The H4 subtype of avian influenza virus(AIV)is prevalent worldwide,but only receives little attention due to its low pathogenicity in poultry.Consequently,it remains largely unclear whether H4 AIVs pose a potential th...The H4 subtype of avian influenza virus(AIV)is prevalent worldwide,but only receives little attention due to its low pathogenicity in poultry.Consequently,it remains largely unclear whether H4 AIVs pose a potential threat to the poultry industry and public health.During the period from 2011 to 2022,we conducted an active surveillance programm.A total of 154,762 swab samples were collected across various provinces,and 427 H4 viruses were detected,resulting in a positivity rate of 0.28%.All H4 viruses were isolated from poultry,primarily from ducks in live poultry markets.We further investigated the genetic evolutionary characteristics and pathogenicity of 20 H4Nx viruses isolated in our program.Phylogenetic analysis revealed that the 20 H4Nx viruses belonged to the Eurasian lineage and exhibited significant genetic diversity,with 19 distinct genotypes identified.Molecular characterization indicated that these viruses were low-pathogenicity AIVs with limited binding affinity to human receptors,yet they contained mutations associated with enhanced viral replication and pathogenicity in mammals.Pathogenicity tests conducted in ducks demonstrated that H4 viruses were weakly pathogenic,exhibiting limited replication and transmission capabilities.However,some viruses were able to replicate effectively in mice and induce weight loss.For instance,DK/AH/AG61/11(H4N6)can replicate efficiently in MDCK cells,indicating a potential threat to mammals.These findings underscore the importance of ongoing surveillance of H4 AIVs to better understand their evolution and transmission dynamics and to prevent potential public health risks.展开更多
Toxoplasma gondii(T.gondii)is a globally distributed parasite that can infect a diversity of warm-blooded animals,including swine and humans.Infection in swine poses a considerable threat to food safety and public hea...Toxoplasma gondii(T.gondii)is a globally distributed parasite that can infect a diversity of warm-blooded animals,including swine and humans.Infection in swine poses a considerable threat to food safety and public health.The aim of this meta-analysis was to estimate the seroprevalence of T.gondii infection in the swine population in China from 2000 to 2023 and to examine potential factors associated with infection.A total of 112 studies were included,collectively involving 145,152 swine samples originating from 26 provinces.The pooled seroprevalence was 26.0%(95%CI:23.3%–28.7%).Stratified analysis based on diagnostic methods revealed that studies using the indirect hemagglutination assay(IHA)reported a seroprevalence of 19.7%(95%CI:17.2%–22.2%),whereas those utilizing the enzyme-linked immunosorbent assay(ELISA)reported a higher seroprevalence of 35.5%(95%CI:29.1%–41.8%).Geographical analysis indicated higher seroprevalence in the South Central and Southwest regions,whereas the East and Northwest areas reported the lowest seroprevalence.Chongqing Province reported the highest seroprevalence,reaching 44.9%(95%CI:43.4%–46.0%),followed by Xinjiang,Hainan,and Guizhou,whereas the lowest was observed in Shandong Province(3.5%,95%CI:1.7%–6.1%).These findings provide important epidemiological evidence that can inform strategies for the prevention and control of T.gondii infection in swine populations,with a focus on high-risk populations and geographical areas.This imperative contributes substantially to the improvement of both food safety and public health.展开更多

Genetic analysis revealed that Sw/SD/3/2005 was a triple-reassortant virus with a PB2 gene from human-like H1N1, NS from classical swine H1N1, and the remaining genes from human-like H3N2 virus. These findings further support the concept that swine can serve as reservoir or mixing vessels of influenza virus strains and maintain genetic and antigenic stability of viruses. Furthermore, we have successfully established a reverse genetics system based on eight plasmids and rescued Sw/SD/3/2005 through cell transfection. HI tests and RT-PCR confirmed that the rescued virus maintained the biological properties of the wild type Sw/SD/3/2005. The successful establishment of the reverse genetics system of Sw/SD/3/2005 will enable us to conduct extensive studies of the molecular evolution of H3N2 influenza viruses in swine.展开更多

During our surveillance of chicken from 2019 to 2021 in Shandong Province, China, we isolated 11 H9N2AIVs. Phylogenetic analyses showed that the eight gene segments of the 11 isolates were closely related to several sublineages of Eurasian lineage: BJ/94-like clades(HA and NA genes), G1-like clades(PB2 and M genes), and SH/F/98-like clades(PB1, PA, NP and NS genes). The isolates showed mutation sites that preferentially bind to humanlike receptors(HA) and mammalian fitness sites(PB2, PB1 and PA), as well as mutations in antigen and drug resistance sites. Moreover, studies with mice revealed four isolates with varying levels of pathogenicity. The average antibody titer of the H9N2 AIVs was 8.60 log2. Based on our results, the epidemiological surveillance of H9N2 AIVs should be strengthened.展开更多

