Based on the field data obtained during cruises on the shelf of the East China Sea from 1997 to 1999, seasonal variations of coastal upwelling on the inner shelf are discussed by using cross-shelf transect profiles and horizontal distributions of chemical and hydrographic variables. Results show that the coastal upwelling was year-round, but the areas and intensities of the upwelling were quite different in season. The coastal upwelling occurred in all of the coastal areas of the region in spring and summer, but in autumn only in the area off Zhejiang Province, and in winter in the area off Fujian Prov- ince. It was the strongest in summer and the weakest in winter. Geographically, it was the strongest in the area off Zhejiang Province and the weakest in the southmost or northmost parts of the East China Sea. The estimated nutrient fluxes upward into euphotic zone through coastal upwelling were quite large, es- pecially for phosphate, which contributed significantly to primary production and improved the nutrient structure of the coastal ecosystem in the East China Sea. 展开更多

The effects of changing salinity and nitrogen limitation on dimethylsulfoniopropionate(DMSP) and dimethylsulfide(DMS) concentrations were investigated in batch cultures of coastal diatom Skeletonema costatum,an ecologically important species.Changes in salinity from 20-32 caused no measurable variation in cell growth or culture yield,but increased intracellular DMSP per cell by 30%.Nitrogen limitation caused up to a two-fold increase in total DMSP per cell and up to a three-fold increase in DMS per cell.These changes in DMSP and DMS per cell in the Skeletonema costatum cultures with nitrogen limitation and changing salinity were primarily attributed to the physiological functions of DMSP as an osmolyte and an antioxidant.The data obtained in this study indicated that nitrogen limitation and salinity may play an important role in climate feedback mechanisms involving biologically derived DMS. 展开更多

For understanding more about the water exchange between the Kuroshio and the East China Sea,We studied the variability of the Kuroshio in the East China Sea(ECS) in the period of 1991 to 2008 using a three-dimensional circulation model,and calculated Kuroshio onshore volume transport in the ECS at the minimum of 0.48 Sv(1 Sv ;106 m3/s) in summer and the maximum of 1.69 Sv in winter.Based on the data of WOA05 and NCEP,The modeled result indicates that the Kuroshio transport east of Taiwan Island decreased since 2000.Lateral movements tended to be stronger at two ends of the Kuroshio in the ECS than that of the middle segment.In addition,we applied a spectral mixture model(SMM) to determine the exchange zone between the Kuroshio and the shelf water of the ECS.The result reveals a significantly negative correlation(coefficient of-0.78) between the area of exchange zone and the Kuroshio onshore transport at 200 m isobath in the ECS.This conclusion brings a new view for the water exchange between the Kuroshio and the East China Sea.Additional to annual and semi-annual signals,intra-seasonal signal of probably the Pacific origin may trigger the events of Kuroshio intrusion and exchange in the ECS. 展开更多

An open reading frame (lcn61) of lymphocystis disease virus China (LCDV-cn), probably responsible for encoding putative zinc-finger proteins was amplified and inserted into pET24a (+) vector. Then it expressed in E. coli BL21 (DE3), and His-tag fusion protein of high yield was obtained. It was found that the fusion protein existed in E. coli mainly as inclusion bodies. The bioinformatics analysis indicates that LCN61 is C2H2 type zinc-finger protein containing four C2H2 zinc-finger motifs. This work provides a theory for functional research of lcn61 gene. 展开更多

Immunostimulants may improve disease resistance of aquaculture animals by promoting the nonspecific immunity response of the organisms. Five types of saccharides, including chitosan, yeast polysaccharide, burdock oligosaccharide, seaweed polysaccharide and lentinus edodes polysaccharide, were screened for potential use as immunostimulants by using spectrophotometry. The saccharides were injected into Apostichopusjaponicus, a sea cucumber, and the lysozyme and superoxide dismutase (SOD) activities of the coelomic fluid and epidermal slime were monitored in six consecutive days. The results show that the lysozyme activity of the animal's coelomic fluid was significantly stimulated on day 2, day 4 and day 6 after the injection of the saccharides (P〈0.05). The effects of chitosan and yeast polysaccharide were the most notable. The lysozyme activity of the epidermal slime was significantly increased by chitosana, yeast polysaccharide, seaweed polysacchafide, and burdock oligosaccharide on day 1 and day 2 after the injection (P〈0.05). The SOD activity of the coelomic fluid was significantly promoted by the saccharides on day 2 and day 4 post-injection (P〈0.05), while the SOD activity of the epidermal slime increased on day 2. These findings indicate that chitosan and yeast polysaccharide are the most effective immunostimulants and potential healthy anti-disease feedstuff for A. japonicus. 展开更多

