Economical,stable,and corrosion-resistant catalytic electrodes are still urgently needed for the oxygen evolution reaction(OER)in water and seawater.Herein,a mild electroless plating strategy is used to achieve large-...Economical,stable,and corrosion-resistant catalytic electrodes are still urgently needed for the oxygen evolution reaction(OER)in water and seawater.Herein,a mild electroless plating strategy is used to achieve large-scale preparation of the“integrated”phosphorus-based precatalyst(FeP-NiP)on nickel foam(NF),which is in situ reconstructed into a highly active and corrosion-resistant(Fe)NiOOH phase for OER.The interaction between phosphate anions(PO_(x)^(y-))and iron ions(Fe^(3+))tunes the electronic structure of the catalytic phase to further enhance OER kinetics.The integrated FeP-NiP@NF electrode exhibits low overpotentials for OER in alkaline water/seawater,requiring only 275/289,320/336,and 349/358 mV to reach 0.1,0.5,and 1.0 A cm^(−2),respectively.The in situ reconstructed PO_(x)^(y-)anion electrostatically repels Cl−in seawater electrolytes,allowing stable operation for over 7 days at 1.0 A cm^(−2) in extreme electrolytes(1.0 M KOH+seawater and 6.0 M KOH+seawater),demonstrating industrial-level stability.This study overcomes the complex synthesis limitations of P-based materials through innovative material design,opening new avenues for electrochemical energy conversion.展开更多
BACKGROUND Type 2 diabetes mellitus(T2DM)often leads to vascular complications,such as albuminuria.The role of insulin autoantibodies(IAA)and their interaction with D-dimer in this context remains unclear.AIM To inves...BACKGROUND Type 2 diabetes mellitus(T2DM)often leads to vascular complications,such as albuminuria.The role of insulin autoantibodies(IAA)and their interaction with D-dimer in this context remains unclear.AIM To investigate the characteristics of IAA and its effect on albuminuria in T2DM patients.METHODS We retrospectively analyzed clinical data from 115 T2DM patients with positive IAA induced by exogenous insulin,and 115 age-and sex-matched IAA-negative T2DM patients as controls.Propensity scores were calculated using multivariate logistic regression.Key variables were selected using the least absolute shrinkage and selection operator(LASSO)algorithm.We constructed a prediction model and analyzed the association between IAA and albuminuria based on demographic and laboratory parameters.RESULTS The IAA-positive group had significantly higher D-dimer levels[0.30(0.19-0.55)mg/L vs 0.21(0.19-0.33)mg/L,P=0.008]and plasma insulin levels[39.1(12.0-102.7)μU/mL vs 9.8(5.5-17.6)μU/mL,P<0.001]compared to the IAA-negative group.Increases in the insulin dose per weight ratio,diabetes duration,and urinary albumin-to-creatinine ratio(UACR)were observed but did not reach statistical significance.The LASSO model identified plasma insulin and D-dimer as key factors with larger coefficients.D-dimer was significantly associated with UACR in the total and IAA-positive groups but not in the IAA-negative group.The odds ratio for D-dimer elevation(>0.5 g/L)was 2.88(95%confidence interval:1.17-7.07)in the IAA-positive group(P interaction<0.05).CONCLUSION D-dimer elevation is an independent risk factor for abnormal albuminuria and interacts with IAA in the development of abnormal albuminuria in T2DM patients.展开更多

Background:Osteoarthritis(OA)is a long-term degenerative joint disease worsen-ing over time.Aging and chondrocyte senescence contribute to OA progression.MicroRNAs have been confirmed to regulate different cellular pr...Background:Osteoarthritis(OA)is a long-term degenerative joint disease worsen-ing over time.Aging and chondrocyte senescence contribute to OA progression.MicroRNAs have been confirmed to regulate different cellular processes.They con-tribute to OA pathology and may help to identify novel biomarkers and therapies for OA.Methods:This study used bioinformatics and experimental investigations to analyze and validate differentially expressed miRNAs in OA that might affect chondrocyte apoptosis and senescence.Results:miR-6779 was found to be significantly down-regulated in OA.Seventy-six of the predicted and miR-6779 targeted genes and the OA-associated disease genes overlapped,and these were enriched in cell proliferation,cell apoptosis,and cell cycle.miR-6779 overexpression remarkably attenuated IL-1βeffects on chon-drocytes by reducing MMP3 and MMP13 levels,promoting cell apoptosis,suppress-ing cell senescence,and increasing caspase-3,caspase-9 and reducing P16 and P21 levels.miR-6779 targeted inhibition of X-linked inhibitor of apoptosis protein(XIAP)expression.XIAP knockdown partially improved IL-1β-induced chondrocyte senes-cence and dysfunction.Lastly,when co-transfected with a miR-6779 agomir,the XIAP overexpression vector partially attenuated the effects of miR-6779 overexpression on chondrocytes;miR-6779 improved IL-1β-induced senescence and dysfunction in chondrocytes through targeting XIAP.Conclusion:miR-6779 is down-regulated,and XIAP is up-regulated in OA cartilage and IL-1β-treated chondrocytes.miR-6779 inhibits XIAP expression,thereby promot-ing senescent chondrocyte cell apoptosis and reducing chondrocyte senescence and ECM loss through XIAP.展开更多