[Methods] The species, quantities, distribution and seasonal variations of insects and wild animals in related zones were investigated from 2008 to 2010, and the collected samples of the insects and wild animals were tested in laboratory for related equine diseases. [Results] The investigations indicated that there were some mosquitoes, flies, horseflies, punkies, ticks, bats, wild birds and wild bears in equastrianism venue and peripheral regions of disease-free zone of the Guangzhou Asian Games, the laboratory results of Japa- nese encephalitis, vesicular stomatitis, Nipah virus disease, West Nile fever, and Trypanosomiasis evansi, were all negative. According to the in- vestigations and test results, the risk assessments of insect and wild animal vectors in the prevalence of related equine diseases were made to con- firm that the risk was relatively low or very low, and comprehensive prevention and control measures with prevention as major measures and insecti- cides application and environment managements as supplementary means were made on the basis of the risk assessment conclusions. Florfenicol(FLO)is a chemically synthesized broad-spectrum antimicrobial agent of amide alcohols for animals,which is one of the most widely used antimicrobials in livestock,poultry,and aquaculture.With the use of FLo...Florfenicol(FLO)is a chemically synthesized broad-spectrum antimicrobial agent of amide alcohols for animals,which is one of the most widely used antimicrobials in livestock,poultry,and aquaculture.With the use of FLo,more and more attention has been paid to its hematopoietic toxicity,immunotoxicity,genotoxicity,and embryotoxicity.In this study,SPF chicks.at the age of 3 d began to drink water with the FLO at a dose of 100 mg L^(-1)for 6 consecutive days,and the growth performance of chicks was monitored,the effect of FLO on immune organs was detected by pathological examination and TdT-mediated dUTP nick-end labeling(TUNEL)apoptosis staining.In order to evaluate the level of organism immunity,the level of Newcastle disease virus antibody in serum was detected by hemagglutination inhibition test,the content of cytokines(IL-1,IL-2,IL-6,TNF-α,IFN-γ)in serum was detected by enzyme linked immunosorbent assay(ELISA),and the transcription of interferon-related genes(IRF-7,2′-5′OAS,Mx1)and cytokine genes(IL-6,TNF-α,IFN-γ)in immune organs were detected by real time fluorescence quantitative PCR.The results showed that the early application of FLO could inhibit the growth and development of chicks,and the body weight and immune organ index of the treatment group were lower than those of the control group.Histopathological examination showed that there was a decrease in the number of lymphocytes in the bursa of Fabricius in the treatment group in the early stage of drug withdrawal,and the results of TUNEL apoptosis staining in the bursa of Fabricius showed that obvious lymphocyte apoptosis occurred in the FLO treatment group.Compared with the control group,the transcription levels of interferon-related genes IRF-7,2′-5′OAS,and cytokine genes IL-6,TNF-αand IFN-γin FLO treatment group decreased to a certain extent,while the transcription level of Mx1 gene had no significant difference at all time points.The level of serum Newcastle disease virus(NDV)antibody and the contents of cytokines IL-1,IL-2 and IFN-γin the FLO treatment group were significantly lower than those in the control group in the early stage of drug withdrawal,but recovered gradually in the later stage.This study showed that FLO has a certain degree of effect on the immune function of chicks,and the results of the study laid the foundation for further research on the mechanism of FLO-induced immunotoxicity.展开更多
A real-time RT-PCR assay using Taq Man-MGB probes was developed to detect and type the bovine viral diarrhea virus(BVDV) in cattle.Universal primers and Taq Man-MGB probes were designed from the 5′-untranslated reg...A real-time RT-PCR assay using Taq Man-MGB probes was developed to detect and type the bovine viral diarrhea virus(BVDV) in cattle.Universal primers and Taq Man-MGB probes were designed from the 5′-untranslated region of known pestiviral sequences.Prior to optimizing the assay, c RNAs were transcribed in vitro from the BVDV 1 and BVDV 2 RTPCR products to make standard curves.The detection limit of the assay was 1.72×102 copies for BVDV 1 and 2.14×102copies for BVDV 2.The specificity of the assay evaluated on several BVDV strains including bovine herpesvirus 1(BHV 1), foot and mouth disease virus(FMDV) and several classical swine fever virus(CSFV) strains showed specific detection of the positive virus over 40 cycles.The assay was highly reproducible with the coefficient of variance ranging from 1.04 to 1.33% for BVDV 1 and from 0.83 to 1.48% for BVDV 2, respectively.Using this method, we tested a total of 2 327 cattle from three dairy farms for the presence of BVDV persistently infected(PI) animals.In this assay, each RT-PCR template contained a mixture of ten samples from different animals.The occurrence rate of PI cattle in three farms ranging from 0.9 to 2.54% could represent partly the PI rates in cattle farm in China.In conclusion, using our real-time PCR assay, we could effectively detect and type BVDV and identify PI cattle in a rapid and cost-effective manner.展开更多
Platycodon grandiflorum(Jacq.)A.DC.is a famous medicinal plant commonly used in East Asia.Triterpene saponins isolated from P.grandiflorum are the main biologically active compounds,among which polygalacin D(PGD)has b...Platycodon grandiflorum(Jacq.)A.DC.is a famous medicinal plant commonly used in East Asia.Triterpene saponins isolated from P.grandiflorum are the main biologically active compounds,among which polygalacin D(PGD)has been reported to be an anti-tumor agent.However,its anti-tumor mechanism against hepatocellular carcinoma is unknown.This study aimed to explore the inhibitory effect of PGD in hepatocellular carcinoma cells and related mechanisms of action.We found that PGD exerted significant inhibitory effect on hepatocellular carcinoma cells through apoptosis and autophagy.Analysis of the expression of apoptosis-related proteins and autophagy-related proteins revealed that this phenomenon was attributed to the mitochondrial apoptosis and mitophagy pathways.Subsequently,using specific inhibitors,we found that apoptosis and autophagy had mutually reinforcing effects.In addition,further analysis of autophagy showed that PGD induced mitophagy by increasing BCL2 interacting protein 3 like(BNIP3L)levels.In vivo experiments demonstrated that PGD significantly inhibited tumor growth and increased the levels of apoptosis and autophagy in tumors.Overall,our findings showed that PGD induced cell death of hepatocellular carcinoma cells primarily through mitochondrial apoptosis and mitophagy pathways.Therefore,PGD can be used as an apoptosis and autophagy agonist in the research and development of antitumor agents.展开更多

