With the development of nanotechnology,gold(Au) and graphene oxide(GO) nanoparticles have been widely used in various fields,resulting in an increased release of these particles into the environment.The released nanoparticles may eventually accumulate in sediment,causing possible ecotoxicological effects to benthic invertebrates.However,the impact of Au-NPs and GO-NPs on the cosmopolitan oligochaete,Tubifex tubifex,in sediment exposure is not known.Mortality,behavioral impact(GO-NP and Au-NP) and uptake(only Au-NP) of sediment-associated Au-NPs(4.9±0.14 nm) and GO-NPs(116±0.05 nm) to T.tubifex were assessed in a number of 5-day exposure experiments.The results showed that the applied Au-NP concentrations(10 and 60 μg Au/g dry weight sediment) had no adverse effect on T.tubifex survival,while Au bioaccumulation increased with exposure concentration.In the case of GO-NPs,no mortality of T.tubifex was observed at a concentration range of 20 and180 μg GO/g dry weight sediment,whereas burrowing activity was significantly reduced at 20 and 180 μg GO/g dry weight sediment.Our results suggest that Au-NPs at 60 μg Au/g or GO-NPs at 20 and 180 μg GO/g were detected by T.tubifex as toxicants during short-term exposures. 展开更多

The contaminants of the ground are potentially harmful agents and when they are released in this medium, their persistence becomes an important concern. Because of the expressed interest, a certain number of pesticides and important chemicals and their toxicity are described in this article. The studies went on the determination of the concentration, the lethal amount of the organochlorinated compounds, chemical organophosphates, carbamates and compounds. One summer recorded 3 pesticides in 5 samples of the grounds of Sikasso and Segou (Mali). Their concentration varies from 20 (atrazine) with 45 g/kg of ground. The lethal amounts of the revealed poisons variable from 338 for phtalates to 28.710 mg/kg for hexane (alkane) thus evaluate their impact on the food chain. Organophosphates and the carbamates (insecticidal) involve a reduction of 34.2% of the number of Cyprinus carpio of fresh water. The atrazine contaminates drinking water, but the diuron modifies the behavior and the reproduction of fish by deteriorating their system of olfactive perception of natural substances. Important mortalities of birds are noted around the corn fields of Bougouni treated by the carbofuran. The pesticides involve at the man a reduction in fruitfulness, an increase in the risk of miscarriage of premature birth, congenital malformations and cancers. 展开更多

NDRG2 gene copy number is not altered in colorectal carcinoma

AIM To investigate if the down-regulation of N-myc Downstream Regulated Gene 2(NDRG2) expression in colorectal carcinoma(CRC) is due to loss of the NDRG2 allele(s).METHODS The following were investigated in the human colorectal cancer cell lines DLD-1, Lo Vo and SW-480: NDRG2 mRNA expression levels using quantitative reverse transcriptionpolymerase chain reaction(qRT-PCR); interaction of the MYC gene-regulatory protein with the NDRG2 promoter using chromatin immunoprecipitation; and NDRG2 promoter methylation using bisulfite sequencing.Furthermore, we performed qPCR to analyse the copy numbers of NDRG2 and MYC genes in the above three cell lines, 8 normal colorectal tissue samples and 40 CRC tissue samples.RESULTS As expected, NDRG2 mRNA levels were low in the three colorectal cancer cell lines, compared to normal colon.Endogenous MYC protein interacted with the NDRG2 core promoter in all three cell lines.In addition, the NDRG2 promoter was heavily methylated in these cell lines, suggesting an epigenetic regulatory mechanism.Unaltered gene copy numbers of NDRG2 were observed in the three cell lines.In the colorectal tissues, one normal and three CRC samples showed partial or complete loss of one NDRG2 allele.In contrast, the MYC gene was amplified in one cell line and in more than 40% of the CRC cases.CONCLUSION Our study suggests that the reduction in NDRG2 expression observed in CRC is due to transcriptional repression by MYC and promoter methylation, and is not due to allelic loss. 展开更多

The feedbacks are represented by non-specific, bounded, non-negative C<sup>1</sup> functions. The main result is the formulation and proof of an easily applicable criterion for existence of a globally stable fixed point of the system. The proof relies on the contraction mapping theorem. Applications of this type of systems are numerous in biology, e.g., models of the hypothalamic-pituitary-adrenal axis and testosterone secretion. Some results important for modelling are: 1) Existence of an attractive trapping region. This is a bounded set with non-negative elements where solutions cannot escape. All solutions are shown to converge to a “minimal” trapping region. 2) At least one fixed point exists. 3) Sufficient criteria for a unique fixed point are formulated. One case where this is fulfilled is when the feedbacks are negative. 展开更多

Accurate, automatic annotation of peptidases with hotpep-protease

Peptidases are essential for intracellular protein processing,signaling and homeostasis,physiological processes and for digestion of food.Moreover,peptidases are important biotechnological enzymes used in processes such as industrial food processing,leather manufacturing and the washing industry.Identification of peptidases is a crucial step in their characterization but peptidase annotation is not a trivial task due to their large sequence diversity.In the present study short,conserved sequence profiles were generated for all peptidase families with more than four members in the comprehensive Merops peptidase database.The sequence profiles were combined with the Homology to Peptide Pattern(Hotpep)method for automatic annotation of peptidases.This method is a standalone software that annotates protease sequences to Merops family and subgroup and is suitable for large-scale sequence analysis.Compared to the Mammalian Degradome Database Hotpep-protease had an accuracy of 92%and a sensitivity of 96%for annotation of the human degradome.Annotation by commonly used methods(Blast and conserved domains)had an accuracy of 69%and a sensitivity of 78%.For fungal genomes,there were large differences between annotation with Hotpep-protease,Blast-and Hidden Markov Model-based annotation and the Merops annotation,which confirms the difficulty of large-scale peptidase annotation.Manual annotation indicated that Hotpep-protease had a positive prediction rate of 0.90 compared to a positive prediction rate of 0.67 for Blast search.Hence,Hotpep-protease is highly accurate method for fast and accurate annotation of peptidases. 展开更多