Methods: Fifty elderly women with reduced balance function, admitted to the Qingbar Elderly Care Center of Chongqing Medical University from December 2022 to December 2023, were randomly selected and divided into two groups. The aerobic exercise group (25 patients) received traditional treatment and rehabilitation nursing, while the balance rhythm dance intervention group (25 patients) received the balance rhythm dance intervention in addition to traditional treatment and rehabilitation nursing. The Unified Parkinson’s Disease Rating Scale (UPDRS) and Berg Balance Scale (BBS) were used as evaluation indicators to compare the intervention effects between the two groups. Results: The data revealed that the balance rhythm dance intervention significantly improved the motor ability and balance function of elderly women in the intervention group (P < 0.01), with statistically significant differences observed. Conclusion: The balance rhythm dance program plays a critical role in promoting the rehabilitation of motor function and balance ability in elderly women, effectively enhancing their quality of life.展开更多
Previous studies have found associations between color discrimination deficits and cognitive impairments besides aging.However,investigations into the microstructural pathology of brain white matter(WM)associated with...Previous studies have found associations between color discrimination deficits and cognitive impairments besides aging.However,investigations into the microstructural pathology of brain white matter(WM)associated with these deficits remain limited.This study aimed to examine the microstructural characteristics of WM in the non-demented population with abnormal color discrimination,utilizing Neurite Orientation Dispersion and Density Imaging(NODDI),and to explore their correlations with cognitive functions and cognition-related plasma biomarkers.The tract-based spatial statistic analysis revealed significant differences in specific brain regions between the abnormal color discrimination group and the healthy controls,characterized by increased isotropic volume fraction and decreased neurite density index and orientation dispersion index.Further analysis of region-of-interest parameters revealed that the isotropic volume fraction in the bilateral anterior thalamic radiation,superior longitudinal fasciculus,cingulum,and forceps minor was significantly correlated with poorer performance on neuropsychological assessments and to varying degrees various cognition-related plasma biomarkers.These findings provide neuroimaging evidence that WM microstructural abnormalities in non-demented individuals with abnormal color discrimination are associated with cognitive dysfunction,potentially serving as early markers for cognitive decline.展开更多
BACKGROUND Young-onset type 2 diabetes mellitus(T2DM)is associated with adverse health outcomes and increased mortality.Vitamin D(VitD)deficiency is likewise linked to various adverse health outcomes and is significan...BACKGROUND Young-onset type 2 diabetes mellitus(T2DM)is associated with adverse health outcomes and increased mortality.Vitamin D(VitD)deficiency is likewise linked to various adverse health outcomes and is significantly associated with lipid metabolism in patients with T2DM.However,little is known regarding the me-chanisms of interaction between VitD and apolipoprotein A1(apoA1)in young-onset T2DM.AIM To evaluate the relationship between VitD and apoA1 levels in patients with young-onset T2DM.METHODS This cross-sectional study was conducted at Zhejiang Provincial People’s Hospital between January 2019 and December 2023.A total of 642 patients with T2DM who aged 18-40 years were included and matched with 642 individuals without diabetes(controls)based on age and sex.No specific intervention was applied,and data were collected from medical records and laboratory tests.The re-lationship between VitD and apoA1 levels was examined using Spearman’s correlation and logistic regression models.RESULTS We found that VitD levels were significantly lower in patients with T2DM compared to controls(15.9 ng/mL vs 17.4 ng/mL,P<0.001),with a notable positive correlation between VitD deficiency and reduced apoA1 levels.Multifactor logistic regression analysis identified that severe VitD deficiency was an independent risk factor for apoA1 in young-onset T2DM patients(odds ratio=3.43,95%confidence interval:1.16-10.20,β=1.23,P=0.026).CONCLUSION Our findings reveal an association between VitD and apoA1 in young-onset T2DM,suggesting that VitD may play a crucial role in metabolic regulation and cardiovascular risk management.展开更多
Osteosarcoma(OS)is the most frequent primary bone sarcomas with high recurrence and poor prognosis.Emerging evidence indicates that membraneless organelles stress granules(SGs),whose assemblies are driven by scaffold ...Osteosarcoma(OS)is the most frequent primary bone sarcomas with high recurrence and poor prognosis.Emerging evidence indicates that membraneless organelles stress granules(SGs),whose assemblies are driven by scaffold protein G3BP1,are extensively involved in tumor,especially in OS.However,how SGs behave and communicate with organelles,particularly nucleoli and mitochondria,during drug challenges remain unknown.This study revealed that chemotherapeutic drugs activated the cysteine protease asparagine endopeptidase(AEP)to specifically cleave the SG core protein G3BP1 at N258/N309 in OS and malignant glioma.tG3BP1-Ns modulated SG dynamics by competitively binding to full-length G3BP1.Strikingly,tG3BP1-Cs,containing a conserved RNA recognition motif CCUBSCUS,sequestered mRNAs of ribosomal proteins and oxidative phosphorylation genes in the nucleoli and mitochondria to repress translation and oxidative stress.Moreover,the inhibition of AEP promoted the tumor-suppressing effect of chemotherapeutic drugs,whereas AEP-cleaved G3BP1 rescue reversed the effect in both OS and glioma models.Cancerous tissues exhibited high levels of AEP and G3BP1 truncations,which were strongly associated with poor prognosis.展开更多

An expanded versio...The theory-based intervention of individualized music has been evaluated clinically and empirically leading to advancement and refinement of an evidence-based protocol, currently in its 5th edition. An expanded version of the protocol was written for professional health care providers with a consumer version tailored for family caregivers. The underlying mid-range theory is presented along with a seminal study that was followed by further research in the United States, Canada, Great Britain, France, Sweden, Norway, Japan and Taiwan. Key studies are summarized. Given its efficacy when implemented by research staff, studies have advanced to testing the intervention under real-life conditions when implemented and evaluated by trained nursing assistants in long-term care facilities and visiting family members. In addition, one study evaluated the implementation of music by family members in the home setting. Initial research focused on agitation as the dependent variable with subsequent research indicating a more holistic response such as positive affect, expressed satisfaction, and meaningful interaction with others. The article advances by describing on-line programs designed to train health care professionals in the assessment, implementation and evaluation of individualized music. In addition, Gerdner has written a story for apicture book intended for children and their families(in press). The story models principles of individualized music to elicit positive memories, reduce anxiety and agitation, and promote communication. Recent studies have revealed that lipid droplets accumulate in neurons after brain injury and evoke lipotoxicity,damaging the neurons.However,how lipids are metabolized by spinal cord neurons after spinal cord injury ...Recent studies have revealed that lipid droplets accumulate in neurons after brain injury and evoke lipotoxicity,damaging the neurons.However,how lipids are metabolized by spinal cord neurons after spinal cord injury remains unclear.Herein,we investigated lipid metabolism by spinal cord neurons after spinal cord injury and identified lipid-lowering compounds to treat spinal cord injury.We found that lipid droplets accumulated in perilesional spinal cord neurons after spinal cord injury in mice.Lipid droplet accumulation could be induced by myelin debris in HT22 cells.Myelin debris degradation by phospholipase led to massive free fatty acid production,which increased lipid droplet synthesis,β-oxidation,and oxidative phosphorylation.Excessive oxidative phosphorylation increased reactive oxygen species generation,which led to increased lipid peroxidation and HT22 cell apoptosis.Bromocriptine was identified as a lipid-lowering compound that inhibited phosphorylation of cytosolic phospholipase A2 by reducing the phosphorylation of extracellular signal-regulated kinases 1/2 in the mitogen-activated protein kinase pathway,thereby inhibiting myelin debris degradation by cytosolic phospholipase A2 and alleviating lipid droplet accumulation in myelin debris-treated HT22 cells.Motor function,lipid droplet accumulation in spinal cord neurons and neuronal survival were all improved in bromocriptine-treated mice after spinal cord injury.The results suggest that bromocriptine can protect neurons from lipotoxic damage after spinal cord injury via the extracellular signal-regulated kinases 1/2-cytosolic phospholipase A2 pathway.展开更多
BACKGROUND Increased homocysteine levels are associated with the risk of cardiovascular disease(CVD)and death.However,their prevention has not been effective in decreasing CVD risk.This study investigated the individu...BACKGROUND Increased homocysteine levels are associated with the risk of cardiovascular disease(CVD)and death.However,their prevention has not been effective in decreasing CVD risk.This study investigated the individual and combined associations of hyperhomocysteinemia and hypertension with incident CVD events and all-cause death in the Chinese elderly population without a history of CVD.METHODS This prospective study was conducted among 1,257 elderly participants(mean age:69 years).A questionnaire survey,physical examinations,and laboratory tests were conducted to collect baseline data.Hyperhomocysteinemia was defined as homocysteine level≥15μmol/L.H-type hypertension was defined as concomitant hypertension and hyperhomocysteinemia.Multivariate Cox regression analysis was used to evaluate individual and combined associations of hyperhomocysteinemia and hypertension with the risks of incident CVD events and all-cause death.RESULTS Over a median of 4.84-year follow-up,hyperhomocysteinemia was independently associated with incident CVD events and all-cause death.The hazard ratios(HRs)were 1.45(95%CI:1.01−2.08)for incident CVD events and 1.55(95%CI:1.04−2.30)for all-cause death.After adjustment for confounding factors,H-type hypertension had the highest HRs for incident CVD events and all-cause death.The fully adjusted HRs were 2.44 for incident CVD events(95%CI:1.28−4.65),2.07 for stroke events(95%CI:1.01−4.29),8.33 for coronary events(95%CI:1.10−63.11),and 2.31 for all-cause death(95%CI:1.15−4.62).CONCLUSIONS Hyperhomocysteinemia was an independent risk factor,and when accompanied by hypertension,it contrib-uted to incident CVD events and all-cause death in the Chinese elderly population without a history of CVD.展开更多

