Thymoquinone(TQ)and gallic acid(GA)are known for counter-tumorigenic characteristics.GA inhibits cancer cell proliferation by interfering with many apoptotic signaling pathways,producing more reactive oxygen species(ROS),focusing on the cell cycle,and suppressing the expression of oncogenes and matrix metalloproteinases(MMPs).In this study,TQ(after reducing to thymohydroquinone)and GA are esterified to form thymohydroquinyl gallate(a prodrug).Thymohydroquinyl gallate(THQG)possesses enhanced antineoplastic efficacy and targeted delivery potential.The chemical and spectroscopic analysis confirms ester synthesis.Gold nanoparticles(AuNPs)are employed as nanocarriers due to their physicochemical and optical characteristics,biocompatibility,and low toxicity.As an efficient drug transporter,(AuNPs) shield conjugated drugs from enzymatic digestion.The prodrug acts as a reducing agent for Au metal atoms and is loaded onto it after reduction.The nano drug is radiolabeled with 99mTc and 131I to monitor the drug biodistribution in animals using a gamma camera and single-photon emission computerized tomography(SPECT).131I is an antineoplastic that helps enhance the drug's efficiency.Chromatographic results reveal promising radiolabeling percentages.In vitro,drug release shows sustained release at pH~5.8.In vitro 3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide(MTT)cytotoxicity assay reveals drug potency on CAL 27 and MCF 7 cell lines. 展开更多

Overall 5-years survival of pancreatic cancer patients is nearly 5%,making this cancer type one of the most lethal neoplasia.Furthermore,the incidence rate of pancreatic cancer has a growing trend that determines a constant increase in the number of deceases caused by this pathology.The poor prognosis of pancreatic cancer is mainly caused by delayed diagnosis,early metastasis of tumor,and resistance to almost all tested cytotoxic drugs.In this respect,the identification of novel potential targets for new and efficient therapies should be strongly encouraged in order to improve the clinical management of pancreatic cancer.Some studies have shown that the mitochondrial uncoupling protein 2(UCP2) is over-expressed in pancreatic cancer as compared to adjacent normal tissues.In addition,recent discoveries established a key role of UCP2 in protecting cancer cells from an excessive production of mitochondrial superoxide ions and in the promotion of cancer cell metabolic reprogramming,including aerobic glycolysis stimulation,promotion of cancer progression.These observations together with the demonstration that UCP2 repression can synergize with standard chemotherapy to inhibit pancreatic cancer cell growth provide the molecular rationale to consider UCP2 as a potential therapeutic target for pancreatic cancer.In this editorial,recent advances describing the relationship between cancer development and mitochondrial UCP2 activity are critically provided. 展开更多

Genital dissatisfaction is well known in female and adults.Less is known about male adolescents and their genital satisfaction.The aim of this study was to investigate and report the role of surgery in male adolescents to improve the evaluation of their genitalia.We considered all patients treated for external genital pathology in the period of adolescence.Inclusion and exclusion criteria were created.Patients underwent an evaluation test before and after surgery.During the study period,137 patients were treated,and at the end of the study,98 cases were considered for analysis.The most frequent pathologies were webbed penis and penile curvature.A postoperative score improvement was noted and patients with concealed penis and webbed penis showed a better postoperative outcome.Overweight was considered an important factor associated with a worse preoperative score.Evaluation of the external genitalia is important in adolescents,and it is an understudied problem.Overweight may be associated with a worse evaluation of one’s genital and should be clinically considered to avoid related social problems in adulthood.Therefore,cosmetic genital surgery should be considered even in male adolescents. 展开更多

