The cotton genomics community has utilized whole genome sequencing data to construct an elite gene pool in which functional genes are related to agronomic traits. However, the functional validation of these genes is hindered by time-consuming and inefficient genetic transformation methods. Thus, establishing a transient transformation system of high efficiency is necessary for cotton genomics. Results To improve the efficiency of transient transformation, we used the protoplasts isolated from the etiolated cotyledon as recipient. The enzymatic digestion buffer comprised 1.5%(w/v) cellulase, 0.75%(w/v) macerozyme, and 1% hemicellulase, osmotically buffered with 0.4 mol·L^(-1) mannitol. After 5 h of dark incubation at 25℃, uniform cotton protoplasts were successfully isolated with a yield of 4.6 × 10^(6) protoplasts per gram(fresh weight) and 95% viability. We incubated 100 μL protoplasts(2.5 × 10^(5)·m L^(-1)) with 15 μg plasmid in the solution of 0.4 mol·L^(-1) mannitol and 40% PEG 4000 for 15 min, ultimately achieving an optimal transient transfection efficiency of 71.47%.Conclusions This transient system demonstrated effective utility in cellular biology research through successful applications in subcellular localization analyses, bimolecular fluorescence complementation(Bi FC) verification, and prime editing vector validation. Through systematic optimization, we established an efficient and expedited protoplast-based transient transformation system and successfully applied this platform to cotton functional genomics studies.展开更多

文摘Litchi has great economic significance as a global fruit crop. However, the advancement of litchi functional genomics has encountered substantial obstacles due to its recalcitrance to stable transformation. Here,we present an efficacious Agrobacterium tumefaciens-mediated transformation system in somatic embryos of‘Heiye'litchi. This system was developed through the optimization of key variables encompassing explant selection,A.tumefaciens strain delineation, bacterium concentration, infection duration, and infection methodology. The subsequent validation of the transformation technique in litchi was realized through the ectopic expression of LcMYB1,resulting in the generation of transgenic calli. However, the differentiation of transgenic calli into somatic embryos encountered substantial challenges. To delineate the intricate molecular underpinnings of LcMYB1's inhibitory role in somatic embryo induction,a comprehensive transcriptome analysis was conducted that encompassed embryogenic calli(C),globular embryos(G),and transgenic calli(TC).A total of 1,166 common differentially expressed genes(DEGs)were identified between C-vs.-G and C-vs.-TC.Gene Ontology(GO)annotation and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis revealed that these common DEGs were mostly related to plant hormone signal transduction pathways.Furthermore,RT-qPCR corroborated the pronounced down-regulation of numerous genes that are associated with somatic embryo induction within the transgenic calli. The development of this transformation system provides valuable support for functional genomics research in litchi.

文摘Background Cotton is an important crop providing the most natural fibers all over the world. Through systematic optimization, we established an efficient and expedited protoplast-based transient transformation system and successfully applied this platform to cotton functional genomics studies.

文摘This study aims to understand the behavior of the Bugis community in developing vernacular house models in the coastal areas of South Sulawesi Province, along with the factors influencing this behavior, such as knowledge, motivation, attitude, locus of control, commitment, concern, and income level.Bugis vernacular houses reflect local wisdom that is adapted to environmental,socio-cultural conditions, and the challenges of modernization. This correlational research was conducted in the regencies of Pangkajene and Kepulauan,Barru, and Bone, with a sample of 300 household heads. The variables analyzed include knowledge, motivation, attitude, locus of control, commitment, concern, and income level.Data were collected through knowledge tests and questionnaires and were analyzed using simple and multiple regression tech-niques.The results show that the Bugis community’s behavior in developing vernacular houses is moderate.Individually, motivation, locus of control, commitment, and concern significantly influence behavior, while knowledge, attitude, and income do not have a significant impact. However, collectively, all independent variables(knowledge, motivation, attitude, locus of control, commitment, concern, and income)affect the behavior of the Bugis community in developing vernacular houses in coastal areas. This study provides valuable insights into the environmental and social adaptations of the Bugis coastal community,as well as efforts to preserve culture through the sustainable development of vernacular houses.
文摘BACKGROUND Helicobacter pylori(H.pylori)eradication treatment for primary gastric mucosaassociated lymphoid tissue(MALT)lymphoma has already been established.However,t(11;18)(q21;q21)/API2-MALT1 translocation-positive lesions are a type of primary gastric MALT lymphoma in which a response to eradication treatment is difficult to achieve. In addition, trisomy 18 may be associated with diffuse large B-cell lymphoma(DLBCL)transformation of gastric MALT lymphoma.CASE SUMMARY A 66-year-old man was diagnosed with MALT lymphoma in the ascending colon by colonoscopy and biopsy. Two years later, esophagogastroduodenoscopy revealed chronic atrophic gastritis that was positive for H.pylori, and eradication treatment was administered. Two years and nine months later(at the age of 70),a new ulcerative lesion suggestive of MALT lymphoma appeared in the gastric body, and six months later,a similar lesion was also found in the fundus. One year later(4 years and 3 months after H.pylori eradication),at the age of 72,the lesion in the gastric body had become deeper and had propagated.A biopsy revealed a pathological diagnosis of DLBCL.Both MALT lymphoma lesions in the ascending colon and DLBCL lesions in the stomach were positive for the t(11;18)(q21;q21)/API2-MALT1 translocation, and trisomy 18q21 was also detected. After 6 courses of R-CHOP(rituximab,cyclophosphamide,doxorubicin,vincristine and prednisone)chemotherapy, all of the above lesions disappeared[complete remission(CR)],and CR has been maintained for more than 3 years. In addition, both the colonic and gastric lesions were proven to have the same clonality.CONCLUSION Because the patient had a MALT1 translocation with trisomy 18q21,it was thought that this gastric MALT lymphoma developed independently of H.pylori infection and progressed.