Our result showed that Hsp90 location in the tumor tissue was signiifcantly associated with the tumor cell and most in the cytoplasm of the tumor cell, and Hsp90 expression level in the tissue and the antibody titer level in the serum was most signiifcantly increased with the development of tumor. This is the ifrst report to show the presence of Hsp90 in tumor tissues induced by the Marek’s disease, with its expression correlated to the tumoral grading. These data may also be valuable for developing new molecular anti-cancer therapies.展开更多

Several types of hemolysins produced by Vp have been characterized as major virulence factors.They are thermostable direct hemolysin (TDH),TDH-related hemolysin (TRH) and thermolabile hemolysin (TLH). In this study, we cloned tdh, trh, and tlh genes from the genome DNA of VP by polymerase chain reaction (PCR).We ligated the three genes into prokaryotic expression vector pET-28a (+),and transformed the recombinant plasmids into Es-cherichia coli BL21 (DE3). The expression of recombinant proteins was induced by isopropyl-β-D-thiogalacto-pyranoside (IPTG). The recombinant proteins were expressed in a form of inclusion bodies and thus purified with Ni-NTA affinity chromatography. Western blotting results showed that recombinant proteins,TDH, TRH and TLH, could be recognized by rabbit anti-VP serum. The three purified proteins were renatured by gradient dialysis.The renatured proteins exhibited hemolytic activity except for TLH in the presence of phosphatidylcholine. These results not only are helpful for better understanding these genes' functions under a single factor level, but also provide evidence for VP vaccine engineering.展开更多

Therefore, reduci...BACKGROUND The prevalence of constipation in the Chinese population over 60 years of age is 11.5%, and this prevalence increases with age, which seriously affects the quality of life in older adults. Therefore, reducing the incidence of constipation in older adults is necessary to promote a healthy lifestyle as well as biochemical health.AIM To explore the value of preoperative guidance and education to improve the effects of bowel cleaning in older adult patients undergoing colonoscopy.METHODS In this study, 160 older adult patients with constipation requiring colonoscopy at Shandong Provincial Hospital between January 2019 and March 2021 were selected and randomly divided into a study group and a control group, with 80patients in each group. The study group received medication guidance and targeted educational guidance before the operation, while the control group received only medication and dietary guidance. The baseline data, colonoscopy duration, bowel preparation compliance, Boston bowel preparation(BBPS)assessment score, intestinal bubble score, the incidence of adverse reactions during bowel preparation, and nursing appointment satisfaction were compared between the two groups.RESULTS The colonoscopy duration times and intestinal bubble scores of the study group were shorter than those of the control group, with statistically significant differences. The BBPS scores for the right, left, and interrupted colon in the study group were also higher than those in the control group, and the difference was statistically significant. Additionally, the study group had a higher rate of liquid diet one day before the examination, higher rate of correct bowel-clearing agent dilution method, higher rate of accurate time of ingesting the bowel-clearing agent, and a higher proportion of patients ingesting bowelclearing agent at the specified time than the control group, with statistically significant differences.The incidence of nausea and vomiting during bowel clearance in the study group was significantly lower than that in the control group. The incidence of abdominal pain, abdominal distension,dizziness, and fatigue was compared between the two groups, but the difference was not statistically significant. The scores of service attitude, detailed notification of dietary precautions, clear and easy-to-understand health educational content, and receiving care and comfort in the study group were significantly higher than those in the control group.CONCLUSION Preoperative special guidance and education were shown to significantly improve bowel clearance and compliance and reduce the incidence of adverse reactions in older adult patients with constipation undergoing colonoscopy. These factors are beneficial for improving patient satisfaction with nursing services.展开更多

It is comprised of 1 550 bp and an open reading frame (ORF) of 241 amino acids, with a predicted molecular mass of approximately 28 kDa. In situ hybridization histochemistry revealed a tissue-specific expression pattern of Amphi-PhLP with the high levels in the ovary, and at a lower level in the hind grit and testis, hepatic caecum, gill, endostyle, and epipbaryngeal groove, while it was absent in the muscle, neural tube and notochord. In the Chinese Hamster Ovary (CHO) cells transfected with the expression plasmid pEGFP-NI/Amphi-PhLP, the fusion protein was targeted in the cytoplasm of CHO cells, suggesting that Amphi-PhLP is a cytosolic protein. This work may provide a framework for further understanding of the physiological function of Amphi-PhLP in B. belcheri.展开更多

