A machine learning model for predicting abnormal liver function induced by a Chinese herbal medicine preparation(Zhengqing Fengtongning)in patients with rheumatoid arthritis based on real-world study

Objective Rheumatoid arthritis(RA)is a systemic autoimmune disease that affects the small joints of the whole body and degrades the patients’quality of life.Zhengqing Fengtongning(ZF)is a traditional Chinese medicine preparation used to treat RA.ZF may cause liver injury.In this study,we aimed to develop a prediction model for abnormal liver function caused by ZF.Methods This retrospective study collected data from multiple centers from January 2018 to April 2023.Abnormal liver function was set as the target variable according to the alanine transaminase(ALT)level.Features were screened through univariate analysis and sequential forward selection for modeling.Ten machine learning and deep learning models were compared to find the model that most effectively predicted liver function from the available data.Results This study included 1,913 eligible patients.The LightGBM model exhibited the best performance(accuracy=0.96)out of the 10 learning models.The predictive metrics of the LightGBM model were as follows:precision=0.99,recall rate=0.97,F1_score=0.98,area under the curve(AUC)=0.98,sensitivity=0.97 and specificity=0.85 for predicting ALT<40 U/L;precision=0.60,recall rate=0.83,F1_score=0.70,AUC=0.98,sensitivity=0.83 and specificity=0.97 for predicting 40≤ALT<80 U/L;and precision=0.83,recall rate=0.63,F1_score=0.71,AUC=0.97,sensitivity=0.63 and specificity=1.00 for predicting ALT≥80 U/L.ZF-induced abnormal liver function was found to be associated with high total cholesterol and triglyceride levels,the combination of TNF-αinhibitors,JAK inhibitors,methotrexate+nonsteroidal anti-inflammatory drugs,leflunomide,smoking,older age,and females in middle-age(45-65 years old).Conclusion This study developed a model for predicting ZF-induced abnormal liver function,which may help improve the safety of integrated administration of ZF and Western medicine. 展开更多

Efficient stabilizing agent-free synthesis of gold nanoparticles via square-wave pulse deposition for enhanced catalytic performance in ethanol electrooxidation

The pressing environmental concerns and the depletion of fossil fuel reserves necessitate a transition toward sustainable energy sources.Ethanol,a renewable biomass-derived fuel,is a promising alternative due to its availability and high energy density.This study investigates the synthesis of gold nanoparticles(Au NPs)via a square-wave pulse deposition technique,aiming to enhance catalytic activity for ethanol electrooxidation.By varying pulse durations,we were able to exert precise control over Au NP size and distribution without stabilizing agents.Characterization using field emission scanning electron microscopy and X-ray diffraction techniques confirmed the formation of clustered nanoparticles of metallic gold phase.Electrochemical characteristics analyses revealed that Au NPs synthesized with a 900 ms pulse duration exhibited the lowest charge transfer resistance and the highest electrochemically active surface area.The electrocatalytic performance test of these Au NPs demonstrated an anodic current density of 2.5 mA cm^-(2)and a Tafel slope of 78 mV dec^(-1),indicating superior catalytic performance and reaction kinetics.Additionally,the Au NPs showed high resistance to poisoning,as evidenced by a low j_(b)/j_(f)ratio of 0.28 and stable chronoamperometric response.These findings underscore the potential of this synthesis method for producing high-performance electrocatalysts utilized in exploiting ethanol's potential as an environmentally friendly energy carrier. 展开更多

Halogenated organic pollutants in aquatic invertebrate–Fish food webs:Global distributions and trophic magnification

Persistent halogenated organic pollutants(HOPs)are globally concerned owing to their persistence,bioaccumulation,and potential toxicity to aquatic organisms.The trophic transfer of HOPs can affect their toxic effects on both humans and ecosystems.Previous studies mostly focused on HOPs biomagnification within specific regional or ecosystem contexts,however,the variation in trophicmagnification factors(TMFs)among different HOPs across a spectrum of diverse aquatic systems globally remains unclear.By collating literatures encompassing TMFs of polychlorinated biphenyls(PCBs),polybrominated diphenyl ethers(PBDEs),or perfluoroalkyl acids(PFAAs)along invertebrate–fish food webs on a global scale,our analysis revealed that Asia,North America,and Europe exhibited the highest pollution levels of HOPs.PCBs were found to have the highest trophicmagnification capability(TMF=3.14),followed by PBDEs(TMF=2.48)and PFAAs(TMF=1.97).TMF values and the probability of TMF exceeding 1 for PCB,PBDE and PFAA congeners showed a parabolic relationship with halogenation degree and carbon chain length.Specifically,PCBswith 6 chlorines,PBDEs with 5 bromines,and PFAAs with 12 carbons exhibited maximum trophic transfer capability and potential along food webs.Fractionation of PCBs and PFAAs was observed,with the proportions of higher chlorination degree and longer chain PFAAs increasing while the others decreased with trophic levels.Surprisingly,we also found a positive relationship between site latitude and TMF values of HOP congeners.This study provides a valuable basis for gauging ecological and health risks of HOPs,facilitating the development of robust standards for aquatic environment management. 展开更多

