Neuronal cell death is a common outcome of multiple pathophysiological processes and a key factor in neurological dysfunction after subarachnoid hemorrhage.Neuronal ferroptosis in particular plays an important role in...Neuronal cell death is a common outcome of multiple pathophysiological processes and a key factor in neurological dysfunction after subarachnoid hemorrhage.Neuronal ferroptosis in particular plays an important role in early brain injury.Bromodomain-containing protein 4,a member of the bromo and extraterminal domain family of proteins,participated in multiple cell death pathways,but the mechanisms by which it regulates ferroptosis remain unclear.The primary aim of this study was to investigate how bromodomain-containing protein 4 affects neuronal ferroptosis following subarachnoid hemorrhage in vivo and in vitro.Our findings revealed that endogenous bromodomain-containing protein 4 co-localized with neurons,and its expression was decreased 48 hours after subarachnoid hemorrhage of the cerebral cortex in vivo.In addition,ferroptosis-related pathways were activated in vivo and in vitro after subarachnoid hemorrhage.Targeted inhibition of bromodomain-containing protein 4 in neurons increased lipid peroxidation and intracellular ferrous iron accumulation via ferritinophagy and ultimately led to neuronal ferroptosis.Using cleavage under targets and tagmentation analysis,we found that bromodomain-containing protein 4 enrichment in the Raf-1 promoter region decreased following oxyhemoglobin stimulation in vitro.Furthermore,treating bromodomain-containing protein 4-knockdown HT-22 cell lines with GW5074,a Raf-1 inhibitor,exacerbated neuronal ferroptosis by suppressing the Raf-1/ERK1/2 signaling pathway.Moreover,targeted inhibition of neuronal bromodomain-containing protein 4 exacerbated early and long-term neurological function deficits after subarachnoid hemorrhage.Our findings suggest that bromodomain-containing protein 4 may have neuroprotective effects after subarachnoid hemorrhage,and that inhibiting ferroptosis could help treat subarachnoid hemorrhage.展开更多

