Exercise preconditioning alleviates ischemia-induced memory deficits by increasing circulating adiponectin

Cerebral ischemia is a major health risk that requires preventive approaches in addition to drug therapy.Physical exercise enhances neurogenesis and synaptogenesis,and has been widely used for functional rehabilitation after stroke.In this study,we determined whether exercise training before disease onset can alleviate the severity of cerebral ischemia.We also examined the role of exercise-induced circulating factors in these effects.Adult mice were subjected to 14 days of treadmill exercise training before surgery for middle cerebral artery occlusion.We found that this exercise pre-conditioning strategy effectively attenuated brain infarct area,inhibited gliogenesis,protected synaptic proteins,and improved novel object and spatial memory function.Further analysis showed that circulating adiponectin plays a critical role in these preventive effects of exercise.Agonist activation of adiponectin receptors by Adipo Ron mimicked the effects of exercise,while inhibiting receptor activation abolished the exercise effects.In summary,our results suggest a crucial role of circulating adiponectin in the effects of exercise pre-conditioning in protecting against cerebral ischemia and supporting the health benefits of exercise. 展开更多

Aerobic exercise mitigates high-fat diet-induced cognitive decline through suppressing liver-brain-axis-mediated ATF3 and neuroinflammation

It is well known that appropriate aerobic exercise can effectively alleviate fatty liver and enhance brain function.The concept of multi-organ crosstalk coordinating disease progression has become the current research hot topic.However,there remains an urgent need to elucidate its specific mechanisms.This study aimed to explore the impact of a high-fat diet(HFD)on liver health and cognitive function,and to further uncover the regulatory effect of aerobic exercise by liver-specific activating transcription factor 3(Atf3)knockout(ATF3cKO)mice in a“liver-brain”axis mode.The 5-week-old C57BL/6 and ATF3cKO mice were fed with HFD for 32 weeks,and sequentially subjected to aerobic exercise intervention at the 20th week for another 12 consecutive weeks.Meanwhile,C57BL/6 mice were provided with a normal diet as the control group.The functional parameters of liver and brain of all mice were assessed.Cognitive capacity of all mice was assessed by the Morris water maze(MWM).Inflammatory factors in the serum and brain of mice were quantified using enzyme-linked immunosorbent assay(ELISA),and the expression of inflammasomes was detected by immunohistochemistry(IHC).Additionally,the activation of nuclear factor-κB(NF-κB)and phosphoinositide 3-kinase(PI3K)signal pathways was analyzed by Western blotting.In this study,HFD impaired hepatic and brain functions,while aerobic exercise and liver-specific Atf3 knockout suppressed inflammatory factors in the peripheral circulation through hepatoprotective mechanisms,thereby attenuating cerebral inflammation and preserving neurological integrity,as well as mitigating HFD-induced cognitive decline. 展开更多

Nutrigenomics is an emerging research field to investigate the interactions between food components and genes and elucidate their impacts on health and diseases.Exercise,another critical factor influencing human health,is intricately linked with nutrition and genetics,and is influenced by a combination of genetic background,lifestyle choice,and environmental factors.The high-throughput sequencing technologies and advanced bioinformatic tools have offered researchers to delve deeper into how individual genetic expression responses to exercise and how dietary interventions can enhance exercise performance and recovery through the gene expression changes and modifications.Among these mechanisms,gene expression emerges as the primary focus,highlighting its role in linking nutrients and exercise to metabolic pathways,muscle synthesis,and inflammation.While epigenetic modifications are briefly considered,the emphasis remains on how direct and indirect influences on gene expression drive physiological adaptations.Herein,the article reviews recent advancements in nutrigenomics within the context of exercise science,exploring gene-regulating mechanisms affected by specific nutrients and dietary patterns on exercise capacity,muscle synthesis,and damage repair.It also outlines the potential applications of personalized nutrition plans in athletic training and general fitness.In addition,this article addresses current challenges and potential issues in the practical implementation of nutrigenomics in exercise-induced health promotion.Future studies should emphasize interdisciplinary collaboration to unravel the intricate gene-nutrition-exercise interaction networks,thereby providing a solid theoretical foundation and technological support for the advancement of precision exercise nutrition. 展开更多

Objective This study is aimed to report the development,the reliability and validity of the Chinese Children Physical Activity Questionnaire(CCPAQ)which was designed for the assessment of physical activity pattern in young population in China.Methods The CCPAQ was administered for two times in 119 children(mean age 13.1±2.4 years;boys 47%)to examine reliability by using intraclass correlation coefficients.Validity was determined in 106 participants by agreement with the CCPAQ measures and the objective method,the ActiGraph accelerometer.Data on physical activity patter n including time spe nt on d iff ere nt intensi ties and total physical activity,sedentary behavior as well as physical activity energy expenditure were used to assess the validity with Spearman's correlation coefficient and the Bland-Altman plots.Results The reliability coefficient of the CCPAQ ranged from 0.63-0.93(Intra-class correlation coefficient).Spearman's correlation coefficient for validity of time spent on total physical activity and sedentary behavior were all 0.32(P<0.001),and for physical activity energy expenditure was 0.58(P<0.001).Time spent on moderate-to-vigorous physical activity and light physical activity showed a relatively low correlation with the accelerometer(rho=0.20,P=0.040;rho=0.19,P=0.054).Conclusion The CCPAQ appears to be a promising and feasible method to assess physical activity pattern in Chinese children. 展开更多

