Therapeutic effects of Pueraria lobata(Willd.)Ohwi root and Hovenia dulcis Thunb.extracts on alcoholic liver disease:Network pharmacology and experimental validation 被引量:1

Objective:To investigate the protective effects of the combined concentrated liquid extract of Pueraria lobata(Willd.)Ohwi root(P.lobata,Ge Gen)and Hovenia dulcis Thunb.(H.dulcis,Zhi Ju Zi)against ethanol-induced liver damage in vitro,using a human hepatoma cell line G2(HepG2)cell model.Methods:HepG2 cells were cultured in medium containing 4%ethanol to establish a model of alcoholic liver damage.The cells were then treated with the combined extract obtained via cryogenic extraction.Biochemical assays and Western blot analyses were performed to assess the levels of oxidative stress markers,antioxidant enzymes,and inflammatory cytokines.In addition,activation of the phosphoinositide 3-kinase/protein kinase B(PI3K/AKT)pathway was examined to elucidate the mechanisms underlying the effects of the extract.Results:Treatment with the extract contributed to a significant reduction in the release of nitric oxide and reactive oxygen species in the ethanol-treated HepG2 cells;promoted the elevated expression of superoxide dismutase,catalase,and glutathione,indicating enhanced antioxidant defenses;and showed strong free radical-scavenging activity against 1,1-diphenyl-2-picrylhydrazyl radicals.In addition,by activating the PI3K/AKT pathway,treatment promoted increases in the expression of nuclear factor erythroid 2-related factor 2 and its downstream targets,subsequently inhibiting apoptosis.Moreover.inflammatory responses were mitigated,as indicated by reductions in the expression of tumor necrosis factor-alpha and interleukin-6,and we detected reduction in the levels of alanine aminotransferase and aspartate aminotransferase,thereby indicating hepatoprotective effects.Conclusion:The combined P.lobata root and H.dulcis extract was established to have notable antioxidative and anti-inflammatory properties,effectively alleviating ethanol-induced liver damage in vitro.These findings highlight the potential applicability of this extract as a candidate for treating alcoholic liver disease. 展开更多

In the context of a surging demand for functional foods,this study utilized Sophora japonica L.(SL)and Rosa rugosa Thunb.(RT),which are rich in polyphenols(with flavonoids as the core subclass).High-purity extracts(SLE and RTE)were obtained through ethanol reflux extraction and macroporous resin purification,and then formulated with maltodextrin and erythritol to prepare a composite solid beverage.This beverage exhibited excellent antioxidant capabilities.At a concentration of 1 mg/mL,the scavenging rates of DPPH,ABTS,and hydroxyl radicals reached 82.4%,94.6%,and 49.2%,respectively.Network pharmacology indicated that quercetin andβ-sitosterol could modulate lipid metabolism pathways.Moreover,the beverage showed potential for lipid-lowering.Its cholate adsorption capacity was 589.4±2.9 mg/g at pH 7.0,and the IC50 value for pancreatic lipase inhibition was 32.55 mg/mL.However,a 60-day storage stability test revealed that the moisture content approached 5%,likely due to polyphenol-flavonoid reactivity,extending dissolution time to 30.88 s.These changes were attributed to polyphenols(with flavonoids as the core active subclass,and non-flavonoids such as phenolic acids as auxiliary),resulting in color alterations and reduced solubility.This study confirmed the dual functions of the SL-RT beverage in antioxidant and lipid-lowering aspects.Nevertheless,it also pointed out the need to optimize the formula and process to enhance stability,providing an important basis for the development of stable functional beverages. 展开更多

Objective:To evaluate the quality of Lygodium japonicum(Thunb.)Sw.(L.japonicum,Hai Jin Sha)by comparing its components without stewed(W)and stewed(S)using ultra-high-performance liquid chromatography(UHPLC)and chemometric analysis.Additionally,network pharmacology was employed to investigate the possible mechanisms of action of L.japonicum in the urinary calculi(UC)treatment.Methods:A fingerprinting method was established to identify components through UHPLC-tandem mass spectrometry.Chemometric techniques were used to compare the L.japonicum extraction methods.Furthermore,various network pharmacological approaches were used to identify and analyze the potential targets of the identified components in relation to UC.Results:The W and S extracts were distributed into two distinct clusters.Significant differences in the levels of protocatechuic aldehyde,caffeic acid,and p-coumaric acid were observed between S and W.Network pharmacology analysis revealed that the primary targets of L.japonicum in the UC treatment were serum albumin and epidermal growth factor receptors,with potential active components including protocatechuic acid and caffeic acid.Conclusion:This study comprehensively examined the therapeutic components of L.japonicum before and after boiling,shedding light on its potential mechanisms of action in UC treatment.These findings offer valuable insights into the development and utilization of L.japonicum resources. 展开更多