BACKGROUND Adipose-derived mesenchymal stem cells(ASCs)are characterized by long-term self-renewal and a high proliferation rate.Under adequate conditions,they may differentiate into cells belonging to mesodermal,endodermal or ectodermal lineages.Pericytes support endothelial cells and play an important role in stabilizing the vessel wall at the microcirculation level.The loss of pericytes,as occurs in diabetic retinopathy,results in a breakdown of the blood-retina barrier(BRB)and infiltration of inflammatory cells.In this context,the use of pericytelike differentiated ASCs may represent a valuable therapeutic strategy for restoring BRB damage.AIM To test in vitro strategies to obtain pericyte-like differentiation of human ASCs(hASCs).METHODS Different culture conditions were tested:hASCs cultured in a basal medium supplemented with transforming growth factorβ1;and hASCs cultured in a specific pericyte medium(PM-hASCs).In a further sample,pericyte growth supplement was omitted from the PM.In addition,cultures of human retinal pericytes(hRPCs)were used for comparison.Pericyte-like differentiation of hASCs was tested by immunocytochemical staining and western blotting to evaluate the expression ofα-smooth muscle actin(α-SMA)and neural/glial antigen 2(NG2).Interactions between human retinal endothelial cells(hRECs)and different groups of hASCs were investigated in co-culture experiments.In these cases,the expression of typical junctional proteins such as vascular endothelial-Cadherin,zonula occludens-1 and Occludin were assessed in hRECs.In an in vitro model of the BRB,values of trans-endothelial electrical resistance were measured when hRECs were co-cultured with various groups of pretreated hASCs.The values observed were compared with co-cultures of hRECs and hRPCs as well as with cultures of hRECs alone.Three-dimensional co-cultures of hRECs and hRPCs or pericyte-like hASCs in Matrigel were designed to assess their reciprocal localization.RESULTS After 3-6 d of culture,α-SMA and NG2 immunocytochemistry showed that the closest pericyte-like phenotype was observed when hASCs were cultured in Pericyte Medium(PM-hASCs).In particular,α-SMA immunoreactivity,already visible at the basal level in pericytes and ASCs,was strongly increased only when transforming growth factor was added to the culture medium.NG2 expression,almost undetectable in most conditions,was substantially increased only in PMhASCs.Immunocytochemical results were confirmed by western blot analysis.The presence of pericyte growth supplement seems to increase NG2 expression rather thanα-SMA,in agreement with its role in maintaining pericytes in the proliferative state.In co-culture experiments,immunoreactivity of vascular endothelial-Cadherin,zonula occludens-1 and Occludin was considerably increased in hRECs when hRPCs or PM-hASCs were also present.Supporting results were found by trans-endothelial electrical resistance measurements,gathered at 3 and 6 d of co-culture.The highest resistance values were obtained when hRECs were co-cultured with hRPCs or PM-hASCs.The pericyte-like phenotype of PM-hASCs was also confirmed in three-dimensional co-cultures in Matrigel,where PM-hASCs and hRPCs similarly localized around the tubular formations made by hRECs.CONCLUSION PM-hASCs seem able to strengthen the intercellular junctions between hRECs,likely reinforcing the BRB;thus,hASC-based therapeutic approaches may be developed to restore the integrity of retinal microcirculation. 展开更多

BACKGROUND Hepatitis C virus(HCV)infection and its consequent complications are undeniably a public health burden worldwide,particularly in Egypt.Emerging evidence suggests that many lncRNAs have relevant roles in viral infections and antiviral responses.AIM To investigate the expression profiles of circulating lncRNAGAS5,lncRNAHEIH,lncRNABISPR and mRNABST2 in naïve,treated and relapsed HCV Egyptian patients,to elucidate relation to HCV infection and their efficacy as innovative biomarkers for the diagnosis and prognosis of HCV GT4.METHODS One hundred and thirty HCV-infected Egyptian patients and 20 healthy controls were included in this study.Serum lncRNAs and mRNABST2 were measured using quantitative real-time polymerase chain reaction(qRT-PCR).RESULTS Our results indicated that serum lncRNAGAS5 and LncRNABISPR were upregulated,whereas mRNA BST2 and LncRNA HEIH were downregulated in naïve patients.In contrast,HCV patients treated with sofosbuvir and simeprevir;with sofosbuvir and daclatasvir;or with sofosbuvir,daclatasvir and ribavirin exhibited lower levels of lncRNAGAS5 and lncRNABISPR with higher mRNABST2 compared to naïve patients.Notably,patients relapsed from sofosbuvir and simeprevir showed higher levels of these lncRNAs with lower mRNABST2 compared to treated patients.LncRNAGAS5 and lncRNABISPR were positively correlated with viral load and ALT at P<0.001,whereas mRNABST2 was negatively correlated with viral load at P<0.001 and ALT at P<0.05.Interestingly,a significant positive correlation between lncRNA HEIH and AFP was observed at P<0.001.CONCLUSION Differential expression of these RNAs suggests their involvement in HCV pathogenesis or antiviral response and highlights their promising roles in diagnosis and prognosis of HCV. 展开更多

