Anwei decoction alleviates chronic atrophic gastritis by modulating the gut microbiota-metabolite axis and NLRP3 inflammasome activity

BACKGROUND Chronic atrophic gastritis(CAG)is a clinically refractory gastric disease often characterized by high recurrence rates and adverse drug reactions.Anwei decoction(AWD),a traditional Chinese medicine formula,has been shown to significantly improve clinical symptoms in patients with CAG,as demonstrated by a multicenter cohort study(overall effective rate:82.5%,P<0.01).However,the unclear molecular mechanisms and therapeutic targets of AWD limit its international acceptance.AIM To investigate the therapeutic mechanisms of AWD against CAG from an integrated perspective.METHODS In this study,N-methyl-N’-nitro-N-nitrosoguanidine was used to establish a CAG rat model.Serum-derived constituents transferred from AWD were first identified using ultra-high-performance liquid chromatography coupled with tandem mass spectrometry.The concentrations of inflammatory cytokines in serum samples were determined by enzyme-linked immunosorbent assay.Moreover,gastric mucosal tissues were analyzed by quantitative realtime polymerase chain reaction to measure messenger RNA(mRNA)levels of the NLRP3 inflammasome.Western blotting was used to detect the protein expression of NLRP3,caspase-1,and interleukin(IL)-1β.To elucidate the regulatory mechanisms underlying AWD treatment,structural alterations of the gut microbiota(GM)and associated metabolites were analyzed using integrated high-throughput sequencing(16S rRNA)and liquid chromatography-mass spectrometry based untargeted metabolomics.This comprehensive approach systematically clarified AWD’s multi-target therapeutic mechanisms against CAG.RESULTS AWD notably reduced serum levels of pro-inflammatory cytokines,such as IL-1β,IL-18,tumor necrosis factor-α,and lipopolysaccharide,demonstrating significant statistical differences(all P<0.01).Additionally,AWD substantially inhibited NLRP3 mRNA expression in gastric mucosal tissue(P<0.01)and concurrently decreased the protein abundance of NLRP3,IL-1β,and caspase-1(all P<0.01),thereby suppressing inflammasome signaling activation.GM analysis indicated that AWD intervention significantly increased the relative abundance of beneficial bacteria.Associated microbial metabolites likely inhibited the NLRP3 inflammasome pathway by modulating immune cell function.Non-targeted metabolomics further indicated that AWD exerted anti-inflammatory effects by regulating critical metabolic pathways,including the Kaposi’s sarcoma-associated herpesvirus infection pathway,autophagy processes,and glycosylphosphatidylinositol-anchor biosynthesis.CONCLUSION AWD alleviates the pathological progression of CAG through multi-target synergistic mechanisms.On one hand,AWD directly suppresses gastric mucosal inflammation by inhibiting NLRP3 inflammasome activation.On the other hand,AWD remodels intestinal microbiota-metabolite homeostasis,enhances intestinal barrier function,and regulates mucosal immune responses. 展开更多

Radiator thermal performance test equipment plays a key role in the processing of developing a new type of heat radiator and its application products.The precise of temperature controlling,temperature measuring andflow measuring are the vital factors for a radiator thermal performance test equipment.Based on the above back-ground,this paper improves the measurement and control system of radiator thermal performance test equip-ment,which improves the accuracy of the radiator thermal performance test equipment.This paper also optimizes the software and hardware system simultaneously so as to improve the precision of the auto-test system of test equipment.Theflow rate ranges from 175 kg/h to 178 kg/h under different conditions.The average is 176.5 kg/h and the deviation rates are from 1.62%to 1.97%.The heat produced under various conditions is different.The maximum is 4.3 kW and the minimum is 4.2 kW for condition 1,the maximum is 3.3 kW and the minimum is 3.2 kW for condition 2 and the maximum is 1.95 kW and the minimum is 1.89 kW for condition 3.However,the deviation rate is about 2.9%,which shows that the device has high stability and high precision.This paper studies a new electronic heat cost allocate meter test method by radiator thermal performance test equipment at the same time.This paper tests temperature changes through four measures points and gets a result appeared as a heat backup which should be avoided when using in the test of electronic heat cost allocate meter.Some experiences and references could be gained for further research in the heating system from this test and research. 展开更多

