Objective:To investigate the anticancer effects of Xi Huang Capsule(XH)in vivo,thirty-two Nu/Nu mice inoculated with human breast cancer SKBR-3 cells and twenty-four Nu/Nu mice inoculated with murine breast cancer 4T1...Objective:To investigate the anticancer effects of Xi Huang Capsule(XH)in vivo,thirty-two Nu/Nu mice inoculated with human breast cancer SKBR-3 cells and twenty-four Nu/Nu mice inoculated with murine breast cancer 4T1 cells were randomized into the control group,XH group,5-FU group and combination of XH and 5-FU group in a 1:1:1:1 ratio.Method:The 5-FU group was injected with 5-FU at 30 mg/kg intraperitoneally every third day;XH group received doses of 0.25g/kg of XH by gastric perfusion each day;The control group was injected daily with normal saline(N.S.)intraperitoneally and the combination group was treated with 5-FU and XH on the same schedules as above.All treatments lasted for 15 days in human SKBR-3 breast cancer cells and 11 days in mice breast cancer 4T1 cells.Tumor volume,tumor weight,organ index,and change in body weight of nude mice were measured,respectively.PCNA and vimentin protein expression were examined by immunohistochemical assay.Results:In SKBR-3 cell xenograft tumor experiments,the XH group,5-FU group and the combination group had significantly smaller tumor volumes(966.39±80.23mm3,892.21±150.77mm3,817.93±162.47 mm3,respectively),and lower tumor weights(0.90±0.14g,0.84±0.32,0.86±0.24g,respectively),as compared with the control group(all P<0.05).The combination group had the highest tumor inhibition rate(38.7%).The similar results emerged in 4T1 cell xenograft tumor.Only the combination group had the least body weight increase of SKBR-3 cells xenograft tumor(P<0.05 as compared with the control group).In SKBR-3 cell xenograft tumor experiments,the 5-FU group had a lower Liver index(43.02±5.00mg/g versus 50.95±4.59mg/g)as compared with the control group(P<0.05),whereas the combination group reversed the changes in the 5-FU group with Liver index of 49.69±4.81 mg/g(P<0.05).The combination group had a higher Spleen index(5.95±1.62 versus 4.72±0.66mg/g)as compared with the control group,and had a higher Spleen index as compared with the 5-FU group(4.54±0.79 mg/g,P<0.05).In 4T1 cell xenograft tumor experiments,the 5-FU group and the combination group had a lower Liver index(47.69±6.41,49.87±5.96 versus 58.95±7.33),but Liver index of XH group had no significantly difference as compared with the control group.The Spleen index was the same to that in SKBR-3 cells xenograft tumor.PCNA and vimentin expression of XH group was significantly lower than that of the control group.Conclusion:The treatment of XH was equally effective in the inhibition of tumor growth,and may have potentially additional benefit in improving the general condition and immunity of the mice not only in human breast cancer cell but also in rat mammary carcinoma in vivo.展开更多
Background:High altitude disease(HAD)can reduce combat effectiveness and damage the health of soldiers at high altitudes.The objective of this hypothesis study is to build a four-period prevention model for high altit...Background:High altitude disease(HAD)can reduce combat effectiveness and damage the health of soldiers at high altitudes.The objective of this hypothesis study is to build a four-period prevention model for high altitude disease that can be applied at high altitudes of over 3000 m.Presentation of the hypothesis:We divided the time at high altitude into nine periods,with three stages from the ascent preparation to the descent to the plain,and applied a continuous dynamic and systematic four-period prevention model across the nine periods.Each period of three stages has its own different measures and targets high altitude health care services for the prevention of high altitude disease.A standard four-period prevention model for high altitude disease was constructed for the high altitude health services at the population level.Testing the hypothesis:Our hypothesized HAD prevention model represents a continuous dynamic and systematic four-period prevention model across the nine periods.This hypothesis can be tested from three aspects.The first one is assessment of soldiers’operating efficacies.The second is comparison of the long-term high altitude population health basic data and development and utilization of big data.The third is descent population health status comparative study and historical retrospective study on prevention.Implications:As we know,it is necessary to protect soldiers’health through the ascent and descent.Through the standard four-period model,we can protect soldiers’health by preventing high altitude diseases,screening the susceptible population,securely tracking their location and maintaining soldiers’health statuses;we also maintain their operational capabilities,eliminate their psychological fears and ease their family troubles.展开更多
