Background Sperm cryopreservation is widely used in the cattle industry,as it allows for disassociating the localiza-tion of sires and the collection of semen from the timing of artificial insemination.While freeze-th...Background Sperm cryopreservation is widely used in the cattle industry,as it allows for disassociating the localiza-tion of sires and the collection of semen from the timing of artificial insemination.While freeze-thawing is known to impair sperm DNA integrity,whether the damage induced consists of single-(SSB)or double-strand breaks(DSB)has not been determined.In addition,no previous study has addressed if DNA breaks preferentially reside in specific genome regions such as those forming the toroid linker regions,or are rather spread throughout the regions linked to protamines.The main aim of the present work,therefore,was to elucidate the type and localization of the DNA damage generated by cryopreservation and to evaluate its impact on artificial insemination outcomes in cattle.Results The incidence of SSB and DSB was evaluated in 12 ejaculates before and after cryopreservation with the Comet assay,and the localization of the DNA breaks was assessed using pulsed-field gel electrophoresis(PFGE).Before cryopreservation,the incidence of SSB was 10.99%±4.62%and involved 20.56%±3.04%of sperm cells,whereas these figures significantly(P<0.0001)increased up to 34.11%±3.48%and 53.36%±11.00%in frozen-thawed sperm.In contrast,no significant differences in the incidence of DSB were observed(P>0.990)before and after cryopreservation(before:incidence of 13.91%±1.75%of sperm DNA affecting 56.04%±12.49%of sperm cells;after:incidence of 13.55%±1.55%of sperm DNA involving 53.36%±11.00%of sperm cells).Moreover,PFGE revealed that the percentage of sperm DNA fragments whose length was shorter than a toroid(<31.5 kb)was greater(P<0.0001)after(27.00%±4.26%)than before freeze-thawing(15.57%±4.53%).These differences indicated that the DNA breaks induced by cryopreservation affect the regions condensed in protamines,which are structured in toroids.On the other hand,in vivo fertility rates were associated to the incidence of SSB and DSB in frozen-thawed sperm(P=0.032 and P=0.005),but not with the size of the DNA fragments resulting from these breaks(P>0.05).Conclusion Cryopreservation of bovine sperm generates single-strand DNA breaks,which are mainly located in protamine-condensed toroidal regions.The incidence of DNA breaks in cryopreserved sperm has an impact on cat-tle fertility,regardless of the size of generated fragments.展开更多
Toroidal torques,generated by the resonant magnetic perturbation(RMP)and acting on the plasma column,are numerically systematically investigated for an ITER baseline scenario.The neoclassical toroidal viscosity(NTV),i...Toroidal torques,generated by the resonant magnetic perturbation(RMP)and acting on the plasma column,are numerically systematically investigated for an ITER baseline scenario.The neoclassical toroidal viscosity(NTV),in particular the resonant portion,is found to provide the dominant contribution to the total toroidal torque under the slow plasma flow regime in ITER.While the electromagnetic torque always opposes the plasma flow,the toroidal torque associated with the Reynolds stress enhances the plasma flow independent of the flow direction.A peculiar double-peak structure for the net NTV torque is robustly computed for ITER,as the toroidal rotation frequency is scanned near the zero value.This structure is found to be ultimately due to a non-monotonic behavior of the wave-particle resonance integral(over the particle pitch angle)in the superbanana plateau NTV regime in ITER.These findings are qualitatively insensitive to variations of a range of factors including the wall resistivity,the plasma pedestal flow and the assumed frequency of the rotating RMP field.展开更多
The exploration of exotic shapes and properties of atomic nuclei,e.g.,αcluster and toroidal shape,is a fascinating field in nuclear physics.To study the decay of these nuclei,a novel detector aimed at detecting multi...The exploration of exotic shapes and properties of atomic nuclei,e.g.,αcluster and toroidal shape,is a fascinating field in nuclear physics.To study the decay of these nuclei,a novel detector aimed at detecting multipleα-particle events was designed and constructed.The detector comprises two layers of double-sided silicon strip detectors(DSSD)and a cesium iodide scintillator array coupled with silicon photomultipliers array as light sensors,which has the advantages of their small size,fast response,and large dynamic range.DSSDs coupled with cesium iodide crystal arrays are used to distinguish multipleαhits.The detector array has a compact and integrated design that can be adapted to different experimental conditions.The detector array was simulated using Geant4,and the excitation energy spectra of someα-clustering nuclei were reconstructed to demonstrate the performance.The simulation results show that the detector array has excellent angular and energy resolutions,enabling effective reconstruction of the nuclear excited state by multipleαparticle events.This detector offers a new and powerful tool for nuclear physics experiments and has the potential to discover interesting physical phenomena related to exotic nuclear structures and their decay mechanisms.展开更多
