Evaluation of Sensitivities and Specificities of SARS-CoV Detection by Real-time Quantitative Reverse Transcription-PCR Assays

The etiological agent of severe acute respiratory syndrome(SARS) was identified as a new coronavirus,termed SARS-CoV. Establishment of an efficient and sensitive diagnostic system of SARS-CoV genetic materials is crucial for SARS control. In this study,we quantified SARS-CoV mRNAs in both infected cell culture lysate and in supernatant by using Real-time quantitative revere transcription-PCR based on EvaGreenTM dye and Taqman-MGB probes. For extensive evaluation of sensitivities and specificities,13 pairs of primers and 4 probes were designed based on different genes of SARS-CoV. Glyceraldehydes-3-phosphate dehydrogenase(GAPDH) was selected as the internal control gene. Results showed that S-gene-specific PCR was the most sensitive for detection,but because of its sequence variability in the different viral strains,primers and a probe based on the N gene were suitable substitutions. Meanwhile,we found the mRNA concentrations in cell culture lysates were much higher than in cell supernatant and facilited more sensitive detection of the SARS-CoV. 展开更多

In this work,we demonstrate that the strength of anion specificities of thermosensitive polymers is determined by the affinity of direct anion binding to the polymers.We have prepared a series of thermosensitive statistical copolymers with distinct thermoresponsive behaviors.The anions can specifically interact with the different types of thermosensitive polymers in very different strengths.A similar strength of specific anion effects on thermoresponsive behaviors can be observed at very different salt concentrations for the different types of thermosensitive polymers.A stronger anion binding to the thermosensitive polymers gives rise to a more obvious anion specificity and vice versa.The work presented here opens up opportunities for the application of ion binding affinity to modulate the strength of ion specificities of thermosensitive polymers. 展开更多

In this case, how and up to what point will they manage to collect all the data related there, to exploit them and to place them at the disposal of the public? We chose a methodology of investigation according to the research methods developed for the study of the industrial and agricultural systems (Bear, Gibbon, Haggblade, & Ritchie, 1993; Boomgard, Davies, Haggblade, & Mead, 1992; Laurent, 1993; Montigaud, 1992) in order to establish the cartography of the media, the actors, the sets of themes to the menu. The genus Actinidia is primarily functionally dioecious,and early sex identification plays a crucial role in improving breeding efficiency and reducing production costs.In this study,the accuracy of three sex-linked molecular markers(SyGI[Shy Girl],FrBy[Friendly Boy],and SmY1)in sex identification was evaluated in various Actinidia species.The selected marker products were subsequently cloned and sequenced in six wild Actinidia species.Ninety-six wild A.chinensis chinensis accessions and 74 A.chinensis deliciosa accessions,most of which were wild,with only one cultivated,were used for comprehensive primer validation.Thirty-three juvenile A.chinensis chinensis hybrid seedlings were used for practical application tests.The results showed that the marker SyGI accurately identified the sex of 20 samples from six Actinidia species and 96 A.chinensis chinensis accessions with 100%reliability.For Actinidia chinensis deliciosa,the identification accuracy reached 98.65%.Sequence analysis revealed that SyGI shared the highest similarity with the male-specific genomic region.Furthermore,SyGI achieved 100%accuracy in identifying the sex of 33 juvenile A.chinensis chinensis individuals.The findings confirm that the SyGI marker possesses high accuracy,strong specificity,and broad applicability,making it a valuable tool for kiwifruit breeding programs.The cloned sequences from wild Actinidia species also provide important references for future research on the mechanisms of sexual evolution and determination. 展开更多

This study investigates the performance boundaries of ramjet and scramjet engines fueled by boronbased propellant through full-scale engine modeling and three-dimensional computational fluid dynamics simulations.Results show that the performance boundary between ramjets and scramjets occurs near Mach 7.Specifically,at Mach 6,the ramjet exhibits a 1290 m/s higher specific impulse than the scramjet;however,at Mach 7,their performance becomes comparable.The ramjet's higher static temperature promotes boron particle vaporization and B_(2)O_(2) dissociation,limiting the total temperature increase,unlike in scramjets.The boron vapor mass fraction significantly impacts this temperature difference,with ramjets exhibiting values 8.5 and 3.9 times higher than scramjets at Mach 6 and Mach 7,respectively.Despite lower total temperatures,ramjets achieve more efficient boron combustion due to the combined effects of higher pressures and longer particle residence times.These findings offer valuable insights for engine designers in selecting ramjet or scramjet configurations for boron-fueled propulsion systems. 展开更多