[ Method] Tumor animal model was successfully established with chickens infected by MDV. Real-time RT-PCR, histopathological and immunohistochemistrical methods were used to detect the pathological lesion, transcription level, expression level and distribution of HSP60 in the kidney. [ Result] Obvious pathological changes appeared in kidney tissue of chicken after injection of MDV for 21 d. HSP60 was mainly distributed in the cytoplasm of tumor cell and interstitial macrophages. The expression of HSP60 in challenge group was always higher than blank control group and vaccine control group, and the difference reached extremely significant level from 28 to 86 day-ages ( P 〈 0.01 ) ; the expression of HSP60 in challenge group at 28 day-age was about 8.608 and 12.752 times of blank control group and vaccine control group, respectively. The mRNA transcription levels of HSP60 in challenge group showed a downward parabola during 7 to 42 day-ages; the mRNA transcription level of HSP60 in challenge group was rising in early stage (7 -21 d), and then gradually dropped; it reached the peak value at 21 d, being 1.222 and 1.179 times of blank control group and vaccine control group, respectively. The mRNA transcription level of HSP60 was higher than two control groups throughout the entire experiment, and the difference reached extremely significant level from 14 to 28 day-agas (P 〈0.01 ). [ Conclusion] HSP60 expression in kidney tissue and its mRNA transcription level can reflect MDV infection, tumor formation and development progress, with close relationship with pathogenesis of tumor diseases, which is expected to be used as one of the determination indicators for diagnosis and morbidity process of tumor diseases in chicken.展开更多

Distribution data of sexes, ages, professions and regions of the cases were analyzed in this report. The results showed that the elderly cases, especially the male elderly, were significantly more than expected, which is different from human cases of H5N1 avian influenza and human cases of the pandemic H1N1 influenza. The outbreak was rated as a Grade Ⅲ (severe) outbreak, and it would evolve into a Grade IV (very severe) outbreak soon, using a method reported previously. The H7N9 AIV will probably circulate in humans, birds and pigs for years. Moreover, with the driving force of natural selection, the virus will probably evolve into highly pathogenic AIV in birds, and into a deadly pandemic influenza virus in humans. Therefore, the H7N9 outbreak has been assumed severe, and it is likely to become very or extremely severe in the future, highlighting the emergent need of forceful scientific measures to eliminate any infected animal flocks. We also described two possible mild scenarios of the future evolution of the outbreak.展开更多
Chicken embryo fibroblasts (CEFs) are among the most commonly used cells for the study of interactions between chicken hosts and H5N1 avian influenza virus (AIV).In this study,the expression of eleven housekeeping gen...Chicken embryo fibroblasts (CEFs) are among the most commonly used cells for the study of interactions between chicken hosts and H5N1 avian influenza virus (AIV).In this study,the expression of eleven housekeeping genes typically used for the normalization of quantitative real-time PCR (QPCR) analysis in mammals were compared in CEFs infected with H5N1 AIV to determine the most reliable reference genes in this system.CEFs cultured from 10-day-old SPF chicken embryos were infected with 100 TCID50 of H5N1 AIV and harvested at 3,12,24 and 30 hours post-infection.The expression levels of the eleven reference genes in infected and uninfected CEFs were determined by real-time PCR.Based on expression stability and expression levels,our data suggest that the ribosomal protein L4 (RPL4) and tyrosine 3-monooxygenase tryptophan 5-monooxygenase activation protein zeta polypeptide (YWHAZ) are the best reference genes to use in the study of host cell response to H5N1 AIV infection.However,for the study of replication levels of H5N1 AIV in CEFs,the β-actin gene (ACTB) and the ribosomal protein L4 (RPL4) gene are the best references.展开更多

By means of FQ-PCR, the transcription changes of HSP70 and HSP90 mRNA in the livers and hearts of transport stressed pigs were studied. The level of HSP70 mRNA transcription increased continuously from the beginning of transportation. The inductions of HSP70 mRNA transcription in the livers and hearts of 10 h transport stressed pigs were 2.5 and 4.1 times higher than that of the un-transport stressed pigs (P〈0.01). However, the transcription levels of HSP90 mRNA in the livers and hearts decreased with the transport stress.展开更多

文摘The H4 subtype of avian influenza virus(AIV)is prevalent worldwide,but only receives little attention due to its low pathogenicity in poultry.Consequently,it remains largely unclear whether H4 AIVs pose a potential threat to the poultry industry and public health.During the period from 2011 to 2022,we conducted an active surveillance programm.A total of 154,762 swab samples were collected across various provinces,and 427 H4 viruses were detected,resulting in a positivity rate of 0.28%.All H4 viruses were isolated from poultry,primarily from ducks in live poultry markets.We further investigated the genetic evolutionary characteristics and pathogenicity of 20 H4Nx viruses isolated in our program.Phylogenetic analysis revealed that the 20 H4Nx viruses belonged to the Eurasian lineage and exhibited significant genetic diversity,with 19 distinct genotypes identified.Molecular characterization indicated that these viruses were low-pathogenicity AIVs with limited binding affinity to human receptors,yet they contained mutations associated with enhanced viral replication and pathogenicity in mammals.Pathogenicity tests conducted in ducks demonstrated that H4 viruses were weakly pathogenic,exhibiting limited replication and transmission capabilities.However,some viruses were able to replicate effectively in mice and induce weight loss.For instance,DK/AH/AG61/11(H4N6)can replicate efficiently in MDCK cells,indicating a potential threat to mammals.These findings underscore the importance of ongoing surveillance of H4 AIVs to better understand their evolution and transmission dynamics and to prevent potential public health risks.

