Background:The study aimed to investigate the protective effect and mechanism of total flavonoids of Scutellaria baicalensis(TFSB)on acute myocardial ischemia(AMI)rats by using functional metabonomics.Methods:Rats wer...Background:The study aimed to investigate the protective effect and mechanism of total flavonoids of Scutellaria baicalensis(TFSB)on acute myocardial ischemia(AMI)rats by using functional metabonomics.Methods:Rats were divided into the Control,Model,AMI positive control(Propranolol hydrochloride,30 mg/kg),low dose TFSB(50 mg/kg),and high dose TFSB(100 mg/kg)groups.Rats received the corresponding treatment by intragastric administration once daily for 10 consecutive days.Electrocardiogram,myocardial enzyme,triphenyltetrazolium chloride staining,hematoxylin-eosin,and enzyme-linked immunosorbent assay were performed to evaluate the protective effect of TFSB on AMI rats.Then,the UHPLC-Q-Orbitrap MS method based on serum metabolomics was utilised to search for metabolic biomarkers and metabolic pathways.Subsequently,Western blot and RT-PCR techniques were employed to identify the respective genes and proteins.Results:Pharmacodynamics revealed that TFSB could ameliorate AMI in rats.The results of the metabolomics analysis indicated that the alterations in metabolic profile observed in rats with AMI were partially improved by treatment with TFSB.Moreover,the mRNA expression levels of 5-lipoxygenase(5-LOX)and 15-lipoxygenase(15-LOX)and the protein expression levels of 5-LOX,15-LOX,interleukin-1β(IL-1β),and NF-κB p65 were reduced following treatment with TFSB.Conclusion:The potential treatment of TFSB in AMI may be ascribed to its ability to regulate arachidonic acid metabolism.展开更多

Methods The static and dynamic adsorption/desorption methods were used, and the separation and purification process was evaluated by measuring the concentration of total flavonoid in the fractions with UV spectrophotometer. Results The SP70 macroporous resin was the most effective compared with other macroporous resins. The optimum conditions were screened, which were 0.5 g· mL^- 1 corresponding to crude drug for concentration of extract, pH 5 - 6, and appended 60 times the volume of the resin bed (BV) with the adsorption speed 2 BV·h^-1, and the volume of aq. 70% (V/V) ethanol as eluant was 5 BV with desorption speed 2 BV·h^-1. By this method, the final contents of total flavonoids exceeded 80%. Conclusion The SP70 macroporous resin is the most effective one for large-scale isolation and purification of flavonoids from Radix Pueraria, which meets industrial needs.展开更多

TFE (300 mg/kg) was administered once daily to male Sprague-Dowley rats by gavage for fifteen days. The total cytochrome P450 content and its main isoforms CYP1A2, CYP3A4 and CYP2E1 activities in rat liver microsomes were detected. The activity of CYP1A2 was measured by fluorometry and the activities of CYP3A4 and CYP2E1 were determined by measuring the amount of methanal and p-aminophenol formed using UV/Vis spectrophotometer, respectively. Administration of TFE significantly increased the total CYP450 content and activities ofCYP1A2, CYP3A4 and CYP2E1 in rat liver microsomes, compared with the control group. Particularly, the activities of CYP1A2 and CYP2E1 were enhanced significantly (P〈0.01). TFE induced the increase in total CYP450 content and its main isoforms CYP1A2, CYP3A4 and CYP2E1 activities in rat liver microsomes.展开更多
Suaeda salsa an annual herb of Chenopodiaceae Suaeda well accepted as both a folk medicinal and food plant.In the current study,the extraction of total flavonoids from S.salsa and the in vitro antioxidant,hypoglycemic...Suaeda salsa an annual herb of Chenopodiaceae Suaeda well accepted as both a folk medicinal and food plant.In the current study,the extraction of total flavonoids from S.salsa and the in vitro antioxidant,hypoglycemic and hypolipidemic activities of the extracts were investigated.The total flavonoids was extracted by refluxing with ethanol and the selection and optimization of main experimental parameters were carried out using single factor experiments combined with central composite design-response surface methodology(CCD-RSM).The total flavonoids extracted from S.salsa were tested for hydroxyl and oxygen radical scavenging activity,and inhibitory activities againstα-amylase and lipase in vitro.Results revealed that the optimal conditions for the extraction of total flavonoids were as following:liquid-tosolid ratio,31.40:1;ethanol concentration,83.12%;reflux temperature,90℃,and reflux time,128.40 min.Under these optimized conditions,the predicted and actual values of total flavonoids yield were 0.896%and 0.932%,respectively.The in vitro antioxidant tests revealed that the total flavonoids could scavenge hydroxyl and oxygen radicals and showed inhibitory activity againstα-amylase and lipase.The optimal extraction conditions obtained in this experiment are simple to perform and have good reproducibility.The results have demonstrated that the total flavonoids from S.salsa can be used as a potential source of natural antioxidant,hypoglycemic and hypolipidemic ingredients,which provides new pharmacological basis for the development of S.salsa.展开更多

