Our aim was to explore whether LIFtransfected NSCs(LIF-NSCs) can ameliorate brain injury and promote neuroprotection in a rat model of cerebral ischemia. To accomplish this goal, we transfected NSCs with a lentivirus carrying the LIF gene to stably overexpress LIF. The LIF-NSCs reduced caspase 3 activation under conditions of oxygen-glucose deprivation in vitro.Transient cerebral ischemia was induced in rats by 2 h of middle cerebral artery occlusion(MCAo), and LIF-NSCs were intravenously injected at 6 h post-ischemia. LIF-NSC treatment reduced the infarction volume and improved neurological recovery. Moreover, LIF-NSCs improved glial cell regeneration and ameliorated white matter injuryin the MCAo rats. The NSCs acted as carriers and increased the expression of LIF in the lesions to protect against cerebral infarction, suggesting that LIF-NSCs could be a potential treatment for cerebral infarction.展开更多

OBJECTIVE: To evaluate CVR by detecting the increase rate of blood flow volume of middle cerebral artery (MCA) before and after breathholding in diabetic patients with hypertension, and observe the effects of hypertension on cerebrovascular reserve capacity of diabetic patients. DESIGN: Controlled observation. SETTINGS: Department of Function, Affiliated Hospital of Hebei University; Department of Special Diagnosis, the 202 Hospital of Chinese PLA. PARTICIPANTS: Inpatients or outpatients with type 2 diabetes mellitus (DM) or primary hypertension admitted to Departments of Gastroenterology and Cardiology, Affiliated Hospital of Hebei University and the 202 Hospital of Chinese PLA from April to December 2004 were involved in this experiment. Inclusive criteria: type 2 DM met the criteria of the report on diabetes diagnosis announced in 1999 by WHO expert committee, totally 88 patients were involved. Primary hypertension met the diagnosis criteria announced in 1999 by WHO/ISH, totally 42 patients were involved. Another group of 43 concurrent subjects who received physical examination served as controls. According to the disease condition, the involved patients were assigned into 3 groups: DM group (only diabetic patients), hypertension group (only hypertension patients) and DM complicated with hypertension group (diabetic patients with hypertension). Informed consent for the examination was obtained from all the involved subjects. METHODS: Before MCA of subjects was detected, bilateral carotid artery was routinely detected by high-frequency ultrasonography. The mean velocity and diameter of blood flow were detected under the quiescent condition and 25 s after breath holding with transcranial color-coded duplex sonography. The volume of blood flow of MCA was calculated according to the following formula: Volume of blood flow of MCA (mL/ min) = (the diameter of blood flow /2) 2×π× mean blood flow velocity×60. The increase rates of mean blood flow velocity and blood flow volume of patients with hypercapnia were calculated as compared with under the quiescent condition to evaluate cerebrovascular reactivity. MAIN OUTCOME MEASURES: Mean blood flow velocity, blood flow diameter and MCA blood flow volume as well as the increase rates of them. RESULTS: Among the 173 participates, 103 patients and 39 healthy controls were involved in the final analysis, and the other 31 dropped out due to not accomplishing the examination. ①Comparison of blood flow parameter of MCA under the quiescent condition: The mean blood flow velocity of DM complicated by hypertension group was significantly higher than that of DM group (P 〈 0.05) and control group (P 〈 0.05); The blood flow diameter of DM complicated by hypertension group was significantly smaller than that of control group (P 〈 0.01). Under the quiescent condition, significant difference was not found in the intergroup comparison of blood flow volume of MCA. ② Comparison of increase rate of blood flow parameter: In the DM group, hypertension group, DM complicated by hypertension group and control group, the increase rate of mean blood flow velocity of was (29.34±4.50)%,(29.35±4.35)%,(26.68±4.99)% and (30.99±3.54)%, respectively, the increase rate of blood flow volume of MCA was (35.32±5.08)%, (35.36±6.16)%,(31.78±7.11)% and(37.26±4.17)%, respectively, and the increase rate of blood flow diameter was (2.29±1.09)%,(2.27±0.95)%,(1.97±1.05)% and(2.36±0.46)%, respectively. The increase rate of mean blood flow velocity and that of blood flow volume of MCA in the DM complicated byhypertension group were significantly lower than those in the other 3 groups (P 〈 0.05). CONCLUSION: CVR is markedly lowered and cerebrovascular reserve capacity is damaged in diabetic patients with hypertension.展开更多

This transient plasticity may involve NMDA receptors containing NR2B, the major sub unit expressed at early developmental stages. The main objective of the present study was to investigate the effects of NR2B gene knockdown on the functional integration of the adult-born granule cells gen- erated from the subgranule zone (SGZ) in the hippocampus. The small interfering RNA (siRNA) was used to knock down the NR2B gene in the adult-born hippocampal neurons. In the functional integration test, the mice were exposed to a novel environment (open field arena), and the expression of c-fos was immunohistochemically detected in the hippocampus. After exposure to the novel environment, siRNA-NR2B mice were significantly different from control mice in either the number of squares or the number of rears they crossed, showing decreased horizontal and vertical activity (P〈0.05). Moreover, the c-fos expression was increased in both control and siRNA-NR2B mice after open field test. But, it was significantly lower in siRNA-NR2B neurons than in control neurons. It was concluded that the neu- ral activity of newborn neurons is regulated by their own NR2B-containing NMDA glutamate receptors during a short, critical period after neuronal birth.展开更多

