The sulfate-reducing bacteria(SRB)corrosion of H70 brass,H80 brass and T2 copper was systematically studied using microstructure characterizations and electrochemical measurements.The results showed that H70 brass,H80...The sulfate-reducing bacteria(SRB)corrosion of H70 brass,H80 brass and T2 copper was systematically studied using microstructure characterizations and electrochemical measurements.The results showed that H70 brass,H80 brass and T2 copper exhibited good corrosion resistance in the sterile environment,and the corrosion products were mainly metal oxides,such as Cu_(2)O,CuO and ZnO.The SRB metabolism sharply accelerated the corrosion process of three types of copper alloys,especially the T2 copper.In the inoculated environment,an additional mixture of Cu_(2)S,ZnS and CuSO_(4)existed in the corrosion products.Pitting corrosion was the main corrosion style for the H70 brass and H80 brass,while general corrosion and pitting corrosion simultaneously dominated the corrosion process of the T2 copper in this environment.The results provide a new insight to the microbiological corrosion of copper alloys.展开更多

When the concentration of COD and Cd 2+ in the influent were 270 5mg/L and 100mg/L respectively and hydraulic retention time was 4 hours, the removal rate of COD and Cd 2+ were higher than 73 8% and 99 8% respectively. The reactor can treat as high as 1000mg/L of concentration of Cd 2+ . The highest removal velocity rate of Cd 2+ reached 2999 1mg/(L·d). The results showed that serious pitting corrosion took place when aluminum alloys were exposed in the mediums containing SRB, whereas no pitting corrosion were found on the surfaces of aluminum alloys only exposed in blank mediums non containing SRB. Batch experiments suggest that combining ZVI with SRB can enhance the removal of U(Ⅵ) synergistically. The removal rate of U(Ⅵ) in the ZVI+SRB combining system is obviously higher than the total rate of ZVI system and SRB system with a difference of 13.4% at t=2 h and 29.9% at t=4 h. Column experiments indicate that the reactor filled with both ZVI and SRB biofilms is of better performance than the SRB bioreactor in wastewater basification, desulfurization and U(Ⅵ) fixation. The results imply that the ZVI+SRB permeable reactive barrier may be a promising method for treating subsurface uranium contamination.展开更多

Distributions of total mercury (THg) and methylmercury (MeHg) in mangrove...Estuaries are important sites for mercury (Hg) methylation, with sulfate-reducing bacteria (SRB) thought to be the main Hg methylators. Distributions of total mercury (THg) and methylmercury (MeHg) in mangrove sediment and sediment core from Jiulong River Estuary Provincial Mangrove Reserve, China were determined and the possible mechanisms of Hg methylation and their controlling factors in mangrove sediments were investigated. Microbiological and geochemical parameters were also determined. Results showed that SRB constitute a small fraction of total bacteria (TB) in both surface sediments and the profile of sediments. The content of THg, MeHg, TB, and SRB were (350 ± 150) ng/g, (0.47 ± 0.11) ng/g, (1.4× 10^011 ± 4.1 × 10^9) cfu/g dry weight (dw), and (5.0× 10^6 d: 2.7 × 10^6) cfu/g dw in surficial sediments, respectively, and (240 ± 24) ng/g, (0.30 ± 0.15) ng/g, (1.9 × 10^11 ± 4.2 × 10^9) cfu/g dw, and (1.3 × 10^6 ± 2.0 × 10^6) cfu/g dw in sediment core, respectively. Results showed that THg, MeHg, TB, MeHg/THg, salinity and total sulfur (TS) increased with depth, but total organic matter (TOM), SRB, and pH decreased with depth. Concentrations of MeHg in sediments showed significant positive correlation with THg, salinity, TS, and MeHg/THg, and significant negative correlation with SRB, TOM, and pH. It was concluded that other microbes, rather than SRB, may also act as main Hg methylators in mangrove sediments.展开更多

