文摘Spinal cord injury represents a severe form of central nervous system trauma for which effective treatments remain limited.Microglia is the resident immune cells of the central nervous system, play a critical role in spinal cord injury.Previous studies have shown that microglia can promote neuronal survival by phagocytosing dead cells and debris and by releasing neuroprotective and anti-inflammatory factors. However, excessive activation of microglia can lead to persistent inflammation and contribute to the formation of glial scars, which hinder axonal regeneration.Despite this, the precise role and mechanisms of microglia during the acute phase of spinal cord injury remain controversial and poorly understood. To elucidate the role of microglia in spinal cord injury,we employed the colony-stimulating factor 1 receptor inhibitor PLX5622 to deplete microglia. We observed that sustained depletion of microglia resulted in an expansion of the lesion area, downregulation of brain-derived neurotrophic factor, and impaired functional recovery after spinal cord injury.Next,we generated a transgenic mouse line with conditional overexpression of brain-derived neurotrophic factor specifically in microglia. We found that brain-derived neurotrophic factor overexpression in microglia increased angiogenesis and blood flow following spinal cord injury and facilitated the recovery of hindlimb motor function.Additionally,brain-derived neurotrophic factor overexpression in microglia reduced inflammation and neuronal apoptosis during the acute phase of spinal cord injury.Furthermore, through using specific transgenic mouse lines,TMEM119,and the colony-stimulating factor 1 receptor inhibitor PLX73086,we demonstrated that the neuroprotective effects were predominantly due to brain-derived neurotrophic factor overexpression in microglia rather than macrophages. In conclusion, our findings suggest the critical role of microglia in the formation of protective glial scars.Depleting microglia is detrimental to recovery of spinal cord injury, whereas targeting brain-derived neurotrophic factor overexpression in microglia represents a promising and novel therapeutic strategy to enhance motor function recovery in patients with spinal cord injury.
文摘In this paper,we present a circuit model of single-quantum-well InGaN/GaN light-emitting diodes based on the standard rate equations. Two rate equations describe carrier transport processes occurring in sep-arate confinement heterostructure and quantum well respectively, and the third equation describes the varied photons in quantum well.By using the presented model, impacts of quantum well thickness on the static and dynamic performances are investigated.Simulated results show that LED with 4 nm well exhibits better lightcurrent(L-I)performance,but LED with 3 nm well presents wider 3 dB modulation bandwidth. It reveals that high carrier density in quantum well is detrimental to the static performance, but beneficial to the dynamic performance.

文摘The production of light(anti-)nuclei in high-energy collisions has long posed an apparent paradox:How can loosely bound systems such as the anti-deuteron with a binding energy of only 2.23 MeV be formed and survive in the extreme hot and dense hadronic environment emerging from proton–proton(pp)and heavy-ion collisions, where characteristic thermal energies exceed 100 MeV?A new femtoscopy analysis published on Nature[1]by the ALICE Collaboration at the Large Hadron Collider(LHC)delivers the clearest answer to date.

文摘Vestibular hair cells(HCs)in the inner ear, crucial for balance and spatial orientation, are classified into type I and type II subtypes, but the mechanisms regulating their differentiation remain unclear. In this study,we examined the role of Pou4f3,an important transcription factor,in vestibular HC differentiation using Pou4f3^(DTR/DTR)(deficient)and Pou4f3CreER/CreER(knockout)mouse models. In Pou4f3-deficient mice, the HC number decreased, and immature HCs failed to develop type I characteristics, indicating a developmental arrest. While type II HCs differentiated normally,Pou4f3 deficiency disrupted HC bundle formation and cell polarity.Findings from knockout models further confirmed the essential role of Pou4f3 in vestibular HC subtype specification. This study underscores the critical role of Pou4f3 in determining vestibular HC subtypes and offers insights into potential strategies for restoring vestibular function through HC regeneration.

