BACKGROUND The high mortality rate and recurrence/metastasis remain major challenges in the clinical management of gastric cancer(GC)patients.To optimize treatment stratification and management,there is an urgent need...BACKGROUND The high mortality rate and recurrence/metastasis remain major challenges in the clinical management of gastric cancer(GC)patients.To optimize treatment stratification and management,there is an urgent need for efficient and non-invasive biomarkers.A meta-analysis on the prognostic role of circulating tumor cells(CTCs)in GC revealed a strong association between CTCs and patient prognosis.Among CTC subtypes,Interstitial CTCs(I-CTCs)exhibited the strongest invasiveness.This study innovatively investigated the expression profile of I-CTCs in advanced GC patients to evaluate their clinical utility.AIM To evaluate the clinical utility of I-CTCs as a non-invasive prognostic biomarker in advanced GC.To investigate the correlation between I-CTC count thresholds and chemotherapy efficacy in advanced GC patients.To establish the potential of preoperative I-CTC profiling for optimizing treatment stratification and postoperative surveillance.METHODS This study retrospectively analyzed 59 patients with advanced GC treated at the General Surgery Clinical Medical Center of Gansu Provincial Hospital between October 2019 and October 2020.The expression levels of I-CTCs were measured,and patient survival was monitored.The receiver operating characteristic curve was plotted to determine the optimal cut-off value for I-CTCs expression levels.Based on this cut-off value,59 GC patients were grouped into positive and negative groups.The differences in clinicopathological characteristics between the two groups were analyzed.Patient survival was follow-up and recorded until October 2022.Plotting survival curves and performing univariate and multifactorial analyses of patient prognostic factors.The Kaplan-Meier method and Cox regression model were used,respectively.RESULTS A total of 59 patients were included in this study,and receiver operating characteristic curve analysis showed that the best cut-off value for I-CTCs was 5,with an area under the curve of 0.8356(95%CI:0.7122-0.9590).The I-CTC count of≥5 defines the positive group,while counts<5 are classified as the negative group.Positive I-CTCs correlated with the degree of tumor differentiation and disease progression(P<0.05).16 of 59 patients received neoadjuvant chemotherapy.There were divided into progressive disease and disease control groups based on response to neoadjuvant chemotherapy.Patients in the I-CTCs-negative group had longer overall survival and disease-free survival than those in the positive group(P<0.05).Multifactorial analysis revealed that I-CTCs positivity(HR=13.323,95%CI:1.675-105.962,P=0.014)was an independent risk factor for survival in patients with advanced GC.CONCLUSION In patients with advanced GC,an I-CTC count of≥5 is associated with both poor prognosis and reduced chemotherapy efficacy.I-CTCs may serve as a valuable preoperative biomarker for predicting the prognosis of advanced GC.展开更多
Mitochondria are well-acknowledged as ideal targets for tumor therapy due to their important role in energy supply and cellular signal regulation.Mitochondria-specific photosensitizers have been reported to be critica...Mitochondria are well-acknowledged as ideal targets for tumor therapy due to their important role in energy supply and cellular signal regulation.Mitochondria-specific photosensitizers have been reported to be critical for inducing cell apoptosis.Two-photon fluorescence imaging provides a new technique for delineating biological structures and activities in deep tissues.Herein,we developed a new aggregation-induced emission(AIE)active photosensitizer by attaching a pyridinium group for mitochondrial target-ing.The rationally designed photosensitizer(TTTP)exhibited excellent photophysical properties,good biocompatibility,reactive oxygen species(ROS)stimulation ability,anticancer efficacy,and two-photon imaging properties.TTTP was highly taken up by cells and accumulated specifically in mitochondria but was selectively cytotoxic to cancer cells.Under light irradiation,the generation of ROS was significantly boosted,leading to actively induced apoptosis.The in vivo tumor photodynamic therapeutic efficacy of TTTP showed significant inhibition of tumor growth.Furthermore,the underlying mechanism of TTTP tu-mor suppression revealed that the apoptosis agonist Bax was markedly up-regulated while the antagonist Bcl-xL was down-regulated.This research provides a potential mitochondrial-targeted phototherapeutic agent for effective therapy and two-photon fluorescence imaging.展开更多

