BACKGROUND Rotavirus(RV),a primary cause of diarrhea-related mortality in 2021,has been shown to damage intestinal epithelial cells while upregulating intestinal stem cells(ISCs)activities.ISCs within the crypt niche drive the continuous self-renewal of intestinal epithelium,preserving its barrier functions.Paneth cells secrete antimicrobial peptide and signaling molecules within the intestine crypt,thereby playing a crucial role in intestinal immune defense and providing ISCs functional support.However,the regulatory function of Paneth cells under pathological conditions,such as RV infection,remains unclear.AIM To determine the impact of RV infection on Paneth cells and how Paneth cells regulate ISCs during intestinal injury repair.METHODS We constructed a reference genome for the RV enteric cytopathogenic human orphan virus strain and reanalyzed published single-cell RNA sequencing data to investigate Paneth cell responses to RV-induced intestinal injury.We derived Paneth-ISC communication networks using CellChat,tracked ISC differentiation with pseudotime analysis,and validated our findings in leucine-rich repeat-containing G protein-coupled receptor 5-enhanced green fluorescent protein-internal ribosomal entry site-Cre recombinase estrogen receptor variant 2 mice and organoids via immunofluorescence,flow cytometry,and reverse transcription quantitative polymerase chain reaction.RESULTS We found that RV directly infects Paneth cells,leading to a reduction in mature Paneth cells and an increase in kallikrein 1-high immature Paneth cells.Paneth-ISC communication was significantly enhanced.In particular,the bone morphogenic protein 7(BMP7)-activin A receptor type 2B/BMP receptor type 1A-Smad pathway was upregulated post-infection,suggesting that Paneth cells suppress excessive ISC proliferation.Functional validation confirmed activation of this pathway.CONCLUSION Paneth cells regulate ISC proliferation during RV infection by activating BMP7 signaling,limiting excessive stem cell expansion and preserving crypt homeostasis for effective epithelial repair. 展开更多

Stroke is a major cause of death and disability worldwide.It is characterized by a highly interconnected and multiphasic neuropathological cascade of events,in which an intense and protracted inflammatory response plays a crucial role in worsening brain injury.Neuroinflammation,a key player in the pathophysiology of stroke,has a dual role.In the acute phase of stroke,neuroinflammation exacerbates brain injury,contributing to neuronal damage and blood–brain barrier disruption.This aspect of neuroinflammation is associated with poor neurological outcomes.Conversely,in the recovery phase following stroke,neuroinflammation facilitates brain repair processes,including neurogenesis,angiogenesis,and synaptic plasticity.The transition of neuroinflammation from a harmful to a reparative role is not well understood.Therefore,this review seeks to explore the mechanisms underlying this transition,with the goal of informing the development of therapeutic interventions that are both time-and context-specific.This review aims to elucidate the complex and dual role of neuroinflammation in stroke,highlighting the main actors,biomarkers of the disease,and potential therapeutic approaches. 展开更多

To enhance the anti-resistance efficacy of our previously disclosed naphthyl-triazine 5,structure-based drug design strategy was rationally conducted to design a series of novel biphenyl-piperidine-triazinecontaining non-nucleoside reverse transcriptase inhibitors.Remarkably,several of these compounds demonstrated single-digit nanomolar antiviral potency against both wild-type(WT)human immunodeficiency virus-1(HIV-1)and five clinically relevant mutant strains.Among these,compound 11s emerged as the most potent inhibitor,showing remarkable efficacy against WT HIV-1(50%effective concentration(EC_(50))=2 nmol/L)and five mutant strains(EC_(50)=0.003-0.073μmol/L),which was significantly superior to that of compound 5.This optimized derivative demonstrated substantially improved pharmacological properties compared to existing drugs etravirine(ETR)and rilpivirine(RPV),showing a 4-fold reduction in cytotoxicity alongside 6-fold enhancement in selectivity index(50%cytotoxic concentration(CC_(50))=19.69μmol/L,selectivity index(SI)=7438).The compound's metabolic profile revealed exceptional stability,with an elimination half-life(t_(1/2)=41.4 min)more than double that of RPV(t_(1/2)=16.03min).Comprehensive safety evaluation indicated minimal cytochrome P450(CYP)enzymes interference,low cardiac ion channel activity,and no observable acute toxicity,collectively suggesting a reduced risk profile for therapeutic applications.These promising characteristics significantly advance the development potential of biphenyl-piperidine-triazine derivatives as next-generation non-nucleoside reverse transcriptase inhibitors(NNRTIs),offering enhanced efficacy,improved safety,and favorable pharmacokinetic properties for antiretroviral therapy. 展开更多

