Perciformes,the largest order of vertebrates with 20 suborders,is the most diverse fish order that dominates vertebrate ocean life.The complete mitochondrial control region(CR) of Trichiurus japonicus(Trichiuridae,Sco...Perciformes,the largest order of vertebrates with 20 suborders,is the most diverse fish order that dominates vertebrate ocean life.The complete mitochondrial control region(CR) of Trichiurus japonicus(Trichiuridae,Scombroidei) and Pampus sp.(Stromateidae,Stromateoidei) were amplified and sequenced.Together with data from GenBank,the tandem repeats in the mitochondrial CR from 48 species,which covered nine suborders of Perciformes,are reported in this study.The tandem repeats tend to be long in the suborder Percoidei and Stromateoidei.The identical repeats in 21 species of Cichlidae suggest a common origin and have existed before species divergence.Larimichthys crocea shows tandem repeats instead of the typical structure of the central conserved sequence blocks,which was first reported in Perciformes and vertebrates.This might have resulted from interruption of the polymerase activity during the H-strand synthesis.The four broader patterns presented here for the tandem repeats,including those in both the 5' and 3' ends,only in the either 5' or 3' end,and in the central conserved domain of the control region,will be useful for understanding the evolution of species.展开更多

Flow cytometry isused to identify the ploidy in different developmental stages. For induction of triploidy in fertilized eggs in vitro, thehighest induction rate observed in blastula by cytochalasin B, 6-DMAP and KCl is 49.1%, 51.7% and 77.5%,respectively. In the KCl treatment of pregnant crabs with the fertilized eggs, the highest triploid induction rateobserved in the zoea is 85.3%. For induction of tetraploidy, the highest induction rate observed in the blastula bycytochaslasin B, 6-DMAP and KCl is 50.3%, 54.9% and 79.8% respectively. In the KCl treatment of pregnant crabswith the fertilized eggs, the highest induction rate in zoea is 27.3%. Through this study such difficulty as in vitroculture is overcome. One hundred and three differential cDNAs were isolated by SSH. Sequencing and BLAST analysis showed 90 inserts shared more than 95% homolog with sequences in the GenBank/EMBL database. WS9326A is a peptide antibiotic containing a highly unusual N-methyl-E-2-3-dehydrotyrosine(NMet-Dht)residue that is incorporated during peptide assembly on a non-ribosomal peptide synthetase(NRPS).The cytochrome P450 ...WS9326A is a peptide antibiotic containing a highly unusual N-methyl-E-2-3-dehydrotyrosine(NMet-Dht)residue that is incorporated during peptide assembly on a non-ribosomal peptide synthetase(NRPS).The cytochrome P450 encoded by sas16(P450Sas)has been shown to be essential for the formation of the alkene moiety in NMet-Dht,but the timing and mechanism of the P450Sas-mediatedα,β-dehydrogenation of Dht remained unclear.Here,we show that the substrate of P450Sas is the NRPS-associated peptidyl carrier protein(PCP)-bound dipeptide intermediate(Z)-2-pent-1′-enyl-cinnamoyl-Thr-N-Me-Tyr.We demonstrate that P450Sas-mediated incorporation of the double bond follows N-methylation of the Tyr by the N-methyl transferase domain found within the NRPS,and further that P450Sas appears to be specific for substrates containing the(Z)-2-pent-1’-enyl-cinnamoyl group.A crystal structure of P450Sas reveals differences between P450Sas and other P450s involved in the modification of NRPS-associated substrates,including the substitution of the canonical active site alcohol residue with a phenylalanine(F250),which in turn is critical to P450Sas activity and WS9326A biosynthesis.Together,our results suggest that P450Sas catalyses the direct dehydrogenation of the NRPS-bound dipeptide substrate,thus expanding the repertoire of P450 enzymes that can be used to produce biologically active peptides.展开更多
