticals and cosmetics. 展开更多
