BACKGROUND Thyroglobulin(Tg)is one of the markers of thyroid cancer, and its concentration may be elevated in patients with malignant thyroid tumors.Thyroid-stimulating hormone(TSH)is secreted by the pituitary gland, which has a significant impact on thyroid gland function.Excessively high or low TSH levels may be associated with an increased risk of thyroid cancer.Thus,in-depth studies on the association of serum Tg and TSH levels with thyroid cancer risk in patients with thyroid nodules are warranted. This can help determine whether Tg and TSH levels can predict the degree of malignancy of thyroid nodules, which can in turn guide doctors in making accurate diagnoses and treatment decisions.Furthermore, such studies can provide more accurate diagnostic methods for thyroid nodules and help patients become aware of the presence of thyroid cancer as early as possible, thereby improving the success rate of treatment and prognosis.AIM To investigate the association of serum Tg and TSH levels with the risk of thyroid cancer in patients undergoing thyroid nodule surgery.METHODS The clinical data and laboratory examination results of 130 patients who underwent thyroid nodule surgery were retrospectively analyzed.Furthermore, their preoperative serum Tg and TSH levels were recorded.Histopathological examination conducted during follow-up revealed the presence of thyroid cancer.Correlation analysis were used to analyze the association of Tg and TSH levels with the risk of thyroid cancer.RESULTS Of the 130 patients,60 were diagnosed with thyroid cancer.Statistical analysis revealed a significant positive correlation between serum Tg levels and the risk of thyroid cancer(P<0.05).This suggests that high serum Tg levels are associated with an increased risk of thyroid cancer. However,no significant correlation was observed between serum TSH levels and the risk of thyroid cancer(P>0.05).CONCLUSION In patients who underwent thyroid nodule surgery, serum Tg levels exhibited a significant correlation with the risk of thyroid cancer but serum TSH levels did not. These findings suggest that serum Tg can serve as an important biomarker for assessing the risk of thyroid cancer in these patients. 展开更多

Thyroid-stimulating hormone(TSH)is important for the thyroid gland, development, growth, and metabolism.Defects in TSH production or the thyrotrope cells within the pituitary gland cause congenital hypothyroidism(CH),resulting in growth retardation and neurocognitive impairment. While human TSH is known to display rhythmicity, the molecular mechanisms underlying the circadian regulation of TSH and the effects of TSH-thyroid hormone(TH)signaling on the circadian clock remain elusive. Here we show that TSH,thyroxine(T4),triiodothyronine(T3),and tshba display rhythmicity in both larval and adult zebrafish and tshba is regulated directly by the circadian clock via both E′-box and D-box.Zebrafish tshba^(−/−)mutants manifest congenital hypothyroidism, with the characteristics of low levels of T_(4)and T_(3)and growth retardation.Loss or overexpression of tshba alters the rhythmicity of locomotor activities and expression of core circadian clock genes and hypothalamic-pituitary-thyroid(HPT)axis-related genes.Furthermore,TSH-TH signaling regulates clock2/npas2 via the thyroid response element(TRE)in its promoter, and transcriptome analysis reveals extensive functions of Tshba in zebrafish.Together, our results demonstrate that zebrafish tshba is a direct target of the circadian clock and in turn plays critical roles in circadian regulation along with other functions. 展开更多