文摘Diabetic retinopathy is a prominent cause of blindness in adults, with early retinal ganglion cell loss contributing to visual dysfunction or blindness. In the brain, defects inγ-aminobutyric acid synaptic transmission are associated with pathophysiological and neurodegenerative disorders, whereas glucagon-like peptide-1 has demonstrated neuroprotective effects. However,it is not yet clear whether diabetes causes alterations in inhibitory input to retinal ganglion cells and whether and how glucagon-like peptide-1 protects against neurodegeneration in the diabetic retina through regulating inhibitory synaptic transmission to retinal ganglion cells. In the present study,we used the patch-clamp technique to recordγ-aminobutyric acid subtype A receptor-mediated miniature inhibitory postsynaptic currents in retinal ganglion cells from streptozotocin-induced diabetes model rats.We found that early diabetes(4 weeks of hyperglycemia)decreased the frequency of GABAergic miniature inhibitory postsynaptic currents in retinal ganglion cells without altering their amplitude, suggesting a reduction in the spontaneous release ofγ-aminobutyric acid to retinal ganglion cells.Topical administration of glucagon-like peptide-1 eyedrops over a period of 2 weeks effectively countered the hyperglycemia-induced downregulation of GABAergic mIPSC frequency, subsequently enhancing the survival of retinal ganglion cells.Concurrently, the protective effects of glucagon-like peptide-1 on retinal ganglion cells in diabetic rats were eliminated by topical administration of exendin-9-39,a specific glucagon-like peptide-1 receptor antagonist,or SR95531,a specific antagonist of theγ-aminobutyric acid subtype A receptor.Furthermore, extracellular perfusion of glucagon-like peptide-1 was found to elevate the frequencies of GABAergic miniature inhibitory postsynaptic currents in both ON-and OFF-type retinal ganglion cells. This elevation was shown to be mediated by activation of the phosphatidylinositol-phospholipase C/inositol 1,4,5-trisphosphate receptor/Ca2+/protein kinase C signaling pathway downstream of glucagon-like peptide-1 receptor activation. Moreover, multielectrode array recordings revealed that glucagon-like peptide-1 functionally augmented the photoresponses of ON-type retinal ganglion cells.Optomotor response tests demonstrated that diabetic rats exhibited reductions in visual acuity and contrast sensitivity that were significantly ameliorated by topical administration of glucagon-like peptide-1.These results suggest that glucagon-like peptide-1 facilitates the release ofγ-aminobutyric acid onto retinal ganglion cells through the activation of glucagon-like peptide-1 receptor, leading to the de-excitation of retinal ganglion cell circuits and the inhibition of excitotoxic processes associated with diabetic retinopathy.Collectively, our findings indicate that theγ-aminobutyric acid system has potential as a therapeutic target for mitigating early-stage diabetic retinopathy.Furthermore, the topical administration of glucagon-like peptide-1 eyedrops represents a non-invasive and effective treatment approach for managing early-stage diabetic retinopathy.

文摘Unmanned Aerial Vehicle(UAV)plays a prominent role in various fields, and autonomous navigation is a crucial component of UAV intelligence.Deep Reinforcement Learning(DRL)has expanded the research avenues for addressing challenges in autonomous navigation.Nonetheless, challenges persist, including getting stuck in local optima, consuming excessive computations during action space exploration, and neglecting deterministic experience. This paper proposes a noise-driven enhancement strategy. In accordance with the overall learning phases,a global noise control method is designed, while a differentiated local noise control method is developed by analyzing the exploration demands of four typical situations encountered by UAV during navigation. Both methods are integrated into a dual-model for noise control to regulate action space exploration.Furthermore, noise dual experience replay buffers are designed to optimize the rational utilization of both deterministic and noisy experience. In uncertain environments, based on the Twin Delay Deep Deterministic Policy Gradient(TD3)algorithm with Long Short-Term Memory(LSTM)network and Priority Experience Replay(PER),a Noise-Driven Enhancement Priority Memory TD3(NDE-PMTD3)is developed. We established a simulation environment to compare different algorithms, and the performance of the algorithms is analyzed in various scenarios. The training results indicate that the proposed algorithm accelerates the convergence speed and enhances the convergence stability. In test experiments, the proposed algorithm successfully and efficiently performs autonomous navigation tasks in diverse environments, demonstrating superior generalization results.

文摘The micro-riblet structures have been demonstrated effective in controlling the Total Pressure Loss(TPL)of aero-engine blades. However, due to the considerable scale gap between micro-texture and an actual aero-engine blade, wind tunnel tests and numerical simulations with massive grids directly describing the global flow field are costly for aerodynamic evaluation.Furthermore, the fine micro surface structure brings unavoidable manufacturing errors, and the probability prediction contributes to gaining the confidence interval of the results. Therefore,a novel relay-based probabilistic model for multi-fidelity scenarios in the TPL prediction of a compressor cascade with micro-riblet surfaces is proposed to trade off accuracy and efficiency.Combined with the low-fidelity flow data generated by an aerodynamic solution strategy using the boundary surrogate model and the high-fidelity flow data from the experiment, the relay-based modeling has been achieved through knowledge transferring, and the confidence interval can be provided by the Gaussian Process Regression(GPR)model. The TPL of compressor cascades with micro-riblet surfaces under different surface structures at March number Ma=0.64,0.74,0.84 have been evaluated using the Relay-Based Probabilistic(RBP)model. The results illustrate that the RBP model could provide higher accuracy than the Single-Fidelity-Data-Driven(SFDD)prediction model, which show the promising potential of multi-fidelity scenarios data fusion in the aerodynamic evaluation of multi-scale configurations.