BACKGROUND Colorectal cancer (CRC) is the third most prevalent malignancy and has the fourth highest global cancer mortality rate. Early diagnosis and prompt medical attention can improve quality of life and the prognosis of CRC patients. Accumulating evidence reveals that long non-coding RNAs (lncRNAs) function as oncogenes or anti-oncogenes, as well as biomarkers in various cancers. AIM To investigate the levels and molecular mechanism of the lncRNA maternally expressed gene 3 (MEG3) in CRC. METHODS The levels of lncRNA MEG3 in CRC tissue, serum and cell line samples were explored via qRT-PCR. The relationship between MEG3 levels and clinicopathological features in CRC was investigated. The diagnostic and prognostic values of serum MEG3 levels were analyzed with ROC curves and KaplanMeier survival curves, respectively. RESULTS Significant decreased levels of MEG3 existed in CRC tissue, cell lines and serum. CRC patients with down-regulated serum MEG3 levels had larger tumor sizes, and advanced clinical stages. The sensitivity and specificity of serum MEG3 levels in CRC detection was 0.667 and 0.875, respectively. Tumor size, T stages, and serum MEG3 levels are indie factors that produce an effect on CRC patients' prognosis. KaplanMeier survival curves suggested that CRC patients with high levels of MEG3 had a remarkably better overall survival rate. CONCLUSION LncRNA MEG3 is down-regulated in CRC, and regulates cell functions by targeting adenosine deaminase’s effect on RNA 1 in CRC. 展开更多

Following the previous work,in this paper,the antireflective films thicknesses,refractive indexes and reflectance spectra of different color categories of the polycrystalline silicon cells are tested and compared.It is found that the color difference of polycrystalline silicon cells is mainly caused by the antireflective film.Then the matrix transfer method is used to simulate the reflection spectra according to the actual tested parameters of the samples,and the effectiveness of the simulation is verified.Finally,according to the distribution of the spectral solar irradiance,the total solar absorption of the polycrystalline silicon cells with different antireflective film thicknesses is simulated.The optimal value of the antireflective film thickness of the polycrystalline silicon cell is calculated.This study has important guiding significance for photovoltaic(PV)enterprises to realize the optimal production of plasma enhanced chemical vapor deposition(PECVD)process in production. 展开更多

[Objectives]To establish a quality standard method for the determination of quercetin in leaves of Heritiera littoralis.[Methods]Quercetin was extracted from the leaves of H.littoralis by alcohol extraction,and the quercetin was quantitatively analyzed by HPLC.[Results]There was a good linear relationship between the peak area(A)and the content(M)of quercetin in the range of 0.048-0.168μg,A=2615.5M+1.5286,r=0.9999.The method was applied to the determination of ethanol extract of H.littoralis leaves,the average recovery rate was 98.77%and RSD was 1.57%.[Conclusions]The method is simple,convenient,reproducible and accurate,and provides an idea for the determination of quercetin in leaves of H.littoralis. 展开更多