Prognostic Value of Circulating Tumor Cells and Cancer Associated Macrophage-Like Cells in Metastatic Non-Small Cell Lung Cancer Patients:A Retrospective Exploratory Analysis

Objectives:Although immune checkpoint inhibitors(ICIs)and targeted therapies have reshaped treatment non-small cell lung cancer(NSCLC)paradigms,prognosis remains poor for many patients due to delayed diagnosis and resistance mechanisms.Liquid biopsy offers a minimally invasive approach to monitoring tumor evolution.Among circulating biomarkers,circulating tumor cells(CTCs)and cancer-associated macrophage-like cells(CAM-Ls)may provide complementary prognostic insights.The study aimed to evaluate the prognostic role of CTC and CAM-Ls dynamic in metastatic NSCLC patients.Methods:We retrospectively analyzed 77 patients with metastatic NSCLC who underwent CTC and CAM-L evaluation via the CellSearch^(R)system at baseline(T0)and after three months of first-line treatment(T1)including chemotherapy,targeted therapy,or ICIs.Survival outcomes were analyzed using Kaplan-Meier and Cox regression analyses.Results:Conversion to CTC-negative status at T1 was associated with improved outcomes,with median overall survival(OS)and progression-free survival(PFS)of 33 and 18 months,respectively,vs.10 and 6 months in persistently positive patients(both p<0.001).CTC negativity at T1 remained an independent prognostic factor for OS(HR:6.68)and PFS(HR:5.91,both p<0.0001).CAM-L positivity at T1 also correlated with longer OS(30 vs.12 months)and PFS(13 vs.6 months,both p<0.0001),particularly among ICI-treated patients.Combined CTC and CAM-L assessment further refined risk stratification.Conclusions:Dynamic monitoring of CTCs and CAM-Ls provides actionable prognostic information in metastatic NSCLC.CTC-negative status predicted longer OS and PFS,while CAM-L positivity at T1 was associated with improved outcomes,particularly in ICI-treated patients.Combined assessment of both biomarkers may directly inform therapeutic decision-making,through early detection of outcomes. 展开更多

Harmful algal blooms (HABs) have been affecting negatively the shellfish and aquaculture industries around the world. Though a lot of efforts have been made to disclose the changes of environmental factors involved and their effects on the HABs events, the molecular mechanism of this process remains unclear. To address this problem, proliferating cell nuclear antigen gene (pcna) was isolated and characterized from Alexandrium catenella. It showed high homology to those of other dinoflagellates (89% and 91% homology to Pfiesteria piscicid and Pyrocystis lunula, respectively), and also 42%–43% homology to those of plant and animals. The expression level of pcna revealed by quantitative real time PCR was the lowest at the late lagging cell growth phase, increased to the highest at the late exponential phase, and then decreased at the stationary phase. Though the cell growth rate was also changing, no positive correlation between pcna expression level and cell growth rate was displayed throughout the whole cell growth stages (r 2 =0.024 6). However, the pcna expression level had the similar trend with the change of cell growth rate throughout the whole growing process, e.g., from increasing at the earlier cell growth stage to decreasing at the following stages, though slightly lagging to the latter. 展开更多

Objective:To evaluate the effects of ethanol extract from Ardisia gigantifolia leaves on cell proliferation and cancer stem cell(CSC)number in gastric cancer.Methods:The inhibitory effect of Ardisia gigantifolia extract on the proliferation of MKN45 and MKN74 gastric cancer cells was assessed using 3-(4,5-dimethylthiazole-2-yl)-2,5-diphenyltetrazolium bromide assay.Non-adherent culture(3D)model was used to evaluate the effect of the extract on tumorsphere size and number.Moreover,the expression of CD44,ALDH,and p21 was determined by immunofluorescence analysis.Flow cytometric analysis was performed to evaluate cell cycle arrest and the expression of gastric CSC markers CD44 and ALDH.Real-time PCR analysis was also carried out to assess the effect of the extract on the expression of cell cycle-regulated genes.Results:Ardisia gigantifolia extract effectively inhibited cell proliferation with an IC_(50)of 55.7μg/m L in MKN45 cells and 123.6μg/m L in MKN74 cells.The extract also arrested cell cycle in the G_(0)/G_(1)phase as well as significantly reduced the size and number of tumorspheres.The markedly increased expression of p21 was observed at both m RNA and protein levels in the extract-treated adherent cells and tumorspheres.In addition,Ardisia gigantifolia extract significantly reduced the number of CD44-and/or ALDH-expressing gastric CSC.Conclusions:The development of gastric CSC can be inhibited by the ethanol extract of Ardisia gigantifolia. 展开更多

