BACKGROUND Radial head fractures constitute approximately one-third of all elbow fractures,significantly impacting the young and active population.While open reduction and internal fixation is the preferred treatment ...BACKGROUND Radial head fractures constitute approximately one-third of all elbow fractures,significantly impacting the young and active population.While open reduction and internal fixation is the preferred treatment for displaced fractures,its high complication rate in comminuted fractures has led to the increasing use of radial head arthroplasty(RHA).RHA provides improved functional outcomes with fewer complications,yet its long-term efficacy remains a topic of debate.AIM To evaluate the functional outcomes of patients undergoing RHA with a modular metallic prosthesis for comminuted Mason type III and IV radial head fractures.METHODS A prospective and retrospective hospital-based study was conducted at Dayanand Medical College and Hospital,Ludhiana over 32 months(January 2021-August 2023).A total of 26 patients with Mason type III and IV fractures were included,with six retrospective and 20 prospective cases.Functional outcomes were assessed using the Mayo Elbow Performance Score(MEPS),elbow range of motion,pain via Visual Analog Scale,and activities of daily living at immediate postoperative,three-month,and six-month follow-ups.RESULTS MEPS at 6 months follow up for 4 cases(15.38%)had good scores,and 22 cases(84.62%)had excellent scores,with a mean±SD of 97.31±6.67.Comparisons showed significant improvement from immediate post-operative to 3 months(P<0.0001),from immediate post-operative to 6 months(P<0.0001),and between 3 months and 6 months(P<0.0001).None of the patients had elbow instability after radial head replacement and 22 cases(84.62%)had no complications,while 3 cases(11.54%)had a stiff elbow,and 1 case(3.85%)had heterotopic ossification.CONCLUSION RHA is an effective treatment for comminuted radial head fractures,providing stable elbow function with minimal complications.展开更多

Crimean-Congo hemorrhagic fever virus(CCHFV)is a biosafety level-4(BSL-4)pathogen that causes Crimean-Congo hemorrhagic fever(CCHF)characterized by hemorrhagic manifestation,multiple organ failure and high mortality r...Crimean-Congo hemorrhagic fever virus(CCHFV)is a biosafety level-4(BSL-4)pathogen that causes Crimean-Congo hemorrhagic fever(CCHF)characterized by hemorrhagic manifestation,multiple organ failure and high mortality rate,posing great threat to public health.Despite the recently increasing research efforts on CCHFV,host cell responses associated with CCHFV infection remain to be further characterized.Here,to better understand the cellular response to CCHFV infection,we performed a transcriptomic analysis in human kidney HEK293 cells by high-throughput RNA sequencing(RNA-seq)technology.In total,496 differentially expressed genes(DEGs),including 361 up-regulated and 135 down-regulated genes,were identified in CCHFV-infected cells.These regulated genes were mainly involved in host processes including defense response to virus,response to stress,regulation of viral process,immune response,metabolism,stimulus,apoptosis and protein catabolic process.Therein,a significant up-regulation of type III interferon(IFN)signaling pathway as well as endoplasmic reticulum(ER)stress response was especially remarkable.Subsequently,representative DEGs from these processes were well validated by RT-qPCR,confirming the RNA-seq results and the typical regulation of IFN responses and ER stress by CCHFV.Furthermore,we demonstrate that not only type I but also type III IFNs(even at low dosages)have substantial anti-CCHFV activities.Collectively,the data may provide new and comprehensive insights into the virus-host interactions and particularly highlights the potential role of type III IFNs in restricting CCHFV,which may help inform further mechanistic delineation of the viral infection and development of anti-CCHFV strategies.展开更多
In order to enrich the research about type III secretion system (T3SS) injectisome of Vibrio alginolyticus, vscX gene was cloned from E algianlyticus strain HY9901 for bioinformatics analysis and expression analysis...In order to enrich the research about type III secretion system (T3SS) injectisome of Vibrio alginolyticus, vscX gene was cloned from E algianlyticus strain HY9901 for bioinformatics analysis and expression analysis. Specific primers were designed according to the full-length geanme sequence of V. alginolyticus in GenBank. vscX gene (C, enBank accession number: FR780679) contained a 378 bp open reading frame (ORF), encoding a putative protein of 125 amino acids. The theoretical molecular weight was 14.209 2 kD and theoretical pI was 5.75. By using Signal 4.1 Server and TMHMM Server 2.0, it was predicted