Soil salinization is a major abiotic stress that hampers plant development and significantly reduces agricultural productivity,posing a serious challenge to global food security.Akebia trifoliata(Thunb.)Koidz,a species within the genus Akebia Decne.,is valued for its use in food,traditionalmedicine,oil production,and as an ornamental plant.Curcumin,widely recognized for its pharmacological properties including anti-cancer,anti-neuroinflammatory,and anti-fibrotic effects,has recently drawn interest for its potential roles in plant stress responses.However,its impact on plant tolerance to saline-alkali stress remains poorly understood.In this study,the effects of curcumin on saline-alkali resistance in A.trifoliata were examined by subjecting plants to a saline-alkali solution containing 150 mmol/L sodium ions(a mixture of Na_(2)SO_(4),Na_(2)CO_(3),and NaHCO_(3)).Curcumin treatment under these stress conditions leads to anatomical improvements in leaf structure.Furthermore,A.trifoliatamaintained a favorable Na^(+)/K^(+)ratio through increased potassium uptake and reduced sodium accumulation.Biochemical analysis revealed elevated levels of proline,soluble sugars,and soluble proteins,along with improved activities of antioxidant enzymes such as superoxide dismutase(SOD),catalase(CAT),and peroxidase(POD).Similarly,the concentrations of hydrogen peroxide(H_(2)O_(2))and malondialdehyde(MDA)were significantly reduced.Transcriptome analysis under saline-alkali stress conditions showed that curcumin influenced seven keymetabolic pathways annotated in the Kyoto Encyclopedia of Genes and Genomes(KEGG)database,with differentially expressed unigenes primarily enriched in transcription factor families such as MYB,AP2/ERF,NAC,bHLH,and C2C2.Moreover,eight differentially expressed genes(DEGs)associated with plant hormone signal transduction were linked to the auxin and brassinosteroid pathways,critical for cell elongation and plant growth.These findings indicate that curcumin increases saline-alkali stress tolerance in A.trifoliata by modulating physiological,biochemical,and transcriptional responses,ultimately supporting improved growth under adverse conditions. 展开更多

Based on the meteorological data of 24 automatic stations in Xinfeng County,combined with the climatic conditions for the growth of Morinda officinalis F.C.How and Sarcandra glabra(Thunb.)Nakai,the climate suitability zoning indicators of M.officinalis and S.glabra in Xinfeng County were constructed by relevant statistical methods,and the climate suitability zoning indicators were divided into four grades:most suitable,suitable,sub-suitable and unsuitable.According to the results of expert scores,the relevant growth impact factors were assigned weight values.Based on geographic information system(GIS),the climate suitability zoning of M.officinalis and S.glabra planting in Xinfeng County was carried out.The results show that most areas of Xinfeng County were suitable for planting M.officinalis and S.glabra.Among them,the most suitable planting areas of M.officinalis were mainly distributed in most of Huilong Town,from Jiangnan Community of Fengcheng Street to Yuntianhai area of Meikeng Town,Matouwantian Village and areas to the south,and areas to the south of Zhutong Village,Fengcheng Street.The most suitable areas for S.glabra planting were mainly distributed in Puchang Village of Huilong Town,the northeast of Huangzhai Town,the south of Fengcheng Street,and the central-southern part of Matou Town.The zoning results provide a scientific basis for the rational layout of M.officinalis and S.glabra planting in Xinfeng County. 展开更多

The dark blue pigment(DBP)is a health ingredient from Vaccinium bracteatum Thunb.leaves used as a functional food supplement.However,the details of transepithelial absorption on the intestinal epithelial cells are barely understood.This study aimed to clarify the absorption properties of DBP in the Caco-2 cell monolayer model and evaluate the effect on the endo-metabolism and barrier function of Caco-2 cells.The results showed that the DBP did not show the dose-dependent toxic effect to Caco-2 cells between 0.25 and 1.5 mg/mL,which did not cause disorder in the normal cell metabolism and absorption activity.The Caco-2 cell monolayer model could absorb DBP by passive and active transport,and the absorptive pattern was dosedependent when the concentration was more than 0.25 mg/mL.During DBP absorption,an increase in m RNA and protein expressions of glucose transporters demonstrated that the glucose transporters were the potential transporter of DBP.But the glucose transport amounts were significantly lowered after 30 min of DBP treatment,indicating that DBP owned the inhibitory effect on glucose transportation.Furthermore,DBP also owned protective effects on the barrier function of intestinal epithelial cells. 展开更多