Although the certified power conversion efficiency(PCE)of single-junction perovskite solar cells(PSCs)has achieved a high level of 27%,approaching the single-crystalline silicon solar cells,the device stability remain...Although the certified power conversion efficiency(PCE)of single-junction perovskite solar cells(PSCs)has achieved a high level of 27%,approaching the single-crystalline silicon solar cells,the device stability remains an urgent issue to be resolved for the commercialization.Defect passivation emerged as a viable approach to enhance the operational stability of the solar devices.Herein,phenylthiourea(PhTu)derivatives are selected as effective passivation agents to enhance the optoelectronic properties of printed methylammonium lead iodide(MAPbI_(3))films.It is demonstrated that incorporating a small amount of 1-(4-carboxyphenyl)-2-thiourea(PhTu-COOH)significantly reduces the trap-state density and leads to longer carrier lifetime of the perovskite films.As a result,the inverted solar device made of Ph Tu-COOH-modified MAPbI_(3) perovskite film shows remarkably improved efficiency(from 17.29%to 20.22%)and obviously increased open-circuit voltage(V_(OC))(from 1.043 to 1.143 V),as compared with the pristine device.Moreover,the Ph Tu-COOH-modified PSCs exhibit enhanced operational stability due to the significantly reduced trap-state density.Finally,the optimized solar module fabricated with an active area of 11.28 cm^(2) delivers a high PCE of 17.07%with negligible V_(OC)loss,demonstrating the feasibility of the blade-coating method for large-area perovskite film deposition.展开更多
Atmospheric black carbon(BC)significantly impacts climate change,atmospheric environment,and human health.To explore the temporal variation of BC concentration and its health effects during different periods in urban ...Atmospheric black carbon(BC)significantly impacts climate change,atmospheric environment,and human health.To explore the temporal variation of BC concentration and its health effects during different periods in urban Jinan from 2021 to 2022,we analyzed BC concentration characteristics and assessed the population exposure risk using Monte Carlo simulation.Additionally,we studied the role of driving factors on BC using SHapley Additive exPlanations interpretation algorithm.A clear temporal trend was observed in BC levels,with the highest BC concentration in winter(2.40±1.76μg/m^(3))and the lowest in summer(1.46±0.89μg/m^(3)),and a bimodal pattern in diurnal variation.Our results indicated that the carcinogenic risk of BC for adults exceeded the acceptable threshold in winter.With emissions playing a dominant role,meteorological conditions also significantly affected BC,with wind speed,relative humidity,and boundary layer height playing major roles in variations of BC concentration.Meteorological conditions during the haze periods favored BC accumulation,hours with elevated PM_(2.5) levels were associated with high BC concentrations and increased carcinogenic effects.Understanding the temporal variations and drivers of BC is crucial for formulating effective policies to mitigate air pollution.Our study may provide theoretical methods and practical guidance for addressing BC levels under varying meteorological conditions and enhance understanding of atmospheric characteristics and population exposure to BC.展开更多
Recent studies have indicated that stroke can lead to neuronal iron overload and lipid peroxidation.Lycium barbarum glycopeptide,which has a low molecular weight and potent antioxidant properties,may mitigate ferropto...Recent studies have indicated that stroke can lead to neuronal iron overload and lipid peroxidation.Lycium barbarum glycopeptide,which has a low molecular weight and potent antioxidant properties,may mitigate ferroptosis in stroke.We hypothesized that Lycium barbarum glycopeptide can effectively mitigate iron overload within ischemic neurons due to its robust antioxidant properties.The aims of this study were to investigate the effects of Lycium barbarum glycopeptide on ferroptotic damage following brain ischemia and explore the underlying mechanisms.A rat model of middle cerebral artery occlusion was established using the intraluminal filament method,and the rats were treated with Lycium barbarum glycopeptide for 7 consecutive days,beginning 24 hours after ischemia.Liproxstatin-1,a ferroptosis inhibitor,and Erastin,a ferroptosis activator,were used as controls.We found that treatment with Lycium barbarum glycopeptide resulted in significant reductions in infarct volume(as detected by triphenyltetrazolium chloride staining staining and magnetic resonance imaging)and neuronal death(as measured by Nissl staining),as well as improvements in sensory and motor functions in rats subjected to middle cerebral artery occlusion.Furthermore,treatment with Lycium barbarum glycopeptide alleviated anxiety and depression-like behaviors and improved memory.Additionally,Lycium barbarum glycopeptide effectively reduced the iron ion content in the ischemic penumbra of the cortex.Moreover,treatment with Lycium barbarum glycopeptide downregulated the expression of ferroptotic and oxidant proteins such as transferrin receptor 1,divalent metal transporter 1,and Acyl-CoA synthetase long-chain family member 4 and upregulated the expression of anti-ferroptotic and antioxidant proteins such as ferroportin 1,solute carrier family 7 member 11,glutathione,and glutathione peroxidase 4.However,these beneficial effects were reversed when ferroptosis was induced with the activator Erastin.Therefore,the positive effects of Lycium barbarum glycopeptide in ischemic stroke are likely mediated through activation of the anti-ferroptotic pathway and the antioxidative System Xc-glutathione-glutathione peroxidase 4 pathway.Overall,our findings highlight the potential use of Lycium barbarum glycopeptide as a neuroprotective agent targeting both ferroptosis and oxidation to decrease ischemic brain damage.展开更多
In-situ heating conversion is the most practical recovery method for lacustrine low-to-medium maturity shale oil.However,the energy output-input ratio must exceed the economic threshold to achieve commercial developme...In-situ heating conversion is the most practical recovery method for lacustrine low-to-medium maturity shale oil.However,the energy output-input ratio must exceed the economic threshold to achieve commercial development.This paper systematically investigates the mechanism of super-rich accumulation of organic matter in continental shale,sweet spot evaluation,optimal heating windows,and appropriate well types and patterns from the perspectives of enhancing energy output and reducing energy input.(1)The super-rich accumulation of organic matter in lacustrine shale is primarily controlled by the intensity,frequency,and preservation of external material inputs,and is related to moderate volcanic and hydrothermal activities,marine transgressions,with total organic carbon content greater than or equal to 6%.(2)The quality of organic-rich intervals is related to the type of source material and hydrocarbon generation potential.The in-situ conversion-derived hydrocarbon quality index(HQI)is established,and the zones exhibiting HQI>450 are defined as sweet spots.(3)Considering the characteristics of the organic matter conversion material field and seepage field,the temperature interval 300-370℃is recommended as the optimal heating window for the Chang 7_(3)sub-member of the Triassic Yanchang Formation in the Ordos Basin.Based on the advantages of thermal conductivity,permeability,and hydrocarbon expulsion efficiency along the bedding direction during in-situ heating,the“horizontal well heating+vertical well development”scheme is proposed,which has demonstrated significant enhancement in both recovery factor and energy output-input ratio,making it the optimal in-situ conversion process.The research findings provide a theoretical and technical foundation for the economical and efficient development of low-to-medium maturity shale oil.展开更多
This study investigated the heterogeneous responses of organic matter(OM)in highly-to over-mature source rocks during thermal maturation.An integrated analysis was conducted on the Raman spectroscopic and geochemical ...This study investigated the heterogeneous responses of organic matter(OM)in highly-to over-mature source rocks during thermal maturation.An integrated analysis was conducted on the Raman spectroscopic and geochemical signatures of shales from the Lower Silurian Longmaxi Formation and the Lower Cambrian Qiongzhusi Formation,as well as anthracites from the Lower Permian Shanxi–Formation and the Upper Carboniferous Taiyuan Formation(collectively referred to as the Shanxi Taiyuan Formations).Additionally,burial and thermal evolution modeling was employed to support the analysis.A systematic assessment of Raman spectral parameters(e.g.,the positions and intensity ratio of the D and G bands)revealed robust correlations between the thermal history patterns of source rocks and molecular structural evolution parameters.The subsequent mechanistic quantification demonstrated that the maturation state of the source rocks was subjected to the hierarchical control of three principal factors:Peak heating temperature,the duration of sustained thermal intensity,and effective maturation duration.In addition,comparative analyses demonstrated that the anthracites attained higher structural ordering under sustained thermal conditions.This contrasts with the disordered carbon matrices observed in the intermittently heated shales.Raman spectroscopy further revealed broader variations in the D and G band intensities of the Longmaxi Formation compared to the Qiongzhusi Formation.This difference is associated with their different thermal histories.The thermal burial histories confirm that shales in the Longmaxi Formation underwent thermal exposure at lower peak temperatures over a shorter duration compared to those in the Qiongzhusi Formation.Finally,this study established a maturity calibration model for over-mature source rocks through a systematic correlation between Raman peak height ratios(R_(D/G))and vitrinite reflectance(R_(o)).展开更多
Photoreceptor degeneration is a major cause of vision impairment in retinal diseases,for which no effective treatment currently exists.Previous research by our team demonstrated that Lycium barbarum glycopeptide and l...Photoreceptor degeneration is a major cause of vision impairment in retinal diseases,for which no effective treatment currently exists.Previous research by our team demonstrated that Lycium barbarum glycopeptide and luteolin can independently promote photoreceptor survival and function in degenerated mouse retinas,although with limited efficacy.This study evaluated whether a combination of Lycium barbarum glycopeptide and luteolin provides enhanced therapeutic benefits compared with either compound alone.Wild-type mice received a daily oral gavage of Lycium barbarum glycopeptide and luteolin for 7 days prior to intraperitoneal injection of N-nitroso-N-methylurea to induce photoreceptor damage.The treatment continued for an additional week after injury.Retinal structure and function were subsequently assessed using electroretinogram recordings,visual behavior testing,and immunostaining.Western blot analysis was conducted to investigate the underlying protective mechanisms.The results showed that the Lycium barbarum glycopeptide-luteolin mixture significantly increased photoreceptor survival,improved retinal light response,and enhanced visual behavior.Importantly,the combination outperformed either compound alone in protective efficacy.Mechanistic analysis indicated that the mixture suppressed retinal inflammation and modulated the extracellular signal-regulated kinase and Bcl-2-associated X protein/B-cell lymphoma 2 signaling pathways.These findings suggest that the combination of Lycium barbarum glycopeptide and luteolin represents a promising therapeutic strategy for photoreceptor degeneration.展开更多
Because the pathogenesis of Alzheimer’s disease is multifactorial and complex,integrated multi-level omics analysis is essential to comprehensively elucidate its molecular alterations.We therefore utilized the well-e...Because the pathogenesis of Alzheimer’s disease is multifactorial and complex,integrated multi-level omics analysis is essential to comprehensively elucidate its molecular alterations.We therefore utilized the well-established amyloid precursor protein/presenilin 1 mouse model to carry out an integrated multi-omics study using transcriptomic,proteomic,N^(6)-methyladenosine epitranscriptomic,and phosphoproteomic analyses.The results revealed substantial molecular alterations across multiple biological dimensions and the alteration in the expression of several key genes,such as GFAP,APP,and RTN4,in a mouse model of Alzheimer’s disease.The pronounced elevation of RTN4 in reactive astrocytes is indicative of its involvement in Alzheimer’s disease pathogenesis.Furthermore,we identified dysregulation of pathways related to endocytosis,highlighting the critical role of this process in disease progression.Our findings underscore the significant impact of post-transcriptional(N^(6)-methyladenosine methylation)and post-translational(phosphorylation)protein modifications,which have been underrepresented in Alzheimer’s disease research.The significant contribution made by this study is the integrated,multi-level omics analysis that we carried out to investigate the complex biological changes that occur in Alzheimer’s disease.Our findings provide novel insights into Alzheimer’s disease pathogenesis and suggest potential therapeutic targets,such as RTN4.展开更多