The present study aimed to determine the factors associated with developing postreperfusion hyperkalemia in liver transplantation from DCD. METHODS: One hundred thirty-one consecutive adult pa-tients who underwent orthotopic liver transplantation from DCD were retrospectively studied. Based on serum potassium within 5 minutes after reperfusion, recipients were divided into two groups: hyperkalemia and normokalemia. According to preoperative biopsy results, the DCD graft livers were clas-siifed into ifve categories. Univariate analysis was performed using Chi-square test to identify variables that were signiif-cantly different between two groups. Multivariate logistic regression was used to conifrm the risk factors of developing hyperkalemia and postreperfusion syndrome. Correlation analysis was used to identify the relationship between the serum concentration of potassium within 5 minutes after re-perfusion and the difference in mean arterial pressure values before and within 5 minutes after reperfusion. RESULTS: Twenty-two of 131 liver recipients had hyperkale-mia episodes within 5 minutes after reperfusion. The rate of hyperkalemia was signiifcantly higher in recipients of macro-steatotic DCD graft liver (78.6%,P<0.001) than that in recipi-ents of non-macrosteatotic DCD graft liver. The odds ratio of developing postreperfusion hyperkalemia in recipients of macrosteatotic DCD graft liver was 51.3 (P<0.001). Macroste-atosis in the DCD graft liver was an independent risk factor of developing hyperkalemia within 5 minutes after reperfusion. The highest rate of postreperfusion syndrome also occurred in the recipients with macrosteatotic DCD graft liver (71.4%, P<0.001). A strong relationship existed between the serum po-tassium within 5 minutes after reperfusion and the difference in mean arterial pressure values before and within 5 minutes after reperfusion in macrosteatotic DCD graft liver recipients. CONCLUSION: Macrosteatosis in the DCD graft liver was an independent risk factor of developing hyperkalemia and postreperfusion syndrome in the recipients.展开更多
Recent studies have shown that the microtubule disrupting protein Stathmin 1(STMN1)is differentially expressed in AML patients and healthy control.The aim of this study was to explore the effects and molecular mechani...Recent studies have shown that the microtubule disrupting protein Stathmin 1(STMN1)is differentially expressed in AML patients and healthy control.The aim of this study was to explore the effects and molecular mechanism of STMN1 in AML.Here,the expression of STMN1 in peripheral blood cells(PBMCs)and bone marrow of AML patients and healthy volunteers was detected by RT-PCR and Western blot.STMN1 expression was regulated by transfected with STMN1 overexpressed plasmid or shRNA in two human leukemia cell lines K562 and HL60.Cell proliferation was examined by CCK8 and Edu staining.Annexin V and TUNEL assays were applied to test cell apoptosis.Flow cytometry was used to test the cell cycle distribution.The activation of the PI3K signaling pathway and the expression levels of cell cycle and cell apoptosis-related protein were determined by Western blot.In this study,we found that STMN1 was overexpressed in PBMCs and bone marrow of AML patients.STMN1 expression was closely related to FAB subtypes,risk stratification,disease-free survival,and overall survival of AML.Functional assays showed that overexpression of STMN1 in HL60 and K562 cells enhanced cell proliferation,decreased cell apoptosis,and caused G1 phase arrest.In contrast,suppression of STMN1reduced cell proliferation and enhanced cell apoptosis in both HL60 and K562 cells.Moreover,the PI3K/Akt pathway was activated by STMN1,while suppression of STMN1 dysregulated the PI3K/Akt pathway and upregulating the levels of caspases3 and Bax expression.In conclusion,STMN1 was confirmed to promote the proliferation and inhibit the apoptosis of HL60 and K562 cells by modulating the PI3K/Akt pathway.STMN1 might be a novel molecular target for treating AML.展开更多
Diabetic kidney disease(DKD)is a common complication of diabetes mellitus that contributes to the risk of end-stage kidney disease(ESKD).Wide glycemic var-iations,such as hypoglycemia and hyperglycemia,are broadly fou...Diabetic kidney disease(DKD)is a common complication of diabetes mellitus that contributes to the risk of end-stage kidney disease(ESKD).Wide glycemic var-iations,such as hypoglycemia and hyperglycemia,are broadly found in diabetic patients with DKD and especially ESKD,as a result of impaired renal metabolism.It is essential to monitor glycemia for effective management of DKD.Hemoglobin A1c(HbA1c)has long been considered as the gold standard for monitoring glycemia for>3 months.However,assessment of HbA1c has some bias as it is susceptible to factors such as anemia and liver or kidney dysfunction.Continuous glucose monitoring(CGM)has provided new insights on glycemic assessment and management.CGM directly measures glucose level in interstitial fluid,reports real-time or retrospective glucose concentration,and provides multiple glycemic metrics.It avoids the pitfalls of HbA1c in some contexts,and may serve as a precise alternative to estimation of mean glucose and glycemic variability.Emerging studies have demonstrated the merits of CGM for precise monitoring,which allows fine-tuning of glycemic management in diabetic patients.Therefore,CGM technology has the potential for better glycemic monitoring in DKD patients.More research is needed to explore its application and management in different stages of DKD,including hemodialysis,peritoneal dialysis and kidney transplantation.展开更多