However, the mechanism of action of SST...Previous experimental studies have shown that cerebral infarction can be effectively reduced following treatment with scutellaria baicalensis stem-leaf total flavonoid (SSTF). However, the mechanism of action of SSTF as a preventive drug to treat cerebral infarction remains unclear. In this study, Sprague-Dawley rats were pretreated with 50, 100, 200 mg/kg SSTF via intragastric ad- ministration for 1 week prior to the establishment of focal cerebral ischemia/reperfusion injury. The results showed that pretreatment with SSTF effectively improved neurological function, reduced brain water content and the permeability of blood vessels, ameliorated ischemia-induced morphology changes in hippocampal microvessels, down-regulated Fas and FasL protein expression, elevated the activity of superoxide dismutase and glutathione peroxidase, and decreased malondialdehyde content. In contrast to low-dose SSTF pretreatment, the above changes were most obvious after pretreatment with moderateand high-doses of SSTF. Experimental findings indicate that SSTF pretreatment can exert protective effects on the brain against cerebral ischemia/reperfusion injury. The underlying mechanisms may involve reducing brain water content, increasing microvascular recanalization, inhibiting the apoptosis of hippocampal neurons, and attenuating free radical damage.展开更多

This study aimed to explore the effects of Scutellaria baicalensis stem-leaf total flavonoid on amyloid beta-peptide-induced neuronal apoptosis and the expression of apoptosis-related proteins in the rat hippocampus. Male Wistar rats were given intragastric administration of Scutellaria baicalensis stem-leaf total flavonoid, 50 or 100 mg/kg, once per day. On day 8 after administration, 10 pg amyloid beta-peptide (25-35) was injected into the bilateral hippocampus of rats to induce neuronal apoptosis. On day 20, hippocampal tissue was harvested and probed with the terminal deoxyribonucleotidyl transferase-mediated biotin-16-dUTP nick-end labeling assay. Scutellaria baicalensis stem-leaf total flavonoid at 50 and 100 mg/kg reduced neuronal apoptosis induced by amyloid beta-peptide (25-35) in the rat hippocampus. Immunohistochemistry and western blot assay revealed that expression of the pro-apoptotic protein Bax, cytochrome c and caspase-3 was significantly diminished by 50 and 100 mg/kg Scutellaria baicalensis stem-leaf total flavonoid, while expression of the anti-apoptotic protein Bcl-2 was increased. Moreover, 100 mg/kg Scutellana baicalensis stem-leaf total flavonoid had a more dramatic effect than the lower dosage. These experimental findings indicate that Scutellaria baicalensis stem-leaf total flavonoid dose-dependently attenuates neuronal apoptosis induced by amyloid beta-peptide in the hippocampus, and it might mediate this by regulating the expression of Bax, cytochrome c, caspase-3 and Bcl-2.展开更多
Flavonoids have been reported to have therapeutic potential for spinal cord injury.Hawthorn leaves have abundant content and species of total flavonoids,and studies of the effects of the total flavonoids of hawthorn l...Flavonoids have been reported to have therapeutic potential for spinal cord injury.Hawthorn leaves have abundant content and species of total flavonoids,and studies of the effects of the total flavonoids of hawthorn leaves on spinal cord injury have not been published in or outside China.Therefore,Sprague-Dawley rats were used to establish a spinal cord injury model by Allen's method.Rats were intraperitoneally injected with 0.2 m L of different concentrations of total flavonoids of hawthorn leaves(5,10,and 20 mg/kg)after spinal cord injury.Injections were administered once every 6 hours,three times a day,for 14 days.After treatment with various concentrations of total flavonoids of hawthorn leaves,the Basso,Beattie,and Bresnahan scores and histological staining indicated decreases in the lesion cavity and number of apoptotic cells of the injured spinal cord tissue;the morphological arrangement of the myelin sheath and nerve cells tended to be regular;and the Nissl bodies in neurons increased.The Basso,Beattie,and Bresnahan scores of treated spinal cord injury rats were increased.Western blot assays showed that the expression levels of pro-apoptotic Bax and cleaved caspase-3 were decreased,but the expression level of the anti-apoptotic Bcl-2 protein was increased.The improvement of the above physiological indicators showed a dose-dependent relationship with the concentration of total flavonoids of hawthorn leaves.The above findings confirm that total flavonoids of hawthorn leaves can reduce apoptosis and exert neuroprotective effects to promote the recovery of the motor function of rats with spinal cord injury.This study was approved by the Ethics Committee of the Guangxi Medical University of China(approval No.201810042)in October 2018.展开更多