A fundamental requirement for proper measurement of well-being in diverse contexts is the appropriate translation of well-being measures into the languages spoken by the specific population.The aim of this paper is to...A fundamental requirement for proper measurement of well-being in diverse contexts is the appropriate translation of well-being measures into the languages spoken by the specific population.The aim of this paper is to identify measures of well-being that have been translated into African languages up to the year 2019 and make suggestions for researchers who are faced with the challenge of translating well-being instruments into local languages.Online databases were searched to identify published studies reporting the translation of well-being instruments into African languages.Some researchers were further contacted and requested to provide relevant studies.A total of 352 publications were retrieved and 44 translated measures met the inclusion criteria.Findings showed that all the translated African language versions of existing measures were published between 2006 and 2019.Although the published translations were increasingly widespread,the distribution of available studies was uneven,with very low published translation activity in East and North Africa and a preponderance of publications on translated instruments in South Africa.The authors suggest deriving population norms for relevant translations;increasing funding and other resources for translation projects;developing cross-national collaborations on translations;and making the translated versions of well-being instruments more accessible for use by other researchers.展开更多

Extracellular matrix(ECM)stiffening is a typical characteristic of cartilage aging,which is a quintessential feature of knee osteoarthritis(KOA).However,little is known about how ECM stiffening affects chondrocytes an...Extracellular matrix(ECM)stiffening is a typical characteristic of cartilage aging,which is a quintessential feature of knee osteoarthritis(KOA).However,little is known about how ECM stiffening affects chondrocytes and other molecules downstream.This study mimicked the physiological and pathological stiffness of human cartilage using polydimethylsiloxane(PDMS)substrates.It demonstrated that epigenetic Parkin regulation by histone deacetylase 3(HDAC3)represents a new mechanosensitive mechanism by which the stiffness matrix affected chondrocyte physiology.We found that ECM stiffening accelerated cultured chondrocyte senescence in vitro,while the stiffness ECM downregulated HDAC3,prompting Parkin acetylation to activate excessive mitophagy and accelerating chondrocyte senescence and osteoarthritis(OA)in mice.Contrarily,intra-articular injection with an HDAC3-expressing adeno-associated virus restored the young phenotype of the aged chondrocytes stimulated by ECM stiffening and alleviated OA in mice.The findings indicated that changes in the mechanical ECM properties initiated pathogenic mechanotransduction signals,promoted the Parkin acetylation and hyperactivated mitophagy,and damaged chondrocyte health.These results may provide new insights into chondrocyte regulation by the mechanical properties of ECM,suggesting that the modification of the physical ECM properties may be a potential OA treatment strategy.展开更多

Prostate cancer(PCa)exhibits epidemiological and molecular heterogeneity.Despite extensive studies of its phenotypic and genetic properties in Western populations,its molecular basis is not clear in Chinese patients.T...Prostate cancer(PCa)exhibits epidemiological and molecular heterogeneity.Despite extensive studies of its phenotypic and genetic properties in Western populations,its molecular basis is not clear in Chinese patients.To determine critical molecular characteristics and explore correlations between genomic markers and clinical parameters in Chinese populations,we applied an integrative genetic/transcriptomic assay that combines targeted next-generation sequencing and quantitative real-time PCR(qRT-PCR)on samples from 46 Chinese patients with PCa.Lysine(K)-specific methyltransferase 2D(KMT2D),zinc finger homeobox 3(ZFHX3),A-kinase anchoring protein 9(AKAP9),and GLI family zinc finger 1(GLI1)were frequently mutated in our cohort.Moreover,a clinicopathological analysis showed that RB transcriptional corepressor 1(RB1)deletion was common in patients with a high risk of disease progression.Remarkably,four genomic events,MYC proto-oncogene(MYC)amplification,RB1 deletion,APC regulator of WNT signaling pathway(APC)mutation or deletion,and cyclin-dependent kinase 12(CDK12)mutation,were correlated with poor disease-free survival.In addition,a close link between KMT2D expression and the androgen receptor(AR)signaling pathway was observed both in our cohort and in The Cancer Genome Atlas Prostate Adenocarcinoma(TCGA-PRAD)data.In summary,our results demonstrate the feasibility and benefits of integrative molecular characterization of PCa samples in disease pathology research and personalized medicine.展开更多