MIC of carbon steel caused by SRB is the most intens...Sulfate reducing bacteria(SRB) are often the culprits of microbiologically influenced corrosion(MIC) in anoxic environments because sulfate is a ubiquitous oxidant. MIC of carbon steel caused by SRB is the most intensively investigated topic in MIC because of its practical importance. It is also because biogenic sulfides complicate mechanistic SRB MIC studies, making SRB MIC of carbon steel is a long-lasting topic that has generated considerable confusions. It is expedient to think that biogenic H_2S secreted by SRB acidifies the broth because it is an acid gas. However, this is not true because endogenous H_2S gets its H^+ from organic carbon oxidation and the fluid itself in the first place rather than an external source. Many people believe that biogenic H_2S is responsible for SRB MIC of carbon steel. However, in recent years,well designed mechanistic studies provided evidence that contradicts this misconception. Experimental data have shown that cathodic electron harvest by an SRB biofilm from elemental iron via extracellular electron transfer(EET) for energy production by SRB is the primary cause. It has been demonstrated that when a mature SRB biofilm is subjected to carbon source starvation, it switches to elemental iron as an electron source and becomes more corrosive. It is anticipated that manipulations of EET related genes will provide genetic-level evidence to support the biocathode theory in the future. This kind of new advances will likely lead to new gene probes or transcriptomics tools for detecting corrosive SRB strains that possess high EET capabilities.展开更多
The study is aimed at the problem of high content of Cr^(6+),Cr^(3+)and SO_(4)^(2-)is high and low pH value in acid mine drainage(AMD).Moreover,treatment of AMD by sulfate reducing bacteria(SRB)requires the addition o...The study is aimed at the problem of high content of Cr^(6+),Cr^(3+)and SO_(4)^(2-)is high and low pH value in acid mine drainage(AMD).Moreover,treatment of AMD by sulfate reducing bacteria(SRB)requires the addition of carbon source,while the treating effectiveness is not good enough on its own.The sugarcane slag,the corn cob and the sunflower straw were selected as the SRB carbon source cooperating with iron scrap to construct the dynamic columns 1,2 and 3.The mechanism of removing Cr^(6+),Cr^(3+),SO_(4)^(2-)and H+and the regularity of sustained release of carbon source and TFe release was studied in AMD.The removal efficiency of heavy metal ions,the ability of sustained release of carbon source,and the ability of adjusting acid by the three dynamic columns were compared.The result shows that the average removal rates of Cr^(6+),Cr^(3+)and SO_(4)^(2-)in effluent of dynamic column 1,filled by sugarcane slag,iron scrap and SRB,were 96.9%,67.1%and 54.3%.The average release of TFe and chemical oxygen demand(COD)were 4.4 and 287.3 mg/L.Its average pH was 6.98.Compared with the performance of dynamic columns 1,2 and 3,dynamic column 1 performed best in removing Cr^(6+),Cr^(3+)and SO_(4)^(2-)from AMD and controlling the release of COD and TFe,adjusting the pH of the solution.The study is of significance in treatment of AMD by taking for biomass materials as SRB carbon source in cooperation with iron scrap.展开更多

Background: Among the gut microbiota,sulfate-reducing bacteria(SRB) is a kind of hydrogen-utilizing functional bacteria that plays an important role in intestinal hydrogen and sulfur metabolism.However,information is ...Background: Among the gut microbiota,sulfate-reducing bacteria(SRB) is a kind of hydrogen-utilizing functional bacteria that plays an important role in intestinal hydrogen and sulfur metabolism.However,information is lacking regarding diversity and community structure of SRB in the gut of piglets.Middle cecum contents were collected from 6 Yorkshire and 6 Meishan piglets at postnatal days(PND) 14,28 and 49.Piglets were weaned at PND28.Real-time quantitative PCR was performed to detect the number of SRB in the cecum based on dissimilatory sulfite reductase subunit A(dsrA) gene.Prior to real-time PCR,plasmid containing the dsrA gene was constructed and used as external standard to create a standard curve,from which the gene copies of dsrA were calculated.H2S concentration in the cecal contents was measured.Illumina PE250 sequencing of dsrA gene was used to investigate SRB diversity in cecum contents.Results: The qPCR results showed that the number of SRB at PND49 was significantly higher than that at PND28 in Meishan piglets.The concentration of H2S has no significant difference between piglet breeds and between different ages.The Illumina sequencing analysis revealed that the Chao1 richness index was significantly higher at PND49 than that at PND14 and PND28 in Yorkshire piglets.Based on dsrA gene similarities,Proteobacteria,Actinobacteria,and Firmicutes were identified at the phylum level,and most sequences were classified as Proteobacteria.At the genus level,most of sequences were classified as Desulfovibrio.At the species level,Desulfovibrio intestinalis was the predominant SRB in the piglet cecum.The relative abundance and the inferred absolute abundance of Faecalibacterium prausnitzii at PND49 were significantly higher than that at PND14 in Yorkshire piglets.Pig breeds did not affect the dsrA gene copies of SRB,diversity index and community pattern of SRB.Conclusions: Sulfate-reducing bacteria are widely colonized in the cecum of piglets and D.intestinalis is the dominant SRB.The age of piglets,but not the pig breeds affects the diversity and community pattern of SRB.展开更多

