Base editing, as an expanded clustered regularly interspaced short palindromic repeats(CRISPR)-Cas genome editing strategy, permits precise and irreversible nucleotide conversion. SaKKH, an efficient variant of a Cas9 ortholog from Staphylococcus aureus(SaCas9), is important in genome editing because it can edit sites with HHHAAT protospacer adjacent motif(PAM) that the canonical Streptococcus pyogenes Cas9(SpCas9) or its variants(e.g. xCas9, Cas9-NG) cannot. However, several technical parameters of SaKKH involved base editors have not been well defined and this uncertainty limits their application. We developed an effective multiplex cytosine base editor(SaKKHn-pBE) and showed that it recognized NNARRT, NNCRRT, NNGRGT, and NNTRGT PAMs. Based on 27 targets tested, we defined technical parameters of SaKKHn-pBE including the editing window, the preferred sequence context, and the mutation type. The editing efficiency was further improved by modification of the SaKKH sgRNA. These advances can be applied in future research and molecular breeding in rice and other plants. 展开更多

On the basis of theoretical analyses and calculations of high speed continuous impact force and tool notch surface temperature acted upon by burr and serrated chip edge, a notch wear model of low stress value and temperature impact fatigue was established. Saw-tooth-shaped burr and fin-shaped chip edge continuously impacts the rake face and flank face at high speed and high fre-quency, which results in a V-shaped notch wear. An experiment was done to validate that the saw-tooth-shaped burr does affect the notch wear. This model can be utilized to solve reasonably many problems that cannot be explained by any other theoretical assumptions. 展开更多

Case Based Reasoning(CBR)is one of the artificial intelligent methodologies that is widely used in problem solving by reusing the most similar previous experiences stored in the library.A framework of ECO-CBR for Life Cycle Assessment data collection has been used and the process was carried out using SolidWorks program.The practicality of the tool has been validated using case study,which then provides solution.The output enable researchers to determine forecast error and forecast accuracy,by valuing the calculation from Total Carbon Footprint,Energy Consumption,Air Acidification,and Water Eutrophication.ECO-CBR is able to assist designers in product design.Due to the limitation of environmental impact consideration in product sustainability,there is a demand to propose a tool that can assist designers to reduce environmental impact of product design at early stage.The model works as an essential guideline to lessen repeated mistakes in the product development process and help designers measure the risks before concluding ideal decisions.Minor errors that occur through the study showed that ECO-CBR is reliable to be implemented in order to find a better solution. 展开更多

It is a measure of the amount of algae growing in a waterbody and can be used to monitor the trophic condition of a waterbody. We studied the pre and post effects of marine ranch construction in Chl-a concentration in Zhelin Bay, Southern China using Normalized Difference Chlorophyll Index (NDCI) and a web-based tool (https://mapcoordinates.info/). We used 8 day composite MODIS image collections of 500 m resolution and randomly selected two stations to extract the chlorophyll-a concentration values through the web-based tool. We recorded the slight increase in NDCI values in all stations after the construction of marine ranch which is a good indicator of the marine organisms’ reproduction and survival. 展开更多

Utilization and Uptake of the UpToDate Clinical Decision Support Tool in Five Medical Schools in Uganda (August 2022-August 2023): A Partnership with the Better Evidence Program

