Using the back skin of the mice as the test object,UPLC was used to determine the retention rate of Glabridin(GLA)in the skin and subcutaneous fat pf mice with different dosage forms and different administration times,and to investigate the transdermal absorption effect of liquid crystal cream.The experimental results showed that the 0.5,1,2,4,6 and 8h skin retention rates of GLA in the liquid crystal cream group were 1.67,0.79,1.73,1.47,1.17 and 1.15 times higher than those of the ordinary cream group under the same dosage of administration,respectively.The 0.5,1,2,4,6 and 8 h fat retention rates of GLA in the normal cream group were 0.86,1.87,1.37,1.20,1.35 and 1.19 times higher than those in the liquid crystal group,respectively.Liquid crystal cream can significantly improve the skin retention of GLA,with certain slow and controlled release,and possesses certain skin targeting properties. 展开更多

为研究饲粮添加山楂叶黄酮和甜菊糖苷对蛋鸡产蛋性能、蛋品质和血清生化的影响,试验选取健康的66周龄京粉6号蛋鸡360只,随机分为4组,每组6个重复,每个重复15只鸡。对照组饲喂基础日粮,试验组在基础日粮中分别添加120 mg/kg山楂叶黄酮、1... 为研究饲粮添加山楂叶黄酮和甜菊糖苷对蛋鸡产蛋性能、蛋品质和血清生化的影响,试验选取健康的66周龄京粉6号蛋鸡360只,随机分为4组,每组6个重复,每个重复15只鸡。对照组饲喂基础日粮,试验组在基础日粮中分别添加120 mg/kg山楂叶黄酮、120 mg/kg甜菊糖苷及120 mg/kg山楂叶黄酮+120 mg/kg甜菊糖苷的复配组合。预试期2周,正试期4周。结果显示:与对照组相比,山楂叶黄酮组、甜菊糖苷组和复配组68~72周龄生产性能差异不显著(P>0.05)。与对照组相比,山楂叶黄酮组、甜菊糖苷组和复配组蛋黄颜色显著提高(P<0.05)。与对照组相比,山楂叶黄酮组血清中甘油三酯(TG)和低密度脂蛋白(LDL)的含量分别降低22.03%和13.88%(P<0.05);甜菊糖苷组血清中TG和LDL的含量分别降低26.83%和9.80%(P<0.05);复配组血清中TG、LDL和胆固醇(TC)的含量分别降低34.54%、52.65%和28.93%(P<0.05)。与对照组相比,山楂叶黄酮组、甜菊糖苷组及复配组肝脏脂滴占比分别降低64.60%、67.60%和81.88%(P<0.05)。综上所述,饲粮添加山楂叶黄酮和甜菊糖苷对68~72周龄蛋鸡的生产性能无显著影响,但能够改善蛋黄颜色、血清生化指标和肝脏脂肪沉积。 展开更多

The experiment was conducted to evaluate the differences of plant growth as well as sweetener material of two types of Stevia rebaudiana under conditions of absence and presence of drought stress. In vitro, the experiment was conducted as drought traits using four levels of polyethylene glycol (0.0, 10,000, 20,000, 30,000 ppm). It was observed that survival % and No. of shoots should be considered as the important factors associated with stevioside content in stevia plant. According to path coefficient analysis and positive direct effect of studied traits, survival % recorded the highest value (0.5386), whereas No. of shoots recorded the highest value (– 0.8827) as negative direct effect in related to stevioside content. The results showed that all traits were affected, especially under 30,000 ppm level of polyethylene glycol. Generally, the most of studies traits exhibited a recorded clearly difference between the two types and used drought levels on stevia plants. Therefore, selection based on survival % would be more effective to improving stevioside content of stevia plants in drought stress conditions. 展开更多

In order to improve the bioavailability of lutein(LUT),a novel lutein-stevio side nanoparticle(LUT-STE)were prepared previously,but the information about LUT-STE on protecting of eye health was limited.This study investigated the effect of LUT-STE on antioxidant activity of H_(2)O_(2)-induced human retinal pigment epithelial(ARPE)cells.LUT and LUT-STE(final concentration of 5μg/mL)significantly enhanced cell viability from(74.84±5.10)%to(81.92±10.01)%(LUT)and(89.33±4.34)%(LUT-STE),and inhibited the cell apoptosis(P<0.05).After pretreatment with LUT-STE in ARPE cells,the levels of superoxide dismutase(SOD),catalase(CAT)and glutathion peroxidase(GSH-Px)in ARPE cells were significantly increased(P<0.05),the contents of reactive oxygen species(ROS)and malondialdehyde(MDA)were decreased.In addition,the vascular endothelial growth factor(VEGF)levels were inhibited by 13.61%and 17.39%,respectively,pretreatment with LUT and LUT-STE.Western blotting results showed that the pretreatment with LUT-STE inhibited the expression of caspase-9 and caspase-3 and up-regulated Bcl-2/Bax pathway to inhibit H_(2)O_(2)-induced apoptosis.In summary,the novel delivery LUT-STE had more pronounced inhibitory effect on H_(2)O_(2)-induced damage in human ARPE cells. 展开更多

