METHODS Rats with acetic acid indu...AIM To study the short term effect of Danshen ( Salvia miltiorrhiza ) on acetic acid induced chronic gastric ulcer in rats and its long term effect in preventing recurrence. METHODS Rats with acetic acid induced gastric ulcer were treated with Danshen and cimetidine for 30 days. Traditional gastric mucosal auto radiography and 3H TdR incorporation into gastric mucosa in vitro were employed to study the effects of Danshen in rat acetic acid induced chronic gastric ulcer, including ulcer index (UI), ulcer inhibitory rate (IR) and label rate (LR). RESULTS On the day 5, 30 and 126 of ulcer making, the UI in the Danshen group was obviously lower than that in the cimetidine group and the control group (42 3±3 9, 3 6±1 2, 4 4±2 3; 49 1±3 6, 5 9±1 4, 9 2±1 3; 61 0±3 8, 8 9±2 5, 12 4±2 4, respectively, P <0 01), the IR (%) in the Danshen group was obviously higher than that in the cimetidine group (31, 59, 64 8; 19, 33, 26, respectively), and the LR in the Danshen group was obviously higher than that in the cimetidine group and the control group (10 0±0 5, 16 2±0 8, 15 0±0 6; 9 0±0 5, 13 9±0 6, 10 8±0 7; 6 5±0 7, 10 1±0 5, 8 0±0 7, respectively, P <0 01). There was no obvious difference in UI in the Danshen group on day 30 as compared with that on day 126. CONCLUSION Danshen is effective in promoting ulcer healing and preventing recurrence. The mechanism of action is to strengthen the gastric mucosal barrier and to promote the gastric mucosal cell proliferation along the edge of the ulcer.展开更多

The present study analyzed the anti-inflammatory characteristics of curcumin via myeloperoxidase activity and nitric oxide content after 2-hour ischemia/24-hour reperfusion in Sprague Dawley rats. In addition, expressions of nuclear factor kappa B, tumor necrosis factor-α and interleukin-lβ protein were measured. Curcumin significantly reduced myeloperoxidase and nitric oxide synthase activities and suppressed expressions of nuclear factor kappa B, tumor necrosis factor-α, and interleukin-lβ in ischemia/reperfusion brain tissue. Results suggested that the neuroprotective effect of curcumin following cerebral ischemia/reperfusion injury could be associated with inhibition of inflammatory reactions.展开更多

The present study investigated whether Sal B has a neuroprotective effect on secondary spinal cord injury when administrated alone. In addition, the effects of Sal B on attenuating expression of tumor necrosis factor-α (TNF-α) following acute spinal cord injury were analyzed, as well as the effects of combined treatment of Sal B and etanercept. Immunohistochemical staining demonstrated that Sal B significantly reduced matrix metalloproteinase-1 and c-Fos expression at 24 hours after spinal cord injury, and decreased tissue edema was detected using the dry-wet weight method at 3 days after injury. In addition, Sal B significantly promoted recovery of motor function in rats. These effects were most significant at a dose of 20 mg/kg Sal B. At 24 hours after spinal cord injury, reverse transcription-polymerase chain reaction and western blot assay results showed that Sal B, etanercept, or the combination significantly suppressed increased TNF-α mRNA and protein expression, although the combination resulted in more significant outcomes. These results suggested that Sal B exerted neuroprotective effects against secondary spinal cord injury by reducing expression of matrix metalloproteinase-1, c-Fos, and TNF-α. Moreover, Sal B combined with etanercept resulted in more significant anti-inflammatory effects.展开更多

Objective To observe the effects and mechanism of Yiqi Wenyang Xuoxue Huatan Formula(YWHHF)on hypoxia-hypercapnia pulmonary hypertension rats.Methods Totally 55 SD rats were randomly divided normoxia control(N)group,h...Objective To observe the effects and mechanism of Yiqi Wenyang Xuoxue Huatan Formula(YWHHF)on hypoxia-hypercapnia pulmonary hypertension rats.Methods Totally 55 SD rats were randomly divided normoxia control(N)group,hypoxia-hypercapnia(HH)group,high-dose YWHHF(YH)group,middle-dose YWHHF(YM)group,low-dose YWHHF(YL)group,10 in each group.Rats in N group were raised outside the chamber,rats in the rest groups were raised in the oxygen chamber with low oxygen concentration and high CO2 concentration for 8 h daily,6 days a week for 4 weeks.Rats in YH,YM and YL groups were fed with YWHHF at a daily dose of 0.6,0.3 and 0.15 g/kg respectively by gastrogavage 0.5 h before entering chamber,equal volume of normal saline was administered to rats in the N group and HH group by gastrogavage.All treatment lasted for 4 successive weeks.The mean pulmonary artery pressure(mPAP),mean carotid artery pressure(mCAP)and right ventricular hypertrophy index(RVHI)were measured by right heart catheterization.The morphological changes of pulmonary arteries were observed under light microscope and electron microscope.The ratio of pulmonary vascular wall area and total area(WA/TA),the ratio of vascular lumen area and total area(LA/TA)were determined.Reverse transcription polymerase chain reaction(RT-PCR)and Western blot were used to detect the expression of ERS related protein and mRNA(GRP78,CHOP,JNK,Caspase-12).The apoptotic index of pulmonary artery smooth muscle cells was detected by TUNEL.Results Compared with the N group,mPAP,RVHI and WA/TA were increased in the HH group,LA/TA were decreased(P<0.01),expression of ERS related protein and mRNA(GRP78,JNK,Caspase-12,CHOP)were increased in the HH group(P<0.01),and pulmonary artery smooth muscle cells apoptosis rate was decreased(P<0.01).Compared with the HH group,mPAP,RVHI and WA/TA in YH,YM and YL groups were decreased,LA/TA were increased(P<0.05,P<0.01),and expression of ERS related protein and mRNA were decreased in the YH,YM and YL groups(P<0.05,P<0.01).The apoptotic rates of pulmonary artery smooth muscle cells in the YH and YM group were significantly higher than those of in the HH group(P<0.05,P<0.01).Compared with the YL group,the above indexes of YH and YM groups were significantly improved(P<0.05,P<0.01).Conclusion YWHHF can inhibit JNK,Caspase-12 and CHOP to reduce the hypoxia-hypercapnia pulmonary hypertension in rats.Comprehensive consideration,the effect of medium dose(0.3 g/kg)is the most appropriate.展开更多
