Synaptic mitochondria in aging and neurodegenerative diseases:Functional decline and vulnerability

Aging is a physiological and complex process produced by accumulative age-dependent cellular damage,which significantly impacts brain regions like the hippocampus,an essential region involved in memory and learning.A crucial factor contributing to this decline is the dysfunction of mitochondria,particularly those located at synapses.Synaptic mitochondria are specialized organelles that produce the energy required for synaptic transmission but are also important for calcium homeostasis at these sites.In contrast,non-synaptic mitochondria primarily involve cellular metabolism and long-term energy supply.Both pools of mitochondria differ in their form,proteome,functionality,and cellular role.The proper functioning of synaptic mitochondria depends on processes such as mitochondrial dynamics,transport,and quality control.However,synaptic mitochondria are particularly vulnerable to age-associated damage,characterized by oxidative stress,impaired energy production,and calcium dysregulation.These changes compromise synaptic transmission,reducing synaptic activity and cognitive decline during aging.In the context of neurodegenerative diseases such as Alzheimer’s,Parkinson’s,and Huntington’s,the decline of synaptic mitochondrial function is even more pronounced.These diseases are marked by pathological protein accumulation,disrupted mitochondrial dynamics,and heightened oxidative stress,accelerating synaptic dysfunction and neuronal loss.Due to their specialized role and location,synaptic mitochondria are among the first organelles to exhibit dysfunction,underscoring their critical role in disease progression.This review delves into the main differences at structural and functional levels between synaptic and non-synaptic mitochondria,emphasizing the vulnerability of synaptic mitochondria to the aging process and neurodegeneration.These approaches highlight the potential of targeting synaptic mitochondria to mitigate age-associated cognitive impairment and synaptic degeneration.This review emphasizes the distinct vulnerabilities of hippocampal synaptic mitochondria,highlighting their essential role in sustaining brain function throughout life and their promise as therapeutic targets for safeguarding the cognitive capacities of people of advanced age. 展开更多

The intricate landscape of neurodegenerative diseases complicates the search for effective therapeutic approaches.Photoreceptor degeneration,the common endpoint in various retinal diseases,including retinitis pigmentosa and age-related macular degeneration,leads to vision loss or blindness.While primary cell death is driven by genetic mutations,oxidative stress,and neuroinflammation,additional mechanisms contribute to disease progression.In retinitis pigmentosa,a multitude of genetic alterations can trigger the degeneration of photoreceptors,while other retinopathies,such as agerelated macular degeneration,are initiated by combinations of environmental factors,such as diet,smoking,and hypertension,with genetic predispositions.Nutraceutical therapies,which blend the principles of nutrition and pharmaceuticals,aim to harness the health benefits of bioactive compounds for therapeutic applications.These compounds generally possess multi-target effects.Polyphenols and flavonoids,secondary plant metabolites abundant in plant-based foods,are known for their antioxidant,neuroprotective,and anti-inflammatory properties.This review focuses on the potential of polyphenols and flavonoids as nutraceuticals to treat neurodegenerative diseases such as retinitis pigmentosa.Furthermore,the importance of developing reliable delivery methods to enhance the bioavailability and therapeutic efficacy of these compounds will be discussed.By combining nutraceuticals with other emerging therapies,such as genetic and cell-based treatments,it is possible to offer a more comprehensive approach to treating retinal degenerative diseases.These advancements could lead to a viable and accessible option,improving the quality of life for patients with retinal diseases. 展开更多

The development of clinical candidates that modify the natural progression of sporadic Parkinson's disease and related synucleinopathies is a praiseworthy endeavor,but extremely challenging.Therapeutic candidates that were successful in preclinical Parkinson's disease animal models have repeatedly failed when tested in clinical trials.While these failures have many possible explanations,it is perhaps time to recognize that the problem lies with the animal models rather than the putative candidate.In other words,the lack of adequate animal models of Parkinson's disease currently represents the main barrier to preclinical identification of potential disease-modifying therapies likely to succeed in clinical trials.However,this barrier may be overcome by the recent introduction of novel generations of viral vectors coding for different forms of alpha-synuclein species and related genes.Although still facing several limitations,these models have managed to mimic the known neuropathological hallmarks of Parkinson's disease with unprecedented accuracy,delineating a more optimistic scenario for the near future. 展开更多