Objective:Though Chinese medicine(CM)has showed its clinical efficacy for the treatment of obstructive sleep apnea(OSA)in China,no systematic reviews or meta-analyses provide evidences for its therapeutic effects on O...Objective:Though Chinese medicine(CM)has showed its clinical efficacy for the treatment of obstructive sleep apnea(OSA)in China,no systematic reviews or meta-analyses provide evidences for its therapeutic effects on OSA and the long-term safety.The aim of the present study is to evaluate the effectiveness and safety of CM on OSA using meta-analysis.Methods:We used search items of"Chinese Medicine"AND"obstructive sleep apnea"to retrieve the randomized control trials(RCTs)of CM treatments for OSA in PubMed,Embase,Cochrane Library,China National Knowledge Infrastructure(CNKI),VIP database,and Wanfang database from their respective inception dates to December 2019.Only RCTs of CM therapy versus no treatment which could be quantitatively synthesized were included.Finally,20 studies representing 1,297 participants were included after extraction.Two investigators independently extracted and analyzed the data using RevMan5.3 software.Results:The treatment group using CM decoctions or CM granules presented significantly ameliorative effects on apnoea-hypopnoea index(AHI)compared with the control group(MD:-2.58,95%CI:-3.59 to-1.56,P<0.00001 and MD:-5.47,95%CI:-6.75 to-4.19,P<0.00001,respectively)in the sensitivity analysis.However,there were non significant differences in the duration of treatment between subgroups,indicating that the duration of treatment has no impacts on the therapeutic effects on AHI.CM granules also showed significantly ameliorative effects on the lowest nocturnal oxygen saturation(LSaO2)(MD:2.76,95%CI:1.85 to 3.68,P<0.00001).CM decoctions exhibited significantly improved Epworth Sleepiness Scale(ESS)scores compared with the control group in a sensitivity analysis(MD:-1.50,95%CI:-2.13 to-0.88,P<0.00001).CM granules showed a better improvement of ESS than the control group(MD:-1.35,95%CI:-1.92 to-0.78,P<0.00001).Mild adverse reactions occurred only in five patients and disappeared without special treatment.Conclusion:This study showed favorable therapeutic efficacy of CM on OSA.However,in consideration of the low methodological quality of the included RCTs,more rigorous designed,large sample size RCTs are recommended for providing more high-quality evidences.展开更多

The results of our mathematical analysis indicate that the basic reproduction number plays an important role in studying the dynamics of the system. If the basic reproduction number is less than unity, it is shown that the disease-free equilibrium is globally asymptotically stable by comparison arguments. If it is greater than unity, the system is permanent and there is a unique endemic equilibrium. In this case, sufficient conditions are established to guarantee the global stability of the endemic equilibrium by the theory of the compound matrices. Mitochondrial calcium uptake 1 (MICU1) has been demonstrated as a key regulator of...Background: Mitochondrial Ca2+ uptake is a pivotal pathophysiological process for neuronal survival when subjected to ischemic insult. Mitochondrial calcium uptake 1 (MICU1) has been demonstrated as a key regulator of the mitochondrial calcium uniporter (MCU), identified as a tetrameric highly specific channel that modulates mitochondrial Ca2+ uptake. Methods: Adult male Sprague–Dawley (SD) rats underwent middle cerebral artery occlusion (MCAO) to create the standard focal cerebral ischemia model. The permanent MCAO approach utilized the intraluminal approach. Neurological examination, and subsequent histological characterization of cerebral infarcts using triphenyltetrazolium chloride staining, as well as Western blot, immunohistochemical staining, and real-time quantitative polymerase chain reaction assays were employed to assess the functional effects of MICU1 and its expression in the brain.Results: Animals exposed to MCAO displayed the typical neurological deficit accompanied by cortical and subcortical infarction at 72 h post-stroke. The expression of MICU1, with co-localization with neurons, was detected at different time points (6 h and 12 h) after ischemic damage. Altogether, these observations revealed an up-regulation of MICU1 expression in the early stages of cerebral ischemia.Conclusion: The results demonstrated that MICU1 was upregulated in neurons at the acute phase of ischemic stroke. Because MICU1 has been previously shown to participate in mitochondrial Ca2+ uptake mediated by MCU, our study further implicates the involvement of MICU1 in calcium overload-induced cell death which is closely associated with stroke.展开更多

