Knee osteoarthritis(KOA)represents one of the most common causes of chronic pain. The high prevalence and disability rates of KOA impose a severe burden on both individuals and society. In contrast to cutaneous pain,KOA-induced joint pain is characterized as a deep tissue pain that potentially involves distinct subgroups of peripheral sensory neurons and central processing mechanisms.Furthermore,KOA pain is closely related to locomotion activity.Impaired sensorimotor integration and pain mutually reinforce each other in KOA,forming a vicious cycle that exacerbates disease progression. In this review,we highlight the key differences between KOA pain and cutaneous pain, and the latter has been extensively studied in the pain field. We hope to offer new insights into the central mechanisms and development of new treatment strategies for KOA based on the interactions between impaired sensorimotor integration and chronic joint pain. 展开更多

Alzheimer's disease,a progressively degenerative neurological disorder,is the most common cause of dementia in the elderly. While its precise etiology remains unclear, researchers have identified diverse pathological characteristics and molecular pathways associated with its progression.Advances in scientific research have increasingly highlighted the crucial role of non-coding RNAs in the progression of Alzheimer's disease. These non-coding RNAs regulate several biological processes critical to the advancement of the disease, offering promising potential as therapeutic targets and diagnostic biomarkers. Therefore, this review aims to investigate the underlying mechanisms of Alzheimer's disease onset, with a particular focus on microRNAs,long non-coding RNAs,and circular RNAs associated with the disease. The review elucidates the potential pathogenic processes of Alzheimer's disease and provides a detailed description of the synthesis mechanisms of the three aforementioned non-coding RNAs.It comprehensively summarizes the various non-coding RNAs that have been identified to play key regulatory roles in Alzheimer's disease,as well as how these noncoding RNAs influence the disease's progression by regulating gene expression and protein functions. For example,miR-9 targets the UBE4B gene, promoting autophagy-mediated degradation of Tau protein, thereby reducing Tau accumulation and delaying Alzheimer's disease progression.Conversely, the long non-coding RNA BACE1-AS stabilizes BACE1 mRNA,promoting the generation of amyloid-βand accelerating Alzheimer's disease development.Additionally, circular RNAs play significant roles in regulating neuroinflammatory responses.By integrating insights from these regulatory mechanisms, there is potential to discover new therapeutic targets and potential biomarkers for early detection and management of Alzheimer's disease. This review aims to enhance the understanding of the relationship between Alzheimer's disease and non-coding RNAs,potentially paving the way for early detection and novel treatment strategies. 展开更多

Neuronal cell death is a common outcome of multiple pathophysiological processes and a key factor in neurological dysfunction after subarachnoid hemorrhage.Neuronal ferroptosis in particular plays an important role in early brain injury.Bromodomain-containing protein 4,a member of the bromo and extraterminal domain family of proteins, participated in multiple cell death pathways, but the mechanisms by which it regulates ferroptosis remain unclear. The primary aim of this study was to investigate how bromodomain-containing protein 4 affects neuronal ferroptosis following subarachnoid hemorrhage in vivo and in vitro. Our findings revealed that endogenous bromodomain-containing protein 4 co-localized with neurons, and its expression was decreased 48 hours after subarachnoid hemorrhage of the cerebral cortex in vivo.In addition,ferroptosis-related pathways were activated in vivo and in vitro after subarachnoid hemorrhage.Targeted inhibition of bromodomain-containing protein 4 in neurons increased lipid peroxidation and intracellular ferrous iron accumulation via ferritinophagy and ultimately led to neuronal ferroptosis.Using cleavage under targets and tagmentation analysis,we found that bromodomain-containing protein 4 enrichment in the Raf-1 promoter region decreased following oxyhemoglobin stimulation in vitro.Furthermore, treating bromodomain-containing protein 4-knockdown HT-22 cell lines with GW5074,a Raf-1 inhibitor, exacerbated neuronal ferroptosis by suppressing the Raf-1/ERK1/2 signaling pathway. Moreover, targeted inhibition of neuronal bromodomain-containing protein 4 exacerbated early and long-term neurological function deficits after subarachnoid hemorrhage. Our findings suggest that bromodomain-containing protein 4 may have neuroprotective effects after subarachnoid hemorrhage, and that inhibiting ferroptosis could help treat subarachnoid hemorrhage. 展开更多