Functional foods,namely as natural or processed foods containing bioactive compounds,can provide health-promoting effects beyond basic nutrition,or offer the prevention or supplementary treatment of chronic diseases.The bioactive components in functional foods usually have pleiotropic effects,including antioxidant,anti-inflammatory,hypolipidemic,glycemic-regulating,cytoprotective,and neuroprotective functions.Autophagy is one of the highly conserved cellular processes for the clearance of aberrant components in eukaryotic cells,and plays an essential role in health promotion and prevention and treatment of a series of chronic diseases.Once the cells are in the stress environment,the induced autophagy will accelerate the clearance of cellular damaged or toxic protein aggregates or dysfunctional cellular organelles to maintain homeostasis in cells.In this article,we summarize several widely investigated bioactive components used as functional foods,such as resveratrol,epigallocatechin-3-gallate,curcumin and trehalose,with the regulatory function for autophagy during the intervention of chronic diseases,which will provide the references or novel thoughts for the development of functional foods with the modulation of autophagy. 展开更多

Obesity,caused by excessive energy,leads to body weight gain and various diseases,including cognitive impairment.Current studies suggest that diet restriction such as optimal fasting and regular exercise are crucial for improving cognitive capacity.However,further exploration is needed to understand the specific mechanisms of high fat diet(HFD)-induced cognitive decline in obesity.In the present study,4-month-old mice were subjected to HFD feeding for 18 weeks,followed by aerobic exercise and high-intensity intermittent exercise,regular diet feeding,and intermittent fasting for 8 weeks,and then used to evaluate cognitive capacity,inflammation,compromised insulin signaling pathway,and apoptosis in hippocampal tissue,as well as AMPK/SIRT1 and TLR4 signal pathways.Obese mice revealed impaired cognitive capacity as compared with mice fed with regular diets.In contrast,aerobic exercise,high-intensity intermittent exercise,regular diet,and intermittent fasting could inhibit apoptosis caused by inflammation-mediated compromised insulin signaling pathway in hippocampal tissues through activating the AMPK/SIRT1 signal pathway and suppressing the TLR4 signal pathway,thereby rescuing the cognitive impairment of obese mice.Therefore,diet restriction and exercise interventions may play a positive role in reverting obesity-induced cognitive impairment. 展开更多

Ferroptosis is a programmed cell death,and its mechanism involves multiple metabolic pathways,such as iron and lipid metabolism,and redox homeostasis.Exerkines are important mediators that optimize cellular homeostasis and maintain physiological health during exercise stim-ulation.This article comprehensively examines the mechanisms and regulatory networks for governing ferroptosis and summarizes the impact of exercise and exerkines on ferroptosis under varying load intensities and disease contexts.Notably,despite its significant efficacy and minimal side effects, the therapeutic and prognostic potential of exercise in ferroptosis-related diseases remains largely unexplored.This article,by summarizing recent progresses in the regulation of exerkines-mediated ferroptosis,could further uncover the preventive or alleviative mecha-nisms of some diseases upon exercise interventions,which will be beneficial to design exercise interventional strategies for alleviating disease progression through the regulation of ferroptosis. 展开更多

Objective This study was aimed at examining the trends and correlates of physical activity(PA)and sedentary behaviors among Chinese children.Methods A total of 4,341 subjects(6,936 observations)aged 6–17 years who participated in the China Health and Nutrition Survey(2004–2015)were included.Of the subjects,41%participated in the survey twice or more.Random-effects ordinal regression models and repeated-measures mixed-effects models were used to examine the PA trends.Quantile regression models were applied to examine the factors influencing PA and sedentary behaviors.Results From 2004 to 2015,the prevalence of physical inactivity among Chinese children aged 6–17 years increased by 5.5%[odds ratio(OR),1.51;95%confidence interval(CI),1.19–1.90;P<0.001].The PA volume declined by 5.8 metabolic equivalent of task-hr/week(P<0.001),and the time spent in sedentary behaviors increased by 1.8 hr/week(P<0.001).Age,ethnicity,and region showed significant effects on the PA volume across the quartiles(P<0.001).Across the quartiles,sedentary time was significantly higher in the children residing in urban areas(P<0.001)or areas with high urbanization levels(P≤0.005)than in their counterparts.Conclusions A declining PA trend among Chinese children aged 6–17 years was observed from 2004 to2015,and certain subgroups and geographical areas are at higher risk of physical inactivity. 展开更多