Physiological-biochemical analysis showed that the strain could utilize polyacrylamide as sole carbon and nitrogen source. The sequence analysis of 16S rDNA illustrated that the similarity of F8 and Desulfovibrio desulfuricans (AF192153) was 99%, and the similarity sequence of dissimilatory sulfite reductase gene (DSR) cloned from the strain and Desulfovibrio desulfuricans (AF273034) was 98%. Their phylogenitic analysis was basically anastomosed, and thus temporarily named as Desulfovibrio desulfuricans F8. The DSR cloned from F8 strain was 2740 bp in length consisting of three ORF, DSRA, DSRB and DSRD as a single operon (DSRABD) regulated by the same operator. DSRA contained typical conservative box of sulfate—sulfite reducing enzyme (SiteⅠand SiteⅡ), which could bind siroheme and [Fe4S4]. DSRB retained a [Fe4S4] binding site, with an uncomplimentary structure for siroheme binding. There was no conservative box in DSRD. Sequence analysis of DSR will provide a theoretical basis for quantitative detection, metabolic pathway modification through gene engineering, and sulfate reducing bacteria (SRB) suppression.展开更多

The pro...Sulfate reducing bacteria(SRB) play significant roles in anaerobic environments in oil sands mature fine tailings(MFTs). Hydrogen sulfide(H2S) is produced during the biological sulfate reduction process. The production of toxic H2S is one of the concerns because it may hinder the landscape remediation efficiency of oil sands tailing ponds. In present study, the in situ activity and the community structure of SRB in MFT and gypsum amended MFT in two settling columns were investigated. Combined techniques of H2S microsensor and dissimilatory sulfite reductase β-subunit(dsrB) genes-based real time quantitative polymerase chain reaction(qPCR) were applied to detect the in situ H2S and the abundance of SRB. A higher diversity of SRB and more H2S were observed in gypsum amended MFT than that in MFT, indicating a higher sulfate reduction activity in gypsum amended MFT; in addition, the activity of SRB varied as depth in both MFT and gypsum amended MFT: the deeper the more H2S produced. Long-term plans for tailings management can be assessed more wisely with the information provided in this study.展开更多

However, as previous studies to address corrosion in the marine environment have largely focused on anaerobic bacteria, limited attention has been paid to the composition and function of aerobic and facultative bacteria in this process. For analysis in this study, ten samples were collected from rust layers on steel plates that had been immersed in seawater for diff erent periods (i.e., six months and eight years) at Sanya and Xiamen, China. The cultivable aerobic bacterial community structure as well as the number of sulfate-reducing bacteria (SRB) were analyzed in both cases, while the proportion of facultative SRB among the isolated aerobic bacteria in each sample was also evaluated using a novel approach. Bacterial abundance results show that the proportions are related to sea location and immersion time;abundances of culturable aerobic bacteria (CAB) and SRB from Sanya were greater in most corrosion samples than those from Xiamen, and abundances of both bacterial groups were greater in samples immersed for six months than for eight years. A total of 213 isolates were obtained from all samples in terms of CAB community composition, and a phylogenetic analysis revealed that the taxa comprised four phyla and 31 genera. Bacterial species composition is related to marine location;the results show that Firmicutes and Proteobacteria were the dominant phyla, accounting for 98.13% of the total, while Bacillus and Vibrio were the dominant genera, accounting for 53.06% of the total. An additional sixfacultative SRB strains were also screened from the isolates obtained and were found to encompass the genus Vibrio (four strains), Staphylococcus (one strain), and Photobacterium (one strain). It is noteworthy that mentions of Photobacterium species have so far been absent from the literature, both in terms of its membership of the SRB group and its relationship to corrosion.展开更多

Research on the interrelationship between fouling organism and calcareous deposits is very important but very limited, especially sulfate-reducing bacteria(SRB). SRB is a kind of very important fouling organism that causes microbial corrosion of metals. A study of the influence of calcareous deposit on corrosion behavior of Q235 carbon steel in SRB-containing culture medium was carried out using electrochemical impedance spectroscopy(EIS), scanning electron microscopy(SEM) and surface spectroscopy(EDS). The calcareous deposit was formed with good crystallinity and smooth surface under the gradient current density of -30 μA cm^(-2) in natural seawater for 72 h. Our results can help elucidate the formation of calcareous deposits and reveal the interrelationship between SRB and calcareous deposits under cathodic protection. The results indicate that the corrosion tendency of carbon steel was obviously affected by Sulfate-reducing Bacteria(SRB) metabolic activity and the calcareous deposit formed on the surface of carbon steel under cathodic protection was favourable to reduce the corrosion rate. Calcareous deposits can promote bacterial adhesion before biofilm formation. The results revealed the interaction between biofouling and calcareous deposits, and the anti-corrosion ability was enhanced by a kind of inorganic and organic composite membranes formed by biofilm and calcareous deposits.展开更多