However, the treatment options are very limited. The aim of this study was to investigate effects of electroacupuncture (EA) on visceral hypersensitivity in a rat model of IBS and to explore the underlying mechanisms of EA effects. Visceral hypersensitivity was established by neonatal maternal deprivation (NMD) in male rats on postnatal days 2 - 15. Behavioral experiments were conducted at the age of 7 weeks. Treatment with EA at Zusanli (stomach-36, ST-36) significantly reduced abdominal withdrawal reflex (AWR) scores in NMD rats but not in age-matched healthy control rats. In addition, EA treatment hyperpolarized resting membrane potentials, increased the rheobase and reduced the numbers of action potentials evoked by 2 and 3 times rheobase current stimulation of dorsal root ganglion (DRG) neurons innervating the colon. NMD markedly enhanced expression of TRPV1 in colon related DRGs while EA treatment drastically suppressed the expression of TRPV1 in DRGs of NMD rats. These data suggest that EA treatment produced an analgesic effect, which might be mediated at least in a part by suppression of TRPV1 expression and by inhibition of neuronal excitability of primary sensory neurons in rats with chronic visceral pain.展开更多

METHODS: A keyword search was conducted on the SCIE journals on the Thomson Reuters website(http://www.thomsonscientific.com.cn) with the search formula "Alternative OR Complementary OR Integrative OR Chinese Medicine OR Medicinal OR Ethnopharmacology OR Manipulative OR Natural Medicine OR Natural Product OR Phytotherapy OR Phytomedicine OR Homeopathy OR Acupuncture OR Planta OR Traditional OR Ayurvedic OR Herb OR Herbal". RESULTS: A total of 32 kinds of traditional medicine journals were collected by SCI, of which 30 are in English and the rest are in German.The subject of traditional medical journals covered by SCI covers traditional medicine, supplementary and alternative therapies, botanicals, and natural medicines. Chinese medicine professionals in China can take these factors into consideration when using literature or choosing the direction of paper submission. CONCLUSION: The IF of traditional medicine related journals are relative low, while those of biology and natural drugs are relative high. IF should not be considered as the only factor to evaluate the impact and academic level of journals.展开更多

Organ-on-a-chip stands as a pivotal platform for skeletal muscle research while constructing 3D skeletal muscle tissues that possess both macroscopic and microscopic structures remains a considerable challenge.This st...Organ-on-a-chip stands as a pivotal platform for skeletal muscle research while constructing 3D skeletal muscle tissues that possess both macroscopic and microscopic structures remains a considerable challenge.This study draws inspiration from LEGO-like assembly,employing a modular approach to construct muscle tissue that integrates biomimetic macroscopic and microscopic structures.Modular LEGO-like hybrid nanofibrous scaffold bricks were fabricated by the combination of 3D printing and electrospinning techniques.Skeletal muscle cells cultured on these modular scaffold bricks exhibited a highly orientated nanofibrous structure.A variety of construction of skeletal muscle tissues further enabled development by various assembling processes.Moreover,skeletal muscle-on-a-chip(SMoC)was further assembled as a functional platform for electrical or perfusion stimuli investigation.The electrical stimulus was conveniently applied and tuned in such a SMoC platform to significantly enhance the differentiation of skeletal muscle tissues.Additionally,the effect of perfusion stimulation on skeletal muscle vascularization within the SMoC platform was also demonstrated.These findings highlight the potential of these assembled SMoCs as functional ex vivo platforms for skeletal tissue engineering and drug research applications,and such a LEGO-like assembly strategy could also be applied to the other engineering organ-on-chips fabrication,which facilitates the development of bionic functional platforms for various biomedical research applications.展开更多