Stevioside, extracted from the leaves of Stevia rebaudiana Bertoni, is a natural, high intensity, lowcaloric sweetener with wide therapeutic activities. Conventional stevioside extraction methodologies involve the use of non-green solvents, supercritical fluids, microwaves, etc., however, all these processes are expensive, time-consuming and eco-unfriendly. Therefore an alternative process is desired for the isolation of stevioside. In this study, a novel enzyme-mediated extraction (EME) method has been developed. The dry stevia leaves were pre-treated with hydrolytic enzymes aided by transition metal salts (FeCl3). This was followed by pressurized hot water extraction (PHWE) to release stevioside. The crude extract was purified and clarified through multi-stage membrane filtration. The results confirm that metal salt-assisted cellulase pre-treatment enhanced the yield of stevioside to 72%, with 98% purity, which was higher in comparison to existing methods. Thus, the methodology developed establishes a simple, “green”, enzyme-mediated process for the efficient isolation of stevioside under economical and eco-friendly conditions. 展开更多

Determination of Rebaudioside A and Stevioside in Leaves of S.rebaudiana Bertoni Grown in Mexico by a Validated HPLC Method

Stevia rebaudiana is a plant with high sweetening capacity due to its content of glycosides, mainly stevioside and rebaudioside A. Several techniques have been used to determine the concentrations of glycosides in Stevia, although an HPLC method is recommended by the FAO/WHO-JECFA. Varieties of Stevia have been recently grown in Mexico, with no previous report of glycosides by a validated method. The aim of this study was to validate an isocratic HPLC method for content determination of main glycosides in the leaves of Stevia cultivated in Mexico. HPLC method was performed using a C18 column (250 mm × 4.6 mm, 5 μm) and UV detector set at 210 nm. The mobile phase consisted of 32:68 (v/v) mixture of acetonitrile and sodium-phosphate buffer (10 mmol/L, pH 2.6), set to a flow rate of 1.0 mL/min. Rebaudioside A and stevioside were determined in two Stevia varieties: Morita II and Criolla, and also validation parameters were calculated. Rebaudioside A content (g/100g) in Morita II was 15.15 ± 0.02 while stevioside was 3.97 ± 0.003;in the case of Criolla they were 4.03 ± 0.01 and 8.80 ± 0.14, respectively (p < 0.001). The recoveries of fortified samples were 100% ± 10% and precision RSD was ≤6.27%. The criteria of validation showed accuracy, linearity (≥0.99), and precision;therefore, the determination of glycosides was performed with reliability. 展开更多

选择硼、锌、镁肥进行复配,设置16个施肥处理分析了不同处理对甜叶菊农艺性状、生理指标、糖苷含量及产量的影响,为河西地区甜叶菊叶面喷施中微肥提供科学依据。结果表明,T7处理(硼、锌、镁肥施用量分别为7.5、7.5、22.5 kg/hm^(2))甜... 选择硼、锌、镁肥进行复配,设置16个施肥处理分析了不同处理对甜叶菊农艺性状、生理指标、糖苷含量及产量的影响,为河西地区甜叶菊叶面喷施中微肥提供科学依据。结果表明,T7处理(硼、锌、镁肥施用量分别为7.5、7.5、22.5 kg/hm^(2))甜叶菊株高最高、分枝数最多,较T16处理(对照,不施用硼、锌、镁肥)分别增加21.2%、92.5%。生物量方面,T4处理(硼、锌、镁肥施用量分别为22.5、7.5、7.5 kg/hm^(2))茎鲜质量最大,较对照增加116.4%;T7处理叶鲜质量和叶干质量最高,较对照分别增加139.6%和125.1%。生理指标方面,T8处理(硼、锌、镁肥施用量分别为7.5、7.5、7.5 kg/hm^(2))叶绿素总量和超氧化物歧化酶(SOD)活性最高,较对照分别提高18.3%和15.7%。糖苷累积方面,T5处理(硼、锌、镁肥施用量分别为7.5、22.5、22.5 kg/hm^(2))莱鲍迪苷A(RA)含量最高,较对照增加26.2%;T2处理(硼、锌、镁肥施用量分别为22.5、22.5、7.5 kg/hm^(2))甜菊苷(Stv)含量最高,较对照增加223.2%;T8处理总糖苷含量最高,较对照增加29.6%;T7处理实现了RA(83.13 g/kg)和Stv(34.20 g/kg)协同高产。产量显示,T7处理产量达11788.5 kg/hm^(2),较对照增加125.1%。相关性分析显示,RA含量与叶绿素总量、SOD活性呈显著正相关关系;Stv含量与叶绿素总量、SOD活性呈极显著负相关关系;总糖苷含量与SOD活性呈显著负相关关系;可溶性蛋白含量与叶绿素总量、SOD活性、过氧化物酶(POD)活性呈显著或极显著正相关关系;可溶性糖含量与POD活性呈显著正相关关系。方差分析显示,锌肥、镁肥对RA和Stv含量影响极显著,硼肥、锌肥对株高影响极显著,镁肥对叶绿素总量影响极显著。通过隶属函数法综合评价得出T7处理效果最优,即硼、锌、镁肥施用量分别为7.5、7.5、22.5 kg/hm^(2)时,最有利于促进甜叶菊的生长发育和品质提升。 展开更多

