Objective: To test the antioxidant, antimicrobial and a-glucosidase inhibitory activities of the roots extracts from Hertia cheirifolia(H. cheirifolia) L.Methods: Total phenolics and total flavonoids content of the di...Objective: To test the antioxidant, antimicrobial and a-glucosidase inhibitory activities of the roots extracts from Hertia cheirifolia(H. cheirifolia) L.Methods: Total phenolics and total flavonoids content of the different extracts were determined by colorimetric methods and reverse phase high-performance liquid chromatography(RP-HPLC) was performed to identify various chemical components.The different extracts were evaluated for antioxidant activities by 2,2-diphenyl-1-picrylhydrazyl(DPPH), 2,2-azino-bis-3-ethylenebenzothiozoline-6-sulfonic acid(ABTS·^+) and β-carotene bleaching tests and α-glucosidase inhibitory properties. The antimicrobial activity was carried out in vitro by the broth dilution method.Results: Trans-cinnamic acid, rutin hydrate, naringin and quercetin were the main compounds of the ethyl acetate extract from H. cheirifolia L. This extract has significant scavenging activity to decrease free radicals especially for DPPH and ABTS radicals. As well as, the ethyl acetate extract exhibited an antimicrobial property against bacterial strains. Bacillus licheniformis and Salmonella enterica were the most sensitive strains with minimum inhibitory concentration values of 0.156 mg/mL.Conclusion: The ethyl acetate extract was found to be selectively antioxidant and antimicrobial.展开更多
Background: Chagas disease is still a public health problem because of the remaining high prevalence, the expansion of the disease into developed countries and the re-emergences by oral transmission outbreaks. Chagas ...Background: Chagas disease is still a public health problem because of the remaining high prevalence, the expansion of the disease into developed countries and the re-emergences by oral transmission outbreaks. Chagas cardiomyopathy evolves as a consequence of an autonomic un-balance where the parasympathetic tone is undermined. Objective: To determine the functionality and expression of muscarinic cholinergic receptors in acute Chagas disease. Methodology: 62 male, 3-week-old Sprague Dawley rats were assayed;32 were infected with Trypanosoma cruzi trypo-mastigotes and 30 were healthy controls. Electrocardiographic studies were conducted in the absence or presence of direct muscarinic (oxotremorine and McN-A-343) or indirect agonists (phenylephrine) or antagonist (pirenzepine). Muscarinic M1 and M2 receptor expression was determined by radioligand [3H]-QNB binding assay and immunoblot. Results: Chagasic acute myocarditis was sustained by electrocardiographic signs and histopathological findings. Bradycardia induced by oxotremorine was significantly higher in healthy rats (HR) and the differences were enhanced by CsCl. In the absence of the agonist, CsCl induced a greater bradycardia in chagasic rats (ChR). In HR McN-A-343 induced tachycardia, however it induces bradycardia in the presence of a acetylcholinesterase inhibitor (neostigmine);no effects were observed in ChR. Pirenzepine induced a higher tachycardia in HR. Phenylephrine in the presence of pirenzepine induced a similar bradycardia in both groups, but recovery was faster in ChR. Levan is a fructose homopolymer obtained from both plants and microorganisms.Microbial levan is typically produced in a variety of molecular weights(MW).Previous studies have shown that different MW of levan exhibit d...Levan is a fructose homopolymer obtained from both plants and microorganisms.Microbial levan is typically produced in a variety of molecular weights(MW).Previous studies have shown that different MW of levan exhibit different biological activities.Though levan was reported for its immunomodulatory effect by increasing nitric oxide(NO)production in RAW264.7 macrophage cells,whether its MW can affect this activity remains unknown.In this study,the effect of different levan MW from Bacillus subtilis on its immunoregulatory properties was investigated.B.subtilis was cultivated using various cultivation parameters to obtain levan of different MW.Three weight average MW of levan,>8×10^(5) Da(M1),1.5×10^(4) Da(M2)and 3.7×10^(3) Da(M3),were selected to test for their effect on modulating NO levels in RAW264.7 cells.Our results showed that M2 increased NO production in a dose-dependent manner,and more effectively than that of M1 and M3.Consistent with these findings,M2 significantly stimulated inducible nitric oxide synthase(iNOS)and cyclooxygenase-2(COX2)protein expression,while less pronounced effects were observed in the cells treated with M1 and M3.Therefore,our results demonstrate that various MW of levan have varying effects on its immunoregulatory activities in RAW264.7 cells.Furthermore,our findings suggest that the size of levan should be taken into account when employing levan as one of the active ingredients in functional foods and immunomodulatory products.展开更多