The hybrid scenario,which has good confinement and moderate MHD instabilities,is a proposed operation scenario for international thermonuclear experimental reactor(ITER).In this work,the effect of plasma rotation on t...The hybrid scenario,which has good confinement and moderate MHD instabilities,is a proposed operation scenario for international thermonuclear experimental reactor(ITER).In this work,the effect of plasma rotation on the HL-3 hybrid scenario is analyzed with the integrated modeling framework OMFIT.The results show that toroidal rotation has no obvious effect on confinement with a high line averaged density of n_(bar)~(7)×10^(19)m^(-3).In this case,the ion temperature only changes from 4.7 keV to 4.4 keV with the rotation decreasing from 10^(5) rad/s to 10^(3) rad/s,which means that the turbulent heat transport is not dominant.While in the scenarios characterized by lower densities,such as n_(bar)~4×10^(19)m^(-3),turbulent transport becomes dominant in determining heat transport.The ion temperature rises from 3.8 keV to 6.1 keV in the core as the rotation velocity increases from 10^(3) rad/s to 10^(5) rad/s.Despite the ion temperature rising,the rotation velocity does not obviously affect electron temperature or density.Additionally,it is noteworthy that the variation in rotation velocity does not significantly affect the global confinement of plasma in scenarios with low density or with high density.展开更多

Terahertz metamaterial biosensors have attracted significant attention in the biological field due to their advantages of label-free,real-time and in situ detection.In this paper,a highly sensitive metamaterial sensor...Terahertz metamaterial biosensors have attracted significant attention in the biological field due to their advantages of label-free,real-time and in situ detection.In this paper,a highly sensitive metamaterial sensor with semi-ring mirror symmetry based on toroidal dipole resonance is designed for a new metamaterial biosensor.It is shown that a refractive index sensitivity of 337.5 GHz per refractive index unit can be achieved under an analyte of saturated thickness near a 1.33 THz transmission dip.For biosensor samples where aflatoxin B1 is dropped on the metamaterial surface in our experiment,dip amplitudes of transmission varying from 0.1904 to 0.203 and 0.2093 are observed as aflatoxin B1 concentrations are altered from 0 to 0.001μg·ml-1 and to 0.01μg·ml-1,respectively.Furthermore,when aflatoxin B1 concentrations are 0.1μg·ml-1,1μg·ml-1,10μg·ml-1 and 100μg·ml-1,dip amplitudes of 0.2179,0.226,0.2384 and 0.2527 and dip redshifts of 10.1 GHz,20.1 GHz,27.7 GHz and 37.6 GHz are respectively observed.These results illustrate high-sensitivity,label-free detection of aflatoxin B1,enriching the applications of sensors in the terahertz domain.展开更多
Density limit has long been a widely studied issue influencing the operating range of tokamaks.The rapid growth of the m/n=2/1(where m and n are poloidal and toroidal mode numbers,respectively)tearing mode is generall...Density limit has long been a widely studied issue influencing the operating range of tokamaks.The rapid growth of the m/n=2/1(where m and n are poloidal and toroidal mode numbers,respectively)tearing mode is generally regarded as a primary precursor to the density limit disruption.In this experiment,the coupling of the m/n=1/1 mode and the m/n=2/1 mode in highdensity plasma was observed.During a sawtooth cycle,the frequencies of the two modes gradually converge until they become equal.After that,toroidal coupling occurs between the 1/1 and 2/1 modes,resulting in a mutually fixed phase relationship.With the occurrence of toroidal coupling,the 2/1 mode is stabilized.Prior to the disruption,the cessation of the 1/1 and 2/1 mode coupling,along with the rapid growth in the amplitude of the 2/1 mode,can be observed.Additionally,under the same parameters,comparing discharges with or without the 1/1 mode,it is found that the presence of the 1/1 mode leads to higher plasma density and temperature parameters.展开更多