为了解决单一甜味剂在口感方面存在的不足,如甜味不纯正、明显的后余味和苦味等问题,通过采用单因素试验和正交试验将甜菊糖苷Reb-A(rebaudioside A)、赤藓糖醇、麦芽糖醇、L-谷氨酸钠和索马甜以不同比例混合优化,配制出口感近似蔗糖、... 为了解决单一甜味剂在口感方面存在的不足,如甜味不纯正、明显的后余味和苦味等问题,通过采用单因素试验和正交试验将甜菊糖苷Reb-A(rebaudioside A)、赤藓糖醇、麦芽糖醇、L-谷氨酸钠和索马甜以不同比例混合优化,配制出口感近似蔗糖、甜味纯正、营养、天然安全的复配甜味剂。结果表明,赤藓糖醇和麦芽糖醇对甜菊糖苷Reb-A的后余味和苦味有明显的改善作用,提高了其甜味品质。甜味蛋白质索马甜增加了复配产品的营养功能,而L-谷氨酸钠能够抑制甜菊糖苷Reb-A过高的甜味强度,并且改善了复配产品的风味。复配甜味剂的最佳复配质量分数为甜菊糖苷Reb-A 0.016%、赤藓糖醇1.2%、麦芽糖醇2.5%、索马甜0.003%、L-谷氨酸钠0.2%。通过与市场上已售其他复配型甜味剂的比较,该复配甜味剂具有甜味纯正、热量低、口感近似蔗糖、营养安全等优点,可作为一种家用普适性复配甜味剂推广使用。 展开更多

基于紫外可见-近红外光谱技术建立在线检测甜菊糖生产外排水中甜菊糖苷浓度的方法。采用透射方式获得系列浓度的甜菊糖生产外排水样品及光谱数据;借助主成分分析法剔除异常样本,采用D1、SG、SNV及其组合方法对原始光谱数据进行预处理;... 基于紫外可见-近红外光谱技术建立在线检测甜菊糖生产外排水中甜菊糖苷浓度的方法。采用透射方式获得系列浓度的甜菊糖生产外排水样品及光谱数据;借助主成分分析法剔除异常样本,采用D1、SG、SNV及其组合方法对原始光谱数据进行预处理;分析并选择不同预处理方法下载荷高、相关性高的变量作为特征变量,利用PLS方法建立甜菊糖苷外排水中TSG含量模型。结果表明:SNV+FD+SG预处理方法结合PLS因子载荷和Important Variables特征变量筛选方法建模效果最优,模型的RMSECV和RMSECP均≤0.0250,Rv^(2)和Rp^(2)均≥0.96,说明方法可以将甜菊糖生产外排水中的甜菊糖苷含量检测误差控制到±0.05%以内,并将甜菊糖苷生产外排水中甜菊糖苷含量波动范围由0.001~0.30 mg/mL,缩小至0.04~0.12 mg/mL,实现了甜菊糖吸附外排水中甜菊糖苷含量的实时监测和精准控制,有利于提高树脂柱利用率和减少甜菊糖苷外排,对植物提取行业吸附过程控制极具借鉴意义。 展开更多

研究壳聚糖絮凝剂在甜叶菊提取液澄清过程中的作用。分别以甜叶菊水提液的澄清率和甜菊苷的损失率为指标,考察不同因素对絮凝效果的影响。通过正交试验法确定壳聚糖絮凝的较优工艺条件为:1%壳聚糖溶液投加量2mL/g甜叶菊干叶、甜叶菊提... 研究壳聚糖絮凝剂在甜叶菊提取液澄清过程中的作用。分别以甜叶菊水提液的澄清率和甜菊苷的损失率为指标,考察不同因素对絮凝效果的影响。通过正交试验法确定壳聚糖絮凝的较优工艺条件为:1%壳聚糖溶液投加量2mL/g甜叶菊干叶、甜叶菊提取液质量浓度1g生药/10mL提取液、pH6.0、温度45℃,絮凝时间4h。在该条件下,甜叶菊水提液的澄清率为86.63%,甜菊苷的损失率为6.89%,与膜分离法澄清的效果相近,且无需特殊设备。与传统化学絮凝法相比,壳聚糖絮凝法能更有效地保留水提液中的有效成分,具有絮凝效果好、易生物降解、对环境安全、无二次污染等优点。 展开更多