Eucalyptus staigeriana essential oil(EsEO)has well-known anthelmintic activity in small ruminants.However,its volatility limits its therapeutic action.The aim of this study was to develop a water-in-oil sodium alginatebased nanoemulsion with an effective in vitro effect on the eggs and larvae of Haemonchus contortus,a gastrointestinal parasite of sheep and goats.Four oil-in-water sodium alginate-based emulsions were prepared using a high-energy method with different proportions of Tween 80,EsEO,and sodium alginate(ALG)4%.The physical-chemical characterization included stability,particle size,zeta potential and infrared spectra.The effects of the emulsions were evaluated against H.contortus via the egg hatching test(EHT)and larval development test(LDT).The results showed that the emulsions were stable over 7 days,nanometer scale particles(218.8 to 371.5 nm)predominating and with negative zeta potentials(−28.9 to−46.9 mV).All four emulsions were more effective than EsEO in the EHT,with 50%effective concentrations(EC50)of 0.088 to 0.15 mg/mL for the emulsions and 0.308 mg/mL for EsEO.However,in the LDT,the emulsions and essential oil had similar effects,with EC50 values of 3.91 to 4.60 mg/mL for the emulsions and 4.17 mg/mL for EsEO.Emulsion 2,with low Tween 80/EsEO and ALG/EsEO ratios,was considered better overall in terms of physical,chemical and anthelmintic assessment and is a promising candidate for further in vivo assays against adult H.contortus. 展开更多

Acalypha gaumeri(Euphorbiaceae)is the only endemic species of the genus in the Yucatan Peninsula.It is dioecious and has antifungal properties against various phytopathogens.In the present study,molecular identification of A.gaumeri was performed using the rbcL region,confirming its belonging to the Acalypha genus.Its genetic diversity was evaluated using 10 SPAR markers(ISSR and DAMD)from 60 individuals collected from female and male plants of the Kiuic,Tinum and Yaxcaba ex-situ populations.The results showed a high level of genetic polymorphism(PIC=0.980)and significant differences among the populations.Ethanol and aqueous extracts from leaves,stems,and roots of both genders and three populations were evaluated against three phytopathogenic fungi.Only the ethanol extracts of the roots showed inhibitory antifungal activity,whereas Kiuic and Tinum,both male and female individuals,showed inhibitory effects at 1000μg/mL against the three pathogens.The ethanol extract of the female flowering plant of Kiuic showed activity at minimum inhibitory concentrations of 250μg/mL against Alternaria chrysanthemi CICY004 and 500μg/mL against Colletotrichum gloeosporioides CICY002 and Penicillium oxalicum ITC25.Chromatographic profile of the ethanol extracts confirmed the presence of alkaloidal components in the ex-situ cultivated plants,which were analyzed by HPLC.The results revealed that the peaks at T_(R) of 7.60,7.88,and 8.49 min were the most abundant components(9.95%-21.93%),with differences between female and male plants of the three populations.This research confirms that A.gaumeri cultivated and genetically characterized is a potential raw material to develop an eco-friendly product for the control of fungal diseases in crops. 展开更多

The mechanisms leading to neurological and neurodegenerative diseases are not completely known,and new,more effective,therapeutic treatments are necessary for most neurological pathologies.The treatment of neurological and neurodegenerative diseases is complicated due to the blood-brain barrier,which makes it difficult for drugs to access the brain areas in which they must act to improve the pathology.A tool that can help to overcome this difficulty is the use of extracellular vesicles,which can easily cross the blood-brain barrier.The extracellular vesicles are considered a main way of communication between the brain and the rest of the body,with important implications for the physiopathology and therapy of neurological diseases.In recent years,the involvement of microbiota in many neurological pathologies,as well as its possible therapeutic role,has also become evident.A key mediator in the pathologic and beneficial effects of microbiota seems to be the bacterial extracellular vesicles.There is an important communication between the brain and the intestinal microbiota(the gut-brain axis),by which the microbiota influences brain function,impacts on mental health,and plays a role in different neurological and neurodegenerative diseases.The identification of the mechanisms involved in this gut-brain axis is essential to understanding the mechanisms of neurological pathologies and to developing more effective treatments for these diseases.Bacterial extracellular vesicles would play a relevant role in these processes.This review compiles the recent information and evidence on the role of bacterial extracellular vesicles in brain pathologies and on the therapeutic utility of bacterial extracellular vesicles in neurological and neurodegenerative diseases.One advantage of bacterial extracellular vesicles compared to extracellular vesicles derived from other cell types,such as stem cells,is that bacterial extracellular vesicles are generally easier to produce and modify.Bacterial extracellular vesicles may be easily modified to target a specific pathology and/or to enhance its therapeutic efficacy.Although the studies are still scarce,they open a wide field of possibilities for future studies,which will lead to a deeper understanding of the role of microbiota and bacterial extracellular vesicles in neurological pathologies and the underlying mechanisms,as well as to the development of new treatments based on the use of bacterial extracellular vesicles in neurological diseases. 展开更多