The effects of equilibrium toroidal rotation during edge-localized mode(ELM)mitigated by resonant magnetic perturbation(RMP)are studied with the experimental equilibria of the EAST tokamak based on the four-field mode...The effects of equilibrium toroidal rotation during edge-localized mode(ELM)mitigated by resonant magnetic perturbation(RMP)are studied with the experimental equilibria of the EAST tokamak based on the four-field model in the BOUT++code.As the two main parameters to determine the toroidal rotation profiles,the rotation shear and magnitudes were separately scanned to investigate their roles in the impact of RMPs on peeling-ballooning(P-B)modes.On one hand,the results show that strong toroidal rotation shear is favorable for the enhancement of the self-generated E×B shearing rate<ω_(E×B)>with RMPs,leading to significant ELM mitigation with RMP in the stronger toroidal rotation shear region.On the other hand,toroidal rotation magnitudes may affect ELM mitigation by changing the penetration of the RMPs,more precisely the resonant components.RMPs can lead to a reduction in the pedestal energy loss by enhancing the multimode coupling in the turbulence transport phase.The shielding effects on RMPs increase with the toroidal rotation magnitude,leading to the enhancement of the multimode coupling with RMPs to be significantly weakened.Hence,the reduction in pedestal energy loss by RMPs decreased with the rotation magnitude.In brief,the results show that toroidal rotation plays a dual role in ELM mitigation with RMP by changing the shielding effects of plasma by rotation magnitude and affecting<ω_(E×B)>by rotation shear.In the high toroidal rotation region,toroidal rotation shear is usually strong and hence plays a dominant role in the influence of RMP on P-B modes,whereas in the low rotation region,toroidal rotation shear is weak and has negligible impact on P-B modes,and the rotation magnitude plays a dominant role in the influence of RMPs on the P-B modes by changing the field penetration.Therefore,the dual role of toroidal rotation leads to stronger ELM mitigation with RMP,which may be achieved both in the low toroidal rotation region and the relatively high rotation region that has strong rotational shear.展开更多
The CLT code was used to quantitatively study the impact of toroidal mode coupling on the explosive dynamics of the m/n=3/1 double tearing mode.The focus of this study was on explosive reconnection processes,in which ...The CLT code was used to quantitatively study the impact of toroidal mode coupling on the explosive dynamics of the m/n=3/1 double tearing mode.The focus of this study was on explosive reconnection processes,in which the energy bursts and the main mode no longer dominates when the separation between two rational surfaces is relatively large in the medium range.The development of higher m and n modes is facilitated by a relatively large separation between two rational surfaces,a small q_(min)(the minimum value of the safety factor),or low resistivity.The relationships between the higher m and n mode development,explosive reconnection rate,and position exchange of 3/1 islands are summarized for the first time.Separation plays a more important role than q_(min)in enhancing the development of higher m and n modes.At a relatively large separation,the good development of higher m and n modes greatly reduces the reconnection rate and suppresses the development of the main mode,resulting in the main mode not being able to develop sufficiently large to generate the position changes of 3/1 islands.展开更多
In this paper, a logistical regression statistical analysis (LR) is presented for a set of variables used in experimental measurements in reversed field pinch (RFP) machines, commonly known as “slinky mode” (SM), ob...In this paper, a logistical regression statistical analysis (LR) is presented for a set of variables used in experimental measurements in reversed field pinch (RFP) machines, commonly known as “slinky mode” (SM), observed to travel around the torus in Madison Symmetric Torus (MST). The LR analysis is used to utilize the modified Sine-Gordon dynamic equation model to predict with high confidence whether the slinky mode will lock or not lock when compared to the experimentally measured motion of the slinky mode. It is observed that under certain conditions, the slinky mode “locks” at or near the intersection of poloidal and/or toroidal gaps in MST. However, locked mode cease to travel around the torus;while unlocked mode keeps traveling without a change in the energy, making it hard to determine an exact set of conditions to predict locking/unlocking behaviour. The significant key model parameters determined by LR analysis are shown to improve the Sine-Gordon model’s ability to determine the locking/unlocking of magnetohydrodyamic (MHD) modes. The LR analysis of measured variables provides high confidence in anticipating locking versus unlocking of slinky mode proven by relational comparisons between simulations and the experimentally measured motion of the slinky mode in MST.展开更多

