Axonal growth inhibitors and their receptors in spinal cord injury:from biology to clinical translation 被引量:4

Axonal growth inhibitors are released during traumatic injuries to the adult mammalian central nervous system, including after spinal cord injury. These molecules accumulate at the injury site and form a highly inhibitory environment for axonal regeneration. Among these inhibitory molecules, myelinassociated inhibitors, including neurite outgrowth inhibitor A, oligodendrocyte myelin glycoprotein, myelin-associated glycoprotein, chondroitin sulfate proteoglycans and repulsive guidance molecule A are of particular importance. Due to their inhibitory nature, they represent exciting molecular targets to study axonal inhibition and regeneration after central injuries. These molecules are mainly produced by neurons, oligodendrocytes, and astrocytes within the scar and in its immediate vicinity. They exert their effects by binding to specific receptors, localized in the membranes of neurons. Receptors for these inhibitory cues include Nogo receptor 1, leucine-rich repeat, and Ig domain containing 1 and p75 neurotrophin receptor/tumor necrosis factor receptor superfamily member 19(that form a receptor complex that binds all myelin-associated inhibitors), and also paired immunoglobulin-like receptor B. Chondroitin sulfate proteoglycans and repulsive guidance molecule A bind to Nogo receptor 1, Nogo receptor 3, receptor protein tyrosine phosphatase σ and leucocyte common antigen related phosphatase, and neogenin, respectively. Once activated, these receptors initiate downstream signaling pathways, the most common amongst them being the Rho A/ROCK signaling pathway. These signaling cascades result in actin depolymerization, neurite outgrowth inhibition, and failure to regenerate after spinal cord injury. Currently, there are no approved pharmacological treatments to overcome spinal cord injuries other than physical rehabilitation and management of the array of symptoms brought on by spinal cord injuries. However, several novel therapies aiming to modulate these inhibitory proteins and/or their receptors are under investigation in ongoing clinical trials. Investigation has also been demonstrating that combinatorial therapies of growth inhibitors with other therapies, such as growth factors or stem-cell therapies, produce stronger results and their potential application in the clinics opens new venues in spinal cord injury treatment. 展开更多

Acute respiratory infections(ARIs)are the main cause of morbidity and mortality worldwide,especially among children.The human bocavirus(HBoV)is a nonenveloped DNA virus that was recently identified as a respiratory pathogen associated with respiratory tract infections(RTIs),predominantly in infants and young children.It is also detected from the gastrointestinal tract in children.The prevalence of HBoV1 acute respiratory tract infection varies across age groups,ranging from 10.3%to 12.51%in individuals under 3 years of age.The spectrum of clinical presentation includes mild upper RTIs,acute exacerbation of asthma,bronchitis,bronchiolitis,pneumonia,and multi-organ failure.Although HBoV is often detected in patients with ARIs who have other respiratory viruses(17%-85%),recent studies have identified it as the sole aetiology for mild to severe ARIs.Children with pre-existing medical conditions infected with HBoV often have a risk of severe illness.HBoV infection is diagnosed primarily by detecting viral DNA in respiratory samples using molecular methods.Currently,there is no specific antiviral treatment for HBoV infections and the cases are managed symptomatically.General preventive measures used for the prevention of viral RTIs are applicable,as there is no effective vaccine against this virus.The HBoV has been implicated in RTIs,particularly in children,and has also been detected in cases of gastroenteritis.Despite its global prevalence,the exact pathogenic role of HBoV remains unclear due to frequent co-infections with other viruses.This minireview discusses the virology,epidemiology,clinical manifestations,diagnosis,and potential treatment approaches related to HBoV infections. 展开更多

Establishing the etiological diagnosis can be challenging and often requires various investigations. This cross-sectional study, conducted in Cotonou among 35 azoospermic men, aimed to assess the role of biochemical analysis of seminal fluid in determining the etiological diagnosis. The results revealed significant heterogeneity in biochemical profiles. Most of the patients had normal levels of alpha-glucosidase, fructose, and zinc, indicating normal accessory gland function and suggesting non-obstructive azoospermia. However, a significant subgroup exhibited abnormalities in these markers, suggesting a possible obstructive azoospermia. The heterogeneity in biochemical profiles observed in this study highlights the need for further studies to better characterize azoospermia in Benin. 展开更多