Introduction:The regulation of thyroid-stimulating hormone(TSH)synthesis involves neurotransmitters, with melatonin being a subject of ongoing debate.TSH transcription, synthesis, and secretion from the pituitary pars distalis(PD)is primarily regulated in a photoperiodic manner by thyrotropin-releasing hormone(TRH).In contrast,in the pituitary pars tuberalis(PT),mRNA transcription and alpha/beta chain synthesis, but not secretion,of a TSHlike product is regulated by melatonin.Conversely,non-photoperiodic melatonin might also affect the secretion of a TSH-like product from the PT.Nevertheless, the impact of exogenous melatonin on the underlying PD-TSH synthesis remains unclear.Casein kinase 1α(CK1α)plays a negative regulatory role in TSH synthesis in the mouse pituitary. Objective:We investigated whether non-photoperiodic melatonin affects PD-TSH synthesis through its interaction with CK1α.Methods:Immunohistochemistry and immunofluorescence staining detected the colocalization of the melatonin receptor MT1 with CK1αand TSH-βin the PD.RT-qPCR,western blotting, and ELISA revealed the effect of melatonin on Tshb mRNA,MTNR1A mRNA,Csnk1a1 mRNA,CK1αprotein,MT1 protein, and TSH levels. Results:Robust colocalization of the melatonin receptor MT1 with CK1αand TSH-βin the PD.Tshb mRNA and CK1αprotein expression levels peaked at opposite phases of the 24-h light:dark cycle.Exogenous melatonin administration promoted pituitary TSH synthesis, while concurrently inhibiting CK1αactivity.The upregulation of endogenous CK1αactivity in primary pituitary cells significantly blunted the melatonin stimulatory impact on Tshb mRNA and TSH levels.Mechanistically, the CK1αagonist pyrvinium abrogated melatonin-induced activation of p-PKC and p-CREB expression in vitro. Conclusion:The CK1α/PKC signaling pathway mediates the regulation of melatonin in pituitary TSH synthesis. We demonstrate an important theoretical and experimental basis for understanding the mechanism of endocrine system diseases caused by abnormal TSH synthesis in the pituitary. 展开更多

A simple and feasible potentiometric immunosensing platform based on enzymatic biocatalytic precipitation technique was designed for the sensitive detection of thyroid-stimulating hormone(TSH;a typical kind of biomarkers for thyroid carcinoma),using horseradish peroxidase(HRP)-loaded liposome for the signal amplification. To construct such an assay system,a sandwich-type immunoreaction was readily carried out on monoclonal anti-TSH capture antibody-coated electrode by using polyclonal antiTSH secondary antibody-conjugated HRP-loaded liposome.Accompanying the formation of sandwichtype immunocomplex, the carried liposome was lysed through the added Triton X-100 to release the entrapped HRP molecules, which catalyzed the oxidation of 4-chloro-1-naphthol to produce an insoluble and uncharged organic precipitation on the electrode surface, thereby causing the change of the local electrical potential. Two labeling protocols with and without the liposome were investigated for detection of target TSH,improved analytical features were achieved with HRP-entrapped liposome.Under optimal conditions, the potentiometric immunosensor had good responses for TSH detection within the linear range of 0.01-30 p IU/mL at a detection limit of 0.0067 p IU/mL.Good reproducibility, high specificity and long-time stability were acquired during the assay procedure.Importantly,a wellmatched accuracy between the potentiometric immunosensor and commercial human TSH ELISA kit was gave for the analysis of human serum samples. 展开更多

A wavelength-dependent three-dimensional(3D)superlocalization imaging method on gold nanoislands(GNIs)chip was developed as a supersensitive single-molecule thyroid-stimulating hormone(TSH)nanobiosensor.Scattered and fluorescent signals from gold nanoislands on the substrate and quantum dots(QDs)nanoprobes were simultaneously isolated and acquired within an evanescent field layer generated by total internal reflection(TIR)of incident light using a dual-view device. The 3D TIR fluorescence images of TSH-bound QDs on the GNIs were obtained using z-axis optical sectioning at 10nm intervals before/after immunoreaction to identify the optimal conditions for detection. The localized centroid position of QD nanoprobes and GNI were distinguished at a subdiffraction limit resolution using 3D Gaussian fitting to the point spread function. The QD TSH nanobiosensor using wavelength-dependent 3D TIR fluorescence-based single-molecule localization microscopy(3D TIRF-SLM)imaging technique showed an excellent detection limit of 90 yoctomoles(~54 molecules)and a wide linear dynamic range of 1.14 zmol/L-100 pmol/L for TSH.The detection sensitivity was about 4.4×10^(9)times higher than conventional enzyme-linked immunosorbent assay and could successfully quantify TSH in human serum. The wavelength-dependent 3D TIRF-SLM technique may emerge as a reliable platform for ultrahigh-sensitive nanobiosensors at the single-molecule level and early diagnosis with quantification of disease-related ultra-tracebiomolecules. 展开更多