Furthermore, full account is taken for the high...We will give an overview of results obtained by our reactive fluid model. It is characterised as a fluid model where all moments with sources in the experiment are kept. Furthermore, full account is taken for the highest moments appearing in unexpanded denominators also including full toroidicity. It has been demonstrated that the strength of zonal flows is dramatically larger in reactive fluid closures than in those which involve dissipation. This gives a direct connection between the fluid closure and the level of excitation of turbulence. This is because zonal flows are needed to absorb the inverse cascade in quasi 2D turbulence. This also explains the similarity in structure of the transport coefficients in our model with a reactive closure in the energy equation and models which have a reactive closure because of zero ion temperature such as the Hasegawa-Wakatani model. Our exact reactive closure unifies several well-known features of tokamak experiments such as the L-H transition, internal transport barriers and the nonlinear Dimits upshift of the critical gradient for onset of transport. It also gives transport of the same level as that in nonlinear gyrokinetic codes. Since these include the kinetic resonance this confirms the validity of the thermodynamic properties of our model. Furthermore, we can show that while a strongly nonlinear model is needed in kinetic theory a quasilinear model is sufficient in the fluid description. Thus our quasilinear fluid model will be adequate for treating all relevant problems in bulk transport. This is finally confirmed by the reproduction by the model of the experimental power scaling of the confinement time Te ~ P-2/3. This confirms the validity of our reactive fluid model. This also gives credibility to our ITER simulations including the H-mode barrier. A new result is here, that alpha heating strongly reduces the slope of the H-mode barrier. This should significantly reduce the effects of ELM's.展开更多

文摘Density limit has long been a widely studied issue influencing the operating range of tokamaks.The rapid growth of the m/n=2/1(where m and n are poloidal and toroidal mode numbers,respectively)tearing mode is generally regarded as a primary precursor to the density limit disruption.In this experiment,the coupling of the m/n=1/1 mode and the m/n=2/1 mode in highdensity plasma was observed.During a sawtooth cycle,the frequencies of the two modes gradually converge until they become equal.After that,toroidal coupling occurs between the 1/1 and 2/1 modes,resulting in a mutually fixed phase relationship.With the occurrence of toroidal coupling,the 2/1 mode is stabilized.Prior to the disruption,the cessation of the 1/1 and 2/1 mode coupling,along with the rapid growth in the amplitude of the 2/1 mode,can be observed.Additionally,under the same parameters,comparing discharges with or without the 1/1 mode,it is found that the presence of the 1/1 mode leads to higher plasma density and temperature parameters.

文摘The effects of equilibrium toroidal rotation during edge-localized mode(ELM)mitigated by resonant magnetic perturbation(RMP)are studied with the experimental equilibria of the EAST tokamak based on the four-field model in the BOUT++code.As the two main parameters to determine the toroidal rotation profiles,the rotation shear and magnitudes were separately scanned to investigate their roles in the impact of RMPs on peeling-ballooning(P-B)modes.On one hand,the results show that strong toroidal rotation shear is favorable for the enhancement of the self-generated E×B shearing rate<ω_(E×B)>with RMPs,leading to significant ELM mitigation with RMP in the stronger toroidal rotation shear region.On the other hand,toroidal rotation magnitudes may affect ELM mitigation by changing the penetration of the RMPs,more precisely the resonant components.RMPs can lead to a reduction in the pedestal energy loss by enhancing the multimode coupling in the turbulence transport phase.The shielding effects on RMPs increase with the toroidal rotation magnitude,leading to the enhancement of the multimode coupling with RMPs to be significantly weakened.Hence,the reduction in pedestal energy loss by RMPs decreased with the rotation magnitude.In brief,the results show that toroidal rotation plays a dual role in ELM mitigation with RMP by changing the shielding effects of plasma by rotation magnitude and affecting<ω_(E×B)>by rotation shear.In the high toroidal rotation region,toroidal rotation shear is usually strong and hence plays a dominant role in the influence of RMP on P-B modes,whereas in the low rotation region,toroidal rotation shear is weak and has negligible impact on P-B modes,and the rotation magnitude plays a dominant role in the influence of RMPs on the P-B modes by changing the field penetration.Therefore,the dual role of toroidal rotation leads to stronger ELM mitigation with RMP,which may be achieved both in the low toroidal rotation region and the relatively high rotation region that has strong rotational shear.