that VscX protein had no transmembrane domain or signal peptide. The results of prediction using SoftBerry-Psite software showed that VscX protein contained three casein ki- nase lI phosphorylation sites, one N-myristoylation site and three C-terminal targeting signal sites. Subcellular localization revealed that VscX might be located in cytoplasm with the possibility of 56.5%. According to SMART prediction, VscX had one Pfam ( 1 - 125 aa) domain. Phylogenetic analysis revealed that VscX from V. alginolyticus and VscX from E parahemolyticus were clustered into the same group. Network interaction analysis showed that vscX was adjacent to vseY, vopB and sycN. By real-time fluorescent quantitative PCR technique and 2-△△△ method, the differences in expression levels of VscX mRNA in V. alginolyticus strain HY9901, T3SS deletion strain AvscO and complementary strain C-vscO at different growth stages were analyzed. The results showed that the expression levels of VscX mRNA in V. algianlyticus strain HY9901 and C-vscO were significantly up-regulated at stable growth stage ( P 〈0.01 ) ; the expression levels of VscX mRNA in deletion strain △vscO were significantly up-regulated at late growth stage ( P 〈0.01 ). Interfacial superconductivity(IS)has been a topic of intense interest in condensed matter physics,due to its unique properties and exotic photoelectrical performance.However,there are few reports about IS systems cons...Interfacial superconductivity(IS)has been a topic of intense interest in condensed matter physics,due to its unique properties and exotic photoelectrical performance.However,there are few reports about IS systems consisting of two insulators.Here,motivated by the emergence of an insulator-metal transition in type-Ⅲ heterostructures and the superconductivity in some“special”two-dimensional(2D)semiconductors via electron doping,we predict that the 2D heterostructure SnSe_(2)/PtTe_(2) is a model system for realizing IS by using firstprinciples calculations.Our results show that due to slight but crucial interlayer charge transfer,SnSe_(2)/PtTe_(2) turns to be a type-Ⅲ heterostructure with metallic properties and shows a superconducting transition with the critical temperature(T_(c))of 3.73 K.Similar to the enhanced electron–phonon coupling(EPC)in the electrondoped SnSe_(2) monolayer,the IS in the SnSe_(2)/PtTe_(2) heterostructure mainly originates from the metallized SnSe_(2) layer.Furthermore,we find that its superconductivity is sensitive to tensile lattice strain,forming a domeshaped superconducting phase diagram.Remarkably,at 7%biaxial tensile strain,the superconducting T_(c) can increase more than twofold(8.80 K),resulting from softened acoustic phonons at the𝑀point and enhanced EPC strength.Our study provides a concrete example for realizing IS in type-Ⅲ heterostructures,which waits for future experimental verification.展开更多
Collagen made a tremendous impact in the field of regenerative medicine as a bioactive material.For decades,collagen has been used not only as a scaffolding material but also as an active component in regulating cells...Collagen made a tremendous impact in the field of regenerative medicine as a bioactive material.For decades,collagen has been used not only as a scaffolding material but also as an active component in regulating cells’biological behavior and phenotype.However,animal-derived collagen as a major source suffered from problems of immunogenicity,risk of viral infection,and the unclear relationship between bioactive sequence and function.Recombinant humanized collagen(rhCol)provided alternatives for regenerative medicine with more controllable risks.However,the characterization of rhCol and the interaction between rhCol and cells still need further investigation,including cell behavior and phenotype.The current study preliminarily demonstrated that recombinant humanized collagen type III(rhCol III)conformed to the theoretical amino acid sequence and had an advanced structure resembling bovine collagen.Furthermore,rhCol III could facilitate basal biological behaviors of human skin fibroblasts,such as adhesion,proliferation and migration.rhCol III was beneficial for some extracellular matrix-expressing cell phenotypes.The study would shed light on the mechanism research of rhCol and cell interactions and further understanding of effectiveness in tissue regeneration.展开更多