AIM:To investigate the proliferative effect of advanced glycation end-products(AGEs) and the role of their cellular receptor(RAGE) on hepatocellular carcinoma(HCC) cells,and the inhibitory effects of MK615,an extract from Japanese apricot,against AGEs were also evaluated.METHODS:Two HCC cell lines,HuH7 and HepG2,were used.Expression of RAGE was investigated by poly-merase chain reaction,Western blotting,and flow cytemetry(FACS).The effect of MK615 on RAGE expression was also evaluated by FACS.The proliferative effects of a control(unglycated bovine serum albumin),glucosederived AGEs(Glc-AGE),and glyceraldehyde-derived AGEs(Glycer-AGE),and the anti-proliferative effect of MK615 against AGEs,were evaluated using MTT assays.RESULTS:Expression of RAGE was confirmed at both the mRNA and protein levels in both HuH7 and HepG2.FACS revealed that the level of RAGE expression was higher in HuH7 than in HepG2.Treatment with 0.1 μg/mL MK615 decreased the expression level of RAGE from 24.3% to 3.7% in HuH7 and from 6.2% to 4.8% in HepG2.The growth indices for the control,Glc-AGE,and Glycer-AGE were 1.06 ± 0.08,0.99 ± 0.04,and 1.38 ± 0.05,respectively,in HuH7(P = 0.037),and were 1.03 ± 0.04,1.04 ± 0.03,and 1.07 ± 0.05,respectively,in HepG2(P > 0.05).When the cells were cultured simultaneously with Glycer-AGE and MK615,MK615 abrogated the proliferative effect of Glycer-AGE in HuH7.CONCLUSION:Only Glycer-AGE has a proliferative effect on HuH7,which expresses a higher level of RAGE.MK615 suppresses the proliferative effect of GlycerAGE on HuH7 by decreasing the expression of RAGE. 展开更多

AIM To examine whether hemodialysis(HD) patients with restless legs syndrome(RLS) are subjects of greater fatigue and impaired quality of life(QoL) compared to HD patients without RLS.METHODS Eighty five stable HD patients participated in this study. According to their RLS status, the patients were dividedinto the RLS group(n = 23) and the non-RLS group(n = 62). QoL, fatigue, sleep quality, daily sleepiness and depression symptoms were assessed by using various questionnaires. Finally, biochemical parameters including iron, ferritin, hemoglobin, hematocrit and parathormone were assessed. RESULTS The HD patients with RLS scored worse in all the questionnaires used in the study(P < 0.05). The patients with RLS were more likely to receive the HD therapy on the morning shift, whilst 43.5% of the RLS patients reported to experience the RLS symptoms also during HD. The severity of RLS was correlated with fatigue, depression score and sleep quality(P < 0.05). CONCLUSION HD patients with RLS are subject to lower QoL related parameters and greater fatigue compared to HD patients without RLS. RLS should be successfully managed in order to improve the QoL of the sufferers. 展开更多

Quorum sensing is the biochemical communication system within bacterial cells which may instigate their virulence factors,and hence,is imperative for pathogenesis.Such a system optimizes metabolic and behavioral activities played by the densely populated bacterial community as well as the production of the concomitant response against diffusible or secreted signals.Thus,the multi-layered signal transduction networks of quorum sensing help bacteria acclimatize to the disadvantages by increasing their confrontation ability aided by biofilm formation.Previous studies unraveled the impact of quorum sensing in the microbial interactions both within a single species and between several species.The development of the synthetic ecological models has added advantages in the studies of microbial sociability in natural habitats as well as aided in drug designing.Current review highlighted the up-to-date knowledge on the basic mechanisms of quorum sensing and its influence to accelerate the bacterial resistance against different environmental stimuli or synthetic drugs along with the factors affecting the quorum sensing system,and the harmful role of quorum sensing in food spoilage.The information gathered in this review would be beneficial to understand the metabolic cross talk as well as the virulence traits of the pathogenic microorganisms in course of their pathogenesis augmented by quorum sensing. 展开更多