Currently,stem cell transplantations in cardiac repair are limited owing to disadvantages,such as immunological rejection and poor cell viability.Although direct injection of exosomes can have a curative effect simila...Currently,stem cell transplantations in cardiac repair are limited owing to disadvantages,such as immunological rejection and poor cell viability.Although direct injection of exosomes can have a curative effect similar to that of stem cell transplantation,high clearance hinders its application in clinical practice.Previous reports suggested that induction of coronary collateralization can be a desired method of adjunctive therapy for someone who had missed the optimal operation time to attenuate myocardial ischemia.In this study,to mimic the paracrine and biological activity of stem cells,we developed artificial stem cells that can continuously release Tβ4-exosomes(Tβ4-ASCs)by encapsulating specific exosomes within microspheres using microfluidics technology.The results show that Tβ4-ASCs can greatly promote coronary collateralization in the periphery of the myocardial infarcted area,and its therapeutic effect is superior to that of directly injecting the exosomes.In addition,to better understand how it works,we demonstrated that the Tβ4-ASC-derived exosomes can enhance the angiogenic capacity of coronary endothelial cells(CAECs)via the miR-17-5p/PHD3/Hif-1αpathway.In brief,as artificial stem cells,Tβ4-ASCs can constantly release functional exosomes and stimulate the formation of collateral circulation after myocardial infarction,providing a feasible and alternative method for clinical revascularization.展开更多

In this study, we evaluated the T cell response to melanoma-associated ...Background: Development of innovative immunotherapy is imperative to improve the poor survival of the nasopharyngeal carcinoma (NPC) patients. In this study, we evaluated the T cell response to melanoma-associated antigen (MAGE)-A1, MAGE-A3, or synovial sarcoma X-2 (SSX-2) in the peripheral blood of treatment-naive NPC patients. The relationship of responses among the three proteins and the human leukocyte antigen (HLA)-A types were analyzed to provide evidence of designing novel therapy. Methods: Sixty-one NPC patients admitted into the Tumor Hospital affiliated to the Xinjiang Medical University between March 2015 and July 2016 were enrolled. Mononuclear cells were isolated from the peripheral blood before any treatment. HLA-A alleles were typed with Sanger sequence-based typing technique. The T cell response to the MAGE-A1, MAGE-A3, or SSX-2 was evaluated with the Enzyme-Linked ImmunoSpot assay. Mann-Whitney U-test was used to compare the T cell responses from different groups. Spearman's rank correlation was used to analyze the relationship of T cell responses. Results: HLA-A*02:01, A*02:07, and A*24:02 were the three most frequent alleles (18.9%, 12.3%, and 11.5%, respectively) among the 22 detected alleles. 31.1%, 19.7%, and 16.4% of the patients displayed MAGE-A1, MAGE-A3, or SSX-2-specific T cell response, respectively. The magnitudes of response to the three proteins were 32.5, 38.0, and 28.7 SFC/106 peripheral blood mononuclear cells, respectively. The T cell response against the three proteins correlated with each other to different extent. The percentage of A*02:01 and A*24:02 carriers were significantly higher in patients responding to any of the three proteins compared to the nonresponders. Conclusion: MAGE-A1, MAGE-A3, or SSX-2-specific T cell responses were detectable in a subgroup of NPC patients, the frequency and magnitude of which were correlated.展开更多