In this paper, the effect of sulfate-reducing bacteria (SRB) on corrosion behavior of mild steel in sea mud was studied by weight loss, dual-compartment cell, electronic probe microanalysis (EPMA), transmission electron microscopy (TEM) combined with energy dispersive X-ray analysis (EDX) and electrochemical impedance spectroscopy (EIS). The results showed that corrosion rate and galvanic current were influenced by the metabolic activity of SRB. In the environment of sea mud containing SRB, the original corrosion products, ferric (oxyhydr) oxide, transformed to iron sulfide. With the excess of the dissolved H2S, the composition of the protective layer formed of FeS transformed to FeS2 or other non-stoichiometric polysulphide, which changed the state of the former layer and accelerated the corrosion process.展开更多

The size of microspheres can be controlled by changing reaction time and temperature. The acid concentration was critical for the formation of microspheres with smooth surfaces. The excellent antibacterial performance of PNMA in novolac epoxy coating to sulfate reducing bacteria was demonstrated. Moreover, in API media, PNMA inhibited growth of SRB and then reduced the corrosion rate of carbon steel remarkably.展开更多

The 10-methylhexadecanoic acid (10Mel6:0) has been detected from most of the samples using GC-MS based on relative retention time and co-injection of standard 10Me16:0 compound. Samples with higher 10Me16: 0 concentrations, show higher U contents. The presence of 10Mel6:0 along with iso-ω7-cis-heptadecenoic acid (i17:1ω7c) and ω7-cisoctadecenoic acid (18:1ω7c) in the fluid inclusions may indicate the occurrence of SRB during the U mineralization. Saturated fatty-acids and unsaturated fatty-acids detected in this study have individual δ^13C values from-30.3‰ to-28.5‰ and-30.5‰ to-27.9‰, respectively. These values are close to those of n-alkanes of the associated oils and the bulk oils, but significantly distinct from methane, thus the oils are concluded to have been used as substrate for the microorganisms to survive on. This proposal is supported by oil biodegradation and ore-stage calcite cement with δ^13C values from-1.4‰ to-17.2‰.展开更多

The results show that inoculation of SRB into the culture medium significantly affects the anodic polarization behavior of NSSAM3 and accelerates anodic depolarization process, however, it has little effect on cathodic polarization curves of NSSAM3. Under the same exposure time, the anodic polarization curves of NSSAM3 in culture medium with SRB are in anodic active dissolution state when anodic polarization potential value is below 0 V(SCE), whose anodic polarization current density is bigger than that of in culture medium without SRB. Moreover, when the concentration of Cu^2+ in SRB medium increases, anodic polarization current density of NSSAM3 decreases and polarization resistance increases with increasing time. Scanning electron microscope (SEM) observations indicate that SRB unevenly attaches on the surface of NSSAM3, and induces the sensitivity to local corrosion.展开更多

These zones are different by the degree of pollution in order ...In this study, the electrochemical corrosion behavior of copper was investigated in seawater collected from four different marine zones of Agadir coastal. These zones are different by the degree of pollution in order to study the effect of this pollution on the copper corrosion, especially the microbial pollution by sulfate reducing-bacteria (SRB). So, to prove this relationship, the microbiological analyses researching the SRB are realized. In parallel, the electrochemical impedance measurement and atomic absorption analysis are established to compare the microbiological evolution cycles with the electrochemical behavior of copper during the immersion period. In the results, we found a good correlation between the growth cycle of marine sulfate-reducing bacteria and the copper corrosion rate by the sulfur and extracellular polymeric substances (EPS) produced as bacteria metabolites. Additionally, this corrosion rate depends on the immersed time: it is maximal after the first or second month depending on the marine zone.展开更多

The results showed that serious pitting corrosion took place when aluminum alloys were exposed in the mediums containing SRB, whereas no pitting corrosion were found on the surfaces of aluminum alloys only exposed in blank mediums non containing SRB. It was demonstrated with EDXA that corrosion of aluminum alloys exposed in the solutions containing SRB, whereas the corrosion in the solution non containing microorganisms was attributed to the presence of chloride ions(Cl ).