文摘Toxoplasma gondii(T.gondii)is a globally distributed parasite that can infect a diversity of warm-blooded animals,including swine and humans.Infection in swine poses a considerable threat to food safety and public health.The aim of this meta-analysis was to estimate the seroprevalence of T.gondii infection in the swine population in China from 2000 to 2023 and to examine potential factors associated with infection.A total of 112 studies were included,collectively involving 145,152 swine samples originating from 26 provinces.The pooled seroprevalence was 26.0%(95%CI:23.3%–28.7%).Stratified analysis based on diagnostic methods revealed that studies using the indirect hemagglutination assay(IHA)reported a seroprevalence of 19.7%(95%CI:17.2%–22.2%),whereas those utilizing the enzyme-linked immunosorbent assay(ELISA)reported a higher seroprevalence of 35.5%(95%CI:29.1%–41.8%).Geographical analysis indicated higher seroprevalence in the South Central and Southwest regions,whereas the East and Northwest areas reported the lowest seroprevalence.Chongqing Province reported the highest seroprevalence,reaching 44.9%(95%CI:43.4%–46.0%),followed by Xinjiang,Hainan,and Guizhou,whereas the lowest was observed in Shandong Province(3.5%,95%CI:1.7%–6.1%).These findings provide important epidemiological evidence that can inform strategies for the prevention and control of T.gondii infection in swine populations,with a focus on high-risk populations and geographical areas.This imperative contributes substantially to the improvement of both food safety and public health.
文摘One influenza H3N2 virus, A/swine/Shandong/3/2005 (Sw/SD/3/2005), was isolated from pigs with respiratory disease on a farm in eastern China. Genetic analysis revealed that Sw/SD/3/2005 was a triple-reassortant virus with a PB2 gene from human-like H1N1, NS from classical swine H1N1, and the remaining genes from human-like H3N2 virus. These findings further support the concept that swine can serve as reservoir or mixing vessels of influenza virus strains and maintain genetic and antigenic stability of viruses. Furthermore, we have successfully established a reverse genetics system based on eight plasmids and rescued Sw/SD/3/2005 through cell transfection. HI tests and RT-PCR confirmed that the rescued virus maintained the biological properties of the wild type Sw/SD/3/2005. The successful establishment of the reverse genetics system of Sw/SD/3/2005 will enable us to conduct extensive studies of the molecular evolution of H3N2 influenza viruses in swine.
文摘H9N2 avian influenza virus(AIV) has widely circulated in poultry worldwide and sporadic infections in humans and mammals. During our surveillance of chicken from 2019 to 2021 in Shandong Province, China, we isolated 11 H9N2AIVs. Phylogenetic analyses showed that the eight gene segments of the 11 isolates were closely related to several sublineages of Eurasian lineage: BJ/94-like clades(HA and NA genes), G1-like clades(PB2 and M genes), and SH/F/98-like clades(PB1, PA, NP and NS genes). The isolates showed mutation sites that preferentially bind to humanlike receptors(HA) and mammalian fitness sites(PB2, PB1 and PA), as well as mutations in antigen and drug resistance sites. Moreover, studies with mice revealed four isolates with varying levels of pathogenicity. The average antibody titer of the H9N2 AIVs was 8.60 log2. Based on our results, the epidemiological surveillance of H9N2 AIVs should be strengthened.

文摘[ Objective] In order to construct the specific equine disease-free zone of of the Guangzhou Asian Games, to ensure that the equestrian events of Guangzhou Asian Games be smoothly held. [Methods] The species, quantities, distribution and seasonal variations of insects and wild animals in related zones were investigated from 2008 to 2010, and the collected samples of the insects and wild animals were tested in laboratory for related equine diseases. [Results] The investigations indicated that there were some mosquitoes, flies, horseflies, punkies, ticks, bats, wild birds and wild bears in equastrianism venue and peripheral regions of disease-free zone of the Guangzhou Asian Games, the laboratory results of Japa- nese encephalitis, vesicular stomatitis, Nipah virus disease, West Nile fever, and Trypanosomiasis evansi, were all negative. According to the in- vestigations and test results, the risk assessments of insect and wild animal vectors in the prevalence of related equine diseases were made to con- firm that the risk was relatively low or very low, and comprehensive prevention and control measures with prevention as major measures and insecti- cides application and environment managements as supplementary means were made on the basis of the risk assessment conclusions. [ Conclu- sions] This research has laid a solid foundation for the successful building of the first specific equine disease-free zone in our country, ensured the smooth holding of the 16th equastdan competition in Guangzhou Asian Games.
文摘In order to reduce the risks for the spread of disease in cattle movements, we investigated China’s cattle breeding and movement pattern, analyzed risk factors of disease infection caused by long-distance movement, explored the relevant risk management measures and conjectured the direction of cattle movement based on the regional distribution of cattle inventory and beef price. We also constructed a market value chain in live cattle movements and qualitatively analyzed the risks for unapparent infection in the process of movement. Meanwhile, we put forward a long-term policy of reducing the risks for unapparent infection and animal health supervision measures.

