Surface Expressions of Rayleigh-Taylor Instability in Continental Interiors

Two-dimensional thermal-mechanical numerical models show that Rayleigh-Taylor-type (RT) gravitational removal of high-density lithosphere may produce significant surface deformation (vertical deflection 〉1000 m) in the interior of a continental plate.A reasonable range of crustal strengths and thicknesses,representing a variation from a stable continental interior to a hot orogen with a thick crust,is examined to study crustal deformation and the surface deflection in response to an RT instability.In general,three types of surface deflection are observed during the RT drip event:(1) subsidence and negative topography; (2) uplift and positive topography; (3) subsidence followed by uplift and inverted topography.One key factor that determines the surface expression is the crustal thickness.Models with a thin crust mainly show subsidence and develop a basin.In the thick crust models,surface expressions are more variable,depending on the crustal strength and depth of highdensity anomaly.With weak crust and a deep high-density anomaly,the RT drip is decoupled from the overlying crust,and the surface exhibits uplift or little deflection,as the RT drip induces contraction and thickening of the overlying crust.In contrast,with a strong crust and shallow anomaly,the surface is more strongly coupled with the drip and undergoes subsidence,followed by uplift. 展开更多

Cloning and Prokaryotic Expression of P23 Major Surface Protein Gene from Theileria sergenti 被引量:2

[Objective] The aim was to study cloning and prokaryotic expression of P23 major surface protein gene of Theileria sergenti. [Method] A pair of specific primers was designed according to the sequence of P23 major surface protein of T. sergenti (D84447).The P23 gene was amplified by PCR from genomic DNA of T. sergenti and cloned into pMD18-T vector to construct recombinant clonal vector pMD18-P23. Positive clones were identified by PCR screening and restriction digestion. A recombinant expression plasmid pGEX-4T-P23 was constructed by subcloning the cloned P23 gene into the linearized pGEX-4T-1 vector and transformed into E. coli BL21. After introduction by IPTG,the expressed fusion protein was identified by SDS-PAGE and Western-blotting. [Result] The cloned gene has a total length of 507 bp. Sequencing result showed that the nucleotide sequence of the cloned P23 gene shared 99.4% identity with that of P23 published in GenBank (D84447). The expressed fusion protein was 46 ku in molecular mass. Induction opportunity of zhours after culture inoculation was the best,the induction time of 6 h was the best,and induction temperature of 34 ℃ was the best as well,IPTG of 1 mmol/L had little effect on the expression. Western-blotting indicated that recombinant protein was recognized by specific antibody. [Conclusion] This study would lay a foundation for further research on the prevention and diagnose of T. sergenti. 展开更多

G protein-coupled receptors (GPCRs) are involved in all human physiological systems where they are responsible for transducing extracellular signals into cells. GPCRs signal in response to a diverse array of stimuli... G protein-coupled receptors (GPCRs) are involved in all human physiological systems where they are responsible for transducing extracellular signals into cells. GPCRs signal in response to a diverse array of stimuli including light, hormones, and lipids, where these signals affect downstream cascades to impact both health and disease states. Yet, despite their importance as therapeutic tar- gets, detailed molecular structures of only 30 GPCRs have been determined to date. A key challenge to their structure determination is adequate protein expression. Here we report the quantification of protein expression in an insect cell expression system for all 826 human GPCRs using two different fusion constructs. Expression char- acteristics are analyzed in aggregate and among each of the five distinct subfamilies. These data can be used to identify trends related to GPCR expression between dif- ferent fusion constructs and between different GPCR families, and to prioritize lead candidates for future structure determination feasibility. 展开更多