A lung cancer early-warning risk model based on facial diagnosis image features

Objective To explore the feasibility of constructing a lung cancer early-warning risk model based on facial image features,providing novel insights into the early screening of lung cancer.Methods This study included patients with pulmonary nodules diagnosed at the Physical Examination Center of Shuguang Hospital Affiliated to Shanghai University of Traditional Chinese Medicine from November 1,2019 to December 31,2024,as well as patients with lung cancer diagnosed in the Oncology Departments of Yueyang Hospital of Integrated Traditional Chinese and Western Medicine and Longhua Hospital during the same period.The facial image information of patients with pulmonary nodules and lung cancer was collected using the TFDA-1 tongue and facial diagnosis instrument,and the facial diagnosis features were extracted from it by deep learning technology.Statistical analysis was conducted on the objective facial diagnosis characteristics of the two groups of participants to explore the differences in their facial image characteristics,and the least absolute shrinkage and selection operator(LASSO)regression was used to screen the characteristic variables.Based on the screened feature variables,four machine learning methods:random forest,logistic regression,support vector machine(SVM),and gradient boosting decision tree(GBDT)were used to establish lung cancer classification models independently.Meanwhile,the model performance was evaluated by indicators such as sensitivity,specificity,F1 score,precision,accuracy,the area under the receiver operating characteristic(ROC)curve(AUC),and the area under the precision-recall curve(AP).Results A total of 1275 patients with pulmonary nodules and 1623 patients with lung cancer were included in this study.After propensity score matching(PSM)to adjust for gender and age,535 patients were finally included in the pulmonary nodule group and the lung cancer group,respectively.There were significant differences in multiple color space metrics(such as R,G,B,V,L,a,b,Cr,H,Y,and Cb)and texture metrics[such as gray-levcl co-occurrence matrix(GLCM)-contrast(CON)and GLCM-inverse different moment(IDM)]between the two groups of individuals with pulmonary nodules and lung cancer(P<0.05).To construct a classification model,LASSO regression was used to select 63 key features from the initial 136 facial features.Based on this feature set,the SVM model demonstrated the best performance after 10-fold stratified cross-validation.The model achieved an average AUC of 0.8729 and average accuracy of 0.7990 on the internal test set.Further validation on an independent test set confirmed the model’s robust performance(AUC=0.8233,accuracy=0.7290),indicating its good generalization ability.Feature importance analysis demonstrated that color space indicators and the whole/lip Cr components(including color-B-0,wholecolor-Cr,and lipcolor-Cr)were the core factors in the model’s classification decisions,while texture indicators[GLCM-angular second moment(ASM)_2,GLCM-IDM_1,GLCM-CON_1,GLCM-entropy(ENT)_2]played an important auxiliary role.Conclusion The facial image features of patients with lung cancer and pulmonary nodules show significant differences in color and texture characteristics in multiple areas.The various models constructed based on facial image features all demonstrate good performance,indicating that facial image features can serve as potential biomarkers for lung cancer risk prediction,providing a non-invasive and feasible new approach for early lung cancer screening. 展开更多

Gastric organoids are models created in the laboratory using stem cells and sophisticated three-dimensional cell culture techniques.These models have shown great promise in providing valuable insights into gastric physiology and advanced disease research.This review comprehensively summarizes and analyzes the research advances in culture methods and techniques for adult stem cells and induced pluripotent stem cell-derived organoids,and patient-derived organoids.The potential value of gastric organoids in studying the pathogenesis of stomach-related diseases and facilitating drug screening is initially discussed.The construction of gastric organoids involves several key steps,including cell extraction and culture,three-dimensional structure formation,and functional expression.Simulating the structure and function of the human stomach by disease modeling with gastric organoids provides a platform to study the mechanism of gastric cancer induction by Helicobacter pylori.In addition,in drug screening and development,gastric organoids can be used as a key tool to evaluate drug efficacy and toxicity in preclinical trials.They can also be used for precision medicine according to the specific conditions of patients with gastric cancer,to assess drug resistance,and to predict the possibility of adverse reactions.However,despite the impressive progress in the field of gastric organoids,there are still many unknowns that need to be addressed,especially in the field of regenerative medicine.Meanwhile,the reproducibility and consistency of organoid cultures are major challenges that must be overcome.These challenges have had a significant impact on the development of gastric organoids.Nonetheless,as technology continues to advance,we can foresee more comprehensive research in the construction of gastric organoids.Such research will provide better solutions for the treatment of stomach-related diseases and personalized medicine. 展开更多
