Unveiling the role of VcCOMT38 as a specific O-methyltransferase for enhancing lignin biosynthesis:insights from blueberry and cross-species analysis

Lignin is a significant secondary metabolite produced through the phenylpropanoid pathway.As a vital component of the plant cell wall,lignin affects various fruit characteristics,including size,seed quantity,and firmness.In this study,we conducted comprehensive identification and phylogenetic analysis of 265 Caffeic acid O-methyltransferase(COMT)genes across ten different plant species,including Vaccinium corymbosum and four other Vaccinium species.The results reveal that VcCOMT38 is a promising structural gene for the biosynthesis of lignin in blueberry.An in vitro enzymatic assay of VcCOMT38 demonstrated that it is a special enzyme in the lignin biosynthesis pathway and prefers to use caffeic acid as a substrate over 5-hydroxyferulic acid.Transient overexpression and silencing of VcCOMT38 in Vaccinium corymbosum‘Northland’fruits demonstrated that VcCOMT38 participates in lignin biosynthesis and contributes to both an increased number of immature seeds and enhanced fruit firmness.The heterologous overexpression of VcCOMT38 in Nicotiana benthamiana revealed that this gene could increase the lignin content and the syringyl/guaiacyl(S/G)ratio,which determines the maximum monomer yield during lignin depolymerization.These results highlight VcCOMT38 as a crucial gene in lignin biosynthesis and its potential for improving lignin production in industry through genetically modified woody plants. 展开更多

Longitudinal study of peer bullying victimization and its psychological effects on adolescents

BACKGROUND Peer bullying victimization(PBV)is a significant public health issue that adolescents often face,with approximately one-third having experienced PBV.Understanding its interrelationships with mental health problems is crucial for effective intervention.This study aims to examine the longitudinal relationships between internalizing problems(depression and anxiety),externalizing problems(aggression),and PBV among middle school students using cross-lagged panel network analysis.AIM To examine the longitudinal relationships between internalizing problems(depression and anxiety),externalizing problems(aggression),and PBV among middle school students using cross-lagged panel network analysis.METHODS A total of 1260 middle school students(54.6%male)participated in this study.Data were collected at two time points(time 1 and time 2)using self-report questionnaires to assess PBV,depression,anxiety,and aggression.Cross-lagged panel network analysis was applied to examine the bi-directional relationships between these variables over time.RESULTS Depression,particularly a“sad mood,”was the most significant predictor of both PBV and aggression.Cyberbullying victimization also emerged as a key factor influencing depression and anxiety.While anxiety had weaker and less consistent effects on PBV,aggression was predominantly influenced by depressive symptoms and sleep disturbances.The analysis also identified key symptoms,such as a“sad mood”and sleep disturbances,as crucial targets for intervention to mitigate the cycle of PBV,depression,anxiety,and aggression.CONCLUSION This study provides important insights for bullying victimization prevention and intervention strategies:(1)Early identification and intervention targeting depression,particularly a“sad mood,”and sleep disturbances;(2)The importance of addressing cyberbullying as a distinct factor influencing mental health;and(3)The need for comprehensive,longitudinal approaches to understanding and intervening in the interconnected mental health issues among adolescents. 展开更多

