Few-layer nanosheets(NSs)of hexagonal boron nitride(h-BN)and molybdenum disulfide(MoS_(2))display notable piezoelectric properties.Yet,their integration into polymers typically yields non-piezoelectric composites due ...Few-layer nanosheets(NSs)of hexagonal boron nitride(h-BN)and molybdenum disulfide(MoS_(2))display notable piezoelectric properties.Yet,their integration into polymers typically yields non-piezoelectric composites due to NSs’random distribution.We introduce a facile method for fabricating intrinsic piezoelectric composites incorporated with NSs without electric poling.Our innovative process aligns NSs within polyvinyl alcohol polymer,leveraging ice-water interfacial tension,water crystallization thrust,and directional cross-linking during freezing.The resulting PE composites exhibit a maximum piezoelectric coefficient of up to 25.5-28.4 pC N^(-1),comparable to polyvinylidene difluoride(PVDF),with significant costefficiency,safety,and scalability advantages over conventional materials.Using this composite,we develop highly sensitive wearable pressure and strain sensors,and an ultrasound energy harvester.These sensors detect finger bending and differentiate between walking and running,while the harvester generates1.18 V/2.31μA under 1Wcm^(-2)ultrasound input underwater.This universal method offers a novel manufacturing technique for piezoelectric composites,demonstrating remarkable effectiveness in synthesizing intrinsic piezoelectric composites based on 2D materials.Moreover,its potential extends to applications in wearable electronics and energy harvesting,promising significant advancements in these fields.展开更多

The problem of effluent total nitrogen(TN)at most of the wastewater treatment plants(WWTPs)in China is important for meeting the related water quality standards,even under the condition of high energy consumption.To a...The problem of effluent total nitrogen(TN)at most of the wastewater treatment plants(WWTPs)in China is important for meeting the related water quality standards,even under the condition of high energy consumption.To achieve better prediction and control of effluent TN concentration,an efficient prediction model,based on controllable operation parameters,was constructed in a sequencing batch reactor process.Compared with previous models,this model has two main characteristics:①Superficial gas velocity and anoxic time are controllable operation parameters and are selected as the main input parameters instead of dissolved oxygen to improve the model controllability,and②the model prediction accuracy is improved on the basis of a feedforward neural network(FFNN)with algorithm optimization.The results demonstrated that the FFNN model was efficiently optimized by scaled conjugate gradient,and the performance was excellent compared with other models in terms of the correlation coefficient(R).The optimized FFNN model could provide an accurate prediction of effluent TN based on influent water parameters and key control parameters.This study revealed the possible application of the optimized FFNN model for the efficient removal of pollutants and lower energy consumption at most of the WWTPs.展开更多

METHODS: The serum pharmacological method was used to avoid interference from administration of the crude medicinal herbs. Eight purebred NewZealand rabbits were used for preparation of serum containing various concentrations of AETC. For- ty-eight Balb/c-nu mice were used for in vivo experi- ments. The effects of serum containing AETC on the proliferation of A549 cells and expression levels of the epidermal growth factor receptor/mito- gen-activated protein kinase (EGFR/MAPK) path- way-related proteins in vitro were investigated. Ad- ditionally, the effects on the growth of A549 xeno- grafts in nude mice, and expression levels of the EG- FR/MAPK pathway-related proteins in the xeno- grafts, were investigated. RESULTS: 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl- tetrazolium bromide (MTT) assay revealed that the serum containing AETC significantly decreased the viability of A549 cells in a dose-dependent manner. Western blot showed that the serum containing various concentrations of AETC strongly reduced the levels of phospho-Jun N-terminal kinase (p-JNK) and phospho-extracellular signal-regulated kinasel/2 (ERK1/2) while it increased the level of p-p38. However, no significant effects on the ex- pression levels of JNK, ERK1/2, and p38 MAPK were found. In addition, an anticancer effect from AETC was observed in vivo in the Balb/c-nu mice bearing A549 xenografts.CONCLUSION: AETC has significant effects on the growth of A549 xenografts and on the activity of the EGFR/MAPK pathway. Therefore, AETC may be beneficial in lung carcinoma treatment.展开更多
Objective:Integration of risk stratification into fecal immunochemical test(FIT)might aid in the suboptimal detection of advanced neoplasms by FIT in colorectal cancer(CRC)screening.A comparative study was conducted t...Objective:Integration of risk stratification into fecal immunochemical test(FIT)might aid in the suboptimal detection of advanced neoplasms by FIT in colorectal cancer(CRC)screening.A comparative study was conducted to evaluate the participation and diagnostic yield of the parallel combination of questionnaire-based risk assessment(QRA)and FIT,FIT-only and QRA-only strategies in a CRC screening program in China.Methods:The study included 29,626 individuals aged 40-74 years and invited to participate in a CRC screening program in China.Participants were first invited to undertake QRA and one-time FIT(OC-sensor).Participants with positive QRA or FIT were deemed to be high-risk individuals who were recommended for subsequent colonoscopy.Participation,detection rate,and resource demand for colonoscopy were calculated and compared.Results:Of the 29,626 invitees,20,203 completed the parallel combination,8,592 completed the QRA-only,and11 completed the FIT-only strategy.For the parallel combination,FIT-only,and QRA-only strategies,the overall positivity rates were 10.2%(2,928/28,806),5.4%(1,096/20,214),and 6.8%(1,944/28,795),respectively;the yield of advanced neoplasm per 10,000 invitees were 46.9[95%confidence interval(95%CI):39.8-55.4],36.8(95%CI:30.5-44.4),and 12.2(95%CI:8.8-16.8),respectively;the positive predictive values for detecting advanced neoplasms among participants who completed colonoscopy were 4.7%(95%CI:4.0%-5.6%),9.9%(95%CI:8.3%-11.9%),and 1.9%(95%CI:1.3%-2.6%),respectively;the number of colonoscopies required to detect one advanced neoplasm was 11.4(95%CI:9.8-13.4),5.7(95%CI:4.8-6.7),and 28.4(95%CI:20.7-39.2),respectively.Conclusions:The parallel combination of QRA and FIT did not show superior efficacy for detecting advanced neoplasm compared with FIT alone in this CRC screening program.展开更多