Type III antifreeze proteins(AFPIIIs)are a group of small globular proteins found in some polar fishes to protect them against freezing damage.Transgenic expression of AFPs has been shown to confer cold tolerance to c...Type III antifreeze proteins(AFPIIIs)are a group of small globular proteins found in some polar fishes to protect them against freezing damage.Transgenic expression of AFPs has been shown to confer cold tolerance to commercially important plants and animals.We have previously isolated multiple AFPIII genes in the Antarctic eelpout(Lycodichthys dearborni)that encode larger AFPIII isoforms with up to 12 of the conventional domains.Here we have introduced the fish AFPIII genes that encode for the monomer(ld1),dimer(ld2),trimer(ld3)and tetramer(ld4)AFPIII isoforms in tobacco plants.Pot-grown 4-week-old transgenic tobacco plants were exposed to cold stress at 4◦C for 30 days and the results show that ld1,ld2,ld3 and ld4 transgenic plants present relatively lower electrolyte leakage and lower content of malondialdehyde(MDA),but accumulated higher content of proline when compared to control plants.This indicates considerable improved membrane integrity under low temperature stress and improvement of the plant cold resistance.The plants transformed with the AFPIII tetramer-and trimer-domains demonstrated a higher cold-tolerant levels when compared with plants transformed with the dimer-and monomer AFPIII domains.Our study further supports that fish AFPIIIs,especially the multidomain proteins,protect cells from non-freezing hypothermic stresses,apart from there well-known function as ice inhibitors molecules at freezing temperature.展开更多
In this paper,we propose and analyze a delayed predator-prey model with Holling type III functional response taking into account cooperation behavior in predators.The time delay is introduced in the attack rate to rep...In this paper,we propose and analyze a delayed predator-prey model with Holling type III functional response taking into account cooperation behavior in predators.The time delay is introduced in the attack rate to represent the time necessary to trigger the attack.Each analytical result is followed by an ecological interpretation.We investigate the effect of hunting cooperation on both the number and the level of positive steady states.We observe that the level of the positive equilibrium decreases when increasing the hunting cooperation parameter.Then,we study the impact of the delay as well as the cooperation in hunting on the dynamics of the system.We prove that the presence of delay in the attack rate induces stability switches around the coexisting equilibrium when predators cooperate.In addition,we consider the discrete delay as a bifurcation parameter and prove that the model undergoes a Hopf-bifurcation at the coexisting equilibrium when the delay crosses some critical values.Numerical simulations are presented to confirm our analytical findings.展开更多
BACKGROUND Treatment response to direct-acting antivirals(DAAs)is a challenging issue and the identification of non-responders patients is very important.AIM To evaluate the relation between baseline serum levels of h...BACKGROUND Treatment response to direct-acting antivirals(DAAs)is a challenging issue and the identification of non-responders patients is very important.AIM To evaluate the relation between baseline serum levels of hyaluronic acid(HA)and type III procollagen N-peptide(PIIINP)with direct-acting antivirals treatment failure in Egyptian patients with chronic hepatitis C.METHODS Hepatitis C patients(responders and non-responders to sofosbuvir/daclatasvir)were tested for HA and PIIINP using sensitive chemiluminescent immunoassay.RESULTS There were distinctly higher PIIINP(P=0.0003)and HA(P<0.0001)levels in non-responders than responders patients with a good ability for distinguishing non-responders from patients with sustained virological response(area under the curve=0.766 for HA and 0.684 for PIIINP).Logistic regression analysis revealed that the HA×PIIINP is the model with the highest predictive ability(area under the curve=0.809).Diagnostic performances were superior to each marker alone with good sensitivity(74.7%),specificity(74%),positive predictive(68.3%),negative predictive values(79.6%)and accuracy(74.3%).The multiplication of HA×PIIINP is correlated significantly(P<0.05)with elevated liver enzymes(r=0.212),decreased albumin(r=-0.26),elevated aspartate aminotransferase-platelet ratio index(r=0.223)and elevated fibrosis-4 score(r=0.216)scores.CONCLUSION These findings suggested the remarkable role of fibrogensis markers HA and PIIINP in the prediction of hepatitis C virus DAAs treatment response.Multiplying HA with PIIINP values increase the sensitivity to detect treatment success and thus may aim to improve treatment duration and the disease control.展开更多