By using comparison arguments, it is proved when the threshold value is less than unity, the diseasefree equilibrium is globally asymptoti cally stable. When the threshold value is greater than unity, by using an iteration scheme and by constructing appropriate Lyapunov functional, respectively, sufficient conditions are derived for the global stability of the endemic equilibrium of the model. In gynecological oncology,ovarian cancer(OC)remains the most lethal,highlighting its significance in public health.Our research focused on the role of long non-coding RNA(lncRNA)in OC,particularly senescence-related l...In gynecological oncology,ovarian cancer(OC)remains the most lethal,highlighting its significance in public health.Our research focused on the role of long non-coding RNA(lncRNA)in OC,particularly senescence-related lncRNAs(SnRl-ncRNAs),crucial for OC prognosis.Utilizing data from the genotype-tissue expression(GTEx)and cancer genome Atlas(TCGA),SnRlncRNAs were discerned and subsequently,a risk signature was sculpted using co-expression and differential expression analyses,Cox regression,and least absolute shrinkage and selection operator(LASSO).This signature's robust-ness was validated through time-dependent receiver operating characteristics(ROC),and multivariate Cox regression,with further validation in the international cancer genome consortium(ICGC).Gene set enrichment analyses(GSEA)unveiled pathways intertwined with risk groups.The ROC,alongside the nomogram and calibration outcomes,attested to the model's robust predictive accuracy.Of particular significance,our model has demonstrated superiority over several commonly utilized clinical indicators,such as stage and grade.Patients in the low-risk group demonstrated greater immune infiltration and varied drug sensitivities compared to other groups.Moreover,consensus clustering classified OC patients into four distinct groups based on the expression of 17 SnRlncRNAs,showing diverse survival rates.In conclusion,these findings underscored the robustness and reliability of our model and highlighted its potential for facilitating improved decision-making in the context of risk assessment,and demonstrated that these markers potentially served as robust,efficacious biomarkers and prognostic tools,offering insights into predicting OC response to anticancer therapeutics.展开更多

文摘Background:High altitude disease(HAD)can reduce combat effectiveness and damage the health of soldiers at high altitudes.The objective of this hypothesis study is to build a four-period prevention model for high altitude disease that can be applied at high altitudes of over 3000 m.Presentation of the hypothesis:We divided the time at high altitude into nine periods,with three stages from the ascent preparation to the descent to the plain,and applied a continuous dynamic and systematic four-period prevention model across the nine periods.Each period of three stages has its own different measures and targets high altitude health care services for the prevention of high altitude disease.A standard four-period prevention model for high altitude disease was constructed for the high altitude health services at the population level.Testing the hypothesis:Our hypothesized HAD prevention model represents a continuous dynamic and systematic four-period prevention model across the nine periods.This hypothesis can be tested from three aspects.The first one is assessment of soldiers’operating efficacies.The second is comparison of the long-term high altitude population health basic data and development and utilization of big data.The third is descent population health status comparative study and historical retrospective study on prevention.Implications:As we know,it is necessary to protect soldiers’health through the ascent and descent.Through the standard four-period model,we can protect soldiers’health by preventing high altitude diseases,screening the susceptible population,securely tracking their location and maintaining soldiers’health statuses;we also maintain their operational capabilities,eliminate their psychological fears and ease their family troubles.