In addition to the organic matter type,abundance,thermal maturity,and shale reservoir space,the preservation conditions of source rocks play a key factor in affecting the quantity and quality of retained hydrocarbons ...In addition to the organic matter type,abundance,thermal maturity,and shale reservoir space,the preservation conditions of source rocks play a key factor in affecting the quantity and quality of retained hydrocarbons in source rocks of lacustrine shale,yet this aspect has received little attention.This paper,based on the case analysis,explores how preservation conditions influence the enrichment of mobile hydrocarbons in shale oil.Research showns that good preservation conditions play three key roles.(1)Ensure the retention of sufficient light hydrocarbons(C_(1)–C_(13)),medium hydrocarbons(C_(14)–C_(25))and small molecular aromatics(including 1–2 benzene rings)in the formation,which enhances the fluidity and flow of shale oil;(2)Maintain a high energy field(abnormally high pressure),thus facilitating the maximum outflow of shale oil;(3)Ensure that the retained hydrocarbons have the miscible flow condition of multi-component hydrocarbons(light hydrocarbons,medium hydrocarbons,heavy hydrocarbons,and heteroatomic compounds),so that the heavy hydrocarbons(C_(25+))and heavy components(non-hydrocarbons and asphaltenes)have improved fluidity and maximum flow capacity.In conclusion,in addition to the advantages of organic matter type,abundance,thermal maturity,and reservoir space,good preservation conditions of shale layers are essential for the formation of economically viable shale oil reservoirs,which should be incorporated into the evaluation criteria of shale oil-rich areas/segments and considered a necessary factor when selecting favorable exploration targets.展开更多
This research presents a numerical simulation methodology for optimizing circular composite overlays’dimensions and pressure characteristics with orthotropic mechanical properties,specifically,for metal conduits with...This research presents a numerical simulation methodology for optimizing circular composite overlays’dimensions and pressure characteristics with orthotropic mechanical properties,specifically,for metal conduits with temperature-dependent elastoplastic behavior.The primary objective of the proposed method is to prevent crack propagation during pressure surges from operational to critical levels.This study examines the“Beineu-Bozoy-Shymkent”steel gas conduit,examining its performance across a temperature range of−40 to+50℃.This work builds on prior research on extended avalanche destruction in steel gas conduits and crack propagation prevention techniques.Theanalysis was conducted using a dynamic finite-element approach with the ANSYS-19.2/ExplicitDynamics software.Simulations of unprotected conduits revealed that increasing gas-dynamic pressure can convert a partial-depth crack into a through-crack,extending longitudinally to approximately seven times its initial length.Notably,at T=+50℃,the developed crack length was 1.2%longer than that at T=−40℃,highlighting the temperature sensitivity of crack progression.The modeling results indicate that crack propagation can be effectively controlled using a circular composite overlay with a thickness between 37.5%and 50%of the crack depth and a length approximately five times that of the initial crack,centered symmetrically over the crack.In addition,preliminary stress analysis indicated that limiting the overlay-induced pressure to 5%of the operational pressure effectively arrested crack growth without generating significant stress concentrations near the overlay boundaries,thereby preventing conduit integrity.展开更多