BACKGROUND The diagnosis of sepsis combined with acute respiratory distress syndrome(ARDS)has increased owing to the enhanced awareness among medical profes-sionals and the continuous development of modern medical tec...BACKGROUND The diagnosis of sepsis combined with acute respiratory distress syndrome(ARDS)has increased owing to the enhanced awareness among medical profes-sionals and the continuous development of modern medical technologies,while early diagnosis of ARDS still lacks specific biomarkers.One of the main patho-genic mechanisms of sepsis-associated ARDS involves the actions of various pathological injuries and inflammatory factors,such as platelet and white blood cells activation,leading to an increase of surface adhesion molecules.These adhesion molecules further form platelet-white blood cell aggregates,including platelet-mononuclear cell aggregates(PMAs).PMAs has been identified as one of the markers of platelet activation,here we hypothesize that PMAs might play a potential biomarker for the early diagnosis of this complication.METHODS We selected 72 hospitalized patients diagnosed with sepsis as the study population between March 2019 and March 2022.Among them,30 patients with sepsis and ARDS formed the study group,while 42 sepsis patients without ARDS comprised the control group.After diagnosis,venous blood samples were imme-diately collected from all patients.Flow cytometry was employed to analyze the expression of PMAs,platelet neutrophil aggregates(PNAs),and platelet aggregates(PLyAs)in the serum.Additionally,the Acute Physiology and Chronic Health Evaluation(APACHE)II score was calculated for each patient,and receiver operating characteristic curves were generated to assess diagnostic value.RESULTS The study found that the levels of PNAs and PLyAs in the serum of the study group were higher than those in the control group,but the difference was not statistically significant(P>0.05).However,the expression of PMAs in the serum of the study group was significantly upregulated(P<0.05)and positively correlated with the APACHE II score(r=0.671,P<0.05).When using PMAs as a diagnostic indicator,the area under the curve value was 0.957,indicating a high diagnostic value(P<0.05).Furthermore,the optimal cutoff value was 8.418%,with a diagnostic sensitivity of 0.819 and specificity of 0.947.CONCLUSION In summary,the serum levels of PMAs significantly increase in patients with sepsis and ARDS.Therefore,serum PMAs have the potential to become a new biomarker for clinically diagnosing sepsis complicated by ARDS.展开更多