Objective:To assess the burden of Group B Streptococcus(GBS)and analyze the distribution of scrotypes in relation to their source.The review highlights data gaps in transmission dynamics and regional food consumption practices,which are esscntial for designing cffective public health strategies and advancing vaccine development.Methods:Scarches were conducted in Web of Science,MEDLINE,Science Direct,PubMcd,and Scopus databases to find studies related to GBS during 1990-2025.Eligible studies were those that described prevalence,scrotype distribution or scquence type(ST)of GBS in Southeast Asian countries.Random-cffects meta-analysis was used to pool data.Results:A total of 26 studies met the inclusion criteria from cight countries.The pooled estimate of maternal GBS colonization was 15.1%,with scrotypesII,v,ⅡI,V,and I a accounting for the majority of cases(91.24%)in the Southcast Asia studies.Data on ST was limited;however,ST1 was found to be predominant in Malaysia and Thailand,while ST283 was notably linked to the consumption of raw fish.Conclusions:The pooled estimate of the maternal colonization with GBS was 15.1%which is cquivalent to many other primary and review reports worldwide.Distribution of scrotype and ST is needed to be studied in Southcast Asian countries to devise cffective preventive measures.These findings underscore thc importance of surveillance and tailored prevention strategies to combat GBS infections in Southcast Asia. 展开更多

Despite the proven beneficial role of type 2 cytokines in diabetes and obesity,IL-9,a predominant Th2 cytokine,has not been investigated in this context.The present study characterized the role of IL-9 signaling in obesity and metabolic dysfunction.We found decreased IL-9 levels in human type 2 diabetes patients and decreased IL-9 signaling in high-fat diet(HFD)-induced obese mice.On the other hand,recombinant IL-9(rIL-9)treatment reversed insulin insensitivity and inflammation following HFD consumption.IL-9R knockout(KO)mice fed a HFD presented faster weight gain,impaired glucose and insulin tolerance,defective insulin signaling,increased adipocyte size,and decreased energy expenditure.In the adipose tissue of HFD-fed IL-9R KO mice,a significant increase in the number of CD11c+macrophages and a decrease in the number of RELMα+macrophages,eosinophils and ILC2s were observed,along with increased TNF,decreased adiponectin production and increased expression of NLRP3.In vitro treatment of human and mouse macrophages with rIL-9 decreased the release of NLRP3-induced IL-1βand IL-18.In vivo treatment of HFD-fed IL-9R KO mice with a pharmacological inhibitor of the NLRP3 inflammasome rescued body weight,insulin sensitivity and adipose tissue inflammation.Mechanistically,the STAT5 protein was found to be important for the IL-9-induced inhibition of the NLRP3 inflammasome in adipose tissue.In addition,we also demonstrated a potential role for IL-9 in the protective effects of helminth immunomodulation during obesity and insulin resistance in filaria-infected humans and in an animal model.Taken together,the results of this study highlight that IL-9 signaling improves insulin signaling by inhibiting NLRP3-induced inflammation. 展开更多

The main adaptations to endurance exercise include the correction of electrolyte imbalance, a decrease in glycogen storage and the increase of oxidative stress, intestinal permeability, muscle damage, and systemic inflammatory response. Adaptations to exercise might be influenced by the gut microbiota, which plays an important role in the production, storage, and expenditure of energy obtained from the diet as well as in inflammation,redox reactions, and hydration status.Methods: A systematic and comprehensive search of electronic databases, including MEDLINE, Scopus, Clinical Trials.gov, Science Direct,Springer Link, and EMBASE was done. The search process was completed using the keywords: "endurance", "exercise", "immune response","microbiota", "nutrition", and "probiotics".Results: Reviewed literature supports the hypothesis that intestinal microbiota might be able to provide a measureable, effective marker of an athlete's immune function and that microbial composition analysis might also be sensitive enough to detect exercise-induced stress and metabolic disorders. The review also supports the hypothesis that modifying the microbiota through the use of probiotics could be an important therapeutic tool to improve athletes' overall general health, performance, and energy availability while controlling inflammation and redox levels.Conclusion: The present review provides a comprehensive overview of how gut microbiota may have a key role in controlling the oxidative stress and inflammatory responses as well as improving metabolism and energy expenditure during intense exercise. 展开更多

