Screening and Identification of Strains Converting Trans-anethole to Anisic Acid

[Methods] According to the traditional microbial culture method, with trans-anethole as the sole carbon source, through enrichment culture and separation and purification, preliminary screening by thin layer chromatography and re-screening by high-performance liquid chromatography, strains that degraded trans-anethole to produce anisic acid were obtained, and 16 S rDNA sequencing and phylogenetic tree construction were performed for genetic analysis. [Results] Eleven strains that degraded trans-anethole to produce anisic acid were obtained, among which strain NT2 that produced anisic acid with a relatively high efficiency was initially identified as Pseudomonas sp. The strain’s trans-anethole degradation rate was 45.41%, and the molar production rate and cumulative concentration of anisic acid were 21.80% and 1.96 g/L, respectively. [Conclusions] Strain NT2 has a strong ability to degrade trans-anethole to produce anisic acid, and can enrich strain resources for degradation of trans-anethole to anisic acid through microbial conversion. 展开更多

To determine whether they could yield10-Hydroxy-2-decenoic acid during fermentation, gas chromatography and gas chromatography-mass spectrometry methodswere used. The results showed that the strains BH002 and BH004, were both identified as Crytococcaceae, where BH002was primarily classified into Candida for possessing the abilities. The 10-HDA productivity of Candida BH002 and that ofBH004 were 0.327% and 0.2648% respectively. 展开更多

Antibiotics residues have been accumulating in the environment day by day due to overuse of antibiotics.Recalcitrant antibiotic residues,such as tylosin(TYL),can cause serious environmental problems,which makes it important to eliminate TYL from the environment.It is important to eliminate TYL from the environment.In this study,a strain was isolated and purified from fermentation by-product that came from a TYL production factory.The TYL degrading strain was identified by its morphology,physiological and biochemical reactions and sequencing the PCR-amplified fragments of its 16 S r DNAcoding genes.The temperature,shaking speed,initial TYL concentration,p H and inoculum sizes were investigated under simulated conditions by using single factor tests.The results showed that TYL2,a high efficient strain was isolated and was identified as Brevibacillus borstelensis.The degradation rate of TYL by this strain could reach to 75%with an initial concentration of 25 mg L^-1 within 7 days under conditions of 7%B.borstelensis(v/v,2×108 CFU m L^-1)at p H 7.0 and at 35°C.It is interesting that this strain has a very strong ability to degrade the TYL in natural sewage with the degradation rate of 65%within 7 days.This result could be helpful for the degradation of TYL and provide guidance for the degradation of other antibiotics. 展开更多

Screening influence on the Stark effect of impurity states in strained wurtzite GaN/Al_xGa_(1-x)N heterojunctions under pressure 被引量:1

The screening effect of the random-phase-approximation on the states of shallow donor impurities in free strained wurtzite GaN/AlxGa1-xN heterojunctions under hydrostatic pressure and an external electric field is investigated by using a variational method and a simplified coherent potential approximation. The variations of Stark energy shift with electric field, impurity position, A1 component and areal electron density are discussed. Our results show that the screening dramatically reduces both the blue and red shifts as well as the binding energies of impurity states. For a given impurity position, the change in binding energy is more sensitive to the increase in hydrostatic pressure in the presence of the screening effect than that in the absence of the screening effect. The weakening of the blue and red shifts, induced by the screening effect, strengthens gradually with the increase of electric field. Furthermore, the screening effect weakens the mixture crystal effect, thereby influencing the Stark effect. The screening effect strengthens the influence of energy band bending on binding energy due to the areal electron density. 展开更多

[Objective] The aim was to screen strains producing alkaline protease from soil and study the conditions for enzyme production.[Method] Eight strains producing alkaline protease were isolated from soil through plate isolation,and the ability of enzyme production was measured by filter paper and Folin-phenol method.The strain with the strongest ability of enzyme production was screened as a candidate strain,then the factors influencing the ability of enzyme production was studied,finally the conditions for enzyme production was optimized through orthogonal test.[Result] No.5 strain was screened as a candidate strain due to its strong ability of enzyme production(6.00 U/ml),which accounted for 134.1% of that of Bacillus licheniformis,and it was gram-positive bacterium belonging to Clostridium.Orthogonal test showed that the optimal condition for producing protease was an environment with pH=11,0.3% of sucrose and 0.8% of peptone in the fermentation medium,and inoculation amount was 105 cfu/ml.In addition,peptone had significant impact on the level of enzyme production.[Conclusion] The study could provide theoretical references for the screening of strains producing alkaline protease. 展开更多