The micro-RNA (miR...This study was performed to investigate a potential marker for the presence of spermatozoa in the ejaculate following varicocelectomy in Chinese men with nonobstructive azoospermia and varicoceles. The micro-RNA (miR)-192a levels in seminal plasma and testicular tissue were evaluated by quantitative real-time polymerase chain reaction from 60 men with nonobstructive azoospermia and varicoceles (Group A: 27 men with spermatozoa found in the ejaculate after surgery; Group B: 33 men without spermatozoa found in the ejaculate after surgery) and 30 controls. The seminal plasma and testicular tissue miR-192a levels were higher in Group B than in Group A and the controls (P〈 0.001), and there was no significant difference between Group A and the controls (P〉 0.05). Apoptosis and proliferation assays with miR mimics and inhibitors showed that miR-192a induced GC-2 cell apoptosis through the activation of Caspase-3 protein. Thus, seminal plasma miR-192a appears to be a potential marker for successfully indicating spermatozoa in the ejaculate following microsurgical varicocelectomy in men with nonobstructive azoospermia and varicoceles. Seminal plasma miR-192a may be a useful clinical marker for prescreening to determine which patients with nonobstructive azoospermia and varicoceles would benefit from varicocelectomy.展开更多
Ectopic insulinoma is a very rare and dormant tumor.Here we report the case of a 79-year-old female who presented with repeated episodes of hypoglycemia and was diagnosed with insulinoma based on laboratory and imagin...Ectopic insulinoma is a very rare and dormant tumor.Here we report the case of a 79-year-old female who presented with repeated episodes of hypoglycemia and was diagnosed with insulinoma based on laboratory and imaging examinations.Computed tomography and positron emission tomography revealed a tumor in the retroperitoneum under and left of the hepatoduodenal ligament, which was resected successfully using a laparoscopic approach.Pathologic results revealed an ectopic insulinoma, which was confirmed immunohistochemically.Ectopic insulinomas are accompanied by hypoglycemia that can be misdiagnosed as drug- or disease-induced.These tumors are difficult to diagnose and locate, particularly in atypical cases or for very small tumors.Synthetic or targeted examinations, including low blood glucose, elevated insulin, proinsulin, and C-peptide levels, 48-h fasting tests, and relevant imaging methods should be considered for suspected cases of insulinoma.Surgery is the treatment of choice for patients with insulinoma, and laparoscopic resection is a feasible and effective method for select ectopic insulinoma cases.展开更多
BACKGROUND Pancreatic cancer(PC)is one of the most lethal malignancies worldwide.It is known that the proliferation of PC cells is a critical process in the disease.Previous studies have failed to identify the key gen...BACKGROUND Pancreatic cancer(PC)is one of the most lethal malignancies worldwide.It is known that the proliferation of PC cells is a critical process in the disease.Previous studies have failed to identify the key genes associated with PC cell proliferation,using bioinformatic analysis,genome-wide association studies,and candidate gene testing.AIM To investigate the function of the chromobox 8(CBX8)/receptor substrate 1(IRS1)/AKT axis in PC.METHODS A genome-wide CRISPR-Cas9 screening was performed to select genes that could facilitate PC cell proliferation.Quantitative reverse transcription-polymerase chain reaction was used to detect the expression of CBX8 in PC tissues and cells.The regulatory roles of CBX8 in cell proliferation,migration,and invasion were verified by in vivo and in vitro functional assays.RESULTS CBX8 was upregulated in PC tissues and shown to drive PC cell proliferation.Higher expression of CBX8 was correlated with worse outcomes of PC patients from two independent cohorts comprising a total of 116 cases.CBX8 was also proved to serve as a promising therapeutic target for a PC xenograft model.We demonstrated that hypoxia-inducible factor(HIF)-1a induced CBX8 transcription by binding to the promoter of CBX8.CBX8 efficiently activated the PI3K/AKT signaling by upregulating insulin IRS1.CONCLUSION CBX8 is a key gene regulated by HIF-1α,and activates the IRS1/AKT pathway,which suggests that targeting CBX8 may be a promising therapeutic strategy for PC.展开更多