Thyroid-stimulating hormone (TSH) assay is considered a decisive diagnostic method for thyroid disorders. The lack of different TSH immunoassay standardization often leads to misdiagnosis of subclinical hypothyroidism (SCH). This study aims to differentiate between two analytical platforms for TSH measurement, namely Abbott Laboratories and Snibe Diagnostics. This study employed 173 thyroid profile samples to detect SCH. Initially, the sample was run on Abbott and then by Snibe immunoassay. Statistical analysis showed a significant difference of 0.87 mIU/L (P < 0.001) between the TSH values of the two assays. The proposed study corroborates with previous literature by manifesting notable differences in the TSH measurements of two different assays. The findings revealed some insights regarding the differences between the two assays, which conclude that the diagnosis of SCH requires vigilance because of inadequate assay standardization. Thus, future research involving larger cohorts with standardization efforts is imperative to improve the reliability of TSH measurements using different analytical platforms. 展开更多

Objective:To study the effects of preconceptional thyroid-stimulating hormone(TSH)levels on antral follicle count(AFC)and pregnancy outcomes in a firstin vitro fertilization(IVF)/intracytoplasmic sperm injection(ICSI)cycle. Methods:A retrospective cohort study was conducted to explore the effects of preconceptional TSH levels on AFC and pregnancy outcomes in 1340 patients. The patients were divided into three groups:group 1,0.55≤TSH<2.5μIU/mL(n=868);group 2,2.5≤TSH<4.0μIU/mL(n=334);and group 3,TSH≥4.0μIU/mL(n=138).Results:There were significant differences in AFC(P=0.002),number of zygotes(P=0.008),and fertilization rate(P=0.015)among the three groups.Among the three groups, there were no obvious differences in live birth(P=0.559),high-quality embryo(P=0.108),or clinical pregnancy(P=0.939)rates.Binary logistics regression analysis showed that TSH correlated with AFC when TSH was≥4μIU/mL(P=0.033);however,TSH levels had no effect on live birth or clinical pregnancy rate. Conclusions:Although the preconceptional TSH level negatively related with AFC,number of zygotes, and fertility rate, especially when TSH was>4μIU/mL,TSH was not an independent factor on the live birth rate among fertile women undergoing their first IVF/ICSI-embryo transfer treatment. The effect of TSH levels on couples undergoing assisted reproductive technology requires further investigation. 展开更多

Background Little information about the current management of patients with thyroid-stimulating hormone (TSH)-secreting pituitary adenomas or about the usefulness of the somatostatin analogue octreotide was contained in the literature. This study aimed to report the efficacy and safety of the long-acting octreotide formulation in patients with TSH-secreting pituitary adenomas after incomplete surgery and octreotide treatment failure. Methods Fifteen patients with TSH-secreting pituitary adenomas (8 men and 7 women), who previously underwent incomplete surgical resection and/or adjuvant radiotherapy (n=12) and failure of octreotide treatment (n=15), followed between 2007 and 2010 in Beijing Tiantan Hospital were included in this study. All patients received 1- to 2-months of the long-acting octreotide formulation treatment after the above combination of treatment. Paired samples t-test was used to analysis the variables. Results After two-month duration of the long-acting octreotide formulation treatment, the mean serum free or unbound thyroxine (FT4) ((16.02±1.72) pmol/L) and free triiodothyronine (FT3) ((2.87±0.43) pmol/L) levels of 15 patients significantly decreased compared with those after octreotide-treatment (FT4, (35.36±7.42) pmol/L, P 〈0.001; FT3, (17.85±7.22) pmol/L, P 〈0.001). Mean TSH levels stayed in the normal range after the long-acting octreotide formulation treatment ((0.72±0.21) mUlL) and were significantly lower than the pretreatment value ((5.27±1.04) mUlL, P 〈0.001), post-surgery value ((3.37±0.31) mU/L, P 〈0.001) and post-octreotide-treatment value ((4.52±0.41) mU/L, P 〈0.001). In these patients with TSH-secreting pituitary adenomas there was no evidence of tachyphylaxis. Conclusion The long-acting octreotide formulation may be a useful and safe therapeutic tool to facilitate the medical treatment of TSH-secreting pituitary adenomas in patients who underwent incomplete surgery or need long-term somatostatin analog therapy. 展开更多