Probiotics can regulate the body’s immune system through both non-specific and specific immunity,thereby regulating host health.In terms of non-specific immune regulation,probiotics can activate the intrinsic immune system,regulate the mucosal barrier function,and play an immune role by influencing the activity of intrinsic immune cells such as macrophages,dendritic cells and natural killer cells,as well as their differentiation and maturation;in terms of specific immune regulation,probiotics play a role in regulating the immunoglobulin level and the maturation of B cells.Probiotics can also regulate T-cell differentiation according to the condition of the body,thus regulating specific immunity.Many studies have focused on the role of probiotics in metabolism and nutrition,and the mechanisms involved in the immunomodulatory role of probiotics have only been partially described.This review summarises the role of common probiotics such as Lactobacillus plantarum and Lactobacillus rhamnosus in immunomodulation as well as their mechanisms,describing the currently known mechanisms of immunomodulation by probiotics in improving the host immune system.A deeper understanding of probiotics and their specific mechanisms of action will facilitate the use of probiotics for immunomodulation in clinical medicine,functional foods,and other areas.This will also contribute to the development and research of engineered probiotics,next-generation probiotics,and other new functional probiotics with immunomodulatory effects. 展开更多

The specific surface area(S S)and pore size(D)exhibit an inherent trade-off in the microscale design of bone implants:larger pores typically correlate with reduced surface area and vice versa.This relationship has attracted notable attention because of its critical role in the regulation of cell adhesion and osteogenesis.However,it remains largely unclear how S S and D affect the generated bone tissue and dynamically change during long-term osteogenesis.Herein,by applying rigorous geometric mapping to minimal surfaces,we constructed precisely partitioned and layer-by-layer thickened tissue models to simulate osteogenesis across different temporal scales and thereby track the dynamic evolution of geometric characteristics,permeability,and mechanobiological tissue differentiation.The high-S S samples were found to facilitate the rapid formation of new bone tissue in the early stages.However,their smaller pores tended to cause occlusions,hindering further tissue development.In contrast,low-S S samples showed slower bone regeneration,but their larger pores provided adequate physical space for tissue regeneration and mass transport,ultimately promoting bone formation in the long term.Mechanobiological regulation suggests that fibrous tissue formation inhibits additional bone formation,establishing a dynamic equilibrium between osteogenesis and pore space to sustain nutrient/waste exchange throughout the regenerative process.Overall,smaller pores are preferable in implants for minimally loaded osteoplasty procedures focused on early-stage bone consolidation,whereas larger pores are preferable in dynamically loaded implants requiring prolonged mechanical stability. 展开更多

The establishment of plantations has become a critical approach for reducing greenhouse gas emissions,particularly in fragile environments with carbon sequestration potential.In karst areas,plantations based on fastgrowing afforestation species made significant contributions to enhancing carbon sequestration.However,the impact of understory vegetation on carbon accumulation remains unclear.Especially,the carbon accumulation associated with litter produced during the replacement of understory species receives insufficient attention,which leads to the neglect of the carbon sequestration potential in plantations of karst areas.Leaf is a crucial organ that links the litter production.To explore how leaf traits adapt to competitive environments and drive litter carbon accumulation during understory species replacement,this study observed leaf traits and litter carbon content changes in three types of plantations in the Liujiang River Basin,a typical karst area.A total of 37 sampling plots were selected for field investigation over a twoyear period.Leaf traits,species diversity,vegetation coverage,and litter carbon characteristics in understory vegetation were measured.Variance analysis,allometric equations,and path analysis were used for data analysis.The results showed that most understory species adopted a biomass conservation strategy under high-coverage conditions(>44.27%)and expanded competitive leaf area under low-coverage conditions(<44.27%).However,Bidens pilosa and Miscanthus floridulus exhibited strong competitiveness during understory species replacement.They showed an expansion of competitive leaf area under high-coverage conditions.This competitive strategy reduced species diversity and community specific leaf area.But the rapid expansion of Bidens pilosa and Miscanthus floridulus increased understory vegetation coverage,and their increased specific leaf area facilitated leaf shedding,resulting in significant litter weight accumulation(P<0.05),thereby enhancing litter carbon content per unit area.These competitive strategies were key driving factors for the increase in litter carbon content per square meter,which reached a maximum of 49.6% higher than that in natural grasslands.And the maximum increase in litter carbon accumulation derived from understory vegetation reached 3.37 times from 2023 to 2024 in plantations.In the understory vegetation of plantations,the competitive strategies reflected by leaf adaptation of key competitive species are critical factors influencing litter carbon accumulation.Future research could deeply explore the carbon sequestration effects resulting from the dynamic changes in competition within the understory vegetation of plantations. 展开更多