BACKGROUND A patient with type Ⅲ Kummell’s disease had a ruptured posterior cortex of the fractured vertebral body, which caused spinal cord compression. An open surgery was considered the best choice of operation. ...BACKGROUND A patient with type Ⅲ Kummell’s disease had a ruptured posterior cortex of the fractured vertebral body, which caused spinal cord compression. An open surgery was considered the best choice of operation. However, the patient and her family refused open surgery and instead demanded a minimally invasive surgical treatment such as percutaneous vertebroplasty(PVP). After preoperative discussion, we finally adopted the novel therapy of traditional Chinese medicine manipulative reduction(TCMMR) combined with PVP.CASE SUMMARY A patient with type Ⅲ Kummell’s disease exhibiting bone block-induced spinal cord compression was admitted to our hospital. She suffered from a variety of medical disorders but refused open surgery, and instead asked for PVP surgery. TCMMR, in parallel with PVP, was used to restore the height of the compressed vertebral body and reduce the symptoms of spinal cord compression by the bone block in order to strengthen the vertebral body and prevent further collapse. The surgery was very successful. The height of the compressed vertebra was restored, and the symptom of spinal cord compression by bone block was reduced successfully via TCMMR. The fractured vertebra was solidified by the PVP. The pain visual analog score declined from preoperative 7 scores to postoperative 2 scores, and the Frankel spinal cord scale increased from preoperative D degree to postoperative E degree.CONCLUSION The new method has advantages in treating patients with type Ⅲ Kummell’s disease who cannot be treated with open surgery.展开更多
BACKGROUND Gustilo III fractures have a high incidence and are difficult to treat.Patients often experience difficulty in wound healing.Negative pressure drainage technology can help shorten wound healing time and has...BACKGROUND Gustilo III fractures have a high incidence and are difficult to treat.Patients often experience difficulty in wound healing.Negative pressure drainage technology can help shorten wound healing time and has positive value in improving patient prognosis.AIM To explore the clinical value of the negative pressure sealing drainage technique in wound healing of Gustilo IIIB and IIIC open fractures.METHODS Eighty patients with Gustilo IIIB and IIIC open fractures with skin and soft tissue injuries who were treated in the Second People’s Hospital of Dalian from March 2019 to December 2021 were selected as the research subjects.They were divided into a study group(n=40,healed with negative pressure closed drainage)and a control group(n=40,healed with conventional dressing changes)according to the variation in the healing they received.The efficacy of the clinical interventions,the variations in the regression indicators(time to wound healing,time to fracture healing,time to hospitalization),and the conversion and healing of bacterial wounds were compared 1-3 mo after the intervention.RESULTS The total effective rate of patients among the study group was 95.00%(38/40),which was notably higher than 75.00%(30/40)among the control group(P<0.05).The wound healing time,fracture healing time,and hospital stay of the patients in the study group was shorter than the control group(P<0.05).After the intervention,the negative bacterial culture at the wound site rate and wound healing rate of the patients among the study group increased compared to the control group(P<0.05).CONCLUSION Negative pressure sealing and drainage technology has a good therapeutic effect on patients with Gustilo IIIB and IIIC open fracture wounds with skin and soft tissue injury.It can notably enhance the wound healing rate and the negative rate of bacteria on the wound surface and help to speed up the recovery process of patients.展开更多

文摘Collagen made a tremendous impact in the field of regenerative medicine as a bioactive material.For decades,collagen has been used not only as a scaffolding material but also as an active component in regulating cells’biological behavior and phenotype.However,animal-derived collagen as a major source suffered from problems of immunogenicity,risk of viral infection,and the unclear relationship between bioactive sequence and function.Recombinant humanized collagen(rhCol)provided alternatives for regenerative medicine with more controllable risks.However,the characterization of rhCol and the interaction between rhCol and cells still need further investigation,including cell behavior and phenotype.The current study preliminarily demonstrated that recombinant humanized collagen type III(rhCol III)conformed to the theoretical amino acid sequence and had an advanced structure resembling bovine collagen.Furthermore,rhCol III could facilitate basal biological behaviors of human skin fibroblasts,such as adhesion,proliferation and migration.rhCol III was beneficial for some extracellular matrix-expressing cell phenotypes.The study would shed light on the mechanism research of rhCol and cell interactions and further understanding of effectiveness in tissue regeneration.