文摘Objective:Though Chinese medicine(CM)has showed its clinical efficacy for the treatment of obstructive sleep apnea(OSA)in China,no systematic reviews or meta-analyses provide evidences for its therapeutic effects on OSA and the long-term safety.The aim of the present study is to evaluate the effectiveness and safety of CM on OSA using meta-analysis.Methods:We used search items of"Chinese Medicine"AND"obstructive sleep apnea"to retrieve the randomized control trials(RCTs)of CM treatments for OSA in PubMed,Embase,Cochrane Library,China National Knowledge Infrastructure(CNKI),VIP database,and Wanfang database from their respective inception dates to December 2019.Only RCTs of CM therapy versus no treatment which could be quantitatively synthesized were included.Finally,20 studies representing 1,297 participants were included after extraction.Two investigators independently extracted and analyzed the data using RevMan5.3 software.Results:The treatment group using CM decoctions or CM granules presented significantly ameliorative effects on apnoea-hypopnoea index(AHI)compared with the control group(MD:-2.58,95%CI:-3.59 to-1.56,P<0.00001 and MD:-5.47,95%CI:-6.75 to-4.19,P<0.00001,respectively)in the sensitivity analysis.However,there were non significant differences in the duration of treatment between subgroups,indicating that the duration of treatment has no impacts on the therapeutic effects on AHI.CM granules also showed significantly ameliorative effects on the lowest nocturnal oxygen saturation(LSaO2)(MD:2.76,95%CI:1.85 to 3.68,P<0.00001).CM decoctions exhibited significantly improved Epworth Sleepiness Scale(ESS)scores compared with the control group in a sensitivity analysis(MD:-1.50,95%CI:-2.13 to-0.88,P<0.00001).CM granules showed a better improvement of ESS than the control group(MD:-1.35,95%CI:-1.92 to-0.78,P<0.00001).Mild adverse reactions occurred only in five patients and disappeared without special treatment.Conclusion:This study showed favorable therapeutic efficacy of CM on OSA.However,in consideration of the low methodological quality of the included RCTs,more rigorous designed,large sample size RCTs are recommended for providing more high-quality evidences.

文摘The human insulin gene modified with a C-peptide was synthesized according to the plant-preferred codon,and a fusion gene expression vector of insulin combined with green fluorescent protein(GFP)was constructed.The optimization of the flax callus culturing was undertaken,and a more efficient Agrobacterium-mediated genetic transformation of the flax hypocotyls was achieved.The critical concentration values of hygromycin on the flax hypocotyl development,as well as on its differentiated callus,were explored by the method of antibiotic gradient addition,and the application of antibiotic screening for the verification of positive calluses was assessed.The fusion gene of insulin and GFP was successfully inserted into the flax genome and expressed,as confirmed through polymerase chain reaction and Western blotting.In conclusion,we have established a flax callus culture system suitable for insulin expression.By optimizing the conditions of the flax callus induction,transformation,screening,and verification of a transgenic callus,we have provided an effective way to obtain insulin.Moreover,the herein-employed flax callus culture system could provide a feasible,cheap,and environmentally friendly platform for producing bioactive proteins.

文摘Although cancer chemopreventive agents have been confirmed to effectively protect high-risk populations from cancer invasion or recurrence,only over ten drugs have been approved by the U.S.Food and Drug Administration.Therefore,screening potent cancer chemopreventive agents is crucial to reduce the constantly increasing incidence and mortality rate of cancer.Considering the lengthy prevention process,an ideal chemopreventive agent should be nontoxic,inexpensive,and oral.Natural compounds have become a natural treasure reservoir for cancer chemoprevention because of their superior ease of availability,cost-effectiveness,and safety.The benefits of natural compounds as chemopreventive agents in cancer prevention have been confirmed in various studies.In light of this,the present review is intended to fully delineate the entire scope of cancer chemoprevention,and primarily focuses on various aspects of cancer chemoprevention based on natural compounds,specifically focusing on the mechanism of action of natural compounds in cancer prevention,and discussing in detail how they exert cancer prevention effects by affecting classical signaling pathways,immune checkpoints,and gut microbiome.We also introduce novel cancer chemoprevention strategies and summarize the role of natural compounds in improving chemotherapy regimens.Furthermore,we describe strategies for discovering anticancer compounds with low abundance and high activity,revealing the broad prospects of natural compounds in drug discovery for cancer chemoprevention.Moreover,we associate cancer chemoprevention with precision medicine,and discuss the challenges encountered in cancer chemoprevention.Finally,we emphasize the transformative potential of natural compounds in advancing the field of cancer chemoprevention and their ability to introduce more effective and less toxic preventive options for oncology.