文摘The exploration of the coupling and coordination between urban innovation capability(IC)and industrial transformation(IF)serves as a novel perspective for interpreting the increasingly severe phenomenon of population shrinkage during the industrialization process. This study investigated the evolutionary characteristics of IC and IF in shrinking and growing cities in Northeast China from 2010 to 2020.It uses entropy weighted model, coupling coordination degree, the Dagum Gini coefficient, and geographic detectors to analyze the coordinated development of IC and IF in the context of population shrinkage. The study analyzed the spatiotemporal patterns and driving mechanisms for their coordinated development. The results show that:1)both urban IC and IF exhibited an overall positive trend during the study period.Shrinking cities depend more on IF to address the challenges of population shrinkage, while growing cities mainly rely on innovation-driven development.2)The coupling gap between IC and IF in shrinking and growing cities has widened over time, with the coordination level of shrinking cities steadily decreasing.Cities with serious disorder are concentrated in northern Heilongjiang Province, while most cities in Jilin Province experience moderate disorder.Liaoning Province, however, shows generally good coupling coordination.3)Human capital is the key factor driving coupling coordination in both types of cities.Shrinking cities rely on economic and financial development, with the‘repair-type’logic that emphasizes short-term economic growth and resource compensation. In contrast, the coupling of growing cities relies on basic support capabilities, with the‘optimization-type’logic focused on enhancing endogenous resilience and systemic coordination.Exploring the coordination between urban innovation capabilities and industrial transformation can provide a new perspective for research on population shrinkage, which holds certain theoretical and practical significance for implementing the new round of revitalization strategy in Northeast China.

文摘Objectives:Electronic health records(EHRs)offer valuable real-world data(RWD)for Chinese medicine research. However, significant methodological challenges remain in developing integrative Chinese-Western medicine(ICWM)databases. This study aims to establish a best-practice methodological framework, referred to as BRIDGE,to guide the construction of ICWM databases using EHRs.Methods:We developed the methodological framework through a comprehensive process, including systematic literature review, synthesis of empirical experiences, thematic expert discussions, and consultation with an external panel to reach consensus. Results:The BRIDGE framework outlines 6 core components for ICWM-EHR database development:Overall design, database architecture, data extraction and linkage, data governance, data verification, and data quality evaluation.Key data elements include variables related to study population, treatment or exposure, outcomes, and confounders. These databases support various research applications, particularly in evaluating the effectiveness and safety of integrative therapies. To demonstrate its practical value,we developed an ICWM-EHR database on women’s reproductive lifespan, encompassing 2,064,482 patients. This database captures women’s health conditions across the life course, from reproductive age to older adulthood. Conclusions:The BRIDGE methodological framework provides a standardized approach to building high-quality ICWM-EHR databases. It offers a unique opportunity to strengthen the methodological rigor and real-world relevance of Chinese medicine research in integrated healthcare settings.
文摘With the first rays of dawn peeking through the rainforest at Shiti Village,Dehong Dai and Jingpo Autonomous Prefecture,Yunnan Province,Xu Xiaolong,a bird-watching guide from the Jingpo ethnic group, led a German bird-watching group into the jungle. His great-grandfather made a living by hunting“weird-looking birds”in the mountains.Today, the birds(Great Hornbill)are attracting tourist dollars from other parts of the world without dodging bullets. In Shiti Village,bird-watchers can enjoy the chirping of Great Hornbills and feel the rumbling in the air as the birds fly past. The village on the Chinese border with Myanmar was formerly poverty-stricken,with an average annual income of barely 2,000 yuan(US$305)per household 10 years ago.Today, the average annual income has grown to 150,000 yuan(US$23,075).It is a shining example of the successful practice of the notion that“lucid waters and lush mountains are invaluable assets.”