Application of the Sk-MSCs in the bridging conduit for repairing long nerve gap injury resulted favorable axonal regeneration, which showing superior effects than gold standard therapy--healthy nerve autograft. AIM To evaluate the efficacy of peripheral blood concentrations of angiopoietins(Ang) as cirrhosis biomarkers of chronic hepatitis C(CHC).METHODS Ang1 and Ang2 serum levels were measured by enzyme-linked immunosorbent assays(ELISA) in samples from 179 cirrhotic and non-cirrhotic CHC patients, classified according to the METAVIR system.Groups were compared by non-parametric MannWhitney U test. Subsequently, the association of peripheral concentrations of angiopoietins with the stage of fibrosis was analyzed using Spearman correlation test. Finally, the accuracy, sensitivity and specificity of circulating angiopoietins for cirrhosis diagnosis were determined by the study of the respective area under the curve of receiver operator characteristics(AUC-ROC).RESULTS Peripheral blood concentrations of Ang1 and Ang2 in CHC patients were significantly related to fibrosis. While Ang1 was decreased in cirrhotic subjects compared to non-cirrhotic(P < 0.0001), Ang2 was significantly increased as CHC progressed to the end stage of liver disease(P < 0.0001). Consequently, Ang2/Ang1 ratio was notably amplified and significantly correlated with fibrosis(P < 0.0001). Interestingly, the individual performance of each angiopoietin for the diagnosis of cirrhosis reached notable AUC-ROC values(above 0.7, both), but the Ang2/Ang1 ratio was much better(AUC-ROC = 0.810) and displayed outstanding values of sensitivity(71%), specificity(84%) and accuracy(82.1%) at the optimal cut-off(10.33). Furthermore, Ang2/Ang1 ratio improved the performance of many other previously described biomarkers or scores of liver cirrhosis in CHC.CONCLUSION Ang2/Ang1 ratio might constitute a useful tool for monitoring the progression of chronic liver disease towards cirrhosis and play an important role as therapeutic target. 展开更多

In infertile males, abnormal karyotypes are more frequent than in the general population. Furthermore, meiotic disorders affecting the germ cell-line have been observed in men with normal somatic karyotypes consulting for infertility. In both cases, the production of unbalanced spermatozoa has been demonstrated. Basically addressed to establish reproductive risks, fluorescence in situ hybridization (FISH) on decondensed sperm heads has become the most frequently used method to evaluate the chromosomal constitution of spermatozoa in carriers of numerical sex chromosome abnormalities, carriers of structural chromosome reorganizations and infertile males with normal karyotype. The aim of this review is to present updated figures of the information obtained through sperm FISH studies with an emphasis on its clinical significance. Varicocele is present in approximately 15% of men, and, although it is the most commonly diagnosed cause of male infertility, nearly two-thirds of men with varicoceles remain fertile. It was decided to make use of the current evidence obtained from the previous meta-analyses between 2004 and 2015 as well as available articles covering this field, preferably randomized controlled articles dealing with the topic of semen analysis before and after repair. Two important meta-analyses were discussed as well as other articles dealing with the topic of semen analysis before and after varicocelectomy. The evidence suggests that all semen parameters improve after varicocele repair. Based on the available evidence, it is clear that there is a benefit in treating men with a palpable varicocele. One can expect that all semen parameters will improve within 3 months after repair. 展开更多

A physiological sequence called autophagy qualitatively determines cellular viability by removing protein aggregates and damaged cyto-plasmic constituents, and contributes significantly to the degree of myocardial ischemia-reperfusion (I/R) injury. This tightly orchestrated cata-bolic cellular‘housekeeping’ process provides cells with a new source of energy to adapt to stressful conditions. This process was first described as a pro-survival mechanism, but increasing evidence suggests that it can also lead to the demise of the cell. Autophagy has been implicated in the pathogenesis of multiple cardiac conditions including myocardial I/R injury. However, a debate persists as to whether autophagy acts as a protec-tive mechanism or contributes to the injurious effects of I/R injury in the heart. This controversy may stem from several factors including the va-riability in the experimental models and species, and the methodology used to assess autophagy. This review provides updated knowledge on the modulation and role of autophagy in isolated cardiac cells subjected to I/R, and the growing interest towards manipulating autophagy to increase the survival of cardiac myocytes under conditions of stress-most notably being I/R injury. Perturbation of this evolutionarily conserved intracellular cleansing autophagy mechanism, by targeted modulation through, among others, mammalian target of rapamycin (mTOR) inhibitors, adenosine monophosphate-activated protein kinase (AMPK) modulators, calcium lowering agents, resveratrol, longevinex, sirtuin activators, the proapoptotic gene Bnip3, IP3 and lysosome inhibitors, may confer resistance to heart cells against I/R induced cell death. Thus, therapeutic ma-nipulation of autophagy in the challenged myocardium may benefit post-infarction cardiac healing and remodeling. 展开更多