In order to explore the reduction pathways of zinc oxide in LiCl molten salt and the optimal process, experiments were conducted in an alumina crucible using metallic lithium as the reducing agent and lithium chloride molten salt as the reaction medium at 923 K.The study assessed the effects of lithium thermochemical reduction and electrolytic reduction of ZnO.The volatilization behavior of metal oxides in molten salts, the equivalent of a reducing agent, reduction time, amount of molten salt, stirring time, and the method of reduction feed were investigated for their impacts on the reduction yield and product composition.X-ray powder diffraction(XRD)analysis of the products showed that lithium reduction of ZnO not only produced metallic Zn but also formed a LiZn alloy.Electrolytic reduction can be used to obtain the metallic Zn product by controlling the potential below-2.2 V(vs Ag/Ag^(+)).Moreover,sintered oxides and higher electrode potentials could enhance the efficiency of electrolysis.Under the optimal reaction conditions determined experimentally, the lithium reduction experiment achieved a yield of 77.2%after a 12-h test, and the electrolytic reduction reached a yield of 85.4%after a 6-h test. 展开更多

Penthorum chinense Pursh has been used for centuries as an herbal medicine and food in East Asia. The main active substances in P.chinense are galloylated macrocyclic polyphenolic compounds, which have excellent medicinal properties.Galloylation and glycosylation are key steps in the formation of polyphenolic compounds,as the glycosylation of flavonoids is required for the acylation of flavonoid glycosides, and the glycosylation of gallic acid is necessary for its role as an acyl donor. Therefore, glycosylation to generate the acyl donor or acceptor is a core step in the biosynthesis of polyphenolic compounds. However, how this glycosylation occurs in P.chinense is unknown. In this study,we determined that the UDP-glucose transferase PcUGT84A82 mediates the glycosylation of gallic acid and pinocembrin to produce 1-O-Galloyl-β-D-glucose and pinocembroside, respectively.Metabolic profiling of polyphenolic compounds using UHPLC-ESI–Q-TOF/MS revealed high levels of polyphenols in flowers, leaves, and roots, and low levels in stems of P.chinense.We performed isoform-sequencing(Iso-seq)to assemble a full-length transcriptome of P.chinense, from which we identified 58 UGT family members.PcUGT84A82 is highly similar to functional UGTs in other plant species, and PcUGT84A82 transcript levels were positively correlated with the levels of various polyphenolic compounds. We validated the function of PcUGT84A82 via in vitro enzyme assays and transient expression in Nicotiana benthamiana leaves.Subcellular localization tests showed that PcUGT84A82 localizes to the nucleus and cytoplasm. In summary,PcUGT84A82 catalyzes the conversion of gallic acid to 1-O-Galloyl-β-D-glucose as the acyl donor and pinocembrin to pinocembroside as the acyl acceptor, mediating the biosynthesis of galloylated macrocyclic polyphenolic compounds in P.chinense.These findings lay the foundation for elucidating the entire biosynthetic pathway of active polyphenols in this important herbal plant species. 展开更多

In light of the pressing global challenges of climate change, declining crop resilience, and hidden hunger,it is imperative to overcome the limitations of conventional crop breeding to enhance both the nutritional quality and stress tolerance of crops.Synthetic metabolic engineering presents innovative strategies for the precision modification and de novo design of metabolic pathways. This approach generally encompasses three essential steps:identifying key metabolites through metabolomics, integrating multi-omics technologies to investigate the synthesis and regulation of these metabolites, and utilizing gene editing or de novo design to modify crop metabolic pathways associated with desirable agronomic traits. This review underscores the vital role of plant metabolite diversity in enhancing crop nutritional quality and stress resilience.Integrated multi-omics analyses facilitate the metabolic engineering by identifying key genes, transporters, and transcription factors that regulate metabolite biosynthesis.Precision modification strategies employ genome editing tools to reprogram endogenous metabolic networks, while de novo design reconstructs metabolic pathways through the introduction of exogenous biological elements—thereby both approaches enable the targeted enhancement of desired traits. These strategies have been effectively implemented in major food crops. However, simultaneously enhancing nutritional quality and stress resilience remains challenging due to inherent trade-offs and resource competition in distinct metabolic pathways within plants.Future research should integrate AI-driven predictive models with multi-omics datasets to decipher dynamic metabolic homeostasis and engineer climate-smart crops that maximize yield while preserving quality and environmental adaptability. 展开更多