Strain sensors for human-motion detection must offer high stretchability, high sensitivity, fast response, and high recovery speed. In this study, we choose silver paste as a sensing material and use a screen printing method to fabricate the strain sensor based upon an electrical-resistance mechanism. After curing elastomeric polyurethane film with a thickness of 150 μm on PET film, the polyester resin mixed with blocked isocyanate curing agent was coated as a masking layer to reduce the film’s stickiness. The effect of the polyester masking layer upon the silver paste screen printing process was examined using a rolling-ball-tack test, TGA analysis of polyester resins, and cured silver-electrode films. The cost-effective strain sensor fabricated by using silver paste and screen printing processes on the stretchable-polyurethane-substrate film showed high sensitivity and fast response in a strain range of up to 100%. 展开更多

[Objective] Strain Biok Av-023 used as the control was employed on screening of high-avermectin yield mutants by rational screening.[Method] With Biok Av-023 as the original strain,the positive mutation strain was firstly screened by routine UV mutagenesis,and then the high-yield avermectin producing strain was selected by the breeding way inferred by L-Ile induction.[Result] UV mutation and L-Ile directional screening had showed that the best L-Ile screening concentration was 0.5%,and the high-yield mutation strain AV60s-32 after re-screening reached the highest titer of 4520 IU/ml,which increased by 23.4% compared with the original strain.[Conclusion] The production of avermectin can be effectively enhanced by the combined way of UV mutation and L-Ile rational breeding. 展开更多

培养料中添加桑枝适栽香菇菌株的筛选评价

本文从健康母乳样品中分离鉴定出21株双歧杆菌,通过脾淋巴细胞增殖试验、药敏性、溶血性、抑菌活性以及黏附能力评价,选定具有最佳活性的菌株。通过16S rRNA进行鉴定后,优化冻干技术并制备冻干微胶囊。结果表明,该菌为动物双歧杆菌乳亚... 本文从健康母乳样品中分离鉴定出21株双歧杆菌,通过脾淋巴细胞增殖试验、药敏性、溶血性、抑菌活性以及黏附能力评价,选定具有最佳活性的菌株。通过16S rRNA进行鉴定后,优化冻干技术并制备冻干微胶囊。结果表明,该菌为动物双歧杆菌乳亚种(Bifidobacterium animalis subsp.lactis),将其命名为BA-9。B.lactis BA-9具有安全性,其疏水性为36.24%,自凝集率为77.4%,可显著(P<0.05)降低RAW264.7细胞中NO活性,具有体外抗炎潜力。海藻酸钠与卡拉胶以1:1制备复合壁材,加入复合冻干剂(11.25%脱脂乳粉、4.93%低聚木糖和2.74%甘油)。冻干后添加冻干保护剂的B.lactis BA-9微胶囊,经过模拟胃肠液消化,活性仅下降1.3 lg CFU/g。在4℃下贮藏28 d存活率可达90.03%,有效提升了储藏稳定性。本研究为筛选具有免疫抗炎调节活性的双歧杆菌及其冻干载体的开发和应用提供了研究基础。 展开更多

基于理化指标和感官评价筛选干酪成熟用腐乳源霉菌

为筛选能为霉菌干酪提供良好风味的霉菌发酵剂,使用从腐乳相关产品中分离出的霉菌菌株制作霉菌干酪样品,对其进行理化指标测定和感官评价,最终筛选出既有干酪成熟能力又能提供良好风味的霉菌菌株。从腐乳相关产品中分离出4株具有潜力制... 为筛选能为霉菌干酪提供良好风味的霉菌发酵剂,使用从腐乳相关产品中分离出的霉菌菌株制作霉菌干酪样品,对其进行理化指标测定和感官评价,最终筛选出既有干酪成熟能力又能提供良好风味的霉菌菌株。从腐乳相关产品中分离出4株具有潜力制作干酪的霉菌,即白地霉(MP07、MP08),卷枝毛霉(FR04),雅致放射毛霉(QF03)。理化指标测定结果表明,4株腐乳源霉菌均具有一定的干酪成熟能力,其制作的干酪在主要成分含量上接近市售卡门贝尔干酪,水分含量在51.43~54.46 g/100 g,蛋白质含量在15.00~16.31 g/100 g,脂肪含量在26.47~29.62 g/100 g;在成熟完成时,其制作的干酪内部pH在6.0左右,外部pH在7.0左右;其制作的干酪中蛋白质发生初级水解和次级水解,菌株FR04制作的样品423和菌株QF03制作的样品365的蛋白质水解指数显著高于其他2组样品(P<0.05),分别为12.55%和11.59%,表明这2株霉菌具有较强的干酪成熟能力;其制作的干酪核心部分的质构参数在成熟过程中发生变化,样品365在成熟完成时的硬度、咀嚼性和回复性显著低于其他3组(P<0.05),表明该样品内部成熟状态最好。感官评价结果表明,4组腐乳源霉菌干酪与市售卡门贝尔在外观、气味、味道和口感上具有显著差异(P<0.05),样品365的平均整体喜好程度得分(5分)在5组霉菌干酪中最高。因此,确定菌株QF03为4株霉菌中最适用于制作霉菌干酪的菌株。本研究为霉菌干酪的制作提供了新的优质发酵菌株,对我国微生物资源的开发和利用具有一定的贡献。 展开更多