Objectives Retinal ischemia-reperfusion(RIR)injury results in irreversible visual impairments.The disruption of the outer blood-retinal barrier(OBRB)is a major ocular pathogenic process that RIR injury affects.Current clinical strategies are limited.This study aimed to elucidate how electroacupuncture(EA)protects the OBRB against RIR injury.Methods Male Wistar rats(7 weeks old,250 g to 280 g)were used in this study.Three independent experiments were conducted.First,Opioid peptide levels were quantified using enzyme-linked immunosorbent assay(ELISA).42 rats were randomly divided into 7 groups(n=6/group):Control:No treatment;high intraocular pressure(HIOP):Acute intraocular pressure elevation-induced RIR injury;HIOP+SHAM EA:RIR injury+sham EA at Xinming(Extra acupoint)and Jingming(BL1)for 30 min(shallow needle insertion but without electric stimulation);HIOP+2 Hz EA:RIR injury+2 Hz EA at Xinming and BL1 for 30 min;HIOP+100 Hz:RIR injury+100 Hz EA at Xinming and BL1 for 30 min;HIOP+2/100 Hz EA:RIR injury+2/100 Hz EA at Xinming and BL1 for 30 min;HIOP+4/20 Hz EA:RIR injury+4/20 Hz EA at Xinming and BL1 for 30 min.Second,retinal morphology was assessed by hematoxylin and eosin(HE)staining.20 rats were randomly allocated into 4 groups(n=5/group):Control:No treatment;HIOP:Acute intraocular pressure elevation-induced RIR injury;HIOP+SHAM EA:RIR injury+sham EA at Xinming and BL1 for 30 min(shallow needle insertion but without electric stimulation);HIOP+2 Hz EA:RIR injury+2 Hz EA at Xinming and BL1 for 30 min.Third,the permeability of OBRB was evaluated using the fluorescein isothiocyanate(FITC)-dextran leakage assay.15 rats were randomly divided into 5 groups(n=3/group):Control:No treatment;HIOP:Acute intraocular pressure elevation-induced RIR injury;HIOP+SHAM EA:RIR injury+sham EA at Xinming and BL1 for 30 min(shallow needle insertion but without electric stimulation);HIOP+2 Hz EA:RIR injury+2 Hz EA at Xinming and BL1 for 30 min;Nal+HIOP+2 Hz EA:Intravitreal injection ofδ-opioid receptor antagonist Naltridole(10µl,100 nM)30 min before RIR injury induction,followed by 2 Hz EA treatment at Xinming and BL1 for 30 min.In vitro studies examined enkephalins'effects on oxygen–glucose deprivation/reperfusion(OGD/R)induced injury in ARPE‐19 cells.Cell viability was evaluated by cell counting kit-8(CCK-8)assay,and morphological changes were recorded by Molecular Devices.Apoptosis was detected by Annexin V-FITC flow cytometry.Delta opioid receptor(DOR)expression in total protein and membrane protein were analyzed by western blotting(WB).Immunofluorescence(IF)staining and WB assessed ZO-1 and Claudin-19.For cell-based assays,n indicates the number of biologically independent replicates.Results It was found that 2 Hz EA treatment increased enkephalins(methionine-enkephalin and leucine-enkephalin)levels(P<0.01),restoring the increased retinal thickness(P<0.05)and mitigating RGCs loss(P<0.05)post-RIR injury.FITC-dextran leakage in the outer retina was ameliorated by 2 Hz EA(P<0.05),reversibly countered by Naltrindole(P<0.05),a DOR antagonist.Treatment with 30µM enkephalins enhanced ARPE-19 cell viability(P<0.001,P<0.0001)and inhibited apoptosis(P<0.0001).Enkephalins elevated DOR levels in total protein(P<0.05)and membrane protein fractions(P<0.001,P<0.0001),as well as elevated ZO-1(P<0.001,P<0.01)and Claudin-19(P<0.0001,P<0.001)levels following OGD/R,counteracted by Naltrindole.Conclusion It was found that 2 Hz EA inhibits the breakdown of OBRB via enkephalins activate DOR in RIR injury. 展开更多