文摘The cardiopulmonary health of children may be affected by acute ozone(O3)exposure during physical activity[1];however,its effects in high-altitude regions such as the Xizang Plateau remain uncertain.In high-altitude areas,lower oxygen levels may cause children to experience shortness of breath or require increased respiratory effort during vigorous activities such as running.This could lead to increased pollutant inhalation,potentially elevating the burden on the cardiovascular system and triggering adverse reactions such as increased heart rate and elevated blood pressure.Furthermore,differences in physiological adaptation between Han children who have migrated to Xizang and Tibetan children who are native to the region may contribute to different reactions to environmental exposure[2].
文摘Florfenicol(FLO)is a chemically synthesized broad-spectrum antimicrobial agent of amide alcohols for animals,which is one of the most widely used antimicrobials in livestock,poultry,and aquaculture.With the use of FLo,more and more attention has been paid to its hematopoietic toxicity,immunotoxicity,genotoxicity,and embryotoxicity.In this study,SPF chicks.at the age of 3 d began to drink water with the FLO at a dose of 100 mg L^(-1)for 6 consecutive days,and the growth performance of chicks was monitored,the effect of FLO on immune organs was detected by pathological examination and TdT-mediated dUTP nick-end labeling(TUNEL)apoptosis staining.In order to evaluate the level of organism immunity,the level of Newcastle disease virus antibody in serum was detected by hemagglutination inhibition test,the content of cytokines(IL-1,IL-2,IL-6,TNF-α,IFN-γ)in serum was detected by enzyme linked immunosorbent assay(ELISA),and the transcription of interferon-related genes(IRF-7,2′-5′OAS,Mx1)and cytokine genes(IL-6,TNF-α,IFN-γ)in immune organs were detected by real time fluorescence quantitative PCR.The results showed that the early application of FLO could inhibit the growth and development of chicks,and the body weight and immune organ index of the treatment group were lower than those of the control group.Histopathological examination showed that there was a decrease in the number of lymphocytes in the bursa of Fabricius in the treatment group in the early stage of drug withdrawal,and the results of TUNEL apoptosis staining in the bursa of Fabricius showed that obvious lymphocyte apoptosis occurred in the FLO treatment group.Compared with the control group,the transcription levels of interferon-related genes IRF-7,2′-5′OAS,and cytokine genes IL-6,TNF-αand IFN-γin FLO treatment group decreased to a certain extent,while the transcription level of Mx1 gene had no significant difference at all time points.The level of serum Newcastle disease virus(NDV)antibody and the contents of cytokines IL-1,IL-2 and IFN-γin the FLO treatment group were significantly lower than those in the control group in the early stage of drug withdrawal,but recovered gradually in the later stage.This study showed that FLO has a certain degree of effect on the immune function of chicks,and the results of the study laid the foundation for further research on the mechanism of FLO-induced immunotoxicity.

文摘A real-time RT-PCR assay using Taq Man-MGB probes was developed to detect and type the bovine viral diarrhea virus(BVDV) in cattle.Universal primers and Taq Man-MGB probes were designed from the 5′-untranslated region of known pestiviral sequences.Prior to optimizing the assay, c RNAs were transcribed in vitro from the BVDV 1 and BVDV 2 RTPCR products to make standard curves.The detection limit of the assay was 1.72×102 copies for BVDV 1 and 2.14×102copies for BVDV 2.The specificity of the assay evaluated on several BVDV strains including bovine herpesvirus 1(BHV 1), foot and mouth disease virus(FMDV) and several classical swine fever virus(CSFV) strains showed specific detection of the positive virus over 40 cycles.The assay was highly reproducible with the coefficient of variance ranging from 1.04 to 1.33% for BVDV 1 and from 0.83 to 1.48% for BVDV 2, respectively.Using this method, we tested a total of 2 327 cattle from three dairy farms for the presence of BVDV persistently infected(PI) animals.In this assay, each RT-PCR template contained a mixture of ten samples from different animals.The occurrence rate of PI cattle in three farms ranging from 0.9 to 2.54% could represent partly the PI rates in cattle farm in China.In conclusion, using our real-time PCR assay, we could effectively detect and type BVDV and identify PI cattle in a rapid and cost-effective manner.

文摘Platycodon grandiflorum(Jacq.)A.DC.is a famous medicinal plant commonly used in East Asia.Triterpene saponins isolated from P.grandiflorum are the main biologically active compounds,among which polygalacin D(PGD)has been reported to be an anti-tumor agent.However,its anti-tumor mechanism against hepatocellular carcinoma is unknown.This study aimed to explore the inhibitory effect of PGD in hepatocellular carcinoma cells and related mechanisms of action.We found that PGD exerted significant inhibitory effect on hepatocellular carcinoma cells through apoptosis and autophagy.Analysis of the expression of apoptosis-related proteins and autophagy-related proteins revealed that this phenomenon was attributed to the mitochondrial apoptosis and mitophagy pathways.Subsequently,using specific inhibitors,we found that apoptosis and autophagy had mutually reinforcing effects.In addition,further analysis of autophagy showed that PGD induced mitophagy by increasing BCL2 interacting protein 3 like(BNIP3L)levels.In vivo experiments demonstrated that PGD significantly inhibited tumor growth and increased the levels of apoptosis and autophagy in tumors.Overall,our findings showed that PGD induced cell death of hepatocellular carcinoma cells primarily through mitochondrial apoptosis and mitophagy pathways.Therefore,PGD can be used as an apoptosis and autophagy agonist in the research and development of antitumor agents.