Objective:To elucidate the anti-melanogenic potential of Carex pumila Thunb.extract(FBCC-EP850).Methods:A collection of 180 plant extracts was tested for inhibition of mushroom tyrosinase activity using an in vitro assay.Among them,FBCC-EP850 exhibited the most promising inhibitory activity.Further analysis was conducted to investigate its mechanisms and therapeutic potential in reducing melanogenesis in B16F10 melanoma cells and zebrafish larvae.Results:FBCC-EP850 inhibited mushroom tyrosinase activity in a dose-dependent manner,with a half-maximal inhibitory concentration of 45.83μg/mL.FBCC-EP850 at concentrations up to 50μg/mL demonstrated minimal cytotoxicity against B16F10 melanoma cells and no adverse effects on zebrafish larvae.Treatment with 50μg/mL of FBCC-EP850 significantly reducedα-melanocyte stimulating hormone-induced melanin production and suppressed cellular tyrosinase activity in B16F10 melanoma cells.Additionally,FBCC-EP850 at 25 and 50μg/mL effectively diminished hyperpigmentation inα-melanocyte stimulating hormone-stimulated zebrafish larvae.Its anti-melanogenic action could be attributed to modulation of the cAMP-CREB-MITF signaling pathway.Conclusions:Carex pumila extract can inhibit melanogenesis by modulating the cAMP-CREB-MITF signaling pathway,which can be used as a promising candidate for treating hyperpigmentation disorders. 展开更多

The analysis of chloroplast gene characteristics in Alpinia japonica(Thunb.)Miq.is of great significance for developing relevant genetic resources.The high-throughput sequencing and bioinformatic research were performed to analyze the chloroplast genome characteristics of A.japonica.The total chloroplast genome length of A.japonica was 161,906 bp,with a typical circular tetrameric structure.And 133 genes were annotated,comprising 87 protein-coding,38 tRNA,and 8 rRNA genes.Furthermore,22 genes contained two copies,and 18 genes owned introns.Repeat sequence analysis showed that it contains 321 simple sequence repeats(SSRs)and 37 long segment repeats.Compared with the chloroplast genomes of eight representative plants in the genus Alpinia,the gene structure,type,and quantity were relatively conservative.Rps12 was the highest variation site in the entire chloroplast gene.A phylogenetic tree showed that the genus Alpinia was the most closely related to the genus Amomum.Meanwhile,A.japonica is the most closely related to Alpinia chinensis belonging to the genus Alpinia.Overall,the chloroplast genome of a new species was reported in the genus Alpinia,and a basis was provided for the utilization of Alpinia plants as a medical resource. 展开更多

[Method] HP-5MS capillary columns were used and column temperature was controlled by a program. MS analysis was performed with EI and quadruple mass analyzer. The volatile constituents in L. japonica Thunb. were identified by NIST02 and Wiley275 libraries, and their relative contents were determined with chromatographic peak area normalization method. [Result] According to GC-MS total ion-current chromatograms, 35 volatile constituents were identified in L. japonica Thunb. from Guangxi Zhuang Autonomous Region, mainly including methyl linolenate, n-hexadecanoic acid and ζ-muurolene; 18 volatile constituents were identified in L. japonica Thunb. from Hunan Province, mainly including n-hexadecanoic acid, linoleic acid and α-curcumene. 大叶苦丁茶(Ilex latifolia Thunb.)含有丰富的咖啡酰奎宁酸。在本研究中,运用胶束电动毛细管色谱法,测定了在不同发育时期的大叶苦丁茶不同器官中的咖啡酰奎宁酸含量。大叶苦丁茶的不同器官在不同发育时期的咖啡酰奎宁酸含量有着显著... 大叶苦丁茶(Ilex latifolia Thunb.)含有丰富的咖啡酰奎宁酸。在本研究中,运用胶束电动毛细管色谱法,测定了在不同发育时期的大叶苦丁茶不同器官中的咖啡酰奎宁酸含量。大叶苦丁茶的不同器官在不同发育时期的咖啡酰奎宁酸含量有着显著的变化。在叶片中,10天叶龄的叶的咖啡酰奎宁酸含量是19.78 g/100 g,而当叶龄达到12月时,其中的咖啡酰奎宁酸含量已经降为1.41 g/100 g。随着发育时间的增长,雌花、雄花中的含量变化也有类似的变化。雌花(果)和雄花在发育期为10天时的咖啡酰奎宁酸含量高:分别20.29 g/100 g和18.06 g/100g,随后在雌花发育成的7个月果实中下降到0.16 g/100 g,而在40天期接近脱落的雄花中其含量降为10.36 g/100 g。大叶苦丁茶的器官中,除了嫩叶,雄花由于高含量的咖啡酰奎宁酸和较多的生物量,也是一种好的咖啡酰奎宁酸来源材料。本文首次报道了大叶苦丁茶的花中含有大量的咖啡酰奎宁酸。MEKC方法也是首次被用于苦丁茶中的咖啡酰奎宁酸类成分的测定,使用这种方法,大叶苦丁茶中的6种咖啡酰奎宁酸成分在40分钟内完全分离。相较于以前文献中报道的高效液相色谱法,本文为苦丁茶中的咖啡酰奎宁酸的质量控制提供了一种新的检测方法。 展开更多