Under these conditions,the extraction rate of total flavonoids was 1. 115%.[Conclusions] The extraction process obtained by response surface methodology was stable,reasonable,accurate and reliable. (TFH) quercetin (Que) and isorhamnetin (Isor) on the intracellular free calcium ([Ca^2+]) in vascular smooth muscle cells (VSMC) of spontaneously hypertensive rats (SHR) and Wistar-Kyoto rats (WKY). Metheds: Fluo 3-acetoxymethylester(Fluo-3/AM) was used to observe the effects of TFH (100mg/L) and its essential monomers, namely Que (10^-4mol/L) and Isor (10^-4mol/L) on changes of [Ca^2+]1 in cultured SHR and WKY VSMC (abbr. to Ca-SHR & Ca-WKY) following exposure to high K^+, norepinephrine (NE) and angiotensin Ⅱ (AngⅡ), and to compare with the effects of verapamil (Ver). Results: (1) TFH, Que and Isor had inhibitory effects on resting Ca-SHR (P〈0.05), but had no significant effects on Ca-WKY (P〉0.05). (2) High K^+ could increase Ca-SHR more significantly than Ca-WKY (P〈0.05); TFH, Que and Isor could inhibit the elevation of [Ca^2+]1 induced by high K^+ -depolarization, with the effects similar to that of Ver, and the effect on Ca-SHR was more significant than that on Ca-WKY (P〈0.05). (3) NE and Ang Ⅱ could increase Ca-SHR more significantly than Ca-WKY (P〈0.05), TFH, Que and Isor had remarkably inhibitory effect on the elevation of Ca-SHR and Ca-WKY induced by NE or Ang Ⅱ. (4) In the absence of extracellular Ca^2+ , TFH, Que and Isor also had certain inhibitory effect on Ca-SHR and Ca-WKY induced by NE, and the effect on the former was more significant than that on the latter(P〈0.05). Ceaclusiea: TFH, Que and Isor might decrease the levels of [Ca^2+], in VSMCs by blocking both voltage-dependent calcium channels (VDC) and receptoroperated calcium channels (ROC) in physiological or pathological state, which may be one of the important mechanisms of their hypotensive and protective effects on target organs in patients with hypertension.展开更多

Rat ischemia models were administered 0.3, 0.15 and 0.075 g/kg Radix Ilicis Pubescentis total flavonoids from 3 days before modeling to 2 days after injury. Results showed that Radix Ilicis Pubescentis total flavonoids could reduce pathological injury in rats with cerebral ischemia/reperfusion injury. The number of Nissl bodies increased, Bax protein expression decreased, Bcl-2 protein expression increased and the number of CD34-positive cells increased. Therefore, Radix Ilicis Pubescentis total flavonoids can improve the bone marrow stem cell mobilization effect, enhance the anti-apoptotic ability of nerve cells, and have a neuroprotective effect on cerebral ischemia/reperfusion injury in rats.展开更多

The optimal conditions for ultrasonic extraction of total flavonoids from A. keiskei were extraction time of 25 min, ethanol concentration of 80%, ultrasonic power of 60 W, solid-to-liquid ratio of 1:20 (g/ml), under which the yield of total flavonoids reached 1.56%. [Results]The optimal extraction process of CPTF was as below: ethanol concentration 54. 76%,extraction temperature 83. 92℃,extraction time 102. 64 min,solid-liquid ratio 1:20,extraction for twice. The effects of PTFCs separately or combined with arsenic trioxide on the proliferation and apoptosis of leukemia cells were determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT), fluorescence microscopy, and flow cytometry. Their effects on the expression levels of apoptosisrelated regulators were determined by Western blot assay. PTFCs combined with arsenic trioxide significantly inhibited the growth of Kasumi-1 cells, and apoptosis was confirmed by flow cytometry analysis. Hoechst 33258 staining showed more significant morphological changes and more apoptosis following the combined treatment. Western blots showed changes in the expression of genes for poly ADP-ribose polymerase(PARP), caspase 3/9, and P65. The results indicated that PTFCs separately or combined with arsenic trioxide inhibited proliferation of leukemia cells in vitro and induced their apoptosis by modulating the expression of apoptosis-related regulator genes.展开更多
In this study, bioassay-guided isolation, identification and biological evaluation of antioxidant components from total flavonoids of Chinese herbal plant Mimenghua were performed. 1, 1-Diphenyl-2-picrylhydrazyl(DPPH)...In this study, bioassay-guided isolation, identification and biological evaluation of antioxidant components from total flavonoids of Chinese herbal plant Mimenghua were performed. 1, 1-Diphenyl-2-picrylhydrazyl(DPPH) radical scavenging test was adopted as the bioassay-guided method, Sephadex LH-20 column chromatography and high performance liquid chromatography were used as the purification tools for the acquisition of antioxidant components. One compound was obtained and identified as acteoside by its physicochemical properties and spectral characteristics. Antioxidant activity in vitro of acteoside was determined by evaluation of the scavenging activity on DPPH radical and hydroxyl radical, reducing power and total antioxidant capability.The results showed that acteoside had significant antioxidative activity and possessed higher activity with the increase of concentration, which provides the potential application of acteoside in food, pharmaceutical and cosmetic industries.展开更多
Objective To observe the effects of total flavonoids of chrysanthemum and medicated serum on the expression of related proteins in the lacrimal tissue and dry-eye cell models of male rabbits with dry eye caused by cas...Objective To observe the effects of total flavonoids of chrysanthemum and medicated serum on the expression of related proteins in the lacrimal tissue and dry-eye cell models of male rabbits with dry eye caused by castration.Methods(1)150 male Japanese rabbits were randomly divided into five groups,with 30 rabbits in each group:normal control group(group A),sham group(group B),model group(group C),androgen control group(group D)and total flavonoids of chrysanthemum treatment group(group E).The androgen deficiency dry-eye model was established by bilateral castration in groups C,D and E.Normal saline was administered to groups A,B and C by gavage;androgen(testosterone propionate)was injected into muscle in group D;and group E was given total flavonoids of chrysanthemum by gavage.All white rabbits were tested the Schirmer I test(SIT)and tear break-up time(BUT).After euthanasia,tear gland tissue was harvested so that we could observe pathological changes in the expression of related inflammatory factors in the lacrimal gland tissue.The expression of interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α)and transforming growth factor-β1(TGF-β1)was detected in the lacrimal gland tissue by immunohistochemistry.Reverse transcription PCR was used to quantitatively detect expression of TGF-β1 mRNA.(2)Male Wistar rat lacrimal epithelial cells were used to establish a model of eye stem cell apoptosis caused by androgen levels.The blank control group was set up without androgen culture,the control group with androgen culture,and the total flavonoids of chrysanthemum group without androgen.The MTT method was used to determine the optimal intervention dosage of drug-containing plasma.Western blot and QPCR were used to detect the expression of AR mRNA,NF-κB phosphorylated protein and TGF-β1 in lacrimal epithelial cells,and the androgen-like effect of total flavonoids of chrysanthemum was observed.Results(1)Immunohistochemistry showed that groups A,B,D and E had significantly lower expression of IL-1βand TNF-αthan group C(P<0.05);among these,group E had slightly higher expression than group D(P>0.05).RT-PCR results showed that the relative expression of TGF-β1 mRNA in groups A,B,D and E was significantly higher than in group C(P<0.05),and the relative expression of TGF-β1 mRNA in groups D and E was higher than that in groups A and B(P<0.05).(2)Using the MTT method,the final concentration of interfering cells was calculated to be 13.2%.The expression of AR protein,NF-κB and TGF-β1 in the chrysanthemum flavonoid plasma intervention and testosterone propionate intervention groups was enhanced,and there were significant differences relative to the blank group(P<0.01).The expression level of NF-κB in the total flavonoid containing plasma intervention group was lower than that in the testosterone propionate intervention group(P<0.01).Conclusions The total flavonoids of chrysanthemum can inhibit IL-1βand TNF-αexpression in the lacrimal gland tissue of castrated male rabbits with dry eye to increase synthesis of TGF-β1 mRNA and TGF-β1,thereby inhibiting the inflammatory response.The medicated plasma with total flavonoids of chrysanthemum promotes expression of AR mRNA,upregulating expression of NF-κB,further promoting upregulation of TGF-β1 protein expression in lacrimal epithelial cells,inhibiting inflammation by regulating related proteins,and ultimately alleviating the symptoms of dry eye.展开更多