BACKGROUND Helicobacter pylori(H.pylori)infection is a global problem,causing significant morbidity and mortality.Furazolidone is recommended to eradicate H.pylori infections in China owing to the highly associated an...BACKGROUND Helicobacter pylori(H.pylori)infection is a global problem,causing significant morbidity and mortality.Furazolidone is recommended to eradicate H.pylori infections in China owing to the highly associated antibiotic resistance.CASE SUMMARY This article presents two cases of lung injury caused by furazolidone treatment of H.pylori infection and the relevant literature review.Two patients developed symptoms,including fever,cough,and fatigue after receiving a course of furazolidone for H.pylori infection.Chest computed tomography showed bilateral interstitial infiltrates.Laboratory studies revealed elevated blood eosinophil count.After discontinuing furazolidone with or without the use of corticosteroids,the symptoms improved rapidly.A PubMed database literature search revealed three reported cases of lung injury suggestive of furazolidone-induced pulmonary toxicity.CONCLUSION Clinicians should be aware of the side effects associated with the administration of furazolidone to eradicate H.pylori infection.展开更多
AIM:To identify factors contributing to visual improvement after treatment of macular edema(ME)secondary to branch retinal vein occlusion(BRVO),and to assess the interaction between laser therapy and intravitreal rani...AIM:To identify factors contributing to visual improvement after treatment of macular edema(ME)secondary to branch retinal vein occlusion(BRVO),and to assess the interaction between laser therapy and intravitreal ranibizumab(IVR).METHODS:We retrospectively reviewed the medical records of patients who had been treated for BRVO-related ME at our hospital.Records were traceable for at least 12 mo,and evaluated factors included age,sex,medical history,smoking history,treatment methods,foveal hemorrhage,and change in visual acuity.Treatments included laser therapy,IVR,sub-Tenon’s capsule injection of triamcinolone(STTA),a combination,or no intervention.Multivariate logistic regression analysis and interaction terms were used to assess the clinical efficacy of the treatments,and odds ratios(OR)and 95%confidence intervals(CI)were calculated.RESULTS:Seventy-three patients(34 men,39 women;73 eyes)with a mean age of 69.4±12.1 y were included.Patients who underwent IVR monotherapy,laser monotherapy,and STTA+laser had significantly higher best corrected visual acuity at 12 mo compared to baseline(P<0.001,<0.001,and 0.019,respectively).Logistic regression analysis without interaction terms found that IVR was a significant visual acuity recovery factor(adjusted OR:3.89,95%CI:1.25-12.1,P=0.019).Adjusted OR using an interaction model by logistic regression was 16.6(95%CI:2.54-108.47,P=0.003)with IVR treatment,and 8.25(95%CI:1.34-50.57,P=0.023)with laser treatment.No interaction was observed(adjusted OR:0.07,95%CI:0.01-0.75,P=0.029).CONCLUSION:IVR contributes to improvements in visual acuity at 12 mo in ME secondary to BRVO.No interaction is observed between laser therapy and IVR treatments.展开更多
Our previous RNA sequencing study showed that the long non-coding RNA ischemia-related factor Vof-16(lncRNA Vof-16)was upregulated after spinal cord injury,but its precise role in spinal cord injury remains unclear.Bi...Our previous RNA sequencing study showed that the long non-coding RNA ischemia-related factor Vof-16(lncRNA Vof-16)was upregulated after spinal cord injury,but its precise role in spinal cord injury remains unclear.Bioinformatics predictions have indicated that lncRNA Vof-16 may participate in the pathophysiological processes of inflammation and apoptosis.PC12 cells were transfected with a pHBLV-U6-MCS-CMV-ZsGreen-PGK-PURO vector to express an lncRNA Vof-16 knockdown lentivirus and a pHLV-CMVIE-ZsGree-Puro vector to express an lncRNA Vof-16 overexpression lentivirus.The overexpression of lncRNA Vof-16 inhibited PC12 cell survival,proliferation,migration,and neurite extension,whereas lncRNA Vof-16 knockdown lentiviral vector resulted in the opposite effects in PC12 cells.Western blot assay results showed that the overexpression of lncRNA Vof-16 increased the protein expression levels of interleukin 6,tumor necrosis factor-α,and Caspase-3 and decreased Bcl-2 expression levels in PC12 cells.Furthermore,we established rat models of spinal cord injury using the complete transection at T10.Spinal cord injury model rats were injected with the lncRNA Vof-16 knockdown or overexpression lentiviral vectors immediately after injury.At 7 days after spinal cord injury,rats treated with lncRNA Vof-16 knockdown displayed increased neuronal survival and enhanced axonal extension.At 8 weeks after spinal cord injury,rats treated with the lncRNA Vof-16 knockdown lentiviral vector displayed improved neurological function in the hind limb.Notably,lncRNA Vof-16 knockdown injection increased Bcl-2 expression and decreased tumor necrosis factor-αand Caspase-3 expression in treated animals.Rats treated with the lncRNA Vof-16 overexpression lentiviral vector displayed opposite trends.These findings suggested that lncRNA Vof-16 is associated with the regulation of inflammation and apoptosis.The inhibition of lncRNA Vof-16 may be useful for promoting nerve regeneration and functional recovery after spinal cord injury.The experiments were approved by the Institutional Animal Care and Use Committee of Guangdong Medical University,China.展开更多