文摘To investigate the dynamic change of heat shock protein 90 (Hsp90) in the genesis and development of tumor, we successfully established tumor animal model using Marek’s disease and then determined the location of Hsp90 in the tumor tissue using immunohistochemistry method, the antibody titer level of Hsp90 in the serum and the expression level in the tissue using enzyme-linked immunosorbent assay (ELISA) method. Our result showed that Hsp90 location in the tumor tissue was signiifcantly associated with the tumor cell and most in the cytoplasm of the tumor cell, and Hsp90 expression level in the tissue and the antibody titer level in the serum was most signiifcantly increased with the development of tumor. This is the ifrst report to show the presence of Hsp90 in tumor tissues induced by the Marek’s disease, with its expression correlated to the tumoral grading. These data may also be valuable for developing new molecular anti-cancer therapies.

文摘Vibrio parahaemolyticus (VP) is one of the pathogenic vibrios endangering net-cage cultured Pseudosciaena crocea,Fennerpenaeus chinensis, and shellfish in coastal areas of China. Several types of hemolysins produced by Vp have been characterized as major virulence factors.They are thermostable direct hemolysin (TDH),TDH-related hemolysin (TRH) and thermolabile hemolysin (TLH). In this study, we cloned tdh, trh, and tlh genes from the genome DNA of VP by polymerase chain reaction (PCR).We ligated the three genes into prokaryotic expression vector pET-28a (+),and transformed the recombinant plasmids into Es-cherichia coli BL21 (DE3). The expression of recombinant proteins was induced by isopropyl-β-D-thiogalacto-pyranoside (IPTG). The recombinant proteins were expressed in a form of inclusion bodies and thus purified with Ni-NTA affinity chromatography. Western blotting results showed that recombinant proteins,TDH, TRH and TLH, could be recognized by rabbit anti-VP serum. The three purified proteins were renatured by gradient dialysis.The renatured proteins exhibited hemolytic activity except for TLH in the presence of phosphatidylcholine. These results not only are helpful for better understanding these genes' functions under a single factor level, but also provide evidence for VP vaccine engineering.
文摘BACKGROUND The prevalence of constipation in the Chinese population over 60 years of age is 11.5%, and this prevalence increases with age, which seriously affects the quality of life in older adults. Therefore, reducing the incidence of constipation in older adults is necessary to promote a healthy lifestyle as well as biochemical health.AIM To explore the value of preoperative guidance and education to improve the effects of bowel cleaning in older adult patients undergoing colonoscopy.METHODS In this study, 160 older adult patients with constipation requiring colonoscopy at Shandong Provincial Hospital between January 2019 and March 2021 were selected and randomly divided into a study group and a control group, with 80patients in each group. The study group received medication guidance and targeted educational guidance before the operation, while the control group received only medication and dietary guidance. The baseline data, colonoscopy duration, bowel preparation compliance, Boston bowel preparation(BBPS)assessment score, intestinal bubble score, the incidence of adverse reactions during bowel preparation, and nursing appointment satisfaction were compared between the two groups.RESULTS The colonoscopy duration times and intestinal bubble scores of the study group were shorter than those of the control group, with statistically significant differences. The BBPS scores for the right, left, and interrupted colon in the study group were also higher than those in the control group, and the difference was statistically significant. Additionally, the study group had a higher rate of liquid diet one day before the examination, higher rate of correct bowel-clearing agent dilution method, higher rate of accurate time of ingesting the bowel-clearing agent, and a higher proportion of patients ingesting bowelclearing agent at the specified time than the control group, with statistically significant differences.The incidence of nausea and vomiting during bowel clearance in the study group was significantly lower than that in the control group. The incidence of abdominal pain, abdominal distension,dizziness, and fatigue was compared between the two groups, but the difference was not statistically significant. The scores of service attitude, detailed notification of dietary precautions, clear and easy-to-understand health educational content, and receiving care and comfort in the study group were significantly higher than those in the control group.CONCLUSION Preoperative special guidance and education were shown to significantly improve bowel clearance and compliance and reduce the incidence of adverse reactions in older adult patients with constipation undergoing colonoscopy. These factors are beneficial for improving patient satisfaction with nursing services.

文摘Dear Editor,Since it first appeared in humans in China in 2013,a novel H7N9influenza virus has caused 1,568 cases of infection,with a mortality rate of 39.2%(Yin et al.,2021).The early H7N9 virus strains had low pathogenicity and did not cause avian disease(Zhang et al.,2013).However,they caused concern about poultry consumption and caused huge economic losses to the poultry industry.Since mid-2016,some H7N9 viruses mutated.