文摘Stroke remains a leading cause of death and disability worldwide, and electroacupuncture has a long history of use in stroke treatment. This meta-analysis and systematic review aimed to evaluate the efficacy of electroacupuncture and explore its potential mechanisms in animal models of ischemic stroke. The PubMed,EMBASE,Web of Science,CENTRAL,and CINAHL databases were comprehensively searched up to May 1,2024.This review included articles on preclinical investigations of the efficacy and mechanisms of electroacupuncture in treating ischemic stroke.Data from 70 eligible studies were analyzed in Stata 18.0,using a random-effects model to calculate the standardized mean difference(Hedge’s g).The risk of bias was assessed using RevMan 5.4 software, and the quality of evidence was rated according to the Grading of Recommendations,Assessment,Development, and Evaluation(GRADE)system.Subgroup analyses were conducted to test the consistency of the results and sensitivity analyses were used to assess their robustness. The quality assessment revealed that most studies adequately handled incomplete data and selective reporting. However, several methodological limitations were identified:only 4 studies demonstrated a low risk of allocation concealment,26 achieved a low risk of outcome assessment bias, and 9 had a high risk of randomization bias.Additionally, there was an unclear risk regarding participant blinding and other methodological aspects. The GRADE assessment rated 12 outcomes as moderate quality and 6 as low quality. The mechanisms of electroacupuncture treatment for ischemic stroke can be categorized as five primary pathways:(1)Electroacupuncture significantly reduced infarct volume and apoptotic cell death(P<0.01)in ischemic stroke models;(2)electroacupuncture significantly decreased the levels of pro-inflammatory factors(P<0.01)while increasing the levels of anti-inflammatory factors(P=0.02);(3)electroacupuncture reduced the levels of oxidative stress indicators(P<0.01)and enhanced the expression of antioxidant enzymes(P<0.01);(4)electroacupuncture significantly promoted nerve regeneration(P<0.01);and(5)electroacupuncture influenced blood flow remodeling(P<0.01)and angiogenesis(P<0.01).Subgroup analyses indicated that electroacupuncture was most effective in the transient middle cerebral artery occlusion model(P<0.01)and in post-middle cerebral artery occlusion intervention(P<0.01).Dispersive waves were found to outperform continuous waves with respect to neuroprotection and anti-inflammatory effects(P<0.01),while scalp acupoints demonstrated greater efficacy than body acupoints(P<0.01).The heterogeneity among the included studies was minimal, and sensitivity analyses indicated stable results.Their methodological quality was generally satisfactory. In conclusion, electroacupuncture is effective in treating cerebral ischemia by modulating cell apoptosis, oxidative stress,inflammation,stroke-induced nerve regeneration, blood flow remodeling, and angiogenesis. The efficacy of electroacupuncture may be influenced by factors such as the middle cerebral artery occlusion model, the timing of intervention onset, waveform, and acupoint selection.Despite the moderate to low quality of evidence, these findings suggest that electroacupuncture has clinical potential for improving outcomes in ischemic stroke.

文摘User identity linkage(UIL)across online social networks seeks to match accounts belonging to the same real-world individual. This cross-platformmapping enables accurate user modeling but also raises serious privacy risks.Over the past decade, the research community has developed a wide range of UIL methods, from structural embeddings tomultimodal fusion architectures. However, corresponding adversarial and defensive approaches remain fragmented and comparatively understudied. In this survey,we provide a unified overview of both mapping and antimappingmethods for UIL.We categorize representativemappingmodels by learning paradigmand datamodality, and systematically compare them with emerging countermeasures including adversarial injection, structural perturbation, and identity obfuscation. To bridge these two threads,we introduce amodality-oriented taxonomy and a formal gametheoretic framing that casts cross-network mapping as a contest between mappers and anti-mappers.This framing allows us to construct a cross-modality dependency matrix, which reveals structural information as themost contested signal, identifies node injection as the most robust defensive strategy, and points to multimodal integration as a promising direction. Our survey underscores the need for balanced,privacy-preserving identity inference and provides a foundation for future research on the adversarial dynamics of social identity mapping and defense.