文摘[Objectives]To analyze the chemical components of Meconopsis integrifolia(Maxim.)Franch total flavonoids(MITF).[Methods]The chemical components of MITF were identified by UPLC-Q-Exactive Orbitrap MS.The column was performed using ACQUITY UPLC HSS T3(100 mm×2.1 mm,1.8μm)with 0.1%formic acid water(A)-acetonitrile(B)with gradient elution at 30℃,injection volume of 2μL,and flow rate of 0.3 mL/min.Electrospray ion source adopted positive and negative ion detection mode with scanning range m/z 100-1000.[Results]A total of 93 compounds were identified from MITF,including 57 flavonoids,25 phenolic acids,9 alkaloids and 2 others.Among them,dimethoxytaxifolin was identified as a possible new compound by SciFinder search,and 67 compounds were first identified in M.integrifolia.[Conclusions]This study provides a scientific foundation for clarifying the material basis of the efficacy of M.integrifolia and improving the quality standards.

文摘Background:The study aimed to investigate the protective effect and mechanism of total flavonoids of Scutellaria baicalensis(TFSB)on acute myocardial ischemia(AMI)rats by using functional metabonomics.Methods:Rats were divided into the Control,Model,AMI positive control(Propranolol hydrochloride,30 mg/kg),low dose TFSB(50 mg/kg),and high dose TFSB(100 mg/kg)groups.Rats received the corresponding treatment by intragastric administration once daily for 10 consecutive days.Electrocardiogram,myocardial enzyme,triphenyltetrazolium chloride staining,hematoxylin-eosin,and enzyme-linked immunosorbent assay were performed to evaluate the protective effect of TFSB on AMI rats.Then,the UHPLC-Q-Orbitrap MS method based on serum metabolomics was utilised to search for metabolic biomarkers and metabolic pathways.Subsequently,Western blot and RT-PCR techniques were employed to identify the respective genes and proteins.Results:Pharmacodynamics revealed that TFSB could ameliorate AMI in rats.The results of the metabolomics analysis indicated that the alterations in metabolic profile observed in rats with AMI were partially improved by treatment with TFSB.Moreover,the mRNA expression levels of 5-lipoxygenase(5-LOX)and 15-lipoxygenase(15-LOX)and the protein expression levels of 5-LOX,15-LOX,interleukin-1β(IL-1β),and NF-κB p65 were reduced following treatment with TFSB.Conclusion:The potential treatment of TFSB in AMI may be ascribed to its ability to regulate arachidonic acid metabolism.