文摘Batch and column experiments were conducted to determine whether zerovalent iron (ZVI) and sulfate reducing bacteria (SRB) can function synergistically and accelerate pollutant removal. Batch experiments suggest that combining ZVI with SRB can enhance the removal of U(Ⅵ) synergistically. The removal rate of U(Ⅵ) in the ZVI+SRB combining system is obviously higher than the total rate of ZVI system and SRB system with a difference of 13.4% at t=2 h and 29.9% at t=4 h. Column experiments indicate that the reactor filled with both ZVI and SRB biofilms is of better performance than the SRB bioreactor in wastewater basification, desulfurization and U(Ⅵ) fixation. The results imply that the ZVI+SRB permeable reactive barrier may be a promising method for treating subsurface uranium contamination.

文摘Estuaries are important sites for mercury (Hg) methylation, with sulfate-reducing bacteria (SRB) thought to be the main Hg methylators. Distributions of total mercury (THg) and methylmercury (MeHg) in mangrove sediment and sediment core from Jiulong River Estuary Provincial Mangrove Reserve, China were determined and the possible mechanisms of Hg methylation and their controlling factors in mangrove sediments were investigated. Microbiological and geochemical parameters were also determined. Results showed that SRB constitute a small fraction of total bacteria (TB) in both surface sediments and the profile of sediments. The content of THg, MeHg, TB, and SRB were (350 ± 150) ng/g, (0.47 ± 0.11) ng/g, (1.4× 10^011 ± 4.1 × 10^9) cfu/g dry weight (dw), and (5.0× 10^6 d: 2.7 × 10^6) cfu/g dw in surficial sediments, respectively, and (240 ± 24) ng/g, (0.30 ± 0.15) ng/g, (1.9 × 10^11 ± 4.2 × 10^9) cfu/g dw, and (1.3 × 10^6 ± 2.0 × 10^6) cfu/g dw in sediment core, respectively. Results showed that THg, MeHg, TB, MeHg/THg, salinity and total sulfur (TS) increased with depth, but total organic matter (TOM), SRB, and pH decreased with depth. Concentrations of MeHg in sediments showed significant positive correlation with THg, salinity, TS, and MeHg/THg, and significant negative correlation with SRB, TOM, and pH. It was concluded that other microbes, rather than SRB, may also act as main Hg methylators in mangrove sediments.

文摘Sulfate reducing bacteria(SRB) are often the culprits of microbiologically influenced corrosion(MIC) in anoxic environments because sulfate is a ubiquitous oxidant. MIC of carbon steel caused by SRB is the most intensively investigated topic in MIC because of its practical importance. It is also because biogenic sulfides complicate mechanistic SRB MIC studies, making SRB MIC of carbon steel is a long-lasting topic that has generated considerable confusions. It is expedient to think that biogenic H_2S secreted by SRB acidifies the broth because it is an acid gas. However, this is not true because endogenous H_2S gets its H^+ from organic carbon oxidation and the fluid itself in the first place rather than an external source. Many people believe that biogenic H_2S is responsible for SRB MIC of carbon steel. However, in recent years,well designed mechanistic studies provided evidence that contradicts this misconception. Experimental data have shown that cathodic electron harvest by an SRB biofilm from elemental iron via extracellular electron transfer(EET) for energy production by SRB is the primary cause. It has been demonstrated that when a mature SRB biofilm is subjected to carbon source starvation, it switches to elemental iron as an electron source and becomes more corrosive. It is anticipated that manipulations of EET related genes will provide genetic-level evidence to support the biocathode theory in the future. This kind of new advances will likely lead to new gene probes or transcriptomics tools for detecting corrosive SRB strains that possess high EET capabilities.