BACKGROUND Natural killer(NK)/T cell lymphoma is a rare and highly aggressive malignant tumor,and is a special form of non-Hodgkin's lymphoma.Although extranodal involvement is frequently found in tissues such as ...BACKGROUND Natural killer(NK)/T cell lymphoma is a rare and highly aggressive malignant tumor,and is a special form of non-Hodgkin's lymphoma.Although extranodal involvement is frequently found in tissues such as the skin,testicular and gastrointestinal tract etc,its presence in skeletal muscle has scarcely been reported in the literature.CASE SUMMARY We report a case of extranodal NK/T cell lymphoma with muscle swelling as the first clinical manifestation.A 42-year-old man,who initially presented with localized swelling in the double lower extremities,demonstrated gradual facial and eyelid swelling,and his imaging results showed multiple sites of muscle damage throughout the body.The final pathological results suggested NK/T cell lymphoma,and immunohistochemistry showed CD20(-),CD3(+),CD30(+),CD56(-),EBER(+),Ki67(60%),TIA-1(+)and CD68(±)staining.The muscle swelling significantly improved after treatment with chemotherapy regimens.CONCLUSION This disease is difficult to diagnose and highly invasive,and should be included in the differential diagnosis of unexplained muscle swelling.展开更多

文摘Economical,stable,and corrosion-resistant catalytic electrodes are still urgently needed for the oxygen evolution reaction(OER)in water and seawater.Herein,a mild electroless plating strategy is used to achieve large-scale preparation of the“integrated”phosphorus-based precatalyst(FeP-NiP)on nickel foam(NF),which is in situ reconstructed into a highly active and corrosion-resistant(Fe)NiOOH phase for OER.The interaction between phosphate anions(PO_(x)^(y-))and iron ions(Fe^(3+))tunes the electronic structure of the catalytic phase to further enhance OER kinetics.The integrated FeP-NiP@NF electrode exhibits low overpotentials for OER in alkaline water/seawater,requiring only 275/289,320/336,and 349/358 mV to reach 0.1,0.5,and 1.0 A cm^(−2),respectively.The in situ reconstructed PO_(x)^(y-)anion electrostatically repels Cl−in seawater electrolytes,allowing stable operation for over 7 days at 1.0 A cm^(−2) in extreme electrolytes(1.0 M KOH+seawater and 6.0 M KOH+seawater),demonstrating industrial-level stability.This study overcomes the complex synthesis limitations of P-based materials through innovative material design,opening new avenues for electrochemical energy conversion.
文摘BACKGROUND Type 2 diabetes mellitus(T2DM)often leads to vascular complications,such as albuminuria.The role of insulin autoantibodies(IAA)and their interaction with D-dimer in this context remains unclear.AIM To investigate the characteristics of IAA and its effect on albuminuria in T2DM patients.METHODS We retrospectively analyzed clinical data from 115 T2DM patients with positive IAA induced by exogenous insulin,and 115 age-and sex-matched IAA-negative T2DM patients as controls.Propensity scores were calculated using multivariate logistic regression.Key variables were selected using the least absolute shrinkage and selection operator(LASSO)algorithm.We constructed a prediction model and analyzed the association between IAA and albuminuria based on demographic and laboratory parameters.RESULTS The IAA-positive group had significantly higher D-dimer levels[0.30(0.19-0.55)mg/L vs 0.21(0.19-0.33)mg/L,P=0.008]and plasma insulin levels[39.1(12.0-102.7)μU/mL vs 9.8(5.5-17.6)μU/mL,P<0.001]compared to the IAA-negative group.Increases in the insulin dose per weight ratio,diabetes duration,and urinary albumin-to-creatinine ratio(UACR)were observed but did not reach statistical significance.The LASSO model identified plasma insulin and D-dimer as key factors with larger coefficients.D-dimer was significantly associated with UACR in the total and IAA-positive groups but not in the IAA-negative group.The odds ratio for D-dimer elevation(>0.5 g/L)was 2.88(95%confidence interval:1.17-7.07)in the IAA-positive group(P interaction<0.05).CONCLUSION D-dimer elevation is an independent risk factor for abnormal albuminuria and interacts with IAA in the development of abnormal albuminuria in T2DM patients.

文摘Diabetic foot ulcers(DFUs)represents a significant public health issue,with a rising global prevalence and severe potential complications including amputation.Traditional treatments often fall short due to various limitations such as high recurrence rates and extensive resource utilization.This editorial explores the innovative use of acellular fish skin grafts as a transformative approach in DFU management.Recent studies and a detailed case report highlight the efficacy of acellular fish skin grafts in accelerating wound closure,reducing dressing changes,and enhancing patient outcomes with a lower socio-economic burden.Despite their promise,challenges such as limited availability,patient acceptance,and the need for further research persist.Addressing these through more extensive randomized controlled trials and fostering a multidisciplinary treatment approach may optimize DFU care and reduce the global health burden associated with these complex wounds.

文摘Background:Osteoarthritis(OA)is a long-term degenerative joint disease worsen-ing over time.Aging and chondrocyte senescence contribute to OA progression.MicroRNAs have been confirmed to regulate different cellular processes.They con-tribute to OA pathology and may help to identify novel biomarkers and therapies for OA.Methods:This study used bioinformatics and experimental investigations to analyze and validate differentially expressed miRNAs in OA that might affect chondrocyte apoptosis and senescence.Results:miR-6779 was found to be significantly down-regulated in OA.Seventy-six of the predicted and miR-6779 targeted genes and the OA-associated disease genes overlapped,and these were enriched in cell proliferation,cell apoptosis,and cell cycle.miR-6779 overexpression remarkably attenuated IL-1βeffects on chon-drocytes by reducing MMP3 and MMP13 levels,promoting cell apoptosis,suppress-ing cell senescence,and increasing caspase-3,caspase-9 and reducing P16 and P21 levels.miR-6779 targeted inhibition of X-linked inhibitor of apoptosis protein(XIAP)expression.XIAP knockdown partially improved IL-1β-induced chondrocyte senes-cence and dysfunction.Lastly,when co-transfected with a miR-6779 agomir,the XIAP overexpression vector partially attenuated the effects of miR-6779 overexpression on chondrocytes;miR-6779 improved IL-1β-induced senescence and dysfunction in chondrocytes through targeting XIAP.Conclusion:miR-6779 is down-regulated,and XIAP is up-regulated in OA cartilage and IL-1β-treated chondrocytes.miR-6779 inhibits XIAP expression,thereby promot-ing senescent chondrocyte cell apoptosis and reducing chondrocyte senescence and ECM loss through XIAP.

文摘Objective: To investigate the effect of a self-developed balance rhythm dance program on the rehabilitation of motor function and the reduction of fall risk in elderly women with diminished balance function. Methods: Fifty elderly women with reduced balance function, admitted to the Qingbar Elderly Care Center of Chongqing Medical University from December 2022 to December 2023, were randomly selected and divided into two groups. The aerobic exercise group (25 patients) received traditional treatment and rehabilitation nursing, while the balance rhythm dance intervention group (25 patients) received the balance rhythm dance intervention in addition to traditional treatment and rehabilitation nursing. The Unified Parkinson’s Disease Rating Scale (UPDRS) and Berg Balance Scale (BBS) were used as evaluation indicators to compare the intervention effects between the two groups. Results: The data revealed that the balance rhythm dance intervention significantly improved the motor ability and balance function of elderly women in the intervention group (P < 0.01), with statistically significant differences observed. Conclusion: The balance rhythm dance program plays a critical role in promoting the rehabilitation of motor function and balance ability in elderly women, effectively enhancing their quality of life.

文摘Previous studies have found associations between color discrimination deficits and cognitive impairments besides aging.However,investigations into the microstructural pathology of brain white matter(WM)associated with these deficits remain limited.This study aimed to examine the microstructural characteristics of WM in the non-demented population with abnormal color discrimination,utilizing Neurite Orientation Dispersion and Density Imaging(NODDI),and to explore their correlations with cognitive functions and cognition-related plasma biomarkers.The tract-based spatial statistic analysis revealed significant differences in specific brain regions between the abnormal color discrimination group and the healthy controls,characterized by increased isotropic volume fraction and decreased neurite density index and orientation dispersion index.Further analysis of region-of-interest parameters revealed that the isotropic volume fraction in the bilateral anterior thalamic radiation,superior longitudinal fasciculus,cingulum,and forceps minor was significantly correlated with poorer performance on neuropsychological assessments and to varying degrees various cognition-related plasma biomarkers.These findings provide neuroimaging evidence that WM microstructural abnormalities in non-demented individuals with abnormal color discrimination are associated with cognitive dysfunction,potentially serving as early markers for cognitive decline.