熊蜂是蜜蜂科熊蜂属昆虫的统称,在维护自然生态系统的稳定与促进农业生产等方面发挥着重要的作用。为了掌握四川贡嘎山国家级自然保护区中熊蜂物种多样性的现状与历史变化情况、评估它们的生态服务功能、鉴定出濒危蜂种进而进行保护,本... 熊蜂是蜜蜂科熊蜂属昆虫的统称,在维护自然生态系统的稳定与促进农业生产等方面发挥着重要的作用。为了掌握四川贡嘎山国家级自然保护区中熊蜂物种多样性的现状与历史变化情况、评估它们的生态服务功能、鉴定出濒危蜂种进而进行保护,本研究在2022-2023年间,分3次在保护区中的23块采样地采用网捕的方式共采集了919头熊蜂样本,并在采样的同时记录了熊蜂取食植物的种类。同时采用形态特征与分子条形码技术相结合的方法对采集到的熊蜂标本进行了物种鉴定、计算了其多度及均匀度指数、分析了其海拔分布特点。研究结果显示,保护区中现有熊蜂21种,隶属于7个亚属,Shannon-Wiener多样性指数为2.2093。这21种熊蜂中有8种为稀有种,其中的灰熊蜂Bombus grahami、卧龙熊蜂B.wolongensis、拉达克熊蜂B.ladakhensis、稳纹熊蜂B.waltoni等4种熊蜂,不仅数量少、而且分布范围也非常狭窄,是保护区中亟需高度关注的蜂种,应尽早采取保护措施。随着海拔的升高保护区中熊蜂的数量和物种丰富度都逐渐增加,并且在海拔3000~4000 m的区间二者均达到峰值。熊蜂在保护区中为超过43种野生、经济植物提供授粉服务。与40年前的调查结果相比,拟熊蜂亚属的两种熊蜂(牛拟熊蜂B.bohemicus与探索拟熊蜂B.expolitus)均没有在保护区中被发现,大概率已经发生了局地灭绝,本研究的这一调查结果也证实了前人的猜测:由于其社会性寄生的生活方式,拟熊蜂亚属的熊蜂应该更容易濒危或灭绝。综上所述,本研究掌握了保护区中熊蜂种质资源的现状及历史变化情况,鉴定出亟需高度关注的蜂种,这为保护区中熊蜂种群的动态监测与保护奠定了重要基础。 展开更多

Size of the ulna and taxonomic order affect vane dimensions of secondary remiges in birds 被引量:1

The number of secondary feathers varies among orders of birds with some orders exhibiting a positive relationship with ulna length,whereas in other orders secondary number is invariant.This difference has implications for scaling of the width of the feather vane within orders.In those species where the number of secondary remiges is invariant with ulna length,vane width should scale isometrically with ulna size to maintain an aerodynamic flight surface.Where feather count increases with increasing ulna length then vane width should exhibit negative allometry.Vane length should also correlate with ulna length,irrespective of the number of feathers.Data were compiled from an online library of images for the vane length and the width of the vane at 50%of the vane length for the fifth secondary feather for 209 bird species from 24 different orders.The results supported the hypotheses that vane width is a function of ulna size,and the number of secondary feathers as associated with different orders.Vane length was unaffected by the number of secondaries but varied between orders.The results suggest that birds have solved the problem of maintaining the aerodynamic surface of the proximal wing in two ways.Hence as ulna length increases the first solution involves more feathers that exhibit negative allometry for vane width,or in the second where feather count doesn't change,the vane width simply scales isometrically.The implications for the mechanical properties of the vane,and how it affects wing function,have not yet been explored in a range of birds. 展开更多