In this study,artificial neural networks(ANNs)were implemented to determine design parameters for an impressed current cathodic protection(ICCP)prototype.An ASTM A36 steel plate was tested in 3.5%NaCl solution,seawater,and NS4 using electrochemical impedance spectroscopy(EIS)to monitor the evolution of the substrate surface,which affects the current required to reach the protection potential(Eprot).Experimental data were collected as training datasets and analyzed using statistical methods,including box plots and correlation matrices.Subsequently,ANNs were applied to predict the current demand at different exposure times,enabling the estimation of electrochemical parameters(limiting voltage values)that can be used to optimize a self-regulating ICCP system.The obtained electrochemical parameters were then used,through Particle Swarm Optimization(PSO),to fine-tune an ANN-based proportional-integral-derivative(PID)controller for the ICCP system. 展开更多

To ensure the efficient use of resources,particularly in water-scarce arid and semi-arid regions where abiotic stress threatens food security,assessing soil and climate suitability for specific crops is crucial.Simultaneously,food production must align with sustainable development goals by minimizing negative environmental impacts.Therefore,establishing agro-climatic suitability using a spatiotemporal approach is essential.This involves three key steps:first,determining the climatically appropriate months based on the species’requirements(temporal suitability),and second,establishing the soil suitability of specific plots(spatial suitability).Following this,quantifying crop evapotranspiration allows for optimized water use.This study used climatic and soil variables from diverse data sources to characterize the study area.Subsequently,suitability classes for Portulaca oleracea were determined based on existing literature.Our analysis concerning temporal suitability revealed that June and July are the optimal months for sowing this species in all of the municipalities.Spatially,approximately 30%of the agricultural land use of the study area exhibits a highly suitable class in most municipalities.Both dimensions,the temporal and the spatially,were validated through Chi square(χ^(2))Goodness-of-Fit test and theχ^(2)test of independence,respectively.Consequently,for a one-month production cycle during periods of high suitability,estimated evapotranspiration values are between 210 and 245 mm.In brief,the study area demonstrates favorable agro-climatic conditions for P.oleracea cultivation in specific months of the year according to parameters used,with potential in a large proportion of agricultural land and achievable water requirements. 展开更多

Mexican oregano(Lippia graveolens Kunth)is an aromatic species of high culinary and medicinal relevance.In Mexico,more than 40 taxa with characteristic aroma and flavor are commercially recognized as oregano,with L.graveolens being the most widely distributed and economically important.Despite its relevance,few domesticated or semi-domesticated cultivars exist,and wild populations remain the main source of raw material,raising concerns regarding sustainability and quality standardization.The essential oil and oleoresins of L.graveolens possess recognized bioactivity,including antioxidant,antifungal,antibacterial,and anti-inflammatory properties,largely attributed to phenolic compounds such as thymol and carvacrol.Given the increasing global demand for natural bioactives and functional foods,optimizing cultivation practices is essential to enhance both the yield and phytochemical quality of this species.This study evaluated the effect of organic fertilization(0,5,10,and 15 t ha^(−1)of vermicompost)on the phenolic profile,flavonoid content,and antioxidant capacity of oregano oleoresin obtained from a semi-domesticated population across three harvests.The highest phenolic concentration(≈500 mg GAE g^(−1)extract)and greatest antioxidant activity(ABTS>3.5×10^(5)μmol TE g^(−1)extract)were observed at the 5 t ha^(−1)dose during the second harvest.Flavonoid content peaked in the third harvest(480-620 mg QE g^(−1)extract),whereas the unfertilized control exhibited the highest DPPH activity in the first harvest.Overall,the results indicate that vermicompost dosage and harvest timing substantially influence the functional quality of L.graveolens.Moderate organic fertilization,particularly 5 t ha^(−1),enhances the biosynthesis of bioactive secondary metabolites,underscoring its potential for sustainable production systems. 展开更多