With the rapid development of Internet technology,REST APIs(Representational State Transfer Application Programming Interfaces)have become the primary communication standard in modern microservice architectures,raising increasing concerns about their security.Existing fuzz testing methods include random or dictionary-based input generation,which often fail to ensure both syntactic and semantic correctness,and OpenAPIbased approaches,which offer better accuracy but typically lack detailed descriptions of endpoints,parameters,or data formats.To address these issues,this paper proposes the APIDocX fuzz testing framework.It introduces a crawler tailored for dynamic web pages that automatically simulates user interactions to trigger APIs,capturing and extracting parameter information from communication packets.A multi-endpoint parameter adaptation method based on improved Jaccard similarity is then used to generalize these parameters to other potential API endpoints,filling in gaps in OpenAPI specifications.Experimental results demonstrate that the extracted parameters can be generalized with 79.61%accuracy.Fuzz testing using the enriched OpenAPI documents leads to improvements in test coverage,the number of valid test cases generated,and fault detection capabilities.This approach offers an effective enhancement to automated REST API security testing. 展开更多

The brain's functions are governed by molecular metabolic networks.However,due to the sophisticated spatial organization and diverse activities of the brain,characterizing both the minute and large-scale metabolic activity across the entire brain and its numerous micro-regions remains incredibly challenging.Here,we offer a high-definition spatially resolved metabolomics technique to better understand the metabolic specialization and interconnection throughout the mouse brain using improved ambient mass spectrometry imaging.This method allows for the simultaneous mapping of thousands of metabolites at a 30 μm spatial resolution across the mouse brain,ranging from structural lipids to functional neurotransmitters.This approach effectively reveals the distribution patterns of delicate microregions and their distinctive metabolic characteristics.Using an integrated database,we annotated 259 metabolites,demonstrating that the metabolome and metabolic pathways are unique to each brain microregion.The distribution of metabolites,closely linked to functionally connected brain regions and their interactions,offers profound insights into the complexity of chemical processes and their roles in brain function.An initial dataset for future metabolomics research might be obtained from the high-definition mouse brain's spatial metabolome atlas. 展开更多

In wireless sensor networks,ensuring communication security via specific emitter identification(SEI)is crucial.However,existing SEI methods are limited to closed-set scenarios and lack the ability to detect unknown devices and perform classincremental training.This study proposes a class-incremental open-set SEI approach.The open-set SEI model calculates radiofrequency fingerprints(RFFs)prototypes for known signals and employs a self-attention mechanism to enhance their discriminability.Detection thresholds are set through Gaussian fitting for each class.For class-incremental learning,the algorithm freezes the parameters of the previously trained model to initialize the new model.It designs specific losses:the RFFs extraction distribution difference loss and the prototype transformation distribution difference loss,which force the new model to retain old knowledge while learning new knowledge.The training loss enables learning of new class RFFs.Experimental results demonstrate that the open-set SEI model achieves state-of-theart performance and strong noise robustness.Moreover,the class-incremental learning algorithm effectively enables the model to retain old device RFFs knowledge,acquire new device RFFs knowledge,and detect unknown devices simultaneously. 展开更多