Background:Hemorrhagic expansion into the fourth ventricle is an independent risk factor for poor outcomes in intraventricular hemorrhage(IVH)patients.However,to date,available animal models of IVH are limited to models of supratentorial ventricular hemorrhage,and there are no specific models of fourth ventricle hemorrhage.This limitation hinders comprehensive basic research and the understanding of the pathophysiological changes that occur following fourth ventricle hemorrhage.Therefore,the development of an animal model of fourth ventricle hemorrhage is highly important.Methods:In this study,a novel rat model of fourth ventricle hemorrhage was established via autologous blood injection through the foramen of Magendie.Anesthetized rats were positioned in a stereotaxic apparatus with their heads tilted downward at an angle of approximately 20°relative to the vertical axis.A needle was inserted through the foramen,and autologous blood obtained from the rat's heart was injected into the fourth ventricle via a microinfusion pump.Systematic evaluations of the model were conducted using small-animal magnetic resonance imaging,histopathological analysis,and neurological function assessment.Results:The rats developed stable and reproducible fourth ventricle hematomas and ventricular dilation.They also exhibited acute-phase hydrocephalus and pathological features of perilesional brain tissue injury,with observed neurological deficits comparable to patients with fourth ventricle hemorrhage.Conclusion:This model successfully recapitulates the clinicopathological and pathophysiological characteristics of patients with fourth ventricle hemorrhage and can be utilized for further investigation into the pathophysiological mechanisms underlying posthemorrhagic hydrocephalus and perilesional brainstem tissue injury. 展开更多

Electron-positron colliders operating in the GeV center-of-mass range,or tau-charm energy region,have been proved to enable competitive frontier research due to several unique features.With the progress of high-energy physics in the last two decades,a new-generation Tau-Charm factory,called the Super Tau-Charm Facility(STCF),has been actively promoted by the particle physics community in China.STCF has the potential to address fundamental questions such as the essence of color confinement and the matter-antimatter asymmetry within the next decades.The main design goals of the STCF are a center-of-mass energy ranging from 2 to 7 GeV and a luminosity surpassing 5×10^(34)cm^(−2)s^(−1)that is optimized at a center-of-mass energy of 4 GeV,which is approximately 50 times that of the currently operating Tau-Charm factory-BEPCII.The STCF accelerator has two main parts:a double-ring collider with a crab-waist collision scheme and an injector that provides top-up injections for both electron and positron beams.As a typical third-generation electron-positron circular collider,the STCF accelerator faces many challenges in both accelerator physics and technology.In this paper,the conceptual design of the STCF accelerator complex is presented,including the ongoing efforts and plans for technological research and develop-ment,as well as the required infrastructure.The STCF project aims to secure support from the Chinese central government for its construction during the 15th Five-Year Plan(2026-2030). 展开更多

[Objectives] This study was conducted on tissue-culture rapid propagation techniques of Spathiphyllum kochii Engl. et Krause. [Methods] With lateral buds of S. kochii as explants, the effects of such four basic media as MS, B5, Nitsch and Wpm and the ratio of two hormones(1, 2, 3 mg/L 6-BA and 0.1, 0.3, 0.5 mg/L NAA) on bud proliferation of S. kochii were studied by the complete test method, and the effect of the ratio of the two hormones(0.25, 0.50, 1.00 mg/L NAA and 1.0, 1.5 mg/L IBA) on rooting of S. kochii as well as the effect of substrate ratio(perlite and peat soil at 1∶9, 2∶8, 3∶7, 4∶6 and 5∶5) on its transplanting survival rate were also studied. [Results] The best basic medium for the rapid propagation of S. kochii was MS. The best hormone ratio suitable for bud proliferation was 2 mg/L6-BA+0.1 mg/L NAA, and the average number of buds proliferated at 45 d was 3.04. The average bud height was 2.05 cm; the most suitable medium for rooting was 1/2 MS+1 mg/L IBA+0.25 mg/L NAA, and its rooting rate was 100%; and the best transplanting substrate was 5∶5 perlite and peat. The soil ratio had a survival rate of 94%. [Conclusions] This study provides a theoretical basis for the improvement of the transplanting survival rate of test-tube S. kochii plantlets. 展开更多