The chemical communication system in Drosophila melanogaster Meigen,1830 plays a major role in courtship and consists of the male-specific cis-Vaccenyl acetate and sex-specific contact pheromones,cuticular hydrocarbon...The chemical communication system in Drosophila melanogaster Meigen,1830 plays a major role in courtship and consists of the male-specific cis-Vaccenyl acetate and sex-specific contact pheromones,cuticular hydrocarbons(CHC),which build up during ontogeny(first 4 days).They replace longer CHCs,common to both sexes and present only after the imaginal eclosion.A detailed quantitative description of the evolution of cuticular unsaturated hydrocarbons with age is presented here for males and females of different D.melanogaster strains,which have been bred in well controlled environments.Monoenes appear in both sexes at around 12 h,before female dienes.The present paper argues that this is likely linked to the switching on of a new set of genes.Ecdysone,which is more abundant in females than in males during this critical period,might control this switch.Parallel behavioral studies show that whereas female of all ages trigger early mature male courtship steps like wing vibration,only females older than 1 day trigger late courtship steps like attempted copulation.This supports the hypothesis that late male courtship steps might be triggered by the CHCs,which build up after this age,especially female-specific(Z,Z)-7,11-and(Z,Z)-5,9-dienes.展开更多
Light sheet fluorescence microscopy(LSFM)is rapidly becoming an essential technology for mesoscopic imaging of samples such as embryos and adult mouse organs.However,LSFM can suffer from optical artifacts for which th...Light sheet fluorescence microscopy(LSFM)is rapidly becoming an essential technology for mesoscopic imaging of samples such as embryos and adult mouse organs.However,LSFM can suffer from optical artifacts for which there is no intrinsic solution.The attenuation of light due to absorbing material causes“shadow”artifacts along both the illumination and detection paths.Several approaches have been introduced to reduce this problem,including scanning illumination and multi-view imaging.However,neither of these approaches completely eliminates the problem.If the distribution of the absorbing material is complex,shadows cannot be avoided.We introduce a new approach that relies on multi-modal integration of two very different mesoscopic techniques.Unlike LSFM,optical projection tomography(OPT)can operate in transmission mode to create a voxel map of the 3D distribution of the sample’s optical attenuation.Here,we demonstrate a hybrid instrument(OPTiSPIM)that can quantify this attenuation and use the information to correct the shadow artifacts of LSFM.展开更多
Exploring halogen engineering is of great significance for reducing the density of defect states in crystals of organic-inorganic hybrid perovskites and hence improving the crystal quality.Herein,high-quality single c...Exploring halogen engineering is of great significance for reducing the density of defect states in crystals of organic-inorganic hybrid perovskites and hence improving the crystal quality.Herein,high-quality single crystals of PEA_(2)PbI_(4)(X=CI,Br,I)and their para-F(p-F)substitution analogs are prepared using the facile solution method to study the effects of both p-F substitution and halogen anion engineering.After p-F substitution,the triclinic PEA_(2)PbX_(4)(X=Cl,Br)and cubic PEA_(2)PbX_(4)(X=I)crystals unifies to monoclinic crystal structure for p-F-PEA_(2)PbX_(4)(X=Cl,Br,I)crystals.The p-F substitution and halogen engineering,together with crystal structure variation,enable the tunability of optoelectrical properties.Experimentally,after the p-F substitution,the energy levels are lowered with increased Fermi levels,and the bandgaps of p-F-PEA_(2)PbX_(4)(X=Cl,Br,I)are slightly reduced.Beneftting from the enhancement of the charge transfer and the reduced trap density by p-F substitution and halogen anion engineering,the average carrier lifetime of the p-F-PEA_(2)PbI_(4)is obviously reduced.Compared with PEA_(2)PbI_(4)the X-ray detector based on p-F-PEA_(2)PbI_(4)single crystals exhibits higher radiation stability under high-dose X-ray irradiation,implying long-term operando stability.展开更多