Neurodegenerative diseases, which are characterized by progressive neuronal loss and the lack of disease-modifying therapies, are becoming a major global health challenge. The existing neuromodulation techniques, such as deep brain stimulation and transcranial magnetic stimulation, show limitations such as invasiveness, restricted cortical targeting, and irreversible tissue effects. In this context,low-intensity transcranial ultrasound has emerged as a promising noninvasive alternative that can penetrate deep into the brain and modulate neuroplasticity. This review comprehensively assesses the therapeutic mechanisms, efficacy, and translational potential of low-intensity transcranial ultrasound in treating neurodegenerative diseases, with emphasis on its role in promoting neuronal regeneration, modulating neuroinflammation, and enhancing functional recovery. We summarize the findings of previous studies and systematically illustrate the potential of low-intensity transcranial ultrasound in regulating cell death mechanisms, enhancing neural repair and regeneration, and alleviating symptoms associated with neurodegenerative diseases.Preclinical findings indicate that low-intensity transcranial ultrasound can enhance the release of neurotrophic factors(e.g.,brain-derived neurotrophic factor),promote autophagy to clear protein aggregates, modulate microglial activation, and temporarily open the blood-brain barrier to facilitate targeted drug delivery.Existing clinical trial data show that low-intensity transcranial ultrasound can reduce amyloid-βplaques,improve motor and cognitive deficits, and promote remyelination in various disease models.Early clinical trials suggest that low-intensity transcranial ultrasound may enhance cognitive scores in Alzheimer’s disease and alleviate motor symptoms in Parkinson’s disease, all while demonstrating a favorable safety profile.Past studies support the notion that by integrating safety, precision, and reversibility,low-intensity transcranial ultrasound can transform the treatment landscape for neurodegenerative disease. However, more advancements are necessary for future clinical application of low-intensity transcranial ultrasound, including optimizing parameters such as frequency, intensity, and duty cycle;considering individual anatomical differences;and confirming long-term efficacy. We believe establishing standardized protocols, conducting larger trials, and investigating the underlying mechanisms to clarify dose-response relationships and refine personalized application strategies are essential in this regard.Future research should focus on translating preclinical findings into clinical practice, addressing technical challenges, and exploring combination therapies with pharmacological or gene interventions. 展开更多

Rice is one of the most important staple crops globally.Rice plant diseases can severely reduce crop yields and,in extreme cases, lead to total production loss.Early diagnosis enables timely intervention, mitigates disease severity, supports effective treatment strategies, and reduces reliance on excessive pesticide use.Traditional machine learning approaches have been applied for automated rice disease diagnosis;however, these methods depend heavily on manual image preprocessing and handcrafted feature extraction, which are labor-intensive and time-consuming and often require domain expertise.Recently,end-to-end deep learning(DL) models have been introduced for this task, but they often lack robustness and generalizability across diverse datasets. To address these limitations,we propose a novel end-toend training framework for convolutional neural network(CNN) and attention-based model ensembles(E2ETCA).This framework integrates features from two state-of-the-art(SOTA) CNN models,Inception V3 and DenseNet-201,and an attention-based vision transformer(ViT) model. The fused features are passed through an additional fully connected layer with softmax activation for final classification. The entire process is trained end-to-end,enhancing its suitability for realworld deployment.Furthermore,we extract and analyze the learned features using a support vector machine(SVM),a traditional machine learning classifier,to provide comparative insights. We evaluate the proposed E2ETCA framework on three publicly available datasets, the Mendeley Rice Leaf Disease Image Samples dataset, the Kaggle Rice Diseases Image dataset, the Bangladesh Rice Research Institute dataset, and a combined version of all three.Using standard evaluation metrics(accuracy, precision, recall, and F1-score),our framework demonstrates superior performance compared to existing SOTA methods in rice disease diagnosis, with potential applicability to other agricultural disease detection tasks. 展开更多