BACKGROUND Various non-steroidal anti-inflammatory drugs(NSAIDs)have been used for juvenile idiopathic arthritis(JIA).However,the optimal method for JIA has not yet been developed.AIM To perform a systematic review and network meta-analysis to determine the optimal instructions.METHODS We searched for randomized controlled trials(RCTs)from PubMed,EMBASE,Google Scholar,CNKI,and Wanfang without restriction for publication date or language at August,2023.Any RCTs that comparing the effectiveness of NSAIDs with each other or placebo for JIA were included in this network meta-analysis.The surface under the cumulative ranking curve(SUCRA)analysis was used to rank the treatments.P value less than 0.05 was identified as statistically significant.RESULTS We included 8 RCTs(1127 patients)comparing 8 different instructions including meloxicam(0.125 qd and 0.250 qd),Celecoxib(3 mg/kg bid and 6 mg/kg bid),piroxicam,Naproxen(5.0 mg/kg/d,7.5 mg/kg/d and 12.5 mg/kg/d),inuprofen(30-40 mg/kg/d),Aspirin(60-80 mg/kg/d,75 mg/kg/d,and 55 mg/kg/d),Tolmetin(15 mg/kg/d),Rofecoxib,and placebo.There were no significant differences between any two NSAIDs regarding ACR Pedi 30 response.The SUCRA shows that celecoxib(6 mg/kg bid)ranked first(SUCRA,88.9%),rofecoxib ranked second(SUCRA,68.1%),Celecoxib(3 mg/kg bid)ranked third(SUCRA,51.0%).There were no significant differences between any two NSAIDs regarding adverse events.The SUCRA shows that placebo ranked first(SUCRA,88.2%),piroxicam ranked second(SUCRA,60.5%),rofecoxib(0.6 mg/kg qd)ranked third(SUCRA,56.1%),meloxicam(0.125 mg/kg qd)ranked fourth(SUCRA,56.1%),and rofecoxib(0.3 mg/kg qd)ranked fifth(SUCRA,56.1%).CONCLUSION In summary,celecoxib(6 mg/kg bid)was found to be the most effective NSAID for treating JIA.Rofecoxib,piroxicam,and meloxicam may be safer options,but further research is needed to confirm these findings in larger trials with higher quality studies. 展开更多

As efficient water treatment agents, a novel series of rectorite-based ZnO and TiO2 hybrid composites(REC/ZnO/TiO2) were synthesized and characterized in this study. Effects of experimental parameters including TiO2 mass ratio, solution p H and catalyst dosage on the removal of methyl blue(MB) were also conducted. The presence of a little mass ratio(2%-6%) of TiO2 highly promoted the photoactivity of REC/ZnO/TiO2 in removal of MB dye from aqueous solution, in which ZnO and REC played a role of photocatalyst and adsorbent. The promotion effects of TiO2 may result from the accelerated separation of electron-hole on ZnO. The observed kinetic constant for the degradation of MB over REC/ZnO and REC/ZnO/TiO2 were 0.015 and 0.038 min^(-1), respectively. The degradation kinetics of MB dye, which followed the Langmuir–Hinshelwood model, had a reaction constant of 0.17 mg/(L min). The decrease of removal ratio of MB after five repetitive experiments was small, indicating REC/ZnO/TiO2 has great potential as an effective and stable catalyst. 展开更多

The crystal belongs to the monoclinic system, space group P21/n with a = 8.736(2), b = 14.850(3), c = 11.775(2)A, β = 102.42(3)°, V = 1491.8(5) A^3, Z = 2, Dc = 1.593 g/cm^3, Mr = 715.36, λ(MoKα) = 0.71073A , μ = 1.669 mm^-1, F(000) = 736, the final R = 0.0338 and wR = 0.0789. A total of 3411 unique reflections were collected, of which 2777 with I 〉 2σ(I) were observed. The molecule of the complex is located on a crystallographic inversion centre. Each Zn atom in the complex is five-coordinated by one Schiff base ligand and two bridging azide groups, forming a trigonal bipyramidal configuration. The Zn···Zn distance is 3.328(2)A . In the crystal structure, molecules are linked through intermolecular C–H…N hydrogen bonds, forming layers parallel to the bc plane. The complex shows low cytotoxic property to both normal and carcinoma cells. 展开更多