文摘The study is aimed at the problem of high content of Cr^(6+),Cr^(3+)and SO_(4)^(2-)is high and low pH value in acid mine drainage(AMD).Moreover,treatment of AMD by sulfate reducing bacteria(SRB)requires the addition of carbon source,while the treating effectiveness is not good enough on its own.The sugarcane slag,the corn cob and the sunflower straw were selected as the SRB carbon source cooperating with iron scrap to construct the dynamic columns 1,2 and 3.The mechanism of removing Cr^(6+),Cr^(3+),SO_(4)^(2-)and H+and the regularity of sustained release of carbon source and TFe release was studied in AMD.The removal efficiency of heavy metal ions,the ability of sustained release of carbon source,and the ability of adjusting acid by the three dynamic columns were compared.The result shows that the average removal rates of Cr^(6+),Cr^(3+)and SO_(4)^(2-)in effluent of dynamic column 1,filled by sugarcane slag,iron scrap and SRB,were 96.9%,67.1%and 54.3%.The average release of TFe and chemical oxygen demand(COD)were 4.4 and 287.3 mg/L.Its average pH was 6.98.Compared with the performance of dynamic columns 1,2 and 3,dynamic column 1 performed best in removing Cr^(6+),Cr^(3+)and SO_(4)^(2-)from AMD and controlling the release of COD and TFe,adjusting the pH of the solution.The study is of significance in treatment of AMD by taking for biomass materials as SRB carbon source in cooperation with iron scrap.

文摘Microbiologically influenced corrosion caused by sulfate-reducing bacteria(SRB) poses a serious threat to marine engineering facilities.This study focused on the interaction between the corrosion behavior of two aluminum alloys and SRB metabolic activity.SRB growth curve and sulfate variation with and with aluminum were performed to find the effect of two aluminum alloys on SRB metabolic activity.Corrosion of 5052 aluminum alloy and Al-Zn-In-Cd aluminum alloy with and without SRB were performed.The results showed that both the presence of 5052 and Al-Zn-In-Cd aluminum alloy promoted SRB metabolic activity,with the Al-Zn-In-Cd aluminum alloy having a smaller promotion effect compared with 5052 aluminum alloy.The electrochemical results suggested that the corrosion of the Al-Zn-In-Cd aluminum alloy was accelerated substantially by SRB.Moreover,SRB led to the transformation of Al-Zn-In-Cd aluminum alloy corrosion product from Al(OH)3 to Al2 S3 and NaAlO2.

文摘Background: Among the gut microbiota,sulfate-reducing bacteria(SRB) is a kind of hydrogen-utilizing functional bacteria that plays an important role in intestinal hydrogen and sulfur metabolism.However,information is lacking regarding diversity and community structure of SRB in the gut of piglets.Middle cecum contents were collected from 6 Yorkshire and 6 Meishan piglets at postnatal days(PND) 14,28 and 49.Piglets were weaned at PND28.Real-time quantitative PCR was performed to detect the number of SRB in the cecum based on dissimilatory sulfite reductase subunit A(dsrA) gene.Prior to real-time PCR,plasmid containing the dsrA gene was constructed and used as external standard to create a standard curve,from which the gene copies of dsrA were calculated.H2S concentration in the cecal contents was measured.Illumina PE250 sequencing of dsrA gene was used to investigate SRB diversity in cecum contents.Results: The qPCR results showed that the number of SRB at PND49 was significantly higher than that at PND28 in Meishan piglets.The concentration of H2S has no significant difference between piglet breeds and between different ages.The Illumina sequencing analysis revealed that the Chao1 richness index was significantly higher at PND49 than that at PND14 and PND28 in Yorkshire piglets.Based on dsrA gene similarities,Proteobacteria,Actinobacteria,and Firmicutes were identified at the phylum level,and most sequences were classified as Proteobacteria.At the genus level,most of sequences were classified as Desulfovibrio.At the species level,Desulfovibrio intestinalis was the predominant SRB in the piglet cecum.The relative abundance and the inferred absolute abundance of Faecalibacterium prausnitzii at PND49 were significantly higher than that at PND14 in Yorkshire piglets.Pig breeds did not affect the dsrA gene copies of SRB,diversity index and community pattern of SRB.Conclusions: Sulfate-reducing bacteria are widely colonized in the cecum of piglets and D.intestinalis is the dominant SRB.The age of piglets,but not the pig breeds affects the diversity and community pattern of SRB.

文摘A sulfate reducing bacteria was isolated from mining sewage of Daqing Oilfield by Hungate anaerobic technology. Physiological-biochemical analysis showed that the strain could utilize polyacrylamide as sole carbon and nitrogen source. The sequence analysis of 16S rDNA illustrated that the similarity of F8 and Desulfovibrio desulfuricans (AF192153) was 99%, and the similarity sequence of dissimilatory sulfite reductase gene (DSR) cloned from the strain and Desulfovibrio desulfuricans (AF273034) was 98%. Their phylogenitic analysis was basically anastomosed, and thus temporarily named as Desulfovibrio desulfuricans F8. The DSR cloned from F8 strain was 2740 bp in length consisting of three ORF, DSRA, DSRB and DSRD as a single operon (DSRABD) regulated by the same operator. DSRA contained typical conservative box of sulfate—sulfite reducing enzyme (SiteⅠand SiteⅡ), which could bind siroheme and [Fe4S4]. DSRB retained a [Fe4S4] binding site, with an uncomplimentary structure for siroheme binding. There was no conservative box in DSRD. Sequence analysis of DSR will provide a theoretical basis for quantitative detection, metabolic pathway modification through gene engineering, and sulfate reducing bacteria (SRB) suppression.