文摘BACKGROUND The high mortality rate and recurrence/metastasis remain major challenges in the clinical management of gastric cancer(GC)patients.To optimize treatment stratification and management,there is an urgent need for efficient and non-invasive biomarkers.A meta-analysis on the prognostic role of circulating tumor cells(CTCs)in GC revealed a strong association between CTCs and patient prognosis.Among CTC subtypes,Interstitial CTCs(I-CTCs)exhibited the strongest invasiveness.This study innovatively investigated the expression profile of I-CTCs in advanced GC patients to evaluate their clinical utility.AIM To evaluate the clinical utility of I-CTCs as a non-invasive prognostic biomarker in advanced GC.To investigate the correlation between I-CTC count thresholds and chemotherapy efficacy in advanced GC patients.To establish the potential of preoperative I-CTC profiling for optimizing treatment stratification and postoperative surveillance.METHODS This study retrospectively analyzed 59 patients with advanced GC treated at the General Surgery Clinical Medical Center of Gansu Provincial Hospital between October 2019 and October 2020.The expression levels of I-CTCs were measured,and patient survival was monitored.The receiver operating characteristic curve was plotted to determine the optimal cut-off value for I-CTCs expression levels.Based on this cut-off value,59 GC patients were grouped into positive and negative groups.The differences in clinicopathological characteristics between the two groups were analyzed.Patient survival was follow-up and recorded until October 2022.Plotting survival curves and performing univariate and multifactorial analyses of patient prognostic factors.The Kaplan-Meier method and Cox regression model were used,respectively.RESULTS A total of 59 patients were included in this study,and receiver operating characteristic curve analysis showed that the best cut-off value for I-CTCs was 5,with an area under the curve of 0.8356(95%CI:0.7122-0.9590).The I-CTC count of≥5 defines the positive group,while counts<5 are classified as the negative group.Positive I-CTCs correlated with the degree of tumor differentiation and disease progression(P<0.05).16 of 59 patients received neoadjuvant chemotherapy.There were divided into progressive disease and disease control groups based on response to neoadjuvant chemotherapy.Patients in the I-CTCs-negative group had longer overall survival and disease-free survival than those in the positive group(P<0.05).Multifactorial analysis revealed that I-CTCs positivity(HR=13.323,95%CI:1.675-105.962,P=0.014)was an independent risk factor for survival in patients with advanced GC.CONCLUSION In patients with advanced GC,an I-CTC count of≥5 is associated with both poor prognosis and reduced chemotherapy efficacy.I-CTCs may serve as a valuable preoperative biomarker for predicting the prognosis of advanced GC.

文摘Mitochondria are well-acknowledged as ideal targets for tumor therapy due to their important role in energy supply and cellular signal regulation.Mitochondria-specific photosensitizers have been reported to be critical for inducing cell apoptosis.Two-photon fluorescence imaging provides a new technique for delineating biological structures and activities in deep tissues.Herein,we developed a new aggregation-induced emission(AIE)active photosensitizer by attaching a pyridinium group for mitochondrial target-ing.The rationally designed photosensitizer(TTTP)exhibited excellent photophysical properties,good biocompatibility,reactive oxygen species(ROS)stimulation ability,anticancer efficacy,and two-photon imaging properties.TTTP was highly taken up by cells and accumulated specifically in mitochondria but was selectively cytotoxic to cancer cells.Under light irradiation,the generation of ROS was significantly boosted,leading to actively induced apoptosis.The in vivo tumor photodynamic therapeutic efficacy of TTTP showed significant inhibition of tumor growth.Furthermore,the underlying mechanism of TTTP tu-mor suppression revealed that the apoptosis agonist Bax was markedly up-regulated while the antagonist Bcl-xL was down-regulated.This research provides a potential mitochondrial-targeted phototherapeutic agent for effective therapy and two-photon fluorescence imaging.
文摘Developing biomimetic hydrogel-based scaffolds that can mimic the native anisotropic skeletal muscle tissue structure for inducing 3D aligned myogenic differentiation remains an on going challenge.Herein,we presented a method to 3D bioprint fibrinogen and GelMA via free printing into the support gel to engineer 3D oriented skeletal muscle scaffolds.A 3D aligned biomimetic scaffold based on this double-network hydrogel bioink was further 3D printed within the Carbopol supported gel by using a printing gel-in-gel strategy to mimic the native anisotropic structure of skeletal muscle tissue,and the cell cultivation experiment performed on myoblast-laden 3D bioprinted aligned biomimetic scaffolds indicated the material's ability to guide 3D myoblast alignment and elongation.
文摘A fundamental requirement for proper measurement of well-being in diverse contexts is the appropriate translation of well-being measures into the languages spoken by the specific population.The aim of this paper is to identify measures of well-being that have been translated into African languages up to the year 2019 and make suggestions for researchers who are faced with the challenge of translating well-being instruments into local languages.Online databases were searched to identify published studies reporting the translation of well-being instruments into African languages.Some researchers were further contacted and requested to provide relevant studies.A total of 352 publications were retrieved and 44 translated measures met the inclusion criteria.Findings showed that all the translated African language versions of existing measures were published between 2006 and 2019.Although the published translations were increasingly widespread,the distribution of available studies was uneven,with very low published translation activity in East and North Africa and a preponderance of publications on translated instruments in South Africa.The authors suggest deriving population norms for relevant translations;increasing funding and other resources for translation projects;developing cross-national collaborations on translations;and making the translated versions of well-being instruments more accessible for use by other researchers.