Few studies have investigated alterations in the immune cell microenvironment of the dorsal root ganglia following spinal cord injury and whether these modifications facilitate axonal regeneration. In this study,we used a single-cell RNA sequencing dataset to create a comprehensive profile of the diverse cell types in the dorsal root ganglia and spinal cord of a mid-thoracic contusion injury model in cynomolgus monkeys.Cell communication analysis indicated that specific signaling events among various dorsal root ganglia cell types occur in response to spinal cord injury.Single-cell analysis using dimensionality reduction clustering identified distinct molecular signatures for nine cell types, including macrophage subpopulations, and differential gene expression profiles between dorsal root ganglia cells and spinal cord cells following spinal cord injury. The macrophage subpopulations were categorized into 11 clusters(MC0-MC10)based on differentially expressed genes, with the top 10 genes being ABCA6,RBMS3,EBF1,LAMA4,ANTXR2,LAMA2,SOX5,FOXP2,GHR,and APOD.MC0,MC1,and MC2 constituted the predominant macrophage populations.MC4,MC6,and MC9 were nearly absent in the spinal cord, but exhibited significant increases in the dorsal root ganglia post-spinal cord injury.Notably, these subpopulations possess a strong capacity for regulating axonal regeneration. The developmental progression of dorsal root ganglia macrophages after spinal cord injury was elucidated using cell trajectory and pseudo-time analyses.Genes such as EBF1(MC6 and MC9 marker),RBMS3(MC6 and MC9 marker),and ABCA6(MC6 marker)showed high expression levels in the critical pathways of macrophage function.Through ligand-receptor pair analysis,we determined that the effects of macrophages on microglia are predominantly mediated through interaction pairs(e.g.,SPP1-CD44,LAMC1-CD44,and FN1-CD44),potentially facilitating specific cellular communications within the immune microenvironment. The single-cell RNA sequencing dataset used in this study represents the first comprehensive transcriptional analysis of the dorsal root ganglia after spinal cord injury in cynomolgus monkeys, encompassing nearly all cell types within the dorsal root ganglia region.Using this dataset,we evaluated diverse subtypes of macrophages in the post-spinal cord injury dorsal root ganglia area and examined the signaling pathways that facilitate interactions among immune response-related macrophages in the dorsal root ganglia.Findings from this study provide a theoretical basis for understanding how the immune microenvironment influences the regenerative capacity of dorsal root ganglia neurons after spinal cord injury and offer novel insights into the complex processes underlying the pathobiology of spinal cord injury. 展开更多

Tunneling nanotubes are crucial structures for cellular communication and are observed in a variety of cell types.Glial cells, the most abundant cells in the nervous system, play a vital role in intercellular signaling and can show abnormal activation under pathological conditions. Our bibliometric analysis indicated a substantial increase in research on tunneling nanotubes over the past two decades, highlighting their important role in cellular communication. This review focuses on the formation of tunneling nanotubes in various types of glial cells, including astrocytes, microglia, glioma cells, and Schwann cells,as well as their roles in cellular communication and cargo transport. We found that glial cells influence the stability of the neural system and play a role in nerve regeneration through tunneling nanotubes.Tunneling nanotubes facilitate the transmission and progression of diseases by transporting pathogens and harmful substances. However, they are also involved in alleviating cellular stress by removing toxins and delivering essential nutrients.Understanding the interactions between glial cells through tunneling nanotubes could provide valuable insights into the complex neural networks that govern brain function and responses to injury. 展开更多