文摘Sulfate reducing bacteria(SRB) play significant roles in anaerobic environments in oil sands mature fine tailings(MFTs). Hydrogen sulfide(H2S) is produced during the biological sulfate reduction process. The production of toxic H2S is one of the concerns because it may hinder the landscape remediation efficiency of oil sands tailing ponds. In present study, the in situ activity and the community structure of SRB in MFT and gypsum amended MFT in two settling columns were investigated. Combined techniques of H2S microsensor and dissimilatory sulfite reductase β-subunit(dsrB) genes-based real time quantitative polymerase chain reaction(qPCR) were applied to detect the in situ H2S and the abundance of SRB. A higher diversity of SRB and more H2S were observed in gypsum amended MFT than that in MFT, indicating a higher sulfate reduction activity in gypsum amended MFT; in addition, the activity of SRB varied as depth in both MFT and gypsum amended MFT: the deeper the more H2S produced. Long-term plans for tailings management can be assessed more wisely with the information provided in this study.

文摘Anaerobic, aerobic, and facultative bacteria are all present in corrosive environments. However, as previous studies to address corrosion in the marine environment have largely focused on anaerobic bacteria, limited attention has been paid to the composition and function of aerobic and facultative bacteria in this process. For analysis in this study, ten samples were collected from rust layers on steel plates that had been immersed in seawater for diff erent periods (i.e., six months and eight years) at Sanya and Xiamen, China. The cultivable aerobic bacterial community structure as well as the number of sulfate-reducing bacteria (SRB) were analyzed in both cases, while the proportion of facultative SRB among the isolated aerobic bacteria in each sample was also evaluated using a novel approach. Bacterial abundance results show that the proportions are related to sea location and immersion time;abundances of culturable aerobic bacteria (CAB) and SRB from Sanya were greater in most corrosion samples than those from Xiamen, and abundances of both bacterial groups were greater in samples immersed for six months than for eight years. A total of 213 isolates were obtained from all samples in terms of CAB community composition, and a phylogenetic analysis revealed that the taxa comprised four phyla and 31 genera. Bacterial species composition is related to marine location;the results show that Firmicutes and Proteobacteria were the dominant phyla, accounting for 98.13% of the total, while Bacillus and Vibrio were the dominant genera, accounting for 53.06% of the total. An additional sixfacultative SRB strains were also screened from the isolates obtained and were found to encompass the genus Vibrio (four strains), Staphylococcus (one strain), and Photobacterium (one strain). It is noteworthy that mentions of Photobacterium species have so far been absent from the literature, both in terms of its membership of the SRB group and its relationship to corrosion.

文摘Cathodic protection is a very effective method to protect metals, which can form calcareous deposits on metal surface. Research on the interrelationship between fouling organism and calcareous deposits is very important but very limited, especially sulfate-reducing bacteria(SRB). SRB is a kind of very important fouling organism that causes microbial corrosion of metals. A study of the influence of calcareous deposit on corrosion behavior of Q235 carbon steel in SRB-containing culture medium was carried out using electrochemical impedance spectroscopy(EIS), scanning electron microscopy(SEM) and surface spectroscopy(EDS). The calcareous deposit was formed with good crystallinity and smooth surface under the gradient current density of -30 μA cm^(-2) in natural seawater for 72 h. Our results can help elucidate the formation of calcareous deposits and reveal the interrelationship between SRB and calcareous deposits under cathodic protection. The results indicate that the corrosion tendency of carbon steel was obviously affected by Sulfate-reducing Bacteria(SRB) metabolic activity and the calcareous deposit formed on the surface of carbon steel under cathodic protection was favourable to reduce the corrosion rate. Calcareous deposits can promote bacterial adhesion before biofilm formation. The results revealed the interaction between biofouling and calcareous deposits, and the anti-corrosion ability was enhanced by a kind of inorganic and organic composite membranes formed by biofilm and calcareous deposits.