To develop a NaI (T1) detector for in situ radioactivity monitoring in the marine environment and enhance the confidence of the probability of the gamma-spectrum analysis, Monte Carlo simulations using the Monte Carlo N-Particle ( MNCP ) code were performed to provide the response spectra of some interested radionuclides and the background spectra originating from the natural radionuclides in seawater recorded by a NaI (T1) detector. A newly developed 75 mm × 75 mm NaI (T1) detector was calibrated using four reference radioactive sources 137Cs, 60Co, 40K and 54Mn in the laboratory before the field measurements in seawater. A simulation model was established for the detector immersed in seawater. The simulated spectra were all broadened with Gaussian pulses to reflect the statistical fluctuations and electrical noise in the real measurement. The simulated spectra show that the single-energy photons into the detector are mostly scattering low-energy photons and the high background in the low energy region mainly originates from the Compton effect of the high energy y-rays of natural radionuclides in seawater. The simulated background spectrum was compared with the experimental one recorded in field measurement and they seem to be in good agreement. The simulation method and spectra can be used for the accurate analysis of the filed measurement results of low concentration radioactivity in seawater. 展开更多

The inhibiting effect of ciprofloxacin,norfloxacin and ofloxacin on the corrosion of mild steel in 1 mol·L-1 HCl and the mechanism were studied at different temperatures using mass loss measurement,electrochemical method,and X-ray photoelectron spectroscopy(XPS) .Effective inhibition was shown by mass loss,potentiodynamic polarization and impedance spectroscopy measurement.The corrosion rate of the metal in the mass loss measurement,and the corrosion reaction on cathode and anode in the electrochemical measurement were accelerated when temperature was increased.XPS results showed that the inhibitors adsorbed effectively on the metal surface. 展开更多

We investigated the distribution of four enzymes involved in the immune response of Apostichopus japonicus. We collected samples of the tentacles, papillate podium, integument, respiratory tree, and digestive tract and stained them for acid phosphatase (ACP), alkaline phosphatase (AKP), non-specific esterase (NSE) and peroxidase (POD) activity. The distribution and content of ACE AKP, NSE, and POD differed among the tissues. The coelomic epithelium of the tentacle, papillate podium, and integument and the mucous layer of respiratory tree were positive for ACP activity. The coelomic epithelium and cuticular layer of the tentacle, papillate podium, and integument and the mucous layer and tunica externa of the respiratory tree and digestive tract stained positive or weakly positive for AKP activity. Almost all the epithelial tissues stained positive, strongly positive, or very strongly positive for NSE activity. The cuticular layer of the tentacle and integument and the mucous layer, tunica submucosa, and tunica externa of the respiratory tree and digestive tract stained positive for POD activity. We hypothesize that these enzymes play a role in the immune response in A. japonicus. 展开更多

A total of 69 strains of thermophilic bacteria were isolated from water, soil and sediment samples from three Indonesia's hot spring areas (Pantai cermin, Kalianda and Banyu wedang) by using Minimal Synthetic Medium (MSM). The extreme thermophile Brevibacillus sp. PLI-1 was found to produce extracellular thermophilic alkaline protease with optimal activity at 70℃ and pH 8.0-9.0. The molecular weight of the protease was estimated to be around 56 kD by SDS-PAGE. The maximum activity of the protease was 26.54 U mL-1. The protease activity did not decrease after 30 min and still retained more than 70% of relative activity after 60 min at 70℃ and pH 8.0. The ion Mg2+ was found to promote protease activity at both low and high concentrations, whereas Cu2+ and Zn2+ could almost completely inhibit the activity. Divalent cation chelator EDTA inhibited the enzyme activity by 55.06% ± 0.27%, while the inhibition caused by PMSF, Leupeptin, Pepstain A and Benzamidine were 66.78% ± 3.25%, 52.37% ± 0.25%, 62.47% ± 2.96% and 50.99% ± 0.24%, respectively. Based on these observations, the enzyme activity was conspicuously sensitive to the serine and cysteine protease inhibitors. All these results indicated that the protease isolated from the strain PLI-1 was a thermophilic protease and had a high-temperature stability and a pH stability. 展开更多