Patients are likely to use information obtained from the internet to control their epilepsy, but little is known about the accuracy of this information. In this survey, we have assessed the feasibility and usability ofinternet-based interventions for the treatment of epilepsy. Methods: Data were collected from an internet search. Different search terms were used to obtain general information on epilepsy together with information about medication, types of epilepsy, treatment, women's health, and other information. The accuracy of the information was evaluated by a group of experts. Results: A total of 1320 web pages were assessed. The majority were websites related to health. A large number (80.2%) of web pages contained content related to the search term. A significant number of web pages 450/1058 (42.5%) claimed to provide information from a credible source: however, only 206/1058 (19.5%) of the information was accurate and complete; 326/1058 (30.8%) was accurate but incomplete; 328/1058 (31.0%) was correct but nonstandard, and 198/1058 (18.8%) was inaccurate. The authenticity of the information was not significantly different between the two search engines (x^2 = 0.009, P - 0.924). No significant difference was observed in the information obtained from a specialist or nonspecialist source (x^2 = 7.538, P- 0.057). There was also no correlation between the quality of the information and the priority (x^2 = 6.880, P = 0,076). Conclusions: Searching for information about epilepsy on the internet is convenient, but the information provided is not reliable. Too much information is inaccurate or for advertisement purposes, and it is difficult for patients to find the useful information. Turning to the internet for medical knowledge may be harmful. Physicians should be aware that their patients may search for information on the internet and guide them to safe, reputable websites.展开更多

Results: LSD1 and PARP1 mRNA expression in ovarian cancer tissue were significantly higher than those in benign ovarian tissue;P21, P27 and E-cadherin protein levels in ovarian cancer tissue were significantly lower than those in benign ovarian tissue while CyclinD1, E2F, Twist1, Snail, Slug and N-cadherin protein levels were significantly higher those in benign ovarian tissue;P21 and P27 protein levels in the ovarian cancer tissue with high LSD1 expression were significantly lower than those in the ovarian cancer tissue with low LSD1 expression while CyclinD1 and E2F protein levels were significantly higher than those in the ovarian cancer tissue with low LSD1 expression;Twist1, Snail, Slug and N-cadherin protein levels in the ovarian cancer tissue with high PARP1 expression were significantly higher than those in the ovarian cancer tissue with low PARP1 expression while E-cadherin protein level was significantly lower than that in the ovarian cancer tissue with low PARP1 expression. Conclusion: The LSD1 and PARP1 highly expressed in ovarian cancer tissue can promote the proliferation and epithelial-mesenchymal transition of cancer cells.
文摘Objective: To investigate the correlation of the carotid intima-media unevenness and stiffness with serum illness indexes in patients with type 2 diabetes mellitus. Methods: A total of 118 patients with type 2 diabetes mellitus who were treated in this hospital between February 2016 and August 2017 were selected as the diabetes group, and 100 healthy volunteers who received physical examination in this hospital during the same period were selected as normal control group. The differences in carotid intima-media unevenness and stiffness levels as well as serum levels of insulin resistance indexes and inflammatory adipocytokines were compared between the two groups of subjects. Pearson test was used to assess the correlation between carotid intima-media parameter levels and above serum illness index levels in patients with type 2 diabetes mellitus. Results: IMIsqrt and PWV levels in diabetes group were higher than those in normal control group;serum FINS and IRI levels were higher than those in normal control group whereas ISI level was lower than that in normal control group;serum APN content was lower than that in normal control group whereas LEP and SAA contents were higher than those in normal control group. Pearson test showed that the IMIsqrt and PWV levels in patients with type 2 diabetes mellitus were directly correlated with serum insulin resistance index levels and inflammatory adipocytokine contents. Conclusion: Carotid intima-media unevenness and stiffness both increase in patients with type 2 diabetes mellitus, and could objectively reflect the insulin resistance and systemic micro-inflammatory state.