Total Hcy plasma level varies in the range of 5-15 μmol/L in the normal population. Our aim in this study was to investigate the possible correlations among homocysteine plasma levels, oxidative stress parameters and clinical evolution of stroke. Fifty patients with large-vessel ischemic stroke were studied. Biochemical determinations were performed at entry (T0) and then repeated one month after stroke (T1). Homocysteine levels were significantly increased at T0 with respect to T1 and showed a significant positive correlation with the expression of oxidative stress markers and a negative correlation with indicators of protective anti stress activity. A significant increase of antioxidant activity occurred from T0 to T1 and changes were associated with the severity of clinical conditions. In particular, the extent of homocysteine and of oxidative stress markers plasmatic levels re- duction and of the contemporary increase in anti stress biochemical activities were associated with a reduction of NIHSS scores. These findings, besides confirming an involvement of oxidative stress in in- fluencing the evolution of stroke, suggest a role for homocysteine as a potentially modifiable biochemical alteration able to modulate some mechanisms in- volved in the production of ischemic damage. 展开更多

The opposite biological functions between Bcl-xL and Bax stem from relatively minor differences in their structures. The C-terminal α helix, that functions in mitochondrial membrane targeting, sits in the hydrophobic BH3 binding pocket proposed previously to mediate heterodimer formation among Bcl-2 family members. Overview of the Prevalence and Associated Risk, Factors of Lifestyle Diseases in University Students

Background: The prevalence of lifestyle diseases is increasing rapidly in Youths (17 - 25 yrs). Factors such as poor diet, little or no physical exercise, and smoking are major contributors to this increase. Lifestyle diseases in youths present a timeline challenge as disease duration is longer and financial burden more costly. Aim: To analyse and synthesize published literature on the prevalence of and risk factors associated with four lifestyle diseases—obesity, hypertension, diabetes and cancer—in university/college students worldwide. Results: The literature indicates that among lifestyle diseases in university/college students, overweight/obesity (Body Mass Index > 25 kg/m2) had the highest prevalence of 45.6% and duration of approximately 5 years. Cardiovascular diseases and hypertension had lower prevalence rates but prehypertension which lingers >4 years was at 32.0%. Type 1 diabetes mellitus (T1DM) had a fairly high prevalence in college students (21.7%);type 2 diabetes mellitus (T2DM) was found in much lower percentage (12.95%). Cancer had the lowest prevalence of >2% but was increasing in most regions based on the presence of multiple risk factors. Conclusion: Increase prevalence of these diseases in youths indicates that young people are at high risk of developing these diseases due to poor early lifestyle habits. Early corrective measures can reduce the burden of many lifestyle diseases. 展开更多

Objective: Scandix pecten-veneris L. is a less studied wild edible herb and is considered an extinct plant species in many parts of the world. This study was designed to evaluate its phytochemical composition and biological potential of S. pecten-veneris L. Methods: Phytochemicals including alkaloids, fiavonoids, polyphenols, and tannins were determined in extracts of S. pecten-veneris. Antioxidant activity was determined using 2,2-diphenyl-1- picrylhydrazyl (DPPH), while reducing power was tested by ferric reducing/antioxidant power (FRAP) assay. Antimi- crobial activity against seven bacterial and four fungal strains was evaluated using agar well diffusion assay. Enzymes inhibition study was performed for urease, phosphodiesterase-I, and catalase-II. Results: S. pecten-veneris showed moderate antiradical activity and reducing potential of hydroxyl radicals to about 20% of the initial value. The antiox- idant activity of various extracts of S. pecten-veneris showed a linear correlation with total phenolic contents in the order of water〉n-butanol〉chloroform〉ethyl acetate〉methanol extracts. S. pecten-veneris leaves showed the highest inhibitory activity against Staphylococcus aureus while the highest antifungal activity was observed against Candida albicans. The plant extract was most potent against urease enzymes but showed moderate activity against phosphodiestrase-I and carbonic anhydrase-II. Conclusions: Our data demonstrate that in addition to its culinary uses, S. pecten-veneris has good medicinal potential and hence could be used for treating some specific health ailments. 展开更多