文摘BACKGROUND Young-onset type 2 diabetes mellitus(T2DM)is associated with adverse health outcomes and increased mortality.Vitamin D(VitD)deficiency is likewise linked to various adverse health outcomes and is significantly associated with lipid metabolism in patients with T2DM.However,little is known regarding the me-chanisms of interaction between VitD and apolipoprotein A1(apoA1)in young-onset T2DM.AIM To evaluate the relationship between VitD and apoA1 levels in patients with young-onset T2DM.METHODS This cross-sectional study was conducted at Zhejiang Provincial People’s Hospital between January 2019 and December 2023.A total of 642 patients with T2DM who aged 18-40 years were included and matched with 642 individuals without diabetes(controls)based on age and sex.No specific intervention was applied,and data were collected from medical records and laboratory tests.The re-lationship between VitD and apoA1 levels was examined using Spearman’s correlation and logistic regression models.RESULTS We found that VitD levels were significantly lower in patients with T2DM compared to controls(15.9 ng/mL vs 17.4 ng/mL,P<0.001),with a notable positive correlation between VitD deficiency and reduced apoA1 levels.Multifactor logistic regression analysis identified that severe VitD deficiency was an independent risk factor for apoA1 in young-onset T2DM patients(odds ratio=3.43,95%confidence interval:1.16-10.20,β=1.23,P=0.026).CONCLUSION Our findings reveal an association between VitD and apoA1 in young-onset T2DM,suggesting that VitD may play a crucial role in metabolic regulation and cardiovascular risk management.

文摘Osteosarcoma(OS)is the most frequent primary bone sarcomas with high recurrence and poor prognosis.Emerging evidence indicates that membraneless organelles stress granules(SGs),whose assemblies are driven by scaffold protein G3BP1,are extensively involved in tumor,especially in OS.However,how SGs behave and communicate with organelles,particularly nucleoli and mitochondria,during drug challenges remain unknown.This study revealed that chemotherapeutic drugs activated the cysteine protease asparagine endopeptidase(AEP)to specifically cleave the SG core protein G3BP1 at N258/N309 in OS and malignant glioma.tG3BP1-Ns modulated SG dynamics by competitively binding to full-length G3BP1.Strikingly,tG3BP1-Cs,containing a conserved RNA recognition motif CCUBSCUS,sequestered mRNAs of ribosomal proteins and oxidative phosphorylation genes in the nucleoli and mitochondria to repress translation and oxidative stress.Moreover,the inhibition of AEP promoted the tumor-suppressing effect of chemotherapeutic drugs,whereas AEP-cleaved G3BP1 rescue reversed the effect in both OS and glioma models.Cancerous tissues exhibited high levels of AEP and G3BP1 truncations,which were strongly associated with poor prognosis.

文摘Diabetic gastrointestinal autonomic neuropathy(DGAN)is a common and debilitating complication of diabetes,characterized by autonomic dysfunction in the gastrointestinal system.The complex pathophysiology of DGAN involves neuronal injury that is intrinsically linked to gut dysbiosis.Multiple factors,including hyperglycemia,oxidative stress,and inflammation,significantly contribute to neuronal damage,manifesting as symptoms such as delayed gastric emptying,diarrhea,and constipation.Recent studies have demonstrated that patients with diabetes experience substantial alterations in gut microbiota composition,potentially exacerbating gastrointestinal symptoms.Microbial metabolites may modulate neurotransmitter synthesis and release,directly affecting autonomic nerve function,while dysbiosis amplifies oxidative stress and inflammation,further compromising the enteric nervous system and worsening DGAN.Advances in multi-omics technologies now provide deeper insights into molecular mechanisms of DGAN and its interactions with microbiota.Early diagnosis leveraging biomarkers,gut microbiota analysis,and advanced imaging promises more effective interventions.Emerging therapeutic strategies targeting oxidative stress,inflammation,and gut microbiota represent promising approaches for managing DGAN.Future research should focus on large-scale,multi-ethnic studies and therapies targeting specific microbial metabolites to refine diagnosis and treatment approaches.
文摘The theory-based intervention of individualized music has been evaluated clinically and empirically leading to advancement and refinement of an evidence-based protocol, currently in its 5th edition. An expanded version of the protocol was written for professional health care providers with a consumer version tailored for family caregivers. The underlying mid-range theory is presented along with a seminal study that was followed by further research in the United States, Canada, Great Britain, France, Sweden, Norway, Japan and Taiwan. Key studies are summarized. Given its efficacy when implemented by research staff, studies have advanced to testing the intervention under real-life conditions when implemented and evaluated by trained nursing assistants in long-term care facilities and visiting family members. In addition, one study evaluated the implementation of music by family members in the home setting. Initial research focused on agitation as the dependent variable with subsequent research indicating a more holistic response such as positive affect, expressed satisfaction, and meaningful interaction with others. The article advances by describing on-line programs designed to train health care professionals in the assessment, implementation and evaluation of individualized music. In addition, Gerdner has written a story for apicture book intended for children and their families(in press). The story models principles of individualized music to elicit positive memories, reduce anxiety and agitation, and promote communication. The article concludes with implications for future research.

文摘Recent studies have revealed that lipid droplets accumulate in neurons after brain injury and evoke lipotoxicity,damaging the neurons.However,how lipids are metabolized by spinal cord neurons after spinal cord injury remains unclear.Herein,we investigated lipid metabolism by spinal cord neurons after spinal cord injury and identified lipid-lowering compounds to treat spinal cord injury.We found that lipid droplets accumulated in perilesional spinal cord neurons after spinal cord injury in mice.Lipid droplet accumulation could be induced by myelin debris in HT22 cells.Myelin debris degradation by phospholipase led to massive free fatty acid production,which increased lipid droplet synthesis,β-oxidation,and oxidative phosphorylation.Excessive oxidative phosphorylation increased reactive oxygen species generation,which led to increased lipid peroxidation and HT22 cell apoptosis.Bromocriptine was identified as a lipid-lowering compound that inhibited phosphorylation of cytosolic phospholipase A2 by reducing the phosphorylation of extracellular signal-regulated kinases 1/2 in the mitogen-activated protein kinase pathway,thereby inhibiting myelin debris degradation by cytosolic phospholipase A2 and alleviating lipid droplet accumulation in myelin debris-treated HT22 cells.Motor function,lipid droplet accumulation in spinal cord neurons and neuronal survival were all improved in bromocriptine-treated mice after spinal cord injury.The results suggest that bromocriptine can protect neurons from lipotoxic damage after spinal cord injury via the extracellular signal-regulated kinases 1/2-cytosolic phospholipase A2 pathway.