A sensitive and specific reversed-phase high performance liquid chromatography (RP-HPLC) method was established for the simultaneous quantitative determination of eight active components, two stilbenes (resveratrol, polydatin) and six flavonoids (rutin, quercilrin, quercetin, luteotin, isoorientin, kaempferol), in Polygonum multiflorum Thunb. The chromatographic separation was performed on a Kromasil C18 column (250 mm^4.6 mm, 5 ~tm), and gradient elution was carried out with water-methanol as the mobile phase at a flow rate of 1 mL/min. Stilbenes and flavonoids were respectively detected at 320 nm and 350 nm with DAD. The correlation coefficients of all the calibration curves were found to be higher than 0.9995. The average recoveries were ranged from 96.8% to 102.5% with RSD less than 4.8% for these components. RP-HPLC was validated to be a robust method for the quantitative determination of active components in Polygonum multiflorum Thunb, and it could be used in the quality control of this traditional medicine. 展开更多

[Objective] The aim was to study the variation of leaf characters from different provenance sources of Polygonum multiflorum Thunb,as well as to carry out cluster analysis on P.multiflorum from different provenance sources to provide basis for the classification,identification,breeding and improved variety selection of P.multiflorum.[Method] Leaf shape characters of 31 copies of germplasm resources in the major distribution region of the whole country were determined,and the genetic variation of P.multiflorum leaves from different producing areas was analyzed.[Result] The leaf characters of single plant of the same experimental provenance source of P.multiflorum were relatively stable,the variation was mainly found on the single leaf area,1/2 leaf width,leaf width and other indicators;the variation of each leaf character among different provenance sources was obvious,and the variation was mainly found on the single leaf weight,leaf area,1/2 leaf width,leaf length and other indicators.The correlation analysis of each leaf character in P.multiflorum suggested that the single leaf area and single leaf weight showed extremely significant positive correlation with leaf length,1/2 leaf width,leaf width,leaf thickness and leaf stem length,while the single leaf area and single leaf weight showed significant negative correlation with WWR(leaf width/1/2 leaf width)and LWR(leaf length/1/2 leaf length),in addition,several macroscopic leaf characters such as leaf length,1/2 leaf width,leaf width,leaf stem length showed extremely positive correlation.The main component analysis result suggested that the contribution rate of accumulation variance of the front three main components was up to 97.4%,which could better reflect the comprehensive performance of leaf characters of different provenance sources of P.multiflorum.The cluster analysis showed that the experimental 31 copies of P.multiflorum provenance sources should be divided into three classes,the first class was distributed in the Middle,Western of Guizhou,northwestern of Guangxi and western areas with higher altitude;the second class was distributed in Hunan,Hubei,Sichuan,Guangdong and the most area of Guangxi;the third class was distributed in Anhui,Jiangsu and Henan and Shandong.[Conclusion] Cluster analysis of leaf characters indicated that the kinds of provenance sources which the geographical position was closer could be got together.The study had provided a certain basis for the classification of P.multiflorum. 展开更多

[Objective] The aim was to explore the effects of different factors on rapid propagation and seedling survival of Lonicera japonica Thunb. [Method] The cuttings of 1, 2 or 3 years old, with different length, were soaked in different hormones at different concentrations for different time and planted at different dates. Finally, their rooting rate and survival rate in different treatments were measured and compared. [Result] The rooting rate and survival rate of one-year old cuttings with length of 10-13 cm and planted on September 20 and October 20, 2012 were relatively higher among all the treatments. Hormone treatment could significantly promote the callus formation and accelerate the rooting of the cuttings, and improve their survival rate. The cuttings treated with 120-200 mg/L ABT1 or NAA for 35-45 min had higher rooting rate and survival rate. Among them, the survival rate of cuttings treated with 160 mg/L ABT1 for 35 min was the highest, up to 92.5%. [Conclusion] The study provided basis for improving the survival rate of L. japonica by cuttage, and for optimizing the rapid propagation technique. 展开更多