耐乙醇酶制备及一步法高效提取文冠果叶皂苷

为解决目前使用酶和超声两步法提取存在有机试剂消耗量高和提取时间长的问题,实现文冠果叶皂苷高效提取,本研究从腐殖土壤中定向筛选耐乙醇的产酶菌株,根据其发酵后获得混合酶的耐乙醇特性,设计一步超声-酶-乙醇提取文冠果叶皂苷工艺并... 为解决目前使用酶和超声两步法提取存在有机试剂消耗量高和提取时间长的问题,实现文冠果叶皂苷高效提取,本研究从腐殖土壤中定向筛选耐乙醇的产酶菌株,根据其发酵后获得混合酶的耐乙醇特性,设计一步超声-酶-乙醇提取文冠果叶皂苷工艺并与超声辅助酶解提取(两步法)进行比较。通过单因素及响应面试验优化条件,采用超高效液相色谱-串联质谱法(Ultra-performance liquid chromatography-tandem mass spectrometry,UPLCMS/MS)、环境扫描电子显微镜(Scanning electron microscopy,SEM)、傅立叶变换红外光谱(Fourier transform infrared spectroscopy,FT-IR)和X射线衍射(X-ray diffraction,XRD)表征和解析了文冠果叶残渣的结构特征。结果表明,黑曲霉Aspergillus niger 42发酵的混合酶耐乙醇效果最佳,其分泌的耐乙醇纤维素混合酶(内切葡聚糖酶酶活254±0.05 U/mL)在40%乙醇中稳定性显著(P<0.001)(β-葡萄糖苷酶和木聚糖酶相对酶活分别保留78.67%±0.25%和55.34%±4.00%),且不产生赭曲霉毒素,安全性高。响应面试验显示,以混合酶作为溶液,无水乙醇作为溶质,在最优条件下(超声温度55℃、功率300 W、乙醇浓度40%、时间30 min),皂苷得率达14.98%±0.08%,较超声乙醇提取(7.42%±0.23%)提升101.89%,且乙醇用量较两步法减少30%,皂苷得率提高59%(9.42%±0.04%)。UPLC-Q-TOF MS证实一步法提取物的皂苷组成未变且较两步法含量显著增加。SEM、FT-IR和XRD结果分析表明,一步法较两步法处理对原料表观结构的破坏更大,对结构中的O-H、C=C和C-OC等化学键被破坏更明显且无定形区被充分降解,该一步法兼具皂苷得率高、溶剂消耗低、操作条件温和及环境友好等优势,为文冠果叶皂苷的绿色高效提取提供了新解决方案,对其他天然活性物质的提取提供借鉴。 展开更多

【背景】直接利用纤维素原料作微生物碳源来合成聚羟基脂肪酸酯(polyhydroxyalknoates,PHA),能有效降低其生产成本,但目前仍缺乏合适的微生物菌种。【目的】获得能够直接利用纤维素原料作碳源且高效合成PHA的微生物菌种,对分离菌种进行... 【背景】直接利用纤维素原料作微生物碳源来合成聚羟基脂肪酸酯(polyhydroxyalknoates,PHA),能有效降低其生产成本,但目前仍缺乏合适的微生物菌种。【目的】获得能够直接利用纤维素原料作碳源且高效合成PHA的微生物菌种,对分离菌种进行鉴定并对其发酵特性进行研究。【方法】利用羧甲基纤维素(sodium carboxymethyl cellulose,CMC)-Na为唯一碳源,结合苏丹黑B染色和尼罗红染色法从自然环境中筛选目的菌株,研究分离菌株的滤纸崩解及纤维素酶活情况,通过形态学观察、生理生化反应及16S rRNA基因序列同源性比较对菌株进行分类鉴定,对菌株利用纤维素发酵产PHA的相关发酵条件如氮源种类、温度、pH、接种量等进行优化,最后分别利用傅里叶红外光谱(Fourier transform infrared spectroscopy,FT-IR)和核磁共振氢谱(nuclear magnetic resonance hydrogen spectroscopy,1H-NMR)对发酵产物进行分析。【结果】筛选到1株高效利用纤维素产PHA的菌株FX-9,该菌能够让纤维素滤纸在短时间内崩解明显,并表现出较高的纤维素酶活性。经鉴定,该菌株为产碱杆菌属(Cupriavidus)。优化结果表明,当氮源为酵母粉,温度为30℃,pH 7.0,接种量为10%时,该分离菌株经84 h发酵后PHA浓度达到最大值为0.49 g/L,此时细胞内PHA含量为42.44%。经FT-IR、1H-NMR鉴定该菌株利用纤维素原料发酵所提取的产物为聚羟基丁酸酯(polyhydroxybutyrate,PHB)。【结论】分离菌株Cupriavidus sp.FX-9能直接利用纤维素原料作为碳源且高效合成PHA,这将为实现直接利用木质纤维素原料发酵生产PHA提供菌种资源及经验借鉴。 展开更多