Proton exchange membrane fuel cells (PEMFCs) provide an appealing sustainable energy system,with the solid-electrolyte membrane playing a crucial role in its overall performance.Currently,sulfonated poly(1,4-phenylene ether-ether sulfone)(SPEES),an aromatic hydrocarbon polymer,has garnered considerable attention as an alternative to Nafion polymers.However,the long-term durability and stability of SPEES present a significant challenge.In this context,we introduce a potential solution in the form of an additive,specifically a core–shell-based amine-functionalized iron titanate (A–Fe_(2)TiO_(5)),which holds promise for improving the lifetime,proton conductivity,and power density of SPEES in PEMFCs.The modified SPEES/A–Fe_(2)TiO_(5)composite membranes exhibited notable characteristics,including high water uptake,enhanced thermomechanical stability,and oxidative stability.Notably,the SPEES membrane loaded with 1.2 wt%of A–Fe_(2)TiO_(5)demonstrates a maximum proton conductivity of 155 mS ccm^(-1),a twofold increase compared to the SPEES membrane,at 80°C under 100%relative humidity (RH).Furthermore,the 1.2 wt%of A–Fe_(2)TiO_(5)/SPEES composite membranes exhibited a maximum power density of 397.37 mW cm^(-2)and a current density of 1148 mA cm^(-2)at 60°C under 100%RH,with an opencircuit voltage decay of 0.05 m V/h during 103 h of continuous operation.This study offers significant insights into the development and understanding of innovative SPEES nanocomposite membranes for PEMFC applications. 展开更多

Peripheral nerve injuries induce a severe motor and sensory deficit. Since the availability of autologous nerve transplants for nerve repair is very limited, alternative treatment strategies are sought, including the use of tubular nerve guidance conduits(tNGCs). However, the use of tNGCs results in poor functional recovery and central necrosis of the regenerating tissue, which limits their application to short nerve lesion defects(typically shorter than 3 cm). Given the importance of vascularization in nerve regeneration, we hypothesized that enabling the growth of blood vessels from the surrounding tissue into the regenerating nerve within the tNGC would help eliminate necrotic processes and lead to improved regeneration. In this study, we reported the application of macroscopic holes into the tubular walls of silk-based tNGCs and compared the various features of these improved silk^(+) tNGCs with the tubes without holes(silk^(–) tNGCs) and autologous nerve transplants in an 8-mm sciatic nerve defect in rats. Using a combination of micro-computed tomography and histological analyses, we were able to prove that the use of silk^(+) tNGCs induced the growth of blood vessels from the adjacent tissue to the intraluminal neovascular formation. A significantly higher number of blood vessels in the silk^(+) group was found compared with autologous nerve transplants and silk^(–), accompanied by improved axon regeneration at the distal coaptation point compared with the silk^(–) tNGCs at 7 weeks postoperatively. In the 15-mm(critical size) sciatic nerve defect model, we again observed a distinct ingrowth of blood vessels through the tubular walls of silk^(+) tNGCs, but without improved functional recovery at 12 weeks postoperatively. Our data proves that macroporous tNGCs increase the vascular supply of regenerating nerves and facilitate improved axonal regeneration in a short-defect model but not in a critical-size defect model. This study suggests that further optimization of the macroscopic holes silk^(+) tNGC approach containing macroscopic holes might result in improved grafting technology suitable for future clinical use. 展开更多

Inflammatory bowel disease,particularly Crohn's disease(CD),has been linked to modifications in mesenteric adipose tissue(MAT)and the phenomenon known as"creeping fat"(CrF).The presence of CrF is believed to serve as a predictor for early clinical recurrence following surgical intervention in patients with CD.Notably,the incorporation of the mesentery during ileocolic resection for CD has been correlated with a decrease in surgical recurrence,indicating the significant role of MAT in the pathogenesis of CD.While numerous studies have indicated that dysbiosis of the gut microbiota is a critical factor in the development of CD,the functional implications of translocated microbiota within the MAT of CD patients remain ambiguous.This manuscript commentary discusses a recent basic research conducted by Wu et al.In their study,intestinal bacteria from individuals were transplanted into CD model mice,revealing that fecal microbiota trans-plantation(FMT)from healthy donors alleviated CD symptoms,whereas FMT from CD patients exacerbated these symptoms.Importantly,FMT was found to affect intestinal permeability,barrier function,and the levels of proinflammatory factors and adipokines.Collectively,these findings suggest that targeting MAT and CrF may hold therapeutic potential for patients with CD.However,the study did not evaluate the composition of the intestinal microbiota of the donors or the subsequent alterations in the gut microbiota.Overall,the gut microbiota plays a crucial role in the histopathology of CD,and thus,targeting MAT and CrF may represent a promising avenue for treatment in this patient population. 展开更多