A later heading date generally leads to higher grain yield in favorable ecological regions;however,grain yield reaches a limit as the heading date exceeds a certain threshold.Ghd7 is the first cloned major gene that r...A later heading date generally leads to higher grain yield in favorable ecological regions;however,grain yield reaches a limit as the heading date exceeds a certain threshold.Ghd7 is the first cloned major gene that regulates heading date,plant height and grain number.Here,we investigated the relationship between Ghd7 and florigen genes Hd3a and RFT1,to determine their roles in regulating heading date and grain number under different photoperiods.Our results revealed that under long-day(LD)conditions,Hd3a acts prior to RFT1 to promote heading while negatively regulating plant height and grain number.In contrast,Ghd7 positively regulates heading date,plant height,and grain number by inhibiting both Hd3a and RFT1.Under short-day(SD)conditions,the functions of Hd3a and RFT1 remain consistent with those under LD conditions,but Ghd7 does not inhibit their expression,resulting in a weaker phenotypic effect compared to Hd3a.Additionally,under both LD and SD conditions,increased Ghd7 expression enhances its inhibitory effect on Hd3a and RFT1,leading to later heading and increased grain number;however,once the heading date exceeds 94 d,grain number no longer increases.Moreover,the gn1a allele increased grain number by 16.5%to 42.5%,while combinations of the elite alleles from Ghd7,Hd3a,RFT1,and Gn1a significantly increased grain number by up to 240.9%.Therefore,we propose a new breeding strategy to optimize the heading date and grain number using the Ghd7Hd3aRFT1gn1a combination of Ghd7,Hd3a,RFT1,and Gn1a under LD conditions,and the Ghd7hd3aRFT1gn1a combination under SD conditions.This strategy improved the yield of the high-quality Northeast variety Kongyu 131(KY131)by 69.1%in Beijing and 93.7%in Hainan.This strategy will greatly improve the efficiency of north-to-south adaptation in rice,providing theoretical guidance for expanding the geographical adaptability of rice varieties.展开更多
The article examines the impact of increased aridization of the territory due to an increase in air temperature,reduced precipitation,and the formation of moisture deficiency on grain yields in Northern Kazakhstan.The...The article examines the impact of increased aridization of the territory due to an increase in air temperature,reduced precipitation,and the formation of moisture deficiency on grain yields in Northern Kazakhstan.The most important result of the work is the revealed inverse relationship between grain yields and the temperature of the growing season:low-yielding years are associated with high temperatures and droughts,and high-yielding years are associated with lower temperatures and an optimal ratio of heat and moisture.The novelty of this study is the use of the method of hydrological and climatic calculations in identifying the nature of temperature variability and precipitation in the territory of Northern Kazakhstan for the modern period(1991–2020)compared with the base period(1961–1990).At all the studied meteorological stations,there is a tendency for the average annual temperature and the temperature of the growing season to increase:in the forest-steppe zone with an average warming intensity of 0.3–0.33℃ per decade;in the steppe zone by 0.2–0.43℃ per decade;and in the growing season by 0.2–0.7℃ per decade.The air temperature in the steppe zone is rising more intensively than in the forest-steppe zone,and precipitation in the forest-steppe zone has changed more than in the steppe zone.An increase in the average annual air temperature during the growing season(May–August),combined with a shortage of atmospheric moisture or a constant amount of it,led to an increase in the degree of aridization of the territory,an increase in the frequency of droughts in the steppe zone of Northern Kazakhstan.展开更多
Selenium is one of the important trace elements in the human body.Its deficiency will directly affect human health.With people's attention to health,the content of selenium in food has gradually attracted attentio...Selenium is one of the important trace elements in the human body.Its deficiency will directly affect human health.With people's attention to health,the content of selenium in food has gradually attracted attention.However,detecting selenium compounds in complex samples remains a challenge.In this work,we built an online heating-reaction device.This device combines the electrospray extraction ionization mass spectrometry(EESI-MS)with the heating reaction device,which can simultaneously detect various selenium compounds in complex liquid samples.Under acidic conditions,the sample was heated and catalyzed by a heating reaction device,so that the SeO~(2-)_(3)and O-phenylenediamine(OPD)could generate 1,3-dihydro-2,1,3-benzoselenadiazole.Based on the above reactions,we can detect organic selenium,inorganic selenium and other compounds in liquid samples by organic mass spectrometry.In this experiment,we determined the content of three forms of selenium:selenomethionine(SeMet),l-selenocystine(SeCys(2)),and sodium selenite.The calibration curves for SeMet,SeCys(2),and sodium selenite showed strong linearity within a range of 0.50-50.00μg/L.The limits of detection(LOD)for the three compounds were 0.22,0.27,and 0.41μg/L,respectively.The limits of quantification(LOQ)were 0.68,0.81,and 1.23μg/L,respectively.Spiked recoveries at three levels ranged from 98.8%to 106.1%.In addition,this method can simultaneously detect three selenium compounds and three other specific chemical components in tea infusion samples,providing a rapid and efficient method for identifying tea quality.展开更多

文摘Neuronal cell death is a common outcome of multiple pathophysiological processes and a key factor in neurological dysfunction after subarachnoid hemorrhage.Neuronal ferroptosis in particular plays an important role in early brain injury.Bromodomain-containing protein 4,a member of the bromo and extraterminal domain family of proteins,participated in multiple cell death pathways,but the mechanisms by which it regulates ferroptosis remain unclear.The primary aim of this study was to investigate how bromodomain-containing protein 4 affects neuronal ferroptosis following subarachnoid hemorrhage in vivo and in vitro.Our findings revealed that endogenous bromodomain-containing protein 4 co-localized with neurons,and its expression was decreased 48 hours after subarachnoid hemorrhage of the cerebral cortex in vivo.In addition,ferroptosis-related pathways were activated in vivo and in vitro after subarachnoid hemorrhage.Targeted inhibition of bromodomain-containing protein 4 in neurons increased lipid peroxidation and intracellular ferrous iron accumulation via ferritinophagy and ultimately led to neuronal ferroptosis.Using cleavage under targets and tagmentation analysis,we found that bromodomain-containing protein 4 enrichment in the Raf-1 promoter region decreased following oxyhemoglobin stimulation in vitro.Furthermore,treating bromodomain-containing protein 4-knockdown HT-22 cell lines with GW5074,a Raf-1 inhibitor,exacerbated neuronal ferroptosis by suppressing the Raf-1/ERK1/2 signaling pathway.Moreover,targeted inhibition of neuronal bromodomain-containing protein 4 exacerbated early and long-term neurological function deficits after subarachnoid hemorrhage.Our findings suggest that bromodomain-containing protein 4 may have neuroprotective effects after subarachnoid hemorrhage,and that inhibiting ferroptosis could help treat subarachnoid hemorrhage.