Background: Diabetes is a set of metabolic abnormalities typified by hyperglycemia believed to be associated with aberrant insulin secretion, insulin action, or both. Erectile dysfunction and infertility are globally prevalent complications among male diabetic patients. There is a need to understand the role of a male reproductive hormone in the etiology of infertility in diabetic patients. Objective: This study aims to determine the effect of diabetes mellitus on the Male reproductive hormone (testosterone) on diabetic patients attending the Urology unit of Federal University Teaching Hospital, Abakaliki, Ebonyi State, Nigeria and the effect of industrial sugar consumption in rats model. Materials and Methods: 30 samples (including repeats, n = 3) were collected from known diabetic patients and 30 (including repeats, n = 3) control-non diabetic healthy individuals. Five millilitres (5 ml) of blood was collected from the antecubital vein of each participant and placed into a plain container for serum testosterone measurement using the enzyme-linked immunoassay technique. Blood was also collected from the same subjects in a fluoride oxalate container for plasma enzymatic glucose estimation using a Randox Glucose oxidase kit. For in vivo rat model, the albino rats were grouped into 2 of five (5) rats. The control group (1) was only given normal rat food and water while the test group (2) was treated with 0.4 g of industrial sugar/mL/body weight in addition to rat food and water via oral intubation for 21 days. Blood samples were collected via ocular puncture into plain tubes for testosterone measurement as described above. Results: There is a significant (p Conclusion: In conclusion, data available from this study suggests that increased plasma glucose levels and excessive industrial sugar consumption are associated with decreased testosterone levels in both human and animal models respectively. The mechanism is yet unclear but it could be that DM alters conventional sperm parameters, spermatogenesis, and biosynthesis of testosterone and induces degenerative changes in the testis and epididymis and ejaculatory dysfunction. These mechanisms need to be investigated. 展开更多

BACKGROUND Noonan syndrome(NS)is an autosomal dominant, multisystem disorder with a prevalence of 1 in 1000-2500.Multiple etiologies have been proposed for short stature in NS,including resistance to growth hormone(GH)and GH deficiency(GHD).Irrespective of the presence of GHD,NS is a Food and Drug Administration-approved indication for recombinant-GH therapy.Few case reports of combined anterior pituitary hormone deficiency(CPHD)in NS have been reported.AIM To describe the clinico-biochemical characteristics of NS with CPHD and to assess the response to recombinant GH therapy.METHODS An ambispective case-control study was conducted to compare the clinicohormonal profile and response to recombinant-GH in pediatric patients with NS and CPHD and pediatric patients with NS but without CPHD.RESULTS Five children with NS and CPHD were compared to 6 patients with NS but without CPHD.The most common anterior pituitary hormone involvement in combination with GHD was adrenocorticotrophic hormone deficiency causing hypocortisolemia(n=3,60%),followed by hypogonadotropic hypogonadism and secondary hypothyroidism(n=1 each).Pituitary hypoplasia was seen in the magnetic resonance imaging of all patients with CPHD.Patients with NS and CPHD had lower standard deviation scores of height(-4.18 vs-2.52,P=0.009),bodyweight,and body mass index but a slightly better first year response to recombinant GH(9.2 vs 5.5,P=0.06).There were no differences in dysmorphisms and other anomalies between the two groups.Patients with NS and CPHD had a similar response to GH as patients with CPHD but without NS.One patient with NS and CPHD developed hypocortisolism after GH initiation.CONCLUSION Hypoplasia of the pituitary and GHD with involvement of other pituitary hormones may be seen in NS and may determine response to recombinant GH therapy. 展开更多