文摘Extracellular matrix(ECM)stiffening is a typical characteristic of cartilage aging,which is a quintessential feature of knee osteoarthritis(KOA).However,little is known about how ECM stiffening affects chondrocytes and other molecules downstream.This study mimicked the physiological and pathological stiffness of human cartilage using polydimethylsiloxane(PDMS)substrates.It demonstrated that epigenetic Parkin regulation by histone deacetylase 3(HDAC3)represents a new mechanosensitive mechanism by which the stiffness matrix affected chondrocyte physiology.We found that ECM stiffening accelerated cultured chondrocyte senescence in vitro,while the stiffness ECM downregulated HDAC3,prompting Parkin acetylation to activate excessive mitophagy and accelerating chondrocyte senescence and osteoarthritis(OA)in mice.Contrarily,intra-articular injection with an HDAC3-expressing adeno-associated virus restored the young phenotype of the aged chondrocytes stimulated by ECM stiffening and alleviated OA in mice.The findings indicated that changes in the mechanical ECM properties initiated pathogenic mechanotransduction signals,promoted the Parkin acetylation and hyperactivated mitophagy,and damaged chondrocyte health.These results may provide new insights into chondrocyte regulation by the mechanical properties of ECM,suggesting that the modification of the physical ECM properties may be a potential OA treatment strategy.