文摘BACKGROUND Increased homocysteine levels are associated with the risk of cardiovascular disease(CVD)and death.However,their prevention has not been effective in decreasing CVD risk.This study investigated the individual and combined associations of hyperhomocysteinemia and hypertension with incident CVD events and all-cause death in the Chinese elderly population without a history of CVD.METHODS This prospective study was conducted among 1,257 elderly participants(mean age:69 years).A questionnaire survey,physical examinations,and laboratory tests were conducted to collect baseline data.Hyperhomocysteinemia was defined as homocysteine level≥15μmol/L.H-type hypertension was defined as concomitant hypertension and hyperhomocysteinemia.Multivariate Cox regression analysis was used to evaluate individual and combined associations of hyperhomocysteinemia and hypertension with the risks of incident CVD events and all-cause death.RESULTS Over a median of 4.84-year follow-up,hyperhomocysteinemia was independently associated with incident CVD events and all-cause death.The hazard ratios(HRs)were 1.45(95%CI:1.01−2.08)for incident CVD events and 1.55(95%CI:1.04−2.30)for all-cause death.After adjustment for confounding factors,H-type hypertension had the highest HRs for incident CVD events and all-cause death.The fully adjusted HRs were 2.44 for incident CVD events(95%CI:1.28−4.65),2.07 for stroke events(95%CI:1.01−4.29),8.33 for coronary events(95%CI:1.10−63.11),and 2.31 for all-cause death(95%CI:1.15−4.62).CONCLUSIONS Hyperhomocysteinemia was an independent risk factor,and when accompanied by hypertension,it contrib-uted to incident CVD events and all-cause death in the Chinese elderly population without a history of CVD.
文摘BACKGROUND: With the increasing use of donation after cardiac death (DCD), especially of the graft liver with steatosis or other pathological changes, the frequency of postreperfu-sion hyperkalemia in liver transplantation has increased sig-niifcantly. The present study aimed to determine the factors associated with developing postreperfusion hyperkalemia in liver transplantation from DCD. METHODS: One hundred thirty-one consecutive adult pa-tients who underwent orthotopic liver transplantation from DCD were retrospectively studied. Based on serum potassium within 5 minutes after reperfusion, recipients were divided into two groups: hyperkalemia and normokalemia. According to preoperative biopsy results, the DCD graft livers were clas-siifed into ifve categories. Univariate analysis was performed using Chi-square test to identify variables that were signiif-cantly different between two groups. Multivariate logistic regression was used to conifrm the risk factors of developing hyperkalemia and postreperfusion syndrome. Correlation analysis was used to identify the relationship between the serum concentration of potassium within 5 minutes after re-perfusion and the difference in mean arterial pressure values before and within 5 minutes after reperfusion. RESULTS: Twenty-two of 131 liver recipients had hyperkale-mia episodes within 5 minutes after reperfusion. The rate of hyperkalemia was signiifcantly higher in recipients of macro-steatotic DCD graft liver (78.6%,P<0.001) than that in recipi-ents of non-macrosteatotic DCD graft liver. The odds ratio of developing postreperfusion hyperkalemia in recipients of macrosteatotic DCD graft liver was 51.3 (P<0.001). Macroste-atosis in the DCD graft liver was an independent risk factor of developing hyperkalemia within 5 minutes after reperfusion. The highest rate of postreperfusion syndrome also occurred in the recipients with macrosteatotic DCD graft liver (71.4%, P<0.001). A strong relationship existed between the serum po-tassium within 5 minutes after reperfusion and the difference in mean arterial pressure values before and within 5 minutes after reperfusion in macrosteatotic DCD graft liver recipients. CONCLUSION: Macrosteatosis in the DCD graft liver was an independent risk factor of developing hyperkalemia and postreperfusion syndrome in the recipients.

文摘Recent studies have shown that the microtubule disrupting protein Stathmin 1(STMN1)is differentially expressed in AML patients and healthy control.The aim of this study was to explore the effects and molecular mechanism of STMN1 in AML.Here,the expression of STMN1 in peripheral blood cells(PBMCs)and bone marrow of AML patients and healthy volunteers was detected by RT-PCR and Western blot.STMN1 expression was regulated by transfected with STMN1 overexpressed plasmid or shRNA in two human leukemia cell lines K562 and HL60.Cell proliferation was examined by CCK8 and Edu staining.Annexin V and TUNEL assays were applied to test cell apoptosis.Flow cytometry was used to test the cell cycle distribution.The activation of the PI3K signaling pathway and the expression levels of cell cycle and cell apoptosis-related protein were determined by Western blot.In this study,we found that STMN1 was overexpressed in PBMCs and bone marrow of AML patients.STMN1 expression was closely related to FAB subtypes,risk stratification,disease-free survival,and overall survival of AML.Functional assays showed that overexpression of STMN1 in HL60 and K562 cells enhanced cell proliferation,decreased cell apoptosis,and caused G1 phase arrest.In contrast,suppression of STMN1reduced cell proliferation and enhanced cell apoptosis in both HL60 and K562 cells.Moreover,the PI3K/Akt pathway was activated by STMN1,while suppression of STMN1 dysregulated the PI3K/Akt pathway and upregulating the levels of caspases3 and Bax expression.In conclusion,STMN1 was confirmed to promote the proliferation and inhibit the apoptosis of HL60 and K562 cells by modulating the PI3K/Akt pathway.STMN1 might be a novel molecular target for treating AML.