This study presents the design,construction,and thermal evaluation of a solar-powered cocoa roaster based on a Parabolic Cylinder Collector(PCC)with dual-axis solar tracking.The system integrates three functional subsystems:the cylindrical-parabolic reflecting surface,the stainless-steel absorber tube,and a microcontrollerbased tracking mechanism.The prototype enables continuous acquisition of key thermal variables(solar irradiance,ambient temperature,absorber surface temperature,and bean temperature),allowing a detailed characterization of heat transfer processes during roasting.Roasting experiments were conducted at controlled durations of 40,55,and 70 min between 10:00 and 14:00 h.Maximum roasting temperatures of 125℃–137℃ were reached under average irradiance levels of 685.7–930.5 W m−2.The lowest final moisture content was 2.19%,within the recommended range for high-quality cocoa.Longer roasting durations promoted thermal energy accumulation within the absorber tube,enhancing convective and radiative heat transfer to the bean mass even under fluctuating irradiance.The experimental trends reveal a strong coupling between irradiance variability,absorber temperature,and internal air-beam heat transfer.Comparison with reference parabolic trough collector studies indicate that,although the process-level roasting efficiency(3.83%–7.45%)is lower than conventional collector-level thermal efficiencies,the operating temperatures and moisture-reduction rates align with the thermal requirements of food-processing systems rather than high-enthalpy solar applications.These results also demonstrate the potential of coupling PCC-based solar concentration with lowtemperature convective–radiative roasting processes.Overall,the findings confirm the feasibility of implementing PCC-based roasting technologies in rural or off-grid regions,where solar-driven heat transfer offers a sustainable,low-cost alternative to fossil-fuel-based roasting systems,enabling a controlled thermophysical environment for cocoa transformation. 展开更多

Backgroud Efficient communication between the embryo and the endometrium is essential for the successful establishment and maintenance of pregnancy.Uterine-derived extracellular vesicles(EVs)contribute to embryomaternal communication,supporting early embryonic development.This study aimed to:(i)compare the protein cargo of uterine fluid EVs(UF-EVs)from CYCLIC and PREGNANT heifers;(ii)characterize the protein profile of conditioned medium(CM)-EVs from endometrial explants cultured alone(EXPL)or co-cultured with five d 7 blastocysts(EXPL+EMB)in vitro;and(iii)compare the EV protein cargo between the in vivo and in vitro models(i.e.,EXPL vs.CYCLIC and EXPL+EMB vs.PREGNANT).Results We identified 1,459 and 1,752 proteins in the UF-EVs of CYCLIC and PREGNANT heifers,respectively.Among these,12 were exclusive to CYCLIC,and 18 were exclusive to PREGNANT.Among the 1,329 proteins identified in both groups,16 were differently abundant;ten were more abundant,and six were less abundant in UF-EVs from PREGNANT heifers.In vivo,the changes in UF-EV protein cargo induced by the presence of a blastocyst were related to inflammatory and immune responses,endometrial receptivity,and support of early embryonic development by promoting cell polarity,cell–cell adhesion,and stem cell differentiation.In vitro,we identified 1,501 proteins in the CM-EVs from EXPL,1,975 in the CM-EVs from EXPL+EMB,and 82 in the CM-EVs from EMB.Additionally,50 proteins were unique to EXPL+EMB,and another 33 were differentially abundant due to the synergistic interaction between the embryo and the endometrium.These proteins are involved in embryonic development,regulation of stem cell differentiation,establishment and maintenance of cell polarity,interferon tau(IFNT)-mediated cell signaling,endometrial receptivity,and immune modulation.Although there are qualitative and quantitative differences between in vivo and in vitro-derived EVs,UF-EVs from CYCLIC heifers compared to CM-EVs from EXPL,as well as UFEVs from PREGNANT heifers compared to CM-EVs from EXPL+EMB shared common proteins.Conclusions These findings highlight the pivotal role of EVs in embryo-maternal communication,suggesting that their protein cargo may actively contribute to the modulation of the uterine environment to support early embryonic development.Understanding these molecular interactions could provide valuable insights into the mechanisms of implantation and pregnancy establishment. 展开更多

Background:The Colorectal Cancer(CRC)pathogenesis and therapeutic efficacy are influenced by the gut microbiome,making it a promising biomarker for predicting treatment responses and adverse effects.This systematic review aims to outline the gut microbiome composition in individuals with CRC undergoing the same therapeutic regimen and evaluate interindividual microbiome profile variations to better understand how these differences may influence therapeutic outcomes.Methods:Key studies investigating the microbiome’s role in therapeutic approaches for CRC were searched in both PubMed and Cochrane databases on 12 and 22 March 2025,respectively.Eligible studies included free full-text English-language randomized clinical trials and human observational studies reporting on gut microbiome composition and treatment outcomes.RoB 2 and ROBINS-I were employed in the evaluation of bias for randomized trials and observational studies,respectively.Data extracted was narratively analyzed.Results:Six studies involving a total of 361 individuals were included.Therapeutic interventions,either standard treatments and/or those targeting the gut microbiome,generally increased probiotic taxa and reduced pro-carcinogenic bacteria.However,no consistent pattern of improved clinical outcomes was observed,suggesting that treatment mechanisms,the tumor’s nature,and individual characteristics play critical roles in microbiome modulation.Conclusion:The gut microbiome holds significant potential in clinical settings.Nonetheless,further research is needed to better understand its functional aspects and to consider the influence of treatment mechanisms,the tumor’s nature,and individual characteristics as modulators,in order to optimize clinical outcomes. 展开更多