Groundwater modeling remains challenging due to heterogeneity and complexity of aquifer systems, necessitating endeavors to quantify Groundwater Levels(GWL)dynamics to inform policymakers and hydrogeologists. This study introduces a novel Fuzzy Nonlinear Additive Regression(FNAR)model to predict monthly GWL in an unconfined aquifer in eastern Iran, using a 19-year(1998–2017)dataset from 11 piezometric wells.Under three distinct scenarios with progressively increasing input complexity, the study utilized readily available climate data, including Precipitation(Prc),Temperature(Tave),Relative Humidity(RH),and Evapotranspiration(ETo).The dataset was split into training(70%)and validation(30%)subsets.Results showed that among three input scenarios,Scenario 3(Sc3,incorporating all four variables)achieved the best predictive performance, with RMSE ranging from 0.305 m to 0.768 m,MAE from 0.203 m to 0.522 m,NSE from 0.661 to 0.980,and PBIAS from 0.771%to 0.981%,indicating low bias and high reliability.However,Sc2(excluding ETo)with RMSE ranging from 0.4226 m to 0.9909 m,MAE from 0.3418 m to 0.8173 m,NSE from 0.2831 to 0.9674,and PBIAS from−0.598%to 0.968%across different months offers practical advantages in data-scarce settings. The FNAR model outperforms conventional Fuzzy Least Squares Regression(FLSR)and holds promise for GWL forecasting in data-scarce regions where physical or numerical models are impractical.Future research should focus on integrating FNAR with deep learning algorithms and real-time data assimilation expanding applications across diverse hydrogeological settings. 展开更多

BACKGROUND Robotic assistance is increasingly used for donor and recipient hepatectomy in liver transplantation, yet existing evidence is fragmented and variably indirect.AIM To evaluate clinical outcomes, surgical performance, and economic effects of robotic-assisted donor and recipient hepatectomy in the transplant pathway.METHODS Following Preferred Reporting Items for Systematic reviews and Meta-Analyses 2020 and a priori registration, systematic reviews were included with or without meta-analysis.Four databases were searched through July 2025.Methodological quality was appraised with a measurement tool to assess systematic reviews(AMSTAR 2),and certainty was graded with grading of recommendations assessment, development and evaluation(GRADE).Evidence overlap was calculated via a citation-matrix-based corrected covered area(CCA).Effect sizes were prespecified as risk ratios(RR)for dichotomous outcomes and mean differences for continuous outcomes.RESULTS Five reviews met the inclusion criteria, four with meta-analyses and one consensus review used only for context.Donor(direct)findings were more favorable for robotics in terms of estimated blood loss(≈-117 mL)and length of stay(≈-0.6 days),although with longer operative time(≈+105 minutes).Absolute risks for donor complications were not estimable from ratio-only data.Recipient(indirect)meta-analysis indicated robotics to be favorable in terms of conversion(RR≈0.41)and severe morbidity(RR≈0.81),with a trend toward lower overall morbidity(RR≈0.92)and no difference in 30-day mortality.Differences in length of stay and operative time were small and heterogeneous.Economic evidence(indirect, network meta-analysis)suggested higher procedural costs for robotic vs laparoscopic intervention, but lower hospitalization costs vs open intervention, with laparoscopy the least expensive overall.AMSTAR 2 ratings were moderate-to-high across the reviews,GRADE certainty was low for key donor continuous outcomes, and low-to-moderate for recipient and economic outcomes.Overlap was slight(graded-corpus CCA=0.0%;including a contextual non-transplant review increased CCA to≈1.25%).CONCLUSION Robotic donor hepatectomy confers perioperative advantages at the cost of longer operative time.Recipient and economic findings are indirect and considered hypothesis-generating.Transplant-specific,prospective comparisons using a minimum standardized dataset and uniform outcome definitions are needed to resolve remaining uncertainties and to clarify the cost-utility correlation. 展开更多

Background Frailty is common and significantly impacts prognosis in heart failure(HF). The Vulnerable Elders Survey-13(VES-13), widely used in oncogeriatrics and public health, remains unexplored as a frailty screening tool in HF outpatients. In this study, we prospectively evaluated VES-13 against a multimodal screening assessment in detecting frailty and predicting individual risk of adverse prognosis. Methods Frailty was assessed at the initial visit using both a multimodal approach, incorporating Barthel Index, Older American Resources and Services scale, Pfeiffer Test, abbreviated Geriatric Depression Scale, age > 85 years, lacking support systems, and VES-13. Patients scoring ≥ 3 on VES-13 or meeting at least one multimodal criterion were classified as frail. Endpoints included all-cause mortality, a composite of death or HF hospitalization, and recurrent HF hospitalizations. Results A total of 301 patients were evaluated. VES-13 identified 40.2% as frail and the multimodal assessment 33.2%. In Cox regression analyses, frailty identified by VES-13 showed greater prognostic significance than the multimodal assessment for allcause mortality(HR = 3.70 [2.15–6.33], P < 0.001 vs. 2.40 [1.46–4.0], P = 0.001) and the composite endpoint(HR = 3.13 [2.02–4.84], P< 0.001 vs. 1.96 [1.28–2.99], P = 0.002). Recurrent HF hospitalizations were four times more frequent in VES-13 frail patients while two times in those identified as frail by the multimodal assessment. Additionally, stratifying patients by VES-13 tertiles provided robust risk differentiation. Conclusions VES-13, a simple frailty tool, outperformed a comprehensive multimodal assessment and could be easily integrated into routine HF care, highlighting its clinical utility in identifying patients at risk for poor outcomes. 展开更多