文摘Type III antifreeze proteins(AFPIIIs)are a group of small globular proteins found in some polar fishes to protect them against freezing damage.Transgenic expression of AFPs has been shown to confer cold tolerance to commercially important plants and animals.We have previously isolated multiple AFPIII genes in the Antarctic eelpout(Lycodichthys dearborni)that encode larger AFPIII isoforms with up to 12 of the conventional domains.Here we have introduced the fish AFPIII genes that encode for the monomer(ld1),dimer(ld2),trimer(ld3)and tetramer(ld4)AFPIII isoforms in tobacco plants.Pot-grown 4-week-old transgenic tobacco plants were exposed to cold stress at 4◦C for 30 days and the results show that ld1,ld2,ld3 and ld4 transgenic plants present relatively lower electrolyte leakage and lower content of malondialdehyde(MDA),but accumulated higher content of proline when compared to control plants.This indicates considerable improved membrane integrity under low temperature stress and improvement of the plant cold resistance.The plants transformed with the AFPIII tetramer-and trimer-domains demonstrated a higher cold-tolerant levels when compared with plants transformed with the dimer-and monomer AFPIII domains.Our study further supports that fish AFPIIIs,especially the multidomain proteins,protect cells from non-freezing hypothermic stresses,apart from there well-known function as ice inhibitors molecules at freezing temperature.
文摘In this paper,we propose and analyze a delayed predator-prey model with Holling type III functional response taking into account cooperation behavior in predators.The time delay is introduced in the attack rate to represent the time necessary to trigger the attack.Each analytical result is followed by an ecological interpretation.We investigate the effect of hunting cooperation on both the number and the level of positive steady states.We observe that the level of the positive equilibrium decreases when increasing the hunting cooperation parameter.Then,we study the impact of the delay as well as the cooperation in hunting on the dynamics of the system.We prove that the presence of delay in the attack rate induces stability switches around the coexisting equilibrium when predators cooperate.In addition,we consider the discrete delay as a bifurcation parameter and prove that the model undergoes a Hopf-bifurcation at the coexisting equilibrium when the delay crosses some critical values.Numerical simulations are presented to confirm our analytical findings.