文摘Prostate cancer(PCa)exhibits epidemiological and molecular heterogeneity.Despite extensive studies of its phenotypic and genetic properties in Western populations,its molecular basis is not clear in Chinese patients.To determine critical molecular characteristics and explore correlations between genomic markers and clinical parameters in Chinese populations,we applied an integrative genetic/transcriptomic assay that combines targeted next-generation sequencing and quantitative real-time PCR(qRT-PCR)on samples from 46 Chinese patients with PCa.Lysine(K)-specific methyltransferase 2D(KMT2D),zinc finger homeobox 3(ZFHX3),A-kinase anchoring protein 9(AKAP9),and GLI family zinc finger 1(GLI1)were frequently mutated in our cohort.Moreover,a clinicopathological analysis showed that RB transcriptional corepressor 1(RB1)deletion was common in patients with a high risk of disease progression.Remarkably,four genomic events,MYC proto-oncogene(MYC)amplification,RB1 deletion,APC regulator of WNT signaling pathway(APC)mutation or deletion,and cyclin-dependent kinase 12(CDK12)mutation,were correlated with poor disease-free survival.In addition,a close link between KMT2D expression and the androgen receptor(AR)signaling pathway was observed both in our cohort and in The Cancer Genome Atlas Prostate Adenocarcinoma(TCGA-PRAD)data.In summary,our results demonstrate the feasibility and benefits of integrative molecular characterization of PCa samples in disease pathology research and personalized medicine.
文摘Irritable bowel syndrome (IBS) is characterized by chronic visceral hypersensitivity that companied by altered bowel movement. However, the treatment options are very limited. The aim of this study was to investigate effects of electroacupuncture (EA) on visceral hypersensitivity in a rat model of IBS and to explore the underlying mechanisms of EA effects. Visceral hypersensitivity was established by neonatal maternal deprivation (NMD) in male rats on postnatal days 2 - 15. Behavioral experiments were conducted at the age of 7 weeks. Treatment with EA at Zusanli (stomach-36, ST-36) significantly reduced abdominal withdrawal reflex (AWR) scores in NMD rats but not in age-matched healthy control rats. In addition, EA treatment hyperpolarized resting membrane potentials, increased the rheobase and reduced the numbers of action potentials evoked by 2 and 3 times rheobase current stimulation of dorsal root ganglion (DRG) neurons innervating the colon. NMD markedly enhanced expression of TRPV1 in colon related DRGs while EA treatment drastically suppressed the expression of TRPV1 in DRGs of NMD rats. These data suggest that EA treatment produced an analgesic effect, which might be mediated at least in a part by suppression of TRPV1 expression and by inhibition of neuronal excitability of primary sensory neurons in rats with chronic visceral pain.

文摘OBJECTIVE: To give a general introduction of journals on traditional medicine, which have been collected by the Science Citation Index(SCI). METHODS: A keyword search was conducted on the SCIE journals on the Thomson Reuters website(http://www.thomsonscientific.com.cn) with the search formula "Alternative OR Complementary OR Integrative OR Chinese Medicine OR Medicinal OR Ethnopharmacology OR Manipulative OR Natural Medicine OR Natural Product OR Phytotherapy OR Phytomedicine OR Homeopathy OR Acupuncture OR Planta OR Traditional OR Ayurvedic OR Herb OR Herbal". RESULTS: A total of 32 kinds of traditional medicine journals were collected by SCI, of which 30 are in English and the rest are in German.The subject of traditional medical journals covered by SCI covers traditional medicine, supplementary and alternative therapies, botanicals, and natural medicines. Chinese medicine professionals in China can take these factors into consideration when using literature or choosing the direction of paper submission. CONCLUSION: The IF of traditional medicine related journals are relative low, while those of biology and natural drugs are relative high. IF should not be considered as the only factor to evaluate the impact and academic level of journals.

文摘Organ-on-a-chip stands as a pivotal platform for skeletal muscle research while constructing 3D skeletal muscle tissues that possess both macroscopic and microscopic structures remains a considerable challenge.This study draws inspiration from LEGO-like assembly,employing a modular approach to construct muscle tissue that integrates biomimetic macroscopic and microscopic structures.Modular LEGO-like hybrid nanofibrous scaffold bricks were fabricated by the combination of 3D printing and electrospinning techniques.Skeletal muscle cells cultured on these modular scaffold bricks exhibited a highly orientated nanofibrous structure.A variety of construction of skeletal muscle tissues further enabled development by various assembling processes.Moreover,skeletal muscle-on-a-chip(SMoC)was further assembled as a functional platform for electrical or perfusion stimuli investigation.The electrical stimulus was conveniently applied and tuned in such a SMoC platform to significantly enhance the differentiation of skeletal muscle tissues.Additionally,the effect of perfusion stimulation on skeletal muscle vascularization within the SMoC platform was also demonstrated.These findings highlight the potential of these assembled SMoCs as functional ex vivo platforms for skeletal tissue engineering and drug research applications,and such a LEGO-like assembly strategy could also be applied to the other engineering organ-on-chips fabrication,which facilitates the development of bionic functional platforms for various biomedical research applications.