Background and Objective The effectiveness of radiofrequency ablation(RFA)in improving long-term survival outcomes for patients with a solitary hepatocellular carcinoma(HCC)measuring 5 cm or less remains uncertain.Thi...Background and Objective The effectiveness of radiofrequency ablation(RFA)in improving long-term survival outcomes for patients with a solitary hepatocellular carcinoma(HCC)measuring 5 cm or less remains uncertain.This study was designed to elucidate the impact of RFA therapy on the survival outcomes of these patients and to construct a prognostic model for patients following RFA.Methods This study was performed using the Surveillance,Epidemiology,and End Results(SEER)database from 2004 to 2017,focusing on patients diagnosed with a solitary HCC lesion≤5 cm in size.We compared the overall survival(OS)and cancer-specific survival(CSS)rates of these patients with those of patients who received hepatectomy,radiotherapy,or chemotherapy or who were part of a blank control group.To enhance the reliability of our findings,we employed stabilized inverse probability treatment weighting(sIPTW)and stratified analyses.Additionally,we conducted a Cox regression analysis to identify prognostic factors.XGBoost models were developed to predict 1-,3-,and 5-year CSS.The XGBoost models were evaluated via receiver operating characteristic(ROC)curves,calibration plots,decision curve analysis(DCA)curves and so on.Results Regardless of whether the data were unadjusted or adjusted for the use of sIPTWs,the 5-year OS(46.7%)and CSS(58.9%)rates were greater in the RFA group than in the radiotherapy(27.1%/35.8%),chemotherapy(32.9%/43.7%),and blank control(18.6%/30.7%)groups,but these rates were lower than those in the hepatectomy group(69.4%/78.9%).Stratified analysis based on age and cirrhosis status revealed that RFA and hepatectomy yielded similar OS and CSS outcomes for patients with cirrhosis aged over 65 years.Age,race,marital status,grade,cirrhosis status,tumor size,and AFP level were selected to construct the XGBoost models based on the training cohort.The areas under the curve(AUCs)for 1,3,and 5 years in the validation cohort were 0.88,0.81,and 0.79,respectively.Calibration plots further demonstrated the consistency between the predicted and actual values in both the training and validation cohorts.Conclusion RFA can improve the survival of patients diagnosed with a solitary HCC lesion≤5 cm.In certain clinical scenarios,RFA achieves survival outcomes comparable to those of hepatectomy.The XGBoost models developed in this study performed admirably in predicting the CSS of patients with solitary HCC tumors smaller than 5 cm following RFA.展开更多
The potentials of rare earth-based nanocomposite alloys have never been realized due to strict microstructural constraints.Owing to the easy demagnetization it is challenging to increase the soft magnetic phase conten...The potentials of rare earth-based nanocomposite alloys have never been realized due to strict microstructural constraints.Owing to the easy demagnetization it is challenging to increase the soft magnetic phase content.To avoid the easy demagnetization,Pr-Fe-B/Alnico magnets were fabricated and reported in this manuscript.The content of the Alnico phase is increased from 0 to 25 wt%,while the content of Pr element is reduced to below the sub-stoichiometry of the 2:14:1 main phase.The maximum magnetic energy product,which is the figure-of-merit for permanent magnets,is increased from 122 kJ/m^(3) for the standard alloy to 146 kJ/m^(3) for the alloy with 15 wt% Alnico which shows a significant improvement considering the fact that the Curie point of the magnet is also increased by~66 K.The special microstructure contains distinctly and heterogeneously distributed 2:14:1 and Alnico phases.The dimensions of neither the 2:14:1 nor the Alnico phases meet the dimensional requirements of the nanocomposite magnets,but still the smooth demagnetization curves are noted for the alloys.The behavior of effective anisotropy,the performance of the magnets in applied magnetic field and the magnetic interactions among the various constituent grains were quantitatively studied by reversible susceptibility,irreversible susceptibility and re coil loop openness.This study may provide some guiding principles for the development of nanocomposite magnetic alloys with excellent magnetic properties by using much less RE elements.展开更多
The highly conserved target of rapamycin(TOR)pathway plays an important role in aging across species.Previous studies have established that inhibition of the TOR complex 1(TORC1)significantly extends lifespan in Caeno...The highly conserved target of rapamycin(TOR)pathway plays an important role in aging across species.Previous studies have established that inhibition of the TOR complex 1(TORC1)significantly extends lifespan in Caenorhabditis elegans.However,it has not been clear whether TORC1 perturbation affects aging in a spatiotemporal manner.Here,we apply the auxin-inducible degradation tool to knock down endogenous DAF-15,the C.elegans ortholog of regulatory associated protein of TOR(Raptor),to characterize its roles in aging.Global or tissue-specific inhibition of DAF-15 during development results in various growth defects,whereas neuron-specific knockdown of DAF-15 during adulthood significantly extends lifespan and healthspan.The neuronal DAF-15 deficiency-induced longevity requires the intestinal activities of DAF-16/FOXO and PHA-4/FOXA transcription factors,as well as the AAK-2/AMP-activated protein kinaseαcatalytic subunit.Transcriptome profiling reveals that the neuronal DAF-15 knockdown promotes the expression of genes involved in protection.These findings define the tissue-specific roles of TORC1 in healthy aging and highlight the importance of neuronal modulation of aging.展开更多

In this paper,we consider a downlink non-orthogonal multiple access(NOMA)network assisted by two reconfigurable intelligent surfaces(RISs)over Rician fading channels,in which each user communicates with the base stati...In this paper,we consider a downlink non-orthogonal multiple access(NOMA)network assisted by two reconfigurable intelligent surfaces(RISs)over Rician fading channels,in which each user communicates with the base station by the virtue of a RIS to enhance the reliability of the received signal.To evaluate the system performance of our proposed RIS-NOMA network,we first derive the exact and asymptotic expressions for the outage probability and ergodic rate of two users.Then,we derive the exact and asymptotic upper bound expressions for the ergodic rate of the nearby user.Based on asymptotic analytical results,the diversity orders for the outage probability and the high signal-to-noise ratio(SNR)slopes for the ergodic rate of the two users are obtained in the high SNR regime.Moreover,we derive the system throughputs of the proposed RIS-NOMA network in delay-limited and delay-tolerant transmission modes.Numerical results confirm our analysis and demonstrate that:1)The outage probability and ergodic rate of RIS-NOMA networks are superior to that of RIS-assisted orthogonalmultiple access(OMA)networks;2)The RIS-NOMA networks have ability to achieve a larger system throughput compared to RIS-OMA networks;and 3)The system performance of RIS-NOMA networks can be significantly improved as the number of reflecting elements and Rician factor increases.展开更多

文摘Few-layer nanosheets(NSs)of hexagonal boron nitride(h-BN)and molybdenum disulfide(MoS_(2))display notable piezoelectric properties.Yet,their integration into polymers typically yields non-piezoelectric composites due to NSs’random distribution.We introduce a facile method for fabricating intrinsic piezoelectric composites incorporated with NSs without electric poling.Our innovative process aligns NSs within polyvinyl alcohol polymer,leveraging ice-water interfacial tension,water crystallization thrust,and directional cross-linking during freezing.The resulting PE composites exhibit a maximum piezoelectric coefficient of up to 25.5-28.4 pC N^(-1),comparable to polyvinylidene difluoride(PVDF),with significant costefficiency,safety,and scalability advantages over conventional materials.Using this composite,we develop highly sensitive wearable pressure and strain sensors,and an ultrasound energy harvester.These sensors detect finger bending and differentiate between walking and running,while the harvester generates1.18 V/2.31μA under 1Wcm^(-2)ultrasound input underwater.This universal method offers a novel manufacturing technique for piezoelectric composites,demonstrating remarkable effectiveness in synthesizing intrinsic piezoelectric composites based on 2D materials.Moreover,its potential extends to applications in wearable electronics and energy harvesting,promising significant advancements in these fields.

