A novel alkali-soluble polysaccharide from Lepista sordida(LSAP)mycelia with antioxidative and hepatoprotective activities was characterized.The weight-average molecular weight and number-average molecular weight of LSAP were 1.442×10^(3) and 6.05×10^(2) kDa,respectively.LSAP was consisted of glucose(57.9%),xylose(31.8%),and small amounts of rhamnose,arabinose,galactose,glucuronic acid,and galacturonic acid(1.2%–3.1%).The FT-IR and 2D NMR confi rmed that LSAP was composed of Xylp,Araf,4-O-Me-α-D-GlcpA,(1→4)-linkedβ-D-Glcp,and(1→4)-α-D-GalA,andβ-glycosidic linkages between these sugar units.LSAP displayed notable effects on 1,1-dephenyl-2-picryhydrazyl(DPPH)radical scavenging,hydrogen peroxide scavenging,lipid peroxidation inhibitory ability,reducing power and Fe^(2+)chelating property.These biological effects were further verifi ed by suppressing CCl_(4)-induced oxidative liver damage in mice at doses of 100 and 200 mg/kg.Administration of LSAP in mice prior to CCl_(4) signifi cantly prevented the CCl_(4)-induced elevation in serum alanine aminotransferase,aspartate aminotransferase,and hepatic malondialdehyde.Mice treated with LSAP demonstrated to increase activities in superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)in the liver.We also found out that LSAP prevented CCl_(4)-induced oxidative liver histological alteration.LSAP may exert hepatoprotective effects against CCl_(4)-induced damage through antioxidant mechanisms in model mice. 展开更多

A novel lectin(termed PML)was purified from fruiting bodies of the edible mushroom Phellodon melaleucus(division Basidiomycota)by ion exchange,hydrophobic interaction,and gel filtration chromatographies,with overall titer recovery~60%and 20-fold purification.PML displayed hemagglutination activity 13319 units/mg toward rabbit erythrocytes.SDS-PAGE and gel filtration analyses revealed that PML is a homodimeric lectin with a molecular weight of 28.8 kDa.PML hemagglutination activity was not inhibited by various simple sugars or their derivatives,but was enhanced by cations Ca^(2+),Mg^(2+),Zn^(2+),and Cu^(2+).The activity was stable in pH range 6–9 and in the temperature range 20–60°C.Circular dichroism(CD)spectroscopic analysis showed that PML was composed primarily ofβ-sheets with lowα-helix content.In a B16 melanoma mouse model,PML treatment significantly inhibited tumor growth,and increased cytokine IL-10 content.Our findings suggest that PML is a potential anticancer therapeutic agent. 展开更多

Gene cloning in repeat-rich polyploid genomes remains challenging.Here,we describe a strategy for overcoming major bottlenecks in cloning of the powdery mildew resistance gene(R-gene)Pm69 derived from tetraploid wild emmer wheat.A conventional positional cloning approach was not effective owing to suppressed recombination.Chromosome sorting was compromised by insufficient purity.A Pm69 physical map,constructed by assembling Oxford Nanopore Technology(ONT)long-read genome sequences,revealed a rapidly evolving nucleotide-binding leucine-rich repeat(NLR)R-gene cluster with structural variations.A single candidate NLR was identified by anchoring RNA sequencing reads from susceptible mutants to ONT contigs and was validated by virus-induced gene silencing.Pm69 is likely a newly evolved NLR and was discovered in only one location across the wild emmer wheat distribution range in Israel.Pm69 was successfully introgressed into cultivated wheat,and a diagnostic molecular marker was used to accelerate its deployment and pyramiding with other R-genes. 展开更多
