METHODS: cDNA fragments encoding M2-reactive epitopes of pyruvate dehydrogenase complex Ez (PDCE2), branched chain 2-oxo-acid dehydrogenase complex E2 (BCOADC-E2) and 2-oxo-glutarate dehydrogenase complex E2 (OGDC-E2) were amplified with PCR using total RNA extracted from human peripheral mononuclear blood cells. The fragments were cloned into the plasmid vector pQE-30 and then transferred into E. coliM15 (pREP4) for expression, which was induced by isopropylthio-β-Dgalactoside. The expressed recombinant BPO protein was demonstrated by SDS-PAGE, Western-blotting and Immunoabsorption test, its antigenic reactivity and specificity were identified with seven M2-positive sera confirmed at Euroimmun Research Center (Germany).Using the purified BPO, M2 antibodies in sera from patients with PBC and other liver related diseases were detected with ELISA.
RESULTS: The expressed BPO was observed with both antigenic reactivity and specificity of M2 autoantigens. The determination of M2 antibodies by BPO with ELISA was more sensitive than using the Euroimmun's kit with the coefficients of variation less than 10 % in both interassay and intraassay.With the newly established method, M2 antibodies were found in 100 % (20/20) of patients with PBC. Six cases of liver disease with unknown etiology and 1 patient with drug induced liver injury had detectable levels of serum M2antibodies. There were also 2 patients with autoimmune cholangitis and 1 with autoimmune hepatitis showing M2-antibody positive.
CONCLUSION: Compared with the routine immunofluorescenoe assay and commercially available assay kit using porcine heart mitochondrial protein as the antigen, the detection system established in the present study shows higher sensitivity and specificity and may be used as a powerful tool for the diagnosis of PBC.展开更多

The dynamical behaviors of coherent structures in countercurrent axisymmetric shear flows are experimentally studied.The forward velocity U1 and the velocity ratio R=(U1-U2)/(U1+U2),where U2 denotes the suction veloci...The dynamical behaviors of coherent structures in countercurrent axisymmetric shear flows are experimentally studied.The forward velocity U1 and the velocity ratio R=(U1-U2)/(U1+U2),where U2 denotes the suction velocity,are consldered as the control parameters.Two kinds of vortex structures,i.e.,axisymmetric and helical structures,were discovered with respect to different reginmes in the R versus U1 diagram .In the case of U1 rangjing from 3 to 20m/s and R from 1 to 3,the axisymmetric structures plan an important role.Based on the dynamical behaviors of axisymmetric structures,a critical forward velocity U1cr=6.8m/s was defined,subsequently,the subcritical velocity regime:U1>U1cr and the supercritical velocity regime:U1<U1er,In the subcritical velocity regine,the flow system contains shear layer self-excited oscillations in a certain range of the velocity ratio with respect to any forward velocity.In the supercritical velocity regime,the effect of the velocity ratio could be explained by the relative movement and the spatial evolution of the axisymmetric structure undergoes the following stages:(1) Kelvin-Helmholtz instability leading to vortex rolling up,(2) first time vortex agglomeration.(3) jet colunn self-excited oscillation,(4) shear layer self-excited oscillation,(5)"ordered tearing",(6) turbulence in the case of U1<4m/s (the "ordered tearing" does not exist when U1>4m/s),correspondingly,the spatial evolution of the temporal asymptotic behavior of a dynamical system can be described as follows:(1) Hopt bifurcation,(5) chaos("weak turbulence")in the case of U1<4m/s(superharmonic bifurcation does not exist when U1>4m/s).The proposed new terms,superharmonic and reversed superbarmonic bifurcations,are characterized of the frequency doubling rather than the period doubling.A kind of unfamiliar vortices referred to as the helical structure was discovered experimentally when the forward velocity around 2m/s and the velocity range from 1.1 to 2.3,There are two base frequencies contained in the flow system and they could coexist as indicated by the Wigner-Ville-Distribution and the temporal asymptotic behavior of the dynamical system corresponding to the helical vortex could be described as 2-torus as indicted by the 3D reconstructed phase trajectory and correlation dimension.The scenario of the spatial evolution of helical structures could be described as follows:the jet column is separated into two parts at a certain spatial location and they entangle each other to form the helical vortex until the occurrence of those separated jet columns to reconnect further downstream with the result that the flow system evolves into turbulence in a catastrophic form.Correspondingly,the dynamical system evolves directly into 2-tiorus through the supercritical Hopf bifurcation followed by a transition from a quasi-periodic attractor to a strange attractor.In the case of U1=2m/s,the parametric evolution of the temporal asymptotic behavior of the dynamical system as the velocity ratio increases from 1 to 3 could be described as follows:(1)2-torus(Hopf bifurcation),(2) limit cycle(reversed Hopf bifurcation),(3) strange attractor (subbarmonic bifurcation).展开更多
In this paper, we study the electronic band structure and the ferromagnetic properties of the organic radical p-NPNN by employing density-functional theory with generalized gradient approximation (GGA ) and local-spin...In this paper, we study the electronic band structure and the ferromagnetic properties of the organic radical p-NPNN by employing density-functional theory with generalized gradient approximation (GGA ) and local-spin density approximation (LSDA). The density of states, the total energy, and the spin magnetic moment are calculated. The calculations reveal that the δ-phase of p-NPNN has a stable ferromagnetic ground state. It is found that an unpaired electron in this compound is localized in a single occupied molecular orbital (SOMO) constituted primarily of π^* (NO) orbitals, and the main contribution of the spin magnetic moment comes from the π^* (NO) orbitals. By comparison, we find that the GGA is more suitable to describe free radical systems than LSDA.展开更多

The density of states, total energy, and the spin magnetic moment are calculated. The calculations reveal that the compound MnCu(pbaOH)(H20)3 has a stable metal-ferromagnetic ground state, and the spin magnetic moment per molecule is 2.208 μB, and the spin magnetic moment is mainly from Mn ion and Cu ion. An antiferromagnetic order is expected and the antiferromagnetic exchange interaction of d-electrons of Cu and Mn passes through the antiferromagnetic interaction between the adjacent C, O, and N atoms along the path linking the atoms Cu and Mn.展开更多

The localization length of the syst...We study the localization properties of electrons in a two-dimensional system in a random magnetic field with the average and the amplitude of the magnetic field fluctuations δB. The localization length of the system is calculated by using the finite-size scaling method combined with the transfer-matrix technique. In the case of weak δB, we find that the random magnetic field system is equivalent to the integer quantum Hall effect system, namely, the energy band splits into a series of disorder broadened Landau bands, at the centers of which states are extended with the localization length exponent . With increasing δB, the extended states float up in energy, which is similar to the levitation scenario proposed for the integer quantum Hall effect.展开更多