文摘Although the certified power conversion efficiency(PCE)of single-junction perovskite solar cells(PSCs)has achieved a high level of 27%,approaching the single-crystalline silicon solar cells,the device stability remains an urgent issue to be resolved for the commercialization.Defect passivation emerged as a viable approach to enhance the operational stability of the solar devices.Herein,phenylthiourea(PhTu)derivatives are selected as effective passivation agents to enhance the optoelectronic properties of printed methylammonium lead iodide(MAPbI_(3))films.It is demonstrated that incorporating a small amount of 1-(4-carboxyphenyl)-2-thiourea(PhTu-COOH)significantly reduces the trap-state density and leads to longer carrier lifetime of the perovskite films.As a result,the inverted solar device made of Ph Tu-COOH-modified MAPbI_(3) perovskite film shows remarkably improved efficiency(from 17.29%to 20.22%)and obviously increased open-circuit voltage(V_(OC))(from 1.043 to 1.143 V),as compared with the pristine device.Moreover,the Ph Tu-COOH-modified PSCs exhibit enhanced operational stability due to the significantly reduced trap-state density.Finally,the optimized solar module fabricated with an active area of 11.28 cm^(2) delivers a high PCE of 17.07%with negligible V_(OC)loss,demonstrating the feasibility of the blade-coating method for large-area perovskite film deposition.
文摘Atmospheric black carbon(BC)significantly impacts climate change,atmospheric environment,and human health.To explore the temporal variation of BC concentration and its health effects during different periods in urban Jinan from 2021 to 2022,we analyzed BC concentration characteristics and assessed the population exposure risk using Monte Carlo simulation.Additionally,we studied the role of driving factors on BC using SHapley Additive exPlanations interpretation algorithm.A clear temporal trend was observed in BC levels,with the highest BC concentration in winter(2.40±1.76μg/m^(3))and the lowest in summer(1.46±0.89μg/m^(3)),and a bimodal pattern in diurnal variation.Our results indicated that the carcinogenic risk of BC for adults exceeded the acceptable threshold in winter.With emissions playing a dominant role,meteorological conditions also significantly affected BC,with wind speed,relative humidity,and boundary layer height playing major roles in variations of BC concentration.Meteorological conditions during the haze periods favored BC accumulation,hours with elevated PM_(2.5) levels were associated with high BC concentrations and increased carcinogenic effects.Understanding the temporal variations and drivers of BC is crucial for formulating effective policies to mitigate air pollution.Our study may provide theoretical methods and practical guidance for addressing BC levels under varying meteorological conditions and enhance understanding of atmospheric characteristics and population exposure to BC.

文摘Recent studies have indicated that stroke can lead to neuronal iron overload and lipid peroxidation.Lycium barbarum glycopeptide,which has a low molecular weight and potent antioxidant properties,may mitigate ferroptosis in stroke.We hypothesized that Lycium barbarum glycopeptide can effectively mitigate iron overload within ischemic neurons due to its robust antioxidant properties.The aims of this study were to investigate the effects of Lycium barbarum glycopeptide on ferroptotic damage following brain ischemia and explore the underlying mechanisms.A rat model of middle cerebral artery occlusion was established using the intraluminal filament method,and the rats were treated with Lycium barbarum glycopeptide for 7 consecutive days,beginning 24 hours after ischemia.Liproxstatin-1,a ferroptosis inhibitor,and Erastin,a ferroptosis activator,were used as controls.We found that treatment with Lycium barbarum glycopeptide resulted in significant reductions in infarct volume(as detected by triphenyltetrazolium chloride staining staining and magnetic resonance imaging)and neuronal death(as measured by Nissl staining),as well as improvements in sensory and motor functions in rats subjected to middle cerebral artery occlusion.Furthermore,treatment with Lycium barbarum glycopeptide alleviated anxiety and depression-like behaviors and improved memory.Additionally,Lycium barbarum glycopeptide effectively reduced the iron ion content in the ischemic penumbra of the cortex.Moreover,treatment with Lycium barbarum glycopeptide downregulated the expression of ferroptotic and oxidant proteins such as transferrin receptor 1,divalent metal transporter 1,and Acyl-CoA synthetase long-chain family member 4 and upregulated the expression of anti-ferroptotic and antioxidant proteins such as ferroportin 1,solute carrier family 7 member 11,glutathione,and glutathione peroxidase 4.However,these beneficial effects were reversed when ferroptosis was induced with the activator Erastin.Therefore,the positive effects of Lycium barbarum glycopeptide in ischemic stroke are likely mediated through activation of the anti-ferroptotic pathway and the antioxidative System Xc-glutathione-glutathione peroxidase 4 pathway.Overall,our findings highlight the potential use of Lycium barbarum glycopeptide as a neuroprotective agent targeting both ferroptosis and oxidation to decrease ischemic brain damage.

文摘In-situ heating conversion is the most practical recovery method for lacustrine low-to-medium maturity shale oil.However,the energy output-input ratio must exceed the economic threshold to achieve commercial development.This paper systematically investigates the mechanism of super-rich accumulation of organic matter in continental shale,sweet spot evaluation,optimal heating windows,and appropriate well types and patterns from the perspectives of enhancing energy output and reducing energy input.(1)The super-rich accumulation of organic matter in lacustrine shale is primarily controlled by the intensity,frequency,and preservation of external material inputs,and is related to moderate volcanic and hydrothermal activities,marine transgressions,with total organic carbon content greater than or equal to 6%.(2)The quality of organic-rich intervals is related to the type of source material and hydrocarbon generation potential.The in-situ conversion-derived hydrocarbon quality index(HQI)is established,and the zones exhibiting HQI>450 are defined as sweet spots.(3)Considering the characteristics of the organic matter conversion material field and seepage field,the temperature interval 300-370℃is recommended as the optimal heating window for the Chang 7_(3)sub-member of the Triassic Yanchang Formation in the Ordos Basin.Based on the advantages of thermal conductivity,permeability,and hydrocarbon expulsion efficiency along the bedding direction during in-situ heating,the“horizontal well heating+vertical well development”scheme is proposed,which has demonstrated significant enhancement in both recovery factor and energy output-input ratio,making it the optimal in-situ conversion process.The research findings provide a theoretical and technical foundation for the economical and efficient development of low-to-medium maturity shale oil.

