Cartilage tissue engineering offers hope for tracheal cartilage defect repair.Establishing an anti-inflammatory microenvironment stands as a prerequisite for successful tracheal cartilage restoration,especially in imm...Cartilage tissue engineering offers hope for tracheal cartilage defect repair.Establishing an anti-inflammatory microenvironment stands as a prerequisite for successful tracheal cartilage restoration,especially in immunocompetent animals.Hence,scaffolds inducing an anti-inflammatory response before chondrogenesis are crucial for effectively addressing tracheal cartilage defects.Herein,we develop a shell–core structured PLGA@ICA-GT@KGN nanofilm using poly(lactic-co-glycolic acid)(PLGA)and icariin(ICA,an antiinflammatory drug)as the shell layer and gelatin(GT)and kartogenin(KGN,a chondrogenic factor)as the core via coaxial electrospinning technology.The resultant PLGA@ICA-GT@KGN nanofilm exhibited a characteristic fibrous structure and demonstrated high biocompatibility.Notably,it showcased sustained release characteristics,releasing ICA within the initial 0 to 15days and gradually releasing KGN between 11 and 29days.Subsequent in vitro analysis revealed the potent anti-inflammatory capabilities of the released ICA from the shell layer,while the KGN released from the core layer effectively induced chondrogenic differentiation of bone marrow stem cells(BMSCs).Following this,the synthesized PLGA@ICA-GT@KGN nanofilms were loaded with BMSCs and stacked layer by layer,adhering to a‘sandwich model’to form a composite sandwich construct.This construct was then utilized to repair circular tracheal defects in a rabbit model.The sequential release of ICA and KGN facilitated by the PLGA@ICA-GT@KGN nanofilm established an anti-inflammatory microenvironment before initiating chondrogenic induction,leading to effective tracheal cartilage restoration.This study underscores the significance of shell–core structured nanofilms in temporally regulating anti-inflammation and chondrogenesis.This approach offers a novel perspective for addressing tracheal cartilage defects,potentially revolutionizing their treatment methodologies.展开更多
The ability to control mammalian cells such that they self-organize or enact therapeutic effects as desired has incredible implications.Not only would it further our understanding of native processes such as developme...The ability to control mammalian cells such that they self-organize or enact therapeutic effects as desired has incredible implications.Not only would it further our understanding of native processes such as development and the immune response,but it would also have powerful applications in medical fields such as regenerative medicine and immunotherapy.This control is typically obtained by synthetic circuits that use synthetic receptors,but control remains incomplete.The synthetic juxtacrine receptors(SJRs)are widely used as they are fully modular and enable spatial control,but they have limited gene expression amplification and temporal control.As these are integral facets to cell control,I therefore designed transcription factor based amplifiers that amplify gene expression and enable unidirectional temporal control by prolonging duration of target gene expression.Using a validated in silico framework for SJR signaling,I combined these amplifiers with SJRs and show that these SJR amplifier circuits can direct spatiotemporal patterning and improve the quality of self-organization.I then show that these circuits can improve chimeric antigen receptor(CAR)T cell tumor killing against various heterogenous antigen expression tumors.These amplifiers are flexible tools that improve control over SJR based circuits with both basic and therapeutic applications.展开更多

The user control over the life cycle of data is of an extreme importance in clouds in order to determine whether the service provider adheres to the client’s pre-specified needs in the contract between them or n...The user control over the life cycle of data is of an extreme importance in clouds in order to determine whether the service provider adheres to the client’s pre-specified needs in the contract between them or not, significant clients concerns raise on some aspects like social, location and the laws to which the data are subject to. The problem is even magnified more with the lack of transparency by Cloud Service Providers (CSPs). Auditing and compliance enforcement introduce different set of challenges in cloud computing that are not yet resolved. In this paper, a conducted questionnaire showed that the data owners have real concerns about not just the secrecy and integrity of their data in cloud environment, but also for spatial, temporal, and legal issues related to their data especially for sensitive or personal data. The questionnaire results show the importance for the data owners to address mainly three major issues: Their ability to continue the work, the secrecy and integrity of their data, and the spatial, legal, temporal constraints related to their data. Although a good volume of work was dedicated for auditing in the literature, only little work was dedicated to the fulfillment of the contractual obligations of the CSPs. The paper contributes to knowledge by proposing an extension to the auditing models to include the fulfillment of contractual obligations aspects beside the important aspects of secrecy and integrity of client’s data.展开更多
Objective:We assessed the trends in lung cancer incidence over a 25-year period by socioeconomic groups for men in New South Wales(NSW),Australia.Methods:Men diagnosed with lung cancer between 1987 and 2011 were d...Objective:We assessed the trends in lung cancer incidence over a 25-year period by socioeconomic groups for men in New South Wales(NSW),Australia.Methods:Men diagnosed with lung cancer between 1987 and 2011 were divided into five quintiles according to an Index of Education and Occupation(IEO).We assessed relative socioeconomic differences over time by calculating age-standardized incidence ratios(SIRs)by 5-year period of diagnosis,and estimated absolute differences by comparing the observed and expected numbers of cases using the highest IEO quintile as the reference.Results:Lung cancer incidence for men decreased from 1987 to 2011 for all IEO quintiles,with a greater rate of decline for men living in the highest IEO areas.Thus,the relative disparity increased significantly over the 25-year period(P=0.0006).For example,the SIR for the lowest IEO quintile increased from 1.28 during 1987–1991 to 1.74during 2007–2011.Absolute differences also increased with the proportion of"potentially preventable"cases doubling from 14.5% in 1987–1991 to 30.2% in 2007–2011.Conclusions:Despite the overall decline in lung cancer incidence among men in NSW over the past 25 years,there was a significant increase in disparity across socioeconomic areas in both relative and absolute terms.展开更多