文摘A full length amphioxus cDNA, encoding a novel phosducin-like protein (Amphi-PhLP), was identified for the first time from the gut cDNA library of Branchiostoma belcheri. It is comprised of 1 550 bp and an open reading frame (ORF) of 241 amino acids, with a predicted molecular mass of approximately 28 kDa. In situ hybridization histochemistry revealed a tissue-specific expression pattern of Amphi-PhLP with the high levels in the ovary, and at a lower level in the hind grit and testis, hepatic caecum, gill, endostyle, and epipbaryngeal groove, while it was absent in the muscle, neural tube and notochord. In the Chinese Hamster Ovary (CHO) cells transfected with the expression plasmid pEGFP-NI/Amphi-PhLP, the fusion protein was targeted in the cytoplasm of CHO cells, suggesting that Amphi-PhLP is a cytosolic protein. This work may provide a framework for further understanding of the physiological function of Amphi-PhLP in B. belcheri.

文摘[ Objective] The paper was to investigate the relationship between pathological lesion and transcription, expression and distribution of heat shock protein 60 (HSP60) in kidney of chickens infected by Marek's disease virus (MDV). [ Method] Tumor animal model was successfully established with chickens infected by MDV. Real-time RT-PCR, histopathological and immunohistochemistrical methods were used to detect the pathological lesion, transcription level, expression level and distribution of HSP60 in the kidney. [ Result] Obvious pathological changes appeared in kidney tissue of chicken after injection of MDV for 21 d. HSP60 was mainly distributed in the cytoplasm of tumor cell and interstitial macrophages. The expression of HSP60 in challenge group was always higher than blank control group and vaccine control group, and the difference reached extremely significant level from 28 to 86 day-ages ( P 〈 0.01 ) ; the expression of HSP60 in challenge group at 28 day-age was about 8.608 and 12.752 times of blank control group and vaccine control group, respectively. The mRNA transcription levels of HSP60 in challenge group showed a downward parabola during 7 to 42 day-ages; the mRNA transcription level of HSP60 in challenge group was rising in early stage (7 -21 d), and then gradually dropped; it reached the peak value at 21 d, being 1.222 and 1.179 times of blank control group and vaccine control group, respectively. The mRNA transcription level of HSP60 was higher than two control groups throughout the entire experiment, and the difference reached extremely significant level from 14 to 28 day-agas (P 〈0.01 ). [ Conclusion] HSP60 expression in kidney tissue and its mRNA transcription level can reflect MDV infection, tumor formation and development progress, with close relationship with pathogenesis of tumor diseases, which is expected to be used as one of the determination indicators for diagnosis and morbidity process of tumor diseases in chicken.

文摘Dozens of human cases infected with H7N9 subtype avian influenza virus (AIV) have been confirmed in China since March, 2013. Distribution data of sexes, ages, professions and regions of the cases were analyzed in this report. The results showed that the elderly cases, especially the male elderly, were significantly more than expected, which is different from human cases of H5N1 avian influenza and human cases of the pandemic H1N1 influenza. The outbreak was rated as a Grade Ⅲ (severe) outbreak, and it would evolve into a Grade IV (very severe) outbreak soon, using a method reported previously. The H7N9 AIV will probably circulate in humans, birds and pigs for years. Moreover, with the driving force of natural selection, the virus will probably evolve into highly pathogenic AIV in birds, and into a deadly pandemic influenza virus in humans. Therefore, the H7N9 outbreak has been assumed severe, and it is likely to become very or extremely severe in the future, highlighting the emergent need of forceful scientific measures to eliminate any infected animal flocks. We also described two possible mild scenarios of the future evolution of the outbreak.

文摘Chicken embryo fibroblasts (CEFs) are among the most commonly used cells for the study of interactions between chicken hosts and H5N1 avian influenza virus (AIV).In this study,the expression of eleven housekeeping genes typically used for the normalization of quantitative real-time PCR (QPCR) analysis in mammals were compared in CEFs infected with H5N1 AIV to determine the most reliable reference genes in this system.CEFs cultured from 10-day-old SPF chicken embryos were infected with 100 TCID50 of H5N1 AIV and harvested at 3,12,24 and 30 hours post-infection.The expression levels of the eleven reference genes in infected and uninfected CEFs were determined by real-time PCR.Based on expression stability and expression levels,our data suggest that the ribosomal protein L4 (RPL4) and tyrosine 3-monooxygenase tryptophan 5-monooxygenase activation protein zeta polypeptide (YWHAZ) are the best reference genes to use in the study of host cell response to H5N1 AIV infection.However,for the study of replication levels of H5N1 AIV in CEFs,the β-actin gene (ACTB) and the ribosomal protein L4 (RPL4) gene are the best references.
文摘The RNA transcripted in vitro was used as the standard quantitative template to make the standard curve and establish the fluorescence quantitative RT-PCR (FQ-PCR) method. By means of FQ-PCR, the transcription changes of HSP70 and HSP90 mRNA in the livers and hearts of transport stressed pigs were studied. The level of HSP70 mRNA transcription increased continuously from the beginning of transportation. The inductions of HSP70 mRNA transcription in the livers and hearts of 10 h transport stressed pigs were 2.5 and 4.1 times higher than that of the un-transport stressed pigs (P〈0.01). However, the transcription levels of HSP90 mRNA in the livers and hearts decreased with the transport stress.