Background: Clinical decision support tools provide suggestions to support healthcare providers and clinicians, as they attend to patients. Clinicians use these tools to rapidly consult the evidence at the point of care, a practice which has been found to reduce the time patients spend in hospitals, promote the quality of care and improve healthcare outcomes. Such tools include Medscape, VisualDx, Clinical Key, DynaMed, BMJ Best Practice and UpToDate. However, use of such tools has not yet been fully embraced in low-resource settings such as Uganda. Objective: This paper intends to collate data on the use and uptake of one such tool, UpToDate, which was provided at no cost to five medical schools in Uganda. Methods: Free access to UpToDate was granted through the IP addresses of five medical schools in Uganda in collaboration with Better Evidence at The Global Health Delivery Project at Harvard and Brigham and Women’s Hospital and Wolters Kluwer Health. Following the donation, medical librarians in the respective institutions conducted training sessions and created awareness of the tool. Usage data was aggregated, based on logins and content views, presented and analyzed using Excel tables and graphs. Results: The data shows similar trends in increased usage over the period of August 2022 to August 2023 across the five medical schools. The most common topics viewed, mode of access (using either the computer or the mobile app), total usage by institution, ratio of uses to eligible users by institution and ratio of uses to students by institution are shared. Conclusion: The study revealed that the tool was used by various user categories across the institutions with similar steady improved usage over the year. These results can inform the librarians as they encourage their respective institutions to continue using the tool to support uptake of point-of-care tools in clinical practice. 展开更多

针对数控机床电机座加工过程中存在的基准转换误差累积、装夹次数多及成组零件互换性差等问题,以T5.2数控车床Z轴电机座为研究对象,提出集成工艺优化与夹具创新的一体化解决方案。通过分析电机座结构特征及精度要求,确定以底面为统一基... 针对数控机床电机座加工过程中存在的基准转换误差累积、装夹次数多及成组零件互换性差等问题,以T5.2数控车床Z轴电机座为研究对象,提出集成工艺优化与夹具创新的一体化解决方案。通过分析电机座结构特征及精度要求,确定以底面为统一基准的工艺策略,结合时效处理与余量分级控制,优化粗、半精及精加工工序链。设计多工位铣削夹具、粗精镗孔一体化夹具及共基准组配夹具,实现关键特征(如轴承孔、定位端面)一次装夹加工,消除重复定位误差。工艺验证表明:采用共基准组配夹具后,成组电机座与轴承座中心高一致性误差≤0.005mm,安装修配时间缩短63.3%,互换合格率提升至98.3%;单件加工周期压缩50%以上,关键尺寸标准差降低66%~75%,设备利用率提升25%。研究结果为复杂箱体类零件的高效精密加工提供了理论依据与工程实践参考。 展开更多

Product customization is a trend in the current market-oriented manufacturing environment. However, deduction from customer requirements to design results and evaluation of design alternatives are still heavily reliant on the designer's experience and knowledge. To solve the problem of fuzziness and uncertainty of customer requirements in product configuration, an analysis method based on the grey rough model is presented. The customer requirements can be converted into technical characteristics effectively. In addition, an optimization decision model for product planning is established to help the enterprises select the key technical characteristics under the constraints of cost and time to serve the customer to maximal satisfaction. A new case retrieval approach that combines the self-organizing map and fuzzy similarity priority ratio method is proposed in case-based design. The self-organizing map can reduce the retrieval range and increase the retrieval efficiency, and the fuzzy similarity priority ratio method can evaluate the similarity of cases comprehensively. To ensure that the final case has the based on grey correlation analysis is proposed to evaluate similar cases best overall performance, an evaluation method of similar cases to select the most suitable case. Furthermore, a computer-aided system is developed using MATLAB GUI to assist the product configuration design. The actual example and result on an ETC series machine tool product show that the proposed method is effective, rapid and accurate in the process of product configuration. The proposed methodology provides a detailed instruction for the product configuration design oriented to customer requirements. 展开更多