文摘This study investigated the heterogeneous responses of organic matter(OM)in highly-to over-mature source rocks during thermal maturation.An integrated analysis was conducted on the Raman spectroscopic and geochemical signatures of shales from the Lower Silurian Longmaxi Formation and the Lower Cambrian Qiongzhusi Formation,as well as anthracites from the Lower Permian Shanxi–Formation and the Upper Carboniferous Taiyuan Formation(collectively referred to as the Shanxi Taiyuan Formations).Additionally,burial and thermal evolution modeling was employed to support the analysis.A systematic assessment of Raman spectral parameters(e.g.,the positions and intensity ratio of the D and G bands)revealed robust correlations between the thermal history patterns of source rocks and molecular structural evolution parameters.The subsequent mechanistic quantification demonstrated that the maturation state of the source rocks was subjected to the hierarchical control of three principal factors:Peak heating temperature,the duration of sustained thermal intensity,and effective maturation duration.In addition,comparative analyses demonstrated that the anthracites attained higher structural ordering under sustained thermal conditions.This contrasts with the disordered carbon matrices observed in the intermittently heated shales.Raman spectroscopy further revealed broader variations in the D and G band intensities of the Longmaxi Formation compared to the Qiongzhusi Formation.This difference is associated with their different thermal histories.The thermal burial histories confirm that shales in the Longmaxi Formation underwent thermal exposure at lower peak temperatures over a shorter duration compared to those in the Qiongzhusi Formation.Finally,this study established a maturity calibration model for over-mature source rocks through a systematic correlation between Raman peak height ratios(R_(D/G))and vitrinite reflectance(R_(o)).

文摘Photoreceptor degeneration is a major cause of vision impairment in retinal diseases,for which no effective treatment currently exists.Previous research by our team demonstrated that Lycium barbarum glycopeptide and luteolin can independently promote photoreceptor survival and function in degenerated mouse retinas,although with limited efficacy.This study evaluated whether a combination of Lycium barbarum glycopeptide and luteolin provides enhanced therapeutic benefits compared with either compound alone.Wild-type mice received a daily oral gavage of Lycium barbarum glycopeptide and luteolin for 7 days prior to intraperitoneal injection of N-nitroso-N-methylurea to induce photoreceptor damage.The treatment continued for an additional week after injury.Retinal structure and function were subsequently assessed using electroretinogram recordings,visual behavior testing,and immunostaining.Western blot analysis was conducted to investigate the underlying protective mechanisms.The results showed that the Lycium barbarum glycopeptide-luteolin mixture significantly increased photoreceptor survival,improved retinal light response,and enhanced visual behavior.Importantly,the combination outperformed either compound alone in protective efficacy.Mechanistic analysis indicated that the mixture suppressed retinal inflammation and modulated the extracellular signal-regulated kinase and Bcl-2-associated X protein/B-cell lymphoma 2 signaling pathways.These findings suggest that the combination of Lycium barbarum glycopeptide and luteolin represents a promising therapeutic strategy for photoreceptor degeneration.

文摘Because the pathogenesis of Alzheimer’s disease is multifactorial and complex,integrated multi-level omics analysis is essential to comprehensively elucidate its molecular alterations.We therefore utilized the well-established amyloid precursor protein/presenilin 1 mouse model to carry out an integrated multi-omics study using transcriptomic,proteomic,N^(6)-methyladenosine epitranscriptomic,and phosphoproteomic analyses.The results revealed substantial molecular alterations across multiple biological dimensions and the alteration in the expression of several key genes,such as GFAP,APP,and RTN4,in a mouse model of Alzheimer’s disease.The pronounced elevation of RTN4 in reactive astrocytes is indicative of its involvement in Alzheimer’s disease pathogenesis.Furthermore,we identified dysregulation of pathways related to endocytosis,highlighting the critical role of this process in disease progression.Our findings underscore the significant impact of post-transcriptional(N^(6)-methyladenosine methylation)and post-translational(phosphorylation)protein modifications,which have been underrepresented in Alzheimer’s disease research.The significant contribution made by this study is the integrated,multi-level omics analysis that we carried out to investigate the complex biological changes that occur in Alzheimer’s disease.Our findings provide novel insights into Alzheimer’s disease pathogenesis and suggest potential therapeutic targets,such as RTN4.

文摘The algorithm is designed to solve the global problem of multi-objective optimization with constraints in the context of greenhouse gas assessment and mitigation.Artificial intelligence provides unique opportunities for analyzing large amounts of data and identifying hidden relationships between various factors affecting emissions.The use of AI makes it possible to develop effective emission reduction strategies,predict the consequences of various scenarios,and evaluate the effectiveness of decisions made.Machine learning algorithms are capable of modeling complex systems such as energy infrastructure,transportation,and industry to determine the best ways to minimize emissions.The greenhouse effect and related climate change pose one of the most serious threats to our future.Innovative approaches and modern technologies are needed to effectively combat these problems.Government intelligence,in particular,Giga Chat,offers a variety of services for analysts,forecasting,and user support.Their use can significantly accelerate the transition to sustainable development and achieve the goals of the Paris Agreement to limit global temperature growth to 1.5℃.However,realizing the potential of AI requires careful preparation and consideration of many factors,including data quality,ethics,and technical aspects.Only through the joint efforts of scientists,politicians,and society will we be able to overcome the challenge of climate change and build a future that is safe for future generations.