A surface pyrolysis and gas-phase combustion of the Ammonium Perchlorate(AP)/Hydroxy Terminated Polybutadiene(HTPB)composite propellant reaction kinetic mechanism with five-step chemical reaction is adopted.The effects of helium injection on the burning rate and combustion of AP/HTPB propellant are analyzed in details,and the characteristics of motor performance are obtained.The numerical simulation results demonstrate that helium injection enhances the combustion chamber pressure,thereby increasing the burning rate of propellant.However,the primary combustion reaction of the AP/HTPB propellant takes place within a thin layer on the burning surface,so the low-temperature helium has minimal impact on the gasphase combustion.Ultimately,the helium not only elevates the nozzle exit velocity,resulting in specific impulse gain,but also reduces the exhaust plume temperature.With an increase of helium mass flow rate,the area of the velocity increase zone at the nozzle exit continuously decreases,but the average velocity in the motor exit continuously increases.Overall,when the helium flow rate is 2.5 kg/s,the specific impulse can reach 10.5%.Reducing the helium injection hole diameter enhances mixing of helium and combustion gas and expands the velocity increase zone,thereby maximizing the exit velocity gain in average velocity at the nozzle exit.When the injection hole diameter is reduced from 100 mm to 20 mm,the specific impulse gain increases from 3.1%to 10.6%.Furthermore,increasing helium injection temperature greatly boosts the velocity of the mixed gas with the same helium mass fraction ultimately improving specific impulse. 展开更多

Hierarchical porous MgO is a promising adsorbent for dye removal because of its large Brunauer-Emmett-Teller specific surface area(SBET)and abundant low-coordinated oxygen anions(LCO)sites.As hierarchical porous MgO particles with large SBET values are typically prepared by complicated procedures under harsh conditions,such as high temperatures and high pressures,their large-scale production is impractical.Consequently,the preparation of hierarchical porous MgO with a large SBET value under mild conditions is highly desirable.In this study,a morphology transition engineering strategy is introduced to change the morphology of simple MgO microspheres to an embroidered ball-shaped with a larger SBET value via hydrolysis and calcination without using surfactants as sacrificial templates.During hydrolysis,numerous Mg(OH)_(2)sheets form and attach to the MgO surface,thus increasing the SBET value of the newly obtained MgO that forms by calcination(denoted as NM-x,where x is the hydrolysis time in hours).The sizes of the crystalline sheets were tuned by controlling the hydrolysis time.NM-12 exhibited the highest density of small-sized sheets on its surface and the largest SBET value of 180.17 m^(2)g^(-1),which was 3.51 times that of the MgO precursor(51.89 m^(2)g^(-1)).However,NM-24(134.07 m^(2)g^(-1))had a higher adsorption efficiency for Congo red(CR)than NM-12,despite having a smaller SBET value,which indicates that other factors are involved.NM-24 exhibited a lower probability of exposed(200)and(220)facets which were verified to repulse CR molecules by molecular dynamics simulations,and a greater number of LCO sites,which contributed to adsorption.Thus,this study introduces a facile method for preparing hierarchical porous MgO and examines the effects of LCO sites and exposed facet probabilities on its adsorption properties. 展开更多

Coconut(Cocos nucifera L.),a major oil and fruit crop of the Arecaceae family,is extensively cultivated across the Asia—Pacific region.Despite its agricultural importance,genome assembly in coconut remains challenging due to its large genome size and high proportion of repetitive sequences.Allele-specific expression(ASE)plays a key role in regulating plant development and evolution,yet research on ASE in coconut is limited(Shao et al.,2019;Li et al.,2021;Zhang et al.,2021;Hu et al.,2022).Among phenotypic traits,fruit color is especially important as an indicator of maturity,guiding harvest timing and post-harvest processes(Kapoor et al.,2022).While prior studies have explored various coconut traits such as salt tolerance,fiber content,and plant height(Wang et al.,2021;Yang et al.,2021),investigations into ASE and fruit color remain scarce. 展开更多