文摘Aim To screen the optimum macroporous resin and conditions for the isolation and purification of flavonoids from Radix Puerariae. Methods The static and dynamic adsorption/desorption methods were used, and the separation and purification process was evaluated by measuring the concentration of total flavonoid in the fractions with UV spectrophotometer. Results The SP70 macroporous resin was the most effective compared with other macroporous resins. The optimum conditions were screened, which were 0.5 g· mL^- 1 corresponding to crude drug for concentration of extract, pH 5 - 6, and appended 60 times the volume of the resin bed (BV) with the adsorption speed 2 BV·h^-1, and the volume of aq. 70% (V/V) ethanol as eluant was 5 BV with desorption speed 2 BV·h^-1. By this method, the final contents of total flavonoids exceeded 80%. Conclusion The SP70 macroporous resin is the most effective one for large-scale isolation and purification of flavonoids from Radix Pueraria, which meets industrial needs.

文摘To assess the potential effect of total flavonoids of epimedium (TFE) on cytochrome P450 and activity of its main isoforms in rat liver microsomes. TFE (300 mg/kg) was administered once daily to male Sprague-Dowley rats by gavage for fifteen days. The total cytochrome P450 content and its main isoforms CYP1A2, CYP3A4 and CYP2E1 activities in rat liver microsomes were detected. The activity of CYP1A2 was measured by fluorometry and the activities of CYP3A4 and CYP2E1 were determined by measuring the amount of methanal and p-aminophenol formed using UV/Vis spectrophotometer, respectively. Administration of TFE significantly increased the total CYP450 content and activities ofCYP1A2, CYP3A4 and CYP2E1 in rat liver microsomes, compared with the control group. Particularly, the activities of CYP1A2 and CYP2E1 were enhanced significantly (P〈0.01). TFE induced the increase in total CYP450 content and its main isoforms CYP1A2, CYP3A4 and CYP2E1 activities in rat liver microsomes.
文摘Suaeda salsa an annual herb of Chenopodiaceae Suaeda well accepted as both a folk medicinal and food plant.In the current study,the extraction of total flavonoids from S.salsa and the in vitro antioxidant,hypoglycemic and hypolipidemic activities of the extracts were investigated.The total flavonoids was extracted by refluxing with ethanol and the selection and optimization of main experimental parameters were carried out using single factor experiments combined with central composite design-response surface methodology(CCD-RSM).The total flavonoids extracted from S.salsa were tested for hydroxyl and oxygen radical scavenging activity,and inhibitory activities againstα-amylase and lipase in vitro.Results revealed that the optimal conditions for the extraction of total flavonoids were as following:liquid-tosolid ratio,31.40:1;ethanol concentration,83.12%;reflux temperature,90℃,and reflux time,128.40 min.Under these optimized conditions,the predicted and actual values of total flavonoids yield were 0.896%and 0.932%,respectively.The in vitro antioxidant tests revealed that the total flavonoids could scavenge hydroxyl and oxygen radicals and showed inhibitory activity againstα-amylase and lipase.The optimal extraction conditions obtained in this experiment are simple to perform and have good reproducibility.The results have demonstrated that the total flavonoids from S.salsa can be used as a potential source of natural antioxidant,hypoglycemic and hypolipidemic ingredients,which provides new pharmacological basis for the development of S.salsa.

文摘Previous experimental studies have shown that cerebral infarction can be effectively reduced following treatment with scutellaria baicalensis stem-leaf total flavonoid (SSTF). However, the mechanism of action of SSTF as a preventive drug to treat cerebral infarction remains unclear. In this study, Sprague-Dawley rats were pretreated with 50, 100, 200 mg/kg SSTF via intragastric ad- ministration for 1 week prior to the establishment of focal cerebral ischemia/reperfusion injury. The results showed that pretreatment with SSTF effectively improved neurological function, reduced brain water content and the permeability of blood vessels, ameliorated ischemia-induced morphology changes in hippocampal microvessels, down-regulated Fas and FasL protein expression, elevated the activity of superoxide dismutase and glutathione peroxidase, and decreased malondialdehyde content. In contrast to low-dose SSTF pretreatment, the above changes were most obvious after pretreatment with moderateand high-doses of SSTF. Experimental findings indicate that SSTF pretreatment can exert protective effects on the brain against cerebral ischemia/reperfusion injury. The underlying mechanisms may involve reducing brain water content, increasing microvascular recanalization, inhibiting the apoptosis of hippocampal neurons, and attenuating free radical damage.

文摘Scutellaria baicalensis stem-leaf total flavonoid might attenuate learning/memory impairment and neuronal loss in rats induced by amyloid beta-peptide. This study aimed to explore the effects of Scutellaria baicalensis stem-leaf total flavonoid on amyloid beta-peptide-induced neuronal apoptosis and the expression of apoptosis-related proteins in the rat hippocampus. Male Wistar rats were given intragastric administration of Scutellaria baicalensis stem-leaf total flavonoid, 50 or 100 mg/kg, once per day. On day 8 after administration, 10 pg amyloid beta-peptide (25-35) was injected into the bilateral hippocampus of rats to induce neuronal apoptosis. On day 20, hippocampal tissue was harvested and probed with the terminal deoxyribonucleotidyl transferase-mediated biotin-16-dUTP nick-end labeling assay. Scutellaria baicalensis stem-leaf total flavonoid at 50 and 100 mg/kg reduced neuronal apoptosis induced by amyloid beta-peptide (25-35) in the rat hippocampus. Immunohistochemistry and western blot assay revealed that expression of the pro-apoptotic protein Bax, cytochrome c and caspase-3 was significantly diminished by 50 and 100 mg/kg Scutellaria baicalensis stem-leaf total flavonoid, while expression of the anti-apoptotic protein Bcl-2 was increased. Moreover, 100 mg/kg Scutellana baicalensis stem-leaf total flavonoid had a more dramatic effect than the lower dosage. These experimental findings indicate that Scutellaria baicalensis stem-leaf total flavonoid dose-dependently attenuates neuronal apoptosis induced by amyloid beta-peptide in the hippocampus, and it might mediate this by regulating the expression of Bax, cytochrome c, caspase-3 and Bcl-2.