Aim: To evaluate the protective effects of Hibiscus sabdariffa (Roselle) and Zingiber officinale (Ginger) against cisplatin-induced reproductive toxicity in rats and to study the mechanisms underlying these effects. Methods: Ethanol extracts of H. sabdariffa or Z. officinale [1 g/(kg·day)] were given p.o. to male albino rats for 26 days, which began 21 days before a single cisplatin i.p. injection (10 mg/kg body weight). Results: Extracts of H. sabdariffa and Z. offcinale reduced the extent of cisplatin-induced sperm abnormality and enhanced sperm motility. Both extracts restored the control level of malondialdehyde (MDA) (lipid peroxidation marker) in the cisplatin-treated testis. The cisplatin injection induced decline in the levels of superoxide dismutase (SOD), reduced glutathione (GSH) and catalase (CAT) were significantly reversed to control levels in groups where cisplatin was preceded by the administration of either H. sabdariffa or Z. officinale. Conclusion: Both H. sabdariffa and Z. officinale treatment increased the activities of testicular antioxidant enzymes and restored sperm motility of cisplatin-treated rats. The protective effects of tested plants are, therefore, suggested to be mediated by their potent antioxidant activities. 展开更多

Stat. Mech. 2010 P03018]. Through these parameters, we adjust the anticipated velocity and the acceleration threshold separately. It turns out that the flow rate of synchronized flow mainly changes with the anticipated velocity, and the F →S phase transition feature mainly changes with the acceleration threshold. Therefore, we conclude that the acceleration threshold is the major factor affecting the F → S phase transition. 展开更多

Aim: To assess heine oxygenase-1 (HO-1) activity in the cavemous tissue of sildenafil citrate-treated rats. Methods: One hundred and ninety-two Sprague-Dawley male rats, divided into four equal groups, were investigated. Group 1, the control group, received regular animal chow; group 2 received sildenafil citrate by intragastric tube; group 3 received sildenafil and HO inhibitor (zinc protoporphyrin, ZnPP); and group 4 received sildenafil and nitric oxide synthase (NOS) inhibitor L-nitroarginine methyl ester (L-NAME). Twelve rats from each group were killed after 0.5 h, 1 h, 2 h and 3 h of drug administration. Then HO-1 activity, cGMP levels and NOS enzymatic activity in the cavernous tissues were estimated. Results: In cavemous tissue, HO-1 activity, NOS enzymatic activity and cGMP concentration increased significantly in sildenafil-treated rats compared to other groups throughout the experiment. Rats receiving either HO or NOS inhibitors showed a significant decrease in these parameters. HO- 1 cavemous tissue activity and NOS enzymatic activity demonstrated a positive significant correlation with cGMP levels (r = 0.646, r = 0.612 respectively; P 〈 0.001). Conclusion: The actions of PDE5 inhibitor sildenafil citrate in the cavernous tissue are partly mediated through the interdependent relationship between both HO-1 and NOS activities. 展开更多

Pediatric soft tissue sarcomas are a group of malignant neoplasms arising within embryonic mesenchymal tissues during the process of differentiation into muscle, fascia and fat. The tumors have a biphasic peak for age of incidence. Rhabdomyosarcoma(RMS) is diagnosed more frequently in younger children, whereas adult-type non-RMS soft tissue sarcoma is predominately observed in adolescents. The latter group comprises a variety of rare tumors for which diagnosis can be difficult and typically requires special studies, including immunohistochemistry and molecular genetic analysis. Current management for the majority of pediatric sarcomas is based on the data from large multi-institutional trials, which has led to great improvements in outcomes over recent decades. Although surgery remains the mainstay of treatment, the curative aim cannot be achieved without adjuvant treatment. Pre-treatment staging and risk classification are of prime importance in selecting an effective treatment protocol. Tumor resectability, the response to induction chemotherapy, and radiation generally determine the risk-group, and these factors are functions of tumor site, size and biology. Surgery provides the best choice of local control of small resectable tumors in a favorable site. Radiation therapy is added when surgery leaves residual disease or there is evidence of regional spread. Chemotherapy aims to reduce the risk of relapse and improve overall survival. In addition, upfront chemotherapy reduces the aggressiveness of the required surgery and helps preserve organ function in a number of cases. Long-term survival in low-risk sarcomas is feasible, and the intensity of treatment can be reduced. In high-risk sarcoma, current research is allowing more effective disease control. 展开更多