The stability and evolution of human genetics depend on chromosomes and chromosome-chromosome interactions.We wish to understand the spatial location of chromosomes in dividing cells in order to understand the relationship between chromosome-chromosome interactions and to further investigate the role of chromosomes and their impact on cell biological behavior.In this study,we explored the relative spatial positional relationships of chromosomes[t(9;22)and t(15;17)]in B-ALL cells by using the three-dimensional DNA fluorescent in situ hybridization(3D-FISH)method.The results showed that chromosomes[t(9;22)and t(15;17)]showed relatively stable spatial relationships.The relative stability of the spatial location of chromosomes in dividing cells may be relevant to disease. 展开更多

Cobalt-manganese nano catalysts were prepared by sol-gel method. This research investigated the effects of different cobalt-manganese (Co/Mn = 1/1) loading, pH and calcination conditions on the catalytic performance of Co-Mn/TiO2 catalysts for Fischer-Tropsch synthesis (FTS) in a fixed bed reactor. It was found that the catalyst containing 30wt%(Co-Mn)/TiO2 was an optimal catalyst for the conversion of synthesis gas to light olefins especially propylene. The activity and selectivity of optimal catalyst were studied under different operational conditions. The results showed that the best operational conditions were H2/CO = 1/1 molar feed ratio at 250 ℃ and GHSV = 1300 h-1 un- der atmospheric pressure. Characterization of catalysts was carried out by X-ray diffraction (XRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM), N2 adsorption-desorlation measurements. 展开更多

The catalytic activity of seventeen metal methanesulfonates was compared under the same condition, ferrous methanesufonate proved to be the best. It can be easily recovered and reused for several times without distinct deterioration in catalytic activity. AIM To evaluate reproducibility of pulmonary embolism(PE) clot volume quantification using computed tomography pulmonary angiogram(CTPA) in a multicenter setting.METHODS This study was performed using anonymized data ... AIM To evaluate reproducibility of pulmonary embolism(PE) clot volume quantification using computed tomography pulmonary angiogram(CTPA) in a multicenter setting.METHODS This study was performed using anonymized data in conformance with HIPAA and IRB Regulations(March 2015-November 2016). Anonymized CTPA data was acquired from 23 scanners from 18 imaging centers using each site's standard PE protocol. Two independent analysts measured PE volumes using a semi-automated region-growing algorithm on an FDA-approved image analysis platform. Total thrombus volume(TTV) was calculated per patient as the primary endpoint. Secondary endpoints were individual thrombus volume(ITV), Qanadli score and modified Qanadli score per patient. Inter-and intra-observer reproducibility were assessed using intra-class correlation coefficient(ICC) and BlandAltman analysis. RESULTS Analyst 1 found 72 emboli in the 23 patients with a mean number of emboli of 3.13 per patient with a range of 0-11 emboli per patient. The clot volumes ranged from 0.0041-47.34 cm3(mean +/-SD, 5.93 +/-10.15 cm3). On the second read, analyst 1 found the same number and distribution of emboli with a range of volumes for read 2 from 0.0041 – 45.52 cm3(mean +/-SD, 5.42 +/-9.53 cm3). Analyst 2 found 73 emboli in the 23 patients with a mean number of emboli of 3.17 per patient with a range of 0-11 emboli per patient. The clot volumes ranged from 0.00459-46.29 cm3(mean +/-SD, 5.91 +/-10.06 cm3). Inter-and intraobserver variability measurements indicated excellent reproducibility of the semi-automated method for quantifying PE volume burden. ICC for all endpoints was greater than 0.95 for inter-and intra-observer analysis. Bland-Altman analysis indicated no significant biases.CONCLUSION Semi-automated region growing algorithm for quantifying PE is reproducible using data from multiple scanners and is a suitable method for image analysis in multicenter clinical trials. 展开更多