文摘Rolling characteristics of the canard-controlled rocket with a free-spinning tail are researched based on 3-dimensional Navier–Stokes(N–S)equations.The slide mesh technology of computational fluid dynamics is used to numerically simulate the flow field when tails rotate relative to the rocket body and research the varying pattern of roll moment coefficients of the whole rocket,tail,and canard with Mach number.The comparison between the computation results and a part of experimental data shows that the simulation method is accurate.The convergence criterion during the numerical simulation is put forward;the induced rotation speed of the free-spinning tail and the roll moment coefficient of the rocket under this rotation in actual flight when the canard deflects by 10◦for roll control are obtained and the reason for the free-spinning tail being able to eliminate roll coupling is analyzed.The research provides a reference for the configuration design of canard-controlled rockets with a free-spinning tail and the numerical simulation of their aerodynamic characteristics.

文摘In this paper, an SEIR epidemic model with vaccination is formulated. The results of our mathematical analysis indicate that the basic reproduction number plays an important role in studying the dynamics of the system. If the basic reproduction number is less than unity, it is shown that the disease-free equilibrium is globally asymptotically stable by comparison arguments. If it is greater than unity, the system is permanent and there is a unique endemic equilibrium. In this case, sufficient conditions are established to guarantee the global stability of the endemic equilibrium by the theory of the compound matrices. Numerical simulations are presented to illustrate the main results.

文摘Background: Mitochondrial Ca2+ uptake is a pivotal pathophysiological process for neuronal survival when subjected to ischemic insult. Mitochondrial calcium uptake 1 (MICU1) has been demonstrated as a key regulator of the mitochondrial calcium uniporter (MCU), identified as a tetrameric highly specific channel that modulates mitochondrial Ca2+ uptake. Methods: Adult male Sprague–Dawley (SD) rats underwent middle cerebral artery occlusion (MCAO) to create the standard focal cerebral ischemia model. The permanent MCAO approach utilized the intraluminal approach. Neurological examination, and subsequent histological characterization of cerebral infarcts using triphenyltetrazolium chloride staining, as well as Western blot, immunohistochemical staining, and real-time quantitative polymerase chain reaction assays were employed to assess the functional effects of MICU1 and its expression in the brain.Results: Animals exposed to MCAO displayed the typical neurological deficit accompanied by cortical and subcortical infarction at 72 h post-stroke. The expression of MICU1, with co-localization with neurons, was detected at different time points (6 h and 12 h) after ischemic damage. Altogether, these observations revealed an up-regulation of MICU1 expression in the early stages of cerebral ischemia.Conclusion: The results demonstrated that MICU1 was upregulated in neurons at the acute phase of ischemic stroke. Because MICU1 has been previously shown to participate in mitochondrial Ca2+ uptake mediated by MCU, our study further implicates the involvement of MICU1 in calcium overload-induced cell death which is closely associated with stroke.
文摘In this paper, an SIRS epidemic model with time delay and vaccination is investigated. By analyzing the corresponding characteristic equation, the local stability of diseasefree equilibrium of the model is established. By constructing Lyapunov functional, sufficient conditions are established for the local stability of an endemic equilibrium of the model. Further, a threshold value is obtained. By using comparison arguments, it is proved when the threshold value is less than unity, the diseasefree equilibrium is globally asymptoti cally stable. When the threshold value is greater than unity, by using an iteration scheme and by constructing appropriate Lyapunov functional, respectively, sufficient conditions are derived for the global stability of the endemic equilibrium of the model. Numerical simulations are carried out to illustrate the theoretical results.