文摘Alzheimer’s disease not only affects the brain,but also induces metabolic dysfunction in peripheral organs and alters the gut microbiota.The aim of this study was to investigate systemic changes that occur in Alzheimer’s disease,in particular the association between changes in peripheral organ metabolism,changes in gut microbial composition,and Alzheimer’s disease development.To do this,we analyzed peripheral organ metabolism and the gut microbiota in amyloid precursor protein-presenilin 1(APP/PS1)transgenic and control mice at 3,6,9,and 12 months of age.Twelve-month-old APP/PS1 mice exhibited cognitive impairment,Alzheimer’s disease-related brain changes,distinctive metabolic disturbances in peripheral organs and fecal samples(as detected by untargeted metabolomics sequencing),and substantial changes in gut microbial composition compared with younger APP/PS1 mice.Notably,a strong correlation emerged between the gut microbiota and kidney metabolism in APP/PS1 mice.These findings suggest that alterations in peripheral organ metabolism and the gut microbiota are closely related to Alzheimer’s disease development,indicating potential new directions for therapeutic strategies.
文摘ZnO is a highly significant II-VI semiconductor known for its excellent optoelectronic properties,making it widely applicable and promising for use in light-emitting devices,solar cells,lasers,and photodetectors.The methods for preparing ZnO are diverse,and among them,the hydrothermal method is favored for its simplicity,ease of operation,and low cost,making it an optimal choice for ZnO single-crystal growth.Most studies investigating the effects of different hydrothermal experimental parameters on the morphology and performance of ZnO nano-materials typically focus on only 2—3 variable parameters,with few examining the impact of all possible experimental parameter changes on ZnO nano-mate-rials.The principles of the hydrothermal method and its advantages in nano-material preparation were briefly introduced in this article.The detailed discussion on the influence of various experimental parameters on the preparation of ZnO nano-materials was provided,which including reaction materials,Zn^(2+)/OH^(-)ratio,reaction time and temperature,additives,experimental equipment,and annealing conditions.The review co-vers how different experimental parameters affect the morphology and performance of the materials,as well as how different rare earth doping elements influence the performance of ZnO nano-materials.It is hoped that this work will contribute to future research on the hydrothermal synthesis of nano-materials.

文摘In addition to the organic matter type,abundance,thermal maturity,and shale reservoir space,the preservation conditions of source rocks play a key factor in affecting the quantity and quality of retained hydrocarbons in source rocks of lacustrine shale,yet this aspect has received little attention.This paper,based on the case analysis,explores how preservation conditions influence the enrichment of mobile hydrocarbons in shale oil.Research showns that good preservation conditions play three key roles.(1)Ensure the retention of sufficient light hydrocarbons(C_(1)–C_(13)),medium hydrocarbons(C_(14)–C_(25))and small molecular aromatics(including 1–2 benzene rings)in the formation,which enhances the fluidity and flow of shale oil;(2)Maintain a high energy field(abnormally high pressure),thus facilitating the maximum outflow of shale oil;(3)Ensure that the retained hydrocarbons have the miscible flow condition of multi-component hydrocarbons(light hydrocarbons,medium hydrocarbons,heavy hydrocarbons,and heteroatomic compounds),so that the heavy hydrocarbons(C_(25+))and heavy components(non-hydrocarbons and asphaltenes)have improved fluidity and maximum flow capacity.In conclusion,in addition to the advantages of organic matter type,abundance,thermal maturity,and reservoir space,good preservation conditions of shale layers are essential for the formation of economically viable shale oil reservoirs,which should be incorporated into the evaluation criteria of shale oil-rich areas/segments and considered a necessary factor when selecting favorable exploration targets.

文摘This research presents a numerical simulation methodology for optimizing circular composite overlays’dimensions and pressure characteristics with orthotropic mechanical properties,specifically,for metal conduits with temperature-dependent elastoplastic behavior.The primary objective of the proposed method is to prevent crack propagation during pressure surges from operational to critical levels.This study examines the“Beineu-Bozoy-Shymkent”steel gas conduit,examining its performance across a temperature range of−40 to+50℃.This work builds on prior research on extended avalanche destruction in steel gas conduits and crack propagation prevention techniques.Theanalysis was conducted using a dynamic finite-element approach with the ANSYS-19.2/ExplicitDynamics software.Simulations of unprotected conduits revealed that increasing gas-dynamic pressure can convert a partial-depth crack into a through-crack,extending longitudinally to approximately seven times its initial length.Notably,at T=+50℃,the developed crack length was 1.2%longer than that at T=−40℃,highlighting the temperature sensitivity of crack progression.The modeling results indicate that crack propagation can be effectively controlled using a circular composite overlay with a thickness between 37.5%and 50%of the crack depth and a length approximately five times that of the initial crack,centered symmetrically over the crack.In addition,preliminary stress analysis indicated that limiting the overlay-induced pressure to 5%of the operational pressure effectively arrested crack growth without generating significant stress concentrations near the overlay boundaries,thereby preventing conduit integrity.

文摘Alzheimer's disease,the primary cause of dementia,is characterized by neuropathologies,such as amyloid plaques,synaptic and neuronal degeneration,and neurofibrillary tangles.Although amyloid plaques are the primary characteristic of Alzheimer's disease in the central nervous system and peripheral organs,targeting amyloid-beta clearance in the central nervous system has shown limited clinical efficacy in Alzheimer's disease treatment.Metabolic abnormalities are commonly observed in patients with Alzheimer's disease.The liver is the primary peripheral organ involved in amyloid-beta metabolism,playing a crucial role in the pathophysiology of Alzheimer's disease.Notably,impaired cholesterol metabolism in the liver may exacerbate the development of Alzheimer's disease.In this review,we explore the underlying causes of Alzheimer's disease and elucidate the role of the liver in amyloid-beta clearance and cholesterol metabolism.Furthermore,we propose that restoring normal cholesterol metabolism in the liver could represent a promising therapeutic strategy for addressing Alzheimer's disease.