Integrating information on the glycemic index (GI) and the glycemic load (GL) of diet is limited in C?te d’Ivoire because of the lack of data. Thus, this study was undertaking for the local management and prevention of diabetes mellitus and its complications based on nutritional data (GI and GL values). The study included ten healthy subjects with 7 males, 3 females (28 ± 2 years on average age and 20.5 ± 1.7 on average BMI). Participants tested three different meals with equal carbohydrate load (50 g). Blood samples were obtained at 0, 15, 30, 45, 60, 90 and 120 min before and after consumption for glucose levels determination. GIs were determined using a standard method with glucose as reference food and data were used for GLs calculating. Data showed that GIs value of pounded yam with eggplant sauce and cassava paste with granulates palm nut sauce were high ranging to 94 to 86 respectively while rice with groundnut sauce, have a low GI (GI = 45). Nevertheless, the GLs of the all the test foods are high with the values of 47, 43 and 23 (g) for pounded yam with eggplant sauce, cassava paste with granulates palm nut sauce and rice with groundnut sauce respectively. According to GIs and GLs data, the three test foods must be consumed moderately in a diet. So, it is important to associate GL data to GI data of Ivorian traditional foods for the management and the prevention of diabetes mellitus in C?te d’Ivoire and in others countries sharing the same food tradition. 展开更多

MTT cell viability, dye exclusion, caspase activity and microscopic assessment of apoptotic cells demonstrated that DaJa-3 displayed cytotoxicity to both lung and prostate cancer cells. The A549 cells were more sensitive toward DaJa-3 with an IC</span><sub><span style="font-family:Verdana;">50</span></sub><span style="font-family:Verdana;"> value of 22.28 μg/mL (CI 28.42 to 36.63 μg/mL), compared to that of DU145 cells with an IC</span><sub><span style="font-family:Verdana;">50</span></sub><span style="font-family:Verdana;"> value of 31.45 μg/mL (CI 27.58 to 35.86 μg/mL). The ubiquitousness of naturally occurring boron-containing compounds(BCCs) has led to their constant contact with humankind.Recently,many synthetic BCCs have been elaborated for a broad spectrum of purposes,especially boric,boronic and borinic acids.Although BCCs were once employed primarily as antiseptics and later as antibiotics,they have become an increasingly relevant therapeutic tool.Nevertheless,this potential of BCCs has been drastically limited due to some unfortunate intra-hospital accidents in the 1940 s and 1950 s.The increasing use of BCCs as insecticides,antimicrobials,and other agents is providing new insights into their role in the physiology of several living species and in the pathophysiology of humans.It is becoming clear that BCCs act through a wide range of mechanisms,as do their corresponding boron-free counterparts.When comparing BCCs and similar boron-free compounds,in many cases the former show advantages in the medical field.The current minireview focuses on how BCCs have been developed by means of translational medicine,a process connecting biomedical research with clinical applications.This process of discovery is currently in an exponential stage. 展开更多

Carbon dots(CDs)have emerged as promising nanomaterials for bioimaging and stress monitoring due to their unique optical and functional properties.CDs were synthesized using citric acid and o-phenylenediamine via microwave-assisted heating,named as CP-CDs.High-resolution transmission electron microscopy observed an average particle size of 3.65±0.40 nm with graphitic cores.Raman spectroscopy and Fourier transform infrared spectroscopy confirmed diverse functional groups.The CDs exhibited excitation-dependent fluorescence with a peak emission at 432 nm,a high quantum yield of 54.91%,and a fluorescence lifetime of 9.50±0.15 ns,making them highly suitable for bioimaging.Confocal microscopy demonstrated tissue-specific localization in lettuce plant cells.In stem cells,CP-CDs predominantly targeted mitochondria,confirmed by a colocalization with Mito-Tracker Red.In contrast,leaf cells showed selective accumulation at the stomatal openings.Under salt-and heat-induced stress,stem cells exhibited an increase in mitochondrial fluorescence,indicating stress-responsive interactions,whereas leaf cells maintained consistent stomatal localization.Further,enhanced fluorescence from chloroplasts under stress conditions suggested synergistic effects with chlorophyll.Also,stress conditions caused CP-CDs to accumulate at the cell boundaries in stem cells,highlighting their sensitivity to stress-induced changes.These findings demonstrate the optical properties,tissue-specific uptake,and organelle-level localization of CP-CDs,underlining their potential for bioimaging,stress detection,and targeted delivery systems in plants. 展开更多