文摘BACKGROUND Treatment response to direct-acting antivirals(DAAs)is a challenging issue and the identification of non-responders patients is very important.AIM To evaluate the relation between baseline serum levels of hyaluronic acid(HA)and type III procollagen N-peptide(PIIINP)with direct-acting antivirals treatment failure in Egyptian patients with chronic hepatitis C.METHODS Hepatitis C patients(responders and non-responders to sofosbuvir/daclatasvir)were tested for HA and PIIINP using sensitive chemiluminescent immunoassay.RESULTS There were distinctly higher PIIINP(P=0.0003)and HA(P<0.0001)levels in non-responders than responders patients with a good ability for distinguishing non-responders from patients with sustained virological response(area under the curve=0.766 for HA and 0.684 for PIIINP).Logistic regression analysis revealed that the HA×PIIINP is the model with the highest predictive ability(area under the curve=0.809).Diagnostic performances were superior to each marker alone with good sensitivity(74.7%),specificity(74%),positive predictive(68.3%),negative predictive values(79.6%)and accuracy(74.3%).The multiplication of HA×PIIINP is correlated significantly(P<0.05)with elevated liver enzymes(r=0.212),decreased albumin(r=-0.26),elevated aspartate aminotransferase-platelet ratio index(r=0.223)and elevated fibrosis-4 score(r=0.216)scores.CONCLUSION These findings suggested the remarkable role of fibrogensis markers HA and PIIINP in the prediction of hepatitis C virus DAAs treatment response.Multiplying HA with PIIINP values increase the sensitivity to detect treatment success and thus may aim to improve treatment duration and the disease control.
文摘BACKGROUND A patient with type Ⅲ Kummell’s disease had a ruptured posterior cortex of the fractured vertebral body, which caused spinal cord compression. An open surgery was considered the best choice of operation. However, the patient and her family refused open surgery and instead demanded a minimally invasive surgical treatment such as percutaneous vertebroplasty(PVP). After preoperative discussion, we finally adopted the novel therapy of traditional Chinese medicine manipulative reduction(TCMMR) combined with PVP.CASE SUMMARY A patient with type Ⅲ Kummell’s disease exhibiting bone block-induced spinal cord compression was admitted to our hospital. She suffered from a variety of medical disorders but refused open surgery, and instead asked for PVP surgery. TCMMR, in parallel with PVP, was used to restore the height of the compressed vertebral body and reduce the symptoms of spinal cord compression by the bone block in order to strengthen the vertebral body and prevent further collapse. The surgery was very successful. The height of the compressed vertebra was restored, and the symptom of spinal cord compression by bone block was reduced successfully via TCMMR. The fractured vertebra was solidified by the PVP. The pain visual analog score declined from preoperative 7 scores to postoperative 2 scores, and the Frankel spinal cord scale increased from preoperative D degree to postoperative E degree.CONCLUSION The new method has advantages in treating patients with type Ⅲ Kummell’s disease who cannot be treated with open surgery.

文摘BACKGROUND Gustilo III fractures have a high incidence and are difficult to treat.Patients often experience difficulty in wound healing.Negative pressure drainage technology can help shorten wound healing time and has positive value in improving patient prognosis.AIM To explore the clinical value of the negative pressure sealing drainage technique in wound healing of Gustilo IIIB and IIIC open fractures.METHODS Eighty patients with Gustilo IIIB and IIIC open fractures with skin and soft tissue injuries who were treated in the Second People’s Hospital of Dalian from March 2019 to December 2021 were selected as the research subjects.They were divided into a study group(n=40,healed with negative pressure closed drainage)and a control group(n=40,healed with conventional dressing changes)according to the variation in the healing they received.The efficacy of the clinical interventions,the variations in the regression indicators(time to wound healing,time to fracture healing,time to hospitalization),and the conversion and healing of bacterial wounds were compared 1-3 mo after the intervention.RESULTS The total effective rate of patients among the study group was 95.00%(38/40),which was notably higher than 75.00%(30/40)among the control group(P<0.05).The wound healing time,fracture healing time,and hospital stay of the patients in the study group was shorter than the control group(P<0.05).After the intervention,the negative bacterial culture at the wound site rate and wound healing rate of the patients among the study group increased compared to the control group(P<0.05).CONCLUSION Negative pressure sealing and drainage technology has a good therapeutic effect on patients with Gustilo IIIB and IIIC open fracture wounds with skin and soft tissue injury.It can notably enhance the wound healing rate and the negative rate of bacteria on the wound surface and help to speed up the recovery process of patients.