BACKGROUND Acute pancreatitis(AP)is a severe inflammatory condition of the pancreas that leads to significant morbidity and metabolic disturbances.Hypercalcemia,or elevated serum calcium levels,is a concerning complication that may contribute to kidney stone formation and exacerbate renal issues.Calcium homeostasis is regulated by hormones, such as parathyroid hormone(PTH),calcitonin, and vitamin D,which can be altered in AP.Understanding the interplay among hypercalcemia, kidney stones, and hormonal changes is crucial for improving management strategies and clinical outcomes in patients with AP.AIM To investigate the relationship between hypercalcemia and kidney stone formation in patients with AP,along with changes in calcium-regulating hormone levels.METHODS This study was conducted in 200 patients diagnosed with AP at Ningbo Urology and Nephrology Hospital. The participants underwent regular monitoring of serum calcium levels and kidney stone formation using imaging.Additionally, blood samples were analyzed to measure the levels of calcium-regulating hormones, including PTH,calcitonin, and vitamin D,at baseline and during the course of the disease.RESULTS Of the 185 patients who completed the study,31(16.8%)developed kidney stones during the 7-day follow-up period.Hypercalcemia occurred in 53 patients(28.6%),with a peak incidence on day 3.Among hypercalcemic patients,41.5%developed kidney stones compared to 6.8%in normocalcemic patients(odds ratio=9.67,95%confidence interval:4.12-22.68,P<0.001).A significant positive correlation was found between hypercalcemia and kidney stone formation(P<0.001).Elevated PTH levels were noted in 60%of the patients with hypercalcemia, indicating a strong association between increased PTH levels, hypercalcemia, and kidney stone formation.CONCLUSION This study highlights the complex interplay between AP,calcium metabolism, and renal complications, providing insights that may guide future therapeutic strategies. 展开更多

Growth hormone-secreting pituitary adenomas(GHPAs)cause acromegaly,a condition characterized by persistent excess of growth hormone(GH)and its target hormone,insulin-like growth factor 1(IGF-1).This hormonal imbalance gives rise to diverse comorbidities, ultimately resulting in a shortened lifespan compared to the general population(Piccard et al.,2012).Extensive research has demonstrated that tumors exert a pivotal promoting effects on inflammation, and conversely, inflammatory responses exert reciprocal influence on various biological processes of tumors(Fridlender et al.,2009;Piccard et al.,2012).The microbiome profoundly impacts brain function, behavior, and neuroendocrine responses to stress.Through the hypothalamic-pituitary-adrenal(HPA)axis,the neuroendocrine system modulates the composition and permeability of the gastrointestinal tract, indicating a bidirectional network between gut microbiome and neuroendocrine system(Farzi et al.,2018).In the present study,we explored the microbiome profile in patients with GH adenomas and its potential role in inflammation, utilizing Mendelian randomization(MR)to reveal the casual relationship between microbiome and inflammation markers. 展开更多