Lanthanide ions doped luminescent materials are widely studied for latent fingerprint detection.However,most of these materials are synthesized at very high temperatures and use UV C light for visualization,which is harmful to eye,skin,etc.Herein,the Gd_(0.95)Eu_(0.05)PO_(4) nanorods synthesized by a simple co-precipitation method at 185℃ were reported for latent fingerprint visualization under 395 nm light.The Gd_(0.95)Eu_(0.05)PO_(4) nanomaterial has monoclinic crystal structure and shows rod-shaped morphology.Further,these Gd_(0.95)Eu_(0.05)PO_(4) nanorods exhibit excellent photoluminescence properties and strong fuchsia emission under UV light.These nanorods have been employed for developing latent fingerprints on various porous and non-porous substrates by the powder dusting technique,which exhibits clear and well defined details with high contrast,selectivity and sensitivity under 395 nm UV light.Latent fingerprints developed after 72 h of their deposition also show clear contrast with these nanorods.Therefore,the Gd_(0.95)Eu_(0.05)PO_(4) nanorods can be used for latent fingerprint visualization applications. 展开更多

Background Coronary artery disease(CAD)remains a leading cause of morbidity and mortality.Cytokines play a potential role in atherosclerosis pathogenesis and progression.We investigated the association between high sensitive C-reactive protein(hs CRP)and severity of CAD.Methods CAD patients were stratified according to hs CRP cut-off value into high levels hs CRP group(≥8.4 mg/L)and low levels hs CRP group(<8.4 mg/L).Severity of CAD was assessed according to artery stenosis degree and the number of vessel involved.Statistical analysis was performed using Statistical Package for the Social Sciences(SPSS,version 23.0).Results The mean age was 60.3±11.0 years.The level of hs CRP was increased and ranged from 0.2 to 1020.0 mg/L.Biochemical risk factors and severity of CAD didn’t show significant differences between the two groups.In multivariate linear analysis,cardiac troponin I(c Tn I)and serum amyloid A(SAA)were predictors of hs CRP.As shown in receiver operating characteristic(ROC)curve analysis performed in patients with ST-segment elevation myocardial infarction(STEMI)and compared to myonecrosis biomarkers,hs CRP(area under the curve(AUC):0.905;95%CI:0.844-0.966;P<0.001)could be a powerful predictor marker in evaluating the infarct size after myocardial infarction but not better than c Tn I.Conclusions Hs CRP levels were not associated with the severity of CAD but could be useful in the evaluation of myocardial necrosis in patients with STEMI. 展开更多

Pancreatic cancer(Pa C)shows a clear tendency to increase in the next years and therefore represents an important health and social challenge.Currently,there is an important need to find biomarkers for PaC early detection because the existing ones are not useful for that purpose.Recent studies have indicated that there is a large window of time for PaC early detection,which opens the possibility to find early biomarkers that could greatly improve the dismal prognosis of this tumor.The present manuscript reviews the state of the art of the existing PaC biomarkers.It focuses on the anomalous glycosylation process and its role in PaC.Glycan structures of glycoconjugates such as glycoproteins are modified in tumors and these modifications can be detected in biological fluids of the cancer patients.Several studies have found serum glycoproteins with altered glycan chains in PaC patients,but they have not shown enough specificity for PaC.To find more specific cancer glycoproteins we propose to analyze the glycan moieties of a battery of glycoproteins that have been reported to increase in PaC tissues and that can also be found in serum.The combination of these new candidate glycoproteins with their aberrant glycosylation together with the existing biomarkers could result in a panel,which would expect to give better results as a new tool for early diagnosis of PaC and to monitor the disease. 展开更多