植物乳植杆菌发酵多花黄精提高多糖产量工艺优化及其体外益生元活性

多糖是黄精含量最高的活性成分,为了筛选通过微生物发酵黄精提高多糖含量的乳酸菌及优化发酵工艺,探索发酵物的益生元活性,本文从实验室保存的不同传统发酵食品中分离的12株植物乳植杆菌(Lactiplantibacillus plantarum)发酵黄精,通过... 多糖是黄精含量最高的活性成分,为了筛选通过微生物发酵黄精提高多糖含量的乳酸菌及优化发酵工艺,探索发酵物的益生元活性,本文从实验室保存的不同传统发酵食品中分离的12株植物乳植杆菌(Lactiplantibacillus plantarum)发酵黄精,通过单因素实验,系统考察了发酵时间(0~6 d)、接种量(0~9%v/v)、转速(0~200 r/min)和温度(20~40℃)对黄精发酵液中多糖含量的影响,利用响应面方法对发酵黄精工艺进行优化,并对发酵黄精冻干粉进行益生元活性分析。结果表明:12株菌均能在未添加营养物质的黄精培养基中正常生长,生物量在24~48 h内最高达到1×10^(8) CFU/mL,但48 h后生物量迅速下降。发酵48 h后,筛选到1株多糖含量提高25%的L6菌株。最佳发酵工艺参数为发酵时间3.8 d、接种量2.5%、温度35℃。在此优化条件下,发酵液中多糖含量达到1.30 mg/mL,比优化前显著(P<0.05)提高了57.71%。发酵后多糖分子量明显增加,多糖吡喃环结构发生变化,主要组成多糖的单糖中甘露糖和葡萄糖明显降解,半乳糖和半乳糖醛酸明显增加。此外,发酵物能够明显促进植物乳植杆菌(Lactiplantibacillus plantarum)CICC22213、鼠李糖乳杆菌(Lactobacillus rhamnosus)CICC22175和干酪乳杆菌(Lactobacillus casei)FBKL1.3022这3株益生菌增殖,具有潜在益生元活性,且主要短链脂肪酸乳酸和乙酸含量具有生物量依赖性。这一结果表明,通过工艺优化能够有效提升黄精发酵液中多糖的含量,为开发大健康发酵食品、根茎类药食同源食品提供了技术支撑。 展开更多

为筛选适用于酸木瓜果酒发酵的优良非酿酒酵母,以高酸水果表皮及自然发酵液为原料分离酵母菌,经杜氏管发酵、菌株耐受性能和发酵性能测定试验对菌株进行筛选,通过分子生物学技术对筛选菌株进行鉴定,并通过电子鼻技术结合主成分分析(PCA... 为筛选适用于酸木瓜果酒发酵的优良非酿酒酵母,以高酸水果表皮及自然发酵液为原料分离酵母菌,经杜氏管发酵、菌株耐受性能和发酵性能测定试验对菌株进行筛选,通过分子生物学技术对筛选菌株进行鉴定,并通过电子鼻技术结合主成分分析(PCA)对菌株发酵酸木瓜汁的挥发性风味成分进行分析。结果表明,分离出的144株菌中有4株可同时耐受葡萄糖质量浓度500 g/L、pH 2、SO2质量浓度250 mg/L、乙醇体积分数9%。菌株G26、G28、G29、N1具备优良的产酯产乙醇能力及低产H2S能力。经鉴定,菌株N1为季也蒙迈耶氏酵母(Meyerozyma guilliermonclii)、菌株G26为美极梅奇酵母(Metshnikowia pulcherrima)、菌株G28为异常威克汉姆酵母(Wickerhamomyces anomalus)、菌株G29为耐热拉钱斯氏酵母(Lachancea waltii)。电子鼻分析结果显示,菌株N1和G26发酵酸木瓜汁的醇醛化合物和氮氧化合物变化特征最明显;基于电子鼻分析响应值PCA结果表明,菌株N1发酵样与其他样品的差异最明显。该研究筛选鉴定的适用于酸木瓜发酵的优良非酿酒酵母,为酸木瓜果酒酿造提供了菌种资源与理论依据。 展开更多