文摘Microbiologically influenced corrosion (MIC) is very severe corrosion for constructions buried under sea mud environment. Therefore it is of great importance to carry out the investigation of the corrosion behavior of marine steel in sea mud. In this paper, the effect of sulfate-reducing bacteria (SRB) on corrosion behavior of mild steel in sea mud was studied by weight loss, dual-compartment cell, electronic probe microanalysis (EPMA), transmission electron microscopy (TEM) combined with energy dispersive X-ray analysis (EDX) and electrochemical impedance spectroscopy (EIS). The results showed that corrosion rate and galvanic current were influenced by the metabolic activity of SRB. In the environment of sea mud containing SRB, the original corrosion products, ferric (oxyhydr) oxide, transformed to iron sulfide. With the excess of the dissolved H2S, the composition of the protective layer formed of FeS transformed to FeS2 or other non-stoichiometric polysulphide, which changed the state of the former layer and accelerated the corrosion process.

文摘Microspheres of conducting polymers poly N-methylaniline (PNMA) were successfully synthesized through oxidation of N-methylaniline without any template. The average diameter of the microspheres with a smooth surface was about 0.40 μm when 0.2 M N-methylanUine was oxidized with 0.2 M ammonium persulfate in 0.2 M of HClO4 solution. The size of microspheres can be controlled by changing reaction time and temperature. The acid concentration was critical for the formation of microspheres with smooth surfaces. The excellent antibacterial performance of PNMA in novolac epoxy coating to sulfate reducing bacteria was demonstrated. Moreover, in API media, PNMA inhibited growth of SRB and then reduced the corrosion rate of carbon steel remarkably.

文摘Fatty acids were extracted from fluid inclusions, and analyzed for distribution and individual 613C values to determine if there exists sulfate-reducing bacteria (SRB) and what substrates SRB depleted during U mineralization in the Dongsheng deposit. The 10-methylhexadecanoic acid (10Mel6:0) has been detected from most of the samples using GC-MS based on relative retention time and co-injection of standard 10Me16:0 compound. Samples with higher 10Me16: 0 concentrations, show higher U contents. The presence of 10Mel6:0 along with iso-ω7-cis-heptadecenoic acid (i17:1ω7c) and ω7-cisoctadecenoic acid (18:1ω7c) in the fluid inclusions may indicate the occurrence of SRB during the U mineralization. Saturated fatty-acids and unsaturated fatty-acids detected in this study have individual δ^13C values from-30.3‰ to-28.5‰ and-30.5‰ to-27.9‰, respectively. These values are close to those of n-alkanes of the associated oils and the bulk oils, but significantly distinct from methane, thus the oils are concluded to have been used as substrate for the microorganisms to survive on. This proposal is supported by oil biodegradation and ore-stage calcite cement with δ^13C values from-1.4‰ to-17.2‰.
文摘The electrochemical characteristic of antimicrobial stainless steel bearing copper NSSAM3 in sulfate reducing bacterial (SRB) was investigated by electrochemical impedance spectroscopy (EIS) and potentiodynamic polarization. The results show that inoculation of SRB into the culture medium significantly affects the anodic polarization behavior of NSSAM3 and accelerates anodic depolarization process, however, it has little effect on cathodic polarization curves of NSSAM3. Under the same exposure time, the anodic polarization curves of NSSAM3 in culture medium with SRB are in anodic active dissolution state when anodic polarization potential value is below 0 V(SCE), whose anodic polarization current density is bigger than that of in culture medium without SRB. Moreover, when the concentration of Cu^2+ in SRB medium increases, anodic polarization current density of NSSAM3 decreases and polarization resistance increases with increasing time. Scanning electron microscope (SEM) observations indicate that SRB unevenly attaches on the surface of NSSAM3, and induces the sensitivity to local corrosion.
文摘In this study, the electrochemical corrosion behavior of copper was investigated in seawater collected from four different marine zones of Agadir coastal. These zones are different by the degree of pollution in order to study the effect of this pollution on the copper corrosion, especially the microbial pollution by sulfate reducing-bacteria (SRB). So, to prove this relationship, the microbiological analyses researching the SRB are realized. In parallel, the electrochemical impedance measurement and atomic absorption analysis are established to compare the microbiological evolution cycles with the electrochemical behavior of copper during the immersion period. In the results, we found a good correlation between the growth cycle of marine sulfate-reducing bacteria and the copper corrosion rate by the sulfur and extracellular polymeric substances (EPS) produced as bacteria metabolites. Additionally, this corrosion rate depends on the immersed time: it is maximal after the first or second month depending on the marine zone.