The human species,while buffered against environmental changes by culture and technology,is fundamentally dependent on the flow of ecosystem services. Marine ecosystem services become increasingly valuable as the terrestrial resources become scarce. The value of marine ecosystem services is the monetary flow of ecosystem services on specific temporal and spatial scales,which often changes due to the variation of the goods prices,yields and the status of marine exploitation. Sensitivity analysis is to study the relationship between the value of marine ecosystem services and the main factors which affect it. Uncertainty analysis based on varying prices,yields and status of marine exploitation was carried out. Through uncertainty analysis,a more credible value range instead of a fixed value of marine ecosystem services was obtained in this study. Moreover,sensitivity analysis of the marine ecosystem services value revealed the relative importance of different factors. 展开更多

Based on the data obtained during the 15th Antarctic Expedition of China between November 1998 and February 1999, the paper discusses the water masses distributed on both sides of the Continental Water Boundary (CWB) as well as the spacial variability of the physical characteristics of the central location, the frontal width, the frontal strength, the vertical depth, and the vertical thickness for CWB. The above results are compared with the results from the data obtained during the 9th Antarctic Expedition of China between December 1992 and February 1993, and the temporal variability of those characteristics is discussed. In addition, the inhomogeneity of the kinematic characteristics and the patterns of the dynamic heights for those water masses located on both sides of CWB are illustrated. As it is pointed out in the paper, the trough in the dynamic height field is identified with the frontal location of CWB and the oceanic current shear. Furthermore, the dynamic reason for the upwelling of the circumpolar deep water can be understood by the explanation described in the paper. 展开更多

Lymphocystis disease,caused by the lymphocystis disease virus (LCDV),is a significant worldwide problem in fish industry causing substantial economic losses.In this study,we aimed to develop the DNA vaccine against LCDV,using DNA vaccination technology.We evaluated plasmid pEGFP-N2-LCDV1.3 kb as a DNA vaccine candidate.The plasmid DNA was transiently expressed after liposome transfection into the eukaryotic COS 7 cell line.The distribution and expression of the DNA vaccine (pEGFP-N2-LCDV1.3kb) were also analyzed in tissues of the vaccinated Japanese flounder by PCR,RT-PCR and fluorescent microscopy.Results from PCR analysis indicated that the vaccine-containing plasmids were distributed in injected muscle,the muscle opposite the injection site,the hind intestine,gill,spleen,head,kidney and liver,6 and 25 days after vaccination.The vaccine plasmids disappeared 100 d post-vaccination.Fluorescent microscopy revealed green fluorescence in the injected muscle,the muscle opposite the injection site,the hind intestine,gill,spleen,head,kidney and liver of fish 48 h post-vaccination,green fluorescence did not appear in the control treated tissue.Green fluorescence became weak at 60 days post-vaccination.RT-PCR analysis indicated that the mcp gene was expressed in all tested tissues of vaccinated fish 6-50 days post-vaccination.These results demonstrate that the antigen encoded by the DNA vaccine is distributed and expressed in all of the tissues analyzed in the vaccinated fish.The antigen would therefore potentially initiate a specific immune response.The plasmid DNA was injected into Japanese flounder (Paralichthys olivaceus) intramuscularly and antibodies against LCDV were evaluated.The results indicate that the plasmid encoded DNA vaccine could induce an immune response to LCDV and would therefore offer immune protection against LCD.Further studies are required for the development and application of this promising DNA vaccine. 展开更多

Mineralogical analysis was performed on bulk sediments of 79 surface samples using X-ray diffraction. The analytical results, combined with data on ocean currents and the regional geological background, were used to investigate the mineral sources. Mineral assemblages in sediments and their distribution in the study area indicate that the material sources are complex. (1) Feldspar is abundant in the sediments of the middle Chukchi Sea near the Bering Strait, originating from sediments in the Anadyr River carried by the Anadyr Current. Sediments deposited on the western side of the Chukchi Sea are rich in feldspar. Compared with other areas, sediments in this region are rich in hornblende transported from volcanic and sedimentary rocks in Siberia by the Anadyr Stream and the Siberian Coastal Current. Sediments in the eastern Chukchi Sea are rich in quartz sourced from sediments of the Yukon and Kuskokwim rivers carried by the Alaska Coastal Current. Sediments in the northern Chukchi Sea are rich in quartz and carbonates from the Mackenzie River sediments. (2) Sediments of the southern and central Canada Basin contain little calcite and dolomite, mainly due to the small impact of the Beaufort Gyre carrying carbonates from the Canadian Arctic Islands. Compared with other areas, the mica content in the region is high, implying that the Laptev Sea is the main sediment source for the southern and central Canada Basin. In the other deep sea areas, calcite and dolomite levels are high caused by the input of large amounts of sediment carried by the Beaufort Gyre from the Canadian Arctic Islands (Banks and Victoria). The Siberian Laptev Sea also provides small amounts of sediment for this region. Furthermore, the Atlantic mid-water contributes some fine-grained material to the entire deep western Arctic Ocean. 展开更多