In floodplain wetlands,alterations in hydrological patterns resulting from climate change and human activities could potentially diminish the carbon sequestration capacity of the soils,thereby having a negative impact on global climate change.However,the magnitude of the influence of hydrological regime change on soil carbon remains inadequately monitored.To address this research gap,we collected 306 upper layer(0–20 cm)soil samples from the Dongting Lake floodplain between 2013 and 2022.The random forest(RF)algorithm was used to analyze the spatial distribution of soil organic carbon(SOC)in the upper soil layer of Dongting Lake floodplain and the impact of climate and hydrological changes in the past decade on surface SOC in the East Dongting Lake area was studied.In 2022,the SOC concentration of the Dongting Lake floodplain upper layer soil ranged from 3.34 to 17.67 g kg^(-1),averaging 10.43 g kg^(-1),with a corresponding SOC density of(2.65±0.49)kg m^(-2) and total SOC stock of 6.82 Tg C(2.87–13.48 Tg C).From 2013 to 2022,the SOC concentration of the upper soil layer of the East Dongting Lake area decreased from 18.37 to 10.82 g kg^(-1).This reduction could be attributed to climate and hydrological changes which reduce SOC input by reducing vegetation growth and accelerating SOC decomposition.Above 21.4 m elevation,the amount of SOC loss increased with elevation,the loss being related to the decline in Miscanthus community biomass and greater susceptibility of higher altitude areas to climate and hydrological changes.Our results highlight the need for strengthening wetland SOC management to increase SOC in the soils to help combat climate change. 展开更多

The brain is the most complex human organ,and commonly used models,such as two-dimensional-cell cultures and animal brains,often lack the sophistication needed to accurately use in research.In this context,human cerebral organoids have emerged as valuable tools offering a more complex,versatile,and human-relevant system than traditional animal models,which are often unable to replicate the intricate architecture and functionality of the human brain.Since human cerebral organoids are a state-of-the-art model for the study of neurodevelopment and different pathologies affecting the brain,this field is currently under constant development,and work in this area is abundant.In this review,we give a complete overview of human cerebral organoids technology,starting from the different types of protocols that exist to generate different human cerebral organoids.We continue with the use of brain organoids for the study of brain pathologies,highlighting neurodevelopmental,psychiatric,neurodegenerative,brain tumor,and infectious diseases.Because of the potential value of human cerebral organoids,we describe their use in transplantation,drug screening,and toxicology assays.We also discuss the technologies available to study cell diversity and physiological characteristics of organoids.Finally,we summarize the limitations that currently exist in the field,such as the development of vasculature and microglia,and highlight some of the novel approaches being pursued through bioengineering. 展开更多

However, these invasive... Geotechnical studies are essential in civil engineering for all building and infrastructure projects. Typically, in-situ studies involving soil sample collection through drilling are conducted. However, these invasive methods can be costly when numerous boreholes are needed to assess stratum continuity or are impractical for examining subsurface conditions beneath existing structures. Shallow geophysical exploration offers several non-invasive alternatives for subsurface characterization, with Electrical Resistivity Tomography (ERT) being particularly versatile. ERT provides detailed and accurate subsurface images through a relatively simple and fast field implementation. For this study, four 2D ERT profiles were designed and performed near three buildings at the Centro Universitario de la Costa in Puerto Vallarta, Jalisco, Mexico, using a Gito-1100 V resistivity meter from Hematec with Dipole-Dipole arrays. Basic descriptive statistics were calculated for each data set to establish criteria for outlier removal, optimizing the inversion process in Res2DInv software. The inversion results defined five geoelectric units [UG-1 (1 to 20 Ω‧m), UG-2 (20 to 40 Ω‧m), UG-3 (40 to 100 Ω‧m), UG-4 (100 to 500 Ω‧m), and UG-5 (750 to 1000 Ω‧m)], consistent with previously identified geologic materials. The 2D ERT profiles allowed for the identification of lateral variations in moisture content and saturation and determined the depth of consolidated and possibly cemented materials suitable for future infrastructure projects on the university campus. This work provides a reference framework for implementing the 2D-ERT technique in Puerto Vallarta, supporting its use as a non-invasive alternative for effective subsurface characterization in geotechnical and civil engineering contexts. 展开更多