Prostate cancer(PCa)is a prevalent malignancy in men, traditionally linked to androgen receptor signaling.Emerging evidence suggests thyroid hormones(THs,particularly T3/T4)play a complex role in PCa biology.THs regulate gene transcription via nuclear receptors TRα/β,modulating proliferation, apoptosis, and AR signaling, while non-genomic pathways through integrin αvβ3 activate MAPK/PI3K-Akt signaling, driving metabolic reprogramming, migration, and angiogenesis.Local DIO enzymes fine-tune T3/T4 levels, with DIO2 enhancing proliferation and DIO3 creating a low-TH microenvironment to facilitate immune evasion.Epidemiological studies associate hyperthyroidism or low TSH with elevated PCa risk, whereas experimental models show inconsistent effects, reflecting regulation by hormone levels, receptor distribution, and tumor molecular features.Bibliometric analyses reveal a shift from epidemiological studies to molecular, immune, and metabolic mechanistic research, though clinical translation remains limited. This review synthesizes current knowledge on THs in PCa,highlighting mechanistic insights, evidence gaps, and future directions, aiming to inform early detection, stratification, and therapeutic strategies. 展开更多

Drought is one of the important stress factors affecting the growth and development processes of wheat in China's arid zones, which severely limits the yield. This study examined the impact of deficit irrigation on the flag leaf protection system and yield of drip-irrigated spring wheat during the growth stages in arid zones. In addition, this study aimed to determine the optimal water supply mode for efficient production under drip irrigation conditions and to provide technical support for water-saving and high-yield cultivation of drip-irrigated wheat. The experiment was conducted with a split plot design using the water-sensitive variety Xinchun 22(XC22) and the drought-tolerant variety Xinchun 6(XC6) as the main plots, while a fully irrigated control(CK, 75–80% FC, where FC is field water holding capacity), mild deficit(T1, 60–65% FC) and moderate deficit(T2, 45–50% FC) at the tillering stage, and mild deficit(J1, 60–65% FC) and moderate deficit(J2, 45–50% FC) at the jointing stage were used as the subplots. Systematic studies were conducted on the regulatory effects of deficit irrigation during the tillering and jointing stages on protective substances, membrane lipid metabolism, endogenous hormones in the flag leaf, and yield of spring wheat. Compared with treatments T2 and J2, treatments T1 and J1 were beneficial for increasing the activities of superoxide dismutase(SOD), peroxidase(POD), and catalase(CAT), the levels of proline(Pro), indole-3-acetic acid(IAA), and zeatin riboside(ZR), and the ratios IAA/abscisic acid(ABA), ZR/ABA, IAA/ZR, and(IAA+ZR)/ABA, while reducing the levels of hydrogen peroxide(H2O2), superoxide anion radicals(O2–·), malondialdehyde(MDA), phosphatidic acid(PA), free fatty acids(FFA), ABA, phospholipase D(PLD), and lipoxygenase(LOX), alleviating flag leaf senescence, and increasing yield. Under treatment T1, the SOD, POD, CAT, and Pro levels of flag leaves in XC6 were 11.14, 8.08, 12.98, and 3.66% higher than those of treatment CK, and under treatment J1, they were 6.43, 4.49, 7.36, and 2.50% higher than those of treatment CK. Under treatment T1 in XC6, the IAA, ZR level of the flag leaf, spike number, grains per spike, 1,000-grain weight and yield were 10.50, 5.79, 3.10, 8.84, 3.78, and 10.52% higher than those of treatment CK, and under treatment J1, they were 5.36, 3.94, 2.40, 3.72, 1.37, and 4.46% higher than those of treatment CK. Compared with XC22, XC6 was more conducive to the improvement of flag leaf protective substances, IAA, ZR, dry matter weight, yield components and yield. The correlation analysis showed significant positive correlations between IAA and ZR with SOD, POD, CAT, proline, and yield. IAA and ZR promoted the enhancement of protective enzyme activities, thereby clearing reactive oxygen species to cope with the oxidative stress caused by drought and achieve the effect of delaying senescence. Principal component analysis showed that yield components and dry matter weight, had direct effects on yield. Mild deficiency during the tillering stage without water stress in other stages could effectively optimize yield components, not only achieving high yield while increasing protective substances, but also reducing the reactive oxygen species content. This strategy can be recommended as a water-saving and high-yield production mode for drip irrigation of spring wheat in Xinjiang, China. 展开更多