Dear Editor,Social anxiety disorder(SAD)is a prevalent psychiatric disorder,characterized by persistent and excessive fear of social situations in which the individual may be scrutinized by others[1].Typically emerging in late childhood or adolescence with potential lifelong persistence,SAD significantly impairs interpersonal relationships,educational attainment,and occupational performance[2].Furthermore,SAD demonstrates high comorbidity rates with other mental disorders,including anxiety disorders,substanceuse disorder,and major depressive disorder[3].Current therapeutic interventions remain limited,with suboptimal response to existing psychological and pharmacological treatments in many patients,while the pathological mechanisms underlying SAD continue to elude comprehensive understanding[4].Establishing appropriate animal models is essential to investigating SAD's neural mechanisms and pathological underpinnings and developing effective therapeutic interventions. 展开更多

Prostate-specific membrane antigen(PSMA)is a surface membrane antigen that is highly overexpressed in prostate cancer,with heterogenous expression throughout the natural history of the disease.This has generated significant interest as a potential biomarker for use in early diagnosis and treatment of prostate cancer.We reviewed the literature surrounding PSMA and its current clinical applications in diagnosing and managing early prostate cancer that is confined to the prostate and local lymph nodes.A search on PubMed,Medline,and Web of Science was performed using the following keywords:“PSMA”,“Prostate Specific Membrane Antigen”,“Prostate cancer”,“Biomarker”,“Diagnosis”.We considered all available articles relevant to the topic of PSMA as a biomarker in early prostate cancer when developing this narrative review.Key articles assessing the biology of PSMA,as well as its use as a potential diagnostic and therapeutic target in early prostate cancer,were assessed.The role of PSMA PET as a potential diagnostic and risk stratification tool was assessed.The current use of antibody-drug conjugates and radioligand therapy targeting PSMA was assessed,along with any current evidence to support their use in early prostate cancer.PSMA is heavily expressed throughout the early stages of prostate cancer,and this has significant therapeutic implications.There is a growing body of evidence that shows PSMA PET can play a role in the diagnosis,risk stratification,and prognostication of localised prostate cancer.PSMA-targeted therapies such as Lu-177 currently do not have any proven benefit in treating early prostate cancer;however,this remains an area of ongoing research. 展开更多

Alzheimer’s disease(AD)is the most common form of dementia.In addition to the lack of effective treatments,there are limitations in diagnostic capabilities.The complexity of AD itself,together with a variety of other diseases often observed in a patient’s history in addition to their AD diagnosis,make deciphering the molecular mechanisms that underlie AD,even more important.Large datasets of single-cell RNA sequencing,single-nucleus RNA-sequencing(snRNA-seq),and spatial transcriptomics(ST)have become essential in guiding and supporting new investigations into the cellular and regional susceptibility of AD.However,with unique technology,software,and larger databases emerging;a lack of integration of these data can contribute to ineffective use of valuable knowledge.Importantly,there was no specialized database that concentrates on ST in AD that offers comprehensive differential analyses under various conditions,such as sex-specific,region-specific,and comparisons between AD and control groups until the new Single-cell and Spatial RNA-seq databasE for Alzheimer’s Disease(ssREAD)database(Wang et al.,2024)was introduced to meet the scientific community’s growing demand for comprehensive,integrated,and accessible data analysis. 展开更多

Despite the superior advantages of specific emitter identification in extracting emitter features from in-phase and quadrature(I/Q)signals,challenges persist due to signal-type confusion and background noise interference.To address those limitations,this paper proposes a multi-channel contrast prediction coding and complex-valued residuals network(MCPC-MCVResNet)framework.This model employs contrast prediction techniques to directly extract discriminative features from electromagnetic signal sequences,effectively capturing both amplitude and phase information within I/Q data.A core innovation of this approach is the sphere space softmax(SS-softmax)loss,which optimizes intra-class clustering density of while establishing well-defined boundaries between distinct emitters.The SS-softmax mechanism significantly enhances the model's capacity to discern subtle variations among radiation emitters.Experimental results demonstrate superior identification accuracy,rapid convergence,and exceptional robustness in low signal-to-noise ratio environments. 展开更多