The aquaculture of sea cucumber Apostichopus japonicus (Echinodermata, Holothuroidea) has grown rapidly during recent years and has become an important sector of the marine industry in Northern China. However, with the rapid growth of the industry and the use of non-standard culture techniques, epidemic diseases of A. japonicus now pose increasing problems to the industry. To screen the genes with stress response to bacterial infection in sea cucumber at a genome wide level, we constructed a cDNA library from A. japonicus Selenka (Aspidochirotida: Stichopodidae) after infecting them with Vibrio sp. for 48 h. Total RNA was extracted from the intestine, mesentery and coelomocyte of infected sea cucumber using Trizol and mRNA was isolated by Oligotex mRNA Kits. The ligated cDNAs were transformed into DH5α, and a library of 3.24×105 clones (3.24×105 cfu mL-1) was obtained with the sizes of inserted fragments ranging from 0.8 to 2.5 kb. Sequencing the cDNA clones resulted in a total of 1106 ESTs that passed the quality control. BlastX and BlastN searches have identified 168 (31.5%) ESTs sharing significant homology with known sequences in NCBI protein or nucleotide databases. Among a panel of 25 putative immunity-related genes, serum lectin isoform, complement component 3, complement component 3-like genes were further studied by real-time PCR and they all increased more than 5 fold in response to Vibrio sp. challenge. Our library provides a valuable molecular tool for future study of invertebrate immunity against bacterial infection and our gene expression data indicates the importance of the immune system in the evolution and development of sea cucumber. 展开更多

In light of Wunsch's work, the waves are analytically expressed as superimposition of eigensolutions distribution of energy flux in the shelf region: one is the ratio of water depth in and the other is the ratio of the slope of the internal tide rays to the topographic energy flux distribution: the energy flux is focused around one modenumber or case, the range of modenumbers where energy flux is distributed is narrow. Two parameters have evident effects on the the shelf region to that in the abyssal ocean slope. Generally, there are two patterns of focused around two modenumbers. An analytical study is made on the formation of the Cold Water Mass of the Yellow Sea (CWYS) and the relevant thermally driven circulation. The temperature and velocity field, obtained by solving the coupled equations of motion and heat conduction, show that, in summer, the CWYS has a horizontal cyclonic circulation (component) with vertical upwelling in the middle and downwelling at the edges, that the vertical convection (u-w components) occurs only within a thin layer near the thermocline. and that the deeper layer remains almost motionless. This current structure represents well the mechanisms responsible for the maintenance of the thermocline or CWYS during the wanner months. Comparisons between the theoretical and observed temperatures show very good qualitative and quantitative agreements for corresponding seasons. 展开更多

Conductivity, temperature and depth(CTD) data collected along a zonal hydrographic section from the northern margin of the Amery Ice Shelf on 25-27 February 2008 by the 24th Chinese National Antarctic Research Expedition (CHINARE) cruise in the 2007/2008 austral summer are analyzed to study thermohaline structures. Analysis reveals warm subsurface water in a limited area around the east end of the northern margin, where the temperature, salinity and density have east-west gradients in the surface layer of the hydrographic section. The localization of the warm subsurface water and the causes of the CTD gradients in the 'surface layer are discussed. In addition, the results from these CTD data analyses are compared with those from the 22nd CHINARE cruise in the 2005/2006 austral summer. This comparison revealed that the thermoclines and haloclines had deepened and their strengths weakened in the 2007/2008 austral summer. The difference between the two data sets and the cause for it can be reasonably explained and attributed to the change in ocean-ice-atmosphere interactions at the northern margin of the Amery Ice Shell 展开更多