文摘To the editor:Targeted protein degradation(TPD)drugs are designed to link the protein of interest(POI)and protein degradation regulator(PDR),thus aiding in the degradation of the POI through cellular degradation mechanisms,including the ubiquitineproteasomal and lysosomal pathways.The lysosomal pathway can be further divided into the endosomalelysosomal degradation pathway and the autophagicelysosomal degradation pathway.Proteolysis-targeting chimeras(PROTAC),molecular glue,and chaperone-mediated protein degradation(CHAMP)are designed for targeting the ubiquitineproteasomal degradation pathway.Technologies utilizing the endosomalelysosomal pathway for protein degradation include AbTAC and PROTAB,while those leveraging the autophagicelysosomal pathway include autophagosome-tethering compound(ATTEC),autophagy-targeting chimera(AUTOTAC).

文摘Currently,stem cell transplantations in cardiac repair are limited owing to disadvantages,such as immunological rejection and poor cell viability.Although direct injection of exosomes can have a curative effect similar to that of stem cell transplantation,high clearance hinders its application in clinical practice.Previous reports suggested that induction of coronary collateralization can be a desired method of adjunctive therapy for someone who had missed the optimal operation time to attenuate myocardial ischemia.In this study,to mimic the paracrine and biological activity of stem cells,we developed artificial stem cells that can continuously release Tβ4-exosomes(Tβ4-ASCs)by encapsulating specific exosomes within microspheres using microfluidics technology.The results show that Tβ4-ASCs can greatly promote coronary collateralization in the periphery of the myocardial infarcted area,and its therapeutic effect is superior to that of directly injecting the exosomes.In addition,to better understand how it works,we demonstrated that the Tβ4-ASC-derived exosomes can enhance the angiogenic capacity of coronary endothelial cells(CAECs)via the miR-17-5p/PHD3/Hif-1αpathway.In brief,as artificial stem cells,Tβ4-ASCs can constantly release functional exosomes and stimulate the formation of collateral circulation after myocardial infarction,providing a feasible and alternative method for clinical revascularization.
文摘Background: Development of innovative immunotherapy is imperative to improve the poor survival of the nasopharyngeal carcinoma (NPC) patients. In this study, we evaluated the T cell response to melanoma-associated antigen (MAGE)-A1, MAGE-A3, or synovial sarcoma X-2 (SSX-2) in the peripheral blood of treatment-naive NPC patients. The relationship of responses among the three proteins and the human leukocyte antigen (HLA)-A types were analyzed to provide evidence of designing novel therapy. Methods: Sixty-one NPC patients admitted into the Tumor Hospital affiliated to the Xinjiang Medical University between March 2015 and July 2016 were enrolled. Mononuclear cells were isolated from the peripheral blood before any treatment. HLA-A alleles were typed with Sanger sequence-based typing technique. The T cell response to the MAGE-A1, MAGE-A3, or SSX-2 was evaluated with the Enzyme-Linked ImmunoSpot assay. Mann-Whitney U-test was used to compare the T cell responses from different groups. Spearman's rank correlation was used to analyze the relationship of T cell responses. Results: HLA-A*02:01, A*02:07, and A*24:02 were the three most frequent alleles (18.9%, 12.3%, and 11.5%, respectively) among the 22 detected alleles. 31.1%, 19.7%, and 16.4% of the patients displayed MAGE-A1, MAGE-A3, or SSX-2-specific T cell response, respectively. The magnitudes of response to the three proteins were 32.5, 38.0, and 28.7 SFC/106 peripheral blood mononuclear cells, respectively. The T cell response against the three proteins correlated with each other to different extent. The percentage of A*02:01 and A*24:02 carriers were significantly higher in patients responding to any of the three proteins compared to the nonresponders. Conclusion: MAGE-A1, MAGE-A3, or SSX-2-specific T cell responses were detectable in a subgroup of NPC patients, the frequency and magnitude of which were correlated.