文摘Diabetic kidney disease(DKD)is a common complication of diabetes mellitus that contributes to the risk of end-stage kidney disease(ESKD).Wide glycemic var-iations,such as hypoglycemia and hyperglycemia,are broadly found in diabetic patients with DKD and especially ESKD,as a result of impaired renal metabolism.It is essential to monitor glycemia for effective management of DKD.Hemoglobin A1c(HbA1c)has long been considered as the gold standard for monitoring glycemia for>3 months.However,assessment of HbA1c has some bias as it is susceptible to factors such as anemia and liver or kidney dysfunction.Continuous glucose monitoring(CGM)has provided new insights on glycemic assessment and management.CGM directly measures glucose level in interstitial fluid,reports real-time or retrospective glucose concentration,and provides multiple glycemic metrics.It avoids the pitfalls of HbA1c in some contexts,and may serve as a precise alternative to estimation of mean glucose and glycemic variability.Emerging studies have demonstrated the merits of CGM for precise monitoring,which allows fine-tuning of glycemic management in diabetic patients.Therefore,CGM technology has the potential for better glycemic monitoring in DKD patients.More research is needed to explore its application and management in different stages of DKD,including hemodialysis,peritoneal dialysis and kidney transplantation.
文摘BACKGROUND Helicobacter pylori(H.pylori)infection is a global problem,causing significant morbidity and mortality.Furazolidone is recommended to eradicate H.pylori infections in China owing to the highly associated antibiotic resistance.CASE SUMMARY This article presents two cases of lung injury caused by furazolidone treatment of H.pylori infection and the relevant literature review.Two patients developed symptoms,including fever,cough,and fatigue after receiving a course of furazolidone for H.pylori infection.Chest computed tomography showed bilateral interstitial infiltrates.Laboratory studies revealed elevated blood eosinophil count.After discontinuing furazolidone with or without the use of corticosteroids,the symptoms improved rapidly.A PubMed database literature search revealed three reported cases of lung injury suggestive of furazolidone-induced pulmonary toxicity.CONCLUSION Clinicians should be aware of the side effects associated with the administration of furazolidone to eradicate H.pylori infection.
文摘AIM:To identify factors contributing to visual improvement after treatment of macular edema(ME)secondary to branch retinal vein occlusion(BRVO),and to assess the interaction between laser therapy and intravitreal ranibizumab(IVR).METHODS:We retrospectively reviewed the medical records of patients who had been treated for BRVO-related ME at our hospital.Records were traceable for at least 12 mo,and evaluated factors included age,sex,medical history,smoking history,treatment methods,foveal hemorrhage,and change in visual acuity.Treatments included laser therapy,IVR,sub-Tenon’s capsule injection of triamcinolone(STTA),a combination,or no intervention.Multivariate logistic regression analysis and interaction terms were used to assess the clinical efficacy of the treatments,and odds ratios(OR)and 95%confidence intervals(CI)were calculated.RESULTS:Seventy-three patients(34 men,39 women;73 eyes)with a mean age of 69.4±12.1 y were included.Patients who underwent IVR monotherapy,laser monotherapy,and STTA+laser had significantly higher best corrected visual acuity at 12 mo compared to baseline(P<0.001,<0.001,and 0.019,respectively).Logistic regression analysis without interaction terms found that IVR was a significant visual acuity recovery factor(adjusted OR:3.89,95%CI:1.25-12.1,P=0.019).Adjusted OR using an interaction model by logistic regression was 16.6(95%CI:2.54-108.47,P=0.003)with IVR treatment,and 8.25(95%CI:1.34-50.57,P=0.023)with laser treatment.No interaction was observed(adjusted OR:0.07,95%CI:0.01-0.75,P=0.029).CONCLUSION:IVR contributes to improvements in visual acuity at 12 mo in ME secondary to BRVO.No interaction is observed between laser therapy and IVR treatments.

文摘Our previous RNA sequencing study showed that the long non-coding RNA ischemia-related factor Vof-16(lncRNA Vof-16)was upregulated after spinal cord injury,but its precise role in spinal cord injury remains unclear.Bioinformatics predictions have indicated that lncRNA Vof-16 may participate in the pathophysiological processes of inflammation and apoptosis.PC12 cells were transfected with a pHBLV-U6-MCS-CMV-ZsGreen-PGK-PURO vector to express an lncRNA Vof-16 knockdown lentivirus and a pHLV-CMVIE-ZsGree-Puro vector to express an lncRNA Vof-16 overexpression lentivirus.The overexpression of lncRNA Vof-16 inhibited PC12 cell survival,proliferation,migration,and neurite extension,whereas lncRNA Vof-16 knockdown lentiviral vector resulted in the opposite effects in PC12 cells.Western blot assay results showed that the overexpression of lncRNA Vof-16 increased the protein expression levels of interleukin 6,tumor necrosis factor-α,and Caspase-3 and decreased Bcl-2 expression levels in PC12 cells.Furthermore,we established rat models of spinal cord injury using the complete transection at T10.Spinal cord injury model rats were injected with the lncRNA Vof-16 knockdown or overexpression lentiviral vectors immediately after injury.At 7 days after spinal cord injury,rats treated with lncRNA Vof-16 knockdown displayed increased neuronal survival and enhanced axonal extension.At 8 weeks after spinal cord injury,rats treated with the lncRNA Vof-16 knockdown lentiviral vector displayed improved neurological function in the hind limb.Notably,lncRNA Vof-16 knockdown injection increased Bcl-2 expression and decreased tumor necrosis factor-αand Caspase-3 expression in treated animals.Rats treated with the lncRNA Vof-16 overexpression lentiviral vector displayed opposite trends.These findings suggested that lncRNA Vof-16 is associated with the regulation of inflammation and apoptosis.The inhibition of lncRNA Vof-16 may be useful for promoting nerve regeneration and functional recovery after spinal cord injury.The experiments were approved by the Institutional Animal Care and Use Committee of Guangdong Medical University,China.
文摘Objective:The objective of the study is to construct a training course for physical restraint(PR)evidence‑based practice project and apply it to verify its effect.Methods:A total of 162 nurses from five departments of a general hospital in Beijing were trained to compare the PR knowledge,attitudes,and behaviors of nurses before and after training.Results:The nurses were satisfied with the overall curriculum;the total scores of PR’knowledge,attitude and behavior of nurses after training were higher than before,and the difference was statistically significant(P<0.05);the total scores of nurses who received evidence‑related training before this training were higher than those of nurses who did not receive relevant training,and the difference was statistically significant(P<0.05).Conclusion:Continuing education can effectively improve nurses’knowledge,attitude,and behavior level of PR,ensure patient safety,and promote physical and psychological rehabilitation of patients.

文摘BACKGROUND Natural killer(NK)/T cell lymphoma is a rare and highly aggressive malignant tumor,and is a special form of non-Hodgkin's lymphoma.Although extranodal involvement is frequently found in tissues such as the skin,testicular and gastrointestinal tract etc,its presence in skeletal muscle has scarcely been reported in the literature.CASE SUMMARY We report a case of extranodal NK/T cell lymphoma with muscle swelling as the first clinical manifestation.A 42-year-old man,who initially presented with localized swelling in the double lower extremities,demonstrated gradual facial and eyelid swelling,and his imaging results showed multiple sites of muscle damage throughout the body.The final pathological results suggested NK/T cell lymphoma,and immunohistochemistry showed CD20(-),CD3(+),CD30(+),CD56(-),EBER(+),Ki67(60%),TIA-1(+)and CD68(±)staining.The muscle swelling significantly improved after treatment with chemotherapy regimens.CONCLUSION This disease is difficult to diagnose and highly invasive,and should be included in the differential diagnosis of unexplained muscle swelling.