文摘Two new species,Pangrapta paraumbrosa sp.nov.(Hunan,Shaanxi,Sichuan)and P.hutiaoxiaensis sp.nov.(Yunnan),are described from China.P.paraumbrosa is similar to P.umbrosa,but it can be easily distinguished by a larger wingspan,a grayish white reniform spot,valva without process,vesica with the different size cornuti,and a screwy and membranous ductus bursae;P.hutiaoxiaensis is similar to P.shivula,but it can be easily distinguished by forewing with a big orbicular spot with blackish brown circle,a gray reniform spot with brown circle and partially apparent stripes in the hindwing,and male genitalia with a cluster of cornuti on vesica.Detailed illustrations of adults and genitalia are provided.

文摘The problem of effluent total nitrogen(TN)at most of the wastewater treatment plants(WWTPs)in China is important for meeting the related water quality standards,even under the condition of high energy consumption.To achieve better prediction and control of effluent TN concentration,an efficient prediction model,based on controllable operation parameters,was constructed in a sequencing batch reactor process.Compared with previous models,this model has two main characteristics:①Superficial gas velocity and anoxic time are controllable operation parameters and are selected as the main input parameters instead of dissolved oxygen to improve the model controllability,and②the model prediction accuracy is improved on the basis of a feedforward neural network(FFNN)with algorithm optimization.The results demonstrated that the FFNN model was efficiently optimized by scaled conjugate gradient,and the performance was excellent compared with other models in terms of the correlation coefficient(R).The optimized FFNN model could provide an accurate prediction of effluent TN based on influent water parameters and key control parameters.This study revealed the possible application of the optimized FFNN model for the efficient removal of pollutants and lower energy consumption at most of the WWTPs.

文摘OBJECTIVE: To explore the anticancer mechanism of aqueous extract of Taxus Chinensis (Pilger) Rehd (AETC). METHODS: The serum pharmacological method was used to avoid interference from administration of the crude medicinal herbs. Eight purebred NewZealand rabbits were used for preparation of serum containing various concentrations of AETC. For- ty-eight Balb/c-nu mice were used for in vivo experi- ments. The effects of serum containing AETC on the proliferation of A549 cells and expression levels of the epidermal growth factor receptor/mito- gen-activated protein kinase (EGFR/MAPK) path- way-related proteins in vitro were investigated. Ad- ditionally, the effects on the growth of A549 xeno- grafts in nude mice, and expression levels of the EG- FR/MAPK pathway-related proteins in the xeno- grafts, were investigated. RESULTS: 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl- tetrazolium bromide (MTT) assay revealed that the serum containing AETC significantly decreased the viability of A549 cells in a dose-dependent manner. Western blot showed that the serum containing various concentrations of AETC strongly reduced the levels of phospho-Jun N-terminal kinase (p-JNK) and phospho-extracellular signal-regulated kinasel/2 (ERK1/2) while it increased the level of p-p38. However, no significant effects on the ex- pression levels of JNK, ERK1/2, and p38 MAPK were found. In addition, an anticancer effect from AETC was observed in vivo in the Balb/c-nu mice bearing A549 xenografts.CONCLUSION: AETC has significant effects on the growth of A549 xenografts and on the activity of the EGFR/MAPK pathway. Therefore, AETC may be beneficial in lung carcinoma treatment.
文摘Objective:Integration of risk stratification into fecal immunochemical test(FIT)might aid in the suboptimal detection of advanced neoplasms by FIT in colorectal cancer(CRC)screening.A comparative study was conducted to evaluate the participation and diagnostic yield of the parallel combination of questionnaire-based risk assessment(QRA)and FIT,FIT-only and QRA-only strategies in a CRC screening program in China.Methods:The study included 29,626 individuals aged 40-74 years and invited to participate in a CRC screening program in China.Participants were first invited to undertake QRA and one-time FIT(OC-sensor).Participants with positive QRA or FIT were deemed to be high-risk individuals who were recommended for subsequent colonoscopy.Participation,detection rate,and resource demand for colonoscopy were calculated and compared.Results:Of the 29,626 invitees,20,203 completed the parallel combination,8,592 completed the QRA-only,and11 completed the FIT-only strategy.For the parallel combination,FIT-only,and QRA-only strategies,the overall positivity rates were 10.2%(2,928/28,806),5.4%(1,096/20,214),and 6.8%(1,944/28,795),respectively;the yield of advanced neoplasm per 10,000 invitees were 46.9[95%confidence interval(95%CI):39.8-55.4],36.8(95%CI:30.5-44.4),and 12.2(95%CI:8.8-16.8),respectively;the positive predictive values for detecting advanced neoplasms among participants who completed colonoscopy were 4.7%(95%CI:4.0%-5.6%),9.9%(95%CI:8.3%-11.9%),and 1.9%(95%CI:1.3%-2.6%),respectively;the number of colonoscopies required to detect one advanced neoplasm was 11.4(95%CI:9.8-13.4),5.7(95%CI:4.8-6.7),and 28.4(95%CI:20.7-39.2),respectively.Conclusions:The parallel combination of QRA and FIT did not show superior efficacy for detecting advanced neoplasm compared with FIT alone in this CRC screening program.