BACKGROUND In an era leaning toward a personalized alignment of total knee arthroplasty, coronal plane alignment of the knee(CPAK)phenotypes for each population are studied;furthermore, other possible variables affecting the alignment, such as ankle joint alignment, should be considered.AIM To determine CPAK distribution in the North African(Egyptian)population with knee osteoarthritis and to assess ankle joint line orientation(AJLO)adaptations across different CPAK types.METHODS A cross-sectional study was conducted on patients with primary knee osteoarthritis and normal ankle joints.Radiographic parameters included the mechanical lateral distal femoral angle, medial proximal tibial angle, and the derived calculations of joint line obliquity(JLO)and arithmetic hip-knee-ankle angle(aHKA).The tibial plafond horizontal angle(TPHA)was used for AJLO assessment, where 0°is neutral(type N),<0°is varus(type A),and>0°is valgus(type B).The nine CPAK types were further divided into 27 subtypes after incorporating the three AJLO types.RESULTS A total of 527 patients(1054 knees)were included for CPAK classification, and 435 patients(870 knees and ankles)for AJLO assessment. The mean age was 57.2±7.8 years, with 79.5%females.Most knees(76.4%)demonstrated varus alignment(mean aHKA was-5.51°±4.84°)and apex distal JLO(55.3%)(mean JLO was 176.43°±4.53°).CPAK types I(44.3%),IV(28.6%),and II(10%)were the most common.Regarding AJLO,70.2%of ankles exhibited varus orientation(mean TPHA was-5.21°±6.45°).The most frequent combined subtypes were CPAK type I-A(33.7%),IV-A(21.5%),and I-N(6.9%).A significant positive correlation was found between the TPHA and aHKA(r=0.40,P<0.001).CONCLUSION In this North African cohort, varus knee alignment with apex distal JLO and varus AJLO predominated.CPAK types I,IV,and II were the most common types, while subtypes I-A,IV-A,and I-N were commonly occurring after incorporating AJLO types;furthermore, the AJLO was significantly correlated to aHKA. 展开更多

Ischemic retinopathy is a leading cause of blindness:Ischemic retinopathies including diabetic retinopathy(DR),retinopathy of prematurity, and retinal artery and vein occlusion are major causes of visual impairment.Ischemic retinopathy can be acute, such as in central or branch retinal artery occlusion,or chronic, such as with DR(Figure 1).Although the causes of retinopathies are diverse, one pathogenic event shared by these conditions is the myeloid cell response to retinal ischemia(Shahror et al.,2024a). 展开更多

The flow of a tetra-hybrid Casson nanofluid(Al_(2)O_(3)-CuO-TiO_(2)-Ag/H_(2)O)over a nonlinear stretching sheet is investigated. The Buongiorno model is used to account for thermophoresis and Brownian motion, while thermal radiation is incorporated to examine its influence on the thermal boundary layer. The governing partial differential equations(PDEs)are reduced to a system of nonlinear ordinary differential equations(ODEs)with fully non-dimensional similarity transformations involving all independent variables. To solve the obtained highly nonlinear system of differential equations,a novel Clique polynomial collocation method is applied. The analysis focuses on the effects of the Casson parameter, power index, radiation parameter, thermophoresis parameter,Brownian motion parameter, and Lewis number. The key findings show that thermal radiation intensifies the thermal boundary layer, the Casson parameter reduces the velocity, and the Lewis number suppresses the concentration with direct relevance to polymer processing, coating flows, electronic cooling, and biomedical applications. 展开更多