文摘The computational fluid dynamics(CFD)numerical simulation was used to investigate the aerodynamic characteristic of a canard guided rocket based on the technology of high quality point-to-point multi-block structured grid.The results of numerical simula-tion and experiments showed that the method is highly credible,and the method could be used to the rockets with semblable configurations.The canard guided rocket was designed with different noses,canards and fins.The N-S equations were solved numeri-cally by the finite volume method,and the multi-grid method was adopted to accelerate the convergence.With these strategies the aerodynamic characteristics of rocket,nose,canard and fin were educed,and the change law was summarized.The results will provide the support for the optimization of anard guided rocket in engineering design.
文摘With the development of digital media,feminist thought has demonstrated new modes of dissemination on new media platforms.This paper aims to analyze the interactions between female bloggers and users from a feminist perspective using discourse analysis as a tool.The study explores the deeper motivations and influencing factors behind interactive discourse to foster a more equitable and inclusive discussion on gender issues and a more harmonious social media discourse space.It also seeks to provide theoretical support and methodological reference for scholars studying the dissemination of feminism on new media platforms.The research finds that the pragmatic strategies of users and bloggers differ but are both influenced by subjective emotional tendencies.Furthermore,the study reveals that the discursive identities of users and bloggers in interactions are shaped by the discourse scenarios constructed by bloggers:the closer the content is to the users'lives,the tighter the connection between their discursive identities,and vice versa.Additionally,different types of content play varying roles in the operation of feminist blogger accounts.
文摘In gynecological oncology,ovarian cancer(OC)remains the most lethal,highlighting its significance in public health.Our research focused on the role of long non-coding RNA(lncRNA)in OC,particularly senescence-related lncRNAs(SnRl-ncRNAs),crucial for OC prognosis.Utilizing data from the genotype-tissue expression(GTEx)and cancer genome Atlas(TCGA),SnRlncRNAs were discerned and subsequently,a risk signature was sculpted using co-expression and differential expression analyses,Cox regression,and least absolute shrinkage and selection operator(LASSO).This signature's robust-ness was validated through time-dependent receiver operating characteristics(ROC),and multivariate Cox regression,with further validation in the international cancer genome consortium(ICGC).Gene set enrichment analyses(GSEA)unveiled pathways intertwined with risk groups.The ROC,alongside the nomogram and calibration outcomes,attested to the model's robust predictive accuracy.Of particular significance,our model has demonstrated superiority over several commonly utilized clinical indicators,such as stage and grade.Patients in the low-risk group demonstrated greater immune infiltration and varied drug sensitivities compared to other groups.Moreover,consensus clustering classified OC patients into four distinct groups based on the expression of 17 SnRlncRNAs,showing diverse survival rates.In conclusion,these findings underscored the robustness and reliability of our model and highlighted its potential for facilitating improved decision-making in the context of risk assessment,and demonstrated that these markers potentially served as robust,efficacious biomarkers and prognostic tools,offering insights into predicting OC response to anticancer therapeutics.

文摘This paper is concerned with the existence of traveling wave solutions in a reaction- diffusion predator-prey system with nonlocal delays. By introducing a partially expo- nential quasi-monotonicity condition and a new cross iteration scheme, we reduce the existence of traveling wave solutions to the existence of a pair of upper-lower solutions. By constructing a desirable pair of upper-lower solutions, we establish the existence of traveling wave solutions. Finally, some numerical examples are carried out to illustrate the theoretical results.
文摘A hepatitis B virus (HBV) model with standard incidence and the uninfected cells growing logistically is investigated. By analyzing the corresponding characteristic equations, the local stability of the infection-free and infection equilibria is discussed, respectively. Further, the existence of an orbitally asymptotically stable periodic orbit is also studied. By means of the theory of competitive systems and compound matrices, sufficient conditions are derived for the global stability of the infection-free and infection equilibria, respectively. At last, numerical simulations are carried out to illustrate the main results.