文摘Cancer cells undergo substantial metabolic alterations to sustain increased energy supply and uncontrolled proliferation.As an essential trace element,iron is vital for many biological processes.Evidence has revealed that cancer cells deploy various mechanisms to elevate the cellular iron concentration to accelerate proliferation.Ferroptosis,a form of cell death caused by iron-catalyzed excessive peroxidation of polyunsaturated fatty acids(PUFAs),is a promising therapeutic target for therapyresistant cancers.Previous studies have reported that long noncoding RNA(lncRNA)is a group of critical regulators involved in modulating cell metabolism,proliferation,apoptosis,and ferroptosis.In this review,we summarize the associations among iron metabolism,ferroptosis,and ferroptosis-related lncRNA in tumorigenesis.This information will help deepen understanding of the role of lncRNA in iron metabolism and raise the possibility of targeting lncRNA and ferroptosis in cancer combination therapy.
文摘BACKGROUND The diagnosis of sepsis combined with acute respiratory distress syndrome(ARDS)has increased owing to the enhanced awareness among medical profes-sionals and the continuous development of modern medical technologies,while early diagnosis of ARDS still lacks specific biomarkers.One of the main patho-genic mechanisms of sepsis-associated ARDS involves the actions of various pathological injuries and inflammatory factors,such as platelet and white blood cells activation,leading to an increase of surface adhesion molecules.These adhesion molecules further form platelet-white blood cell aggregates,including platelet-mononuclear cell aggregates(PMAs).PMAs has been identified as one of the markers of platelet activation,here we hypothesize that PMAs might play a potential biomarker for the early diagnosis of this complication.METHODS We selected 72 hospitalized patients diagnosed with sepsis as the study population between March 2019 and March 2022.Among them,30 patients with sepsis and ARDS formed the study group,while 42 sepsis patients without ARDS comprised the control group.After diagnosis,venous blood samples were imme-diately collected from all patients.Flow cytometry was employed to analyze the expression of PMAs,platelet neutrophil aggregates(PNAs),and platelet aggregates(PLyAs)in the serum.Additionally,the Acute Physiology and Chronic Health Evaluation(APACHE)II score was calculated for each patient,and receiver operating characteristic curves were generated to assess diagnostic value.RESULTS The study found that the levels of PNAs and PLyAs in the serum of the study group were higher than those in the control group,but the difference was not statistically significant(P>0.05).However,the expression of PMAs in the serum of the study group was significantly upregulated(P<0.05)and positively correlated with the APACHE II score(r=0.671,P<0.05).When using PMAs as a diagnostic indicator,the area under the curve value was 0.957,indicating a high diagnostic value(P<0.05).Furthermore,the optimal cutoff value was 8.418%,with a diagnostic sensitivity of 0.819 and specificity of 0.947.CONCLUSION In summary,the serum levels of PMAs significantly increase in patients with sepsis and ARDS.Therefore,serum PMAs have the potential to become a new biomarker for clinically diagnosing sepsis complicated by ARDS.
文摘This study was performed to investigate a potential marker for the presence of spermatozoa in the ejaculate following varicocelectomy in Chinese men with nonobstructive azoospermia and varicoceles. The micro-RNA (miR)-192a levels in seminal plasma and testicular tissue were evaluated by quantitative real-time polymerase chain reaction from 60 men with nonobstructive azoospermia and varicoceles (Group A: 27 men with spermatozoa found in the ejaculate after surgery; Group B: 33 men without spermatozoa found in the ejaculate after surgery) and 30 controls. The seminal plasma and testicular tissue miR-192a levels were higher in Group B than in Group A and the controls (P〈 0.001), and there was no significant difference between Group A and the controls (P〉 0.05). Apoptosis and proliferation assays with miR mimics and inhibitors showed that miR-192a induced GC-2 cell apoptosis through the activation of Caspase-3 protein. Thus, seminal plasma miR-192a appears to be a potential marker for successfully indicating spermatozoa in the ejaculate following microsurgical varicocelectomy in men with nonobstructive azoospermia and varicoceles. Seminal plasma miR-192a may be a useful clinical marker for prescreening to determine which patients with nonobstructive azoospermia and varicoceles would benefit from varicocelectomy.
文摘Ectopic insulinoma is a very rare and dormant tumor.Here we report the case of a 79-year-old female who presented with repeated episodes of hypoglycemia and was diagnosed with insulinoma based on laboratory and imaging examinations.Computed tomography and positron emission tomography revealed a tumor in the retroperitoneum under and left of the hepatoduodenal ligament, which was resected successfully using a laparoscopic approach.Pathologic results revealed an ectopic insulinoma, which was confirmed immunohistochemically.Ectopic insulinomas are accompanied by hypoglycemia that can be misdiagnosed as drug- or disease-induced.These tumors are difficult to diagnose and locate, particularly in atypical cases or for very small tumors.Synthetic or targeted examinations, including low blood glucose, elevated insulin, proinsulin, and C-peptide levels, 48-h fasting tests, and relevant imaging methods should be considered for suspected cases of insulinoma.Surgery is the treatment of choice for patients with insulinoma, and laparoscopic resection is a feasible and effective method for select ectopic insulinoma cases.