Substations which are at the center of the electric power transformation from the power plant are changing to IEC 61850 based digital substations. Therefore, within substation, there is a growing demand for the IEC 61850 based Intelligent Electronic Devices (IEDs). The operation of multiple manufacturers of IEDs in a single digital substation network increases the need for IEC 61850 communications specification conformance diagnosis to ensure interoperability for efficient data exchange between IEDs. The IEC 61850-10 presents test items for diagnosing communication specification conformance. There are many test tools available in the market today to test the compliance of the IEC 61850 communications specifications to the IED. In this paper, we propose a model-based diagnostic method for IED communication conformance testing. The proposed model-based software therefore uses the “drag and drop” technique to select the various IEC 61850 communication services (objects) required to design the test case in a user friendly Graphical User Interface (GUI). This makes the service conformance testing more flexible for test engineers and system integrators especially in situations that require test case modifications. Objective:To assess the level of community capacity for dengue prevention and control and to study household environments and larval indices in southern Thailand.Methods:A crosssectional survey was designed for the study,enrolling two communities with higher dengue incidence rate than the standard over the past five years.Data gathering was conducted by the dengue leader group(DLG),including 15 leaders and 15 non-leaders trained by the research team.The dengue community capacity assessment tool(DCCAT) for leaders(115 items.14 domains) and non-leaders(83 items,11 domains).Participants were selected by the DLG based on their communities’ dengue risk.In the low-dengue incidence(LDI) community,32 leaders and 177 non-leaders were selected;while in the high-dengue incidence(HDI) community,31 leaders and 199 non-leaders were chosen.Results:The leaders from the LDI and the HDI communities demonstrated high levels of dengue community-capacity(360.47±58.82,416.22±57.72). Non-leaders in the LDI community demonstrated a moderate level of dengue community capacity competence(205.90±60.76),while the non-leaders in the HDI community had a high level (254.78±50.34).Conclusions:These initial levels of dengue community capacity serves as a baseline for diagnosing each community.For a community that needed to improve its capacity, the DCCAT is essential tool to conduct a pre-post intervention assessment or a serial assessment. A participatory approach is taken to enable local communities to carry out anti-dengue efforts on their own,rather than have intervention by an outsider. 展开更多

Cemented Carbide YG11C and Tool Steel Crl2MoV was blazed with Ni-base amorphous alloys, QG-1011, MBF-20 and MBF-75, using dynamics thermodynamics analogue testing machine Gleeble 1500D. The effects of brazing temperature, holding time and holding pressure on micro-structure and bond strength were investigated. Results showed that YG11C and Crl2MoV were all wetted well by these three Ni-base alloys, and the bond strength was as high as 220MPa, 320MPa, 320MPa respectively. When the blazing temperature was at the point over the melting point 60-70°C of Ni-base alloy, the holding time was about 2-10min, the suitable pressure was benefit for improving the brazing quality. Microanalysis showed Co in cemented carbide diffused into liquid brazing alloy and formed the Fe-Co solid .solution. 展开更多

Recent advancement in nuclear education learning has been through the use of computers and simulation related tasks such as the use of industry codes. Further enhancements in nuclear education are being considered through the use of distance learning technologies. The purpose of this work is to explore distance learning related tools to determine if they can provide an enhanced learning environment for nuclear education. In this work, a set of tools are examined that can be used to augment or replace the traditional lecture method. These tools are Mediasite, Adobe Connect, Elluminate, and Camtasia. All four tools have recording capabilities that allow the students to experience the exchange of information in different ways. This paper compares recent experiences with each of these tools in providing nuclear engineering education and assesses the various constraints and impacts on delivery through direct feedback from students and instructors. In general, the tools were found to be useful for mature students on the condition that the lecturer was comfortable with the tools and in some cases, adequate support from IT groups was provided. 展开更多

The impact force, impact pressure, and impact frequency of the saw-tooth-shaped chip edge on rake face and cutting edge at different speeds were calculated, and the influence of chip edge on notch wear formation was analyzed. Although the Model-Driven paradigm is being accepted in the research environment as a very useful and powerful option for effective software development, its real application in the enterprise context is still a challenge for software engineering. Several causes can be stacked out, but one of them can be the lack of tool support for the efficient application of this paradigm. This paper presents a set of tools, grouped in a suite named NDT-Suite, which under the Model-Driven paradigm offer a suitable solution for software development. These tools explore different options that this paradigm can improve such as, development, quality assurance or requirement treatment.