文摘Background:Carotid intima-media thickness(IMT)and diameter, stiffness, and wave reflections, are independent and important clinical biomarkers and risk predictors for cardiovascular diseases. The purpose of the present study was to establish nationwide reference values of carotid properties for healthy Chinese adults and to explore potential clinical determinants. Methods:A total of 3053 healthy Han Chinese adults(1922 women)aged 18-79 years were enrolled at 28 collaborating tertiary centers throughout China between April 2021 and July 2022.The real-time tracking of common carotid artery walls was achieved by the radio frequency(RF)ultrasound system. The IMT,diameter, compliance coefficient,βstiffness, local pulse wave velocity(PWV),local systolic blood pressure, augmented pressure(AP),and augmentation index(AIx)were then automatically measured and reported.Data were stratified by age groups and sex.The relationships between age and carotid property parameters were analyzed by Jonckheere-Terpstra test and simple linear regressions. The major clinical determinants of carotid properties were identified by Pearson’s correlation, multiple linear regression, and analyses of covariance. Results:All the parameters of carotid properties demonstrated significantly age-related trajectories.Women showed thinner IMT,smaller carotid diameter, larger AP,and AIx than men.Theβstiffness and PWV were significantly higher in men than women before forties, but the differences reversed after that. The increase rate of carotid IMT(5.5µm/year in women and 5.8µm/year in men)and diameter(0.03 mm/year in both men and women)were similar between men and women. For the stiffness and wave reflections, women showed significantly larger age-related variations than men as demonstrated by steeper regression slopes(all P for age by sex interaction<0.05).The blood pressures, body mass index(BMI),and triglyceride levels were identified as major clinical determinants of carotid properties with adjustment of age and sex. Conclusions:The age-and sex-specific reference values of carotid properties measured by RF ultrasound for healthy Chinese adults were established. The blood pressures,BMI,and triglyceride levels should be considered for clinical application of corresponding reference values.

文摘Background:Repetitive transcranial magnetic stimulation (rTMS) and event-related potentials (ERPs) are a noninvasive technique that widely used in neurophysiological field.Although rTMS has shown clinical utility for a number of neurological conditions,Recently, there was little understanding of the the efficacy ofrYMS on Schizophrenia(SZ) and the change of ERP between before and after rTMS treatment. The objective of this study was to investigate the characteristics of N400,mismatch negativity (MMN),and P300 before and after treatment with rTMS in SZ.Methods:One hundred and twenty-seven SZ patients hospitalized in Shanghai Mental Health Center from March 2015 to July 2017,divided into two groups (85 patients were recruited as rTMS group and 42 were recruited as sham rTMS [ShrTMS] group) and 76 normal controls (NCs) who were the staff and refresher staff in our hospital were recruited at the same time.A Chinese-made rTMS and a Runjie W J-1 ERPs instrument were used in the present experiment.N400 was elicited by congruent and noncongruent Chinese idioms. After rTMS treatment,N400,P300,and MMN characteristics were compared with those before treatment and NC group. Results:Compared with NC,the SZ patients exhibited delays in N400,P300,and MMN latency and decreased N400,P300,and MMN amplitudes in their frontal area (P 〈 0.05).After 25 rTMS treatments,N400 amplitudes in the frontal area (elicited by idioms with same phonic and different shape and meaning and with different phonic, shape, and meaning) were increased in the SZ patients (P 〈 0.05).However,there was no significant change in N400 before and after treatment with ShrTMS in SZ patients (P 〉 0.05).Amplitudes for MMN and target P300 also increased in SZ patients after rTMS treatment (P 〈 0.05).Conclusions:Based on our preliminary findings,we believe that the combined usage of N400,MMN,and P300 could be a valuable index and an electrophysiological reference in evaluating the effects ofrTMS treatment in SZ patients.
文摘Background: Most patients with epilepsy want to learn as much as possible about the disease, and many have turned to the internet for information. Physicians should be aware that their patients may search for information on the internet and guide them to safe, reputable websites.

文摘Objective:To build a reference fetal growth chart for the Chinese population based on fetal ultrasound measurements. Methods:This was a multicenter,population-based retrospective cohort study.Longitudinal ultrasound measurement data were collected from 24 hospitals in 18 provinces of China from 1st September through 31st October of 2019.The estimated fetal weight(EFW)was calculated based on head circumference, abdominal circumference, and femur length using Hadlock formula 3.Fetal growth curves were estimated using a two-level linear regression model with cubic splines. All participants were divided into two groups:the northern group(n=5829)and the southern group(n=3246)based on the geographical division of China and male fetus group(n=4775)and female fetus group(n=4300)based on fetal gender. The EFW was compared by fetal gender and geographical group. All statistical models were adjusted for maternal sociodemographic characteristics. Results:A total of 9075 participants with 31,700 ultrasound measurement records were included in this study.Male fetuses demonstrated significantly larger EFW compared to female ones starting at 16 weeks of gestation and extending to delivery(global testP<0.01).The overall geographic difference in EFW was significant(global testP=0.03),and week-specific comparisons showed that the northern group had a greater EFW starting at 15 weeks of gestation and extending to 29 weeks of gestation, although this difference did not extend to the time of delivery.TheZ-score of EFW confirmed that our Chinese fetal growth charts differed from previously published standards. Conclusion:This study provides EFW and ultrasound biometric reference measurements for Chinese fetuses and reveals differences from other fetal growth charts. The chart is worth promoting in more regions of China but should be tested prudently before use.