文摘BACKGROUND Pancreatic cancer(PC)is one of the most lethal malignancies worldwide.It is known that the proliferation of PC cells is a critical process in the disease.Previous studies have failed to identify the key genes associated with PC cell proliferation,using bioinformatic analysis,genome-wide association studies,and candidate gene testing.AIM To investigate the function of the chromobox 8(CBX8)/receptor substrate 1(IRS1)/AKT axis in PC.METHODS A genome-wide CRISPR-Cas9 screening was performed to select genes that could facilitate PC cell proliferation.Quantitative reverse transcription-polymerase chain reaction was used to detect the expression of CBX8 in PC tissues and cells.The regulatory roles of CBX8 in cell proliferation,migration,and invasion were verified by in vivo and in vitro functional assays.RESULTS CBX8 was upregulated in PC tissues and shown to drive PC cell proliferation.Higher expression of CBX8 was correlated with worse outcomes of PC patients from two independent cohorts comprising a total of 116 cases.CBX8 was also proved to serve as a promising therapeutic target for a PC xenograft model.We demonstrated that hypoxia-inducible factor(HIF)-1a induced CBX8 transcription by binding to the promoter of CBX8.CBX8 efficiently activated the PI3K/AKT signaling by upregulating insulin IRS1.CONCLUSION CBX8 is a key gene regulated by HIF-1α,and activates the IRS1/AKT pathway,which suggests that targeting CBX8 may be a promising therapeutic strategy for PC.

文摘The non-Hermitian skin effect has been applied in multiple fields.However,there are relatively few models in the field of thermal diffusion that utilize the non-Hermitian skin effect for achieving thermal regulation.Here,we propose two non-Hermitian Su-Schrieffer-Heeger(SSH)models for thermal regulation:one capable of achieving edge states,and the other capable of achieving corner states within the thermal field.By analyzing the energy band structures and the generalized Brillouin zone,we predict the appearance of the non-Hermitian skin effect in these two models.Furthermore,we analyze the time-dependent evolution results and assess the robustness of the models.The results indicate that the localized thermal effects of the models align with our predictions.In a word,this work presents two models based on the non-Hermitian skin effect for regulating the thermal field,injecting vitality into the design of non-Hermitian thermal diffusion systems.
文摘Background and Objective The effectiveness of radiofrequency ablation(RFA)in improving long-term survival outcomes for patients with a solitary hepatocellular carcinoma(HCC)measuring 5 cm or less remains uncertain.This study was designed to elucidate the impact of RFA therapy on the survival outcomes of these patients and to construct a prognostic model for patients following RFA.Methods This study was performed using the Surveillance,Epidemiology,and End Results(SEER)database from 2004 to 2017,focusing on patients diagnosed with a solitary HCC lesion≤5 cm in size.We compared the overall survival(OS)and cancer-specific survival(CSS)rates of these patients with those of patients who received hepatectomy,radiotherapy,or chemotherapy or who were part of a blank control group.To enhance the reliability of our findings,we employed stabilized inverse probability treatment weighting(sIPTW)and stratified analyses.Additionally,we conducted a Cox regression analysis to identify prognostic factors.XGBoost models were developed to predict 1-,3-,and 5-year CSS.The XGBoost models were evaluated via receiver operating characteristic(ROC)curves,calibration plots,decision curve analysis(DCA)curves and so on.Results Regardless of whether the data were unadjusted or adjusted for the use of sIPTWs,the 5-year OS(46.7%)and CSS(58.9%)rates were greater in the RFA group than in the radiotherapy(27.1%/35.8%),chemotherapy(32.9%/43.7%),and blank control(18.6%/30.7%)groups,but these rates were lower than those in the hepatectomy group(69.4%/78.9%).Stratified analysis based on age and cirrhosis status revealed that RFA and hepatectomy yielded similar OS and CSS outcomes for patients with cirrhosis aged over 65 years.Age,race,marital status,grade,cirrhosis status,tumor size,and AFP level were selected to construct the XGBoost models based on the training cohort.The areas under the curve(AUCs)for 1,3,and 5 years in the validation cohort were 0.88,0.81,and 0.79,respectively.Calibration plots further demonstrated the consistency between the predicted and actual values in both the training and validation cohorts.Conclusion RFA can improve the survival of patients diagnosed with a solitary HCC lesion≤5 cm.In certain clinical scenarios,RFA achieves survival outcomes comparable to those of hepatectomy.The XGBoost models developed in this study performed admirably in predicting the CSS of patients with solitary HCC tumors smaller than 5 cm following RFA.