Background Fatty liver disease causes huge economic losses in the poultry industry due to its high occurrence and lethality rate.Three-dimensional(3D)chromatin architecture takes part in disease processing by regulating tran-scriptional reprogramming.The study is carried out to investigate the alterations of hepatic 3D genome and H3K27ac profiling in early fatty liver(FLS)and reveal their effect on hepatic transcriptional reprogramming in laying hens.Results Results show that FLS model is constructed with obvious phenotypes including hepatic visible lipid deposi-tion as well as higher total triglyceride and cholesterol in serum.A/B compartment switching,topologically associat-ing domain(TAD)and chromatin loop changes are identified by high-throughput/resolution chromosome conforma-tion capture(HiC)technology.Targeted genes of these alternations in hepatic 3D genome organization significantly enrich pathways related to lipid metabolism and hepatic damage.H3K27ac differential peaks and differential expres-sion genes(DEGs)identified through RNA-seq analysis are also enriched in these pathways.Notably,certain DEGs are found to correspond with changes in 3D chromatin structure and H3K27ac binding in their promoters.DNA motif analysis reveals that candidate transcription factors are implicated in regulating transcriptional reprogram-ming.Furthermore,disturbed folate metabolism is observed,as evidenced by lower folate levels and altered enzyme expression.Conclusion Our findings establish a link between transcriptional reprogramming changes and 3D chromatin struc-ture variations during early FLS formation,which provides candidate transcription factors and folate as targets for FLS prevention or treatment. 展开更多

In the structure of 1, each [Ru Ⅱ(ttpy)(CN)3]-connects two [MnⅢ(salen)]+ with two cyanide groups in a cis-conformation, and in turn each[MnⅢ(salen)]+ unit links two [RuⅡ(ttpy)(CN)3]-building blocks in a trans-conformation, resulting in a 1D [-NC-RuⅡ-CN-MnⅢ-]n chain. BACKGROUND Previous studies have suggested that a low albumin-to-alkaline phosphatase ratio(AAPR)is associated with a lower survival rate in patients with various malignancies.However,the relationship between pretreatment AAPR and the prognosis of patients with gastric cancer(GC)remains unclear.AIM To investigate the prognostic value of AAPR in distant metastatic GC.METHODS A total of 191 patients with distant metastatic cancer from a single institute were enrolled in this study.Pretreatment clinical data,including serum albumin and alkaline phosphatase levels,were collected.A chi-square test or Fisher’s exact test was applied to evaluate the correlations between AAPR and various clinical parameters in GC patients.The Kaplan-Meier method and Cox proportional hazards regression model were used to evaluate the prognostic efficacy of AAPR in metastatic GC patients.A two-sided P value lower than 0.05 was considered statistically significant.RESULTS A receiver operating characteristic curve indicated that 0.48 was the optimal threshold value for AAPR.AAPR≤0.48 was significantly associated with bone(P<0.05)and liver metastasis(P<0.05).Patients with high levels of AAPR had better survival in terms of overall survival(OS)and progression-free survival(PFS),regardless of the presence of liver/bone metastasis.Pretreatment AAPR was found to be a favorable predictor of OS and PFS based on a multivariate cox regression model.AAPR-M system,constructed based on AAPR and number of metastatic sites,showed superior predictive ability relative to the number of metastatic sites for predicting survival.CONCLUSION Pretreatment AAPR may serve as an independent prognostic factor for predicting PFS and OS in patients with metastatic GC.Furthermore,AAPR may assist clinicians with individualizing treatment. 展开更多