文摘Flavonoids have been reported to have therapeutic potential for spinal cord injury.Hawthorn leaves have abundant content and species of total flavonoids,and studies of the effects of the total flavonoids of hawthorn leaves on spinal cord injury have not been published in or outside China.Therefore,Sprague-Dawley rats were used to establish a spinal cord injury model by Allen's method.Rats were intraperitoneally injected with 0.2 m L of different concentrations of total flavonoids of hawthorn leaves(5,10,and 20 mg/kg)after spinal cord injury.Injections were administered once every 6 hours,three times a day,for 14 days.After treatment with various concentrations of total flavonoids of hawthorn leaves,the Basso,Beattie,and Bresnahan scores and histological staining indicated decreases in the lesion cavity and number of apoptotic cells of the injured spinal cord tissue;the morphological arrangement of the myelin sheath and nerve cells tended to be regular;and the Nissl bodies in neurons increased.The Basso,Beattie,and Bresnahan scores of treated spinal cord injury rats were increased.Western blot assays showed that the expression levels of pro-apoptotic Bax and cleaved caspase-3 were decreased,but the expression level of the anti-apoptotic Bcl-2 protein was increased.The improvement of the above physiological indicators showed a dose-dependent relationship with the concentration of total flavonoids of hawthorn leaves.The above findings confirm that total flavonoids of hawthorn leaves can reduce apoptosis and exert neuroprotective effects to promote the recovery of the motor function of rats with spinal cord injury.This study was approved by the Ethics Committee of the Guangxi Medical University of China(approval No.201810042)in October 2018.

文摘[Objectives] Response surface analysis methodology was used to optimize the extraction technology of total flavonoids from Mallotus apelta leaves. [Methods] The total flavonoids were extracted using ultrasonic assisted ethanol extraction,and extraction rate of total flavonoids was taken as evaluation index. On the basis of single-factor experiment,Box-Behnken response surface methodology was employed for selecting the optimum extraction process. [Results]The optimum extraction conditions of total flavonoids were as follows: 75% of ethanol concentration,1∶ 25 of ratio of material to liquid,31 min of ultrasonic time. Under these conditions,the extraction rate of total flavonoids was 1. 115%.[Conclusions] The extraction process obtained by response surface methodology was stable,reasonable,accurate and reliable. It was a feasible method to extract the total flavonoids from M. apelta leaves.

文摘As a Chinese medicinal plant,Smilax china is traditionally used in the treatment of pelvic inflammatory disease(PID).Flavonoids have been identified as the anti-inflammatory bioactive fraction.However,the high content of total flavonoids has not been extracted from S.china.The therapeutic effects and mechanisms of total flavonoids are still unknown.In the present study,we aimed to obtain the effective parts of flavonoids in S.china,and elucidate the mechanism of action on PID.The anti-inflammatory effect was evaluated on phenol mucilage-induced pelvic inflammation in rats.This was the first time that total flavonoids in high concentration(up to 55.6%)were obtained in S.china.A total of 15 compounds were separated and identified by NMR,and the total flavonoids were detected with HPLC.TFSC caused a reduction in serum levels of IL-6 and IL-1β.These results suggested that TFSC had a significant anti-PID effect,probably due to inhibition of the inflammatory reaction.

文摘To explore the effects of total flavonoids of Hippophae rhamnoides L. (TFH) quercetin (Que) and isorhamnetin (Isor) on the intracellular free calcium ([Ca^2+]) in vascular smooth muscle cells (VSMC) of spontaneously hypertensive rats (SHR) and Wistar-Kyoto rats (WKY). Metheds: Fluo 3-acetoxymethylester(Fluo-3/AM) was used to observe the effects of TFH (100mg/L) and its essential monomers, namely Que (10^-4mol/L) and Isor (10^-4mol/L) on changes of [Ca^2+]1 in cultured SHR and WKY VSMC (abbr. to Ca-SHR & Ca-WKY) following exposure to high K^+, norepinephrine (NE) and angiotensin Ⅱ (AngⅡ), and to compare with the effects of verapamil (Ver). Results: (1) TFH, Que and Isor had inhibitory effects on resting Ca-SHR (P〈0.05), but had no significant effects on Ca-WKY (P〉0.05). (2) High K^+ could increase Ca-SHR more significantly than Ca-WKY (P〈0.05); TFH, Que and Isor could inhibit the elevation of [Ca^2+]1 induced by high K^+ -depolarization, with the effects similar to that of Ver, and the effect on Ca-SHR was more significant than that on Ca-WKY (P〈0.05). (3) NE and Ang Ⅱ could increase Ca-SHR more significantly than Ca-WKY (P〈0.05), TFH, Que and Isor had remarkably inhibitory effect on the elevation of Ca-SHR and Ca-WKY induced by NE or Ang Ⅱ. (4) In the absence of extracellular Ca^2+ , TFH, Que and Isor also had certain inhibitory effect on Ca-SHR and Ca-WKY induced by NE, and the effect on the former was more significant than that on the latter(P〈0.05). Ceaclusiea: TFH, Que and Isor might decrease the levels of [Ca^2+], in VSMCs by blocking both voltage-dependent calcium channels (VDC) and receptoroperated calcium channels (ROC) in physiological or pathological state, which may be one of the important mechanisms of their hypotensive and protective effects on target organs in patients with hypertension.