OBJECTIVE:To study the morphological basis of the role of Siqi(cold as winter,cool as autumn,warm as spring,hot as summer),Wuwei(five flavors:sweet,pungent,salty,sour,and bitter),and Guijing(meridian tropism) through the use of information integration.METHODS:A14C-2-deoxy-glucose autoradiography method was adopted to determine the overall impact of treatment with 39 herbs on functions of various tissues and organs.Data was measured at 4hs after a single dose and following the last treatment of repeated doses for a week.Least-squares estimation was used and fitted for each herb regression effect of organs and tissues after singleand repeated treatment.The slope of the regression line represented the cumulative trend of the effect of the herbs(β),and the standard deviation of the slope(Sβ) was compared with those of the untreated animals(t 'test).All significantly cumulative effect trends were applied with an artificial neural network,which integrated the relationship among Siqi,Wuwei,and Guijing with tissues and organs.RESULTS:There is a certain relationship among the Siqi,Wuwei,Guijing and the anatomy of organs and tissues,but the different scores indicate that influence of Siqi,Wuwei,Guijing to anatomy of organs and tissues was a nonlinear state.CONCLUSION:Results demonstrated that the effects of Siqi,Wuwei,and Guijing have a morphological basis,and each concept was associated with multiple anatomical structures. 展开更多

Azulene, one of representative nonbenzenoid aromatic hydrocarbons, exhibits unique molecular structure and distinctive physical and chemical properties. Herein, azulenoisoindigo(Az II), an azulene-based isoindigo analogue, is designed and synthesized, which has a twisted molecular backbone and R/Sisomers in single crystals. Interestingly, Az II shows the characteristics of both isoindigo and azulene,such as reversible redox behavior and reversible proton responsiveness. UV-vis-NIR,^(1)H NMR and electron paramagnetic resonance(EPR) measurements were carried out to get insights into the possible mechanism of the proton-responsive property of Az II. The results demonstrated that only one azulenyl moiety of molecule of Az II was protonated and deprotonated, and the protonated Az II can be further oxidized to form azulenium cation radicals. 展开更多

Nanosized Fe-Co catalysts were prepared by co-precipitation method and studied for the conversion of synthesis gas to light olefins.In particular,the effects of a range of preparation variables such as Co/Fe molar ratios of the precipitation solution,pH value of precipitate,temperature of precipitation,promoters and loading of optimum promoter on the structure and catalytic performance are investigated.The optimal nano catalyst for light olefins (C2-C4) production was obtained over the catalyst with Co/Fe molar ratio of 3/1 which promoted with 2 wt% K.The results show that the best operational conditions were GHSV=2200 h^-1 (H2/CO=2/1) at 260℃ under atmospheric pressure.Characterization of catalysts were carried out using X-ray diffraction (XRD),thermal gravimetric analysis (TGA),differential scanning calorimetry (DSC),scanning electron microscopy (SEM),transmission electron microscopy (TEM) and N2 physisorption measurements such as Brunauer-Emmett-Teller (BET) and Barrett-Joyner-Halenda (BJH) methods. 展开更多

Mast cells are the main effector cells in IgE-associated allergic disorders,and we have reported that mucosal mast cells(MMCs)play a more important role in the development of food allergy(FA).IgE with antigen or calcium ionophore stimulation can lead to the activation of MMCs via a calcium-dependent pathway.The purpose of the present study was to identify gene signatures with IgE/antigen(dinitrophenyl-bovine serum albumin,DNP-BSA)or calcium ionophore(A23187)on the activation of MMCs.Differentially expressed genes between the two types of samples were identified with microarray analysis.Gene ontology functional and pathway enrichment analyses of differentially expressed genes were performed using the database for annotation,visualization,and integrated discovery software.The results showed that IgE/antigen and A23187 could induce degranulation,increase vacuoles,and elevate the cytosolic calcium concentration in MMCs.Furthermore,GeneChip analysis showed that the same 134 mRNAs were altered with IgE/DNP-BSA and A23187,suggesting that DNP-BSA/IgE and A23187 affect the same signal pathway partly in degranulation.KEGG analysis showed that the data were enriched in NF-κB,TNF,MAPK,transcription factor activity,DNA binding,and nucleic acid binding,suggesting that activation of MMCs is a complex process.The results provide new insights on MMCs activation. 展开更多

Verlinde has suggested that the gravity has an entropic origin, and a gravitational system could be regarded as a thermodynarnical system. It is well-known that the equipartition law of energy is invalid at very low temperature. Therefore, entropic force should be modified while the temperature of the holographic screen is very low. It is shown that the modified entropic force is proportional to the square of the acceleration, while the temperature of the holographic screen is much lower than the Debye temperature TD. The modified entropic force returns to the Newton's law of gravitation while the temperature of the holographic screen is much higher than the Debye temperature. The modified entropic force is connected with modified Newtonian dynamics (MOND). The constant ao involved in MOND is linear in the Debye frequency WD, which can be regarded as the largest frequency of the bits in screen. We find that there do have a strong connection between MOND and cosmology in the framework of Verlinde's entropic force, if the holographic screen is taken to be bound of the Universe. The Debye frequency is linear in the Hubble constant Ho. 展开更多