文摘Well logging technology has accumulated a large amount of historical data through four generations of technological development,which forms the basis of well logging big data and digital assets.However,the value of these data has not been well stored,managed and mined.With the development of cloud computing technology,it provides a rare development opportunity for logging big data private cloud.The traditional petrophysical evaluation and interpretation model has encountered great challenges in the face of new evaluation objects.The solution research of logging big data distributed storage,processing and learning functions integrated in logging big data private cloud has not been carried out yet.To establish a distributed logging big-data private cloud platform centered on a unifi ed learning model,which achieves the distributed storage and processing of logging big data and facilitates the learning of novel knowledge patterns via the unifi ed logging learning model integrating physical simulation and data models in a large-scale functional space,thus resolving the geo-engineering evaluation problem of geothermal fi elds.Based on the research idea of“logging big data cloud platform-unifi ed logging learning model-large function space-knowledge learning&discovery-application”,the theoretical foundation of unified learning model,cloud platform architecture,data storage and learning algorithm,arithmetic power allocation and platform monitoring,platform stability,data security,etc.have been carried on analysis.The designed logging big data cloud platform realizes parallel distributed storage and processing of data and learning algorithms.The feasibility of constructing a well logging big data cloud platform based on a unifi ed learning model of physics and data is analyzed in terms of the structure,ecology,management and security of the cloud platform.The case study shows that the logging big data cloud platform has obvious technical advantages over traditional logging evaluation methods in terms of knowledge discovery method,data software and results sharing,accuracy,speed and complexity.

文摘A later heading date generally leads to higher grain yield in favorable ecological regions;however,grain yield reaches a limit as the heading date exceeds a certain threshold.Ghd7 is the first cloned major gene that regulates heading date,plant height and grain number.Here,we investigated the relationship between Ghd7 and florigen genes Hd3a and RFT1,to determine their roles in regulating heading date and grain number under different photoperiods.Our results revealed that under long-day(LD)conditions,Hd3a acts prior to RFT1 to promote heading while negatively regulating plant height and grain number.In contrast,Ghd7 positively regulates heading date,plant height,and grain number by inhibiting both Hd3a and RFT1.Under short-day(SD)conditions,the functions of Hd3a and RFT1 remain consistent with those under LD conditions,but Ghd7 does not inhibit their expression,resulting in a weaker phenotypic effect compared to Hd3a.Additionally,under both LD and SD conditions,increased Ghd7 expression enhances its inhibitory effect on Hd3a and RFT1,leading to later heading and increased grain number;however,once the heading date exceeds 94 d,grain number no longer increases.Moreover,the gn1a allele increased grain number by 16.5%to 42.5%,while combinations of the elite alleles from Ghd7,Hd3a,RFT1,and Gn1a significantly increased grain number by up to 240.9%.Therefore,we propose a new breeding strategy to optimize the heading date and grain number using the Ghd7Hd3aRFT1gn1a combination of Ghd7,Hd3a,RFT1,and Gn1a under LD conditions,and the Ghd7hd3aRFT1gn1a combination under SD conditions.This strategy improved the yield of the high-quality Northeast variety Kongyu 131(KY131)by 69.1%in Beijing and 93.7%in Hainan.This strategy will greatly improve the efficiency of north-to-south adaptation in rice,providing theoretical guidance for expanding the geographical adaptability of rice varieties.
文摘The article examines the impact of increased aridization of the territory due to an increase in air temperature,reduced precipitation,and the formation of moisture deficiency on grain yields in Northern Kazakhstan.The most important result of the work is the revealed inverse relationship between grain yields and the temperature of the growing season:low-yielding years are associated with high temperatures and droughts,and high-yielding years are associated with lower temperatures and an optimal ratio of heat and moisture.The novelty of this study is the use of the method of hydrological and climatic calculations in identifying the nature of temperature variability and precipitation in the territory of Northern Kazakhstan for the modern period(1991–2020)compared with the base period(1961–1990).At all the studied meteorological stations,there is a tendency for the average annual temperature and the temperature of the growing season to increase:in the forest-steppe zone with an average warming intensity of 0.3–0.33℃ per decade;in the steppe zone by 0.2–0.43℃ per decade;and in the growing season by 0.2–0.7℃ per decade.The air temperature in the steppe zone is rising more intensively than in the forest-steppe zone,and precipitation in the forest-steppe zone has changed more than in the steppe zone.An increase in the average annual air temperature during the growing season(May–August),combined with a shortage of atmospheric moisture or a constant amount of it,led to an increase in the degree of aridization of the territory,an increase in the frequency of droughts in the steppe zone of Northern Kazakhstan.

文摘Selenium is one of the important trace elements in the human body.Its deficiency will directly affect human health.With people's attention to health,the content of selenium in food has gradually attracted attention.However,detecting selenium compounds in complex samples remains a challenge.In this work,we built an online heating-reaction device.This device combines the electrospray extraction ionization mass spectrometry(EESI-MS)with the heating reaction device,which can simultaneously detect various selenium compounds in complex liquid samples.Under acidic conditions,the sample was heated and catalyzed by a heating reaction device,so that the SeO~(2-)_(3)and O-phenylenediamine(OPD)could generate 1,3-dihydro-2,1,3-benzoselenadiazole.Based on the above reactions,we can detect organic selenium,inorganic selenium and other compounds in liquid samples by organic mass spectrometry.In this experiment,we determined the content of three forms of selenium:selenomethionine(SeMet),l-selenocystine(SeCys(2)),and sodium selenite.The calibration curves for SeMet,SeCys(2),and sodium selenite showed strong linearity within a range of 0.50-50.00μg/L.The limits of detection(LOD)for the three compounds were 0.22,0.27,and 0.41μg/L,respectively.The limits of quantification(LOQ)were 0.68,0.81,and 1.23μg/L,respectively.Spiked recoveries at three levels ranged from 98.8%to 106.1%.In addition,this method can simultaneously detect three selenium compounds and three other specific chemical components in tea infusion samples,providing a rapid and efficient method for identifying tea quality.

文摘The width of rice leaves determines the size of the photosynthetic area.Optimizing rice leaf width can improve the photosynthetic rate,thereby increasing rice yield.In this study,a genome-wide association study(GWAS)was conducted by 225 rice germplasm resources to explore the genetic basis of rice flag leaf width(FLW).We identified nine QTLs associated with FLW(qFLWs),with phenotypic contribution rates ranging from 3.17%to 14.37%.Near-isogenic lines(NILs)were developed for fine-mapping of qFLW11,and the function of FLW11 was further verified.We narrowed down q FLW11 to an 87-kb interval,which contains five genes.