文摘Previous studies have shown that Radix Ilicis Pubescentis total flavonoids have a neuroprotective effect, but it remains unclear whether Radix Ilicis Pubescentis total flavonoids have a synergistic effect with the recombinant human granulocyte colony stimulating factor-mobilized bone marrow stem cell transplantation on cerebral ischemia/reperfusion injury. Rat ischemia models were administered 0.3, 0.15 and 0.075 g/kg Radix Ilicis Pubescentis total flavonoids from 3 days before modeling to 2 days after injury. Results showed that Radix Ilicis Pubescentis total flavonoids could reduce pathological injury in rats with cerebral ischemia/reperfusion injury. The number of Nissl bodies increased, Bax protein expression decreased, Bcl-2 protein expression increased and the number of CD34-positive cells increased. Therefore, Radix Ilicis Pubescentis total flavonoids can improve the bone marrow stem cell mobilization effect, enhance the anti-apoptotic ability of nerve cells, and have a neuroprotective effect on cerebral ischemia/reperfusion injury in rats.

[Methods] The ultrasonic extraction method was adopted and orthogonal design was carried out to study the effects of ethanol concentration( A),ultrasonic time( B),the solid-to-liquid ratio( C) on the extraction of flavonoids from leaves of P. ramosissimus. The Microplate Reader was used to measure the content of flavonoids in leaves of P. ramosissimus. in different extraction conditions,P. ramosissimus. and the optimal extraction conditions were used to measure the content of flavonoids from leaves of P. ramosissimus.[Results]Effects of the ultrasonic extraction method on the extraction rate of flavonoids from leaves of P. ramosissimus: extraction times >ethanol concentration > Ultrasound time > solid-liquid ratio. The screened optimal extraction process was: ethanol concentration of 65%,the solid-to-liquid ratio of 1∶ 25,ultrasonic time of 2 h and extraction times Under this condition,the extraction rate of total flavonoids in leaves of P. ramosissimus. was 2. 56%. [Conclusions] This method is simple and feasibility,reliable,and suitable for rapid measurement with large samples.

文摘[Objective] This study aimed to optimize the ultrasonic extraction process of total flavonoids from Angelica keiskei Koidzumi. [Method] Using the yield of total flavonoids as an indicator, the ultrasonic extraction process of total flavonoids from A. keiskei was optimized by orthogonal experimental design. [ Result ] Among four factors in orthogonal experimental design, extraction time exhibited the most significant effect on the extraction result; ethanol concentration exhib- ited a significant effect on the extraction result; ultrasonic power and solid-to-liquid ratio had no significant effect. The optimal conditions for ultrasonic extraction of total flavonoids from A. keiskei were extraction time of 25 min, ethanol concentration of 80%, ultrasonic power of 60 W, solid-to-liquid ratio of 1:20 (g/ml), under which the yield of total flavonoids reached 1.56%. [ Conclusion] This study provides the basis for further development and utilization of total flavonoids from A. keiskei.

文摘[Objectives] The research aimed to optimize extraction process of Clerodendrum philippinum Schauer var. simplex Mlodenke total flavonoids( CPTF),and provide reference for its development and utilization. [Methods] Based on single-factor test,ethanol concentration,extraction temperature and extraction time were taken as independent variables,and total flavonoids yield was taken as dependent variable. The test was conducted according to central composite design principle. Multivariate linear regression and binomial equation fitting of the result were conducted,and extraction process of CPTF was optimized by using response surface methodology. [Results]The optimal extraction process of CPTF was as below: ethanol concentration 54. 76%,extraction temperature 83. 92℃,extraction time 102. 64 min,solid-liquid ratio 1:20,extraction for twice. [Conclusions] The extraction process of CPTF by central composite design-response surface methodology was simple and feasible,with reliable prediction result,which was suitable for industrial production.

文摘[Objectives]This study aimed to optimize the extraction process of total flavonoids in Cardamine tangutorum O.E.Schulz.[Methods]On the basis of single-factor experiments,the ultrasonic-enzymatic extraction process of total flavonoids in C.tangutorum O.E.Schulz was optimized by Box-Behnken design from aspects of ethanol volume fraction,extraction time,extraction temperature and enzyme addition,with yield of total flavonoids as the evaluation index.[Results]The optimal extraction process of total flavonoids in C.tangutorum O.E.Schulz was as follows:ethanol volume fraction of 70%,ultrasonic extraction time of 43 min,ultrasonic extraction temperature of 37℃and cellulose addition of 0.07 mg.Under the optimal conditions,the yield of total flavonoids from C.tangutorum O.E.Schulz reached 75.90 mg/g.[Conclusions]The optimized extraction process was reasonable and feasible,and it could provide a reference for the extraction of total flavonoids in C.tangutorum O.E.Schulz.This study provided a foundation for the quality control of food/medicinal materials and the development and research of big health products of C.tangutorum O.E.Schulz.