文摘The potentials of rare earth-based nanocomposite alloys have never been realized due to strict microstructural constraints.Owing to the easy demagnetization it is challenging to increase the soft magnetic phase content.To avoid the easy demagnetization,Pr-Fe-B/Alnico magnets were fabricated and reported in this manuscript.The content of the Alnico phase is increased from 0 to 25 wt%,while the content of Pr element is reduced to below the sub-stoichiometry of the 2:14:1 main phase.The maximum magnetic energy product,which is the figure-of-merit for permanent magnets,is increased from 122 kJ/m^(3) for the standard alloy to 146 kJ/m^(3) for the alloy with 15 wt% Alnico which shows a significant improvement considering the fact that the Curie point of the magnet is also increased by~66 K.The special microstructure contains distinctly and heterogeneously distributed 2:14:1 and Alnico phases.The dimensions of neither the 2:14:1 nor the Alnico phases meet the dimensional requirements of the nanocomposite magnets,but still the smooth demagnetization curves are noted for the alloys.The behavior of effective anisotropy,the performance of the magnets in applied magnetic field and the magnetic interactions among the various constituent grains were quantitatively studied by reversible susceptibility,irreversible susceptibility and re coil loop openness.This study may provide some guiding principles for the development of nanocomposite magnetic alloys with excellent magnetic properties by using much less RE elements.

文摘The highly conserved target of rapamycin(TOR)pathway plays an important role in aging across species.Previous studies have established that inhibition of the TOR complex 1(TORC1)significantly extends lifespan in Caenorhabditis elegans.However,it has not been clear whether TORC1 perturbation affects aging in a spatiotemporal manner.Here,we apply the auxin-inducible degradation tool to knock down endogenous DAF-15,the C.elegans ortholog of regulatory associated protein of TOR(Raptor),to characterize its roles in aging.Global or tissue-specific inhibition of DAF-15 during development results in various growth defects,whereas neuron-specific knockdown of DAF-15 during adulthood significantly extends lifespan and healthspan.The neuronal DAF-15 deficiency-induced longevity requires the intestinal activities of DAF-16/FOXO and PHA-4/FOXA transcription factors,as well as the AAK-2/AMP-activated protein kinaseαcatalytic subunit.Transcriptome profiling reveals that the neuronal DAF-15 knockdown promotes the expression of genes involved in protection.These findings define the tissue-specific roles of TORC1 in healthy aging and highlight the importance of neuronal modulation of aging.

文摘Solving intrinsic structural problems such as low conductivity is the main challenge to promote the commercial application of Li_(2)TiSiO_(5).In this study,Li_(2)TiSiO_(5) is synthesized by the sol-gelmethod,and the surface modification of Li_(2)TiSiO_(5) is carried out at different temperatures using low-temperature plasma to enhance its lithium storage performance.The morphological structure and electrochemical tests demonstrate that plasma treatment can improve the degree of agglomeration.The peak position of the plasma-treated Li_(2)TiSiO_(5) is shifted to a lower angle,and the shift angle increases with increasing sputtering power.Li_(2)TiSiO_(5) after 300 W bombardment shows excellent capacity(144.7 mA·hg^(−1)after 500 cycles at 0.1 Ag^(−1))and rate performance(140 mA·hg^(−1)at 5 Ag^(−1)).Electrochemical analysis indicates that excellent electrochemical performance is attributed to the enhancement of electronic and ionic conductivity by plasma bombardment.

文摘In this paper,we consider a downlink non-orthogonal multiple access(NOMA)network assisted by two reconfigurable intelligent surfaces(RISs)over Rician fading channels,in which each user communicates with the base station by the virtue of a RIS to enhance the reliability of the received signal.To evaluate the system performance of our proposed RIS-NOMA network,we first derive the exact and asymptotic expressions for the outage probability and ergodic rate of two users.Then,we derive the exact and asymptotic upper bound expressions for the ergodic rate of the nearby user.Based on asymptotic analytical results,the diversity orders for the outage probability and the high signal-to-noise ratio(SNR)slopes for the ergodic rate of the two users are obtained in the high SNR regime.Moreover,we derive the system throughputs of the proposed RIS-NOMA network in delay-limited and delay-tolerant transmission modes.Numerical results confirm our analysis and demonstrate that:1)The outage probability and ergodic rate of RIS-NOMA networks are superior to that of RIS-assisted orthogonalmultiple access(OMA)networks;2)The RIS-NOMA networks have ability to achieve a larger system throughput compared to RIS-OMA networks;and 3)The system performance of RIS-NOMA networks can be significantly improved as the number of reflecting elements and Rician factor increases.