[Methods] SDS-PAGE and Western blot analysis were employed to detect expression of exogenous antimicrobial peptide in transgenic H. cordata plants. Both wild type and transgenic H. cordata plants were inoculated with different concentrations of Rhizoctonia solani spores for detecting their resistance. [Results] The exogenous antimicrobial peptide was detected at translation level. The optimal parameters for detecting the resistance of transgenic H. cordata plants to R. solani was inoculation of spores at a concentration of 3×105 ind./ml and cultured for three days. The results showed that resistance of transgenic H. cordata plants to R. solani was enhanced in comparison with CKs. [Conclusion] Expression of exogenous antimicrobial peptide can enhance the resistance of transgenic H. cordata plants to stem rot disease. 展开更多

Lour.under single and combined stress of Pb and Cd.[Method] Pot experiment was adopted with O.fragrans seedlings as experimental materials to investigate the effect of single and combined stresses of Pb and Cd on the growth of O.fragrans seedlings under 0,50,100,500,1 000 and 2 000 mg/kg of Pb and 0,1,10,50,100 and 200 mg/kg of Cd.[Result] Under the experimental conditions,the growth of O.fragrans seedlings was significantly inhibited under moderate Pb stress(500 mg/kg) and high Cd stress(100 mg/kg);the decreasing order of Pb uptake rate of different O.fragrans tissues was root stem leaf;the decreasing order of Cd content of different O.fragrans tissues was root stem leaf,while the decreasing order of Cd uptake rate of different O.fragrans tissues was stem root leaf.[Conclusion] Under combined stress,low concentrations of Pb and Cd can mutually promote the uptake and accumulation,while high concentrations of Pb and Cd can mutually inhibit the uptake and accumulation. 展开更多

[Objective] This study was to explore the effect of three precursors on the accumulation of principal volatile oil constituents in tissue culture plantlets of Atractylodes lancea (Thunb.) DC. with the aim to provide references for the improvement of artificial cultivated A. lancea quality. [Method] Three precursors were added into the MS rooting medium for A. lancea tissue culture plantlets and the volatile oil was extracted by ultrasonication after cultured for several days. The content of atractylon, atractylol, β-eucalyptol and atractydin in the volatile oil were determined by using gas chromatography method. [Result] The addition of xylose, isoprene and tetrahydrofuran impacted the growth indicators, yield of volatile oil and relative percentage content of the four constituents of A. lancea tissue culture plantlets. In the 6 g/L xylose optimized medium, the atractylon and β-eucalyptol content reached up to 4.23% and 56.34%, respectively, 1.41% and 1.66% higher than the control; although the addition of isoprene into medium raised the atractylon content, the accumulation of total volatile oil was inhibited that it decreased by 23.67%, 31.06% and 7.10% to the control; for the tetrahydrofuran optimized medium, the content of atractylon, atractylol and atractydin all increased, and the total volatile oil content increased by 49.97% to the control when the concentration of tetrahydrofuran was 0.07 g/L. [Conclusion] The addition of xylose and tetrahydrofuran promoted the accumulation of principal constituents of the volatile oil, whereas the addition of isoprene inhibited the accumulation. 展开更多

青藤是一种重要的中药材,具有较高的药用价值。目前,对于青藤的分子生物学研究极为有限,基因资源比较匮乏。全基因组的破译将为其分子生物学研究的深入开展提供坚实的基础,而对青藤进行全基因组调查以明确其基因组特征可以为全基因测序... 青藤是一种重要的中药材,具有较高的药用价值。目前,对于青藤的分子生物学研究极为有限,基因资源比较匮乏。全基因组的破译将为其分子生物学研究的深入开展提供坚实的基础,而对青藤进行全基因组调查以明确其基因组特征可以为全基因测序方案的制定提供依据。本研究基于Illumina测序平台测定了青藤基因组,结合生物信息学方法得到基因组大小、杂合率、重复序列等特征,并采用SOAPdenovo软件对基因组进行初步组装。过滤得到168.72 G有效数据,青藤基因组大小估计为829.72 Mb;重复序列比率为51.72%,杂合率为0.48%,GC含量为34.57%;K-mer=35进行初步组装得到contigs N50为162 bp,总长为621 033 264 bp,scaffolds N50为489 bp,总长为780 536 487 bp。鉴于青藤基因组高重复序列的特点,其全基因测序可采用Illumina测序技术和Pac Bio测序技术相结合的策略,并采用Hi-C技术以及其相应的组装软件,以此获得青藤全基因组图谱。 展开更多