文摘To investigate the potential effects of pure total flavonoid compounds(PTFCs) from Citrus paradisi Macfadyen separately or combined with arsenic trioxide on the proliferation of human myeloid leukemia cells and the mechanisms underlying the action of PTFCs. The effects of PTFCs separately or combined with arsenic trioxide on the proliferation and apoptosis of leukemia cells were determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT), fluorescence microscopy, and flow cytometry. Their effects on the expression levels of apoptosisrelated regulators were determined by Western blot assay. PTFCs combined with arsenic trioxide significantly inhibited the growth of Kasumi-1 cells, and apoptosis was confirmed by flow cytometry analysis. Hoechst 33258 staining showed more significant morphological changes and more apoptosis following the combined treatment. Western blots showed changes in the expression of genes for poly ADP-ribose polymerase(PARP), caspase 3/9, and P65. The results indicated that PTFCs separately or combined with arsenic trioxide inhibited proliferation of leukemia cells in vitro and induced their apoptosis by modulating the expression of apoptosis-related regulator genes.

文摘In this study, bioassay-guided isolation, identification and biological evaluation of antioxidant components from total flavonoids of Chinese herbal plant Mimenghua were performed. 1, 1-Diphenyl-2-picrylhydrazyl(DPPH) radical scavenging test was adopted as the bioassay-guided method, Sephadex LH-20 column chromatography and high performance liquid chromatography were used as the purification tools for the acquisition of antioxidant components. One compound was obtained and identified as acteoside by its physicochemical properties and spectral characteristics. Antioxidant activity in vitro of acteoside was determined by evaluation of the scavenging activity on DPPH radical and hydroxyl radical, reducing power and total antioxidant capability.The results showed that acteoside had significant antioxidative activity and possessed higher activity with the increase of concentration, which provides the potential application of acteoside in food, pharmaceutical and cosmetic industries.

文摘Objective To observe the effects of total flavonoids of chrysanthemum and medicated serum on the expression of related proteins in the lacrimal tissue and dry-eye cell models of male rabbits with dry eye caused by castration.Methods(1)150 male Japanese rabbits were randomly divided into five groups,with 30 rabbits in each group:normal control group(group A),sham group(group B),model group(group C),androgen control group(group D)and total flavonoids of chrysanthemum treatment group(group E).The androgen deficiency dry-eye model was established by bilateral castration in groups C,D and E.Normal saline was administered to groups A,B and C by gavage;androgen(testosterone propionate)was injected into muscle in group D;and group E was given total flavonoids of chrysanthemum by gavage.All white rabbits were tested the Schirmer I test(SIT)and tear break-up time(BUT).After euthanasia,tear gland tissue was harvested so that we could observe pathological changes in the expression of related inflammatory factors in the lacrimal gland tissue.The expression of interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α)and transforming growth factor-β1(TGF-β1)was detected in the lacrimal gland tissue by immunohistochemistry.Reverse transcription PCR was used to quantitatively detect expression of TGF-β1 mRNA.(2)Male Wistar rat lacrimal epithelial cells were used to establish a model of eye stem cell apoptosis caused by androgen levels.The blank control group was set up without androgen culture,the control group with androgen culture,and the total flavonoids of chrysanthemum group without androgen.The MTT method was used to determine the optimal intervention dosage of drug-containing plasma.Western blot and QPCR were used to detect the expression of AR mRNA,NF-κB phosphorylated protein and TGF-β1 in lacrimal epithelial cells,and the androgen-like effect of total flavonoids of chrysanthemum was observed.Results(1)Immunohistochemistry showed that groups A,B,D and E had significantly lower expression of IL-1βand TNF-αthan group C(P<0.05);among these,group E had slightly higher expression than group D(P>0.05).RT-PCR results showed that the relative expression of TGF-β1 mRNA in groups A,B,D and E was significantly higher than in group C(P<0.05),and the relative expression of TGF-β1 mRNA in groups D and E was higher than that in groups A and B(P<0.05).(2)Using the MTT method,the final concentration of interfering cells was calculated to be 13.2%.The expression of AR protein,NF-κB and TGF-β1 in the chrysanthemum flavonoid plasma intervention and testosterone propionate intervention groups was enhanced,and there were significant differences relative to the blank group(P<0.01).The expression level of NF-κB in the total flavonoid containing plasma intervention group was lower than that in the testosterone propionate intervention group(P<0.01).Conclusions The total flavonoids of chrysanthemum can inhibit IL-1βand TNF-αexpression in the lacrimal gland tissue of castrated male rabbits with dry eye to increase synthesis of TGF-β1 mRNA and TGF-β1,thereby inhibiting the inflammatory response.The medicated plasma with total flavonoids of chrysanthemum promotes expression of AR mRNA,upregulating expression of NF-κB,further promoting upregulation of TGF-β1 protein expression in lacrimal epithelial cells,inhibiting inflammation by regulating related proteins,and ultimately alleviating the symptoms of dry eye.