文摘The CLT code was used to quantitatively study the impact of toroidal mode coupling on the explosive dynamics of the m/n=3/1 double tearing mode.The focus of this study was on explosive reconnection processes,in which the energy bursts and the main mode no longer dominates when the separation between two rational surfaces is relatively large in the medium range.The development of higher m and n modes is facilitated by a relatively large separation between two rational surfaces,a small q_(min)(the minimum value of the safety factor),or low resistivity.The relationships between the higher m and n mode development,explosive reconnection rate,and position exchange of 3/1 islands are summarized for the first time.Separation plays a more important role than q_(min)in enhancing the development of higher m and n modes.At a relatively large separation,the good development of higher m and n modes greatly reduces the reconnection rate and suppresses the development of the main mode,resulting in the main mode not being able to develop sufficiently large to generate the position changes of 3/1 islands.
文摘In this paper, a logistical regression statistical analysis (LR) is presented for a set of variables used in experimental measurements in reversed field pinch (RFP) machines, commonly known as “slinky mode” (SM), observed to travel around the torus in Madison Symmetric Torus (MST). The LR analysis is used to utilize the modified Sine-Gordon dynamic equation model to predict with high confidence whether the slinky mode will lock or not lock when compared to the experimentally measured motion of the slinky mode. It is observed that under certain conditions, the slinky mode “locks” at or near the intersection of poloidal and/or toroidal gaps in MST. However, locked mode cease to travel around the torus;while unlocked mode keeps traveling without a change in the energy, making it hard to determine an exact set of conditions to predict locking/unlocking behaviour. The significant key model parameters determined by LR analysis are shown to improve the Sine-Gordon model’s ability to determine the locking/unlocking of magnetohydrodyamic (MHD) modes. The LR analysis of measured variables provides high confidence in anticipating locking versus unlocking of slinky mode proven by relational comparisons between simulations and the experimentally measured motion of the slinky mode in MST.

文摘We will give an overview of results obtained by our reactive fluid model. It is characterised as a fluid model where all moments with sources in the experiment are kept. Furthermore, full account is taken for the highest moments appearing in unexpanded denominators also including full toroidicity. It has been demonstrated that the strength of zonal flows is dramatically larger in reactive fluid closures than in those which involve dissipation. This gives a direct connection between the fluid closure and the level of excitation of turbulence. This is because zonal flows are needed to absorb the inverse cascade in quasi 2D turbulence. This also explains the similarity in structure of the transport coefficients in our model with a reactive closure in the energy equation and models which have a reactive closure because of zero ion temperature such as the Hasegawa-Wakatani model. Our exact reactive closure unifies several well-known features of tokamak experiments such as the L-H transition, internal transport barriers and the nonlinear Dimits upshift of the critical gradient for onset of transport. It also gives transport of the same level as that in nonlinear gyrokinetic codes. Since these include the kinetic resonance this confirms the validity of the thermodynamic properties of our model. Furthermore, we can show that while a strongly nonlinear model is needed in kinetic theory a quasilinear model is sufficient in the fluid description. Thus our quasilinear fluid model will be adequate for treating all relevant problems in bulk transport. This is finally confirmed by the reproduction by the model of the experimental power scaling of the confinement time Te ~ P-2/3. This confirms the validity of our reactive fluid model. This also gives credibility to our ITER simulations including the H-mode barrier. A new result is here, that alpha heating strongly reduces the slope of the H-mode barrier. This should significantly reduce the effects of ELM's.