The present study examined the protective effect of the ethanol extract of Sarcopyramis nepalensis (EESN) on agents-induced hepatotoxicity in mice and the possible mechanism. Acute liver injury was induced by administration of either CCl4 or D-GalN. The animals were divided into 5 groups in terms of different treatment: normal group, CCl4 or D-GalN group, silymarin or bifendate group, low dose EESN group (10 mg/kg) and high dose EESN group (30 mg/kg). Liver function was evaluated by detecting the levels of serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST). The oxidize stress markers were measured, including malondialdehyde (MDA), glutathione peroxidase (GSH) and superoxide dismutase (SOD). Liver tissues were histopathologically examined by hematoxy-lin-eosin (H&E) staining. The acute toxicity study revealed that there was no toxicity of EESN at the dose of 5 g/kg in mice. The levels of ALT and AST in serum, and the MDA level in live tissues were significantly increased and the activities of SOD and GSH substantially decreased in mice after CCl4 or D-GalN treatment. These biochemical and oxidize stress markers were profoundly improved after treatment with EESN at different doses, which was similar to the results of silymarin or bifendate treatment. The histophathological examination revealed the significant improvement in the pathological changes of the liver in EESN-treated mice as compared to those in CCl4 or D-GalN group. It was concluded that EESN possesses potential antioxidant and hepatoprotective properties and has therapeutic potential for liver diseases. 展开更多

With the development of new synthesis methods and chemistries,a number of new superhard materials have been reported to be harder than diamond.While such materials are highly desirable due to their wide-ranging applications,there are some inherent uncertainties in the methods utilized to determine and define the hardness of such materials.In this paper,we employed the standard Vickers diamond indenter and substitute indenters with the same shape to measure the hardness of nine ceramics and superhard materials within well-defined criteria and methodology,for the assessment of consistency in the hardness testing.The findings and the developed testing method in the current study have broad implications in characterizing new and emerging superhard materials,leading to new discoveries. 展开更多

The Chinese tree shrew(Tupaia belangeri chinensis)has emerged as a promising model for investigating adrenal steroid synthesis,but it is unclear whether the same cells produce steroid hormones and whether their production is regulated in the same way as in humans.Here,we comprehensively mapped the cell types and pathways of steroid metabolism in the adrenal gland of Chinese tree shrews using single-cell RNA sequencing,spatial transcriptome analysis,mass spectrometry,and immunohistochemistry.We compared the transcriptomes of various adrenal cell types across tree shrews,humans,macaques,and mice.Results showed that tree shrew adrenal glands expressed many of the same key enzymes for steroid synthesis as humans,including CYP11B2,CYP11B1,CYB5A,and CHGA.Biochemical analysis confirmed the production of aldosterone,cortisol,and dehydroepiandrosterone but not dehydroepiandrosterone sulfate in the tree shrew adrenal glands.Furthermore,genes in adrenal cell types in tree shrews were correlated with genetic risk factors for polycystic ovary syndrome,primary aldosteronism,hypertension,and related disorders in humans based on genome-wide association studies.Overall,this study suggests that the adrenal glands of Chinese tree shrews may consist of closely related cell populations with functional similarity to those of the human adrenal gland.Our comprehensive results(publicly available at http://gxmujyzmolab.cn:16245/scAGMap/)should facilitate the advancement of this animal model for the investigation of adrenal gland disorders. 展开更多

Recent experimental advancements reported a chemical reaction between antimony and nitrogen under high temperature and high pressure,yielding crystalline antimony nitride(Sb_(3)N_(5))with an orthorhombic structure.Motivated by this statement,we calculate the stability,elastic properties,electronic properties and energy density of the Cmc2_(1) structure for pnictogen nitrides X_(3)N_(5)(X=P,As,Sb,and Bi)using first-principles calculations combined with particle swarm optimization algorithms.Calculations of formation enthalpies,elastic constants and phonon spectra show that P_(3)N_(5),As_(3)N_(5) and Sb_(3)N_(5) are thermodynamically,mechanically and kinetically stable at 35 GPa,whereas Bi_(3)N_(5) is mechanically and kinetically stable but thermodynamically unstable.The computed electronic density of states shows strong covalent bonding between the N atoms and the phosphorus group atoms in the four compounds,confirmed by the calculated electronic localization function.We also calculate the energy densities for Sb_(3)N_(5) and find it to be a potentially high-energy-density material. 展开更多