Objective In this study we aimed to examine the reliability and validity of the wristband activity monitor against the accelerometer for children.Methods A total of 99 children(mean age = 13.0 ± 2.5 y) wore the two monitors in a free-living context for 7 days. Reliability was measured by intraclass correlation to evaluate consistency over time.Repeated-measures analyses of variance was used to detect differences across days. Spearman’s correlation coefficient(rho), median of absolute percentage error, and Bland-Altman analyses were performed to assess the validity of the wristband against the Acti Graph accelerometer. The optimal number of repeated measures for the wristband was calculated by using the Spearman-Brown prophecy formula.Results The wristband had high reliability for all variables, although physical activity data were different across 7 days. A strong correlation for steps(rho: 0.72, P < 0.001), and moderate correlations for time spent on total physical activity(rho: 0.63, P < 0.001) and physical activity energy expenditure(rho: 0.57, P < 0.001) were observed between the wristband and the accelerometer. For different intensities of physical activity, weak to moderate correlations were found(rho: 0.38 to 0.55, P < 0.001).Conclusion The wristband activity monitor seems to be reliable and valid for measurement of overall children’s physical activity, providing a feasible objective method of physical activity surveillance in children. 展开更多

Parkinson’s disease (PD) is a slowly progressive neurodegenerative disorder with motor and non-motorsigns, which begins long before the diagnosis can be made. Pharmacotherapy with dopaminergic drugsis the mainstay of PD therapy. There is no currently available cure or disease-modifying treatment. Dietand various foods have been proposed as possible risk factors for PD. Nutritional supplementation withomega-3 fatty acids, vitamin D, B vitamins, and coenzyme Q may have potential in the management ofPD. In addition, several strategies could possibly lead to the development of medical foods in PD. Forexample, ketogenic diets may have effects complementary to pharmacotherapy. Gut microbial dysbiosisand altered microbial metabolites found in PD may lead to chronic low-grade inflammation in gut andbrain. Interventions targeting gut microbiota, such as the supplementation of probiotics, may providenovel approaches to PD in regard to both symptomatic treatment and disease modification. High qualityrandomized controlled trials need to be conducted in order to evaluate whether these food-basedstrategies have symptomatic efficacy or the potential to delay the progression of PD. 展开更多

The prevalence of childhood obesity has been increasing worldwide and lifestyle changes are the most important strategies in managing this prevalence. This study aimed to describe the intervention effects of nutrition and physical activities offered as an after school short-term on outcomes of healthy nutrition practices, fitness and lowering fatness. This quasi-experimental study was conducted with a convenience sample of 59 caucasian children, aged 7.7 ± 1.4 years old (52.5% girls) registered on a private school of a middle-size town located in Sao Paulo State, Brazil. The school-based 12-week intervention consisted of 2 weekly 60 minutes section, during 3 months, including a nutritional education and a physical activity curriculum. Anthropometric data was measured for all children at baseline and after 12 weeks. All parents were contacted and requested to complete questionnaire about child’s eating behavior at baseline and after the 12-week program. After the intervention, children showed an increasing in the intake of fruits (64.3%), vegetables (61.9%), and water (52.0%). Overall, 83.3% of the children changed eating behavior according to the questionnaire responded by the parents. Waist circumference was significantly lower and abdominal strength improved after participating in the intervention program. Mean height and weight were significantly higher in boys and girls after a 12-week intervention. Although mean values of BMI remained the same after the intervention it was observed a decreasing in the prevalence of obesity among the children. Thus our study showed that a school-based intervention program focused on nutritional education and physical activity program promoted waist circumference reduction and decreased obesity without affecting the height growth along with improved fitness and healthy eating behavior. This intervention program would be feasible and replicable in others schools around the country. 展开更多

Subject-specific spinal musculoskeletal modeling can help understand the spinal loading mechanism during human locomotion.However,existing literature lacks methods to identify the maximum isometric strength of individual spinal muscles.In this study,a muscle strength identification method combining isokinetic testing and musculoskeletal simulations was proposed,and the influence of muscle synergy and intra-abdominal pressure(IAP)on identified spinal muscle strength was further discussed.A multibody dynamic model of the spinal musculoskeletal system was established and controlled by a feedback controller.Muscle strength parameters were adjusted based on the measured isokinetic moments,and muscle synergy vectors and the IAP piston model were further introduced.The results of five healthy subjects showed that the proposed method successfully identified the subject-specific spinal flexor/extensor strength.Considering the synergistic activations of antagonist muscles improved the correlation between the simulated and measured spinal moments,and the introduction of IAP slightly increased the identified spinal extensor strength.The established method is beneficial for understanding spinal loading distributions for athletes and patients with sarcopenia. 展开更多
